Globally destructive crop pathogens often emerge by migrating out of their native ranges. These pathogens are often diverse at their centre of origin and may exhibit adaptive variation in the invaded range via multiple introductions from different source populations. However, source populations are generally unidentified or poorly studied compared to invasive populations. Phytophthora infestans, the causal agent of late blight, is one of the most costly pathogens of potato and tomato worldwide. Mexico is the centre of origin and diversity of P. infestans and migration events out of Mexico have enormously impacted disease dynamics in North America and Europe. The debate over the origin of the pathogen, and population studies of P. infestans in Mexico, has focused on the Toluca Valley, whereas neighbouring regions have been little studied. We examined the population structure of P. infestans across central Mexico, including samples from Michoacán, Tlaxcala and Toluca. We found high levels of diversity consistent with sexual reproduction in Michoacán and Tlaxcala and population subdivision that was strongly associated with geographic region. We determined that population structure in central Mexico has contributed to diversity in introduced populations based on relatedness of U.S. clonal lineages to Mexican isolates from different regions. Our results suggest that P. infestans exists as a metapopulation in central Mexico, and this population structure could be contributing to the repeated re‐emergence of P. infestans in the United States and elsewhere.  相似文献

Odonate diversity in the Coalcomán Mountain Range (CMR), Michoacán State, Mexico, was surveyed, and samplings were made during 2 years in eight streams along an altitudinal gradient. Presence–absence data were analyzed using non-parametric and parametric methods. Beta and gamma diversities were estimated using Whittaker’s and Lande’s formulae, respectively. A total of 2,526 adults and 489 larvae were captured, yielding 116 species (γ diversity), 44 genera and 9 families. Five new species were discovered. The genus Argia was the most important contributor to Zygoptera diversity and total richness (γ diversity), yielding 40.4 and 14.7%, respectively. The non-parametric estimator Chao2 provided the closest theoretical estimate of species richness, and Clench’s model fit the data well (R 2 ranged from 99.44 to 99.99) to explain a high proportion of the variance (98.8). We conclude that beta diversity is important at the landscape scale, supporting the hypothesis that Mexico is a beta diverse country. Our results triple the number of known species of Odonata for Michoacán. Given the considerable richness of odonates at local and landscape scales, our results support the proposal of the Coalcomán Mountain Range as a priority area for conservation and related research.  相似文献

While microsatellites have been used to examine genetic structure in local populations of Neotropical trees, genetic studies based on such high-resolution markers have not been carried out for Mesoamerica as a whole. Here we assess the genetic structure of the Mesoamerican mahogany Swietenia macrophylla King (big-leaf mahogany), a Neotropical tree species recently listed as endangered in CITES which is commercially extinct through much of its native range. We used seven variable microsatellite loci to assess genetic diversity and population structure in eight naturally established mahogany populations from six Mesoamerican countries. Measures of genetic differentiation (FST and RST) indicated significant differences between most populations. Unrooted dendrograms based on genetic distances between populations provide evidence of strong phylogeographic structure in Mesoamerican mahogany. The two populations on the Pacific coasts of Costa Rica and Panama were genetically distant from all the others, and from one another. The remaining populations formed two clusters, one comprised of the northern populations of Mexico, Belize and Guatemala and the other containing the southern Atlantic populations of Nicaragua and Costa Rica. Significant correlation was found between geographical distance and all pairwise measures of genetic divergence, suggesting the importance of regional biogeography and isolation by distance in Mesoamerican mahogany. The results of this study demonstrate greater phylogeographic structure than has been found across Amazon basin S. macrophylla. Our findings suggest a relatively complex Mesoamerican biogeographic history and lead to the prediction that other Central American trees will show similar patterns of regional differentiation.  相似文献

During years 2001, 2002 and 2003 the gravity of the Fusarium wilt in 1000 hectares of melon culture was evaluated in Colima (Mexico). In spite of the soil disinfections with methyl bromide, the losses could reach 25% of the final production. The analysis of 4 soil samples from the fields with ill plants, in a selective medium for Fusarium, allowed to detect the presence of F. oxysporum. By means of the presented technique “soil phytopathometry”, 31 isolates of F. oxysporum f. sp. melonis were obtained from the soil samples. The isolates were inoculated on melon plants to evaluate their pathogenicity. The 31 isolates inoculated, produced the symptoms of chlorosis and wilting, in melon cultivars that allowed us to affirm that all isolates were race 1 of F. oxysporum f. sp. melonis. Being this the first news of the presence of F. oxysporum f. sp. melonis in the state of Colima (Mexico).  相似文献

To investigate the distribution and diversity of the pathogens associated with Fusarium crown rot in the Huanghuai wheat-growing region (HHWGR) of China, we collected wheat samples with symptomatic stem bases from seven provinces in the HHWGR between 2013 and 2016. A total of 1196 isolates obtained from 222 locations were identified as 9 Fusarium species based on morphological and molecular identification. Of these pathogen species, F. pseudograminearum was the dominant species. Furthermore, F. sinensis was isolated from the disease specimens and tested for virulence to wheat. The result of the pathogenicity revealed that an intraspecific differentiation existed in F. pseudograminearum; sequence analysis of the EF-1α gene showed that 194 F. pseudograminearum isolates were differentiated into two distinct clades which closed to the strains from Australia and China respectively, but neither pathogenicity nor EF-1α sequence was related to the geographic origins of these isolates. However, universal rice primers-polymerase chain reaction showed a correlation with the geographical origins of the 194 isolates, which were divided into eight subclusters, the level of genetic diversity was higher within a geographical population than among the different populations. The results of these analyses can be directly used to facilitate disease monitoring and development of control strategies.  相似文献

 In the genus Pinus the internal transcribed spacers (ITS1 and ITS2) and the 5.8s region of the nuclear ribosomal DNA are approximately 3000 bp in length. ITS1 is considerably longer than ITS2 and partial sequences of ITS1 indicate that this region is evolving rapidly and exhibits intraspecific variation. The ITS2 and 5.8s regions are relatively conserved. We surveyed restriction fragment length variability of PCR-amplified fragments (PCR-RFLP) of the ITS region in four populations (86 individuals) of Pinus rzedowskii, a pine endemic to western Michoacán, Mexico. Five of the restriction endonucleases assayed revealed variation, with a total of 13 variants, most of which were length mutations of 300–900 bp. A moderate degree of population differentiation was detected. The average diversity (Shannon’s index) of ITS fragment size patterns was 1.19, with 34% of the variation due to differences among populations and 66% due to differences among individuals within populations. The same individuals were assayed for nine polymorphic isozymes, which gave diversity measures similar to those of each restriction endonuclease. Aim To delimit the distribution of matrilines of the black iguana (Ctenosaura pectinata), detect potential contact zones between them, and test the hypothesis of geographic concordance in the structuring of matrilines with respect to genetic structuring across the nuclear genome. Location Pacific coast of Mexico in the states of Nayarit, Jalisco, Colima, Michoacán in the lowlands adjacent to the western end of the Mexican Volcanic Belt. Methods Tissue samples were obtained every 10–20 km along a transect across the range of three previously described mitochondrial DNA (mtDNA) clades of C. pectinata. Iguanas were genotyped with 11 nuclear microsatellites and maternal lineages were inferred based on mtDNA sequences. Geographic structure and geographic concordance between biparentally and maternally inherited markers were analysed with clustering methods, FST and NST indexes as well as haplotype networks. Results Geographic structure was evident for both markers and the existence of contact zones confirmed. Nevertheless, the distribution of nuclear and mtDNA genetic variation is not geographically concordant. Four matrilines exist in the area, whereas only two nuclear clusters occur. A contact zone between nuclear clusters extends along a distance of c. 60 km, and introgression is evident in individuals from several localities. Main conclusions High levels of genetic diversity were detected on the western coast of Mexico. Historical and contemporaneous processes seem to shape the distribution of genetic variation in C. pectinata. There are no evident geographic, environmental or ecological barriers that coincide with genetic subdivisions, and the observed mtDNA structure is likely to be the result of past climatic changes. The less structured distribution of nuclear genetic variation is consistent with the homogenizing effect of male‐biased dispersal. Our findings have implications for the taxonomy and conservation strategies for this threatened species, and highlight the fact that geographic analyses of both cytoplasmic and nuclear genetic variation are important for the meaningful inference of evolutionary and demographic histories.  相似文献

Fusarium species belonging to the Fusarium fujikuroi species complex (FFSC) are associated with maize in northern Mexico and cause Fusarium ear and root rot. In order to assess the diversity of FFSC fungal species involved in this destructive disease in Sinaloa, Mexico, a collection of 108 fungal isolates was obtained from maize plants in 2007–2011. DNA sequence analysis of the calmodulin and elongation factor 1α genes identified four species: Fusarium verticillioides, F. nygamai, F. andiyazi and F. thapsinum (comprising 79, 23, 4 and 2 isolates, respectively). Differential distribution of Fusarium species in maize organs was observed, that is F. verticillioides was the most frequently isolated species from maize seeds, while F. nygamai predominated on maize roots. Mixed infections with F. verticillioides/F. thapsinum and F. verticillioides/F. nygamai were detected in maize seeds and roots, respectively. Pathogenicity assay demonstrated the ability of the four species to infect maize seedlings and induce different levels of disease severity, reflecting variation in aggressiveness, plant height and root biomass. Isolates of F. verticillioides and F. nygamai were the most aggressive. These species were able to colonize all root tissues, from the epidermis to the vascular vessels, while infection by F. andiyazi and F. thapsinum was restricted to the epidermis and adjacent cortical cells. This is the first report of F. nygamai, F. andiyazi and F. thapsinum infecting maize in Mexico and co‐infecting with F. verticillioides. Mixed infections should be taken into consideration due to the production and/or accumulation of diverse mycotoxins in maize grain.  相似文献

Fusarium proliferatum, F. subglutinans, and F. verticillioides are known causes of ear and kernel rot in maize worldwide. In Mexico, only F. verticillioides and F. subglutinans, have been reported previously as causal agents of this disease. However, Fusarium isolates with different morphological characteristics to the species that are known to cause this disease were obtained in the Highland-Valley region of this country from symptomatic and symptomless ears of native and commercial maize genotypes. Moreover, while the morphological studies were not sufficient to identify the correct taxonomic position at the species level, analyses based in the Internal Transcribed Spacer region and the Nuclear Large Subunit Ribosomal partial sequences allowed for the identification of F. subglutinans, F. solani, and F. verticillioides, as well as four species (F. chlamydosporum, F. napiforme, F. poae, and F. pseudonygamai) that had not previously been reported to be associated with ear rot. In addition, F. napiforme and F. solani were absent from symptomless kernels. Phylogenetic analysis showed genetic changes in F. napiforme, and F. pseudonygamai isolates because they were not true clones, and probably constitute separate sibling species. The results of this study suggest that the biodiversity of Fusarium species involved in ear rot in Mexico is greater than that reported previously in other places in the world. This new knowledge will permit a better understanding of the relationship between all the species involved in ear rot disease and their relationship with maize.  相似文献

Aim A phylogeographic study of the endemic Mexican tulip poppy Hunnemannia fumariifolia (Papaveraceae) was conducted to determine: (1) the historical processes that influenced its geographical pattern of genetic variation; (2) whether isolation by distance was one of the main factors that caused genetic divergence in populations of this species; and (3) whether genetic flow still exists between populations from northern arid zones (Chihuahuan Desert and Sierra Madre Oriental) and those from southern arid zones (Tehuacán‐Cuicatlán Valley) – populations that are separated by the Transvolcanic Belt. Location Xerophytic vegetation in Mexico. Methods Chloroplast DNA (cpDNA) sequences of three regions, trnH‐psbA, rpl32‐trnL(UAG) and ndhF‐rpl32, were obtained for 85 individuals from 17 populations sampled in the field, covering the entire range of H. fumariifolia. The evolutionary history of these populations was investigated using a nested clade phylogeographic analysis and also by conducting various population genetic analyses. Results In total, 17 haplotypes were detected, 14 of which were found in the Sierra Madre Oriental. Differentiation among populations based on cpDNA variation (GST = 0.787, SE 0.0614) indicated population structure in H. fumariifolia, corroborated by a fixation index (FST) of 0.907. Results from analysis of molecular variance found that most of the total variation (90.71%, P < 0.001) was explained by differences among populations. Three regions were determined based on geological correspondence – the Chihuahuan Desert, Sierra Madre Oriental and Tehuacán‐Cuicatlán Valley – and the variation between them was significant (43.39%, P < 0.001). Results of a Mantel test showed a significant correlation between genetic and geographic distances (r = 0.511; P = 0.0001), suggesting a pattern of isolation by distance, which was corroborated by nested clade phylogeographic analysis. Mismatch distribution analysis indicated a sudden demographic expansion. Main conclusions Our study found that isolation by distance influenced genetic divergence in populations of H. fumariifolia. The finding that allopatric fragmentation influenced genetic divergence in populations in the Sierra Madre Oriental may be a reflection of the complex geology of the area. Our results suggest that the areas located in the north of the Sierra Madre Oriental acted as post‐glacial refugia for some populations.  相似文献

Fusarium crown and root rot of tomato (Lycopersicon esculentum) caused by Fusarium oxysporum f. sp. radicis‐lycopersici is a new devastative disease of tomato greenhouse crops in Tunisia. Nothing is known neither about the population of this pathogen in this region, nor about the population of F. oxysporum f. sp. lycopersici the causal agent of Fusarium wilt of tomato. In order to examine the genetic relatedness among the F. oxysporum isolates by intergenic spacer restriction fragment length polymorphism (IGS‐RFLP) analysis and to elucidate the origin of the formae specialesradicis‐lycopersici in Tunisia by looking for genetic similarity of Tunisians isolates with isolates from a foreign source, the genetic diversity among F. oxysporum f. sp. radicis‐lycopersici and F. oxysporum f. sp. lycopersici populations was investigated. A total of 62 isolates of F. oxysporum, obtained from symptomless tomato plants, were characterized using IGS typing and pathogenicity tests on tomato plants. All Fusarium isolates were highly pathogenic on tomato. Fusarium oxysporum f. sp. radicis‐lycopersici isolates were separated into five IGS types. From the 53 F. oxysporum f. sp. radicis‐lycopersici isolates, 34 isolates have the same IGS types (IGS type 25), and the remaining 19 isolates were distributed into four IGS types. However, the only nine isolates of F. oxysporum f. sp. lycopersici have six different IGS types. This difference of diversity between the two formae speciales suggests that F. oxysporum f. sp. radicis‐lycopersici isolates have a foreign origin and may have been accidentally introduced into Tunisia.  相似文献

The columnar cactus Stenocereus stellatus is used in Central Mexico for its edible fruits which are harvested in wild, managed in situ and cultivated populations. Management in situ of wild populations is conducted by selectively sparing and enhancing the abundance of plants with desirable phenotypes when fields are cleared for agricultural use. Cultivation of desirable phenotypes is carried out by vegetative propagation in homegardens. Effects of human management on morphological and genetic variation of S. stellatus were analyzed by comparing morphological diversity indices (MD, based on Simpson’s index) and expected (He) heterozygosity indices from allozyme analysis, in wild, managed in situ, and cultivated populations from La Mixteca and the Tehuacán Valley regions. Morphological diversity was similar among regions, but populations from the wetter La Mixteca region averaged higher genetic variation (He = 0.279) than populations from Tehuacán (He = 0.265). On average, populations manipulated by people had higher levels of variation (MD = 0.479 ± 0.012, He = 0.289 in cultivated populations; MD = 0.461 ± 0.014, He = 0.270 in managed in situ populations) than wild populations (MD = 0.408 ± 0.017, He = 0.253), which is apparently due to a continual introduction and replacement of plant materials in the manipulated populations. The results illustrate that human management may not only maintain but also increase both morphological and genetic diversity of manipulated plant populations in relation to that existing in the wild. Managed in situ and cultivated populations of S. stellatus are important reservoirs of variation, and are crucial for the general maintenance of diversity in wild populations. These populations may play a principal role in designing strategies for the conservation of variation of this cactus.  相似文献

Genetic diversity and recombination underlie the long‐term persistence and evolution of species and are strongly influenced by population size, breeding system and plant longevity. Here, we study genetic structure in the rare Senecio macrocarpus in southeastern Australia to guide current conservation practices. Thirteen neutral microsatellite markers and two chloroplast regions were used to survey the 20 known S. macrocarpus populations and one sympatric S. squarrosus population, a morphologically similar species. All markers showed severe excess or deficit of heterozygotes and linkage disequilibrium was significant. Microsatellite markers revealed 100 multi‐locus genotypes (MLGs) from 523 S. macrocarpus individuals and a further 4 MLGs from 27 S. squarrosus individuals. MLGs varied in frequency and distribution. At the extremes, one MLG was found 108 times across the sampling region and 66 MLGs were found once. The MLGs of all 38 seedlings genotyped were identical to their seed parents implying an asexual origin. Chloroplast regions showed little variation within S. macrocarpus but differed from S. squarrosus. Chromosome counts for S. macrocarpus revealed the same ploidy level as S. squarrosus (2n = 6x = 60) and pollen–ovule ratios were typical of erechthitoid Senecio species showing self‐compatibility. Results suggest that establishment of small populations occur primarily from one extensive source population with indications that both apomixis and selfing may be contributing to its reproduction cycle. We suggest that this species may contribute to future evolutionary processes despite limited genotypic variation and restricted distribution. The loss of genetic diversity in fragmented populations ofself-incompatible plant species may result in sexual reproductive failure andlocal extinctions. Florida ziziphus (Ziziphus celata) is aself-incompatible clonal shrub known only from five genetically depauperatepopulations on the Lake Wales Ridge, Florida, USA. Recovery of this speciesrequires identification of cross-compatible genotypes that can be used to createviable (i.e., sexually reproducing) populations. To further development of arecovery program for this highly imperiled species, we investigated its geneticstructure and sexual reproductive viability. We used random amplifiedpolymorphic DNAs (RAPDs) to investigate genetic variability within remnantpopulations and we conducted experimental compatibility trials to determine thecross-compatibility of remnant genotypes. One hundred and ninety-nine uniquestem samples collected from one ex situ and fivein situ populations were assayed for the presence orabsence of a band for 32 RAPD markers. Based on unweighted pair-group meancluster analysis (UPGMA), only 11 multi-locus genotypes (MLGs) were identified.Eight of these MLGs correspond to MLGs identified in an earlier allozyme study.In addition, we identified three new RAPD-based MLGs. Three of the five naturalpopulations consisted of only one MLG, while the largest and most geneticallydiverse population comprised only four MLGs. Coefficients of similarity rangedfrom 96.6% for the most closely related MLGs to 20.7% for the most distantlyrelated. The compatibility trials demonstrated that most MLGs arecross-incompatible. With 69% of all possible one-way crosses tested (38/55), wehave identified only eight compatible crosses via germination trials. Based onthe results of the compatibility trials, we assigned MLGs toself-incompatibility (SI) mating types. On present evidence, the currentbreeding population of Florida ziziphus may comprise as few as two SI matingtypes. These SI mating types will be used to guide future breeding efforts andan experimental introduction.  相似文献

Recent studies have revealed extensive genetic variation among isolates of Cryptosporidium parvum, an Apicomplexan parasite that causes gastroenteritis in both humans and animals worldwide. The parasite's population structure is influenced by the intensity of transmission, the host-parasite interaction, and husbandry practices. As a result, C. parvum populations can be panmictic, clonal, or even epidemic on both a local scale and a larger geographical scale. To extend the study of C. parvum populations to an unexplored region, 173 isolates of C. parvum collected in Italy from humans and livestock (calf, sheep, and goat) over a 10-year period were genotyped using a multilocus scheme based on 7 mini- and microsatellite loci. In agreement with other studies, extensive polymorphism was observed, with 102 distinct multilocus genotypes (MLGs) identified among 173 isolates. The presence of linkage disequilibrium, the confinement of MLGs to individual farms, and the relationship of many MLGs inferred using network analysis (eBURST) suggest a predominantly clonal population structure, but there is also evidence that part of the diversity can be explained by genetic exchange. MLGs from goats were found to differ from bovine and sheep MLGs, supporting the existence of C. parvum subpopulations. Finally, MLGs from isolates collected between 1997 and 1999 were also identified as a distinct subgroup in principal-component analysis and eBURST analysis, suggesting a continuous introduction of novel genotypes in the parasite population.  相似文献

Ochtodes searlesii Mendoza‐González, Mateo‐Cid et Sentíes sp. nov. is described from Michoacán, tropical Mexican Pacific, on the basis of comparative morphology and rbcL sequence analysis. It is distinguished from other Ochtodes species by its erect axes arising from an encrusting base, its small terete fronds, regularly dichotomously branched axes, and obliquely divided zonate tetrasporangia. Phylogenetic analyses showed that three Pacific Mexican samples, from Caletilla, Zapote and La Majahuita (Michoacán), were identical and formed a distinctive and well supported Clade segregated from other species of Ochtodes from Brazil, Cuba, Ecuador, Guadeloupe and Mexico. The Mexican entity is morphologically distinct from other Ochtodes species as well. On this basis we propose a new Ochtodes species.  相似文献

Fusarium poae is one of the Fusarium species isolated from grains associated with Fusarium head blight (FHB), whose occurrence has increased in the last years. In this study, a total of 105 F. poae isolates from Argentina, Belgium, Canada, England, Finland, France, Germany, Hungary, Italy, Luxembourg, Poland, Switzerland and Uruguay were evaluated using sequence‐related amplified polymorphism (SRAP) to analyse the capacity of this molecular marker to evaluate the F. poae genetic variability. The molecular analysis showed high intraspecific variability within F. poae isolates, and a partial relationship was revealed between variability and the host/geographic origin. Analysis of molecular variance (amova ) indicated a high genetic variability in the F. poae collection, with most of the genetic variability resulting from differences within, rather than between American and European populations. The analysis of sequenced SRAP fragments targets into hypothetical proteins from different Fusarium species showing that the SRAP technique not only allows studying F. poae genetic variability, but also targets coding regions into the F. poae genome. To our knowledge, this is the first report on genetic variability of F. poae using SRAP technique and also demonstrates the efficacy of this molecular marker to amplify open reading frames in fungus.  相似文献

Isolates of Fusarium obtained from Dactylopius opuntiae (Cockerell) (Hemiptera: Dactylopiidae) demonstrated potential as biological control agents against that same insect, which is a pest on Opuntia ficus-indica L Miller. The isolates belong to two species complexes: Fusarium incarnatum-equiseti (FIESC – five species) and Fusarium fujikuroi (FFSC – one species). Twenty-eight isolates of these fungi were characterised using seven Inter Simple Sequence Repeats (ISSR) primers. The UBC841 primer differentiated all the FIESC isolates studied and the single isolate of Fusarium pseudocircinatum O’Donnell & Nirenberg at a level greater than 90% similarity for the fragment sizes. The results indicated high genetic variability among those isolates, an important characteristic for biological control, increasing the chances of finding efficient fungi for insect control. The ISSR markers UBC834 and UBC841 were found to be efficient for characterising and differentiating (DNA fingerprinting) those fungi, and could be used in field monitoring.  相似文献
