The characteristics of the Na+-independent high-affinity binding of [3H]GABA to various types of crude synaptic membranes (CSM) prepared from rat brain cortex were studied. In freshly prepared CSM the content of GABA was so high that the high-affinity [3H]GABA binding could not be determined. In contrast when the frozen-thawed CSM were incubated at 37° for 30 min with or without Triton X-100 or phospholipase C and then washed repeatedly, there was a virtual disappearance of GABA from the supernatant extracts and the binding constants of [3H]GABA to CSM could be determined. Two apparent populations of [3H]GABA binding sites, one with a low- and the other with a high-affinity constant, were detected. The ratio of the number of high- to low-affinity binding sites varies with the method used to prepare the membranes. The lowest value of this ratio was observed with membranes incubated at 37° for 30 min. However, when frozen-thawed CSM were treated with 0.05% Triton X-100 repeatedly, the ratio of the number of high- to low-affinity binding sites increased progressively. This increase in ratio is due to a selective increase in the number of the high-affinity sites without significant changes in the number of the low-affinity sites. The extent of the increase in the number of sites that bind [3H]GABA with high affinity after repeated Triton X-100 treatments was paralleled by a decrease of an endogenous protein which inhibits GABA binding. The reapplication of this endogenous material to membranes repeatedly treated with Triton X-100 reduces the number of high-affinity binding sites for [3H]GABA to values similar to those measured in membranes that were not treated with Triton X-100. The inhibitory preparation extracted from CSM incubated with Triton X-100 was shown to be free of GABA or phospholipids. The gel filtration chromatography reveals the presence of two molecular forms of the inhibitor; of these, the high-molecular-weight material fails to bind GABA, whereas the low-molecular-weight material appears to bind GABA. The high-molecular-weight endogenous inhibitor has been termed GABA modulin.  相似文献

Abstract: The binding of radioactive piperidine-4-sulphonic acid ([3H]P4S) to thoroughly washed, frozen, and thawed membranes isolated from cow and rat brains has been studied. Quantitative computer analysis of the binding curves for four regions of bovine brain revealed the general presence of two binding sites. In these brain regions less satisfactory computer fits were obtained for receptor models showing one or three binding sites or negative cooperativity. With the use of Tris-citrate buffer at 0°C the two affinity classes for P4S in bovine cortex membranes revealed the following binding parameters: KD= 17 ± 7 nM (Bmax= 0.15 ± 0.07 pmol/mg protein) and KD= 237 ± 100 nM (Bmax= 0.80 ± 0.20 pmol/mg protein). Heterogeneity was also observed for association and dissociation rates of [3H]P4S. The slow binding component (kon= 5.6 × 107 or 8.8 × 107 M-1 min-1, kOff= 0.83 min-1, and KD= 14.7 or 9.4 nM, determined by two different methods in phosphate buffer containing potassium chloride) corresponds to the high-affinity component of the equilibrium binding curve (KD= 11 nM, Bmax= 0.12 pmol/mg protein in the same buffer system). The association and dissociation rates for the subpopulation of rapidly dissociating sites, apparently corresponding to the low-affinity sites, were too rapid to be measured accurately. The binding of [3H]P4S appears to involve the same two populations of sites with Bmax values similar to those for [3H]GABA binding to the same tissue, although the kinetic parameters for the two ligands are somewhat different. Furthermore, comparative studies on the inhibition of [3H]P4S and [3H]GABA binding by various GABA analogues, strongly suggest that P4S binds to the GABA receptors. The different effects of P4S and GABA on benzodiazepine binding are discussed.  相似文献

We have studied the developmental sequence of the GABA system in the rabbit retina using an in vitro binding assay to monitor developmental changes in the post-synaptic receptor. A variety of tissue treatments including perchlorate and Triton X-100 were employed to optimize binding and remove endogenous factors which inhibit binding. Pre-treatment of the tissue with 0.05% Triton X-100 revealed high affinity binding for [3H]GABA which increased in a sigmoidal fashion with the post-natal age of the animal. A constant level of binding, at about 16% of adult levels, was noted until day 8, at which time a rapid increase occurred. At 16 days post-natal, the amount of specific binding reached a plateau near adult levels. Kinetic analysis of the GABA receptor showed an increase in the number of receptors (Bmax) with little or no change in the apparent affinity (KD). Our results suggest that the onset of post-synaptic receptor activity is delayed approximately 1 to 2 days, relative to the pre-synaptic components, and the period of rapid increase in GABA receptor binding coincides with the period of maximum increase in retinal synaptic density.  相似文献

The development of the GABA system in the rabbit retina was studied. The number of high- and low-affinity GABA receptor binding sites increased in a sigmoidal manner, with the curve for the low-affinity sites lagging 2–3 days behind that for the high-affinity sites. TheK D for both high-(17.5 nM) and low-affinity (138.0 nM) sites remained constant during development. Treatment of isolated eyecups with the uptake blocker nipecotic acid resulted in an increase in theB max for high-affinity sites in developing tissue with the maximum sensitivity around eye opening; mature tissue exhibited a decrease inB max. In contrast, a gradual decrease in sensitivity to stimulation of the low-affinity sites occurred. These data indicate that the trophic action of GABA is limited to the time when the tissue is developing.  相似文献

An endogenous inhibitor of γ-aminobutyric acid (GABA) receptors was partially purified from bovine brain striatum. It was obtained as a low molecular weight fraction by gel filtration on Biogel P-2 and was adsorbed to Dowex AG 50W-X8, but not to Dowex AG 1-X8. It was ninhydrin-negative, basic, heat-stable substance. It caused dose-dependent inhibition of Na+-independent [3H]GABA bindings. Scatchard plot analysis of the [3H]GABA binding to GABA “B” receptor recognition site showed this inhibitor increased the Kd value (24.1 nM to 3.6 nM) without changing the Bmax. On the other hand, Scatchard plot analysis of the [3H]GABA binding to GABA “A” receptor recognition site showed that the inhibitor decreased number of binding sites (706 fmol/mg protein to 494 fmol/mg protein) without affecting the Kd value. These results suggest that the endogenous inhibitor functions as a modulator for GABAB and GABAA receptors.  相似文献

The kinetics of specific GABA-binding to membranes isolated from cerebellar granule cells, cultured for 12 days from dissociated cerebella of 7-day-old rats was studied using [3H]GABA as the ligand. The granule cells were cultured in the presence of the specific GABA receptor agonist 4, 5, 6, 7-tetrahydroisoxazolo[5,4-c]pyridin-3-ol (THIP, 150 M) or THIP plus the antagonist bicuculline methobromide (150 M of each) or in the absence of the agonist or antagonist. Membranes isolated from granule cells cultured in a medium without the GABA agonist revealed a single binding site for GABA with a binding constant (K D) of 7.9±0.4 nM and aB max of 3.42±0.08 pmol×mg–1 protein. Membranes from cells cultured in the presence of THIP had two binding sites for GABA withK D-values of 6.8±0.9 nM and 476±311 nM, respectively. The correspondingB max values were 4.41±0.42 pmol×mg–1 and 5.81±1.20 pmol×mg–1. The effect of culturing the cells in THIP was antagonized by the simultaneous presence of bicuculline in the culture media, i.e. no significant low-affinity binding for GABA was found on the membranes from granule cells cultured in both THIP and bicuculline. TheK D value (14.3±1.4 nM) for the high affinity binding site was, however, slightly increased compared to the non-treated cells. These findings suggest that the ability of THIP to induce formation of low-affinity GABA receptors is mediated by preexisting high-affinity GABA-receptors on the granule cells.  相似文献

The effect of intrastriatal microinjection of kainic acid (KA) on specific binding of [3H]muscimol to the particulate fractions obtained from corpus striatum (CS), globus pallidus (GP), substantia nigra (SN), and cerebral cortex (CC) was examined. Seven days after the unilateral intrastriatal microinjection of KA, the amount of specifically bound [3H]muscimol was significantly increased at the injected site, whereas no significant alteration of [3H]muscimol binding was found in GP, SN, or CC. Scatchard analysis of striatal binding revealed that microinjection of KA significantly increased the affinity (KD) of GABA receptors on the injected (lesioned) side of the CS without affecting the total number of binding sites (Bmax) therein. This significant increase in [3H]muscimol binding, however, was eliminated by pretreating particulate fractions from the CS with Triton X-100, a non-ionic detergent. No statistically significant difference in amounts of [3H]muscimol binding was detected when the preparations from the KA-treated and non-treated CS were preincubated with 0.05% Triton X-100, respectively. Scatchard analysis using CS preparations treated with 0.05% Triton X-100 revealed that the affinity of the GABA receptor was increased by treatment with Triton X-100, while the total number of binding sites (Bmax) was unchanged by this treatment. These results suggest that neuronal degeneration produced by KA in vivo and pretreatment of particulate preparations with Triton X-100 in vitro may increase the amount of specifically bound [3H]muscimol to CS preparations by a similar molecular mechanism.  相似文献

Effects of bicuculline in vitro, and acute and chronic treatment of a subconvulsive dose of bicuculline on [3H]SR 95531 binding to discrete regions of rat brains were studied in Sprague-Dawley rats. Scatchard analysis of the binding isotherms exhibited two populations of binding sites for [3H]SR 95531 in frontal cortex, cerebellum, striatum and substantia nigra. The apparent KD for high-affinity sites was significantly increased in the frontal cortex and cerebellum in the presence of bicuculline (1 M) with no change in Bmax. In contrast, the apparent affinity for low-affinity sites was not altered in the presence of bicuculline in these regions, whereas the Bmax was significantly decreased in the cerebellum. Following acute (2 mg/kg, i.p.) or chronic (2 mg/kg, i.p. for 10 days) bicuculline treatment, [3H]SR 95531 binding was also investigated in various regions of brains. The acute bicuculline treatment did not affect the [3H]SR 95531 binding in any of the regions studied. In contrast, apparent affinity for [3H]SR 95531 was significantly decreased in low-affinity sites of all regions studied in rats treated chronically with bicuculline. The Bmax values of high and low-affinity sites were significantly increased in the cerebellum with no change in the frontal cortex, striatum and substantia nigra. The present study demonstrates that chronic bicuculline treatment decreases apparent affinity of [3H]SR 95531 binding whereas the treatment increases apparent affinity of [3H]SR 95531 and [3H]muscimol binding in the cerebellum may be due to true up-regulation of GABA binding sites, involving increased de novo synthesis of receptor protein. Abstract: The binding of [3H] γ-aminobutyric acid ([3H]GABA) and [3H]muscimol has been studied in purified synaptic plasma membrane (SPM) preparations from rat brain. Scatchard analysis of specific binding (defined as that displaced by 100 μMγ-aminobutyrate) indicated that the binding of both radiolabelled ligands was best described by a two component Langmuir adsorption isotherm. The apparent KD and Bmax values for [3H]GABA at 4°C were KD1, 20 nM; KD2,165 nM; Bmax1, 0.48 pmol;Bmax2, 6.0 pmol. mg?1; for [3H]muscimol at 4°C they were: KD1, 1.75 nM; KD2, 17.5 nM; Bmaxl, 0.84 pmol. mg?1; Bmax2, 4.8 pmol.mg?1; and for [3H]muscimol at 37°C they were: KD1, 7.0 nM; Km, 60 nM; Bmax], 0.5 pmol-mg?1; Bmax2, 7.2 pmol-mg1. Under the experimental conditions used, the similar Bmilx values for [3H]GABA and [3H]muscimol binding to the SPM preparations suggests that the high- and low-affinity components for the two radiolabeled ligands are identical. The effects of the GAB A antagonist bicuculline on the binding of [3H]muscimol at 4CC and 37°C were studied. At 4°C, antagonism of muscimol binding appeared to be competitive at the high-affinity site but noncompetitive at the low-affinity site. At 37°C, antagonism was again competitive at the high-affinity site but was of a mixed competitive/noncompetitive nature at the low-affinity site. Assuming that binding to the high-affinity site is associated with the pharmacological actions of bicuculline, the apparent KD values obtained suggest a pA2 value of 5.3 against [3H]muscimol at 4°C and 37°C. This figure is in good agreement with several estimates of the potency of bicuculline based on pharmacological measurements. Results from displacement studies using [3H]GABA and [3H]muscimol suggest that [3H]GABA might be a more satisfactory ligand than [3H]muscimol in GABA radioreceptor assays.  相似文献

The effect of early undernutrition and dietary rehabilitation on [3H]gamma-aminobutyric acid ([3H]GABA) binding in rat brain cerebral cortex and hippocampus was examined. Undernourished animals were obtained by exposing their mothers to a protein-deficient diet during both gestation and lactation. Saturation analysis of [3H]GABA binding in the cerebral cortex and hippocampus revealed high- and low-affinity components in the undernourished group, whereas control animals possessed only a low-affinity site. The concentration of low-affinity binding sites was greater in the undernourished animals. Rehabilitation of undernourished animals completely abolished the binding site differences. Treatment of brain membranes with Triton X-100 yielded two binding components in both the undernourished and control animals, although the concentration of lower affinity sites was still greater in the undernourished group. Neither the efficacy nor the potency of GABA to activate benzodiazepine binding in cerebral cortex was modified by undernutrition. These data suggest that early undernourishment modifies the characteristics of [3H]GABA binding, perhaps by reducing the brain content of endogenous inhibitors of the higher affinity binding site. The lack of effect on GABA-activated benzodiazepine binding suggests the possibility that neither the high- nor the low-affinity GABA binding sites are coupled to this receptor component.  相似文献

The aim of the present study was to investigate the effect of environmental conditions such as light-and-dark-adaptation on the plasticity of GABA receptor sites in the chick retina. In chicks exposed to light for 5 hr (light-adapted), specific [3H]GABA binding was increased by 35% in comparison to the binding found in chicks maintained in darkness (dark-adapted). Conversely, in the retina of chicks exposed to darkness for 5 hr, specific [3H]GABA binding was decreased by 28% with respect to that found in chicks kept in the light. Scatchard analysis of the binding data revealed that the affinity of GABA for its receptor binding site was higher in the retinas of light-adapted chicks than in those of dark-adapted chicks (K d values of 19.20±1.23 and 27.20±1.47 nM, respectively). On the contrary, the maximal number of binding sites (Bmax) remained unchanged in light- and dark-adapted chicks (5.2±0.10 and 5.3±0.15 pmol/mg protein, respectively). Gamma amino butyric acid is a major inhibitory neurotransmitter in the central nervous system. In the present study we have investigated the alteration of GABA receptors in the brain stem of rats during pancreatic regeneration. Three groups of rats were used for the study: sham operated, 72 h and 7 days partially pancreatectomised. GABA was quantified by [3H]GABA receptor displacement method. GABA receptor kinetic parameters were studied by using the binding of [3H]GABA as ligand to the Triton X-100 treated membranes and displacement with unlabelled GABA. GABAA receptor activity was studied by using the [3H]bicuculline and displacement with unlabelled bicuculline. GABA content significantly decreased (P < 0.001) in the brain stem during the regeneration of pancreas. The high affinity GABA receptor binding showed a significant decrease in B max (P < 0.01) and K d (P < 0.05) in 72 h and 7 days after partial pancreatectomy. [3H]bicuculline binding showed a significant decrease in B max and K d (P < 0.001) in 72 h pancreatectomised rats when compared with sham where as B max and K d reversed to near sham after 7 days of pancreatectomy. The results suggest that GABA through GABA receptors in brain stem has a regulatory role during active regeneration of pancreas which will have immense clinical significance in the treatment of diabetes.  相似文献

Two commonly used procedures for removing endogenous GABA from brain homogenates were evaluated by measuring residual GABA using high performance liquid chromatography (HPLC). The effect of these treatments on [3H]muscimol binding to the GABA receptor was also determined. Membranes subjected to osmotic lysing and eight washes with Tris-citrate buffer contained significant quantities of residual GABA whereas lysing and incubation with Triton X-100 followed by three buffer washes resulted in GABA levels below the limits of detection. The apparent affinity for [3H]muscimol was significantly higher in the Triton X-100 treated membranes and this was probably a result of the lower amount of GABA present in these membranes. The effect of Triton treatment or buffer washing on residual levels of glutamate, glutamine, aspartate, and taurine were also determined.  相似文献

In two synaptosomal fractions from bovine retina, Triton X-100 and sodium perchlorate specifically enhanced the high affinity binding of 3H-GABA to sites with pharmacological specificity similar to the GABA receptor. Maximal effects were noted at 0.05% Triton X-100 and 100 mM sodium perchlorate. In the fraction enriched in photoreceptor cell synaptosomes from the outer plexiform layer, Triton and perchlorate had similar effects in that two binding sites were observed: a higher affinity site (~20 nMKD) and a lower affinity site (~200 nMKD). However, in the fraction enriched in conventional sized synaptosomes primarily from the inner plexiform layer, the 20 nM site was virtually absent after Triton treatment, but was readily detectable in the presence of perchlorate. These results may suggest that 3H-GABA binding in vitro is inhibited by an endogenous substance which is removed by Triton or perchlorate treatment. The difference in the sensitivity of the two fractions to Triton and perchlorate suggests that in retina this substance (whether it is a membrane peptide or GABA itself) is not uniformly distributed and/or uniformly sensitive to Triton and perchlorate.  相似文献

Abstract The pharmacological and biochemical characteristics of [3H]desipramine binding to rat brain tissue were investigated. Competition studies with noradrenaline, nisoxetine, nortriptyline, and desipramine suggested the presence of more than one [3H]desipramine binding site. Most of the noradrenaline-sensitive binding represented a high-affinity site, and this site appeared to be the same as the high-affinity site of nisoxetine-sensitive binding. The [3H]desipramine binding sites were abolished by protease treatment, a result suggesting that the binding sites are protein in nature. When specific binding was defined by 0.1 μM nisoxetine, the binding was saturable and fitted a single-site binding model with a binding affinity of ~1 nM. This binding fraction was abolished by lesioning of the noradrenaline neurons with the noradrenaline neurotoxin N-(2-chloroethyl)-N-ethyl-2-bromo-benzylamine (DSP4). In contrast, when 10 μM nisoxetine was used to define the specific binding, the binding was not saturable over the nanomolar range, but the binding fitted a two-site binding model with KD values of 0.5 and >100 nM for the high- and low-affinity components, respectively. The high-affinity site was abolished after DSP4 lesioning, whereas the low-affinity site remained. The binding capacity (Bmax) for binding defined by 0.1 μM nisoxetine varied between brain regions, with very low density in the striatum (Bmax not possible to determine), 60-90 fmol/mg of protein in cortical areas and cerebellum, and 120 fmol/mg of protein in the hypothalamus. The binding capacities of these high-affinity sites correlated significantly with the regional distribution of [3H]noradrenaline uptake but not with 5-[3H]hydroxytryptamine uptake. The low-affinity sites did not correlate with the regional distribution of [3H]noradrenaline uptake. Drug inhibition studies showed that noradrenaline inhibits the binding defined by 0.1 μM nisoxetine in a competitive manner. Together, these findings suggest that only a small fraction of the [3H]desipramine binding can be regarded as “specific” binding, and this binding fraction may represent the substrate recognition site for noradrenaline uptake. Assuming that one molecule of desipramine binds to each carrier molecule, the turnover number for the noradrenaline carrier was calculated to be 20/min, i.e., the duration of one transport cycle was 3 s.  相似文献

This study was undertaken to define optimal conditions for exchange of 3H-R-1881 with endogenous hormone bound to androgen receptor (AR) sites in homogenates of rat ventral prostate (RVP) of mature animals. To minimize inactivation of AR binding sites under exchange conditions, extracellular proteases present in RVP were removed by mincing and washing tissue fragments in a Ca2+-free cell culture medium (J-MEM) containing 1% casein, prior to homogenization in a TEDG buffer (50 mm Tris-maleate buffer, pH 7.4; 1.5 mm EDTA; 2.0 mm DTT; and 10% (vv) glycerol) containing 0.5 mm phenylmercuric sulfonyl fluoride (PMSF) and 1.0 mm sodium azide. Na2MoO4 (final concentration, 20 mm) was added to homogenate fractions which then were incubated at 0–4 °C for 1–5 days with a saturating concentration of 3H-R-1881 (20 nm) in the absence and presence of 2 μm radioinert R-1881. Heparin (200 μg/ml) was added to the incubation medium to “solubilize” nuclear chromatin. Free and bound R-1881 were separated by a hydroxylapatite (HAP) batch procedure. Using these conditions, it has been found that (i) incubation periods of 72–96 h at 0–4 °C were required to achieve maximal specific exchange binding of 3H-R-1881 (Bmax) in total homogenates from normal intact rats. Heparin addition (200 μg/ml) did not change Bmax and had little or no effect on the rate of exchange. Mean Bmax was 6.7 ± 1.6 (SD) pmol 3H-R-1881/mg DNA. R-1881 exchange at 24 h of incubation was only about 40% of Bmax. Nonspecific binding, a small fraction (<10%) of Bmax, was near maximal at 2 h. Incubation at 15 °C gave similar R-1881 exchange values to those obtained at 0 °C during the first 24 h, but at 48 h and thereafter R-1881 binding markedly decreased. These Bmax values in total homogenates of normal intact RVP are about 2.5 times greater than the AR values obtained in 1-day castrated rats, when compared on a DNA basis, (ii) Addition of gelatin (0.25%) to inhibit seminin activity had no effect on Bmax. Deletion of either MoO42? or PMSF from the standard exchange medium reduced Bmax values ~20%; if both PMSF and MoO42? were deleted, Bmax was reduced to a greater extent (~35%). Bmax was reduced (40%) when homogenates were prepared without preliminary J-MEM:casein pretreatment and incubated in standard exchange medium with PMSF and MoO42?. (iii) Despite AR stabilization by MoO42? and inhibition of protease activities during exchange incubation, AR inactivation increased exponentially, so that the maximal 3H-R-1881 binding value achieved at 96 h was estimated to represent about 50% of the AR sites originally present. (iv) The binding sites in total homogenates occupied by 3H-R-1881 at 24, 72, and 96 h of exchange exhibited steroid specificity characteristics of AR, as determined by competition studies with a wide variety of steroid hormones and analogues. Scatchard plots of 3H-R-1881 exchange binding in total homogenates of normal intact RVP incubated for 72 or 96 h indicated a single class of affinity sites with apparent Kd of 5 to 6 nm. (v) Sucrose density gradient centrifugation of homogenates incubated for 72 or 96 h showed that the specific 3H-R-1881 binding sites were distributed in two broad peaks associated with low-molecular-weight components. One with S value ~3.5 may be “activated” AR; the other near the top of the gradient (S < 1.6) may include meroreceptor forms of AR.  相似文献

l-[3H]Glutamate binding to synaptic plasma membranes from rat cerebral cortices was carried out at 2–4°C in 50 mM Tris-acetate buffer (pH 7.4) using a microfuge centrifugation method. Binding was increased by repeated freezing-thawing and washing in either crude or partially purified synaptic membranes. Scatchard analysis showed a single binding site (dissociation constant, KD = 697 nM; maximal binding capacity, Bmax = 7.5 pmol/mg protein) in four times distilled water washed crude synaptic membrane. After six times freezing-thawing and washing, a new high affinity site (KD1 = 26 nM, Bmax1 = 1.8 pmol/mg protein) appeared and the number of low affinity site was increased with no apparent change in affinity (KD2 = 662 nM, Bmax2 = 10.5 pmol/mg protein). l-[3H]Glutamate binding was inhibited by acidic amino acid analogues that interact with N-methyl-d-aspartate- and quisqualate-sensitive sites of glutamate receptors. Binding was marginally inhibited by kainate and l-2-amino-4-phosphonobutyrate. These results indicate that repeatedly frozen-thawed and washed synaptic plasma membrane is suitable for studying the subtypes and regulation of glutamate receptors.  相似文献

The influence of activation and inhibition of serotonin receptors by serotonin (5HT) and miancerin on binding of specific nonselective α2-antagonist [3H]RX821002 in rat cerebral cortex membranes was studied. It was shown that the ligand-receptor interaction for α2-adrenoceptors corresponded to the model suggesting the presence of one pool of receptors and binding of two ligand molecules to the receptor. The parameters of the [3H]RX821002 binding to α2-adrenoceptors were as follows: K d = 1.57 ± 0.276 nM, B max = 7.24 ± 1.63 fmol/mg protein, n = 2. In the case of activation of 5HT-receptors by serotonin, the character of ligand binding was different: two pools of receptors were detected with the parameters K d1 = 0.82 ± 0.06; K d2 = 2.65 ± 0.22 nM; B m1 = 1.65 ± 0.23; B m2 = 4.20 ± 0.11 fmol/mg protein; n = 2. The affinity of high-affinity receptors increased twofold and the affininty of low-affinity receptors decreased by 69% as compared to control values. The concentration of high-affinity receptors decreased 4.4-fold, and of low-affinity, 1.7-fold. The value of maximal reaction (B max) decreased by 20%. In the case of miancerin-induced inhibition of 5HT-receptors the character of ligand binding also changed; two pools of receptors were detected with the following parameters: K d1 = 0.48 ± 0.09; K d2 = 3.79 ± 0.71 nM; B 1 = 0.63 ± 0.17; B 2 = 4.75 ± 0.21 fmol/mg protein; n = 2. The affinity of high-affinity receptors pool increased by 70% and that of low-affinity receptors decreased by 76% as compared to control values. The concentration of active high-affinity and low-affinity α2-adrenoceptors decreased by 70% and 141%, respectively. The total amount of the receptors (B max) decreased by 26%. The data suggest that α2-adrenoceptors in rat cerebral cortex exist as dimers. Modulatory effects of serotonin and miancerin on specific antagonist binding to α2-adrenoceptors may be accomplished by altering the character and binding parameters of the nonselective α2-antagonist [3H]RX821002.  相似文献

The method used to prepare crude synaptic membranes (CSMs) from rat brain affects the results obtained for the binding characteristics of 3H-diazepam and the GABA-induced stimulation of 3H-diazepam to CSM. In freshly prepared membranes (rich in GABA and other endogenous inhibitory factors), the KD for 3H-diazepam is approximately 10 nM and the threshold dose of GABA needed to stimulate this binding is approximately 10?5M. Removal of GABA resulted in an increase in the KD values for 3H-diazepam binding. In contrast removal of endogenous inhibitory factors (by treatment of the membranes with Triton X-100) resulted in a decrease of the KD values. In the Tritron X-100 treated membranes the threshold dose of GABA (10?8M) required to stimulate 3H-diazepam binding is in the range of the high affinity component of 3H-GABA binding. Addition of crude preparations of endogenous inhibitor to these membranes increased the KD of 3H-diazepam and inhibited the GABA-induced stimulation of 3H-diazepam binding.  相似文献

The specific binding of GABA (γ-aminobutyric acid) agonist 3H-muscimol, to synaptic membranes from the rat brain showed a significant increase, when the membranous preparations were treated with a low concentration (10?4–10?5M) of mercurial sulfhydryl reagents such as p-chloromercuribenzoate and mercuric chloride. This activation in GABA receptor binding was bicuculline-sensitive, and was partially restored by subsequent treatments with 10 mM cysteine, penicillamine, or mercaptoethanol. Scatchard analysis of the binding revealed that this activation was due to the increase in the affinity of both high and low affinity bindings sites but not in the Bmax values. On the other hand, the treatment of synaptic membranes with hydrophilic sulfhydryl reagents such as N-ethylmaleimide and iodoacetate had no effect on the binding. These hydrophilic sulfhydryl reagents, however, induced an increase of the binding following the pretreatment of synaptic membranes with 0.01% Triton X-100 or 0.5 U/mg prot. of phospholipase A2 (EC 3.1.1.4.). These results suggest that mercurials-sensitive sulfhydryl groups, which are normally masked by membrane lipids, may play a modulating role in GABA receptor binding at central synapses.  相似文献
