Nineteen filamentous fungi, isolated from estuarine sediments in Brazil, were screened for degradation of polycyclic aromatic hydrocarbons (PAH). The fungal isolates were incubated with pyrene. The cultures were extracted and metabolites in the extracts were detected by high performance liquid chromatography (HPLC) and u.v. spectral analyses. Six fungi were selected for further studies using [4,5,9,10-14C]pyrene. Cyclothyrium sp., Penicillium simplicissimum, Psilocybe sp., and a sterile mycelium demonstrated the ability to transform pyrene. Cyclothyrium sp. was the most efficient fungus, transforming 48% of pyrene to pyrene trans-4,5-dihydrodiol, pyrene-1,6-quinone, pyrene-1,8-quinone and 1-hydroxypyrene. This fungus was also evaluated with a synthetic mixture of PAH. After 192 h of incubation, Cyclothyrium sp. was able to degrade simultaneously 70, 74, 59 and 38% of phenanthrene, pyrene, anthracene and benzo[a]pyrene, respectively.  相似文献

通过一次航行对广东湛江湾19个站位的表层和底层海水进行采样。经过对样品的分离纯化,共获得253株丝状真菌菌株。通过测序获得了121个rDNA-ITS序列,并已提交给GenBank。经统计分析发现,湛江湾表层和底层海水真菌数量总体上表现为由湾内向湾口逐渐减少的水平分布格局;湛江湾表层丝状真菌的数量略多于底层,在海水表层和底层中真菌种类和数量的分布规律相似。根据形态特征及rDNA-ITS的Blast同源性分析,这些菌株分属于18个属和32个分类单元,其中包括7个海洋真菌新记录属。结果表明,湛江湾丝状真菌多样性Shannon指数达2.75,物种优势度变化范围为30.90%–0.02%。枝孢属Cladosporium的优势度最高,为湛江湾优势种群,其次是青霉属Penicillium、侧齿霉属Engyodontium、曲霉属Aspergillus、枝顶孢属Acremonium等。湛江湾表层和底层海水真菌群落的Jaccard相似系数为0.42。  相似文献

Eighty-seven soil samples were collected from various areas of Mumbai and its vicinity to determine the prevalence of keratinophilic fungi. From the 55 positive samples (63.21%), a total of four genera with nine species were isolated viz. Chrysosporium indicum (28.73%),C. lobatum (2.29 %), C. sp.I (1.14%), C. sp. II (1.14%),C. tropicum (1.14%),C. zonatum (2.29%),Ctenomyces serratus (2.29%) Malbranchea aurantiaca (2.29%) and Microsporum gypseum complex (21.83%). Cd-tolerant arbuscular mycorrhizal (AM) fungi from heavy-metal polluted soils   总被引:13,自引:1,他引:13  
Spores of arbuscular mycorrhizal (AM) fungi were isolated from two heavy-metal polluted soils in France via trap culture with leek (Allium porrum L.). Preliminary identification showed that the predominant spore type of both cultures (P2 and Cd40) belongs to the Glomus mosseae group. Their sensitivity to cadmium was compared to a laboratory reference strain (G. mosseae) by in vitro germination tests with cadmium nitrate solutions at a range of concentrations (0 to 100 mg L–1) as well as extracts from a metal-polluted and unpolluted soils. Both cultures of AM fungi from heavy-metal polluted soils were more tolerant to cadmium than the G. mosseae reference strain. The graphically estimated EC50 was 0.8 mg L–1 Cd (concentration added to the test device) for G. mosseae and 7 mg L–1 for P2 culture, corresponding to effective Cd concentrations of approximately 50–70 g L–1 and 200–500 g L–1, respectively. The extract of the metal-polluted soil P2 decreased germination of spores from the reference G. mosseae but not from P2 culture. However, the extracts of two unpolluted soils with different physico-chemical characteristics did not affect G. mosseae, whereas germination of P2 spores was markedly decreased in the presence of one of the extracts. These results indicate a potential adaptation of AM fungi to elevated metal concentrations in soil. The tested spores may be considered as metal-tolerant ecotypes. Spore germination results in presence of soil extracts show the difficulty of assessing the ecotoxic effect of metals on AM fungi without considering other soil factors that may interfere in spore germination and hyphal extension.  相似文献

Surfactant nanoemulsions are water in oil preparations that proved to have a broad spectrum biocidal activity against a variety of microorganisms including Gram-positive and Gram-negative bacteria, spores and enveloped viruses. These preparations are non-toxic to the skin, mucous membrane and gastrointestinal tissues at biocidal concentrations. In this study, 0.1% of the nanoemulsion designated X8W60PC has shown fungicidal activity against yeast including Candida albicans and C. tropicalis in 15 minutes. C. tropicalis was more sensitive than C. albicans, which required a longer time or a higher concentration of the nanoemulsion to achieve killing. Neutral to slightly alkaline pH was more effective in killing the yeast cells than acidic pH. Using the minimum inhibitory concentration assay, 0.08% of the nanoemulsion was inhibitory to C. albicans, and parapsilosis and filamentous fungi including Microsporum gypseum,Trichophyton mentagrophytes,Trichophyton rubrum,Aspergillus fumigatus andFusarium oxysporum.None of the individual ingredients was as effective a fungicidal as the nanoemulsion at equivalent concentration. This shows that the nanoemulsion structure is an important factor in the anti-fungal activity. The X8W60PC has great potential as a topical anti-fungal agent and further investigation into the mechanism of fungicidal action is warranted.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

The form and localisation of Cu accumulation in the extraradical mycelium (ERM) of three arbuscular mycorrhizal fungi (AMF), isolated from the same polluted soil contaminated with the Cu and Arsenate, was studied. There were differences in the capacity of the ERM of the three AMF to sorb and accumulate Cu. Glomus caledonium BEG133 had a significantly lower Cu-sorption capacity than Glomus mosseae BEG132 and Glomus claroideum BEG134 isolated from the polluted soil as well as an isolate of G. mosseae BEG25 from a non-polluted soil. This was directly related to the cation exchange capacity (CEC) of the ERM of these fungi. Transmission Electron Microscopy (TEM) and Scanning Electron Microscopy (SEM) linked to an energy dispersive X-ray spectrometer (EDAX) gave more detailed information, showing that the ERM of AMF from the polluted soil was able to accumulate Cu in the mucilaginous outer hyphal wall zone, cell wall and inside the hyphal cytoplasm. The EDAX spectra showed that the accumulated Cu was mainly associated with Fe in the mucilaginous outer hyphal wall zone and in the cell wall. Cu was associated with traces of arsenate inside the cytoplasm of the ERM of Glomus mosseae BEG132 but this was not visible inside the ERM of Glomus caledonium BEG133 or Glomus claroideum BEG134. This work suggests that the ERM of AMF is able to sorb and accumulate Cu, but different tolerance mechanisms exist between the three AMF isolated from the same polluted soil providing further evidence for functional diversity within populations of AMF in soils.  相似文献

The rhizospheric soil microfungi from a native forest (undisturbed and disturbed) were studied using soil dilution plate and soil washing methods. Fungi were isolated using slightly acid and alkaline culture media. 54 taxa were isolated: 49 from undisturbed forest soil and 37 from disturbed forest soil. Acremonium sp., Aspergillus ustus, Coemansia pectinata, Doratomyces stemonitis, Fusarium solani, F. oxysporum, Gliocladium roseum, Humicola fusco-atra, Mortierella sp., Penicillium lilacinum, Trichoderma harzianum, and T. koningii, showed the highest frequency, in both, undisturbed and disturbed forests. In undisturbed soil forest the biodiversity index was 3.97 whereas in disturbed ones was 3.89.  相似文献

Cadmium contamination of soil is a major concern in the biosphere. Beyond the suite of available physico-chemical treatment methods, green and more efficient technologies are desired to reduce cadmium and other heavy metal contaminants to acceptable levels. Elastin-like polypeptides (ELP) composed of a polyhistidine domain (ELPH12) can be used as an environmentally benign chelating agent for ex situ soil washing. However, ELPH12 is relatively non-selective. A biopolymer with metal-binding domains that have stronger affinity, capacity, and selectivity would have distinct advantages. The aim of this work is to investigate the use of a new generation of ELP biopolymer, ELPEC20, containing synthetic phytochelatin (EC) as the metal-binding domain for ex situ soil washing. ELPEC20 was shown to bind cadmium more effectively and selectively than ELPH12. The binding constant of ELPEC20 is an order of magnitude higher and the binding capacity is fivefold higher than ELPH12. In contrast to ELPH12, no decrease in cadmium binding was observed in the presence of other competing metal ions. The improved selectivity and binding capacity provided by ELPEC20 were directly reflected in the enhanced cadmium extraction efficiency from contaminated soil. In batch washing studies up to 62% of the bound cadmium was removed by ELPEC20 while less than 12% was removed by ELPH12. Cadmium was removed not only from the exchangeable fraction but also the oxidizable fraction. The high-affinity binding sites of ELPEC20 also results in very rapid extraction with complete removal achieved within 1 h, suggesting that ELPEC20 could be used as part of a rapid (short retention time) technology with minimum possibility for the biodegradation of biopolymers.  相似文献

Entomopathogenic Hypocreales were isolated from arid soils in Argentina using Tenebrio molitor as bait and tested for their biological performance at 30°C and 45–65% RH. Conidial germination was tested in three vegetable oils (sunflower, olive and maize) at two concentrations (1% and 10%) to evaluate their compatibility for further liquid formulations. According to radial growth and germination results, we selected four isolates to test their pathogenicity against second instar B. tabaci nymphs with the selected oil formulations at 30°C. CEP381 and CEP401 showed the highest radial growth. Isolates CEP381, CEP401, CEP413 and CEP409 (Metarhizium spp.) had similar germination percentages as compared with water control when germinated on either sunflower, olive or maize oils at 10% v/v. The highest mortality of B. tabaci was observed for the isolates CEP381 in sunflower oil and CEP401 in olive oil. Molecular identification of isolates was performed using ITS4–5 primers. All isolates belong to the Metarhizium core group. Tested isolates could grow and infect B. tabaci nymphs at 30°C in some of the vegetable oils as carriers, providing new possibilities for integrated pest management of Bemisia tabaci.  相似文献

Screening was done for the isolation of effective lignin degraders from the forest soil samples, by providing lignin as a carbon source through the enrichment method, which leads to the isolation of 8 effective fungal isolates among 14 isolates. Submerged fermentation was done for the production of ligninolytic enzymes with the effective microorganisms by providing Guiaicol as a carbon source. The assay of laccase, lignin peroxidise activity and specific activity was done after the incubation intervals of 2, 4, 6, 7, 8, 10 and 12 days at 27±2℃ under shake culture condition. Partially purified protein content was estimated by using Lowry's method. Pleurotus sp. and Phanerochaetae chrysosporium are more effective at the 2nd and 7th days of incubation for the production of laccase and lignin peroxidases among the effective isolates.  相似文献

We compared the antigenic characteristics of two thermo-dependent dimorphic fungi isolated from soil in Botucatu, an endemic area of paracoccidioidomycosis (PCM) and Paracoccidioides brasiliensis. The soil isolates grew as cerebriform colonies at 37 °C (yeast form) and as cottonous colonies at 25 °C (mycelial form). No pathogenicity for ddY mice or hamsters were observed. In immunodiffusion test, there were precipitation bands between the 2 soil isolates and pooled PCM patient sera. There were also common precipitation bands at 21, 50 and 58 kDa between the soil isolates antigens and PCM patient sera by Western-blotting, but no gp43 kDa band. No gene for gp 43 kDa protein was detected in the soil isolates by PCR. The fact that these isolates were obtained from an endemic area of PCM and there were some antigenic similarities between the soil isolates and P. brasiliensis in immunodiffusion test and Western-blotting may have some importance in epidemiological surveys done with paracoccidioidin as well interfering with the immune response of the exposed population. Two new species of coelomic filarioid belonging to Litomosoides are described from sigmodontine murids from the Rio de La Plata marshland, Argentina. Litomosoides bonaerensis n. sp., a parasite of Oligoryzomys delticola, belongs to the carinii group and is close to L. silvai, which differs by the head and tail papillae, buccal capsule and cavity, area rugosa, and morphology of the microfilaria. Litomosoides oxymycteri n. sp., from Oxymycterus rufus, belongs to the sigmodontis group. Differential diagnosis is based on the morphology of the buccal capsule, the head and tail papillae, and microfilaria. The ectoparasitic gamasid Ornithonyssus bacoti, in which several Litomosoides species develop, has been recovered from sigmodontines trapped in the study.  相似文献

Thirty-two soil samples were collected from six sites in the vicinity of Lonar lake, a meteorite crater and screened for presence of keratinophilic fungi using hair baiting techniques for isolation. Seventeen isolates were recovered and identified. The cultures were identified by recognition of their macro- and micromorphological features. Their identification was also confirmed by the BLAST search of sequences of ITS1-5.8S-ITS2 rDNA region against the NCBI/Gene bank data and compared with deposited sequences for identification purpose. Seven species of four genera were isolated viz. Aphanoascus durus (9.38%), Aphanoascus punsolae (3.125%), Auxarthron kuehnii (3.125%), Chrysosporium indicum (25.00%), Chrysosporium tropicum (3.125%), Chrysosporium sp. Sediments from eight river mouths along the Catalonian coast (Spain) were surveyed for keratinolytic fungi and related Onygenales. The actidione plating technique was employed. Of 532 actidione-treated sediment samples, 268 (50.3%) were positive for the fungi. Altogether, 384 fungal strains from 35 species were isolated from the samples. Narasimhella marginospora, Aphanoascus fulvescens, Neoarachnotheca keratinophila with its anamorph Myriodontium keratinophilum, Narasimhella hyalinospora, Beauveria alba, Sporothrix schenckii, Chrysosporium lobatum and Gymnoascus littoralis were the predominant species in sediments. Abundance of N. marginospora was clearly correlated with the degree of water pollution with sewage. Selected strains of three species of white rot fungi, Pleurotus ostreatus, Phanerochaete chrysosporium and Trametes versicolor, were grown in sterilized soil from straw inocula. The respective colonization rates and mycelium density values decreased in the above mentioned order. Three- and four-ringed PAHs at 50 ppm inhibited growth of fungi in soil to some extent. The activities of fungal MnP and laccase (units per g dry weight of straw or soil), extracted with 50 mM succinate-lactate buffer (pH 4.5), were 5 to 20-fold higher in straw compared to soil. The enzyme activities per g dry soil in P. ostreatus and T. versicolor were similar, in contrast to P. chrysosporium, where they were extremely low. Compared to the aerated controls, P. ostreatus strains reduced the levels of anthracene, pyrene and phenanthrene by 81–87%, 84–93% and 41–64% within 2 months, respectively. During degradation of anthracene, all P. ostreatus strains accumulated anthraquinone. PAH removal rates in P. chrysosporium and T. versicolor soil cultures were much lower.  相似文献

Soils of 10 poultry farms from Namakkal and 12 feather dumping sites from Chennai were studied for the presence of keratinophilic fungi. A total of 34 species belonging to 19 genera and one non-sporulating fungus were recovered. Sixteen species of fungi and one non-sporulating fungi were common to both sites, eight species were specific to Namakkal and nine species were specific to Chennai. Dermatophytes and closely related fungi were represented by six species belonging to five genera. Fungal species commonly found in the soil samples included Chrysosporium keratinophilum (73%), Trichophyton mentagrophytes (68.2%), Microsporum gypseum (64%), Myceliopthora vellerea (32%), Chrysosporium state of Arthroderma tuberculatum (27.3%) and Geomyces pannorum (23%). Non-dermatophyte fungi were represented by 28 species belonging to 14 genera and one non-sporulating fungus.  相似文献

The keratinophilic fungi of 29 sandpits from kindergarten schools and public parks in the city of Nablus was analysed to evaluate their role in the epidemiology of diseases caused by these fungi. Seventy two species were recovered 28 of which were common to both kindergartens and public parks sandpits. High percentage (57.4%) of fungal isolates found had been identified as the causes of various types of mycoses. Eight species of dermatophytes and closely related fungi were recovered, of which the followings were the most commonly found species in sandpits: Chrysosporium keratinophilum (20.7%), Microsporum gypseum (17.2%), Trichophyton mentagrophytes (6.9%), and C. evolceanui (6.9%).  相似文献

云南高黎贡山是中国生物多样性保护具有全球意义的关键区域,研究该地区土壤中降解纤维素真菌(cellulose-decomposing fungi,CDF)多样性及分布状况对筛选高效CDF和相关功能基因研究具有重要意义.利用羧甲基纤维素(CMC)刚果红平板和PDA平板,分离培养了高黎贡山海拔960~2900m之间0~20cm深度的土壤样品的好氧CDF,通过液体发酵鉴定其纤维素酶活力,并对其rDNA内转录间隔区(ITS)序列进行分析,用Neighbor-Joining法构建系统进化树.结果发现中低海拔地区土壤CDF数量高于高海拔地区,且数量与样地海拔、土壤含水量、温度、有机质和全氮含量呈显著相关(P<0.05);CDF在样地内多样性与土壤有机质、全氮、含水量显著相关(P<0.05);植被类型和人为干扰对CDF的数量和分布都有一定影响.在10个样地中共筛选到35株具有较高降解纤维素类物质能力的真菌,其96h 液体发酵测量CMC酶活力范围在79.6(菌株6)~228.9IU(菌株3).这些株菌分属15个属,其中曲霉属(Aspergillus)、青霉属(Penicillium)、葡萄状穗霉属(Stachybotrys)、毛壳霉属(Chaetomium)的真菌共计20株,占总分离菌株数的57.1%,是高黎贡山土壤中CDF的优势菌群.  相似文献
