The effects of prolonged cycling on neuromuscular parameters were studied in nine endurance-trained subjects during a 5-h exercise sustained at 55% of the maximal aerobic power. Torque during maximal voluntary contraction (MVC) of the quadriceps muscle decreased progressively throughout the exercise (P < 0.01) and was 18% less at the end of exercise compared with the preexercise value. Peak twitch torque, contraction time, and total area of mechanical response decreased significantly (P < 0.05) after the first hour of exercise. In contrast, changes in M-wave characteristics were significant only after the fourth hour of the exercise. Significant reductions (P < 0.05) in electromyographic activity normalized to the M wave occurred after the first hour for the vastus lateralis muscle but only at the end of the exercise for the vastus medialis muscle. Muscle activation level, assessed by the twitch interpolation technique, decreased by 8% (P < 0.05) at the end of the exercise. The results suggest that the time course is such that the contractile properties are significantly altered after the first hour, whereas excitability and central drive are more impaired toward the latter stages of the 5-h cycling exercise.  相似文献

The organophosphate cholinesterase inhibitor paraoxon produces a dose-dependent necrosis in rat skeletal muscle fibers after a single administration. The pathology, which is initiated at the motor end-plate region, is evident as early as 30 minutes after paraoxon administration and is characterized by dilated mitochondria, expanded sarcoplasmic reticulum, fused and widened subsynaptic folds, and coated cleft vesicles. By 24 hours, a generalized breakdown of muscle fiber architecture is evident with an accompanying infiltration of phagocytes. Electrophysiological studies have shown that paraoxon increases neurotransmitter release and causes spontaneous and impulse-related antidromic nerve activity, both of which can be reduced significantly by reactivation of inhibited acetylcholinesterase (AChE) with pyridine-2-aldoxime methiodide. The severity of the myopathy has been found to be positively correlated to the degree and duration of AChE inhibition. It appears that 2 hours of inhibition, with a critical loss in activity, viz., 85%, is necessary to initiate severe muscle fiber necrosis. Prior nerve transection prevents myopathic development and current data support the hypothesis that the induction of skeletal muscle fiber necrosis is triggered by inhibition of a neurally regulated fraction of AChE.  相似文献

The time course of alteration in neuromuscular function of the knee extensor muscles was characterized during a prolonged intermittent exercise. Maximal voluntary contraction (MVC) and surface EMG activity of both vastii were measured during brief interruptions before (T0), during (30, 60, 90, 120, 150 and 180 min: T30, T60, T90, T120, T150, T180) and 30 min after (T+30) a 3 h tennis match in 12 trained players. M-wave and twitch contractile properties were analyzed following single stimuli. Short tetani at 20 Hz and 80 Hz were also applied to six subjects at T0 and T180. Significant reductions in MVC (P < 0.05; −9%) and electromyographic activity normalized to the M wave for both vastii (P < 0.01) occurred with fatigue at T180. No significant changes in M-wave duration and amplitude nor in twitch contractile properties were observed. The ratio between the torques evoked by 20 Hz and 80 Hz stimulation declined significantly (P < 0.001; −12%) after exercise. Central activation failure and alterations in excitation–contraction coupling are probable mechanisms contributing to the moderate impairment of the neuromuscular function during prolonged tennis playing.  相似文献

The aim of the study was to examine alterations in contractile and neural processes in response to an isometric fatiguing contraction performed with EMG feedback (constant-EMG task) when exerting 40% of maximal voluntary contraction (MVC) torque with the knee extensor muscles. A task with a torque feedback (constant-torque task) set at a similar intensity served as a reference task. Thirteen men (26+/-5 yr) attended two experimental sessions that were randomized across days. Endurance time was greater for the constant-EMG task compared with the constant-torque task (230+/-156 s vs. 101+/-32s, P<0.01). Average EMG activity for the knee extensor muscles increased from 33.5+/-4.5% to 54.7+/-21.7% MVC EMG during the constant-torque task (P<0.001), whereas the torque exerted during the constant-EMG task decreased from 42.8+/-3.0% to 17.9+/-5.6% MVC torque (P<0.001). Comparable reductions in knee extensors MVC (-15.7+/-8.7% for the constant-torque task vs. -17.5+/-9.8% for the constant-EMG task, P>0.05) and voluntary activation level were observed at exhaustion. In contrast, excitation-contraction coupling process, assessed with an electrically evoked twitch and doublet, was altered significantly more at the end of the constant-EMG task despite the absence of M-wave changes for both tasks. Present results suggest that prolonged contractions using EMG biofeedback should be used cautiously in rehabilitation programs.  相似文献

The purpose of this study was to compare the cortisol responses from a regular season game and a typical practice session in female National Collegiate Athletic Association Division I collegiate soccer players. Eighteen players were assigned to 2 groups, 10 starters and 8 nonstarters, depending on their playing time. Salivary cortisol concentration, as well as competitive sport anxiety (somatic and cognitive anxiety, self-confidence), was monitored before and after 1 regular season game and 1 typical practice session. Although salivary cortisol levels increased postgame for both starters (+250%) and nonstarters (+140%), they increased to a greater extent for the starters. Practice salivary cortisol did not significantly change (p > 0.05). Cognitive and somatic anxiety was greater pre- and postgame when compared with the pre- and postpractice scores, respectively. These data clearly demonstrate the psychological and physiological differences between soccer competition and practice in collegiate women. It appears that both physiological and psychological variables combine to contribute to the large stress hormone response to an actual competitive game.  相似文献

Evolutionary game theory is a basis of replicator systems and has applications ranging from animal behavior and human language to ecosystems and other hierarchical network systems. Most studies in evolutionary game dynamics have focused on a single game, but, in many situations, we see that many games are played simultaneously. We construct a replicator equation with plural games by assuming that a reward of a player is a simple summation of the reward of each game. Even if the numbers of the strategies of the games are different, its dynamics can be described in one replicator equation. We here show that when players play several games at the same time, the fate of a single game cannot be determined without knowing the structures of the whole other games. The most absorbing fact is that even if a single game has a ESS (evolutionary stable strategy), the relative frequencies of strategies in the game does not always converge to the ESS point when other games are played simultaneously.  相似文献

ABSTRACT: BACKGROUND: Apoptosis is an essential property of all higher organisms that involves extremely complex signaling pathways. Mathematical modeling provides a rigorous integrative approach for analyzing and understanding such intricate biological systems. RESULTS: Here, we constructed a large-scale, literature-based model of apoptosis pathways responding to an external stimulus, cisplatin. Our model includes the key elements of three apoptotic pathways induced by cisplatin: death receptor-mediated, mitochondrial, and endoplasmic reticulum-stress pathways. We showed that cisplatin-induced apoptosis had dose- and time-dependent characteristics, and the level of apoptosis was saturated at higher concentrations of cisplatin. Simulated results demonstrated that the effect of the mitochondrial pathway on apoptosis was the strongest of the three pathways. The cross-talk effect among pathways accounted for approximately 25% of the total apoptosis level. CONCLUSIONS: Using this model, we revealed a novel mechanism by which cisplatin induces dose-dependent cell death. Our finding that the level of apoptosis was affected by not only cisplatin concentration, but also by cross talk among pathways provides in silico evidence for a functional impact of system-level characteristics of signaling pathways on apoptosis.  相似文献

Diaphragmatic contractility was assessed in spontaneously breathing ketamine-anesthetized rabbits by measuring the strength of diaphragmatic contraction in response to bilateral supramaximal phrenic nerve stimulation at frequencies between 10 and 100 Hz. During 10-180 min of inspiratory resistive loading, contractility decreased by approximately 40%, and hypoxemia and both respiratory and lactic acidosis developed. After 10 min of recovery, both the response to high-frequency stimulation (100 Hz) and the arterial PO2 and PCO2 returned to base-line levels, whereas metabolic acidosis and reduced response to low-frequency stimulation (10-20 Hz) persisted. Similar levels of hypoxemia and respiratory acidosis in the absence of inspiratory resistive loading did not alter diaphragmatic contractility. We conclude that in anesthetized rabbits excessive inspiratory resistive loading results in partially reversible diaphragm fatigue of the high- and low-frequency types, accompanied by hypoventilation and lactic acidosis.  相似文献

A new generalized mathematical model for recombinant bacteria which includes inducer effects on cell growth and foreign protein production is developed. The model equation set was applied to a host-vector system, Escherichi coli D1210 and plasmid pSD8. Batch experiments were designed and performed in shake flasks to verify the model. A parameter estimation method was developed and proven to be efficient. Although simple, the model can effectively describe the dynamics of the production of foreign protein in recombinant bacteria and can be used for optimization and control studies to maximize foreign protein production.  相似文献

ABSTRACT

Exposure to light at night results in disruption of endogenous circadian rhythmicity and/or suppression of pineal melatonin, which can consequently lead to acute or chronic adverse health problems. In the present study, we investigated whether exposure to very dim light or very bright light for a short duration influences melatonin suppression, subjective sleepiness, and performance during exposure to constant moderately bright light. Twenty-four healthy male university students were divided into two experimental groups: Half of them (mean age: 20.0 ± 0.9 years) participated in an experiment for short-duration (10 min) light conditions of medium intensity light (430 lx, medium breaks) vs. very dim light (< 1 lx, dim breaks) and the other half (mean age: 21.3 ± 2.5 years) participated in an experiment for short-duration light conditions of medium intensity light (430 lx, medium breaks) vs. very bright light (4700 lx, bright breaks). Each simulated night shift consisting of 5 sets (each including 50-minute night work and 10-minute break) was performed from 01:00 to 06:00 h. The subjects were exposed to medium intensity light (550 lx) during the night work. Each 10-minute break was conducted every hour from 02:00 to 06:00 h. Salivary melatonin concentrations were measured, subjective sleepiness was assessed, the psychomotor vigilance task was performed at hourly intervals from 21:00 h until the end of the experiment. Compared to melatonin suppression between 04:00 and 06:00 h in the condition of medium breaks, the condition of dim breaks significantly promoted melatonin suppression and the condition of bright breaks significantly diminished melatonin suppression. However, there was no remarkable effect of either dim breaks or bright breaks on subjective sleepiness and performance of the psychomotor vigilance task. Our findings suggest that periodic exposure to light for short durations during exposure to a constant light environment affects the sensitivity of pineal melatonin to constant light depending on the difference between light intensities in the two light conditions (i.e., short light exposure vs. constant light exposure). Also, our findings indicate that exposure to light of various intensities at night could be a factor influencing the light-induced melatonin suppression in real night work settings.  相似文献

Critical power (CP) is a theoretical workload representative of an athlete's maximal sustainable pace. Recent research has validated a 3-minute all-out test on a cycle ergometer for determining CP; however, few studies have investigated the sustainability of CP using this test. The purpose of this study was to determine the sustainability of CP established during the 3-minute test and the determinants of sustainability. A group of elite cyclists (N = 21) performed a VO2max test, 3-minute all-out test, and a time to exhaustion (TTE) trial at CP on 3 different days separated by at least 24 hours. Expired gases were collected during all trials and analyzed for VO2 and VCO2. Heart rate was measured by telemetry. Multiple regression was used to determine predictors of sustainability with significance predetermined at p < 0.05. VO2max was measured at 58.9 ± 5.6 ml·kg(-1)·min(-1), ventilation breakpoint at 44.9 ± 5.7 ml·kg(-1)·min(-1) (75% VO2max), and maximum heart rate at 179 ± 10 b·min(-1). Peak power (PP) in the 3-minute all-out test was measured at 738 ± 170 W, and CP was determined at 305 ± 32 W or 79% of VO2max. The VO2 at CP was 55.4 ± 6.9 ml·kg(-1)·min(-1), representing 94% of measured VO2max. The mean TTE at CP was 14.79 ± 8.38 minutes. The difference score of PP - CP significantly predicted TTE (r = 0.65, p < 0.05). No other measured variables contributed to this prediction. Based on sustainability, these data suggest that the 3-minute all-out test may overestimate CP in elite cyclists, which could lead to overtraining if CP determined with this test is used to identify training intensities.  相似文献

We develop a suite of models with varying complexity to predict elk movement behavior during the winter on the Northern Range of Yellowstone National Park (YNP). The models range from a simple representation of optimal patch choice to a dynamic game, and we show how the underlying theory in each is related by the presence or absence of state- and frequency-dependence. We compare predictions from each of the models for three variables that are of basic and applied interest: elk survival, aggregation, and use of habitat outside YNP. Our results suggest that despite low overall forage depletion in the winter, frequency-dependence is crucial to the predictions for elk movement and distribution. Furthermore, frequency-dependence interacts with mass-dependence in the predicted outcome of elk decision-making. We use these results to show how models that treat single movement decisions in isolation from the seasonal sequence of decisions are insufficient to capture landscape scale behavior.  相似文献

Elastic tubes are devices relatively inexpensive, easy to use, portable and safe, which are claimed to provide advantages with respect to training with free weights or weight machine. The aim of this work was to assess neuromuscular modification induced by the use of two different muscle contraction modalities (elastic versus weight resistances) until exhaustion. Fourteen healthy physically active male students (age: 28 ± 6 years; body mass 72.1 ± 11.0 kg; height: 173.5 ± 6.9 cm) were recruited. They were requested to reach exhaustion with dynamic contraction at 70% of their own One Repetition Maximal (1RM) using, in two different days and in random order, Thera Band® tubes or traditional weight plates on an arm machine designed and built for those tasks. Before and after such contractions a standardized fatiguing isometric test (3 Maximal Voluntary Contractions, MVC plus 60 s at 60% MVC) was requested to assess differences induced by the exhaustion sessions. During fatiguing tests surface EMG signals were recorded from biceps brachii muscle with linear arrays of eight electrodes (silver bars 10 mm apart, 5 mm long, and 1 mm diameter) in single differential configuration. Initial values and rates of change of Average Rectified Value (ARV), MeaN power Frequency (MNF) and muscle fiber Conduction Velocity (CV) were calculated to compare the effects of the two contraction modalities. No differences were found between “elastic session” and “weight session” PRE MVCs (31.9 ± 8.8 and 29.9 ± 8.3 nm, respectively) and endurance times (28 ± 6 and 26 ± 7 s, respectively). The same was observed for POST values. During the post-contraction isometric fatigue test, the only parameters influenced by the contraction modality were the initial CV and the rate of change in CV which were 12% and 37% greater (p < 0.05) respectively after contractions performed with elastic resistance compared to free weights. These findings confirm others for which contractions done with the use of elastic band seem to require greater muscle activation; moreover, they highlight a sort of “muscle conditioning” after that specific contraction modality which requires the use of faster motor units. It is thus possible to consider the practical use of such elastic devices to increase the neuromuscular activation, for instance in specific rehabilitation settings where slow movements with minimal risk of injury are requested (post surgery rehabilitation, physical activity with elderly people or children).  相似文献

We generalize chaos game representation (CGR) to higher dimensional spaces while maintaining its bijection, keeping such method sufficiently representative and mathematically rigorous compare to previous attempts. We first state and prove the asymptotic property of CGR and our generalized chaos game representation (GCGR) method. The prediction follows that the dissimilarity of sequences which possess identical subsequences but distinct positions would be lowered exponentially by the length of the identical subsequence; this effect was taking place unbeknownst to researchers. By shining a spotlight on it now, we show the effect fundamentally supports (G)CGR as a similarity measure or feature extraction technique. We develop two feature extraction techniques: GCGR-Centroid and GCGR-Variance. We use the GCGR-Centroid to analyze the similarity between protein sequences by using the datasets 9 ND5, 24 TF and 50 beta-globin proteins. We obtain consistent results compared with previous studies which proves the significance thereof. Finally, by utilizing support vector machines, we train the anticancer peptide prediction model by using both GCGR-Centroid and GCGR-Variance, and achieve a significantly higher prediction performance by employing the 3 well-studied anticancer peptide datasets.

Geldanamycin (GM), a benzoquinone ansamycin antibiotic, is a natural product inhibitor of Hsp90 with potent and broad anti-cancer properties. Because of its adverse effects on liver, its less toxic derivatives 17-(allylamino)-17-demethoxygeldanamycin (17-AAG) and 17-(dimethylaminoethylamino)-17-demethoxygeldanamycin (17-DMAG) are currently being evaluated for the treatment of cancer. Previously, it has been demonstrated that the redox cycling of GM by NADPH-cytochrome P450 reductase leads to the formation of the GM semiquinone and superoxide radicals, the latter being identified using spin-trapping. We hypothesized that the different hepatotoxicity induced by GM, 17-AAG and 17-DMAG reflects the redox active properties of the quinone moiety and possibly the extent of superoxide formation, which may stimulate cellular oxidative injury. Our data demonstrate that superoxide can be efficiently trapped during the reduction of GM, 17-AAG and 17-DMAG by NADPH-cytochrome P450 reductase, and that superoxide formation rate followed the order 17-DMAG > 17-AAG > GM. In the absence of superoxide scavengers, the rate of NADPH oxidation followed the order 17-DMAG > GM > 17-AAG. The half-wave one-electron reduction potentials (E1/2) of GM, 17-AAG and 17-DMAG in DMSO have been determined to be -0.37, -0.13 and -0.015 V (vs. Ag/AgCl), respectively. If the same order of E1/2 follows in neutral aqueous media, thermodynamic considerations imply that 17-DMAG is more readily reduced by the P450 reductase as well as by superoxide. The order of the drug cytotoxicity toward rat primary hepatocytes, as determined by their effect on cell viability and on intracellular oxidant level, was opposite to the order of E1/2 of the respective quinone/semiquinone couples. These results suggest that hepatotoxicity exhibited by the Hsp90 inhibitors belonging to benzoquinone ansamycins could be attributed to superoxide. The apparent discrepancy between the order of toxicity and the orders of superoxide formation rate, which is correlated with E1/2, is discussed.  相似文献

We previously reported that radicicol (Rad) and geldanamycin (Geld), heat shock protein 90 (Hsp90) inhibitors, potentiate neurite growth of cultured sensory neurons from chick embryo. We now show that the antibiotics induce neurite growth in NG108-15 cells. Treatment of the cells with these drugs caused transient decrease in protein levels of Raf1, ERK1/2, phosphorylated ERK1/2, Akt1, and CDK4. The neurite growth of NG108-15 induced by the inhibitors was blocked by actynomycin D, but the neurite growth stimulated by dbcAMP in the cells was not affected. The neurite growth could be due to a change in the synthesis of some specific protein(s) and is speculated to be due to the transient downregulation of particular-signaling molecules stabilized by Hsp90.  相似文献

The purpose of this study was to investigate the effect of time of day and different modalities of recovery (active vs. passive recovery) after intermittent exercise in young soccer players. In randomized order, 16 boys participated in the study, divided into two groups: passive recovery (PRG, n?=?8) and active recovery (ARG, n?=?8). Both groups performed tests at 07:00 and 17:00 h. The results showed that performance in the Sargent jump test (SJT), 10-m sprint, and agility were higher in the evening (17:00 h), around the presumed peak of body temperature.
