Abstract. The present investigations were designed to identify proton pumps in seed coats of Phaseolus vulgaris L. Vacated seed-coat halves were exposed to bathing solutions with indicators for proton pump action and the pH changes in the media were measured. Fusicoccin increased the rate of proton extrusion from the seed coats. Orthovanadate and abscisic acid retarded the proton extrusion evoked by fusicoccin. Abolition of the proton extrusion by parachloromercuriphenylsulphonic acid was partially reversed by diethioerythritol. The extrusion was stimulated by high osmolarities (100 mol m−3 sorbitol), potassium ions (100 mol m−3 KCI) and light. Old seed coats reacted more rapidly to fusicoccin treatments than young ones. Proton pumping in seed coats and cotyledons showed differential responses to fusicoccin, K+ and sucrose. In contrast to seed coats, medium acidification by cotyledons was prohibited by addition of sucrose. The significance of proton pumps for photosynthate transfer in vivo is discussed.  相似文献

We investigated H+ fluxes during circadian rhythmic movements of Samanea saman leaflets by monitoring the pH of a weakly buffered medium bathing extensor or flexor motor tissue excised at different times during 51 hours of darkness. Experiments were made in media of two different osmotic potentials: −0.3 megapascal (control medium) and −1.2 megapascals (control medium supplemented with 0.4 molar mannitol or sorbitol). Both extensor and flexor tissue took up H+ from the control medium at all times when the initial pH was 5.5. Rates of uptake by the extensor varied rhythmically in phase with the leaflet movement rhythm, whereas rates for the flexor were similar at all times. Addition of 0.4`molar mannitol (or sorbitol) to the medium magnified the amplitude of the rhythm in H+ uptake and release by extensor tissue and revealed a rhythm with flexor tissue. In the flexor, mannitol promoted H+ release (or reduced H+ uptake) at all times. We propose that mannitol reduces flexor cell turgor, and that low turgor activates the H+ pump. The magnitude and/or direction of pH changes varied with the initial pH of the medium. The pH values after 60 minutes converged to a narrow range, suggesting that cell wall pH might be regulated.  相似文献

Fusicoccin induced germination in dormant and partially afterripened dormant caryopses of Avena fatua L. The rate of caryopsis germination was slower and final percentage germination lower in the highly dormant inbred line M73 at a given concentration of fusicoccin than in the dormant caryopses of line AN265. Gibberellic acid was more effective than fusicoccin in breaking dormancy in both lines. Promotion of germination of dormant caryopses by fusicoccin was inhibited by a 6-day pretreatment with (2-chloroethyl)trimethylammonium chloride.
The basal rate of proton efflux from embryos isolated from dormant and fully afterripened line AN265 caryopses was similar. Addition of fusicoccin increased the rate of proton efflux from the isolated embryos of dormant and afterripened caryopses by nearly 400%. Gibberellic acid had no effect on the rate of proton extrusion. The uptake of 86Rb+ in dormant and afterripened A. fatua embryos was similar after a 2 h uptake period. The addition of fusicoccin to the medium doubled the uptake of 86Rb4 by dormant and afterripened embryos. Gibberelleic acid had no effect on the uptake of 86Rb+ by isolated embryos from either dormant or afterripened caryopses. The experimental results indicate that gibberellic acid is more versatile in its action than fusicoccin, and gibberellic acid may facilitate dormant A. fatua caryopsis germination by stimulating mechanisms other than the direct H+ efflux and K+ uptake at the membrane level.  相似文献

Summary. We have analysed the incorporation of [3H]sucrose and [3H]mannitol in pulvinar motor cells of Robinia pseudoacacia L. during phytochrome-mediated nyctinastic closure. Pairs of leaflets, excised 2 h after the beginning of the photoperiod, were fed with 50 mM [3H]sucrose or [3H]mannitol, irradiated with red (15 min) or far-red (5 min) light and placed in the dark for 2–3 h. Label uptake was measured in whole pulvini by liquid scintillation counting. The distribution of labelling in pulvinar sections was assessed by both light and electron microautoradiography. [3H]Sucrose uptake was twice that of [3H]mannitol incorporation in both red- and far-red-irradiated pulvini. In the autoradiographs, [3H]sucrose and [3H]mannitol labelling was localised in the area from the vascular bundle to the epidermis, mainly in vacuoles, cytoplasm, and cell walls. Extensor and flexor protoplasts displayed a different distribution of [3H]sucrose after red and far-red irradiation. Far-red light drastically reduced the [3H]sucrose incorporation in extensor protoplasts and caused a slight increase in internal flexor protoplasts. After red light treatment, no differences in [3H]sucrose labelling were found between extensor and flexor protoplasts. Our results indicate a phytochrome control of sucrose distribution in cortical motor cells and seem to rule out the possibility of sucrose acting as an osmoticum. The germination of Paulownia tomentosa Steud. (Empress tree) seeds can be induced either by red light (R) or by exogenously applied gibberellic acid (GA3). The R induced germination is completely suppressed by far red irradiation, abscisic acid (ABA) or by growth retardants such as ancymidol, tetcyclacis and paclobutrazol, though not by AMO 1618 or chloro-choline chloride (CCC). The inhibition caused by far red light (FR) and growth retardants can be overcome by GA3, while the ABA-induced inhibition can be reversed by fusicoccin (FC), which does not reverse the inhibition caused by FR or growth retardants. It is noteworthy that the germination of light insensitive wheat, corn, alfalfa and mung bean seeds is not inhibited by growth retardants.  相似文献

The involvement of Ca(2+) on phytochrome-controlled nyctinastic closure in Albizzia lophantha has been studied by testing the effect of the calcium ionophore 6S-[6alpha(2S(*),3S(*)),8beta(R(*)),9beta,11alpha]-5- methyl-amino)-2-[[3,9,11-trimethyl-8-[-1-methyl-2-oxo-2-(1H-pyrrol-2-yl) ethyl]-1,7-dioxaspiro[5.5]-undec-2yl] methyl]-4-benzoxazolecarboxylic acid (A23187) and the intracellular calcium antagonist 8-(diethylamino)octyl 3,4,5-trimethoxybenzoate hydrochloride (TMB-8). An external supply of Ca(2+) or calcium ionophore A23187 to the Albizzia leaflets emulates the effect of red light irradiation and counteracts the inhibitory effect of far red light. The intracellular calcium antagonist TMB-8 supplied to Albizzia leaflets inhibits the effect of red light, but had no effect on far red irradiated plants. This suggests a dependence between phytochrome action and intracellular free Ca(2+). We suggest that calcium acts as a phytochrome messenger on control of ion fluxes that drive turgor changes in pulvinular motor cells.  相似文献

Summary Effects of fusicoccin alone and together with abscisic acid were observed on the stomatal complex of Commelina communis. The experimental material consisted of isolated epidermal strips incubated in a medium containing the ions required for stomatal opening. Fusicoccin stimulated opening and this was accompanied by potassium entry into the guard cells, and hydrolysis of the starch in their chloroplasts. Abscisic acid alone inhibited potassium entry and starch hydrolysis, but these effects could be almost entirely overcome by fusicoccin.Attempts were made to measure the solute potential of the guard cells under the various treatments. Abscisic acid clearly increased their solute potential, but no absolute measurements could be made in the presence of fusicoccin owing to a failure of plasmolysis even with mannitol solutions of solute potential as low as —35 bars. Experiments using isotopically labelled mannitol indicated a massive uptake into the epidermis in the presence of fusicoccin.The mechanism of stimulation of stomatal opening by fusicoccin probably depends in part on a stimulation of the normal processes associated with opening in the guard cells, but may also involve release of pressure due to destruction of the surrounding cells. The effectiveness of this toxin under natural conditions may depend on its ability to counteract effects of abscisic acid, the stress hormone that induces stomatal closure.  相似文献

The movement patterns of Albizzia julibrissin leaflets transferred from light to darkness differ for leaflets of different age: the older the leaflet, the slower and less completely it closes. Water stress, which enhances ethylene synthesis in other plants, and 1-aminocyclopropane-1-carboxylic acid (ACC), precursor of ethylene, both mimic the effect of aging by reducing the rate and extent of dark-induced closure. Brief far-red compared to red irradiation before darkness does not appear to alter the closure of young leaflets, but far-red preirradiation retards the closure of middle-aged and old leaflets floating on water, and middle-aged leaflets treated with ACC. A change in some membrane property and/or cell wall lignification are suggested as possible explanations for the alteration of leaflet movement.  相似文献

Micromolar concentrations of potassium ferricyanide inhibit light-induced stomatal opening. The extent of the inhibition is dependent on the presence of carbon dioxide and the concentration of potassium ferricyanide needed to obtain 50% inhibition of stomatal opening is 40-fold higher in CO2-free air than in normal air. The fungal toxin, fusicoccin (1 μ M ), overcame the ferricyanide inhibition of stomatal opening indicating that the electron acceptor may interact more or less directly with the activity of the plasma membrane H+-ATPase. Although potassium ferricyanide strongly inhibited stomatal opening, it had only minor effects on stomatal maintaining or stomatal closure due to darkness or ABA.  相似文献

Effects of CO2 on stomatal movements of Commelina communis L. were studied with plants, epidermal strips and guard cell protoplasts. With plants, the stomatal response induced by a blue light pulse was studied for different ambient CO2 concentration ranging from CO2-deprived air to 100 Pa in darkness or under red light. It was observed that the blue light response could be obtained not only under a red light background but also in darkness and CO2-free air, the two responses being quite similar.
With epidermal strips, the effect of CO2 on ferricyanide reductase activity at the guard cell plasmalemma was studied by transmission electron microscopy. In the presence of ferric ions, reduced ferricyanide gives an electron dense precipitate of Prussian Blue. In darkness and air, no precipitate was observed. In darkness and CO2-free air as well as under light and normal air, a precipitate was found along the plasmalemma of the guard cells, indicating a ferricyanide reductase activity. With guard cell protoplasts suspended in a medium either in equilibrium with air or in a CO2-free medium the H+ extrusion induced by a blue light pulse added to a red light background was measured. A low CO2 content was obtained by adding photosynthetic algae to the suspension of guard cell protoplasts. In a CO2-free medium the rate of H+ extrusion was enhanced.
The results are discussed on the basis of a possible competition for reducing power between CO2 fixation and a putative blue light dependent redox chain located on the plasma membrane.  相似文献

Fusicoccin was shown to stimulate the ATP-driven, intravesicular acidification of liposomes reconstituted with crude fusicoccin receptors and the H+-translocating ATPase, both solubilized from maize (Zea mays L.) plasma membrane. The present paper reports optimal conditions for dual reconstitution and fusicoccin activation as well as the biochemical characterization of the effect of fusicoccin on this system. Fusicoccin stimulation of proton pumping was dependent on pH and fusicoccin concentration. Its specificity was demonstrated by the positive effect of two cotylenins that have a high affinity for fusicoccin receptors and by the negative response to 7,9-epideacetylfusicoccin, an inactive fusicoccin derivative. Kinetic measurements at different ATP concentrations showed that fusicoccin increases the Vmax of the enzyme. Fusicoccin stimulation of maize H+-ATPase was also maintained when receptors from maize were substituted by those from spinach (Spinacia oleracea L.).  相似文献

Changes in the contents of ethanol, lactate and malate were determined at different activities of the plasma membrane H+ pump [in the presence and absence of fusicoccin (FC)] and at different O2 availability in cultured cells of Acer pseudoplatanus L. FC induced acidification of the medium under all tested conditions of O2 availability. At low O2 concentrations both ethanolic and lactic fermentations occurred, and FC markedly stimulated lactate production but had no effect on ethanol production. There was also a small, stimulating effect of FC on malate production. At high O2 concentrations no ethanol production was observed and lactate production was reduced. Under these conditions the stimulating effect of FC on lactate production decreased, while that on malate production increased. FC-induced synthesis of lactate and malate is interpreted as depending on the activation of lactate dehydrogenase (EC 1.1.1.27) and phosphoenolpyruvate carboxylase (EC 4.1.1.31) (alkaline pH optima), respectively, due to the alkalinization of the cytoplasmic pH resulting from the stimulation of the H+ pump by FC. These results suggest that the balance between the two pH stat systems depends on the availability of O2.  相似文献

We studied the influence of lysophosphatidylethanolamine (LPE) on the pattern and rate of ethylene production and respiration of tomato ( Lycopersicon esculentum cv. H7155) leaflets and fruit. Leaflets that had been senescing on the plant showed a climacteric-like rise in ethylene production but not in respiration rate which decreased continuously with leaf age. Detached leaflets had a climacteric-like pattern in respiration whether they were incubated in complete darkness or in light. Detached leaflets incubated in the dark had higher rates of ethylene production and CO2 evolution than did light-incubated leaves. There was no change in the pattern of ethylene production or CO2 evolution as a result of LPE treatment. However, LPE-treated attached and detached leaflets had consistently lower rates of CO2 evolution. The reduction in CO2 evolution by LPE was most pronounced at the climacteric-like peak of the detached leaves. LPE-treated leaflets had a higher chlorophyll content and fresh weight and lower electrolyte leakage than the control. LPE-treated fruits had lower rates of ethylene and CO2 production than did the control. LPE-treated fruits also had higher pericarp firmness and lower electrolyte leakage than the control. The results of the present study provide evidence that LPE is able to retard senescence of attached leaves and detached leaves and tomato fruits. Several recent studies suggest that lysolipids can act in a specific manner as metabolic regulators. Our results suggest a specific role of lysolipid LPE in aging and senescence  相似文献

Samanea leaflets usually open in white light and fold together when darkened, but also open and dose with a circadian rhythm during prolonged darkness. Leaflet movement results from differential changes in the turgor and shape of motor cells on opposite sides of the pulvinus; extensor cells expand during opening and shrink during closure, while flexor cells shrink during opening and expand during closure but change shape more than size. Potassium in both open and closed pulvini is about 0.4 N. Flame photometric and electron microprobe analyses reveal that rhythmic and light-regulated postassium flux is the basis for pulvinar turgor movements. Rhythmic potassium flux during darkness in motor cells in the extensor region involves alternating predominance of inwardly directed ion pumps and leakage outward through diffusion channels, each lasting ca 12 h. White light affects the system by activating outwardly directed K+ pumps in motor cells in the flexor region.  相似文献

Fusicoccin receptors are proteins that are widespread in plasmamembrane of higher plants. A critical duration of darkness must be exceeded for the photoperiodic induction of flowering in short-day plants. This requires detection of the light/dark transition at dusk and the coupling of this information to a time-measuring system.
Lowering the Pfr/Ptot, ratio photochemically at the end of the day did not accelerate the onset of dark timing in Pharbitis nil Choisy cv. Violet. Time-measurement was initiated when, with no change in spectral quality, the irradiance fell below a threshold value. Thus, if the light/dark transition at dusk is sensed by a reduction in Pfr, this reduction can be achieved as rapidly through thermal reactions as through photochemical ones. When given at hourly intervals during a 6-h extension of a 24-h main light period in white light, pulses of red light were as effective as continuous red light in delaying the onset of timing; pulses every 2 or 3 h were less effective. The effectiveness of intermittent red light indicates that phytochrome is the photoreceptor and the requirement for frequent exposures suggests that Pfr is lost rapidly in the dark. However, the red light pulses could not be reversed by far-red light, which argues against this hypothesis. An alternative explanation is that the perception of light as being continuous occurs only when "new" Pfr is regenerated sufficiently frequently.
The nature of the coupling of the dusk signal to the time-measuring system is discussed and it is suggested that the effect of each red light pulse is to delay the phase of the photoperiodic rhythm by 1–3 h.  相似文献

Chlorophyll loss in the leaves of cut flowering branches of Alstroemeria pelegrina L. cv. Stajello, placed in water in darkness at 20°, was inhibited by irradiation with red light and by the inclusion of gibberellic acid (GA3) in the water. The effects of red light were abolished when it was followed by far-red light. Effects of GA3 and red light were additive over a range of GA3 concentrations (0. 01–1 μ M ). Chlorophyll breakdown was increased by the inclusion of AMO-1618, ancymidol, or tetcyclasis in the water. The effect of these inhibitors of gibberellin synthesis was fully reversed by GA3. The inhibition of chlorophyll breakdown by red light was absent when AMO-1618, ancymidol or tetcyclasis were included in the water. The results indicate that leaf yellowing is controlled by endogenous gibberellins and that the effect of phytochrome is mediated by gibberellin synthesis.  相似文献

The involvement of phytochrome in stomatal movement in Commelina communis L. is indicated by the following observations: 1) Short irradiation with red or blue light causes opening, of isolated stomata and swelling of guard cell protoplasts. This is reversed by subsequent far red irradiation. 2) In a similar way, stomatal response to prolonged irradiation with red or blue light is decreased by concomitant far red irradiation. 3) Pretreatment with filipin, which interferes with phytochrome binding to membranes, decreases stomatal opening in red and blue light. The stomatal responses to blue and red light are modified by DCMU, N2, CO2-enriched atmosphere, and CO2-free air, which are known to affect, among other processes, chlorophyll fluorescence. Increased chlorophyll fluorescence by DCMU, N2 and CO2-enriched atmosphere enhanced stomatal opening in blue light and inhibited it in red light. CO2-free air, which decreases chlorophyll fluorescence, had the opposite effect.  相似文献

Auxin (IAA, 5 × 10−5 M ) partially prevents the increase in the rate of phosphate uptake during ageing of potato tuber discs ( Solatium tuberosum L. cv. Bintje), whereas fusicoccin (FC, 10−5 M) stimulates it. After the development of enhanced phosphate transport capacity, the response to fusicoccin is greater than with fresh discs. Complementary experiments on K+ (86Rb) absorption show that FC also slightly enhances the rate of K+ uptake, while IAA has no much effect. It is suggested that IAA acts specifically on the development of a mechanism which occurs during the ageing period, while FC action may be more directly linked to the system of phosphate transport itself.  相似文献
