In mating tests of B6 males, and of congenic B6-T1aa males, with paired B6 and B6-T1aa females, there was a preponderance of matings with B6-Th1aa females in both cases. These results conform to a hypothesis that in theQa-T1a region there is anRi (recognition of identity) gene which is expressed in females and influences mating preference; and that anotherRi gene, expressed in males and also influencing mating preference, is situated elsewhere in the MHC region.Abbreviations according to Yamazakiet al. A series of congenic mice on the BALB/c genetic background have been employed to localize a teratocarcinoma transplantation rejection locus, Gt-1, to the K side of the H-2 locus on chromosome 17. Previous studies have placed the Gt-1 sv allele about 8 centimorgans away from the H-2 b or H-2 bv1 locus. Teratocarcinomas derived from 129/sv mice, Gt-1 sv (H-2K bv1/H-2D bv1), are rejected by BALB/c (H-2K d/H-2Dd) and BALB-G mice (H-2K d/H2D-b, but form tumors in BALB-B (H-2K b/H2D b) and BALB/5R5 mice (H-2K b/H2D d). In the reciprocal tumor-rejection test, a BALB/c teratocarcinoma was rejected by immunized BALB·B mice, but formed tumors in the immunized isogenic BALB/c mouse. These studies demonstrate the reciprocal expression of two Gt-1 alleles, one Gt-1 c, in BALB/c mice, and the other, Gt-1 sv, in the congenic BALB·B mice. Shedlovsky and co-workers have placed the Gt-1 locus in a similar location on the K side of the H-2 locus on chromosome 17.  相似文献

The antibody response to the hapten 2,4,6-trinitrophenyl (TNP) conjugated to autogenous mouse serum albumin (MSA) is regulated by anIr gene(s) located within the major histocompatibility complex (MHC). Both the qualitative and quantitative ability of congenic strains to produce TNP-specific antibodies are functions of theH-2 haplotype. Thus, mouse strains may be classified as high (H-2 d), intermediate (H-2 b,H-2 s), and low responders (H-2 a,H-2 k,H-2 n,H-2 p,H-2 q). Antibody responses, as measured by antigen-binding capacities in modified Farr assays, were compared among strains carrying recombinantH-2 haplotypes and their hybrid progenies. Distinct high- and low-responder phenotypes were evident throughout the time course of both primary and secondary antibody responses. The gene locus controlling specific responsiveness to TNP-MSA, now designatedIr-6, was mapped within theI-B subregion of theH-2 complex. Recessive inheritance of high responsiveness was confirmed in hybrid progenies of three different low × high-responder crosses.  相似文献

A.BY, B10.LPa, and B10.129(5M) mice were presensitized in vivo against B10.A(5R) cells and then restimulated in vitro by the same cells in the standard CML assay. The effector cells thus generated lysed not only B10.A(5R), but also C57BL/6 targets, indicating that, in addition to anti-H-2Dd response [measured on the B10.A(5R) targets], response to minor histocompatibility (H) antigens (measured on the C57BL/6 targets) also occurred. The latter response was directed against multiple minor H antigens in the case of the A.BY effectors, and against H-1 and H-3 antigens in the case of B10.129(5M) and B10.LPa effectors, respectively. The sensitization against minor H antigens occurred in the context of H-2Kb H-2Dd antigens, but by testing the response on C57BL/6 targets, only cells reacting with minor H antigens in the context of H-2Kb were assayed. The same effector cells were then tested against H-2b mutant strains, in which theH-2K b allele was replaced by a mutant one. All three effector types [A.BY, B10.LPa, and B10.129(5M)] behaved in a similar way: they all reacted with theH-2 bg1 mutant to the same degree as withH-2 b, they did not react at all or reacted only weakly with theH-2 bd andH-2 bh mutants, and they reacted moderately or strongly with theH-2 ba mutant. The degree of crossreactivity with the mutants reflects, with one exception, the degree of relatedness of these mutants toH-2 b, as established by other methods. The one exception is theH-2 ba mutant, which is the most unrelated toH-2 b, and yet it crossreacted strongly. Further testing, however, suggested that in this instance the crossreactivity was probably directed against H-2 antigens: the anti-H-2Dd effectors apparently crossreacted with the H-2Kba antigens. This finding is an example of cell-mediated crossreactivity between the products of two differentH-2 genes (H-2K andH-2D). It is also an example of anH-2 mutation generating an antigenic determinant known to be present in another strain.  相似文献

A monoclonal antibody (mAb) TP-3 has been established by immunizing rats with the BALB/c mouse thymic epithelial cell line TEL-2. The TP-3 antigen is expressed on stroma cells of thymus, spleen, and lymph node in syngeneic BALB/c mice (H-2 d ). This antigen is also expressed at a low level on the cell surface of immature thymocytes, and at a high level on mature T and B cells. In allogeneic mice such as C57BL/6 (H-2 b ) or C3H (H-2 k ), no cells expressed the TP-3 antigen. Using H-2 congenic mice, reactivity with mAb TP-3 was found to map to a region of H-2D d L d or between D d and Qa, suggesting that TP-3 is a major histocompatibility complex (MHC) class I antigen. However, immunoprecipitation analysis indicated that this antigen is not identical to the classical mouse class I molecules in terms of molecular size, antigenicity, and tissue distribution.  相似文献

Two congenic strains of mice were bred in Konstanz that bear theIg heavy chain allotype gene of the C57BL/6 (Ig-1 b ) in a BALB/c background genome. One line (CB-8K) underwent eight backcross generations to BALB/c before sister-brother mating was initiated. For the other line (CB-16KN) backcrossing was continued for eight further generations, then a homozygousIg-1 b /Ig-1 b strain was produced by sister-brother mating. Both lines were tested for four VH markers of the BALB/c and one of the C57BL parent. The CB-16KN strain expressed the C57BL marker VHNPb together with the C57BL allotype marker, and failed to express the three BALB/c markers, VHDEX, VHS117, VHphOx and VHNPa. It thus resembled the CB-20 strain.Strain CB-8K expressed the VHNPb marker of C57BL but also all the four BALB/c markers that were tested. The strain appeared more heterogeneous than the CB-16KN strain, and a subline was bred from two exceptional mice that did not express the VHNPb marker. This subline (CB-8KN) expressed the BALB/c marker VHNPa regularly, and was negative for the VHNPb marker. It thus resembles the BAB-14 line.The crossing over event thus must have happened in one of the two meioses, which led to the CB-8K line. As BAB-14 is derived in an analogous manner to a branch of the backcross of Potter and Lieberman, which ended up in CB-20, the unexpected finding is discussed that two independent crossing over events (in CB-8KN and BAB 14) within theIg heavy chain gene region have taken place at approximately the same stage of two breeding programs.Recipient of an EMBO fellowship during part of the study.  相似文献

Cytotoxic lymphocytes (CTL) were generated betweenIg-1-congenic strains BALB/c(H-2d,Ig-1a) andC.B-17(H-2d,Ig-1b) by cross-immunization in both directions and rechallenge in vitro. The effector cell populations specifically lysed target cells sharing both theH-2 haplotype and theIg-1 allele of the sensitizing strain. B- and T-cell blasts were equally good targets, suggesting thatH-2-restricted cytotoxic lymphocytes are not directed against serologically defined conventional allotypic determinants, but probably against minor histocompatibility antigens controlled by genes linked to theIg-1 complex. Competition experiments using cold target cells from a series ofIg-1b-congenic strains of the BALB/c background (BAB-14, C.B-17, C.B-26) revealed two not yet described minor histocompatibility loci linked to theIg-1 complex: We could demonstrate that BALB/c anti-C.B-17 effector cells recognize at least two distinct antigenic determinants on C.B-17 target cells, but only one on target cells from BAB-14, which carries a recombinantIg-1 complex. From these results we conclude that one of the minor histocompatibility antigens, designated as H(CH), is encoded by a gene linked to the heavy-chain constant-region (CH) genes, whereas the second minor histocompatibility antigen, designated as H(VH), is coded for by a gene linked to the heavy-chain variable-region (VH) genes. These two new genetic markers may be useful for further analysis of the mouseIg-1 complex because the analysis of the H(CH) and H(VH) genes may facilitate the search for recombinants in that chromosomal region.  相似文献

Influenza nucleoprotein (NP) is an important target antigen for influenza A virus cross-reactive cytotoxic T cells (Tc). Here we examine the NP epitope recognized by cloned and polyclonal BALB/c Tc and the genetics of this recognition pattern. We can define NP residues 147–161 as the epitope seen in conjunction with K d , the only H-2d class I responder allele for NP restriction. H-2 d /H-2 b F1 mice (C57BL × DBA/2) primed by influenza infection lyse only H-2d target cells treated with peptide 147–161 while H-2b targets are recognized only after treatment with NP residues 365–379 (previously found to be recognized by Db restricted Tc cells). Tc cell recognition of NP peptide 147–161 is entirely dictated by expression of K d and not by other B10 or OH background genes of congenic mice. Restriction of a unique NP sequence by each responder class I major histocompatibility complex (MHC) allele suggests that antigen and class I MHC interact for Tc recognition.  相似文献

Lysis of ectromelia- or LCM virus-infected macrophage target cells by virus-specific cytotoxic T cells from mice immunized with the homologous virus occurred only where donors of T cells and target cells shared eitherH-2K orH-2D genes. With both viruses, use of T cell or target cell donors bearing mutations (B6.C-H-2ba, B6-H-2bh, B6-H-2bg1, and B6-H-2bg2), all of which apparently occurred in the same single genetic element in theH-2Kb region, abolished (H-2ba) or impaired (H-2bh,H-2bg1 andH-2bg2) lysis in T cell-target cell combinations that shared (apart from the mutations) all other genes in theK, I-A, orI-B regions of theH-2 complex. The data suggest that virus-induced antigenic patterns on infected B6.C-H- 2ba (mutant) cells are more different antigenically from those on C57BL/6 (wild type) cells than are those on infected cells from the other mutants -B6-H-2bh, B6-H-2bg1, and B6-H-2bg2. (B6.C-H-2ba× B6 -H-2bh)F1 mice behaved like B6-H-2bh, indicating no complementation, and confirming that theH-2K gene(s) involved in recognition of virus-infected cells by virus-specific T cells behave as a single element. These findings are discussed in relation to the nature of virus-induced antigenic patterns that are recognized by virus-specific cytotoxic T cells.  相似文献

Immunogenicity of allogeneic immunoglobulins in mice were studied, measuring the allotype-specific antibody activity by agglutination of allogeneic antibody-coated red blood cells. It was found that the serum from C.B-20 mice (Igh b , BALB/c-congenic) was uniquely immunogenic in BALB/c mice for allotype antibody response. Whereas the C57BL/6 (Igh b ) serum was immunogenic only when heat aggregated and/or combined with adjuvant, the ultracentrifugation-deaggregated C.B-20 serum was definitely immunogenic when administered in a moderate dose (100 μl/mouse). Even more surprising was the fast that very low doses (0.01–0.1 μl) of soluble C.B-20 serum, but not C57BL/6 serum, down regulated the allotype-specific response effectively. Genetic analysis on congenic mice suggested that the immunogenicity is controlled by donorIgh orIgh-V(Id-C.B) inasmuch as the serum from BALB/c-congenic C.B-20 (Igh-V b C b ), but not BALB/c-congenic BAB/14 (Igh-V a C b ), mice was active in BALB/c mice in soluble form. Further studies showed that the Id-C.B was dominantly expressed on the immunoglobulins of (BALB/c×C.B-20)F1 and (C56BL/6×C.B-20)F1 strains, and was originally derived from the C57BL/Ka strain. The major determinant for the antibody production was encoded inIgh-C, but not inIgh-V. It is suggested thatId-C.B controls the allotype-specific antibody response in an unusual manner, possibly acting as a unique determinant activating helper T cells.  相似文献

This study describes the levels of interleukin-1 alpha (IL-1α), tumor necrosis factor alpha (TNFα) and interleukin-6 (IL-6) in the sera and parenchymal organs of various congenic mouse strains infected with Borrelia garinii. A significant elevation of inflammatory cytokine levels was found in the organs of C3H/HeN (H-2k) and B10.BR (H-2k) mice but not in those of BALB/c mice (H-2d). Focally produced cytokines can contribute to antimicrobial defense against these organisms. High levels of IL-1α were observed in the sera of C3H/HeN, B10.BR and B10 (H-2b) mice infected with B. garinii and they were associated with the presence of spirochetes in the skin. Thus, susceptible mice demonstrated a stronger cytokine response than resistant mice. This study presents in vivo evidence that B. garinii infection affects the immunopathogenesis of Lyme disease.  相似文献

TwoH-2 d mutants,H-2 dm2 (H-2L loss mutation) andH-2dm1 (gainplus-loss mutation involving bothH-2L andH-2D) were evaluated for any change in the immunogenicity of marrow stem cells. Grafts of 2 or 4 × 106 BALB/c(C) or BALB/c-H-2dm2 (C-H-2 dm 2) marrow cells were accepted by lethally irradiated B10.D2(H-2 d ) recipients and were rejected by irradiated B10(H-2 b ) recipients. Moreover, both (B6 × C)F1 and (B6 × C-H-2 dm 2)F1 mice, as irradiated recipients, resisted the growth of parental-strain B6(H-2 b ) marrow cells but accepted grafts from C or C-H-2 dm 2 parental-strain donors. Thus, theH-2 mutation involvingH-2L but notH-2D did not affect the expression ofH-2 d -associated Hemopoietic or Hybrid(Hh) antigens of marrow stem cells. Grafts of 2 to 8 × 106 B10.D2 or B10.D2-H-2 dm 1 marrow cells were rejected by B10.BR(H-2 k ) and B6 hosts and were accepted by B10.D2 hosts. However, B10.D2-H-2 dm 1 marrow cells grew to a much greater extent than B10.D2 cells in irradiated (B6 × B10.D2)F1 or (B6 × B10.D2-H-2 dm 1)F1 host mice. Therefore, theH-2 dm 1 mutation has altered the expression of Hh antigens at least quantitatively, resulting in a relative loss of hybrid resistance with the retention of Hh determinants recognized by allogeneic recipient mice which are notH-2 d . Since the Hh determinants of B10.D2 marrow cells have been mapped 16 cM to the right ofH-2, this mutation atH-2D/H-2L may have affected a regulatory gene.  相似文献

Early and late primary IgM antibody responses of mice to Thy-1.1 antigens showed different antigenic and cellular requirements. We studied genetic controls of the early primary responses, which could be induced by subcellular thymocyte antigens independently of host T-cell activity. All Thy-1.2 mouse strains of Igh a(BALB/c and BC8), Igh-V aCb(BAB14), Igh d(AKR/Cum), Igh j(CBA/J, C3H/HeN, C3H.SW, and C3H.JK), and Igh n(NZB) definitely responded early to Thy-1.1 antigens from AKR/J (Igh d), A.Thy-1.1 (Igh e), or B10.Thy-1.1 (Igh b) mice or SD rats, whereas all strains of Igh b(C57BL/6, C57BL/10, B10.D2, B10.BR, B10.A, CB20 and CWB), Igh c(DBA/2), Igh e(A/J), and Igh o(C.AL20) responded poorly to the same antigens. This contrasts with the observation that both strains of Igh j(C3H/HeN) and Igh b(B10.BR) responded well at later times. As was the case for late responses, the matching of H-2 between donor and recipient resulted in early responses of exceptional quality in high-responder strains. To determine the locus in theH-2 complex that affects susceptibility to the development of pulmonary adenomas in mice,H-2 congenic and recombinant strains of mice with A/Wy, BALB/c, C3H, and B10 backgrounds were subjected to treatment with urethane. The average number and the incidence of adenoma foci were recorded five months after the treatment. InH-2 congenic strains on the A/Wy background, the average number of adenoma foci per mouse was significantly higher in mice of the A/Wy, A/J, and A-Tla b (H-2 a ) strains than in A.BY (H-2 b ) mice. In BALB/c and C3H congenic strains, the strains carrying theH-2 k haplotype were more susceptible than those carrying theH-2 b haplotype. InH-2 congenic strains on the B 10 background, the average number and incidence of foci was also higher in haplotypesa, h2, k, andj than in haplotypesb, s, f, d, r, h4, i3, i5, and4. The average numbers of adenoma foci in (A/J × A.BY)F1 (H-2 a /H-2 b ) and (B10 × B10.A)F1 (H-2 b /H-2 a ) were intermediate between the numbers in the parental strains. In [B10.A (4R) × B10.A (3R)]F1 (H-2 h4 /H-2 i3 ) and [B10.A (4R) × B10.A (5R)]F1 (H-2 h4 /H-2 i5 ), the numbers of adenoma foci were higher than in resistant parental recombinants. These patterns of response to urethane matched the patterns of the immune response to lactate dehydrogenase-B (LDH-B) and immunoglobulin gamma 2a (IgG2a) proteins. These differences between mice in their susceptibility to the development of pulmonary adenomas is probably due to the polymorphism of the class II genes in theH-2 complex.  相似文献

A series ofH-2 and non-H-2 congenic resistant (CR) strains on a C57BL/10Sn background were infected with 107 amastigotes ofLeishmania donovani. Non-H-2 congenic strains B10.LP-H-3 b and B10.CE(30NX) and (B10.LP-H-3 b × B10)F1 hybrids showed a very rapid decrease in liver-parasite burdens beyond day 21. Parasite counts for these strains at day 35 were significantly lower than for all other strains tested. The rapid decrease in parasite numbers, massive lymphocellular infiltration into the liver and strong delayed hypersensitivity reactions to parasite antigens in strains congenic for a portion of chromosome 2 indicated that acquired immunity toL. donovani was controlled by a dominant gene at or near theIr-2 locus. In addition, B10.129(10M) mice, which differ from C57BL/10Sn at theH-11 locus, showed highly significant increases in parasite numbers at day 35. Other observations supporting the absence of acquired immunity in B10.129(10M) included negative delayed hypersensitivity tests to parasite antigens and the absence of lymphocellular infiltrate into the liver. Although the differences were not as pronounced,H-2 CR strains withH-2 b ,H-2 a , andH-2 k haplotypes also showed significantly greater decreases in parasite numbers by day 35 as compared to otherH-2 CR strains.  相似文献

Alkylation of 2-methylthiopyrimidin-4(1H)-one (1a) and its 5(6)-alkyl derivatives 1bd as well as theophylline (7) with 2,2-bis(bromomethyl)-1,3-diacetoxypropane (2) under microwave irradia-tion gave the corresponding acyclonucleosides 1-[(3-acetoxy-2-acetoxymethyl-2-bromomethyl)prop-1-yl]-2-methyl-thio pyrmidin-4(1H)-ones 3ad and 7-[(3-acetoxy-2-acetoxymethyl-2-bromomethyl)prop-1-yl]theophylline (8), which upon further irradiation gave the double-headed acyclonucleosides 1,1 ′-[(2,2-diacetoxymethyl)-1,3-propylidene]-bis[(2-(methylthio)-pyrimidin-4(1H)-ones] 4ac, and 7,7 ′-[(2,2-diacetoxymethyl)-1,3-propylidene]-bis(theophylline) (9). The deacetylated derivatives were obtained by the action of sodium methoxide. The activity of deacetylated nucleosides against Hepatitis B virus was evaluated. Compound 5b showed moderate inhibition activity against HBV with mild cytotoxicity.  相似文献

In capping experiments with peripheral T lymphocytes, two anti-H-2.28 sera (AKR anti-AKR.L, anti-Kb, and C3H anti-0H.B10, k anti-b) that do not contain any Qa-2-specific antibodies are able to redistribute not only the H-2.28-positive H-2 molecules, but also Qa-2 molecules. This is due to the capacity of these sera to react with Qa-2 molecules because on cells where all known molecules of the H-2 d haplotype were capped (K1d, K2d, Dd, Md, Ld, L2d), both antisera still reacted when the cells came from a Qa-2 positive Dd strain (B10.A) but not when the cells were of Qa-2 negative strain (BALB/cByA). The reaction with la and non-H-2 antigens was excluded in these experiments. These data show that Qa-2 and H-2 antigens share some specificities of the H-2.28 family. Other anti-private and anti-public anti-H-2 sera failed to react with the Qa-2 molecules.  相似文献

Minimal inhibitory concentrations for 10 new condensed isoindole derivatives were established by the agar diffusion method. Benzo[b]-6H-pyrimido[2,1-a]isoindole-4-one exhibited a broad antifungal effect; the observed MIC (mg/L) were 7 (K. lactis), 8 (C. pseudotropicalis), 17 (W. fluorescens), 22 (E. magnusii), 31 (P. membranaefaciens), 32 (S. alluvius) and 32 (S. cerevisiae). The compound also inhibited the growth ofB. subtilis (MIC 9 mg/L). No effect on the growth ofE. coli was found.  相似文献

Erythrocytes and thymocytes of theH-2 d genotype express between 1.6 and 2.6 times as much glyoxalase I activity per milligram of protein as those of theH-2 b genotype; the activity in theF 1 (H-2b/H-2d) cells is intermediate between these two values. Activity of glyoxalase II is not affected by the H-2 genotype. The H-2d:H-2b ratio of glyoxalase I activity is higher in the BALB background where it is 2.6:1 than in the B10 background where it is 1.6:1. Thermal denaturation kinetics of enzymes from the H-2b and H-2d genotype cells are the same.  相似文献
