Injection of taurodeoxycholate (TDOC) alone or in combination with 10−5 M Ca2+(Ca-EGTA buffer) into newt eggs induced a cup- or furrow-like depression on the egg surface. Reduction of the Ca2+ concentration inhibited the response. These findings imply that TDOC induces a cytoplasmic contraction associated with the membrane and that a micromolar Ca2+ concentration is required for this process. Injection of 10−5 M Ca2+(Ca-EGTA buffer) alone had no effect. On the basis of these findings, the roles of TDOC and Ca2+ in induction of contraction were discussed.  相似文献

Clove oil as an anaesthetic for adult sockeye salmon: field trials   总被引:9,自引:0,他引:9  
Wild migrating sockeye salmon Oncorhynchus nerka exposed to 20, 50 and 80 mg 1−1 of clove oil could be handled within 3 min, recovered within 10 min, and survived 15 min exposure trials. Fish tested at 110 mg 1−1 did not recover from 15 min exposure trials. Response curves developed for induction and recovery time considered the following predictors: clove oil concentration, sex, fish length and depth. A significant positive dependence was observed between induction time and fish length for 20, 50 and 80 mg 1−1 test concentrations; no dependence was observed between induction time and length at 110 and 140 mg 1−1. Recovery time differed as a function of clove oil concentration, but not fish size. A concentration of 50 mg 1−1 is recommended for anaesthetizing sockeye salmon ranging in length from 400 to 550 mm at water temperatures averaging 9–10° C.  相似文献

No significant differences were noted between responses of rainbow trout Oncorhynchus mykiss facial and glossopharyngeal nerves to 15 amino acids. Nine of these amino acids tested at 10−2 M were stimulatory, whereas only two tested at 10−3 M were effective gustatory stimuli. For both nerve systems, ≤10−3 M L-proline was the most stimulatory amino acid, with an estimated threshold of 10−7 M; however, L-α-amino-β-guanidino-propionic acid (estimated threshold of 3×10−3 M), was the most potent compound at 10−2 M. These results indicate that the same amino acids activate taste buds innervated by facial and glossopharyngeal nerves, respectively, and suggest that the same amino acids can be important in chemosensory feeding behaviour in the rainbow trout.  相似文献

The behaviour of juvenile Arctic charr, Salvelinus alpinus (L.) , to an abrupt concentration step of L-amino acids, L-alanine and ammonium chloride was studied by fluviarium technique. The emission rates of these substances were studied. Juvenile Arctic charr emit 8.0 × 10−4 mol total ammonia-N kg−1 h−1 and 3.3 × I0−5 mol amino acids kg−1 h−1. In behaviour tests the charr avoided 5.6x 10−6and 5.6 × 10−7 M ammonium chloride. The 17 L-amino acid mixture, ranked as observed in the analysis of emission, was avoided at 4.6 × 10−7 M, while 100 times dilution of this value gave neither avoidance nor attraction. The charr avoided L-alanine tested alone at the concentration of 4.6 × 10 −7 M. Anosmic charr showed neither avoidance nor attraction to the mixture of 17 amino acids tested at 4.6 × 10−7 M. The results indicate that ammonia as well as emitted amino acids are not responsible for the olfactory mediated attraction to conspecific odour shown earlier in Arctic charr. On the contrary, these substances may have a negative effect by reducing the strength of attraction.  相似文献

The virucidal properties of iodophor, chlorine (sodium hypochlorite), formalin, thimerosal (organic mercurial compound), malachite green, and acriflavine were tested on infectious pancreatic necrosis virus (IPNV). Iodine and chlorine showed good activity, but efficacy depended on the concentration of virus, the presence of organic matter (calf serum), and water pH. Water hardness (0-300 mg 1−1 as CaCO3) did not affect virucidal activity. In a 5 min exposure, 4 mg 1−1 available iodine inactivated 103.9 TCID50 m1−1 IPNV but 16 mg 1−1 iodine were needed for inactivation of 106.3TCID50m1−1. The addition of 0-5% calf serum significantly reduced the iodine concentration and the virucidal activity. In comparison, 4 mg 1−1 chlorine were needed to inactivate 1046 TCID50 m1−1 IPNV in 5 min. However, the addition of 0-07 % serum greatly reduced the chlorine concentration and extended the virucidal contact time to 30 min or more. IPNV at 106.3 TCID60 m1−1 was not inactivated by exposures for 60 min to 0-2% formalin, 10 min to 0-2% thimerosal, 60 min to 5 mg 1−1 malachite green, or 20 min to 500 mg 1−1 acriflavine. However, acriflavine at 0-5 mg 1−1 in cell culture media prevented the development of cytopathology caused by IPNV and may be useful in the treatment of the disease.  相似文献

The role of ethylene in microspore embryogenesis was analyzed by studying the effect of cobalt ions, silver ions, methionine and Ethrel on the androgenic response of in vitro cultured anthers of Datura metel L. Cobalt and silver ions, provided in the form of cobalt chloride and silver nitrate, respectively, decreased the average number of embryos/plantlets developed per anther, at all the concentrations tested. In contrast, methionine (10−5–10−3 M ) and Ethrel (10−6 and 10−5 M ) stimulated embryo formation. The optimal enhancement was recorded at 10−5 M of both methionine and Ethrel.  相似文献

The growth of Fucus vesiculosus L. germlings in chemically defined culture media containing a range of Cu concentrations (20–1000 nM) was monitored simultaneously with measurement of the Cu speciation in the media by competitive equilibrium-adsorptive cathodic stripping voltammetry. Fucus vesiculosus germlings were found to exude Cu-complexing ligands with conditional stability constants of the order of 1.6 × 1011. Ligand concentrations increased with increasing total dissolved Cu concentrations (CuT) until a concentration of 500–800 neq Cu·L−1 was reached. Concentrations of the ligand exceeded CuT in treatments containing 20 and 100 nM Cu, were similar to CuT in the 500-nM Cu treatment, but were less than CuT in the 1000-nM treatment. Therefore, [Cu2+] were calculated to be at concentrations of 10−11− 10−10 M in the 20- and 100-nM treatments, 10−9 M in the 500-nM treatment, and 10−7 M in the 1000-nM treatment. Growth rates were lowest at Cu2+ concentration > 10−9. These results are discussed within the context of the potential roles for exuded copper-complexing ligands.  相似文献

Cultured guinea pig epidermal cells and dermal fibroblasts were chosen as model systems to study possible growth inhibition by cyclic AMP (cAMP)-elevating drugs. The rate of DNA synthesis was used to assay growth rate in control cultures and those treated with agents which increase intracellular cAMP, including dibutyryl cAMP, the phosphodiesterase inhibitors papaverine and theophylline and agents which stimulate adenylate cyclase, iso-proterenol and prostaglandin E2 methyl ester. Treatment for 24 h with dibutyryl cAMP (10−4 to 10−2 M) inhibited cell growth by 50 to 95%, whereas butyrate(10−4M) showed essentially no effect. This inhibition could not be attributed to decreased precursor transport or to drug toxicity. Papaverine (10−6 to 10−4 M) and theophylline (10−4 to 10−3 M) also gave dose-dependent growth inhibition as did isoproterenol and prostaglandinE2methyl ester. Radioautographic analysis of grain density after dibutyryl cAMP treatment and 3H-thymidine incorporation indicated no S-phase inhibition. Abstract It has been hypothesized that vegetation in certain ecosystems inhibits nitrification in soil by producing phenolic compounds that inhibit oxidation of ammonia by nitrifying microorganisms. This hypothesis is based largely on a report that very low concentrations (10−6 M–10−8 M) of several phenolic acids (notably ferulic acid) completely inhibited NO2 production in an aqueous suspension of soil treated with (NH4)2SO4 and a nutrient solution suitable for growth of Nitrosomonas and other autotrophic nitrifying microorganisms. To evaluate this hypothesis, we determined the effects of three ohenolic acids (ferulic acid, caffeic acid, and p -coumaric on nitrite production by representatives of three genera of terrestrial autotrophic nitrifying microorganisms ( Nitrosospira, Nitrosomonas , or Nitrosolubos ) grown on a defined medium containing NH4+. We found that nitrite production by the Nitrososspira was not inhibited by ferulic acid, caffeic acid, or p -coumaric acid at concentrations of 10−6 or 10−5 M and was only slightly inhibited when these acids were at a concentration of 10−4 M. We also found that ferulic acid did not markedly inhibit nitrite production by the three genera of nitrifying microorganisms studied, even when its concentration was as high as 10−3 M. These observations invalidate the hypothesis tested because the phenolic acids studied did not significantly retard ammonia oxidation by autotrophic microorganisms even when their concentration in cultures of these microorganisms greatly exceeded their concentrations in soils.  相似文献

Citrus ( Citrus sinensis L. Osbeck) leaf explants completely abscise within 48 h when exposed to saturating amounts of ethylene at 25°C. When 2,5-norbornadiene was added, 2000 μl 1−1 reduced abscission of explants also exposed to 2 μl 1−1 of ethylene to the level of the control, and 8000 μl 1−1 reduced abscission in explants exposed to 10 μl 1−1 of ethylene to the level of the control, but abscission was complete when 1 000 μl 1−1 of ethylene was used in the presence of 8 000 μl 1−1 of 2,5-norbornadiene. When explants were exposed to 2 μl 1−1 of ethylene, 2000 μl 1−1 of 2,5-norbornadiene prevented abscission if applied up to 10 h after exposure to ethylene. After 18 h, applied 2,5-norbornadiene had little effect on abscission at 48 h. A Lineweaver-Burk plot gave a 1/2 maximum value of 0.12 μl 1−1 for ethylene on abscission, 2,5-Norbornadiene gave competitive kinetics with respect to ethylene with a K1 value of approximately 120 μl 1−1 of 2,5-norbornadiene. The presence of 2,5norbornadiene stimulated ethylene production, which progressively increased as the 2,5-norbornadiene concentration was increased from 250 to 8 000 μl 1−1 2,5-Norbornadiene also suppressed the induction of cellulase and polygalacturonase by ethylene. Together, 2,5-norbornadiene and 2,4-dichlorophenoxyacetic acid were more effective than either alone in reducing abscission. 2,5-Norbornadiene also was effective in preventing the reduction of indole-3-acetic acid transport induced by ethylene.  相似文献

Abstract Anti- Candida activity of 6-amino-2- n -pentylthiobenzothiazole (I), benzylester of (6-amino-2-benzothiazolylthio)acetic acid (II) and of 3-butylthio-(1,2,4-triazolo)-2,3-benzothiazole (III) was followed and compared to that of 2-mercaptobenzothiazole (IV). I and II exhibited good activity against the C. albicans yeast form, similar to IV. They were inhibitorily active against other Candida strains, IC50 values being of the order of 10−5 M, which means better activity than IV. Compound I also exhibited inhibitory activity on germ-tube formation and mycelial growth in the C. albicans strains, while II, III and IV were not active in these tests. III was the least active form of the compounds tested, IC50 values being of the order of 10−4 M. All the compounds tested were highly active on a nystatin-resistant C. albicans mutant, with IC50s of the order of 10−6 M−10−5 M.  相似文献

Abstract. Growth in the presence of retinoids was found to induce erythroid differentiation in Friend murine erythroleukemia (MEL) cells in culture. The program of differentiated functions expressed by retinoid-treated cells was quite similar to that promoted by other inducers of MEL cell differentiation. For example, 70% or more of induced cells synthesized hemoglobin which accumulated to a level of 8 μg–10 μg per 106 cells. The level of acetylcholinesterase activity increased two to five-fold in induced cells, and induction by retinoids, like induction by dimethylsulfoxide (DMSO), promoted the appearance of cell surface lumps or 'blebs'. All-trans retinaldehyde, which promoted maximum hemoglobin and acetylcholinesterase synthesis at a concentration of 5 × 10−7 M, was found to be a more potent inducer than all-trans retinoic acid or retinol, which both showed maximum induction at 1 × 10−5 M. Like differentiation promoted by DMSO, retinoid-induced differentiation was inhibited by 10−7 M dexamethasone.  相似文献

ABSTRACT. The effects of platelet-activating factor (PAF), at doses ranging from 10−6 M to 10−10 M, on cell growth and on cell differentiation of Herpetomonas muscarum muscarum were investigated. Cell differentiation was evaluated by both light and electron microscopy. At the concentrations used, PAF did not interfere with the protozoan growth. However, parasites grown in the presence of PAF (10−6 M) were significantly more differentiated than those grown in the absence of PAF, since the first day of culture. On the first two days of culture, PAF doses ranging from 10−10 M to 10−7 M, did not significantly interfere with the differentiation of these parasites, although after the third day of culture, all PAF doses used significantly increased the protozoan differentiation. Specific PAF receptor antagonists totally abrogated (WEB 2086 and WEB 2170)or significantly decreased (BN 52021) PAF effect on cell differentiation. These findings indicate PAF triggers the process of cell differentiation in Herpetomonas muscarum muscarum and suggest these parasites have receptors for PAF.  相似文献

Abstract: The muscarinic sites in arachnoid and pial vessels were compared by analysis of the binding of quinuclidinyl benzilate (QNB) to membrane preparations. Saturation analysis indicated that the process was saturable, high affinity, and related to protein concentration in both structures. Although the affinities in the two structures [ K D= 0.039 (arachnoid) and 0.097 n M (pial vessels)] were similar, the arachnoid had ∼ 10-fold more binding sites ( B max= 2,100 fmol/mg of protein) than the pial vessels ( B max= 250 fmol/ mg of protein). This difference was found in both bovine and porcine fractions. Pharmacological analysis of [3H]-QNB displacement by muscarinic and nonmuscarinic ligands gave the typical pattern of muscarinic receptors in both structures. Inhibition of binding to pial vessels by the M, antagonist pirenzepine revealed only one low-affinity site ( K i= 7.8 × 10−7 M ), whereas, the arachnoid had a small proportion (21%) of high-affinity sites ( K i= 2.2 × 10−9 M ) associated with low-affinity sites ( K i= 5.50 × 10−7 M ). It is concluded that muscarinic-mediated effects that do not involve the M, subtype are induced in bovine pial vessels by a relatively low concentration of binding sites. The high content of muscarinic binding sites and their diversity in the arachnoid suggest a functional role for muscarinic cholinergic receptors in this structure.  相似文献

Preference responses of zebrafish to 10−3, 10−4 and 10−5M alanine (Ala) were concentration- dependent. Behavioural responses to copper (Cu) and Cu + Ala mixtures were also assessed. Zebrafish avoided 100 and 10 μg Cu l−1, but not 1 μg l−1. Mixtures of 10−3 m Ala+ 100 μg Cu l−1 and 10 4 M Ala + 10 μg Cu 1−1 were avoided as intensely as was Cu alone. Responses to 10−3 M Ala + 10 or 1 μg Cu l−1 and 10 4 M Ala +1 μg Cu l−1 did not differ statistically from controls (no detectable preference or avoidance). These results demonstrate, firstly, that a concentration of a pollutant avoided by itself (10 μg Cu l−1) may not be avoided when encountered with an attractant chemical stimulus (Ala) and may suppress the preference for an attractant stimulus, and secondly, that a concentration of a pollutant not avoided by itself and not considered deleterious (1 μg Cu l−1) suppresses attraction to Ala (an important constituent of prey odours for many fishes).  相似文献

The permeability of internodal cells of Chara australis R. Brown for polyol molecules was determined by using a turgor balance to measure the increase in the osmotic pressure of an internodal cell incubated in artificial pond water containing one of the polyol compounds tested. The permeabilities for ethylene glycol, glycerol, meso -erythritol, xylitol and mannitol were (4. 39 ± 0. 52) × 10−9, (1. 49 ± 0. 40) × 1010, (4. 92 ± 0. 27) × 10−10 (9. 9 ± 3. 4) × 1011 and (7. 6 ± 4. 8) × 10−12 m s−3, respectively. The permeability for glycerol was slightly smaller than that for meso -erythritol, whose molecular weight is larger than that of glycerol in this homologous series: but the reason for this is not clear.  相似文献

Abstract: The effect of pardaxin, a new excitatory neurotoxin, on neurotransmitter release was tested using purely cholinergic synaptosomes of Torpedo marmorata electric organ. Pardaxin elicited the release of acetylcholine with a biphasic dose dependency. At low concentrations (up to 3 × 10−7 M ), the release was calcium-dependent and synaptosomal structure was well preserved as revealed by electron microscopy and measurements of occluded lactate dehydrogenase activity. At concentrations from 3 × 10−7 M to 10−5 M , the pardaxin-induced release of acetylcholine was independent of extracellular calcium, and occluded synaptosomal lactate dehydrogenase activity was lowered, indicating a synaptosomal membrane perturbation. Electron microscopy of 10−6 M pardaxin-treated synaptosomes revealed nerve terminals depleted of synaptic vesicles and containing cisternae. At higher toxin concentrations ( 10−5 M ), there were striking effects on synaptosomal morphology and occluded lactate dehydrogenase activity, suggesting a membrane lytic effect. We conclude that, at low concentrations, this neurotoxin is a promising tool to investigate calcium-dependent mechanisms of neurotransmitter release in the nervous system.  相似文献

Abstract: We investigated the interaction between Pb2+ and protein kinase C (PKC) in the Pb2+-induced release of norepinephrine (NE) from permeabilized adrenal chromaffin cells. Our analysis of endogenous PKC activity in permeabilized cells suggests that Pb2+ interacts with the adrenal enzyme at multiple sites. Pb2+ activates the enzyme through high-affinity ( K A(Pb) = 2.4 × 10−12 M ) interactions and inhibits the enzyme by competitive and noncompetitive interactions with nanomolar-( K i = 7.1 × 10−9 M ) and micromolar- ( K 'i = 2.8 × 10−7 M ) affinity sites, respectively. Activation of PKC by 12- O -tetradecanoylphorbol 13-acetate (TPA) in Ca2+-deficient, Pb2+-containing medium, enhances the Pb2+-induced NE release from permeabilized chromaffin cells by lowering the concentration of Pb2+ required for half-maximal activation of the secretory response from 7.5 × 10−10 to 5.7 × 10−11 M . The PKC inhibitors staurosporine and pseudosubstrate PKC (19–36) abolish the effect of TPA without affecting the Pb2+-induced secretion in the absence of TPA. These results indicate that (a) Pb2+ is a partial agonist of PKC, capable of both activating and inhibiting the enzyme and (b) synergistic activation of PKC by TPA and Pb2+ results in increased sensitivity of exocytosis to Pb2+ but is not obligatory for Pb2+-triggered secretion.  相似文献

Abstract Batch mating experiments were employed to study the kinetics of the conjugal transfer of a TOL plasmid, using the transconjugant strain Pseudomonas aeruginosa PAO 1162 (TOL) as the plasmid donor and Pseudomonas putida PB 2442 and Pseudomonas aeruginosa PAO 1162N as the plasmid recipients. Transfer rates from PAO 1162 (TOL) to PAO 1162N and PB 2442 measured for exponentially grown PAO 1162 (TOL) were 1.81 × 10−14 (standard error (S.E.) 1.25 × 10−15) ml·cell−1min−1 and 3.32 × 10−13 (S.E. 4.42 × 10−14) ml·cell−1min−1, respectively. The instability of the TOL plasmid in PAO 1162 (TOL) was evaluated under conditions that were non-selective for maintenance of the TOL catabolic functions. The measured rates of instability were 6.7 10−6 to 8.3 10−6 min−1, and the loss of the catabolic functions was mainly caused by structural instability of the plasmid.  相似文献
