【目的】大肠杆菌脂多糖(LPS)核心型根据其化学结构的不同分为5种,即R1、R2、R3、R4和K12。通过对禽致病性大肠杆菌(Avian pathogenic Escherichia coli,APEC)安徽、江苏、上海和河南等省市分离株的脂多糖核心型分布情况的研究,分析其与大肠杆菌主要毒力基因之间的潜在联系,以期为APEC的研究和防治提供参考。【方法】对分离到的76株APEC,利用PCR方法开展对LPS核心型分型鉴定和毒力基因检测;分析LPS核心型的分布和毒力基因、致病性之间的相关性。【结果】在76株APEC分离株中,68.4% (52株)为R1核心型,15.8% (12株)为R3型,11.8% (9株)为R4型,3.9% (3株)为R2型,未检测到K12核心型。毒力基因鉴定结果中yijp、mat、fimC、ibeB和ompA的检验阳性率均达到90%以上,可作为APEC的保守基因。其中LPS核心型R1与neuC、cva/cvi、irp2均具有显著正相关性(P<0.05),R3与iroN、irp2均具有显著负相关性(P<0.05),R4核心型与aatA显著正相关(P<0.05)。【结论】APEC的LPS核心型主要为R1。LPS核心型对部分毒力基因分布具有显著影响。  相似文献

[目的]检测禽致病性大肠杆菌IMT5155自分泌黏附素基因等具有代表性的疑似毒力基因在不同来源大肠杆菌中的分布,为进一步研究其致病机理提供依据.[方法]采用PCR和Dot blot,检测疑似毒力基因在不同地区(101株大肠杆菌中国分离株和121株大肠杆菌德国分离株)、不同来源(人源、禽源及猪源)大肠杆菌中的分布,并分析其和大肠杆菌系统进化分群的关系.[结果]自分泌黏附素基因B11等11个疑似毒力基因在禽致病性大肠杆菌中分布率较高,阳性率分别为:A1 36.4%(32/88)、A8 53.4%(47/88)、A1063.6%(56/88)、B1137.5%(33/88)、F3 59.1%(52/88)等,且疑似毒力基因主要存在于大肠杆菌B2进化群中.值得注意的是,D1、E9和F11基因片段在新生儿脑膜炎大肠杆菌中有较高的分布率,分别为60%(6/10)、80%(8/10)和90%(9/10),而在新生儿脑膜炎大肠杆菌中未检测到B11基因.[结论]自分泌黏附素B11等疑似毒力基因与禽致病性大肠杆菌关系密切,但疑似毒力基因D1、E9和F11与新生儿脑膜炎大肠杆菌密切相关,提示禽致病性大肠杆菌可能是新生儿脑膜炎大肠杆菌的毒力基因储库.  相似文献

禽致病性大肠杆菌毒力基因多重PCR方法的建立和应用   总被引:1,自引:0,他引:1  
【目的】建立禽致病性大肠杆菌(Avian pathogenic Escherichia coli,APEC)黏附相关基因、侵袭及毒素相关基因、抗血清存活相关基因及铁转运相关基因的多重PCR方法,实现禽致病性大肠杆菌毒力基因的简便、快速检测。【方法】根据GenBank公布的基因序列,设计合成18对特异性引物,通过条件优化,建立四组多重PCR体系,并通过模板倍比稀释检测各组多重PCR的灵敏性。利用多重PCR检测100株APEC毒力基因的分布,验证多重PCR方法的可行性。【结果】根据PCR扩增片段大小判定,上述四组多重PCR体系均能同时扩增出该组中的各个毒力基因,且灵敏度分别为:103CFU、103CFU、105CFU、105CFU细菌和1ng、1ng、10ng、10ng DNA。100株APEC的毒力因子检测结果显示,多重PCR和单基因PCR结果一致。【结论】建立的四组多重PCR方法能够简便、快速地检测禽致病性大肠杆菌的毒力基因,可用于毒力基因的鉴定以及流行病学调查。  相似文献

【目的】构建禽致病性大肠杆菌(Avian pathogenic Escherichia coli,APEC)rmlA基因缺失株,研究该缺失株的生物学特性。【方法】利用Red重组系统构建rmlA缺失株;比较野生株与缺失株在生长特性、运动性和生物被膜形成能力等方面的差异;运用Real-time PCR技术,比较野生株与rmlA缺失株对APEC部分毒力基因转录的影响。【结果】rmlA缺失株,不影响APEC的生长和运动特性,但生物被膜形成能力显著增强,且使luxS、irp2基因转录水平分别上调2倍、1.8倍,iucD、fyuA则下调25倍。【结论】APEC的rmlA基因可以影响禽致病性大肠杆菌的生物被膜形成能力及部分毒力基因的转录水平;而对APEC的生长、运动特性没有影响。  相似文献

【目的】构建禽致病性大肠杆菌(Avian pathogenic Escherichia coli,APEC)VI型分泌系统2(Type VI secretion system 2,T6SS2)结构基因vgrG缺失株,研究其对APEC生物学特性及致病性的影响。【方法】通过Red同源重组方法构建DE719菌株vgrG基因缺失株,并利用低拷贝质粒pSTV28构建互补株。比较分析野生株、缺失株与互补株的生长特性、运动性、生物被膜形成能力、黏附侵袭能力、动物致病力等差异。【结果】vgr G基因缺失不影响DE719的生长速度、运动能力及生物被膜形成能力。致病性试验表明缺失vgrG导致体内定殖能力及致病力显著下降,然而对DF-1细胞的黏附能力增强。【结论】T6SS2核心组分VgrG在APEC感染过程中发挥重要作用,为了解APEC的致病作用提供参考。  相似文献

【目的】研究gsp L基因缺失对禽致病性大肠杆菌(Avian pathogenic Escherichia coli,APEC)生物学特性的影响。【方法】利用Red重组方法构建禽致病性大肠杆菌DE17株的gsp L缺失株;分析野生株与缺失株的生长特性、黏附和入侵DF1细胞的差异;采用荧光定量PCR的方法比较野生株和缺失株毒力基因转录水平的变化;比较野生株与缺失株的半数致死量(LD50)差异。【结果】gsp L缺失不影响DE17的生长特性,但其黏附和入侵DF1细胞能力显著下调。荧光定量PCR检测结果表明,缺失株毒力基因lux S,pfs,fyu A和iss转录水平明显上调,tsh的转录水平明显下调,而vat,ibe A,stx2f和omp A的转录水平无显著变化;LD50检测结果表明,缺失株比野生株毒力增强了12倍。【结论】gsp L基因的缺失不影响禽致病性大肠杆菌的生长特性,但能减弱其黏附和入侵能力,且可以正调控禽致病性大肠杆菌部分毒力基因的转录水平,推测gsp L基因可能与APEC对宿主的致病性有关。  相似文献

【背景】禽致病性大肠杆菌(Avian pathogenic Escherichia coli, APEC)是禽类主要病原菌之一,群体感应(Quorumsensing,QS)系统可通过信号分子调控其生物学特性。在APEC中信号分子AHL对其生物学特性的影响目前尚不清楚。【目的】研究信号分子AHL对APEC生物学特性的影响。【方法】将含铜绿假单胞菌酰基高丝氨酸内脂合成酶(Acyl-homoserine-lactone synthase,lasI)基因的表达质粒转化至APEC菌株DE17中,构建重组菌株DE17-lasI,利用LasI在DE17中合成AHL。比较野生株和重组菌株产生AHL信号分子、生长特性、生物被膜形成能力、运动性以及耐药性等生物学特性的差异;运用Real-timePCR技术,比较野生株和重组菌株中与生物被膜形成、运动性以及毒力因子相关基因的转录水平。【结果】对重组菌株AHL信号分子检测表明,DE17-lasI能够产生AHL信号分子,与野生株DE17相比,DE17-lasI生物被膜形成能力和运动性显著降低(P0.01),但其生长特性和耐药性无显著变化(P0.05);Real-time PCR检测结果表明,重组菌株的毒力因子fimH转录水平上调了58.8倍,而ompA、iss分别下调了95.4%、77.3%。与生物被膜形成相关基因agn43下调了75%,鞭毛合成基因flhA下调了80.8%。此外,AHL受体sdiA的转录水平上调了19.8倍。【结论】转化lasI至APEC中,能促进其在APEC中合成信号分子AHL,并显著影响APEC的部分生物学特性,为进一步探讨AHL型群体感应系统对APEC的调控作用提供参考。  相似文献

【目的】禽致病性大肠杆菌(Avian pathogenic Escherichia coli,APEC)不仅严重影响全球的养禽业,对人类公共健康也造成巨大的潜在威胁。pag P基因在细菌的抗菌肽抗性和致病性方面发挥重要作用,但关于pag P基因在APEC中的功能尚不清楚。本文构建禽致病性大肠杆菌pag P基因缺失株,对缺失株的抗菌肽抗性和致病性进行研究。【方法】利用Red重组系统构建APEC的pag P基因缺失株,然后利用回复质粒构建回复株。研究pag P基因对细胞黏附与入侵、生物被膜形成能力、外膜渗透性、抗菌肽敏感性、致病性等方面的影响。【结果】成功构建pag P基因缺失株和回复株,抗菌肽抗性试验发现pag P基因缺失株对多粘菌素B、鸡β-防御素2(AVBD2)的敏感性显著增加(P0.01),致病性试验结果表明pag P基因缺失株的毒力显著降低(P0.01)。【结论】APEC的pag P基因对AVBD2的敏感性和APEC的致病性密切相关,为深入研究pag P基因的功能及调控作用奠定了基础。  相似文献

目的了解副溶血性弧菌食物中毒和临床腹泻株Ⅲ型分泌系统的分布以及耐药特征。方法对食物中毒和临床腹泻分离到的共21株副溶血性弧菌进行毒力基因tdh、trh、T3SS1、T3SS2α、T3SS2β和toxR检测,并用VITEK 2 compact全自动微生物鉴定系统进行了耐药性分析。结果 21株菌株中tdh+/trh-占90.48%(19/21),tdh-/trh+和tdh-/trh-分别占4.76%、4.76%,未检测到tdh+/trh+菌株。T3SS1广泛存在于所有菌株中。T3SS2α存在于tdh+/trh-菌株,T3SS2β存在于tdh-/trh+菌株。1株食物中毒菌株毒力基因携带情况为tdh-/trh-/T3SS2α-/T3SS2β-。21株副溶血性弧菌对阿莫西林、头孢吡肟、抗菌素B、庆大霉素、环丙沙星和复方新诺明敏感,对氨苄西林完全耐药。结论食物中毒和临床腹泻分离到的菌株大多携带tdh基因,T3SS2α与tdh相关,而T3SS2β则存在于trh+菌株。未携带tdh和trh基因的食物中毒分离株表明副溶血性弧菌不仅仅依赖TDH和TRH发挥毒力作用,其致病机制具有多样性和复杂性。  相似文献

【背景】禽致病性大肠杆菌(avian pathogenic Escherichia coli,APEC)是禽类主要病原菌之一,大肠杆菌三型分泌系统2 (Escherichia coli type III secretion system 2,ETT2)可通过转录调节子调控其致病性,但在APEC中转录调节子EtrA对其致病性的影响目前尚不清楚。【目的】研究ETT2中转录调节子EtrA对APEC致病性的影响。【方法】利用Red同源重组技术构建ETT2-etrA基因缺失株及回复株。比较生长性能、生物被膜形成、运动性及对血清敏感性的差异,基于RNA-Seq测序及Real-timePCR技术比较野生株和缺失株中与生物被膜形成、运动性以及毒力因子相关基因的转录水平。【结果】与野生株相比,缺失株及回复株生长特性无显著变化(P0.05),但APEC40-ΔetrA生物被膜形成能力和对血清敏感性明显增强(P0.001),运动性较野生株明显下降(P0.01),回复株的表型有所回复。转录组学筛选出7个毒力差异基因,生物被膜形成相关基因显著上调,参与影响细菌运动性的基因显著下调。qRT-PCR验证与转录组学结果一致。【结论】etrA缺失可以显著影响APEC的生物被膜形成、运动性及对血清的敏感性,这可为进一步探讨ETT2对APEC的致病作用提供参考。  相似文献

Autotransporters are secreted bacterial proteins exhibiting diverse virulence functions. Various autotransporters have been identified among Escherichia coli associated with intestinal or extraintestinal infections; however, the specific distribution of autotransporter sequences among a diversity of E. coli strains has not been investigated. We have validated the use of a multiplex PCR assay to screen for the presence of autotransporter sequences. Herein, we determined the presence of 13 autotransporter sequences and five allelic variants of antigen 43 (Ag43) among 491 E. coli isolates from human urinary tract infections, diarrheagenic E. coli, and avian pathogenic E. coli (APEC) and E. coli reference strains belonging to the ECOR collection. Clinical isolates were also classified into established phylogenetic groups. The results indicated that Ag43 alleles were significantly associated with clinical isolates (93%) compared to commensal isolates (56%) and that agn43K12 was the most common and widely distributed allele. agn43 allelic variants were also phylogenetically distributed. Sequences encoding espC, espP, and sepA and agn43 alleles EDL933 and RS218 were significantly associated with diarrheagenic E. coli strains compared to other groups. tsh was highly associated with APEC strains, whereas sat was absent from APEC. vat, sat, and pic were associated with urinary tract isolates and were identified predominantly in isolates belonging to either group B2 or D of the phylogenetic groups based on the ECOR strain collection. Overall, the results indicate that specific autotransporter sequences are associated with the source and/or phylogenetic background of strains and suggest that, in some cases, autotransporter gene profiles may be useful for comparative analysis of E. coli strains from clinical, food, and environmental sources.  相似文献

Escherichia coli strains that cause disease outside the intestine are known as extraintestinal pathogenic E. coli (ExPEC) and include pathogens of humans and animals. Previously, the genome of avian-pathogenic E. coli (APEC) O1:K1:H7 strain O1, from ST95, was sequenced and compared to those of several other E. coli strains, identifying 43 genomic islands. Here, the genomic islands of APEC O1 were compared to those of other sequenced E. coli strains, and the distribution of 81 genes belonging to 12 APEC O1 genomic islands among 828 human and avian ExPEC and commensal E. coli isolates was determined. Multiple islands were highly prevalent among isolates belonging to the O1 and O18 serogroups within phylogenetic group B2, which are implicated in human neonatal meningitis. Because of the extensive genomic similarities between APEC O1 and other human ExPEC strains belonging to the ST95 phylogenetic lineage, its ability to cause disease in a rat model of sepsis and meningitis was assessed. Unlike other ST95 lineage strains, APEC O1 was unable to cause bacteremia or meningitis in the neonatal rat model and was significantly less virulent than uropathogenic E. coli (UPEC) CFT073 in a mouse sepsis model, despite carrying multiple neonatal meningitis E. coli (NMEC) virulence factors and belonging to the ST95 phylogenetic lineage. These results suggest that host adaptation or genome modifications have occurred either in APEC O1 or in highly virulent ExPEC isolates, resulting in differences in pathogenicity. Overall, the genomic islands examined provide targets for further discrimination of the different ExPEC subpathotypes, serogroups, phylogenetic types, and sequence types.  相似文献

AIMS: To assess the abilities of 105 avian pathogenic Escherichia coli (APEC) and 103 avian faecal commensal E. coli (AFEC) to form biofilms on a plastic surface and to investigate the possible association of biofilm formation with the phylotype of these isolates. METHODS AND RESULTS: Biofilm production was assessed in 96-well microtitre plates using three different media, namely, M63 minimal medium supplemented with glucose and casamino acids, brain-heart infusion broth, and diluted tryptic soy broth. Avian E. coli are highly variable in their ability to form biofilms. In fact, no strain produced a strong biofilm in all three types of media; however, most (75.7% AFEC and 55.2% APEC) were able to form a moderate or strong biofilm in at least one medium. Biofilm formation in APEC seems to be mostly limited to nutrient deplete media; whereas, AFEC are able to form biofilms in both nutrient deplete and replete media. Also, biofilm formation in E. coli from phylogenetic groups B2, D and B1 was induced by nutrient deplete conditions; whereas, biofilm formation by members of phylogenetic group A was strongest in a rich medium. CONCLUSIONS: Biofilm formation by APEC and phylotypes B2, D and B1 is induced by nutrient deplete conditions, while AFEC are able to form biofilms in both nutrient rich and deplete media. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study to investigate biofilm formation by a large sample of avian E. coli isolates, and it provides insight into the conditions that induce biofilm formation in relation to the source (APEC or AFEC) and phylogenetic group (A, B1, B2 and D) of an isolate.  相似文献

ABSTRACT: BACKGROUND: Uropathogenic E.coli (UPEC) are among major pathogens causing urinary tract infections. Virulence factors are mainly responsible for the severity of these emerging infections. This study was planned to investigate the distribution of virulence genes and cytotoxic effects of UPEC isolates with reference to phylogenetic groups (B2, B1, D and A) to understand the presence and impact of virulence factors in the severity of infection in Faisalabad region of Pakistan. METHODS: In this study phylogenetic analysis, virulence gene identification and cytotoxicity of 59 uropathogenic E.coli isolates obtained from non-hospitalized patients was studied. RESULTS: Among 59 isolates, phylogenetic group B2 (50%) was most dominant followed by groups A, B1 (19% each) and D (12 %). Isolates present in group D showed highest presence of virulence genes. The prevalence hlyA (37%) was highest followed by sfaDE (27%), papC (24%), cnf1 (20%), eaeA (19%) and afaBC3 (14%). Highly hemolytic and highly verotoxic isolates mainly belonged to group D and B2. We also found two isolates with simultaneous presence of three fimbrial adhesin genes present on pap, afa, and sfa operons. This has not been reported before and underlines the dynamic nature of these UPEC isolates. CONCLUSIONS: It was concluded that in local UPEC isolates from non-hospitalized patients, group B2 was more prevalent. However, group D isolates were most versatile as all were equipped with virulence genes and showed highest level of cytotoxicity.  相似文献

There is scarce data about the importance of phylogroups and virulence factors (VF) in bloodstream infections (BSI) caused by extended-spectrum β-lactamase-producing Escherichia coli (ESBLEC). A prospective multicenter Spanish cohort including 191 cases of BSI due to ESBLEC was studied. Phylogroups and 25 VF genes were investigated by PCR. ESBLEC were classified into clusters according to their virulence profiles. The association of phylogropus, VF, and clusters with epidemiological features were studied using multivariate analysis. Overall, 57.6%, 26.7%, and 15.7% of isolates belonged to A/B1, D and B2 phylogroups, respectively. By multivariate analysis (adjusted OR [95% CI]), virulence cluster C2 was independently associated with urinary tract source (5.05 [0.96-25.48]); cluster C4 with sources other than urinary of biliary tract (2.89 [1.05-7.93]), and cluster C5 with BSI in non-predisposed patients (2.80 [0.99-7.93]). Isolates producing CTX-M-9 group ESBLs and from phylogroup D predominated among cluster C2 and C5, while CTX-M-1 group of ESBL and phylogroup B2 predominantes among C4 isolates. These results suggest that host factors and previous antimicrobial use were more important than phylogroup or specific VF in the occurrence of BSI due to ESBLEC. However, some associations between virulence clusters and some specific epidemiological features were found.  相似文献

Although the number of Escherichia coli bacteria in surface waters can differ greatly between locations, relatively little is known about the distribution of E. coli pathotypes in surface waters used as sources for drinking or recreation. DNA microarray technology is a suitable tool for this type of study due to its ability to detect high numbers of virulence and antimicrobial resistance genes simultaneously. Pathotype, phylogenetic group, and antimicrobial resistance gene profiles were determined for 308 E. coli isolates from surface water samples collected from diverse aquatic ecosystems at six different sites in the St. Clair River and Detroit River areas. A higher frequency (48%) of E. coli isolates possessing virulence and antimicrobial resistance genes was observed in an urban site located downstream of wastewater effluent outfalls than in the other examined sites (average of 24%). Most E. coli pathotypes were extraintestinal pathogenic E. coli (ExPEC) pathotypes and belonged to phylogenetic groups B2 and D. The ExPEC pathotypes were found to occur across all aquatic ecosystems investigated, including riverine, estuarine, and offshore lake locations. The results of this environmental study using DNA microarrays highlight the widespread distribution of E. coli pathotypes in aquatic ecosystems and the potential public health threat of E. coli pathotypes originating from municipal wastewater sources.  相似文献

Bacteremia is the principal way of dissemination of local infections to distant organs. Escherichia coli bacteremia is almost always clinically significant, suggesting an increased risk of developing sepsis syndrome. Fifty-one E. coli bloodstream human isolates were analyzed using PCR technique for several molecular markers associated with extraintestinal virulence, and their phylogenetic group assignment, taking into account the link between the phylogenetic background and the intrinsic virulence of this species. Sixteen virulence genotypes have been identified, the majority of the blood isolates carrying the association of two genes. The genes encoding type 1 fimbria and aerobactin had the highest prevalence. As a confirmation of other studies, the strains assigned to E. coli phylogenetic group B2 exhibited the highest concentration of virulence genes, and represented almost half of the clinical blood isolates. The multifactorial virulence of E. coli strains isolated from invasive infections reflects a phylogenetic inheritance, and supports the concept of ExPEC pathotype as a subset of E. coli population involved in human infectious diseases. The surveillance of geographical variation of E. coli pathogenic clones is useful for epidemiological analysis.  相似文献
