The quantum yield for CO2 uptake was measured in C3 and C4 monocot species from several different grassland habitats. When the quantum yield was measured in the presence of 21% O2 and 340 cm3 m-3 CO2, values were very similar in C3 monocots, C3 dicots, and C4 monocots (0.045–0.056 mole CO2 · mole-1 quanta absorbed). In the presence of 2% O2 and 800 cm3 m-3 CO2, enhancements of the quantum yield values occurred for the C3 plants (both monocots and dicots), but not for C4 monocots. A dependence of the quantum yield on leaf temperature was observed in the C3 grass, Agropyron smithii, but not in the C4 grass, Bouteloua gracilis, in 21% O2 and 340 cm3 m-3 CO2. At leaf temperatures between 22–25°C the quantum yield values were approximately equal in the two species. 相似文献

Species in the Laxa and Grandia groups of the genus Panicum are adapted to low, wet areas of tropical and subtropical America. Panicum milioides is a species with C3 photosynthesis and low apparent photorespiration and has been classified as a C3/C4 intermediate. Other species in the Laxa group are C3 with normal photorespiration. Panicum prionitis is a C4 species in the Grandia group. Since P. milioides has some leaf characteristics intermediate to C3 and C4 species, its photosynthetic response to irradiance and temperature was compared to the closely related C3 species, P. laxum and P. boliviense and to P. prionitis. The response of apparent photosynthesis to irradiance and temperature was similar to that of P. laxum and P. boliviense, with saturation at a photosynthetic photo flux density of about 1 mmol m-2 s-1 at 30°C and temperature optimum near 30°C. In contrast, P. prionitis showed no light saturation up to 2 mmol m-2 s-1 and an optimum temperature near 40°C. P. milioides exhibited low CO2 loss into CO2-free air in the light and this loss was nearly insensitive to temperature. Loss of CO2 in the light in the C3 species, P. laxum and P. boliviense, was several-fold higher than in P. milioides and increased 2- to 5-fold with increases in temperature from 10 to 40°C. The level of dark respiration and its response to temperature were similar in all four Panicum species examined. It is concluded that the low apparent photorespiration in P. milioides does not influence its response of apparent photosynthesis to irradiance and temperature in comparison to closely related C3 Panicum species.Abbreviations AP

The potential for C4 photosynthesis was investigated in five C3-C4 intermediate species, one C3 species, and one C4 species in the genus Flaveria, using 14CO2 pulse-12CO2 chase techniques and quantum-yield measurements. All five intermediate species were capable of incorporating 14CO2 into the C4 acids malate and aspartate, following an 8-s pulse. The proportion of 14C label in these C4 products ranged from 50–55% to 20–26% in the C3-C4 intermediates F. floridana Johnston and F. linearis Lag. respectively. All of the intermediate species incorporated as much, or more, 14CO2 into aspartate as into malate. Generally, about 5–15% of the initial label in these species appeared as other organic acids. There was variation in the capacity for C4 photosynthesis among the intermediate species based on the apparent rate of conversion of 14C label from the C4 cycle to the C3 cycle. In intermediate species such as F. pubescens Rydb., F. ramosissima Klatt., and F. floridana we observed a substantial decrease in label of C4-cycle products and an increase in percentage label in C3-cycle products during chase periods with 12CO2, although the rate of change was slower than in the C4 species, F. palmeri. In these C3-C4 intermediates both sucrose and fumarate were predominant products after a 20-min chase period. In the C3-C4 intermediates, F. anomala Robinson and f. linearis we observed no significant decrease in the label of C4-cycle products during a 3-min chase period and a slow turnover during a 20-min chase, indicating a lower level of functional integration between the C4 and C3 cycles in these species, relative to the other intermediates. Although F. cronquistii Powell was previously identified as a C3 species, 7–18% of the initial label was in malate+aspartate. However, only 40–50% of this label was in the C-4 position, indicating C4-acid formation as secondary products of photosynthesis in F. cronquistii. In 21% O2, the absorbed quantum yields for CO2 uptake (in mol CO2·[mol quanta]-1) averaged 0.053 in F. cronquistii (C3), 0.051 in F. trinervia (Spreng.) Mohr (C4), 0.052 in F. ramosissima (C3-C4), 0.051 in F. anomala (C3-C4), 0.050 in F. linearis (C3-C4), 0.046 in F. floridana (C3-C4), and 0.044 in F. pubescens (C3-C4). In 2% O2 an enhancement of the quantum yield was observed in all of the C3-C4 intermediate species, ranging from 21% in F. ramosissima to 43% in F. pubescens. In all intermediates the quantum yields in 2% O2 were intermediate in value to the C3 and C4 species, indicating a co-function of the C3 and C4 cycles in CO2 assimilation. The low quantum-yield values for F. pubescens and F. floridana in 21% O2 presumably reflect an ineffcient transfer of carbon from the C4 to the C3 cycle. The response of the quantum yield to four increasing O2 concentrations (2–35%) showed lower levels of O2 inhibition in the C3-C4 intermediate F. ramosissima, relative to the C3 species. This indicates that the co-function of the C3 and C4 cycles in this intermediate species leads to an increased CO2 concentration at the site of ribulose-1,5-bisphosphate carboxylase/oxygenase and a concomitant decrease in the competitive inhibition by O2.Abbreviations PEP

The light dependence of quantum yields of Photosystem II (II) and of CO2 fixation were determined in C3 and C4 plants under atmospheric conditions where photorespiration was minimal. Calculations were made of the apparent quantum yield for CO2 fixation by dividing the measured rate of photosynthesis by the absorbed light [A/I=CO2 and of the true quantum yield by dividing the estimated true rate of photosynthesis by absorbed light [(A+Rl)/Ia=CO2·], where RL is the rate of respiration in the light. The dependence of the II/CO2 and II/CO2
* ratios on light intensity was then evaluated. In both C3 and C4 plants there was little change in the ratio of II/CO2 at light intensities equivalent to 10–100% of full sunlight, whereas there was a dramatic increase in the ratio at lower light intensities. Changes in the ratio of II/CO2 can occur because respiratory losses are not accounted for, due to changes in the partitioning of energy between photosystems or changes in the relationship between PS II activity and CO2 fixation. The apparent decrease in efficiency of utilization of energy derived from PS II for CO2 fixation under low light intensity may be due to respiratory loss of CO2. Using dark respiration as an estimate of RL, the calculated II/CO2
* ratio was nearly constant from full sunlight down to approx 5% of full sunlight, which suggests a strong linkage between the true rate of CO2 fixation and PS II activity under varying light intensity. Measurements of photosynthesis rates and II were made by illuminating upper versus lower leaf surfaces of representative C3 and C4 monocots and dicots. With the monocots, the rate of photosynthesis and the ratio of II/CO2 exhibited a very similar patterns with leaves illuminated from the adaxial versus the abaxial surface, which may be due to uniformity in anatomy and lack of differences in light acclimation between the two surfaces. With dicots, the abaxial surface had both lower rates of photosynthesis and lower II values than the adaxial surface which may be due to differences in anatomy (spongy versus palisade mesophyll cells) and/or light acclimation between the two surfaces. However, in each species the response of II/CO2 to varying light intensity was similar between the two surfaces, indicating a comparable linkage between PS II activity and CO2 fixation.Abbreviations A

Grasses using the C4 photosynthetic pathway dominate grasslands and savannahs of warm regions, and account for half of the species in this ecologically and economically important plant family. The C4 pathway increases the potential for high rates of photosynthesis, particularly at high irradiance, and raises water-use efficiency compared with the C3 type. It is therefore classically viewed as an adaptation to open, arid conditions. Here, we test this adaptive hypothesis using the comparative method, analysing habitat data for 117 genera of grasses, representing 15 C4 lineages. The evidence from our three complementary analyses is consistent with the hypothesis that evolutionary selection for C4 photosynthesis requires open environments, but we find an equal likelihood of C4 evolutionary origins in mesic, arid and saline habitats. However, once the pathway has arisen, evolutionary transitions into arid habitats occur at higher rates in C4 than C3 clades. Extant C4 genera therefore occupy a wider range of drier habitats than their C3 counterparts because the C4 pathway represents a pre-adaptation to arid conditions. Our analyses warn against evolutionary inferences based solely upon the high occurrence of extant C4 species in dry habitats, and provide a novel interpretation of this classic ecological association. 相似文献

In this report, the effects of light on the activity and allosteric properties of phosphoenolpyruvate (PEP) carboxylase were examined in newly matured leaves of several C3 and C4 species. Illumination of previously darkened leaves increased the enzyme activity 1.1 to 1.3 fold in C3 species and 1.4 to 2.3 fold in C4 species, when assayed under suboptimal conditions (pH 7) without allosteric effectors. The sensitivities of PEP carboxylase to the allosteric effectors malate and glucose-6-phosphate were markedly different between C3 and C4 species. In the presence of 5 mM malate, the activity of the enzyme extracted from illuminated leaves was 3 to 10 fold higher than that from darkened leaves in C4 species due to reduced malate inhibition of the enzyme from illuminated leaves, whereas it increased only slightly in C3 species. The Ki(malate) for the enzyme increased about 3 fold by illumination in C4 species, but increased only slightly in C3 species. Also, the addition of the positive effector glucose-6-phosphate provided much greater protection against malate inhibition of the enzyme from C4 species than C3 species. Feeding nitrate to excised leaves of nitrogen deficient plants enhanced the degree of light activation of PEP carboxylase in the C4 species maize, but had little or no effect in the C3 species wheat. These results suggest that post-translational modification by light affects the activity and allosteric properties of PEP carboxylase to a much greater extend in C4 than in C3 species. 相似文献

The activities of the carboxylating enzymes ribulose-1,5-biphosphate (RuBP) carboxylase and phosphoenolpyruvate (PEP) carboxylase in leaves of three-week old Zea mays plants grown under phytotron conditions were found to vary according to leaf position. In the lower leaves the activity of PEP carboxylase was lower than that of RuBP carboxylase, while the upper leaves exhibited high levels of PEP carboxylase. Carbon dioxide compensation points and net photosynthetic rates also differed in the lower and upper leaves. Differences in the fine structure of the lowermost and uppermost leaves are shown. The existence of both the C3 and C4 photosynthetic pathways in the same plant, in this and other species, is discussed.Abbreviations PEP

Photosynthetic characteristics and tolerance to photo-oxidation of transgenic rice expressing C4 photosynthesis enzymes 总被引:16,自引:0,他引:16
The photosynthetic characteristics of four transgenic rice lines over-expressing rice NADP-malic enzyme (ME), and maize phosphoenolpyruvate carboxylase (PC), pyruvate,orthophosphate dikinase (PK), and PC+PK (CK) were investigated using outdoor-grown plants.
Relative to untransformed wild-type (WT) rice, PC transgenic rice exhibited high PC activity (25-fold increase) and enhanced
activity of carbonic anhydrase (more than two-fold increase), while the activity of ribulose-bisphosphate carboxylase/oxygenase
(Rubisco) and its kinetic property were not significantly altered. The PC transgenic plants also showed a higher light intensity
for saturation of photosynthesis, higher photosynthetic CO2 uptake rate and carboxylation efficiency, and slightly reduced CO2 compensation point. In addition, chlorophyll a fluorescence analysis indicates that PC transgenic plants are more tolerant to photo-oxidative stress, due to a higher capacity
to quench excess light energy via photochemical and non-photochemical means. Furthermore, PC and CK transgenic rice produced
22–24% more grains than WT plants. Taken together, these results suggest that expression of maize C4 photosynthesis enzymes in rice, a C3 plant, can improve its photosynthetic capacity with enhanced tolerance to photo-oxidation.

Phosphoenolpyruvate carboxylase (PEPCase, EC 4.1.1.3) is a key enzyme of C4 photosynthesis. It has evolved from ancestral non-photosynthetic (C3) isoforms and thereby changed its kinetic and regulatory properties. We are interested in understanding the molecular changes,
as the C4 PEPCases were adapted to their new function in C4 photosynthesis and have therefore analysed the PEPCase genes of various Alternanthera species. We isolated PEPCase cDNAs from the C4 plant Alternanthera pungens H.B.K., the C3/C4 intermediate plant A. tenella Colla, and the C3 plant A. sessilis (L.) R.Br. and investigated the kinetic properties of the corresponding recombinant PEPCase proteins and their phylogenetic
relationships. The three PEPCases are most likely derived from orthologous gene classes named ppcA. The affinity constant for the substrate phosphoenolpyruvate (K
0.5 PEP) and the degree of activation by glucose-6-phosphate classified the enzyme from A. pungens (C4) as a C4 PEPCase isoform. In contrast, both the PEPCases from A. sessilis (C3) and A. tenella (C3/C4) were found to be typical C3 PEPCase isozymes. The C4 characteristics of the PEPCase of A. pungens were accompanied by the presence of the C4-invariant serine residue at position 775 reinforcing that a serine at this position is essential for being a C4 PEPCase (Svensson et al. 2003). Genomic Southern blot experiments and sequence analysis of the 3′ untranslated regions of these genes indicated the existence

The leaf ultrastructure of NADP-malic enzyme type C4 species possessing different anatomical features in the Cyperaceae was examined: types were the Rhynchosporoid type, a normal
Kranz type in which mesophyll cells are adjacent to Kranz cells, and Fimbristyloid and Chlorocyperoid types, unusual Kranz
types in which nonchlorophyllous mestome sheath intervenes between the two types of green cells. They show structural characteristics
basically similar to the NADP-malic enzyme group of C4 grasses, that is, centrifugally located chloroplasts with reduced grana and no increase of mitochondrial frequency in the
Kranz cells. However, the Kranz cell chloroplasts of the Fimbristyloid and Chlorocyperoid types exhibit convoluted thylakoid
systems and a trend of extensive development of peripheral reticulum, although those of the Rhynchosporoid type do not possess
such particular membrane systems. The suberized lamella, probably a barrier for CO2 diffusion, is present in the Kranz cell walls of the Rhynchosporoid type and in the mestome sheath cell walls of the other
two types, and tightly surrounds the Kranz cells (sheaths) that are the sites of the decarboxylation of C4 acids. These ultrastructural features are discussed in relation to C4 photosynthetic function. 相似文献

A computer model comprising light reactions in PS II and PS I, electron-proton transport reactions in mesophyll and bundle
sheath chloroplasts, all enzymatic reactions and most of the known regulatory functions of NADP-ME type C4 photosynthesis has been developed as a system of differential budget equations for intermediate compounds. Rate-equations
were designed on principles of multisubstrate-multiproduct enzyme kinetics. Some of the 275 constants needed (ΔG0′ and K
m values) were available from literature and others (V
m) were estimated from reported rates and pool sizes. The model provided good simulations for rates of photosynthesis and pool
sizes of intermediates under varying light, CO2 and O2. A basic novelty of the model is coupling of NADPH production via NADP-ME with ATP production and regulation of the C3 cycle in bundle sheath chloroplasts. The functional range of the ATP/NADPH ratio in bundle sheath chloroplasts extends from
1.5 to 2.1, being energetically most efficient around 2. In the presence of such stoichiometry, the CO2 concentrating function can be explained on the basis of two processes: (a) extra ATP consumption for starch and protein synthesis
in bundle sheath leads to a faster NADPH and CO2 import compared with CO2 fixation in bundle sheath, and (b) the residual photorespiratory activity consumes RuBP by oxygenation, NADPH and ATP and
causes the imported CO2 to accumulate in bundle sheath cells. As a wider application, the model may be used for predicting results of genetic engineering
of plants.

Most species of the genus Salsola (Chenopodiaceae) that have been examined exhibit C4 photosynthesis in leaves. Four Salsola species from Central Asia were investigated in this study to determine the structural and functional relationships in photosynthesis of cotyledons compared to leaves, using anatomical (Kranz versus non-Kranz anatomy, chloroplast ultrastructure) and biochemical (activities of photosynthetic enzymes of the C3 and C4 pathways, 14C labeling of primary photosynthesis products and 13C/12C carbon isotope fractionation) criteria. The species included S. paulsenii from section Salsola, S. richteri from section Coccosalsola, S. laricina from section Caroxylon, and S. gemmascens from section Malpigipila. The results show that all four species have a C4 type of photosynthesis in leaves with a Salsoloid type Kranz anatomy, whereas both C3 and C4 types of photosynthesis were found in cotyledons. S. paulsenii and S. richteri have NADP- (NADP-ME) C4 type biochemistry with Salsoloid Kranz anatomy in both leaves and cotyledons. In S. laricina, both cotyledons and leaves have NAD-malic enzyme (NAD-ME) C4 type photosynthesis; however, while the leaves have Salsoloid type Kranz anatomy, cotyledons have Atriplicoid type Kranz anatomy. In S. gemmascens, cotyledons exhibit C3 type photosynthesis, while leaves perform NAD-ME type photosynthesis. Whereas leaf gas exchange properties are important to assess carbon and water fluxes in ecosystems worldwide, information of this type is scarce for savanna species. In this study, gas exchange characteristics of 2 C4 grass species (Andropogon canaliculatus and Hyparrhenia diplandra) and 2 C3 tree species (Crossopteryx febrifuga and Cussonia arborea) from the West-African savanna of Lamto (Ivory Coast) were investigated in the field. Measurements were done in order to provide data to allow the parameterization of biochemically-based models of photosynthesis (for C4 and C3 plant metabolic types) and stomatal conductance ; and to compare gas exchange characteristics of coexisting species. No systematic difference was found between grass and tree species for reference stomatal conductance, under standard environmental conditions, or stomatal response to incident light or vapour pressure deficit at leaf surface. Conversely, grass species displayed higher water (1.5-2 fold) and nitrogen (2-5 fold) photosynthetic use efficiencies (WUE and NUE, ratio of net photosynthesis to transpiration and leaf nitrogen, respectively). These contrasts were attributed to the CO2 concentrating mechanism of C4 plants. When looking within plant life forms, no important difference was found between grass species. However, significant contrasts were found between tree species, Cussonia showing higher NUE and reference stomatal conductance than Crossopteryx. These results stress the need to account for functional diversity when estimating ecosystem carbon and water fluxes. In particular, our results suggest that the tree/grass ratio, and also the composition of the tree layer, could strongly affect WUE and NUE at the ecosystem scale in West African savannas. 相似文献

C4 photosynthetic pathway and life form were determined for 159 species in 71 genera and 13 families in the grassland of North China. 45 % of the C4 species were found in Graminae, 19 % in each of Cyperaceae and Chenopodiaceae. More than 51 % of these C4 species were in therophyta and 36 % hemicryptophyta, while fewer species were in nanophanerophyta (9 %) or geophyta (5 %). The numbers of C4 species and their life forms were closely related with grassland deterioration and succession in North China. This indicated that the C4 species had greater capacity to tolerate environmental stress (e.g. drought and salinity) caused by animal grazing and cultivation. 相似文献

The aim of this work was to describe the photosynthetic carbon metabolism of the cooltemperate C4 grass Spartina anglica. With the exception of pyruvate, phosphate dikinase and pyruvate kinase, the maximum catalytic activities in leaves of putative enzymes of the C4 cycle of a phosphoenolpyruvate-carboxykinase C4 plant were considerably in excess of the observed, steady-state rate of photosynthesis, and were comparable with the maximum catalytic activities of key enzymes of the reductive pentose-phosphate pathway. Radioactive carbon from 14CO2 supplied to attached leaves during steady-state photosynthesis appeared first in malate and aspartate from which it moved to intermediates of the reductive pentose-phosphate pathway, and then to sucrose. These experiments show that photosynthetic carbon metabolism in this cool-temperate C4 plant is similar to that of C4 plants of hotter climates. 相似文献

Summary The growth and photosynethetic responses to atmospheric CO2 enrichment of 4 species of C4 grasses grown at two levels of irradiance were studied. We sought to determine whether CO2 enrichment would yield proportionally greater growth enhancement in the C4 grasses when they were grown at low irradiance than when grown at high irradiance. The species studied were Echinochloa crusgalli, Digitaria sanguinalis, Eleusine indica, and Setaria faberi. Plants were grown in controlled environment chambers at 350, 675 and 1,000 l 1-1 CO2 and 1,000 or 150 mol m-2 s-1 photosynthetic photon flux density (PPFD). An increase in CO2 concentration and PPFD significantly affected net photosynthesis and total biomass production of all plants. Plants grown at low PPFD had significantly lower rates of photosynthesis, produced less biomass, and had reduced responses to increases in CO2. Plants grown in CO2-enriched atmosphere had lower photosynthetic capacity relative to the low CO2 grown plants when exposed to lower CO2 concentration at the time of measurement, but had greater rate of photosynthesis when exposed to increasing PPFD. The light level under which the plants were growing did not influence the CO2 compensation point for photosynthesis. 相似文献

Morphological and physiological measurements on individual leaves of Leucaena leucocephala seedlings were used to study acclimation to neutral shading. The light-saturated photosynthetic rate (Pn max) ranged from 19.6 to 6.5 mol CO2 m–2 s–1 as photosynthetic photon flux density (PPFD) during growth decreased from 27 to 1.6 mol m–2 s–1. Stomatal density varied from 144 mm–2 in plants grown in high PPFD to 84 mm–2 in plants grown in low PPFD. Average maximal stomatal conductance for H2O was 1.1 in plants grown in high PPFD and 0.3 for plants grown in low PPFD. Plants grown in low PPFD had a greater total chlorophyll content than plants grown in high PPFD (7.2 vs 2.9 mg g–1 on a unit fresh weight basis, and 4.3 vs 3.7 mg dm–2 on a unit leaf area basis). Leaf area was largest when plants were grown under the intermediate PPFDs. Leaf density thickness was largest when plants were grown under the largest PPFDs. Carbon-isotope ratios were examined as 13C values in several C3, C4, and C3–C4
Flaveria species, and compared to predicted 13C, values generated from theoretical models. The measured 13C values were within 4 of those predicted from the models. The models were used to identify factors that contribute to C3-like 13C values in C3–C4 species that exhibit considerable C4-cycle activity. Two of the factors contributing to C3-like 13C values are high CO2 leakiness from the C4 pathway and pi/pa values that were higher than C4 congeners. A marked break occurred in the relationship between the percentage of atmospheric CO2 assimilated through the C4 cycle and the 13C value. Below 50% C4-cycle assimialtion there was no significant relationship between the variables, but above 50% the 13C values became less negative. These results demonstrate that the level of C4-cycle expression can increase from, 0 to 50% with little integration of carbon transfer from the C4 to the C3 cycle. The photosynthetic response of juveniles of Decussocarpus rospigliosii, an emergent primary forest species and shade tolerant in its juvenile stages and Alchornea triplinervia, a gap-colonizing species of tropical cloud forest in Venezuela was studied. Daily courses of microenvironmental variables and gas exchange under contrasting light conditions (gap and understory) were carried out in their natural environment and transplanted to different light regimes (shade and sun) in the field. The photosynthetic response and some anatomical characteristics of plants from different treatments were analyzed in the laboratory. Photosynthetic rates were low for both species, and were negative during some diurnal periods, related to the low photosynthetically active radiation levels obtained at both gap (6% of total radiation) and understory (2%). A. triplinervia shows higher rates (1.5–3.0 molm-2
-1) than D. rospigliosii (0.7–1.1 molm-2s-1). Both species showed increased photosynthetic rates when grown in gaps. A. triplinervia did not adjust its maximum photosynthetic rates to the prevailing light conditions. In contrast, D. rospigliosii responded to increased light levels. Both species showed low light compensation points when grown under total shade. There was a partial stomatal closure generally during midday in D. rospigliosii. A. triplinervia presented lower leaf conductances, transpiration rates and lesser stomatal control. Some leaf anatomical characteristics, in both species, were affected by variations in the light regime (i.e. increased leaf thickness, leaf specific weight and stomatal density). These results suggest that both species have the ability to respond to variations in their natural light environments, therefore maintaining a favorable carbon balance during the day. 相似文献
