应用IdU-毛玉米油体内SCE技术,以不同剂量的典型诱变剂MMC和CP对70尾黄鳝的脾、肾、血淋巴细胞进行了体内诱发SCE敏感性测试。结果:三种细胞的染色体SCE自发频率均较低,不同剂量MMC和CP诱发黄鳝三种细胞SCE频率均较对照组显著增加。诱变剂剂量与诱发SCE频率呈线性关系。三种细胞染色体SCE对MMC和CP的敏感性次序为肾>脾>血淋巴细胞。与几种鱼和其它动物比较,黄鳝三种细胞的SCE自发频率均较低,对MMC和CP诱发SCE的敏感性均较高,因此认为黄鳝可作为较理想的体内SCE检测系统。  相似文献

Sister chromatid exchange (SCE) frequencies were studied in differentially stained chromosomes from lymphocytes of 17 patients with viral disease. The mean SCE score for the patients was 8.7 +/- 2.9 standard deviations. SCE scores were significantly elevated in the patients compared with the controls (p less than 0.01); however, variability in SCE means was observed in the patients. SCE elevations were also present in long term cultured Epstein Barr virus positive human B lymphocytes.  相似文献

It has been considered by some workers that sister-chromatid exchange (SCE) frequencies are elevated in patients with scleroderma and systemic lupus erythematosus (SLE). However, these observations were based on limited numbers of patients. Other have postulated the presence of a defect in DNA repair in cells from patients with various connective tissue diseases, including scleroderma and SLE. We report our findings from a large survey of SCE frequencies in patients with connective tissue diseases. Their diagnoses are scleroderma, SLE, rheumatoid arthritis, juvenile chronic arthritis, Behcet's syndrome and polyarteritis nodosa. These patients had never received cytotoxic drugs. Healthy individuals, hospital patients with diagnoses other than connective tissue disease and relatives of patients with scleroderma have been used as controls. The results have been analysed by generalized linear modelling, and we have shown that patients with SLE and Behcet's syndrome and controls with viral infections have elevated SCE frequencies, both before and after adjustments have been made for the effect on SCEs of an individual's age, smoking habits, sex and race. The SCEs of patients with scleroderma and their relatives were normal. SCE frequencies increased with age by 4% per decade and the SCE frequencies of smokers were approximately 12% higher than those of nonsmokers of similar age. The sex of an individual did not significantly affect SCEs but individuals from the Middle East were found to have lower counts than those originating from other parts of the world.  相似文献

Chromosomal aberration levels and frequencies of sister chromatid exchanges (SCE) in peripheral blood lymphocytes were investigated for patients with bronchial asthma treated with antilymphocyte globulin (ALG). The significant elevation of chromosomal aberration levels and SCE frequencies were revealed as compared to the healthy controls. The significant differences in chromosomal aberration levels were not observed before and after the courses of ALG therapy. At the same time the SCE frequencies appeared to be significantly decreased after the course of ALG therapy. The similar effects were observed in studies in vitro. The data obtained may suggest that ALG at the used doses does not cause any significant cytogenetic damages in the examined patients.  相似文献

Previous studies of sister-chromatid exchange (SCE) in patients with hepatitis B have been reported. But as far as we know, no such work has been done in children born to parents with hepatitis B, either one or both of whom are infected. In the present study, frequencies of SCE in the peripheral lymphocytes of 30 hepatitis B parents with hepatitis B surface antigen (HBsAg) positive and 40 of their children were observed. SCE frequencies of 20 normal adults and 3 normal children were analysed for comparison with the patients and their children. The results obtained from all of the samples were as follows: The hepatitis B patients with HBsAg positive had a significantly higher SCE frequency than the normal adults (P less than 0.01); the children born after their parents contracted hepatitis B had a significantly higher SCE frequency than normal children (P less than 0.01); there was no significant difference in SCE (P greater than 0.05) between children born after their parents contracted hepatitis B, children born after their mothers acquired it and children born after their fathers acquired it. The above results indicate that hepatitis B patients with HBsAg positive and their children born after they contracted hepatitis B had significantly higher frequencies of SCE; these data might throw new light on the study of genetic factors acting on the mechanism of hepatitis B.  相似文献

It is now possible to examine in detail exchanges between sister chromatids (SCEs) and to attempt to investigate the relationships of such exchanges to aberration formation and DNA-repair mechanisms. The frequency of SCEs is dramatically increased by chemical mutagens and may reflect the level of DNA damage. Lymphocytes from patients with ataxia telangiectasis (AT) show high levels of spontaneous chromosome damage and are hypersentive to ionising radiations and it was of interest to examine the levels of SCE induced in these cells by various mutagens. The frequencies of SCE after treatment with X=rays or three chemical mutagens were equivalent to those in normal cells. The effects of fluorodeoxyuridine and deoxycytidine on SCE frequencies were also tested.  相似文献

Summary Various cigarette smoke condensates (CSC) were analyzed with respect to the induction of sister-chromatid exchanges (SCE) in human lymphocytes in vitro. CSC from a reference cigarette, from three different tobaccos of the reference cigarette, and from a British cigarette induced similar SCE frequencies. Environmental and genetic factors have been implicated as important sources of individual variation in baseline sister-chromatid exchange (SCE) frequency in humans. The current study was designed to test whether the frequency of baseline SCEs in 58 normal blood donors is associated with previously observed variations in SCE frequencies induced by diepoxybutane (DEB). Because 12 subjects were current cigarette smokers and smoking is known to be an in vivo inducer of baseline SCE frequencies, we specifically tested whether higher baseline SCE frequencies in smokers would be associated with in vitro sensitivity to SCE induction by DEB. Analysis of variance showed that DEB-induced SCE frequencies were significantly associated with baseline SCE frequencies; those who were sensitive to SCE induction by DEB were more likely to have higher baseline SCE frequencies. This effect, however, was independent of in vivo induction of SCE by smoking. Chromosomal sensitivity to the induction of SCE by DEB explained approx. 15-20% of the variation in baseline SCE. This was similar in magnitude to the effect of cigarette smoking. Because increased sensitivity to DEB-induced SCEs is common in normal blood donors (approx. 24%) and is associated with an increase in baseline SCEs, it should be investigated as a source of bias and/or a potential marker of sensitivity to environmental mutagens in future cytogenetic studies.  相似文献

In the present paper we have used a rationale based on the development of theoretical equations that define sister-chromatid exchange (SCE) frequencies as a function of two variables, namely the baseline (BrdU-independent) and the BrdU-dependent SCE frequencies. The experimental design includes the estimation of SCE frequencies in second division chromosomes when both cycles occurred in the presence of BrdU and when BrdU incubation took place only during the first cycle in a wide range of BrdU concentrations. The final SCE yields in second division chromosomes could be separated into three different components: (1) The BrdU-independent, ‘spontaneous’ or baseline SCEs, whose low but biologically significant frequency was calculated to be about 0.06 SCEs per pg of DNA; this figure could be similar for most of the cell types; (2) the BrdU-dependent SCEs whose frequency increases with BrdU dose, probably as a result of BrdU substitution for thymidine; (3) the BrdU-dependent SCEs as a consequence of other cellular factors such as disturbance of nucleotide pool sizes. At high BrdU concentrations (300 μM upward) the three components appear to have a significant value in the final SCE yield, whereas at lower BrdU doses the third component seems to be negligible.  相似文献

In vitro treatment with human interferon-alpha (HuIFN-alpha) of hepatitis B virus-infected peripheral lymphocytes from 17 hepatitis B patients induced a decrease in the frequency of sister-chromatid exchanges (SCE). There was a significant difference in mean SCE frequencies between the HuIFN-alpha-treated patients and the control group, but not between acute and chronic hepatitis B patients treated with HuIFN-alpha.  相似文献

Sister-chromatid exchange (SCE) frequencies were studied in blood lymphocytes from 12 patients (3 females and 9 males) with Down's syndrome (DS). The mean frequency of SCE per metaphase for the patients (both sexes) was 9.2 +/- 0.8 which was significantly higher (P less than 0.01) than the mean SCE value (5.1 +/- 0.2) scored for 16 healthy infants (8 females and 8 males). A significant increase in the mean frequency of SCE in 12 parents of infants with DS (8.7 +/- 0.9 SCE/cell) was noticeable when compared with 20 parents of normal infants (6.3 +/- 0.1 SCE/cell). Increases in cellular division with reduction in their replication were also observed in patients with DS. Treatment with mitomycin C (0.05 micrograms/ml), hycanthone (0.1 micrograms/ml) and gamma-radiation (0.1 Gy) revealed a significant (P less than 0.01) increase in frequencies of SCE in DS lymphocytes and in those of their parents as compared to controls. These data may reveal a familial hypersensitivity reaction to these agents. The results indicate a genomic instability and deranged DNA-repair mechanisms which are accentuated by exposure to mutagenic agents, the underlying causal factor for which might be genetic.  相似文献

Summary Sister chromatid exchange (SCE) frequencies were studied in differentially stained lymphocytes from 47 patients with malignant lymphoma. Thirteen patients were untreated when studied. The mean SCE frequency [±standard error (SE)] for these patients was 12.7±0.9 per mitosis. The mean score for 40 controls was 6.1±0.3. SCE mean scores were significantly higher in the untreated patients than in the controls (P<0.001). Seven patients were treated with radiotherapy alone. They demonstrated a mean SCE frequency (8.8±0.8) significantly lower (P<0.01) than that found in untreated patients. Eleven patients received cyclophosphamide within 4 weeks prior to study. They demonstrated a mean SCE frequency (14.3±1.3) significantly higher (P<0.05) than that found in patients who had received regimens that did not contain cyclophosphamide in the prior 4 weeks (11.1±1.3) or who had been off drugs for at least 8 weeks (10.1±0.8). Our data suggest that untreated patients with malignant lymphoma have elevated SCE frequencies, which may be further increased by certain chemotherapeutic agents.  相似文献

Sister-chromatid exchange (SCE) frequency has been studied from peripheral blood lymphocyte cultures of 21 patients with epilepsy on sodium valproate, 20 patients who had not started therapy (untreated) and 20 normal healthy controls. Treated and untreated patients with epilepsy were observed to have higher SCE frequencies (mean 9.05 and 9.82 respectively) than healthy controls (mean 4.8; P < 0.001). There was no significant difference in SCE frequency between treated and untreated patients. This suggests that the disease itself may be associated with an increased frequency of SCEs.  相似文献

Summary Sister chromatid exchange (SCE) frequencies were studied in peripheral lymphocytes from 16 patients with newly diagnosed acute lymphoblastic leukemia (ALL) prior to the initiation of chemotherapy. The mean SCE frequency ( ±SE) for these patients was 12.2±0.2 per metaphase, which was significantly higher (P(0.001) than the mean SCE score for 14 agematched controls, 7.6±0.2. Five of these patients were studied again while they were receiving maintenance therapy consisting primarily of daily 6-mercaptopurine and weekly methotrexate. Their remission SCE levels remained significantly higher than controls (P(0.005). In addition, SCE levels were studied in 7 long-term survivors of ALL. Three of these patients had been receiving continuous maintenance therapy for at least 3 years. Their mean SCE scores were significantly greater than controls (P(0.005). The other 4 patients had finished their final course of chemotherapy at least 8 months prior to the time of sampling, and their mean SCE scores were not significantly different from controls (P>0.10). These data indicate that untreated patients with ALL have increased SCE levels which remain elevated during periods of remission maintained with chemotherapy. However, longterm survivors of ALL who are in remission and off chemotherapy do not demonstrate significantly increased SCE frequencies.  相似文献

Sister-chromatid exchange (SCE) analysis was carried out in different age groups prior to and after therapy with 8-Methoxypsoralen (8-MOP) followed by exposure of the patient to long-wave UV-A (PUVA) and compared to control. The SCE frequencies were increased significantly in PUVA-treated patients as compared to their pre-treatment SCE levels and to controls. A significant increase in SCEs was found in smoking PUVA-treated patients as compared to non-smoking PUVA-treated patients. This study indicates a detectable chromosome-damaging effect of PUVA therapy on its human users.  相似文献

The Brd-U differential staining technique was utilized to examine the induction of sister-chromatid exchanges (SCE) by fluorescent ligt in human fetal lung fibroblasts (IMR-90). Exposure of these cells in media to fluorescent light resulted in an increase in SCE frequencies from a background level of 8.5 SCE/cell to 20.5 SCE/cell. Cellular replication kinetics were also inhibited by fluorescent light exposure. Exposure of cells to fluorescent light in phosphate buffered saline (PBS) resulted in a two-fold increase in SCE levels and incresed inhibition of cell replication, indicating that culture media may have a protective effect. Determinations of SCE frequencies with blocking filters indicated that the fluorescent light wavelengths responsible for SCE induction were in the near-ultraviolet spectrum between 300 and 390 nm. Culturing cell sin media that had been exposed to fluorescent light resulted in a significant increase in SCE levels, 14.5 ± 1.5 vs. 7.5 ± 0.65, demonstrating the contribution of media photoproducts to SCE induction. The role of media photoproducts was further reinforced by finding a significant decline in fluorescent light induced SCE in cells cultured in medium deficient in three known photosensitizers (phenol red, tetracycline and riboflavin) for 2–3 weeks prior to exposure.Since SCE have been shown to be a sensitive indicator of DNA damage, these results indicate that fluorescent light can induce genetic damage in human cells. These findings are also of importance to investigators culturing cells in laboratories with fluorescent illumination.  相似文献

Summary Siter chromatid exchanges (SCE) were analyzed in the cord and postnatal blood of controlled groups of low and high birth weight infants to detect possible associations between abnormal birth weight and SCE frequency. Structural chromosome aberration rates had previously been evaluated for all infants, and possible correlations between aberration and SCE rates were sought.No correlation was found between neonatal or postnatal SCE frequency and birthweight, nor was there evidence of association of chromosome aberration rates with SCE frequency. In all groups of infants, however, mean postnatal SCE frequencies were significantly lower than mean neonatal SCE frequencies.  相似文献

These elevated SCE frequencies tended to decline 3 months after the discontinuation of the pill. Women using a progestogen injectable contraceptive, on the other hand, had unaltered SCE rates.
