The effects of acetate and butyrate during glycerol fermentation to 1,3-propanediol at pH 7.0 by Clostridium butyricum CNCM 1211 were studied. At pH 7.0, the calculated quantities of undissociated acetic and butyric acids were insufficient to inhibit bacterial growth. The initial addition of acetate or butyrate at concentrations of 2.5 to 15 gL−1 had distinct effects on the metabolism and growth of Clostridium butyricum. Acetate increased the biomass and butyrate production, reducing the lag time and 1,3-propanediol production. In contrast, the addition of butyrate induced an increase in 1,3-propanediol production (yield: 0.75 mol/mol glycerol, versus 0.68 mol/mol in the butyrate-free culture), and reduced the biomass and butyrate production. It was calculated that reduction of butyrate production could provide sufficient NADH to increase 1,3-propanediol production. The effects of acetate and butyrate highlight the metabolic flexibility of Cl. butyricum CNCM 1211 during glycerol fermentation. Received: 2 January 2001 / Accepted: 6 February 2001  相似文献

Summary When an aggregate-forming variant of Clostridium butyricum was grown in continuous culture under glucose-limited conditions (pH 5.2, dilution rate = 0.1 h–1, 30° C), glucose was mainly fermented to isopropanol and butanol. Production of solvents was pH dependent and initiated above a threshold concentration of acetic and butyric acid, which probably means that it was triggered by the undissociated concentration of these acids. This was confirmed by studies with a second reactor. Solvent productivity by this variant was found to be stable and the highest ever reported for Clostridium sp. in argued that, due to the aggregated growth mode, this variant is able to reach the threshold concentration of acids at which solventogenesis is initiated at high dilution rates.Offprint requests to: G. 【目的】目前,国内外鲜有关于羊源丁酸梭菌的报道。本课题选用羊源丁酸梭菌HDRy YB1为研究对象,对其发酵工艺进行优化,为该菌株作为饲料添加剂应用于畜牧业生产奠定基础。【方法】采用Plackett-Burman(PB)试验设计法和响应面法分析并优化显著影响HDRy YB1菌株发酵液中芽胞数的培养基成分。【结果】发酵培养基中的面粉浓度、鱼粉浓度和米粉浓度显著影响发酵液中的芽胞数,优化后的发酵培养基组分(质量体积比)为:面粉3.72%、鱼粉0.90%、米粉3.96%、酵母粉0.60%、Na Cl 0.19%、Mg SO4·7H2O 0.19%、KH2PO4 0.01%、Na HCO3 0.01%、Ca CO3 0.48%;培养参数为:37°C,初始p H为7.2-7.4,瓶装量100/250,接种量3%。在此条件下,HDRy YB1菌株发酵完全(18 h)的芽胞数为1.478×108 CFU/m L,是优化前的2.7倍。【结论】HDRy YB1菌株发酵培养基得到了优化,优化后的培养基可用于后期的扩大发酵试验,验证其在实践生产中的应用价值。  相似文献

The ability of Clostridium butyricum cultures in synthetic medium to hydrolyse the tripeptide Leu-Val-Leu was demonstrated using gas chromatography coupled with mass spectrometry. Electrospray mass spectrometry was used to confirm this unknown activity for this C -heterotrophic species listed saccharolytic, non-proteolytic clostridia. The oligopeptide hydrolysis activity is characterised by the detection of the released amino acids and the products of their bioconversion, the corresponding 2-hydroxyacids. This observation opens new aspects in the study of C. butyricum and raised questions about the peptide metabolism by this species. The enzymes implied in the oligopepitide hydrolysis can be regarded as potential virulence factors and being an element in the comprehension of certain pathologies.  相似文献

The ability of Clostridium butyricum cultures to hydrolyze three L-leucine-containing dipeptides (Leu-Leu, Leu-Gly and Gly-Leu) in a synthetic minimal medium is demonstrated by using gas chromatography coupled with mass spectrometry. The 13C nuclear magnetic resonance and a labeled dipeptide L-[1-13C]Leu-Gly were used to confirm this activity.  相似文献

Clostridium pasteurianum fermented glucose to acetate, butyrate, CO2 and H2. In batch cultures the fermentation pattern was only slightly affected by culture pH over the range 8·0 to 5·5. The acetate/butyrate ratio was always higher than or equal to one. Between 2·14 and 2·33 mol H2 was produced per mol glucose fermented. At unregulated pH, more butanol and less butyrate was formed. In a carbon-limited chemostat, the steady-state acetate/butyrate ratio was always lower than one. H2 production was approximately 1·70 mol per mol glucose consumed. Substantial amounts of extracellular protein were formed. With decreasing pH, acetate and formate production decreased, while H2 production was highest at pH 6.0. With increasing dilution rate ( D ), the product spectrum hardly changed, but more biomass was formed. Y glucosemax and Y ATPmax were 55·97 and 31·48 g dry weight per mol glucose or ATP respectively. With increasing glucose input the formation of fatty acids and H2 slightly decreased.
Continuous cultures fermented mannitol to acetate, butyrate, butanol, CO2 and H2. With acetate as co-substrate, butanol production and molar growth yields, Y mannitol and Y ATP, markedly decreased, while the butyrate and H2 production increased. The latter reached a value of 2·21 mol H2 per mol mannitol consumed.  相似文献

Starch fermentation by the porcine caecal anaerobe Clostridium butyricum was examined using gas and volatile fatty acid production as determinants of activity. Potato starch and amylopectin were studied in their native form as well as after retrogradation, which should render them resistant to pancreatic α-amylase digestion. Fermentation of both substrates was enhanced by pancreatin digestion of the native material, possibly due to the removal or disruption of part of the structure of the starch by the pancreatic enzymes. However, pancreatic digestion of retrograded potato starch apparently reduced the amount available for bacterial fermentation, whereas no significant effect was observed with amylopectin. The data suggests that starches which are high in amylopectin would be more likely to influence fermentation in the large intestine in monogastric animals, and that the presence of residual pancreatic enzymes in the lower gut could potentially enhance starch fermentation by this micro-organism.  相似文献

Phosphatidyltransferase from Clostridium butyricum, which catalyzes transfer of the phosphatidyl moiety of phosphatidylethanolamine (PE), phosphatidylglycerol (PG) or phosphatidylserine to primary alcohols such as glycerol, serine and ethanolamine, was tested for its ability to catalyze transfer of the plasmenyl moiety from plasmalogen analogs of PE or PG to glycerol or ethanolamine, respectively. The cell membrane of C. butyricum contains high proportions of these plasmalogens. When diacyl and plasmalogen forms of PE or PG were supplied as donors in equimolar amounts to membrane particles, the diacyl forms were the preferred donors by approx. 7 and 10 to 1, respectively. When the molar ratio of PE its plasmalogen was 1:3.3, the ratio of PG formed to its plasmalogen was 1:0.66. These results show that the enzyme(s) can catalyze transfer of both the diacyl and alkenyl acyl forms of glycerophospholipids, but the diacyl forms are used preferentially.  相似文献

目的 系统评价酪酸梭菌辅助根除幽门螺杆菌(Helicobacter pylori,H. pylori)感染的疗效和安全性。方法 计算机检索PubMed、CENTRAL(2016年7期)、CBM、CNKI、万方数据库、维普数据库,同时追溯纳入文献和相关综述的参考文献,收集关于酪酸梭菌辅助根除H. pylori感染的随机对照试验(RCT),检索时间均从建库至2016年7月30日。由两名评价员根据纳入排除标准筛选文献、提取资料、评价纳入研究的偏倚风险,采取RevMan 5.3进行Meta分析。结果 最终纳入13个RCT,共1 372例患者(其中试验组742例,对照组630例)。Meta分析结果显示:与标准根除方案相比,酪酸梭菌辅助治疗组有利于提高H. pylori的根除率(RR=1.26,95%CI:1.18~1.34,P<0.00001),并且降低总不良反应(RR=0.41,95%CI:0.32~0.52,P<0.00001)、腹泻(RR=0.31,95%CI:0.16~0.58,P=0.0003)、味觉紊乱的发生率(RR=0.35,95%CI:0.17~0.73,P=0.005);然而,在腹胀、恶心的发生率上,酪酸梭菌辅助治疗组虽低于标准方案组,但差异无统计学意义。结论 当前证据显示酪酸梭菌联合标准方案不仅可提高H. pylori的根除率,还能降低总不良反应、腹泻及味觉紊乱的发生。受纳入研究的质量和数量的限制,上述结论有待更多高质量的研究予以验证。  相似文献

Immobilized whole cells of Clostridium butyricum reduced both NAD(+) and NADP(+) in the presence of hydrogen at a pressure of 100 atm. The NAD(+) and NADP(+) reduction activities were 4.45 and 4.30 U/g dry cells, respectively [U = NAD(P)H regenerated, mu mol/min]. The amount of NADH regenerated by immobilized cells increased with increasing hydrogen pressure above 10 atm. Immobilized cells (6 mg dry cells) of Cl. butyricum completely converted NAD(+) (6.4 mumole) to NADH for 5 h, whereas only 60% of NAD(+) were reduced by free cells. Immobilized cells retained 89% activity after the 5-h reactions were repeated 4 times. L-Alanine was continuously produced at the rate of 12.8 mumol/min g dry cells from hydrogen, ammonium, and pyruvate with immobilized Cl. butyricum-alanine dehydrogenase.  相似文献

本研究采用丁酸梭芽孢杆菌(Clostridium butyricum)从甘油发酵制备1,3—丙二醇。该发酵需厌氧培养,发酵培养基为:甘油6%,葡萄糖1%,玉米浆2%,(NH4)2SO40.2%。发酵温度为34℃,pH为6.5~7。在最佳条件下,发酵50h可产1,3—丙二醇40.7g/L,甘油摩尔转化率达68%。  相似文献
