A novel mixed-ligand nickel(II) complex that contains 1-methylimidazole and thiocyanate, Ni(NCS)(2)(Mim)(4) (Mim=1-methylimidazole), was synthesized and its structure was determined by X-ray crystallography, IR spectrum and elemental analysis, etc. Its DNA-binding properties were studied by electronic absorption spectral, viscositive and electrochemical measurements. The absorption spectral and viscositive results suggest that the nickel(II) complex binds to DNA via partial intercalation. The addition of DNA results in the decrease of the peak current of the nickel(II) complex proved their interaction. The slight differences of peak profiles and electrochemical parameters between free and DNA-bound Ni(NCS)(2)(Mim)(4) showed the formation of an electrochemical inactive complex between Ni(NCS)(2)(Mim)(4) and DNA. The binding site and binding constant of the complex to DNA were determined by electrochemical titration method.  相似文献

Novel trinuclear Ni(II) complex [Ni3(pmdien)3(btc)(H2O)3](ClO4)3 · 4H2O, 1 where pmdien = N,N,N′,N′,N″-pentamethyldiethylenetriamine, H3btc = 1,3,5-benzenetricarboxylic (trimesic) acid, has been prepared and structurally characterized. Three nickel atoms are bridged by btc trianion and their coordination sphere is completed by three N atoms of pmdien and O atom of the water molecule. The three nickel(II) magnetic centers are equivalent and their coordination spheres are completed to deformed octahedrons. Magnetic susceptibility was measured over the temperature range 1.8–300 K and zJ = ?0.19 cm?1, D = 3.79 cm?1, g = 2.18 parameters were calculated.  相似文献

A series of mixed-ligand neutral nickel(II) complexes of the general formula [Ni(i-MNT)(2a-5mt)2] (1), [Ni(i-MNT)(2a-2tzn)2] (2) and [Ni(i-MNT)(Im)2] (3), [where i-MNT2? = the dianion of 1,1-dicyano-2,2-ethylenedithiolate, 2a-5mt = 2-amino-5-methyl thiazole, 2a-2tzn = 2-amino-2-thiazoline and Im = imidazole] were prepared and characterized with elemental analyses, spectroscopic (IR, UV–vis) methods, magnetic susceptibility, molar conductivity and cyclic voltammetry measurements. The magnetic data, the electronic spectra and the electrical conductivity measurements indicated mononuclear neutral complexes with square-planar geometry. The X-ray analysis of [Ni(i-MNT)(2a-5mt)2] shows the nickel atom being fourfold coordinated with the two sulfur atoms of the dithiolate (i-MNT) ligand and the endocyclic nitrogen atoms from the two 2a-5mt ring giving rise to a slightly distorted square-planar arrangement. The cyclic voltammograms of the complexes have been recorded and the corresponding redox potentials have been estimated. The DNA-binding studies of the complexes has been evaluated by examining their ability to bind to calf-thymus DNA (CT DNA) with UV spectroscopy and cyclic voltammetry. Both studies have shown that the complexes can bind to CT-DNA by the intercalative and the electrostatic binding mode. Competitive binding studies with ethidium bromide (EB) with fluorescence spectroscopy have also shown that the complexes exhibit the ability to displace the DNA-bound EB indicating that they can bind to DNA in strong competition with EB.  相似文献

Two novel oxovanadium(IV) complexes [VOL1]SO4 (1) and [VOL2]SO4 (2) containing Knoevenagel condensate Schiff base ligand (L1/L2) have been synthesized and characterized by physical, spectral and analytical methods. These complexes are reported as ionic in nature on the basis of elemental composition and molar conductance, and possess square pyramidal geometry around the central metal ions. The binding interactions of (1) and (2) with calf thymus DNA (CT DNA) were explored by absorption spectrophotometric titration, cyclic voltammetry data and viscosity measurements. The calculated intrinsic binding constant values (Kb) for (1) and (2) obtained from UV–Vis absorption studies are 0.4 × 105 and 5.6 × 105 (M−1) respectively. These experimental results indicate that (1) and (2) are intercalative binders and avid binder to CT DNA with different affinities. These complexes exhibit significant oxidative cleavage of supercoiled plasmid (pUC18) DNA in the presence of activators. In particular, the in vitro antimicrobial efficacy of oxovanadium(IV) complexes reveal that they are more active than free ligands. Besides, the in vitro cytotoxic effect of the titled complexes were examined on a bundle of human tumor cell lines such as MCF-7 and HeLa cancerous cell lines by the MTT method. Interestingly, complex (2) exhibits more potent cytotoxic activity than the other complex and standard drug (cisplatin). The mode of cell death was assessed by Hoechst 33258 staining morphological studies.  相似文献

Four novel trinuclear copper(II)/nickel(II) complexes with four trianionic pentadentate ligands, N-(3-t-butylbenzoyl)-5-nitrosalicylhydrazide (H33-t-bbznshz), N-(3,5-dimethylbenzoyl)salicylhydrazide (H33,5-dmbzshz), N-(phenylacetyl)-5-bromosalicylhydrazide (H3pabshz) and N-(3-t-butylbenzoyl)salicylhydrazide (H33-t-bbzshz) have been synthesized and characterized by X-ray crystallography. These trinuclear compounds all have an M–N–N–M–N–N–M core formed by three metal ions and two ligands. The geometries of three Cu(II) ions in compound Cu3(3-t-bbznshz)2(H2O)(DMF)(py)2 · DMF (1) alternate between distorted square pyramidal and square planar, while in compound Cu3(3,5-dmbzshz)2(py)2 (2), they are all square planar. Three Ni(II) ions in compound Ni3(pabshz)2(DMF)2(py)2 (3) and Ni3(3-t-bbzshz)2(py)4 · 2H2O (4) follow square-planar/octahedral/square-planar coordination geometry. Compounds 1, 2 and 4 are bent trinuclear, with the bend angles of 156.4°, 141.49° and 127.1°, respectively, while the three nickel ions in compound 3 are strictly linear, with an angle of 180°. Studies on the trinuclear Ni(II) complexes show that the β-branched N-acylsalicylhydrazide ligands with sterically flexible Cα methylene groups are easier to yield linear trinuclear Ni(II) complexes, while α-branched N-acylsalicylhydrazides ligands tend to form bent trinuclear Ni(II) complexes. Antibacterial screening data indicate that the trinuclear Cu(II) compound 2 is more active than 1 and mononuclear Cu(II) compound, bent trinuclear Ni(II) compound 4 is more active than linear compound 3 and less active than tetranuclear nickel compound in the previous study.  相似文献

A novel copper complex of [Cu(bpy)(pba)2 · H2O] · 0.5H2O (bpy = 2,2′-bipyridine, pba = p-methylbenzoate) was synthesized. The interaction of the complex to native fish sperm DNA was investigated through electrochemistry, electronic absorption spectroscopy and viscosity experiments. In the X-ray crystallography structure, the copper (II) ion is coordinated by two oxygen atoms of two p-methylbenzoate groups, two nitrogen atoms of 2,2′-bipyridine and one water molecule. The observed changes in the physicochemical features of the copper (II) complex on binding to DNA suggested that the complex bind to DNA with intercalation mode via 2,2′-bipyridine ring into DNA base pairs. Electrochemical studies revealed that the complex prefer to bind to DNA in Cu(I) form rather than Cu(II) oxidation state form. Additionally, the nuclease activity of the title complex was assessed by gel electrophoresis assay and the results shown that the copper complex can cleave pBR322 DNA effectively in the presence of ascorbic acid.  相似文献

New 3,5-bis(2-pyridyl)pyrazolato (bpypz) bridged heterometal dinuclear complexes [(nta)Cr(μ-bpypz)MII(picen)]+ (M = Mn(II), Ni(II)) and [(acac)2Cr(μ-bpypz)NiII(picen)]2+ (nta = nitrilotriacetate, picen = N,N′-bis(2-pyridylmethyl)ethylenediamine, acac = acetylacetate) were synthesized and characterized by the X-ray analysis, ESI-MS and the magnetic measurements, and/or 2H NMR spectra. The molecular structures were compared from a viewpoint of the conformation of the picen depending on MII ionic radii or different modes of hydrogen bonds. The picen in [(nta)Cr(μ-bpypz)MnII(picen)]BF4 takes an abnormal conformation with intramolecular bifurcated three-center hydrogen bonds between two carboxylate oxygens of nta and an amine proton of the picen as found for the previously reported corresponding Fe(II) complex [K. Ni-iya, A. Fuyuhiro, T. Yagi, S. Nasu, K. Kuzushita, S. Morimoto, S. Kaizaki, Bull. Chem. Soc. Jpn. 74 (2001) 1891]. On the other hand, for both Ni(II)-nta and Ni(II)-acac complexes, the picen takes a normal conformation with only a two-center hydrogen bond between non-bridging ligands. The magneto-structural relation is discussed for the Cr(III)–Ni(II) complexes in connection with the orthogonality or orbital overlap arising from the difference in distortion around Cr(III) moiety.  相似文献

Selective protein labeling with a small molecular probe is a versatile method for elucidating protein functions under live-cell conditions. In this Letter, we report the design of the binuclear Ni(II)–iminodiacetic acid (IDA) complex for selective recognition and covalent labeling of His-tag-fused proteins. We found that the Ni(II)–IDA complex 1-2Ni(II) binds to the His6-tag (HHHHHH) with a strong binding affinity (Kd = 24 nM), the value of which is 16-fold higher than the conventional Ni(II)–NTA complex (Kd = 390 nM). The strong binding affinity of the Ni(II)–IDA complex was successfully used in the covalent labeling and fluorescence bioimaging of a His-tag fused GPCR (G-protein coupled receptor) located on the surface of living cells.  相似文献

In this study, four mononuclear M(II)-pyridine-2,5-dicarboxylate (M = Co(II), Ni(II), Cu(II) and Zn(II) complexes with pyridine-2,5-dicarboxylic acid or isocinchomeronic acid, 1,10-phenanthroline (phen), [Co(Hpydc)2(phen)]·H2O (1), [Ni(pydc)(phen)2]·6.5H2O (2) [Cu(pydc)(phen)(H2O)2] (3) and [Zn(pydc)(phen)(H2O)2]·H2O (4) have been synthesized. Elemental, thermal and mass analyses, molar conductance, magnetic susceptibilities, IR and UV/vis spectroscopic studies have been performed to characterize the complexes. Subsequently, these ligands and complexes were tested for antimicrobial activity by disc diffusion method on Gram positive, negative bacteria and yeast. In addition, cytotoxic activity tests were performed on rat glioma (C6) cells by MTT viability assay for 24 and 48 h. Antimicrobial activity results demonstrated that when compared to the standard antibiotics, phen displayed the most effective antimicrobial effect. The effect of synthesized complexes was close to phen or less. Cytotoxic activity results showed that IC50 value of phen was determined as 31 μM for 48 h. (1) and (2) compared to the alone ligand had less toxic activity. IC50 values of (3) for 24 and 48 h treatments were 2.5 and 0.6 μM, respectively. IC50 value of (4) for 48 h was 15 μM. In conclusion, phen, (3) and (4) may be useful as antibacterial and antiproliferative agents in the future.  相似文献

5-(4-(N-tert-Butyl-N-aminoxylphenyl))pyrimidine (RL, 4PPN) forms crystallographically isostructural and isomorphic pseudo-octahedral M(RL)2(hfac)2 complexes with M(hfac)2, M = Zn, Cu, Ni, Co, and Mn. Multiple close contacts occur between sites of significant spin density of the organic radical units. Magnetic behavior of the Zn, Cu, Ni, Co complexes appears to involve multiple exchange pathways, with multiple close crystallographic contacts between sites that EPR (of 4PPN) indicates to have observable spin density. Powder EPR spectra at room temperature and low temperature are reported for each complex. Near room temperature, the magnetic moments of the complexes are roughly equal to those expected by a sum of non-interacting moments (two radicals plus ion). As temperature decreases, AFM exchange interactions become evident in all of the complexes. The closest fits to the magnetic data were found for a 1-D Heisenberg AFM chain model in the Zn(II) complex (J/k = (?)7 K), and for three-spin RL—M—RL exchange in the other complexes (J/k = (?)26 K, (?)3 K, (?)6 K, for Cu(II), Ni(II), and Co(II) complexes, respectively).  相似文献

Two dinuclear nickel(II) complexes, [Ni2(L-Et)(N3)(H2O)3](NO3)2 · 2H2O (1) and [Ni2(L-Et)(μ-1,3-N3)(H2O)2](NO3)2 · 4H2O (2) containing (HL-Et = N,N,N′,N′-tetrakis[(1-ethyl-2-benzimidazolyl)methyl]-2-hydroxy-1,3-diaminopropane), have been synthesized and characterized by their IR and UV–Vis spectra and magnetic susceptibilities. The crystal structures of [Ni2(L-Et)(N3)(H2O)3](NO3)2 · CH3OH (1′) and [Ni2(L-Et)(μ-1,3-N3)(H2O)2](NO3)2 · 2C2H5OH (2′) similar to 1 and 2 were determined by X-ray crystallography. In 1′, the two nickel(II) ions are bridged by only an alkoxo group of L-Et, while an azido and an alkoxo connect two nickel(II) ions in 2′. Magnetic susceptibility measurements (2–300 K) showed a weak ferromagnetic exchange coupling between the two nickel(II) ions (2J = 10.1 cm−1) for 1. On the other hand, antiferromagnetic interactions were observed for 2 (2J = −15.8 cm−1).  相似文献

Several peroxidovanadium(V) complexes have been shown as a potent anticancer agents. The aim of this study was to investigate the interaction of monoperoxidovanadium(V) complex Pr4N[VO(O2)(ox)(phen)], (Vphen), [phen = 1,10-phenantroline, ox = oxalate(2?) and Pr4N = tetra(n-propyl)ammonium(1+)] with DNA. UV–Vis spectrophotometry and the alkaline single-cell gel electrophoresis (SCGE, the comet assay) were used to examine the possibility of the vanadium(V) complex to induce changes in DNA. The interaction of Vphen with calf thymus DNA resulted in absorption hyperchromicity in DNA spectrum and shift of the absorption band of DNA to longer wavelengths for the [complex]/[DNA] concentration ratio equals to 4 and after 60 min of incubation. The rise in DNA absorption (by 34%) and bathochromic shift (Δλmax = 6 nm) are indicative of the interaction between DNA and the complex molecules. DNA strand breaks in cellular DNA were investigated using the comet assay. The human lymphocytes were exposed to various concentrations of Vphen for 30 min. The results revealed that Vphen contributed to the DNA damage expressed as DNA strand breaks in concentration dependent manner. The used concentrations of Vphen (ranging from 0.1 to 100 μmol/L) caused higher DNA damage in lymphocytes compared to untreated cells (from 1.2 times for 0.1 μmol/L to 1.8 times for 100 μmol/L). Vphen was screened for its potential antitumor activity towards murine leukemia cell line L1210. Vphen exhibited significant antiproliferative activity depending on its concentration and time of exposure. The IC50 values were 0.247 μg/mL (0.45 μmol/L) for 24 h, 0.671 μg/mL (1.21 μmol/L) for 48 h and 0.627 μg/mL (1.13 μmol/L) for 72 h.  相似文献

We have synthesized a series of M(IPSO)3(BPh4)2 complexes consisting of first row transition metals and a photochromic phenanthroline ligand: (M = Mn(II), Fe(II), Co(II), Ni(II), Zn(II), and Cu(II), IPSO = spiro[indole-phenanthrolinoxazine]). The optical properties associated with photochromic behavior were evaluated by determination of the (i) equilibrium constants (K) for the thermal and photostationary states, (ii) rates of photochemical ring-opening and ring-closure, and (iii) rates of thermal ring-closure. The photocolorability values associated with conversion from the colorless spirooxazine to the colored photomerocyanine isomer are greatly enhanced upon metal coordination. Variable temperature magnetic susceptibility experiments suggest deviations from cubic symmetry associated with desymmetrization induced by a dependence of ligand field strength on the photochromic state of IPSO.  相似文献

From the 2,4-bis(cyanamido)cyclobutane-1,3-dione dianion (2,4-NCNsq2−), two copper complexes [Cu2(PPh3)4(PhCN)2(μ-2,4-NCNsq)] · PhCN (1) and [Cu(dien)(μ-2,4-NCNsq) · H2O]n (2) have been synthesized and characterized by IR and electronic absorption spectroscopies. Their structures have been determined by X-ray crystallography. Complex 1 is a dinuclear copper(I) compound with a 2,4-NCNsq2− ligand bridging two copper atoms through the nitrile nitrogen atoms. Complex 2 appears as a 3D network constituted of copper(II) atoms bridged by 2,4-NCNsq2− dianions. This complex presents an unexpected coordination mode of the bis(cyanamido) ligands which are both coordinated via the nitrile functions and via the amido nitrogen atoms of the NCN groups.  相似文献

A tetrapeptide, l-histidyl-glycyl-glycyl-l-histidine (HGGH), was synthesized and the pUC19 plasmid DNA cleaving activity by copper(II) complex of HGGH (Cu(II)−HGGH) was investigated. Cu(II)−HGGH showed bleomycin-like DNA cleaving activity and, at 50 nM, converted a supercoiled DNA efficiently to a linear DNA in the presence of 500 μM H2O2/sodium ascorbate through an oxidative pathway.  相似文献

The synthesis and characterization by elemental analysis, emission atomic spectroscopy, TG measurements, magnetic measurements, FTIR, 1H NMR, UV–visible spectra and conductivity of a series of iron (II) and nickel (II) complexes with two heterocyclic ligands (L1(SMX): sulfamethoxazole and L2(MIZ): metronidazole) used in pharmaceutical field and with a new ligand derived benzoxazole (L3(MPBO): 2-(5-methylpyridine-2-yl)benzoxazole), were reported. The formulae obtained for the complexes are: [M(L1)2 Cl2nH2O, [M(L2)2Cl2(H2O)2]·H2O and [M(L3)2(OH)2nH2O. Stability constants of these complexes have been determined by potentiometric methods in water–ethanol (90:10, v/v) mixture at a 0.2 mol L?1 ionic strength (NaCl) and at 25.0 ± 0.1 °C. Sirko program was used to determine the protonation constants as well as the binding constants of three species [ML2H2]2+, [ML2] and [ML]2+. The antimicrobial activity of the ligands and complexes was evaluated in vitro against different human bacteria and fungi using agar diffusion method.Iron sulfamethoxazole complex showed a remarkable inhibition of bacteria growth especially on Staphylococcus aureus and P. aeruginosa. The iron metronidazole complex is active against yeasts especially on Candida tropicalis strain. Nickel complexes presented different antibacterial and antifungal behavior's against bacteria and fungal.The acute toxicity study revealed that the iron complexes are not toxic at 2000 mg/kg dose orally administrated.LD50 for nickel complexes was determined using graphical method.No significant differences in the body weights between the control and the treated groups of both rat sexes in subacute toxicity study using for iron complexes. Hematological and clinical blood chemistry analysis revealed no toxicity effects of the iron complexes. Pathologically, neither gross abnormalities nor histopathological changes were observed for these complexes.  相似文献

Two novel heteropolytungstates, [Ni(2,2′-bpy)3]1.5{PW10.79V1.21O40(VO)0.5[Ni(2,2′-bpy)2L]} · 0.5H2O (1) (2,2′-bpy = 2,2′-bipyridine, L = 0.5H2O + 0.5OH?) and K[PW12O40[Ni(1,10-phen)2(OH)]2] · 2H2O (2) (1,10-phen = 1,10-phenanthroline), have been synthesized under hydrothermal conditions and characterized by IR, XPS, and X-ray diffraction analyses. The polyoxoanion of 1 is a mono-capped α-Keggin cluster which supports a metal coordination fragment [Ni(2,2′-bpy)2L]1.5+, while the structure of 2 exhibits a 1D chain constructed from pseudo-Keggin cluster bi-supported transition metal complexes linked by K+ ions. Studies of magnetic properties indicated the presence of paramagnetic behaviours for compounds 1 and 2.  相似文献

BackgroundThe human telomere contains tandem repeat of (TTAGG) capable of forming a higher order DNA structure known as G-quadruplex. Porphyrin molecules such as TMPyP4 bind and stabilize G-quadruplex structure.MethodsIsothermal titration calorimetry (ITC), circular dichroism (CD), and mass spectroscopy (ESI/MS), were used to investigate the interactions between TMPyP4 and the Co(III), Ni(II), Cu(II), and Zn(II) complexes of TMPyP4 (e.g. Co(III)-TMPyP4) and a model human telomere G-quadruplex (hTel22) at or near physiologic ionic strength ([Na+] or [K+]  0.15 M).ResultsThe apo-TMPyP4, Ni(II)-TMPyP4, and Cu(II)-TMPyP4 all formed complexes having a saturation stoichiometry of 4:1, moles of ligand per mole of DNA. Binding of apo-TMPyP4, Ni(II)-TMPyP4, and Cu(II)-TMPyP4 is described by a “four-independent-sites model”. The two highest-affinity sites exhibit a K in the range of 108 to 1010 M 1 with the two lower-affinity sites exhibiting a K in the range of 104 to 105 M 1. Binding of Co(III)-TMPyP4, and Zn(II)-TMPyP4, is best described by a “two-independent-sites model” in which only the end-stacking binding mode is observed with a K in the range of 104 to 105 M 1.ConclusionsIn the case of apo-TMPyP4, Ni(II)-TMPyP4, and Cu(II)-TMPyP4, the thermodynamic signatures for the two binding modes are consistent with an “end stacking” mechanism for the higher affinity binding mode and an “intercalation” mechanism for the lower affinity binding mode. In the case of Co(III)-TMPyP4 and Zn(II)-TMPyP4, both the lower affinity for the “end-stacking” mode and the loss of the intercalative mode for forming the 2:1 complexes with hTel22 are attributed to the preferred metal coordination geometry and the presence of axial ligands.General significanceThe preferred coordination geometry around the metal center strongly influences the energetics of the interactions between the metallated-TMPyP4 and the model human telomeric G-quadruplex. The effect of Cu2+ on the DNA interaction of [(bpy)2Ru(μ-bipp)Ru(bpy)2]4+ (bpy = 2,2′-bipyridine, bipp = 2,9-bis(2-imidazo[4,5-f][1,10]phenanthroline)-1,10-phenanthroline) was studied by electronic absorption and emission methods. Bipp has vacant chelating sites in the middle phenanthroline moiety and can bite a Cu2+ in the absence of DNA, as evidenced by the absorptive and emissive decrease of diruthenium complex. The resulted heterometallic complex binds to DNA via intercalation of the middle part of bipp into the DNA base pairs, demonstrated by the dramatic hypochromism and emission enhancement. The maximum emission enhancement is 1.9-fold, larger than the case without copper. The DNA bound [(bpy)2Ru(μ-bipp)Ru(bpy)2]4+ cannot bite Cu2+ due to the difficult access of the vacant chelating sites of bipp caused by the deep insertion of bipp in the DNA base pairs, as can be told since Cu2+ did not disturb the absorption and emission spectra of the DNA bound diruthenium complex.  相似文献
