The present study aims to evaluate the antihyperlipidaemic, antihypertrophic, and reducing effects of zingerone on isoproterenol‐induced hyperlipidaemia and hypertrophy in rats. Rats were pretreated with zingerone (6 mg/kg body weight) daily for a period of 14 days and then induced myocardial infarction with isoproterenol (100 mg/kg body weight) on days 15 and 16. Isoproterenol increased serum creatine kinase and lactate dehydrogenase activities in the rats. Increased levels/concentrations of serum and heart cholesterol and triglycerides were observed in isoproterenol‐induced myocardial infarcted rats. Isoproterenol also altered serum lipoproteins and the activity of liver 3‐hydroxy‐3‐methyl glutaryl‐coenzyme‐A‐reductase in the rats. The in vitro study revealed a very convincing reducing power of zingerone. Pretreatment with zingerone prevented hyperlipidaemia and cardiac hypertrophy, by virtue of its antihyperlipidaemic, antihypertrophic, and reducing properties in isoproterenol‐induced myocardial infarcted rats.  相似文献

The present study aimed to evaluate the effect of Terminalia pallida fruit ethanolic extract (TpFE) on lipids, lipoproteins, lipid metabolism marker enzymes and paraoxonase (PON) in isoproterenol (ISO)-induced myocardial infarcted rats. PON is an excellent serum antioxidant enzyme which involves in the protection of low density lipoprotein cholesterol (LDL-C) from the process of oxidation for the prevention of cardiovascular diseases. ISO caused a significant increase in the concentration of total cholesterol, triglycerides, LDL-C, very low density lipoprotein cholesterol and lipid peroxidation whereas significant decrease in the concentration of high density lipoprotein cholesterol. ISO administration also significantly decreased the activities of lecithin cholesterol acyl transferase, PON and lipoprotein lipase whereas significantly increased the activity of 3-hydroxy-3-methylglutaryl-coenzyme-A reductase. Oral pretreatment of TpFE at doses 100, 300 and 500?mg/kg body weight (bw) and gallic acid (15?mg/kg bw) for 30?days challenged with concurrent injection of ISO (85?mg/kg bw) on 29th and 30th day significantly attenuated these alterations and restored the levels of lipids, lipoproteins and the activities of lipid metabolizing enzymes. Also TpFE significantly elevated the serum antioxidant enzyme PON. This is the first report revealed that pretreatment with TPFE ameliorated lipid metabolic marker enzymes and increased the antioxidant PON in ISO treated male albino Wistar rats.  相似文献

The effect of doxorubicin (DXR) on the levels of heart, liver and plasma lipids and plasma lipoproteins were studied in rats. Rats were treated with DXR (2.5 mg/kg body weight weekly for 8 weeks, iv) with or without alpha-tocopherol (alpha-TPL) (400 mg/kg body wt daily for 60 days) co-administration. DXR treated rats showed increase in plasma total cholesterol, triglycerides and phospholipids. The activities of lecithin cholesterol-acyl transferase and hepatic and extrahepatic lipoprotein lipase were lowered significantly with concomitant increase in liver and heart lipid peroxide levels in DXR treatment. HDL cholesterol level was found to be decreased significantly in DXR treated rats as a result of which there was an increase of LDLc/HDLc ratio. alpha-TPL coadministration brought back the enzyme activity to near normal and reduced the level of lipid peroxides. The lipid changes were minimum in rats treated with both alpha-TPL and DXR. This study suggests that the toxicity of DXR is reflected in lipids and lipoprotein profile.  相似文献

The present study was designed to evaluate the preventive role of rutin on lipids, lipoproteins, and ATPases in normal and isoproterenol (ISO)-induced myocardial infarction in rats. Rutin (40 and 80 mg/kg) was orally administered to rats for a period of 42 days. After that period, isoproterenol (150 mg/kg) was injected subcutaneously to male wistar rats at an interval of 24 h for 2 days. The weight of heart and the concentrations of total cholesterol, triglycerides, and free fatty acids were increased significantly (p < 0.05), and the concentration of phospholipids was decreased significantly (p < 0.05) in the heart of ISO-treated rats. ISO-treated rats also showed a significant increase (p < 0.05) in the levels of total cholesterol, triglycerides, phospholipids, low-density lipoprotein cholesterol (LDL-C), and very low-density lipoprotein cholesterol (VLDL-C) with a significant decrease (p < 0.05) in high-density lipoprotein cholesterol (HDL-C) level in serum. The activities of sodium potassium dependent adenosine triphosphatase (Na(+)/K(+) ATPase) and magnesium-dependent adenosine triphosphatase (Mg(2+) ATPase) were decreased significantly (p < 0.05), and the activity of calcium-dependent adenosine triphosphatase (Ca(2+)ATPase) was increased significantly (p < 0.05) in the heart in ISO-treated rats. Pretreatment with rutin at doses of 40 or 80 mg/kg to ISO-treated rats showed a significant (p < 0.05) effect in all the parameters studied. Oral administration of rutin to normal rats did not show any significant effect. Thus, the results of our study show that pretreatment with rutin maintained the levels of lipids, lipoproteins, and ATPases in ISO-induced myocardial infarcted rats. The observed effects might be due to the antioxidant potential of rutin.  相似文献

The present study was designed to evaluate the preventive effects of zingerone on circulatory lipid peroxides and nonenzymatic antioxidants in isoproterenol‐induced myocardial infarcted rats. Rats were pretreated with zingerone (6 mg/kg body weight) daily for a period of 14 days and were then induced myocardial infarction with isoproterenol (100 mg/kg body weight) on 15th and 16th day. Increased intensities of serum lactate dehydrogenase isoenzymes 1 and 2 bands enhanced plasma lipid peroxidation products and lowered nonenzymatic antioxidant system were noted in isoproterenol‐induced rats. Pretreatment with zingerone daily for 14 days revealed significant preventive effects on the electrophoretic and biochemical parameters evaluated in isoproterenol‐induced rats. Furthermore, the in vitro study confirmed the potent antioxidant activity of zingerone. The results of our study showed that zingerone protected the rat's heart against isoproterenol‐induced myocardial infarction by its antioxidant effect.  相似文献

This article reports data on the preventive effect of (?)epigallocatechin gallate (EGCG) on lipid metabolism and lipoproteins in isoproterenol (ISO)‐induced myocardial infarction (MI) in Wistar rats. The rats were induced MI by ISO (100 mg/kg) at an interval of 24 h for 2 days. EGCG (30 mg/kg) was given to rats as pretreatment for 21 days orally using an intragastric tube. EGCG significantly reduced the increased serum levels of cholesterol, triglycerides, and free fatty acids in the heart and serum phospholipids (PLs) in ISO‐treated rats. It also significantly increased the reduced levels of heart PLs in ISO‐induced rats. EGCG reduced the levels of serum low‐density lipoprotein cholesterol and very low‐density lipoprotein cholesterol and increased serum high‐density lipoprotein (HDL)‐cholesterol in ISO‐treated rats. It also reduced the increased cholesterol/PL ratio and atherogenic index and significantly increased the reduced ratio of HDL‐cholesterol/total cholesterol. Also EGCG significantly increased the reduced activity of lecithin cholesterol acyl transferase in ISO‐treated rats. Thus, EGCG prevented the accumulation of lipids and altered the levels of lipoproteins in myocardial‐infarcted rats. © 2009 Wiley Periodicals, Inc. This study was designed to evaluate the preventive effect of naringin in isoproterenol (ISO)-induced myocardial infarction (MI) in rats. Rats were pretreated with naringin (10, 20, and 40 mg/kg body weight) orally for a period of 56 days. After the treatment period, ISO (85 mg/kg body weight) was administered subcutaneously to rats at an interval of 24 h for 2 days. There was a significant increase in the levels of total, ester, and free cholesterol, triglycerides (TG), and free fatty acids (FFA) in serum and heart and decrease in heart phospholipids (PL) in ISO-induced rats. Altered levels of lipoproteins and activities of 3-hydroxy-3-methylglutaryl-Coenzyme reductase A in liver and heart, lecithin cholesterol acyl transferase and lipoprotein lipase in plasma were also observed in ISO-induced rats. Pretreatment with naringin (10, 20, and 40 mg/kg) for a period of 56 days significantly decreased the levels of total, ester, and free cholesterol, TG, FFA in serum and heart and increased PL in heart. It also minimized the alterations in serum lipoproteins and lipid metabolic enzymes in ISO-induced rats. Thus, naringin has a lipid-lowering effect in ISO-induced MI rats.  相似文献

The protective action of aspirin in experimental myocardial infraction induced by isoproterenol was studied in rats. Aspirin treated rats showed lower mortality rate and smaller changes in the myocardium on histopathological examination when compared to corresponding animals given isoproterenol alone. Changes were also observed in the different lipid fractions studied. The ratio of cholesterol to phospholipids decreased in the heart in aspirin treated animals when compared to control rats given isoproterenol alone. The levels of lipid peroxide also showed a decrease while the activity of superoxide dismutase (SOD) and catalase registered an increase in the aspirin treated animals given isoproterenol when compared to corresponding animals given isoproterenol alone.  相似文献

The present study was aimed to find out the protective effect of ethanolic extract of E. ribes fruits on homocysteine, lactate dehydrogenase (LDH) and lipid profile in serum, lipid peroxidation (LPO) and non-enzymatic antioxidant glutathione (GSH) levels in brain homogenates and histopathological examination of brain tissue in methionine (1 g/kg body weight, orally for 30 days) induced hyperhomocysteinemic rats. A significant increase in homocysteine, LDH, total cholesterol, triglycerides, low density lipoprotein (LDL-C) and very low density lipoprotein (VLDL-C) levels was observed in serum. Increased LPO levels in brain homogenates with reduced serum high density lipoprotein (HDL-C) levels and decreased GSH content were other salient features observed in methionine treated pathogenic control rats. Administration of ethanolic E. ribes extract (100 mg/kg body weight, orally) for 30 days to methionine-induced hyperhomocysteinemic rats produced a significant decrease in the levels of homocysteine, LDH, total cholesterol, triglycerides, LDL-C, VLDL-C in serum and LPO levels in brain homogenates with significant increase in serum HDL-C levels and GSH content in brain homogenates, when compared with pathogenic control rats. Biochemical observations were further substantiated with histological examination of brain. Degenerative changes of neuronal cells in methionine treated rats were minimized to near normal morphology by ethanolic E. ribes extract administration as evident by histopathological examination. The results provide clear evidence for the first time, that ethanolic E. ribes extract treatment enhances the antioxidant defense against methionine-induced hyperhomocysteinemia and oxidative stress in brain.  相似文献

Hypercholesterolemia, an independent risk factor for increased oxidative renal injury, is associated with the formation of oxidized low-density lipoprotein. Production of reactive oxygen species and nitrogen species have been implicated in diet-induced hypercholesterolemia, principally as means of oxidising low-density lipoproteins. This in turn initiates the accumulation of cholesterol in macrophages, which sets key event in the initiation of atherosclerosis. The aim of the present work is to evaluate the effects of eicosapentaenoic acid (EPA), DL alpha-lipoic acid (LA) and eicosapentaenoate-lipoate derivative (EPA-LA) in controlling the atherogenic disturbances. Four groups of male Wistar rats were fed with a high cholesterol diet (rat chow supplemented with 4% cholesterol and 1% cholic acid; HCD) for 30 days. Among them, 3 groups of rats were treated with either EPA (35 mg/kg body weight/day, oral gavage), LA (20 mg/kg body weight/day, oral gavage) or EPA-LA derivative (50 mg/kg body weight/day, oral gavage) from 16th day to 30th day of the experimental period. Abnormal increase in the levels of reactive oxygen species, 3-nitrotyrosine, malondialdehyde and protein carbonyl as well as an elevation in the activities of xanthine oxidase, lactate dehydrogenase, alkaline phosphatase and acid phosphatase was observed in renal tissue of HCD fed rats. HCD fed rats also showed an increased susceptibility of the apo B-containing lipoproteins to in vitro oxidation. These changes were restored partially in the EPA and LA administered groups. However, the combined derivative EPA-LA almost ameliorated the hypercholesterolemic-oxidative changes in the HCD fed rats.  相似文献

The insulin-like effects of vanadate have been intensively studied in the biological system. Lipids and lipoprotein profiles are altered in diabetes. Rats were made diabetic by a single i.v. injection of streptozotocin (55 mg/kg body weight) in citrate buffer. After the overt of diabetes, the diabetic rats were treated with sodium orthovanadate (0.3 mg/ml) for fifteen days. The altered cholesterol, phospholipids and triglycerides in plasma lipoprotein fractions (HDL, LDL and VLDL) were found to be reverted back to near normal levels in vanadate treated diabetic rats.  相似文献

The aim of the study was to evaluate blood glucose and lipid lowering effects of Umbelliferone (UMB) in streptozotocin (STZ) diabetic rats. Male albino Wistar rats (180 to 200 g) were induced diabetes by administration of STZ (40 mg/kg) intraperitonially. Normal and diabetic rats were treated with UMB in 10 percent dimethyl sulfoxide (DMSO) for 45 days. Diabetic rats had increased plasma glucose and decreased insulin, total proteins (TP), and albumin in addition to decreased food intake and body weight. Elevation in total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), very low density lipoprotein cholesterol (VLDL-C), triglycerides (TG), free fatty acids (FFA), and phospholipids (PL), and reduction in high density lipoprotein cholesterol (HDL-C) in the plasma were observed. Liver and kidney tissues of diabetic rats had elevation in the levels of TC, TG, FFA, and PL. Treatment with UMB decreased plasma glucose and increased insulin, TP, and albumin apart from food intake and body weight. In UMB-treated diabetic rats, plasma and tissue TC, TG, PL and FFA, and plasma LDL-C, VLDL-C, and HDL-C reversed to near normal. Thus, reduction of blood glucose and lipid profiles indicates that UMB has antidiabetic and antihyperlipidemic effects in diabetic rats.  相似文献

Extract of the flower Malva viscus conzattii (M. conzattii) was administered at a dose of 25/50 mg/day/animal to 30 healthy adult male gerbils and 30 adult male house rats to determine its effect on fertility. After 25 days' treatment fin l body weight, and the weights of testes, epididymides, seminal vesicles, and adrenal glands were measured. Testis, epididymides, and seminal vesicles were prepared for histological examination and total protein, RNA, sialic acid, and alkaline phosphatase activity were determined. Quantitative estimation of cholesterol was also made. While overall body weight remained stable during treatment, testicular weight in both animals was drastically decreased. A complete spermatogenic arrest in the testes was evident in house rats treated with 50 mg/day for 20 days and in the gerbil treated with 25 mg/day for 25 days. The seminiferous tubules showed marked degeneration, lined by 1 or 2 cell layers. Epididymides showed degenerative changes as well. RNA contents of the testes, epididydmides, and seminal vesicles of treated anials were significantly lowered as was sialilc acid content. Total cholesterol was increased significantly. M. conzattii causes an effective inhibition of spermatogenesis in gerbils and house rats in 25 states and induces infertility.  相似文献

Ligustrum lucidum Ait. has been used in traditional Chinese medicine for over 1000 years because of its anti-tumor, antimutagenic, antidiabetic, and hepatoprotective properties. The aim of this study was to determine whether oleanolic acid (OA) is the principal active compound of L. lucidum responsible for its antidiabetic properties, and to examine its effect on the expression of thyroid hormones and insulin secretion, thus revealing the mechanism by which L. lucidum modulates insulin levels in diabetes. When rats with streptozotocin-induced diabetes were treated with OA (100 and 200 mg/kg body mass per day, for 40 days), the changes in blood glucose levels and in oral glucose tolerance tests showed that hypoglycemia was more pronounced in OA-treated groups than in the diabetic control rats, and that the levels of triglyceride, total cholesterol, and low-density lipoportein cholesterol in OA-treated rats were lower than those in the diabetic control rats, whose high-density lipoprotein cholesterol increased. OA-treated rats also gained weight, and exhibited increased serum insulin levels. In contrast, OA treatment did not effect the levels of thyroid hormone or TSH in rats with streptozotocin-induced diabetes. These results indicate that OA has hypoglycemic and hypolipidemic effects. OA treatment might stimulate insulin release, and consequently, results in the modulation of glucose levels and regulation of lipid metabolism.  相似文献

Effects of albendazole against larval and adult stages of Angiostrongylus cantonensis in rats were examined. (1) A single oral dose of albendazole of 10, 50 or 100 mg/kg at 7 or 14 days post-infection (PI) showed that the use of drug at 7 days PI was more effective in the larval infection. Rats treated 7 days PI showed significant reductions in the relative wet weight of heart and lungs (g/100 g body wt.), the mean total number of recovered worms and the mean body length of worms, as compared to those in the non-treated control. Similarly, visual morbidity assessment of the lung-tissues revealed a marked reduction in pathological changes in the rats treated 7 days PI. (2) Following two or three successive oral doses of 100 mg/kg at weekly intervals from 6 weeks PI, the first-stage larvae in rat feces completely disappeared 2 weeks post-treatment (PT) and this disappearance lasted during the experiment. In rats treated once, however, larval output reappeared from 3 weeks PT. Both histological sections of the rat lung-tissues and the recovered female worms showed degenerative changes in the female reproductive systems.  相似文献

This study investigated the effect of administration of α-lipoic acid (LA) on lipid metabolism in high fructose–fed insulin-resistant rats. High-fructose feeding (60 g/100 g diet) to normal rats resulted in a significant increase in the concentrations of cholesterol, triglycerides (TGs), free fatty acids (FFAs), and phospholipids in plasma, liver, kidney, and skeletal muscle. Reduced activities of lipoprotein lipase (LPL) and lecithin cholesterol acyl transferase (LCAT) and increased activity of the lipogenic enzyme hydroxymethylglutaryl–coenzyme A (HMG-CoA) reductase were observed in plasma and liver. High-density lipoprotein cholesterol (HDL-C) was significantly lowered and very low-density lipoprotein cholesterol (VLDL-C) and low-density lipoprotein cholesterol (LDL-C) were significantly elevated. Treatment with LA (35 mg/kg body weight intraperitoneal) reduced the effects of fructose. The rats showed near-normal levels of lipid components on plasma and tissues. Activities of key enzymes of lipid metabolism were also restored to normal values. Cholesterol distribution in the plasma lipoproteins was normalized, resulting in a favorable lipid profile. This study demonstrates that LA can alter lipid metabolism in fructose-fed insulin-resistant rats and may have implications in the treatment of insulin resistance.  相似文献

The present study was designed to investigate the antioxidant effect of curcumin on methionine-induced hyperlipidemia and hyperhomocysteinemia in Wistar rats (200-250 g) of either sex. The vehicle control rats were treated with 1% Tween 80 in normal saline (2 ml/kg, po) for 30 days. Hyperlipidemia and hyperhomocysteinemia was induced by methionine administration (1 g/kg, po) for 30 days. A significant increase in total cholesterol, triglycerides, low density lipoprotein cholesterol (LDL-C) and homocysteine levels in serum and thiobarbituric acid reactive substances (TBARS) levels in heart homogenates were observed with a concomitant decrease in serum high density lipoprotein (HDL-C) levels in pathogenic control (i.e. group II) rats, as compared to vehicle control (i.e. group I) rats. Further, curcumin (200 mg/kg, p.o.) treatment in methionine treated rats for 30 days significantly decreased the total cholesterol, triglycerides, LDL-C and homocysteine levels in serum and TBARS levels in heart homogenates and increased serum HDL-C levels, as compared to pathogenic control (i.e. group II) rats. The results of biochemical observations were supplemented by histopathological examination of rat's aortic section. The results of test drug were comparable to that obtained with folic acid (100 mg/kg, p.o.). The results suggest that curcumin has significant antihyperlipidemic and antihyperhomocysteinemic effect against methionine-induced hyperlipidemia and hyperhomocysteinemia in rats.  相似文献

This study was aimed to evaluate the combined effects of quercetin and α-tocopherol on lipid metabolism and glycoprotein components in isoproterenol induced myocardial infarction in Wistar rats. Myocardial infarction in rats was induced by isoproterenol (100 mg/kg) at an interval of 24 h for 2 days. Quercetin (10 mg/kg) and α-tocopherol (10 mg/kg) were given to rats as pretreatment for 14 days orally using an intragastric tube. Quercetin and α-tocopherol significantly reduced the levels of cholesterol, triglycerides and free fatty acids in the serum and heart and serum phospholipids and significantly increased the levels of heart phospholipids in isoproterenol induced rats. They also significantly decreased the activity of plasma and liver 3-hydroxy-3-methylglutaryl-coenzyme-A reductase and increased the activity of plasma and liver lecithin cholesterol acyl transferase in isoproterenol treated rats. In addition to this, they also significantly reduced the levels of hexose, hexosamine, fucose and sialic acid in the serum and heart of isoproterenol treated rats. Quercetin and α-tocopherol also showed significant decrease in plasma lipid peroxidation products (thiobarbituric acid reactive substances and lipid hydroperoxides). Pretreatment with quercetin alone and α-tocopherol alone showed significant effect in all the biochemical parameters in myocardial infarcted rats. But, combined pretreatment with quercetin and α-tocopherol normalized all the above mentioned biochemical parameters in isoproterenol treated myocardial infarction in rats. Thus, the experiment clearly showed that quercetin and α-tocopherol prevented the accumulation of lipids and glycoprotein components in myocardial infarcted rats by their anti-lipid peroxidative effect. This study also showed that combined pretreatment was better than single pretreatment.  相似文献

This study was designed to evaluate the anti-inflammatory and anti-apoptotic effects of the alcoholic extract of the berries of Crataegus oxyacantha (AEC), a medicinal herb, on isoproterenol-induced myocardial infarction (MI) in a rat model. Three groups of Wistar albino rats, each comprising six animals, were selected for this study. Group I rats served as control. Group II rats were given isoproterenol (85 mg/kg body weight) subcutaneously on 59th and 60th days. Group III rats were given AEC (0.5 ml/100 g body weight/day), orally on a daily basis for 60 days, and isoproterenol (85 mg/kg body weight, subcutaneously) was given on 59th and 60th days. On the 61st day, the animals were sacrificed, and marker enzymes like lactate dehydrogenase (LDH) and creatine kinase (CK) were estimated in serum. In the heart tissue sample, antioxidant status, lipid peroxidation and anti-inflammatory properties of AEC were determined. Isoproterenol significantly increased the release of LDH, CK in serum, decreased the antioxidant status in the heart along with an increase in lipid peroxidation. Nitritive stress and apoptosis were seen in isoproterenol-induced rat heart. Pre-treatment with the AEC for 60 days had a significant effect on all the above factors and maintained near normal status. The study confirms the protective effect of AEC against isoproterenol-induced inflammation and apoptosis-associated MI in rats.  相似文献

Globally, human exposure to heavy metals has risen dramatically. Lead (Pb) is one of the most toxic heavy metals to human and other living organisms. Pb affects certain biochemical and physiological activities of the body. Many scientific investigations have documented the therapeutic and antioxidant properties of natural products which isolated from plant sources. The present study was therefore undertaken to evaluate the therapeutic influence of almond oil against Pb toxicity in male rats. The experimental rats were distributed into four groups. The first group was served as control. The second group was treated with 100 mg/kg body weight of Pb. The third group was subjected to almond oil (800 mg/kg body weight) and Pb. The fourth group was supplemented with almond oil. After six weeks, blood serum specimens were analyzed. In the second group, Pb produced a marked increase of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), gamma glutamyl transferase (GGT), total bilirubin, glucose, triglycerides, low density lipoprotein cholesterol (LDL-C), very low density lipoprotein cholesterol (VLDL-C), creatine kinase (CK), lactate dehydrogenase (LDH), creatinine, blood urea nitrogen (BUN), uric acid and malondialdehyde (MDA) levels, while the levels of total protein, albumin, high density lipoprotein cholesterol (HDL-C), glutathione (GSH), superoxide dismutase (SOD) and catalase (CAT) were significantly decreased. In contrast, the treatment with almond oil notably improved the biochemical changes and showed antioxidative effect. The present study disclosed the therapeutic influence of almond oil on the basis of its antioxidant effect against Pb toxicity. Moreover, these new findings indicated that the constituents of almond oil have a promising significant potential in biomedical and pharmacological studies.  相似文献
