The life stages ofEntomophaga grylli (Fresenius) Batko Pathotype 2 were purified and separated by centrifugation in PercollR density-gradient medium. The ranges of buoyant densities for germinated resting spores, germ conidia, and resting spores respectively were: 1.040–1.050, 1.055–1.085, and 1.080–1.120 g/ml. Cuticular invasion by germinated germ conidia was the means by whichMelanoplus grasshoppers became infected. Scanning electron micrographs revealed germination of germ conidia on the visible host integument at 100% RH, but not at 90% RH. Significantly higher mortality (P<0.05) was obtained after 3 weeks with grasshoppers incubated in constant light than in constant dark for 24 h following treatment. Five strains ofMetarhizium anisopliae (Metsch.) Sorokin and one strain ofMetarhizium flavoviride Gams &; Rozsypal originally isolated in Madagascar were studied. Measurements of conidia and, for the first time, also of blastospores produced in a liquid medium were used for species and variety determination. Blastospores ofM. flavoviride were more homogenous in their size than those ofM. anisopliae. Growth at high temperatures between 25° and 40°C showed that 4 isolates ofM. anisopliae grew at 36°C andM. flavoviride grew at 38°C. Using alternating day/night temperatures (8/16 h) the three strains tested could also tolerate 40°/25°C. In bioassays, fiveMetarhizium spp. isolates were tested against third and fourth instar larvae ofLocusta migratoria (L.) at two alternating day/night temperatures of 30°/25°C and 36°/25°C. In the cooler regime, all strains caused a mortality of 50% within 5.9 to 8.5 days (median lethal time), while in the 36°/25°C treatment only the thermophilicM. flavoviride and oneM. anisopliae strain isolated from a soil sample gave comparable results with median lethal times of 6.8 and 7.3 days, respectively.  相似文献

Blastospores of three strains of Metarhizium anisopliae were stored in 18 liquids at 4°C, 20°C and 35°C for 18 weeks, 12 weeks or 9 days respectively. Viability was quantified by determination of their germination. In bioassays the virulence of stored blastospores was studied using adults and third instars of Locusta migratoria migratorioides (R. & F.) and compared to those of freshly produced blastospores and conidia. Generally, there was great variability in the viability of blastospores, depending on the fungal strain and the liquids used. Blastospores survived best at 4°C in 10% hydroxyethyl starch; for example, germination of M. anisopliae strain 97 still amounted to more than 80% after storage for 18 weeks. Other suitable liquids were deionized water, 25% Ringer's solution and 1% sodium alginate. The viability of blastospores stored at 20°C was considerably shorter than at 4°C. During storage for 12 weeks at 20°C the best protective liquids for M. anisopliae strain 97 were 25% Ringer's solution (43% germination), deionized water (23%) and 10% hydroxyethyl starch (23%). At 35°C, 45% of M. anisopliae strain 97 blastospores still germinated after storage for 7 days in 25% glycerol. The bioassays revealed that the virulence of blastospores after storage was comparable to that of fresh ones and even better than that of fresh conidia. In general, the LT50 was about 4–6 days at an alternating day/night temperature of 28/20°C.  相似文献

Athrycia cinerea Coq. is a univoltine parasitoid of the bertha armyworm,Mamestra configurata Walker (Lepidoptera: Noctuidae), in western Canada. This parasitoid overwinters as a pupa in the soil. These pupae are less sensitive than their host to increased cold stress caused by lower temperatures or danger durations of exposure. Exposure to −7.5°C for 140 days did not significantly reduce survival. Survival decreased with exposure to temperatures from −10 to −20°C, but survival was 48% even after 40 days exposure to −20°C. Exploratory activities were done in Syria, Turkey, Iran, Uzbekistan, Kazakhstan, The Kyrghyz Republic, and Russia to locate entomopathogenic fungi of Eurygaster integriceps. Isolates from the entomopathogenic genera Beauveria, Paecilomyces, and Verticillium were collected. Beauveria bassiana was the most commonly recovered species. Thirty-one isolates of the 221 recovered were examined at 20, 25, 30, and 35 °C for 20 days for growth and sporulation. Growth and sporulation were generally highest at 25 °C. None of the isolates grew at 35 °C, and at 30 °C growth was retarded with no conidia being produced. Single- and multiple-concentration bioassays were conducted on greenhouse-grown wheat plants and in pine litter to evaluate virulence of fungi from several sources to E. integriceps. When tested at a single concentration, mortality after 15 days ranged from 66 to >95% in the litter assays and 50 to 91% in the plant assays. There was a distinct concentration response for most of the isolates tested in the multiple-concentration assay, particularly in the in-litter environment. In litter, mortality tended to develop earlier than in on-plant assays. Several isolates of B. bassiana and one Metarhizium anisopliae displayed consistently high virulence against E. integriceps and were more virulent than two commercial strains. Our results demonstrate the potential of entomopathogenic fungi for management of E. integriceps in overwintering sites and in wheat fields.  相似文献

The upper survival temperature for most isolates ofChondrus crispus from localities ranging from northern Norway and Iceland to Spain, and for an isolate from Nova Scotia, was 28 °C after 2 weeks of exposure to temperatures of 20–31 °C at intervals of 1 °C. An upper survival limit of 29 °C was exhibited by a few European isolates from the English Channel, the North Sea, and one Irish isolate from the upper intertidal. The warm-temperate Japanese speciesC. nipponicus andC. giganteus formaflabellatus survived 30 °C, whereas 29 °C was the upper survival limit for the coldtemperatureC. pinnulatus formapinnulatus from northern Japan. A possible origin ofC. crispus in the north Pacific is discussed.  相似文献

The effect of four different temperatures (15, 20, 25 and 30°C) on the in vitro growth of 19 isolates of Pandora blunckii and 14 isolates of Zoophthora radicans from Plutella xylostella larvae was investigated. Both species grew more at 20 and 25°C than the other two temperatures. However, Z. radicans grew more than P. blunckii at 20 and 25°C. Within each species there were differences amongst: all isolates regardless of geographical origin, isolates from different countries and isolates from Mexico. No relationship was found between optimal growth temperature and geographical origin. This represents the first report of the relationship between temperature and the in vitro growth of P. blunckii. The ecological role of this large variability amongst isolates within each species is discussed.  相似文献

This study examined the role of generalist predators in producing higher mortality ofPlutella xylostella L. (Plutellidae) larvae on glossy vs. normal-wax cabbage,Brassica oleracea var.capitata L. To test this, survival and feeding ofP. xylostella were measured on individually caged glossy and normal-wax plants with and without each of three generalist predators,Chrysoperla carnea (Stephens) (Chrysopidae),Orius insidiosus (Say) (Anthocoridae), andHippodamia convergens Guerin-Meneville (Coccinellidae). In the greenhouse, predators always significantly reduced survival ofP. xylostella larvae on glossy plants, but never on normal-wax plants. In the field, predators significantly reducedP. xylostella survival on glossy plants, but onlyC. carnea was effective on normal-wax plants. In similar experiments with excised leaves,O. insidiosus andC. carnea were more effective predators on the glossy leaves, whileH. convergens was equally effective on both kinds of leaves. Patterns for feeding were similar, but significance levels differed from those forP. xylostella survival. The greater effectiveness of predators on glossy plants is apparently due to the reported improved mobility of these animals on glossy leaf surfaces. The data also suggest that reduced mining byP. xylostella exposes the larvae to more predation on glossy plants and contributes some to the resistance. Regardless of the mechanism, resistance toP. xylostella on glossyB. oleracea appears to depend on the action of generalist predators for its full expression. This dependence on predation must be considered in the development and deployment of glossy insect-resistantB. oleracea.  相似文献

Summary In vitro growth rates of transformed (crown gall) and nontransformed cultures ofVinca rosea L. were greater at 32°C than at 25°C. The growth of transformed cells was significantly inhibited by kanamycin, neomycin, and chloramphenicol but not by cycloheximide. Nontransformed cells were inhibited by all four antibiotics., The relative growth rates of transformed cultures induced by four different strains, ofAgrobacterium tumefaciens did not correspond to the relative rates of tumor weight increase observed in vivo nor with the relative weights of tumor tissue in, plants 8 weeks after inoculation with the corresponding bacterial strains.  相似文献

Differential response was noticed when 4 species ofTrichogramma egg parasitoids were stored at 2°, 5° and 10°C for 7 to 49 days. Pupal stage was found to be most appropriate for storage. Emergence ofTrichogramma achaeae Nagaraja and Nagarkatti andTrichogrammatoidea eldanae Viggiani was greater in comparison toTrichogramma chilonis Ishii andTrichogramma japonicum Ashmead at all 3 temperature regimes. Fecundity and longevity declined drastically after storage of 14 days at 2° and 5°C and 21 days at 10°C, when removed to room temperature. 10°C was found to be the best temperature for storage up to 49 days. The influence of simulated sunlight on survival of conidia of 4 species of entomopathogenic Hyphomycetes was investigated. Conidia from 65 isolates ofBeauveria bassiana, 23 ofMetarhizium anisopliae, 14 ofMetarhizium flavoviride and 33 isolates ofPaecilomyces fumosoroseus were irradiated by artificial sunlight (295 to 1,100 nm at an ultraviolet-B irradiance of 0.3 W m–2) for 0, 1, 2, 4 and 8 h. Survival was estimated by comparing the number of colony forming units (CFU) produced by conidia exposed to irradiation to the number of CFUs produced by an unexposed control. Survival decreased with increased exposure to simulated sunlight; exposure for 2 h or more was detrimental to all isolates tested. Overall, isolates ofM. flavoviride were the most resistant to irradiation followed byB. bassiana andM. anisopliae. Conidia ofP. fumosoroseus were most susceptible. In addition to the large interspecies differences in susceptibility to irradiation, there was also an intraspecies variation indicating that strain selection to irradiation tolerance may be important in the development of microbial control agents where increased persistence in an insolated environment is desirable.Abbreviations CFU Colony forming units - UV-B ultraviolet radiation-B  相似文献

Field experiments were conducted to study the influence of cabbage monoculture and mixed cropping on the parasitism of diamondback moth,Plutella xylostella (L.), a destructive pest of all crucifers, by 2 larval parasites,Diadegma semiclausum Hellén andCotesia plutellae Kurdjumov. There was no significant difference in parasitism by either species whether cabbage was planted in insecticide-free monoculture or in mixed cropping with 8 noncrucifers which were sprayed twice a week with chemical insecticides mevinphos, methamidophos and permethrin. Population ofP. xylostella increased as the cabbage plants grew older. Parasitism byC. plutellae was higher soon after cabbage transplanting but decreased as the plants grew older. Parasitism byD. semiclausum was very low soon after cabbage planting but increased as the plants grew older. A significant negative correlation was found betwen parasitism byC. plutellae andD. semiclausum. In a caged field study where only one parasite species was used in an individual cage, parasitism ofP. xylostella by both species decreased as theP. xylostella population increased. This is believed to be due to the absence of competition between the two parasites inside the cage. There was no relationship between host-plant age and parasitism ofP. xylostella larvae by either parasite species.  相似文献

We investigated phagocytosis and intracellular killing of clinical and environmental isolates of Aspergillus spp. by human monocyte-derived macrophages (MDMs). Serial pathogens such as Aspergillus fumigatus, Aspergillus flavus and Aspergillus terreus were examined with a microbiological assay. Phagocytosis for resting conidia of Aspergillus spp. was similar for all isolates tested. During 30 min of incubation phagocytosis ranged from 49.9% to 85.5% for clinical isolates and from 40.3% to 87.1% for environmental isolates. MDMs killed A. fumigatus, A. flavus and A. terreus conidia after ingestion for 120 min, as shown by a decrease in colony forming units (cfu) count of intracellular fungi. The killing index for all isolates of Aspergillus spp., ranged from 12.1 ± 1.1% to 90.3 ± 10.4%; isolate-dependent (P < 0.01) differences against the fungicidal action of MDMs were observed. In conclusion, significant differences were noted for killing indices between several strains of Aspergillus spp. whereas phagocytosis was similar for all isolates tested in vitro. No differences were observed within environmental and clinical isolates.  相似文献

A new biocontrol formulation system was devised that does not require sterile conditions during preparation. It involves mixing vermiculite and powdered wheat bran with wet or dry fermentor biomass of Trichoderma spp. or Gliocladium virens, moistening with 0.05 N HCl, and drying the mixture. Before application to soil, the preparation (VBA‐FB) is activated by re‐moistening with 0.05 N HCl and incubated at room temperature for 2–3 days to stimulate development of young hyphae of the biocontrol fungus. Populations of biocontrol fungi proliferated to greater than 107 colony‐forming units (cfu) per g of soil when activated VBA‐FB was added to soil. In soil artificially infested with Rhizoctonia solani, seven isolates of the 14 studied added as VBA‐FB reduced survival and 12 reduced saprophytic growth of the pathogen. Of these, two isolates of T. hamatum (TRI‐4, Tm‐23) and one of T. harzianum (Th‐87) were the most effective. Preparations formulated with either wet or dry biomass effectively reduced pathogen survival, but activated VBA‐FB was more effective than non‐activated VBA‐FB. Storage of VBA‐FB at 25°C for 24 weeks before activation reduced viability of isolates considerably more than storage at 5°C for 24 weeks. In addition, VBA‐FB stored at 5°C before activation more effectively reduced survival of R. solani than VBA‐FB stored at 25° C. Survival of R. solani was reduced by activated VBA‐FB applied to several soil types (sandy loam, sandy clay loam, clay). Some nitrogen fertilizers increased the efficacy of VBA‐FB preparations of several isolates.  相似文献

The survival of Neozygites cf. floridana (Weiser and Muma) as dry hyphal bodies in mummified cassava green mites, Mononychellus tanajoa (Bondar), at 5.0% RH in the dark was affected by storage temperature. Survival of the fungus in mummies kept at 24±1.0°C could be demonstrated for 6–7 months. When stored at 4°C, the fungus sporulated from 90% of the mummies liberating an average of 186.9 primary conidia per mummy even after a storage period of 16 months, when the experiment was terminated. The temperature, humidity and light condition significantly affected the viability of primary conidia. The percent viability across all factors dropped from 98.4% after 0 h (beginning of the experiment) to 23.4% after a 1 h exposure to the conditions tested. Lower temperatures maintained higher viabilities with 86.3% of the conidia surviving after 18 h at 18°C, whereas almost all conidia died after 12 h at 33°C. Conidia survived less than 1 h when exposed to SDs (saturation deficit) of 2.0 mm Hg or higher at any tested temperature.  相似文献

Laboratory and greenhouse studies were conducted to investigate the suitability of 2 hymenopterous parasites,Diadegma eucerophaga Horstmann andApanteles plutellae Kurdjumov for introduction to control diamondback moth (DBM),Plutella xylostella (L.), a destructive pest of crucifers in tropical to subtropical Southeast Asia. Parasitism byD. eucerophaga was high at temperature range of 15°C to 25°C and that ofA. plutellae, at 20°C to 35°C. Both parasites were active in searching for host and oviposited only during photophase. No parasitism was observed during darkness. WhereasA. plutellae could parasitize all instars of DBM larvae,D. eucerophaga parasitized only the first 3 instars and failed to parasitize the 4th. Parasitism byD. eucerophaga was greater when DBM larvae were feeding on common cabbage (Brassica oleracea var.capitata L.), than on cauliflower (B. oleracea var.italica L.), broccoli (B. oleracea var.botrytis L.) or Chinese cabbage [Brassica campestris L. ssp.pekinensis (Lour) Olsson].A. plutellae parasitism was greater when DBM larvac were feeding on Chinese cabbage than on common cabbage, cauliflower or broccoli. Storage of pupae at 0°C and 4°C to 6°C for up to 2 weeks reduced emergence ofD. eucerophaga adults more than that ofA. plutellae. A non-selective insecticide, deltamethrin, was toxic to adults of both parasites but selective ones such asBacillus thuringiensis, teflubenzuron, and pirimicarb were not. Pupae were more tolerant than adults to insecticides. The insecticide-resistant Luchu strain and susceptible laboratory strain of DBM suffered an equal level of parasitism by both parasites.   相似文献

Laboratory studies were conducted on certain aspects of biology ofDiadegma semiclausum Hellén, a larval parasite of a crucifer pest,Plutella xylostella (L.). Within the range of 15°C to 35°C, the higher temperature, the shorter was the duration of larval and adult stages. Egg hatching and adult emergence were high at 15°C to 30°C but were significantly reduced at 35°C. The higher the temperature, the higher was the proportion of males produced. Temperature threshold was 5.74°C for eggs, 3.80°C for larvae, 5.91°C for pupae and 6.60°C for adults.D. semiclausum oviposition in the first threeP. xylostella larval instars produced more parasite males than females but oviposition in the fourth instar produced significantly more females than males. Parasite adults tended to emerge from their pupae from 06∶00 to 09∶00 hours although some emerged at other hours during the photophase. Adult longevity and production of eggs increased when adults were provided with a food source (honey) compared with no food or provision of water alone. Parasite adults survived and laid eggs for 28 days when provided with food but for only three days when deprived of food.  相似文献

The entomogenous fungusStrongwellsea castrans was isolatedin vitro for the first time, by incubating conidia projected from infected cabbage root flies (Delia radicum) in a simple, semi-defined liquid medium comprising dextrose, yeast extract and lactalbumin hydrolysate buffered to pH 7. The fungus grew as long unitunicate hyphae. After transfer to a solid nutrient medium, multinucleate hyphal bodies were formed which developed a thick, laminated wall. Neither conidia nor resting spores developed in liquid or on solid media and the fungus survived successive sub-culturing only in liquid media. Using the API-ZYM system, tests on extracts on hyphae ofS. castrans were positive for 11 enzymes but there were no consistent differences in enzyme profiles betweenS. castrans and fungi of the related genusErynia.   相似文献

In an attempt to suppress the propagation of citrus greening disease in Indonesia, we explored pathogens ofDiaphorina citri which vectors the disease. At two orange orchards, manyD. citri adults were found to be dead and covered with fungal mycelia. Two fungi,Paecilomyces fumosoroseus andHirsutella citriformis, were consistently isolated from the infected insects. Molecular phylogeny of their 18S rDNA sequences showed that they belong to the ascomycetous clade of the Clavicipitales/Hypocreales, which embraces many entomopathogenic fungi. When healthy adults ofD. citri were inoculated with conidia of theP. fumosoroseus, the insects died within 6 d.  相似文献

Studies were conducted tosearch for fungal strains with potentialpathogenicity against Diabrotica speciosa(Germar) (Coleoptera: Chrysomelidae).Among sixteen fungal isolates screenedthe most virulent was a Beauveria bassiana(Balsamo) Vuillemin isolate (FHD13) thatcaused 70% mortality of D. speciosathird instar larvae. The LC50 value ofB. bassiana isolate FHD13 was3.48 × 1010 conidia/ml.Different temperatures (4, 17 and 26 °C)and vegetable oils (corn, sunflower and canola)used for storage did not significantly affectviability of conidia. A pathogenicity trialagainst D. speciosa larvae performed withthe corn oil formulation (1 × 108 conidia/mlof oil) caused 65% of mortality.  相似文献
