Crude α-amylase preparations from seven Lepidoptera pests were susceptible to inhibition by salt-soluble proteins of bread wheat (Triticum aestivum L.) endosperm. Protein fractions that corresponded to tetrameric, dimeric, and monomeric wheat α-amylase inhibitors, were decreasingly effective against the insect α-amylase activity. To further confirm these results, purified inhibitors were tested against an α-amylase preparation fromEphestia kuehniella (Zeller). This preparation showed decreased activity when increasing amounts of an heterotetrameric inhibitor (reconstituted from its isolated subunits WTAI-CM2, -CM3 and -CM16) were assayed. Activity was only partially inhibited by homodimeric (WDAI-1, synonym 0.53; WDAI-2, synonym 0.19) and monomeric (WMAI-1, synonym 0.28) inhibitors.  相似文献

Insects feeding on stored grains cause considerable damage to harvested cereals and legumes every year. The use of α-amylase inhibitors to interfere with the pest’s digestion process has become an interesting alternative biocontrolling agent. In this study, we have studied the interactions of α-amylase inhibitors from Albizia lebbeck seeds with the amylases of coleopteran and lepidopteran insect pests. We isolated and purified the α-amylase inhibitor using acetone precipitation and gel filtration chromatography. Two prominent activity bands of α-amylase inhibitors were detected in electrophoretic analysis using 8% starch PAGE. We found that the α-amylase inhibitor, isolated as a monomer, had a molecular weight of 14.4 kDa. The α-amylase inhibitor was purified 36.15-fold with gel filtration chromatography. Its specific activity was determined at 14.4 U/mg/min. Feeding analysis of Tribolium confusum larvae on a diet containing purified α-amylase inhibitor from Albizia lebbeck revealed that survival of the larvae was severely affected, with the highest mortality rate occurring on the fifth day of feeding. We found that the isolated α-amylase inhibitor inhibits T. confusum and Helicoverpa armigera α-amylases in electrophoretic analysis as well as in solution assays. The isolated α-amylase inhibitor was found to be resistant to commercial protease as well as T. confusum and H. armigera digestive proteinases. The isolated α-amylase inhibitor was degraded by heating above 60°C. Our results suggest that A. lebbeck α-amylase inhibitor could be a useful future biocontrolling agent.  相似文献

Extracts ofAscophyllum nodosum, Fucus serratus, F. vesiculosus andPelvetia canaliculata contain inhibitors of α-amylase, lipase and trypsin. The inhibitors were isolated and identified by1H NMR spectroscopy as polyphenols which have apparent molecular weights in the range from 30 000 to 100 000 daltons, as determined by ultra-filtration with Amicon membranes. These polyphenols account for the whole of the inhibitory activity in crude algal extracts. The compounds inhibit α-amylase and trypsin in an apparently non-competitive manner, when preincubated with the enzymes, and the inhibition is directly proportional to the concentration of the inhibitor. Starch protects α-amylase when added to the enzyme together with the inhibitors. Under this condition the effectiveness of the inhibitors is reduced ten-fold.  相似文献

The effect of transnitrosation intermediate between S-nitroso-N-acetylcysteine (NACysNO) and cysteine on the growth of vgb-bearing Enterobacter aerogenes was investigated using three parameters: the ratio of the specific growth rates, the inhibition zone, and α-amylase synthesis for the culture exposed to stressors to that of the same stressor-free cultures. The effect of NACysNO/cysteine on the growth of Enterobacter strains was distinctive as compared with the CysNO, NACysNO, and their combination. At a higher concentration (2 mM), the extents of inhibition based on the μNACysNO/cysteineno stress ratio for these cultures were 57%, 62%, and 68% for VHb-expressing, parental, and pUC9-harboring cells, respectively. The inhibition caused by 2 mM NACysNO in the presence of 1 mM cysteine in all bacterial strains was almost twofold that achieved by NACysNO alone. Based on the diameter of the inhibition zone and α-amylase productivity, the four compounds (NACysNO/Cysteine, CysNO, NACysNO, and their combinations) affected the E. aerogenes strains in a concentration-dependent and negative manner. This negative effect was lower in vgb-bearing than vgb-lacking strains. Thus, sulfur-to-sulfur transnitrosation was an efficient NO release and significantly (P < 0.05) affects the growth of Enterobacter strains, to a lesser extent in vgb-bearing strains.  相似文献

Flow cytometric techniques were used to investigate cell size, protein content and cell cycle behavior of recombinantSaccharomyces cerevisiae strains producing human lysozyme (HLZ). Two different signal sequences, the native yeastMFα1 signal sequence and the rat α-amylase signal sequence, were used for secretion of HLZ. The strain containing the rat α-amylase signal sequence showed a higher level of internal lysozyme and lower specific growth rates. Flow cytometric analysis of the total protein content and cell size showed the strain harboring the native yeast signal sequence had a higher total protein content than the strain containing the rat α-amylase signal sequence. Cell cycle analysis indicated that the two lysozyme producing recombinant strains had an increased number of cells in the G2+M phase of the yeast cell cycle compared with the host strain SEY2102.  相似文献

Background  

Plants produce a wide range of proteinaceous inhibitors to protect themselves against hydrolytic enzymes. Recently a novel protein XAIP belonging to a new sub-family (GH18C) was reported to inhibit two structurally unrelated enzymes xylanase GH11 and α-amylase GH13. It was shown to inhibit xylanase GH11 with greater potency than that of α-amylase GH13. A new form of XAIP (XAIP-II) that inhibits α-amylase GH13 with a greater potency than that of XAIP and xylanase GH11 with a lower potency than that of XAIP, has been identified in the extracts of underground bulbs of Scadoxus multiflorus. This kind of occurrence of isoforms of inhibitor proteins is a rare observation and offers new opportunities for understanding the principles of protein engineering by nature.  相似文献

The effect of biomass concentration on the formation of Aspergillus oryzaeα-amylase during submerged cultivation with A. oryzae and recombinant A. nidulans strains has been investigated. It was found that the specific rate of α-amylase formation in chemostats decreased significantly with increasing biomass concentration in the range of approx. 2–12 g dry weight kg−1. When using a recombinant A. nidulans strain in which the gene responsible for carbon catabolite repression of the A. oryzaeα-amylase gene (creA) was deleted, no significant decrease in the specific rate of α-amylase formation was observed. On the basis of the experimental results, it is suggested that the low value of the specific α-amylase productivity observed at high biomass concentration is caused by slow mixing of the concentrated feed solution in the viscous fermentation medium. The 5′-untranslated leader sequence (UTLS) of the slpA gene from Lactobacillus acidophilus contributes to mRNA stabilization by producing a 5′ stem and loop structure, and a high-level expression system for the lactic acid bacteria was developed using the UTLS in this study. A plasmid, which expresses α-amylase under the control of the ldh promoter, was constructed by integrating the core promoter sequence with the UTLS. The role of the UTLS in increasing the copies of the α-amylase mRNA was proved by measuring α-amylase activity in the culture supernatant and the relative expression of α-amylase mRNA was determined by the quantitative real-time PCR analysis. Moreover, several expression systems were constructed by combining the core promoter sequence with the UTLS or with the partially deleted UTLS and the expression level was evaluated. The use of the UTLS led to the success in improving α-amylase expression in the two strains of Lactobacillus casei and Lactococcus lactis. The current study showed that the improvement in protein production using the UTLS could be applied to the expression system in the lactic acid bacteria.  相似文献

Thermostable alkaline α-amylase producing bacterium Bacillus cereus strain isolated from Cuddalore harbour waters grew maximally in both shake flask and fermentor, and produced α-amylase at 35°C, pH 7.5 and 1.0% of substrate concentrations. α-Amylase activity was maximum at 65°C, pH 8.0, 89% of its activity was sustained even at pH 11.0. Added with MnCl2, α-amylase activity showed 4% increase but it was inhibited by EDTA. The molecular weight of the purified α-amylase is 42 kDa.  相似文献

Cell respiratory activity of protoplasts obtained from the wild type of Neurospora crassa and photoreceptor complex WCC—white collar 1 (wc-1) and white collar 2 (wc-2)—mutants of Neurospora crassa strains was investigated. Respiration inhibition by KCN in the presence of 25 mM succinate was similar in all strains and did not exceed 83–85% against control. The significant induction of KCN-resistant respiratory pathway occurred under 1% glucose oxidation in wc-1 and wc-2 mutants if compared with the wild type strains. The inhibitors of the main (cytochrome) pathway of electron transfer in mitochondria—1 mM KCN and antimycin A (4 μg/ml)—blocked the respiration rate of the protoplasts from N. crassa wild type by 75%, while the cell respiration of wc-1 and wc-2 strains was suppressed by approximately 50%. The specific inhibitor of alternative oxidase—10 mM salicylhydroxamic acid (SHAM)—in combination with the blockers of mitochondrial electron transfer chain caused the total suppression of respiratory activity of protoplasts in all studied strains. It is supposed that an increase of KCN-resistance in WCC mutants under glucose oxidation is connected with alternative oxidase activation as the result of failure in reception and signal transduction of active oxygen species.  相似文献

We developed a novel strategy for constructing yeast to improve levels of amylase gene expression and the practical potential of yeast by combining δ-integration and polyploidization through cell fusion. Streptococcus bovis α-amylase and Rhizopus oryzae glucoamylase/α-agglutinin fusion protein genes were integrated into haploid yeast strains. Diploid strains were constructed from these haploid strains by mating, and then a tetraploid strain was constructed by cell fusion. The α-amylase and glucoamylase activities of the tetraploid strain were increased up to 1.5- and tenfold, respectively, compared with the parental strain. The diploid and tetraploid strains proliferated faster, yielded more cells, and fermented glucose more effectively than the haploid strain. Ethanol productivity from raw starch was improved with increased ploidy; the tetraploid strain consumed 150 g/l of raw starch and produced 70 g/l of ethanol after 72 h of fermentation. Our strategy for constructing yeasts resulted in the simultaneous overexpression of genes integrated into the genome and improvements in the practical potential of yeasts.  相似文献

The cloning of α-amylase gene ofS. occidentalis and the construction of starch digestible strain of yeast,S. cerevisiae AS. 2. 1364 with ethanol-tolerance and without auxotrophic markers used in fermentation industry were studied. The yeast/E.coli shuttle plasmid YCEp1 partial library ofS. occidentalis DNA was constructed and α-amylase gene was screened in S.cerevisiae by amylolytic activity. Several transformants with amylolysis were obtained and one of the fusion plasmids had an about 5.0 kb inserted DNA fragment, containing the upstream and downstream sequences of α-amylase gene fromS. occidentalis. It was further confirmed by PCR and sequence determination that this 5.0 kb DNA fragment contains the whole coding sequence of α-amylase. The amylolytic test showed that when this transformant was incubated on plate of YPDS medium containing 1 % glum and 1 % starch at 30°C for 48 h starch degradation zones could be visualized by staining with iodine vapour. α-amylase activity of the culture filtratate is 740–780 mU/mL and PAGE shows that the yeast harboring fusion plasmids efficiently secreted α-amylase into the medium, and the amount of the recombinant α-amylase is more than 12% of the total proteins in the culture filtrate. These results showed that α-amylase gene can be highly expressed and efficiently secreted inS. cerevisiae AS. 2.1364, and the promotor and the terminator of α-amylase gene fromS. occidentalis work well inS. cercvisiac AS. 2.1364.  相似文献

Cultivated varieties of the common bean (Phaseolus vulgaris L.) contain an α-amylase inhibitor (αAI-1) that inhibits porcine pancreatic α-amylase (PPA; EC 3.2.1.1) and the amylases of certain seed weevils, but not that of the Mexican bean weevil, Zabrotes subfasciatus. A variant of αAI-1, called αAI-2, is found in certain arcelin-containing wild accessions of the common bean. The variant αAI-2 inhibits Z. subfasciatus α-amylase (ZSA), but not PPA. We purified αAI-2 and studied its interaction with ZSA. The formation of the αAI-2-ZSA complex is time-dependent and occurs maximally at pH 5.0 or below. When a previously isolated cDNA assumed to encode αAI-2 was expressed in transgenic tobacco seeds, the seeds contained inhibitory activity toward ZSA but not toward PPA, confirming that the cDNA encodes αAI-2. The inhibitors αAI-1 and αAI-2 share 78% sequence identity at the amino acid level and they differ in an important region that is part of the site where the enzyme binds the inhibitor. The swap of a tripeptide in this region was not sufficient to change the specificity of the two inhibitors towards their respective enzymes. The three-dimensional structure of the αAI-1/PPA complex has just been solved and we recently obtained the derived amino acid sequence of ZSA. This additional information allows us to discuss the results described here in the framework of the amino acid residues of both proteins involved in the formation of the enzyme-inhibitor complex and to pinpoint the amino acids responsible for the specificity of the interaction. Seventy-three gene sequences encoding monomeric α-amylase inhibitors were characterized from cultivated wheat “Chinese Spring”, group 6 nullisomic-tetrasomic lines of “Chinese Spring” and diploid putative progenitors of common wheat. The monomeric α-amylase inhibitors from the different sources shared very high homology (99.54%). The different α-amylase inhibitors, which were determined by the 24 single nucleotide polymorphisms (SNPs) of their gene sequences, were investigated. A total of 15 haplotypes were defined by sequence alignment, among which 9 haplotypes were found with only one single sequence sample. Haplotype H02 was found to be the main haplotype occurring in 83 WMAI sequence samples, followed by haplotype H11. The median-joining network for the 15 haplotypes of monomeric α-amylase inhibitor gene sequences from hexaploid wheats was star like, and at least two subclusters emerged. Furthermore evidence of homologous recombination was found between the haplotypes. The relationship between nucleotide substitutions and the amino acid changes in WMAI of hexaploid wheats was summarized. It was clear that only five polymorphic sites in the nucleotide sequence of WMAI resulted in amino acid variations, and that should be the reason for different structure and function of inhibitors. However, little evidence could be found that there were WMAI genes in the A genome of hexaploid wheat, whereas it could conclude from our results that the A genome diploid wheat had WMAI genes. The overall information on the monomeric α-amylase inhibitors from wheat and Aegilops strongly support the view that these inhibitors have evolved from a common ancestral gene through duplication and mutation. Somatic embryo quality is an important factor decisive for the efficiency of somatic embryogenesis. Addition gibberellic acid (GA3) at a concentration of 1 μM to germination medium improved the regeneration of alfalfa somatic embryos. Inhibitory effect of ancymidol, an inhibitor of gibberellin biosynthesis, on germination and conversion may indicate that those processes require endogenous GAs. Since fluridone, an ABA biosynthetic inhibitor, at a concentration of 1 μM, had a slight stimulatory effect on germination of somatic embryos, it may be presumed that embryos contain a too high level of residual ABA after maturation phase (20 μM ABA is used at that phase). The observed improvement of regeneration of somatic embryos by GA3 was correlated with acceleration of starch hydrolysis through α-amylase activity enhancement by GA3. In contrast, the inhibitory effect of ABA on the above processes was probably related to inhibition of α-amylase activity and, in consequence, to delayed starch hydrolysis. It is suggested that α-amylase activity can be considered a good marker of the quality of Medicago sativa L. somatic embryos.  相似文献

Summary Theα-amylase gene ofBacillus amyloliquefaciens has previously been cloned into pUB110 to give the recombinant plasmid, pKTH10 (Palva 1982. Gene 19:81–87). Strains transformed by this plasmid are promising candidates for industrialα-amylase production. The stability of pKTH10 was determined in variousB. subtilis strains possessing specific alleles which affect the level ofα-amylase secretion.B. subtilis strains carrying pKTH10 were cultivated in starch-containing medium for up to 50 generations without antibiotic selection and then screened for the presence of pKTH10. The plasmid proved stable enough (< 1.0% cured after 50 generations) for industrial batchwise enzyme production in two strains, but in asacU9 strain (thesacU9 mutation increases concominantly the production ofα-amylase levansucrase and proteases) 99.9% of cells had lost pKTH10 after 50 generations, although the parental plasmid (pUB110) was stable in this strain (0.09% cured after 50 generations). The instability of pKTH10 in thesacU9 strain seems somehow to be related to high expression of the clonedα-amylase gene: when grown in a medium restrictingα-amylase production, only 0.53% ofsacU9 cells had lost pKTH10 after 50 generations.  相似文献

The expression of the mouse α-amylase gene in the methylotrophic yeast,P. pastoris was investigated. The mouse α-amylase gene was inserted into the multi-cloning site of a Pichia expression vector, pPIC9, yielding a new expression vector pME624. The plasmid pME624 was digested withSalI orBglII, and was introduced intoP. pastoris strain GS115 by the PEG1000 method. Fifty-three transformants were obtained by the transplacement of pME624 digested withSalI orBglII into theHIS 4 locus (38 of Mut+ clone) or into theAOX1 locus (45 of Muts clone). Southern blot was carried out in 11 transformants, which showed that the mouse α-amylase gene was integrated into thePichia chromosome. When the second screening was performed in shaker culture, transformant G2 showed the highest α-amylase activity, 290 units/ml after 3-day culture, among 53 transformants. When this expression level of the mouse α-amylase gene is compared with that in recombinantSaccharomyces cerevisiae harboring a plasmid encoding the same mouse α-amylase gene, the specific enzyme activity is eight fold higher than that of the recombinantS. cerevisiae.  相似文献

It is hypothesized that since protein α-amylase inhibitor (α-AI) and stimulator might be present together in red kidney bean (Phaseolus vulgaris L.) seeds, their in vitro interactions might influence their detection and quantification. Assay of α-AI using extracts from the embryonic axes revealed an unexpected finding in that the extracts stimulated rather than inhibited α-amylase activity. The cotyledon extracts exhibited inhibitory or enhancement effect on α-amylase activity depending on whether prior to the α-amylase assay they had been boiled for 10 min or not. Phytohemagglutinin (PHA-L in particular) is implicated in the present study as a stimulator of α-amylase activity co-extracted with α-AI from red kidney bean cotyledons. The importance of these findings is discussed in relation to the possible widespread occurrence of protein α-amylase stimulator in seeds and other plant parts.  相似文献

A variant of Saccharomyces cerevisiae pep4 strain 20B12, with improved oligotrophic proliferation, cell survival and secretion of heterologous mouse α-amylase, is described. Previously we reported a procedure to enrich NI transformants that are not inhibited by cytotoxic expression of hepatitis B virus surface antigen in the secretion pathway of the protease-A-deficient (pep4) strain. To use the NI cells as a host for heterologous expression, we tried to amend the introduced pYAS/12S vector and obtain a host strain, NI-C, with stable NI phenotype and trp1 marker restored. Southern analysis of genomic DNA of NI-C suggested that the original pYAS/12S was abnormally rearranged and not completely corrected. Further assay showed that the viability and mitotic ability of the NI-C strain were increased. While using the NI-C strain as host for plasmid transformation and heterologous expression of mouse α-amylase, we observed that transformed colonies grew more quickly and secreted more α-amylase than general yeast strains. A further test showed that the NI-C strain was able to use mouse α-amylase as a positive selection marker to form transformed colonies on nitrogen-starved plates that contain starch as the sole carbon source. The results imply that the NI-C variant is an improved pep4 strain that can be used for heterologous expression and for the development of new selective markers in the yeast transformation system.
