Endophytic filamentous actinobacteria were isolated from surface-sterilized roots of wheat plants. Endophytic colonization of germinating wheat seed was examined using one of these endophytes, Streptomyces sp. strain EN27, tagged with the egfp gene. 内生细菌影响兰科植物菌根形成和共生关系的稳定性,在兰科植物的生活史中起着重要作用。内生细菌通过分泌植物激素、采用光合作用、生物固氮或促进矿质营养的循环以及产生铁载体、合成其他活性物质等途径来促进兰科植物生长发育。综述了兰科植物内生细菌物种多样性的研究方法及其对兰科植物的促生机理,基于兰科植物与共生微生物的密切关系,认为内生细菌间、内生细菌与兰科植物菌根真菌间的互作是揭示兰科植物与内生细菌互作机理的重要方向。  相似文献

番茄根内促生放线菌的分离鉴定及其促生效果   总被引:1,自引:0,他引:1  
【背景】植物体内普遍存在一定数量的内生放线菌,对植物的生长发育具有促生作用。【目的】从番茄根内分离、筛选并鉴定出能够有效促进植物生长发育的内生放线菌,为生物菌肥的开发奠定基础。【方法】采用组织研磨培养法和放线菌分离培养基对番茄根内放线菌进行分离,利用Salkowski比色法、钼锑抗比色法和CAS平板检测法进一步筛选出具有较强促生特性的菌株,通过番茄和黄瓜苗期盆栽试验验证其促生效果。结合形态、生理生化以及16S r RNA基因序列相似性和系统发育分析,对菌株进行鉴定。【结果】分离筛选出一株吲哚乙酸(Indole-3-acetic acid,IAA)产量达25.56 mg/L的内生放线菌NEAU-D1,能够产生铁载体并且对多种难溶性磷酸盐具有良好溶解效果,通过16S r RNA基因序列分析,该菌株属链霉菌属。番茄和黄瓜苗期盆栽试验结果表明,接种该菌株的番茄幼苗其根长、株高、植株鲜重和干重较对照分别显著增加了9%、23%、47%和92%,而接种该菌株的黄瓜幼苗根长、株高、植株鲜重和干重较对照分别显著增加了43%、47%、134%和58%。【结论】链霉菌NEAU-D1可以作为潜在的促生菌资源用于设施蔬菜多功能生物菌肥的研发。  相似文献

AIMS: To isolate endophytic Streptomyces strains from tomato and examine their antimicrobial activity. METHODS: Endophytic Streptomyces strains were isolated using surface-sterilization methods and identified by morphological characteristics. Antimicrobial activities were measured by the agar plate sensitivity method. Antifungal activity in vivo was measured by seedling mortality in infested soils. RESULTS: Twenty-one per cent of endophytic streptomycete isolates produced antibacterial metabolites and 41% produced antifungal metabolites in S medium. Sixty-five per cent of the most frequently isolated strains inhibited the growth of Rhizoctonia solani by the antibiosis assay but only 32% produced metabolites active against R. solani in S medium. Growth promotion and enhanced disease resistance of seedlings inoculated with Streptomyces sp. strain S30 were observed in tomato but not in cucumber seedlings. CONCLUSIONS: Endophytic Streptomyces spp. strains were successfully isolated using stringent methods and strain S30 promoted growth and enhanced resistance to R. solani in tomato seedlings. SIGNIFICANCE AND IMPACT OF THE STUDY: Endophytic streptomycetes showing antifungal activity in vitro and in vivo may indicate the potential for their use as biocontrol agents particularly of R. solani disease of tomato seedlings.  相似文献

通过研究桃儿七(Sinopodophyllum hexandrum)与南方山荷叶(Diphylleia sinensis)的内生真菌的生物活性,探讨内生真菌作为生物活性物质资源开发利用的价值。测定内生真菌发酵液的抗细菌、抗真菌、抗卤虫活性后,发现大多数菌株的发酵液都具有不同的生物活性。高活性菌株集中于镰孢霉属、柱孢属、木霉属、色串孢属。桃儿七内生真菌抗真菌活性较强,南方山荷叶内生真菌抗细菌和抗卤虫活性较强。桃儿七和南方山荷叶内生真菌是寻找有价值的活性成分的良好资源。  相似文献

内生拟盘多毛孢是植物 (尤其是木本植物 )内生真菌的重要类群。分离自红豆杉的内生小孢拟盘多毛孢(Pestalotiopsismicrospora)能产生抗癌的代谢产物紫杉醇 ,因而引起了人们对内生拟盘多毛孢研究的兴趣。拟盘多毛孢内生于植物的根、茎、叶及繁殖体中 ,已鉴定的内生拟盘多毛孢有 2 3种。在已调查的植物中 ,内生拟盘多毛孢的定殖率有很大差异 ,在红树 (Rhizophoraapiculata)、椰子 (Cocosnucifer)、西藏红豆杉 (Taxuswallichiana)、茶梅 (Camelliasasanqua)、Fragraeabodenii和Cordemoyaintegrifolia等木本植物中 ,拟盘多毛孢是内生真菌的优势类群。一种拟盘多毛孢可内生于多种植物体内 ,一种植物也可有多种拟盘多毛孢内生 ,其分布与植物所处的生态环境和地域有关。在一定的宿主植物生理条件或外界环境条件下 ,某些内生拟盘多毛孢可转变为寄生或腐生。对植物内生拟盘多毛孢代谢产物的研究表明 ,拟盘多毛孢可产生多种次生代谢产物 ,包括抗癌物质 (紫杉醇和粗榧酸 )和抗菌物质 (环己烷类物质、粗榧酸和 pestaloside)。植物内生拟盘多毛孢的资源调查和开发应用具有广阔的前景。  相似文献

利用组织培养法对秦巴山区虎杖进行内生真菌分离,通过内生真菌的液体发酵,对发酵液和菌丝的乙酸乙酯萃取液进行HPLC分析检测,筛选出一株产白藜芦醇苷的菌株M-56,其产量达1.029 mg/L。根据该菌株的形态特征及菌丝ITS序列分析,将该株菌确定为无性型真菌丝孢纲链格孢菌Alternaria alternaria。  相似文献

AIMS: To isolate endophytic fungi from vegetable plants and examine their in vivo anti-oomycete activity against Phytophthora infestans in tomato plants. METHODS AND RESULTS: Endophytic fungi were isolated from surface-sterilized plant tissues and anti-oomycete activity was measured by in vivo assay using tomato seedlings. Endophytic fungi showing potent anti-oomycete activity were identified by morphological characteristics and nuclear ribosomal ITS1-5.8S-ITS2 sequence analysis. A total of 152 isolates were obtained from 66 healthy tissue samples of cucumber, red pepper, tomato, pumpkin and Chinese cabbage and the fermentation broths of 23 isolates showed potent in vivo anti-oomycete activity against tomato late blight with control values over 90%. The Fusarium oxysporum strain EF119, which was isolated from roots of red pepper, showed the most potent disease control efficacy against tomato late blight. In dual-culture tests, it inhibited the growth of Pythium ultimum, P. infestans and Phytophthora capsici. CONCLUSIONS: Among endophytic fungi isolated from healthy tissues of vegetable plants, F. oxysporum EF119 showed the most potent in vivo anti-oomycete activity against tomato late blight and in vitro anti-oomycete activity against several oomycete pathogens. SIGNIFICANCE AND IMPACT OF THE STUDY: Endophytic fungi showing anti-oomycete activity in vitro and in vivo may be used as biocontrol agents particularly of tomato late blight.  相似文献

对新疆昌吉和石河子两地种植的甜菜内生菌进行了分离、鉴定和分析,结果表明甜菜内生菌多属于细菌,其中假单胞菌 (Pseudomonas sp. )和芽孢菌类(Bacillus sp.)的分离频率分别在33.2%~59.2%和12.7%~28.1%,是甜菜植株中的优势内生菌群.16S rDNA 和 ITS 序列同源性比较和系统发育分析表明内生菌具有丰富的多样性.根中内生菌的多样性高于茎、叶,昌吉地区种植的甜菜中分离出的内生菌种类较多.从感病品种及生长不良甜菜植株中分离出的内生菌种类比较丰富.通过回接分离及利用扫描电镜观察内生菌在植物体内分布发现,内生菌能够定殖于甜菜块根.  相似文献
