The paper presents recent trends in solventless sample preparation techniques for environmental analysis. First, a general classification of solventless methods is given. Next, three of them, treated as preferable techniques, i.e. SPME, SDME and HS, are presented in detail, with respect to their usability and effectiveness for environmental samples. Blackflies are important macroinvertebrate groups from a public health as well as ecological point of view. Determining the biological and environmental factors favouring or inhibiting the existence of blackflies could facilitate biomonitoring of rivers as well as control of disease vectors. The combined use of different predictive modelling techniques is known to improve identification of presence/absence and abundance of taxa in a given habitat. This approach enables better identification of the suitable habitat conditions or environmental constraints of a given taxon. Simuliidae larvae are important biological indicators as they are abundant in tropical aquatic ecosystems. Some of the blackfly groups are also important disease vectors in poor tropical countries. Our investigations aim to establish a combination of models able to identify the environmental factors and macroinvertebrate organisms that are favourable or inhibiting blackfly larvae existence in aquatic ecosystems. The models developed using macroinvertebrate predictors showed better performance than those based on environmental predictors. The identified environmental and macroinvertebrate parameters can be used to determine the distribution of blackflies, which in turn can help control river blindness in endemic tropical places. Through a combination of modelling techniques, a reliable method has been developed that explains environmental and biological relationships with the target organism, and, thus, can serve as a decision support tool for ecological management strategies.  相似文献

Recently there has been a growing demand for information on the biodegradability and microbial toxicity of xenobiotic compounds. The environmental fate and effect of these chemicals in aerobic and anaerobic conditions has been extensively studied using a variety of different methods. This paper reviews the different protocols, equipment and instruments used in anaerobic biodegradation and activity tests, including the more sophisticated automated techniques recently developed. The different systems for detecting biogas production are given particular attention. As well as gasometric methods, the different instrumental techniques used to follow the fate of compounds under anaerobic conditions, via analysis of substrate consumption and product formation are evaluated.  相似文献

Sustainable development of nanotechnology will inevitably require incorporation of life cycle thinking to analyze environmental impacts of nanomanufacturing. While many concerns have been raised regarding the human and ecological health effects of and benefits from using nanoproducts, relatively little attention has been given to the manufacturing phase. Unlike many conventional manufacturing techniques, nanomanufacturing techniques require unique facility and process design as well as operation and control. Accordingly, the environmental burden of most nanomanufacturing techniques may be more profound than that of many other conventional techniques. This article reports on a comprehensive review of the current state of technologies used in the manufacture of nanostructured materials in order to identify those attributes that contribute to environmental impacts. It provides a preliminary analysis of significant attributes of commonly used nanomanufacturing techniques, including strict material purity requirements, less tolerance for defects, low process yields and material utilization efficiencies, repetitive processing steps, the need for specialized environments (such as high temperatures or cryogenic processing), the use of toxic chemicals and solvents, the need for moderate to high vacuum, the use or generation of greenhouse gases, high energy and water consumption, and the potential for chemical exposure.  相似文献

A statistical mechanical model of a bilayer of dipalmitoyl-3-sn-phosphatidylcholine molecules in equilibrium with an aqueous phase saturated with an n-alkane is presented. A mean-field approach developed in previous work on a solventless bilayer (Gruen, Biochim. Biophys. Acta. 595:161--183, 1980) is extended to allow alkane chains to exist in the hydrophobic core of the membrane. As the alkane chains are chemically similar to the lipid chains, much of the analysis follows directly from the solventless model. Novel features of the present model are the inclusion of (a) a term which models the free energy cost of creating space for alkane conformations, (b) a term which constrains the chains to pack evenly in the hydrophobic region of the membrane, and (c) a term which estimates the free energy of mixing of the lipid and alkane molecules in the plane of the bilayer. On uptake of alkane, the dimensions of the bilayer increase. Allowance is made for an increase in thickness and/or an increase in area per lipid. A thermodynamic framework is established which allows evaluation of the free energy of a bilayer of arbitrary dimensions with a view to predicting the equilibrium structure.  相似文献

There is an increasing worldwide concern about the problem of dealing with the waste electrical and electronic equipment (WEEE), given the high volume of appliances that are disposed of every day. In this article, an environmental evaluation of WEEE is performed that combines life cycle assessment (LCA) methodology and multivariate statistical techniques. Because LCA handles a large number of data in its different phases, when one is trying to uncover the structure of large multidimensional data sets, multivariate statistical techniques can provide useful information. In particular, principal‐component analysis and multidimensional scaling are two important dimension‐reducing tools that have been shown to be of help in understanding this type of complex multivariate data set. In this article, we use a variable selection method that reduces the number of categories for which the environmental impacts have to be computed; this step is especially useful when the number of impact categories or the number of products or processes to benchmark increases. We provide a detailed illustration showing how we have used the proposed approach to analyze and interpret the environmental impacts of different domestic appliances.  相似文献

Purpose

In this two-part paper (Background and Initial Assumptions (part 1) and Results of Survey Research (part 2)), we present surveys whose main objective is to determine whether, and to what extent, the life cycle assessment (LCA) technique is used for the identification and assessment of environmental aspects in environmental management systems (EMS) and whether there are any differences in this respect between the companies and countries analysed.

Methods

The survey research was carried out using the computer assisted self-administered interviewing method among selected Polish, German and Swedish organisations which implement EMS in accordance with the requirements of ISO 14001 and/or the EMAS regulation.

Results

The organisations investigated, regardless of their country, are dominated by qualitative and semi-quantitative techniques of assessment and identification of environmental aspects. LCA was used sporadically, although some differences can be observed between the countries analysed.

Conclusions

The environmental managers accustomed to traditional qualitative and semi-quantitative solutions have not been given preparation to enable them to understand and adopt different approaches such as LCA. On the other hand, representatives of the organisations investigated declared that they were ready to accept an even longer timescale for the identification and assessment processes relating to environmental aspects, which represents a potential opportunity for LCA. The more precise understanding and definition of environmental problems that are precisely defined in LCA would represent a novelty for environmental managers. In practice, environmental problems are defined in a general sense and rather ambiguously, as this level of detail is sufficient in the context of qualitative and semi-quantitative techniques commonly used for the identification and assessment of environmental aspects.  相似文献

Planar lipid bilayer membranes are formed from mixtures of pure lipids in the absence of non-biological solvents. The solventless bilayers are characterized by a large specific capacitance (586-957 nF/cm2) comparable to that of cell membranes but considerably greater than that of conventional lipid/decane bilayers. Hydrocarbon solvents, such as n-alkanes or squalene, thicken the bilayer. Membrane dielectric thickness is used as an indicator of bilayer lipid composition. For membranes made from pure monoglyceride/triglyceride mixtures the thickness of the solventless lipid bilayer is independent of both the chain length (11-22 carbons) and mol fraction (0.1-0.9) of triglyceride in the bulk mixture. In contrast, the thickness of the bilayer (2.0-3.3 nm) depends strongly upon the length (16-24 carbons) of the monoglyceride component. Molecular volume considerations lead to the conclusion that the bulk lipid mixture disproportionates to yield bilayer membranes composed of nearly pure monoglyceride. The dielectric thickness of the monoglyceride bilayer is consistent with the notion that the lipid fatty acyl chains are fluid.  相似文献

The term 'heritability', which evokes the image of transmission from parents to children, is used in biology to characterize the resemblance of related individuals in terms of a given characteristic, and to analyse the genetic and environmental causes of this resemblance. In fact, there are three definitions of heritability and it is important to distinguish among these carefully in order to avoid misinterpretations. Various techniques for measuring associated parameters are linked to these definitions. A rigorous analysis of the assumptions which permit the interpretation of parameter estimates is necessary to avoid false conclusions.  相似文献

A simple method of solventless extraction of volatile organic compounds (BTEX) from aqueous samples was developed and validated. A new arrangement of the full volume inside needle capillary adsorption trap (INCAT) device with Porapak Q as a sorbent material and wet alumina as a source of desorptive water vapour flow in a closed analytical system is presented. The analytical characteristics of developed device and of compared purge-and-trap (PTI) device for BTEX compounds are similar; the limits of detection as well as quantification are lower than 1 microg l(-1).  相似文献

Highly active CALB cross-linked enzyme aggregates (CLEAs) were synthesized using a layered methodology based on the synthesis of a cross-linked protein cofeeder core over which an external layer of lipase was later cross-linked. The layered CALB CLEAs were characterized in terms of their catalytic activity in three different test reactions: esterification of oleic acid and ethanol in absence of solvents, esterification of oleic acid and heptanol in organic medium, and hydrolysis of triolein in emulsioned medium. The impact of the cross-linker/protein mass ratio on CLEAs activity, and its evolution with storage time were evaluated in the solventless synthesis of ethyloleate. The amount of cross-linker used showed to be a key parameter for the evolution of the catalytic activity of CLEAs during storage. Under the best conditions found, hyperactivated CALB CLEAs with up to 188% of recovered activity in ethyl oleate synthesis were obtained. In terms of hydrolytic activity mature layered CALB CLEAs showed a retained activity of 68%. The assay of dried mature layered CALB CLEAs in heptyl oleate synthesis showed catalytic activities much higher than the one exhibited by free CALB, reaching 1 h-fatty acid conversions of 14% and 2%, respectively. The high catalytic activity shown by layered CALB CLEAs, suggests that they are an interesting alternative specially for the catalysis of fatty acid esterifications in both organic and solventless medium.  相似文献

微生物生态研究中,对微生物群落结构、群落特征以及其与环境因素的关系的揭示,一直受到广泛关注;适当的数据分析方法有助于更清晰地刻画微生物群落结构特征,明确其与环境因素的关系。结合实例,对微生物生态研究中基于BIOLOG微平板技术的数据分析方法进行梳理,分别介绍数据读取整理、特征指数计算、非限制性排序、限制性排序、聚类分析、环境向量拟合、蒙特尔检验等常用数据操作及生态分析方法;针对不同方法结论,结合研究目标和生态理论给出具有统计学意义的解释,并评价不同方法特点及适用场景;分析过程以R语言实现,并提供全部代码。结果表明,BIOLOG方法产生数据能从多个角度表征微生物群落功能特征,并结合环境指标梯度进行分析;但BIOLOG数据可能不满足正态性分布,在基于正态分布的分析前应提前进行检验;排序分析时应慎用主成分分析,可优先采用其他基于距离矩阵的排序方法;R语言能够简化BIOLOG数据读取及操作,易于完成各类统计分析。本研究能够对微生物生态研究者科学选择应用统计分析方法、提高数据处理效率提供直接参考。  相似文献

Traditional techniques for assessment of microbial numbers and activity generally lack the specificity required for risk assessment following environmental release of genetically engineered microbial inocula. Immunological and molecular-based techniques, such as DNA probing and genetic tagging, were initially used to determine the presence or absence of microorganisms in environmental samples. Increasingly they are being developed for quantification of populations of specific organisms, either indigenous or introduced, in the environment. In addition, they are being used to quantify the activity of particular organisms or groups of organisms, greatly extending the range of techniques available to the microbial ecologist. This article reviews the use of traditional techniques for the quantification of microbial population size and activity and the application of molecular techniques, including DNA probing, genetic marking, use of fluorescent probes, and quantitative PCR, in combination with advanced cell detection techniques such as confocal laser scanning microscopy and flow cytometry.  相似文献

The birth and death transition rates for a population are modelled as functions of both the population size and the environmental condition. An assortment of important theoretical results and techniques that can be utilized to analyze such a population’s behaviour is covered. Consequently, these results and techniques are used to study two examples. Firstly, we study a population with a stable equilibrium state, whose per capita birth and death rates are linearly related to the environmental condition. (The environmental condition in turn is modelled as an Ornstein–Uhlenbeck process.) Secondly, we study a population affected by two interdependent environmental factors.  相似文献

Analytical nuclear physics methods, as well as radioanalytical, X-ray, and radiometric techniques are widely used in the Soviet Union in life sciences studies. Main approaches to the study of natural materials by activation analysis, including bulk analyses, localized analyses, speciation, and in vivo experiments are described. Examples of applications in agriculture to increase protein content of wheat, to improve crop yield, and to enhance resistivity of cotton plants to diseases are given. Activation analysis is used in medicine for prognosis and study of processes of pathogenesis. Detailed information on trace elements is valuable in environmental studies, and general regularities in biogeochemistry may be clarified with activation analysis.

LCA is a system-wide assessment, and the LCIA phase is confronted with the difficulties of local and regional effects in a number of impact categories. We integrate three different environmental techniques to demonstrate how these effects can be addressed in an environmental assessment. The techniques are life cycle inventory, environmental fate models, and an ecological impact assessment using fuzzy expert systems. Results of the LCI are mass and energy flows. In the environmental fate modelling step these mass flows are transformed into concentration and immission values by dispersion-reaction models. A generalised fuzzy expert system for the environmental mechanisms compares calculated exposure with site specific buffering capacities and formulates a generalised dose-response relationship. This generalised fuzzy expert system is used as a template for the assessment of local and regional environmental impacts. An application of this integrated approach is shown for a practical problem: production of magnesium car components. The environmental fate of nitrogen oxides which are released due to the major combustion source within that production system is simulated. Fuzzy expert models for crop damage, soil acidification and eutrophication determine the possible environmental impact of the immited nitrogen oxides. The important methodological extension of this integrated approach is a regionalised impact assessment depending on the spatial distribution of environmental characteristics.  相似文献

Stalagmites, which appear to be deposited by cyanobacterial action, have been identified in two caves in New South Wales (Australia). These have a characteristic morphology, which has given rise to local names likening them to crustaceans. We have studied these speleothems, and the cyanobacteria covering them, by microscopic and chemical techniques. Both calcite precipitation and aeolian sediment trapping are involved in their deposition, which is strongly controlled by environmental conditions. They can be regarded as stromatolites within currently accepted definitions of the term.  相似文献
