During the course of our continuing search for biologically active compounds from Korean medicinal sources, we investigated the white flower of Impatiens balsamina. From the MeOH extract, two new phenolic compounds (12) containing a nitrile group and eleven known phenolic compounds (313) were isolated. The chemical structures of new compounds (12) were determined through NMR, HRMS, and CD data. We tested the isolated compounds (113) for their cytotoxic activities by determining their inhibitory effects on human tumor cell lines (A549, SK-OV-3, SK-MEL-2, and HCT15) in vitro using the sulforhodamine B (SRB) assay. We also investigated their neuroprotective activity by determining their effects on nerve growth factor (NGF) secretion in C6 cells, and anti-neuroinflammatory activity by measuring nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated BV-2 cells.  相似文献

Miliusa sinensis Finet and Gagnep. have recently been subjected to numerous scientific studies pursuing its structure and biological properties. The chromatographic separation of the components from the methanolic extract of M. sinensis leaves yielded three new bisstyryls, namely, sinenbisstyryls A–C (13). The structures of the new compounds were determined via interpretation of their spectroscopic data. Of the isolated compounds, sinenbisstyryl A (1) exhibited cytotoxic activity against the A549 human lung, HepG2 human hepatocyte carcinoma, MCF7 human breast, and DU145 human prostate cancer cell lines, with IC50 values of between 11.46 μM and 19.01 μM in four cases.  相似文献

Two new indole diketopiperazines, namely luteoalbusins A–B (12), along with eight known ones (310), were isolated from the fungus Acrostalagmus luteoalbus SCSIO F457 originated from deep-sea sediment. Their structures were determined by 1D/2D NMR, MS, and CD data analyses. Each of these compounds was evaluated for their cytotoxic activities against SF-268, MCF-7, NCI-H460, and HepG-2 cell lines, and compounds 15 showed significant cytotoxicties against all four cancer cell lines. Moreover, new compounds 1 and 2 had more potent cytotoxicity than the other ones and cisplatin.  相似文献

Nine new 8-O-4′ neolignans, named pinnatifidanin B I–IX (19), together with 9 known analogs (1018) were isolated from the seeds of Crataegus pinnatifida. The structures of 118 were determined by spectroscopic methods, including 1D, 2D NMR, CD and HRESIMS analysis. Compounds 811, 17 and 18 displayed potent cytotoxic activities against human cancer cell lines, and most interestingly, none of the 6 compounds displayed inhibitory activity against human lung cell line (Mrc5). The 6 cytotoxic compounds are considered to be potential as antitumor agents, which could significantly inhibit the cancer cell growth in a dose-dependent manner and are probably safer than positive control drug.  相似文献

A new series of novel Podophyllotoxin-like benzo[b]furo[3,4-e][1,4]diazepin-1-ones possessing structural elements of 4-aza-2,3-didehydropodophyllotoxins with central diazepine ring was designed and synthesized as anti-cancer agents. In initial assessment, the cytotoxic activity of the synthesized compounds was evaluated against three cancer cell lines including MCF-7, PC3 and B16-F10 employing the MTT assay. Some of compounds (12h, 13a, 13c and 14b) showed significant cytotoxic activity. So, we investigated the cytotoxicity of compounds 12h, 13a, 13c and 14b, along with podophyllotoxin as the reference drug in different cancer cell lines including A549, A2780, DU145, HeLa, and normal Huvec cell line. Among these four compounds, 13c showed promising antiproliferative activity against all cancer cells stronger than the other compounds and comparable to reference drug podophyllotoxin in some cancer cells. All these four compounds did not show significant cytotoxicity on normal Huvec cell line. The flow cytometry analysis of the MCF-7, PC3 and A2780 human cancer cell lines treated with 13c showed that 13c, induced apoptosis in the MCF-7, PC3 and A2780 human cancer cell lines, which is in good agreement to its cytotoxic activity as well. Compound 13c did not show significant influence on tubulin assembly and exert its cytotoxic effects via induction of apoptosis and has potent and selective cytotoxic effects in cancer cells.  相似文献

A phytochemical investigation to obtain bioactive substances as lead compounds or agents for cancer led to the obtainment of six new clerodane diterpenoids, designated as kurzipenes A–F (16), from the leaves of Casearia kurzii. Their structures were elucidated on the basis of NMR spectroscopic data analysis and the absolute configurations were confirmed by the time-dependent density functional theory (TDDFT) electronic circular dichroism (ECD) calculations. The cytotoxic activities of compounds 16 were evaluated against human lung cancer A549 cell line, human cervical cancer Hela cell line, and human hepatocellular carcinoma HepG2 cell line. Most diterpenoids showed potent cytotoxicities against the three selected cancer cell lines. The preliminary mechanism studies revealed that the most active compound 2, with an IC50 value of 5.3 μM against Hela cells, induced apoptosis and arrested the Hela cell cycle at the G0/G1 stage to exert cytotoxic effects.  相似文献

Phytochemical investigation of the aerial parts of Siegesbeckia pubescens afforded two new ent-kaurane diterpenoids (1-2), together with sixteen known ent-kaurane and ent-pimarane diterpenoids (3–18). Their structures were elucidated on the basis of extensive spectroscopic methods The absolute configurations of 1–2 and 12 were determined by single-crystal X-ray diffraction analyses. Three new abietane diterpenoids, fleuryinols A–C (13), together with fourteen known compounds, were isolated from the twigs and leaves of Podocarpus fleuryi. Their structures were established by spectroscopic analysis, including 1D- and 2D-NMR spectroscopic techniques. Compounds 18 were tested cytotoxic activity against five human cancer cell lines, HL-60, SMMC-772, A-549, MCF-7, and SW480, of which fleuryinol B (2) and 19-hydroxyferruginol (4) exhibited moderate cytotoxic activity against some cell lines.  相似文献

Two new quassinoids, altissinol A (1) and B (7), together with 12 known quassinoids, were isolated from the 95% ethanol extract of the barks of Ailanthus altissima. The structures of the new compounds (1 and 7) were determined on the basis of the spectroscopic methods including UV, IR, HR-ESI-MS, 1D and 2D NMR. The cytotoxic potential of all isolates were evaluated in vitro against three human hepatoma cell lines. Quassinoids 17 displayed potent cytotoxic activities against human hepatoma Hep3B and HepG2 cell lines. Interestingly, compounds 2, 3, and 5 exhibited cytotoxic activity against multidrug resistance HepG2/ADM cell line with IC50 value 4.3-fold more sensitive to Doxorubicin (DOX).  相似文献

Phytochemical investigation on Drypetes hainanensis has resulted in the isolation of three new phenylpropanoids, named drypetesins A–C (13). Their structures were elucidated by applying various spectroscopic techniques (NMR, UV, IR, MS, and CD). The antiproliferative activities of these compounds were evaluated in vitro against three cultured cancer cell lines. Those new isolates exhibited potent cytotoxic activities against human hepatoma (HepG2), human breast adenocarcinoma (MCF-7), and human lung carcinoma (A-549) cancer cell lines with IC50 value range 5.6–14.8 μM.  相似文献

Five new sesquiterpene lactones (15) were isolated from Inula britannica collected in the wild from Serbia along with five known compounds (610). Sesquiterpene lactones were isolated using centrifugal partition chromatography followed by combination of flash chromatography and semi-preparative HPLC. Isolated compounds were screened for cytotoxic activity on four different human cancer cell lines and their multi-drug resistant counterparts, as well as on normal human keratinocytes. Sesquiterpene lactones showed similar cytotoxic activity toward drug sensitive and drug resistant cancer cell lines.  相似文献

Eight new cardiac glycosides/aglycones (antiaritoxiosides A–G, 17, and antiarotoxinin B, 8), two new coumarins (anticarins A–B, 4142), and two new flavanones (antiarones L–K, 4344) were isolated from trunk bark of Antiaris toxicaria together with 53 known compounds. The new structures were established by extensive analysis of spectroscopic data. Compound 1 (10-carboxy and 3α-hydroxy) and compounds 36 (10-hydroxy) contain unique substituents that are rarely found in cardiac glycosides. The cytotoxic effects of isolated compounds against ten human cancer cell lines, KB, KB-VIN, A549, MCF-7, U-87-MG, PC-3, 1A9, CAKI-1, HCT-9 and S-KMEL-2, were tested using the sulforhodamine B assay. Five compounds (12, 16, 20, 22, and 31) showed significant cytotoxicity against all ten cancer cell lines, with notable potency at the ng/mL level against some cell lines, which merits further development as clinical trial candidates.  相似文献

Two new naphthoquinones, goniothalaminone A (1) and B (2), and a new styryllactone, (?)-8-epi-9-deoxygoniopypyrone acetate (12) together with one known naphthoquinone (3), one known indolequinone (4), one known 1-azaanthraquinone (5), six known styryllactones (611) and one known sesquiterpene (13) were isolated from the roots and leaves of Goniothalamus scortechinii. The structures of the new compounds were elucidated by spectroscopic analysis and of the known compounds by comparison of their physical, UV, IR, 1H and 13C NMR data with those of published compounds. Antiplasmodial, antimycobacterial and cytotoxic activities of the styryllactones were evaluated. Phytochemical investigation of the 70% aqueous EtOH extract of Lycopodium complanatum led to six new polyhydroxy serratene triterpenoids (serrat A-F, 16), along with a known analogue (7). Their structures and configurations were elucidated by data analysis of HRESIMS, 1D and 2D NMR, in combination with comparisons of reported experimental spectroscopic data. All the isolates were evaluated cytotoxic activities against HepG2 cells, MCF-7 cells and series human lung cancer cell lines A549, Calu-6, NCI-H441, NCI-H226 and NCI-H1975. The results indicated that certain compounds inhibited proliferation of human cancer cells. Moreover, all compounds possessed selective cytotoxic activities on MCF-7 cells. Further, possible biosynthesis pathways of these compounds were proposed.  相似文献

Eurycoma longifolia has been widely used for various traditional medicinal purposes in South-East Asia. In this study, five new quassinoids, eurylactone E (1), eurylactone F (2), eurylactone G (3), eurycomalide D (4), and eurycomalide E (5), along with ten known quassinoids (615) were isolated from the roots of E. longifolia. Their structures were determined by extensive spectroscopic methods, including 1D and 2D NMR, and MS spectra data. Among the isolated compounds, 13β-methyl,21-dihydroeurycomanone (6) has been reported as a synthetic derivative. However, it was isolated from the natural product for the first time in this study. The cytotoxic activities of fifteen compounds were evaluated against human lung cancer cell line, A549 and human cervical cancer cell line, HeLa.  相似文献

Two new glycoalkaloids, erianosides A (1) and B (2) along with five known compounds (37) were isolated from the leaves of Solanum erianthum. Their structures were elucidated from analyses of spectroscopic data and all isolates were tested for in vitro cytotoxic activity against human breast cancer cell lines (BT-549, MDA-MB-231, T74D, and MCF-7). Solasonine (5) and solamargine (6) were active against the aforementioned four cancer cell lines with IC50 values of 27.26–35.89 and 5.84–10.13 μM, respectively. Erianoside A (1) (T74D: IC50, 56.39 µM) and solasodine (3) (BT-549 and MDA-MB-231: IC50, 59.15 and 75.63 µM, respectively) had moderate cytotoxic effects towards some cell lines in the panel.  相似文献

Five new triterpenoid saponins, oleiferosides P–T (1–5) were isolated from the EtOH extract of the roots of Camellia oleifera C. Abel. The structures of saponins 1–5 were elucidated on the basis of integrated spectroscopic techniques. All the compounds were characterized to be oleanane-type saponins with sugar moieties linked to the C-3 of the aglycone. By using the MTT assay, an in vitro analysis of the cytotoxic activities of these saponins on the human tumor cell lines (lung adenocarcinoma A549 cells, hepatic carcinoma SMMC-7721 cells and breast cancer MCF-7 cells). Among them, compound 4 showed a certain cytotoxic activity against all the tested cell lines.  相似文献

Three new phenylpropanoids (13) together with six known congeners (49) were isolated from the bark of Juglans mandshurica Maxim using anti-hepatoma activity as a guide. Their structures were determined by comprehensive NMR and HRESIMS spectroscopic data analyses. All the isolated compounds were evaluated for their growth inhibitory activities against two kinds of liver cancer cell lines (HepG2 and Hep3B). Among them, compound 4 showed moderate cytotoxic activities against HepG2 and Hep3B cell lines with IC50 values of 58.58 and 69.87 μM. Compound 5 exhibited 50% cell death rate in HepG2 and Hep3B cell lines at 63.70 and 46.45 μM, respectively. Further observation of morphological changes and Western blot demonstrated that compounds 4 and 5 exhibited their cytotoxic activities through the induction of apoptosis. A structure-activity relationship study suggested that an α, β-unsaturated aldehyde might be the most important functional group.  相似文献
