The demand for automation of liquid-liquid extraction (LLE) in drug analysis combined with the demand for reduced sample preparation time has led to the recent development of liquid-phase microextraction (LPME) based on disposable hollow fibres. In LPME, target drugs are extracted from aqueous biological samples, through a thin layer of organic solvent immobilised within the pores of the wall of a porous hollow fibre, and into an microl volume of acceptor solution inside the lumen of the hollow fibre. After extraction, the acceptor solution is subjected directly to a final analysis either by high performance liquid chromatography (HPLC), capillary electrophoresis (CE), mass spectrometry (MS), or capillary gas chromatography (GC) without any further treatments. Hollow fibre-based LPME may provide high enrichment of drugs and excellent sample clean-up, and probably has a broad application potential within the area of drug analysis. This review focuses on the principle of LPME, and recent applications of three-phase, two-phase, and carrier mediated LPME of drugs from plasma, whole blood, urine, and breast milk.  相似文献

Solid-phase extraction (SPE) procedures for pesticide residues in food and environment are reviewed and discussed. The use of these procedures, which include several approaches such as: matrix solid-phase dispersion (MSPD), solid-phase micro-extraction (SPME) and stir-bar sorptive extraction (SBSE), represents an opportunity to reduce analysis time, solvent consumption, and overall cost. SPE techniques differ from solvent extraction depending on the interactions between a sorbent and the pesticide. This interaction may be specific for a particular pesticide, as in the interaction with an immunosorbent, or non-specific, as in the way a number of different pesticides are adsorbed on apolar or polar materials. A variety of applications were classified according to the method applied: conventional SPE, SPME, hollow-fiber micro-extraction (HFME), MSPD and SBSE. Nowadays there is an evident growing interest in natural antimicrobial compounds isolated from food matrices. According to the type of matrix, different isolation and purification steps are needed and as these active compounds belong to different chemical classes, also different chromatographic and electrophoretic methods coupled with various detectors (the most used diode array detector and mass spectrometer) have to be performed. This review covers recent steps made in the fundamental understanding of sample preparation methods as well as of analytical tools useful for the complete characterization of bioactive food compounds. The most commonly used methods for extraction of natural antimicrobial compounds are the conventional liquid-liquid or solid-liquid extraction and the modern techniques such as pressurized liquid extraction, microwave-assisted extraction, ultrasound-assisted extraction, solid-phase micro-extraction, supercritical fluid extraction, and matrix solid phase dispersion. The complete characterization of the compounds is achieved using both monodimensional chromatographic processes (LC, nano-LC, GC, and CE coupled with different type of detectors) and, recently, using comprehensive two-dimensional systems (LC×LC and GC×GC).  相似文献

An analytical process generally involves four main steps: (1) sample preparation; (2) analytical separation; (3) detection; and (4) data handling. In the bioanalytical field, sample preparation is often considered as the time-limiting step. Indeed, the extraction techniques commonly used for biological matrices such as liquid-liquid extraction (LLE) and solid-phase extraction (SPE) are achieved in the off-line mode. In order to perform a high throughput analysis, efforts have been engaged in developing a faster sample purification process. Among different strategies, the introduction of special extraction sorbents, such as the restricted access media (RAM) and large particle supports (LPS), allowing the direct and repetitive injection of complex biological matrices, represents a very attractive approach. Integrated in a liquid chromatography (LC) system, these extraction supports lead to the automation, simplification and speeding up of the sample preparation process. In this paper, RAM and LPS are reviewed and particular attention is given to commercially available supports. Applications of these extraction supports, are presented in single column and column-switching configurations, for the direct analysis of compounds in various biological fluids.  相似文献

Liquid phase microextraction (LPME), especially hollow fiber liquid-phase microextraction (HF-LPME), and dispersive liquid-liquid microextraction (DLLME) offer high enrichments of target analytes in a single step. The analytical usefulness of these techniques is significantly enhanced by coupling them with suitable derivatization methods. Due to their simplicity, diverse bioanalytical applications have recently been reported. This review focuses on the recent developments of the combined LPME (mainly HF-LPME and single drop microextraction (SDME)) and DLLME techniques with derivatization for the analysis of biological samples. A broad range of sample matrices such as urine, blood, plasma and human hair samples with various derivatization methods for polar or ionizable organic compounds will be considered. These techniques can also be extended to the determination of trace metal ions, such as the heavy metal ions (Hg, Pb, and Co) and Se. Future trends of the techniques will also be discussed.  相似文献

Although the importance of chirality in organophosphorus compounds (OPs) is well recognized in relation to their biological effects, as with most chiral pesticides, OPs are generally marketed, used and released to the environment as racemates (i.e., equimolar mixtures of enantiomers). In addition, research on enantioselective environmental fate and effects of chiral OPs is still limited, particularly in the evaluation of enantioselectivity in their environmental degradation. A large number of OPs are chiral compounds, and yet enantioselectivity in their environmental fate and effects is rarely addressed. This paper highlights the current state of knowledge on the environmental occurrence and behavior of chiral OP pesticides. Developments in enantioselective analytical techniques, specifically gas chromatography (GC), high-performance liquid chromatography (HPLC) and capillary electrophoresis (CE), as applied in the evaluation of enantiomer-specific fate and effects of chiral OPs, are also discussed.  相似文献

Tropane alkaloids like atropine are antidotes applied against organophosphorus intoxications. Atropine is toxic itself and should be closely monitored during treatment. Hence, simple, fast, and sensitive determination methods for tropane alkaloids in serum are desirable. Mostly adopted methods of analysis are gas chromatography (GC); high performance liquid chromatography (HPLC), and capillary electrophoresis (CE). Various liquid and solid capillary fillings used in micellar electrokinetic chromatography, microemulsion electrokinetic chromatography, capillary electrochromatography, and enantioseparation provide high versatility to CE applications. In HPLC, specialised columns enhance separation efficacy. Ultraviolet light detection is common practise, but recently sensitivity and analyte identification were enhanced by coupling GC, HPLC, and CE to mass spectrometry. Apart from medical treatment, tropane alkaloids, cocaine in particular, are abused with various intentions. Forensic analysis of tropane alkaloids and their metabolites comprises the additional difficulty of unequivocal drug identification. Because of severe legal consequences, sophisticated analytical methods were developed and may provide additional techniques for therapeutic drug monitoring. Examples from forensic cocaine analysis and from doping analysis are included in this review.  相似文献

The applicability of two-phase liquid-phase micro-extraction (LPME) in porous hollow polypropylene fiber for the sample preparation and the stereoselective pharmacokinetics of mebeverine (MEB) enantiomers (an antispasmodic drug) in rat after intramuscular administration were studied. Plasma was assayed for MEB enantiomer concentrations using stereospecific high-performance liquid chromatography with ultraviolet detection after a simple, inexpensive, and efficient preconcentration and clean-up hollow fiber-based LPME. Under optimized micro-extraction conditions, MEB enantiomers were extracted with 25 μl of 1-octanol within a lumen of a hollow fiber from 0.5 ml of plasma previously diluted with 4.5 ml alkalized water (pH 10). The chromatographic analysis was carried out through chiral liquid chromatography using a DELTA S column and hexane-isopropyl alcohol (85:15 v/v) containing 0.2% triethylamine as mobile phase. The mean recoveries of (+)-MEB and (-)-MEB were 75.5% and 71.0%, respectively. The limit of detection (LOD) was 3.0 ng/ml with linear response over the concentration range of 10-2500 ng/ml with correlation coefficient higher than 0.993 for both enantiomers. The pharmacokinetic studies showed that the mean plasma levels of (+)-MEB were higher than those of (-)-MEB at almost all time points. Also, (+)-MEB exhibited greater t(max) (peak time in concentration-time profile), C(max) (peak concentration in concentration-time profile), t(1/2) (elimination half-life), and AUC(0-240 min) (area under the curve for concentration versus time) and smaller CL (clearance) and V(d) (apparent distribution volume) than its antipode. The obtained results implied that the absorption, distribution, and elimination of (-)-MEB were more rapid than those of (+)-MEB and there were stereoselective differences in pharmacokinetics.  相似文献

The heterocyclic aromatic amine, 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), has been shown to be carcinogenic in rodents, mice and rats. Following phase I N-hydroxylation and phase II esterification PhIP exerts its carcinogenic effect by binding to DNA purines. Quantitative and qualitative analysis of its bioactivated metabolites as well as it detoxification products is important in studying its biological effects and inter- and intra-individual exposures. A review is presented with an extensive coverage of publications specifically reporting on the analysis of PhIP and its phase I and II metabolites in biological matrices, foodstuff and beverages. Analytical techniques such as liquid and gas chromatography coupled with various detection techniques (mass spectrometry, ultraviolet or fluorescence detection) were mostly applied. We conclude that since the initial identification of PhIP in 1986 a large set of assays has been developed for the analysis of PhIP and its phase I and phase II metabolites in a wide range of matrices, these included food products and biological samples such as plasma, urine and faeces. In addition, it was shown that numerous metabolites were recovered and identified. Thus, we conclude that liquid chromatography coupled to mass spectrometry is clearly the method of choice for sensitive qualitative as well as quantitative analysis with high selectivity and reaching lower quantification levels in the sub pg/mL range. The main aim of this review is that it can be used by other researchers as a resource for method development and optimization of analytical methods of PhIP and its carcinogenic or detoxification products.  相似文献

Pesticides released intentionally into the environment and through various processes contaminate the environment. Although pesticides are associated with many health hazards, there is a lack of monitoring of these contaminants. Traditional chromatographic methods-high-performance liquid chromatography, capillary electrophoresis, and mass spectrometry-are effective for the analysis of pesticides in the environment but have certain limitations such as complexity, time-consuming sample preparation, and the requirement of expensive apparatus and trained persons to operate. Over the past decades, acetylcholinesterase (AChE) inhibition-based biosensors have emerged as simple, rapid, and ultra-sensitive tools for pesticide analysis in environmental monitoring, food safety, and quality control. These biosensors have the potential to complement or replace the classical analytical methods by simplifying or eliminating sample preparation and making field-testing easier and faster with significant decrease in cost per analysis. This article reviews the recent developments in AChE inhibition-based biosensors, which include various immobilization methods, different strategies for biosensor construction, the advantages and roles of various matrices used, analytical performance, and application methods for constructing AChE biosensors. These AChE biosensors exhibited detection limits and linearity in the ranges of 1.0×10(-11) to 42.19 μM (detection limits) and 1.0×10(-11)-1.0×10(-2) to 74.5-9.9×10(3)μM (linearity). These biosensors were stable for a period of 2 to 120days. The future prospects for the development of better AChE biosensing systems are also discussed.  相似文献

The aminoglycosides are a large and diverse class of antibiotics that characteristically contain two or more aminosugars linked by glycosidic bonds to an aminocyclitol component. Structures are presented for over 30 of the most important members of this family of compounds. The use of aminoglycosides in clinical and veterinary medicine and in agriculture is described. Qualitative methods for aminoglycoside analysis include X-ray crystallography, nuclear magnetic resonance (NMR) spectroscopy and mass spectrometry (MS). The major part of this article comprises a comprehensive review of quantitative methods for the determination of aminoglycosides. These are microbiological assay, radiochemical assay, radioimmunoassay, enzyme immunoassay, fluoroimmunoassay and other immunoassays, spectrophotometric and other non-separative methods, gas chromatography (GC), thin-layer chromatography (TLC), high-performance liquid chromatography (HPLC), and capillary electrophoresis (CE). Simple spectrophotometric methods may be adequate for the assay of bulk pharmaceuticals and their formulations. Microbiological assays make useful semi-quantitative screening tests for the analysis of veterinary drug residues in food, but rapid enzyme immunoassays are more suitable for accurate measurements of aminoglycosides in complex matrices. Automated immunoassays are the most appropriate methods for serum aminoglycoside determinations during therapeutic drug monitoring. HPLC techniques provide the specificity and sensitivity required for pharmacokinetic and other research studies, while HPLC–MS is employed for the confirmation of veterinary drug residues. The potential for further development of chromatographic and CE methods for the analysis of biological samples is outlined.  相似文献

Peptide interaction is normally monitored by liquid chromatography (LC), liquid chromatography coupled to mass spectrometry (LC-MS), mass spectrometric (MS) methods such as MALDI-TOF/MS or capillary electrophoresis (CE). These analytical techniques need to apply either high pressure or high voltages, which can cause cleavage of newly formed bondages. Therefore, near infrared reflectance spectroscopy (NIRS) is presented as a rapid alternative to monitor the interaction of glutathione and oxytocin, simulating physiological conditions. Thereby, glutathione can act as a nucleophile with oxytocin forming four new conjugates via a disulphide bondage. Liquid chromatography coupled to UV (LC-UV) and mass spectrometry via an electrospray ionisation interface (LC-ESI-MS) resulted in a 82% and a 78% degradation of oxytocin at pH 3 and a 5% and a 7% degradation at pH 6.5. Capillary electrophoresis employing UV-detection (CE-UV) showed a 44% degradation of oxytocin. LC and CE in addition to the NIRS are found to be authentic tools for quantitative analysis. Nevertheless, NIRS proved to be highly suitable for the detection of newly formed conjugates after separating them on a thin layer chromatography (TLC) plate. The recorded fingerprint in the near infrared region allows for a selective distinct qualitative identification of conjugates without the need for expensive instrumentation such as quadrupole or MALDI-TOF mass spectrometers. The performance of the established NIRS method is compared to LC and CE; its advantages are discussed in detail.  相似文献

The detection, qualitative and quantitative methods based on different chromatographic separations of gas chromatography (GC), high-performance liquid chromatography (HPLC) and capillary electrophoresis (CE) coupled with various detections by ultraviolet absorption (UV), electrochemical detection (ED), fluorescence detection, mass spectrometry (MS) and nuclear magnetic resonance spectroscopy (NMR), as well as non-chromatographic immunoassay are each extensively examined and compared. With the growing interest for peptides and proteins in different kinds of fields, e.g. pharmacy, clinical diagnostics or food industry, the quantification of these compounds is becoming more and more important. Quantitative analysis of these analytes in biological matrices, however, remains a challenging task, due to the complexity of both the matrix and the analytical characteristics of these large bio-molecules. Liquid chromatography coupled to (tandem) mass spectrometry (LC-MS or LC-MS/MS) is the preferred analytical technique for peptide analysis as it allows very selective and sensitive measurements. This article summarizes the numerous published LC-MS applications for the quantification of peptides in biological matrices and discusses all different issues herewith concerned. This includes chromatographic aspects as the selection and effects of mobile and stationary phase, flow rate and temperature, as well as mass spectrometric characteristics such as ionization and detection modes, collision-induced dissociation of peptides and factors influencing the mass spectrometric response. For both techniques the main properties of all described methods have been listed, creating a comprehensive overview with the peptide analytes divided into different classes. Likewise, all other issues concerned with quantitative bioanalysis have been evaluated in detail, including extensive consideration of several different applied sample pre-treatment techniques and reflection of subjects as the choice for an internal standard and assay validation. Furthermore, several issues which are of particular interest for the quantitative bioanalysis of peptide compounds like peptide adsorption and degradation have been regarded.  相似文献

作为高效能的中枢神经抑制剂,γ-羟基丁酸(GHB)其相关物质γ-丁内酯(GBL)和1,4-丁二醇(1,4-BD)的滥用现象越来越严重,由于它们强烈的镇静及健忘效果常常被用作迷奸药,带来了严重的社会问题。由于在体内天然存在GHB,而且其在摄入后消除迅速,这增加了体内GHB及其相关物质的检测和分析评价难度。本文在对GHB及其相关物质的理化性质阐述基础上,综述了它们的提取技术及分析方法研究进展,主要阐述了液.液提取、固相萃取等提取方法与气相色谱法、气相色谱-质谱联用法、高效液相色谱法、液相色谱.质谱联用法、毛细管电泳法等在分析不同检材中GHB及其相关物质的应用,这也为国内法庭案件中GHB及其相关物质的滥用及相关案件提供了可供参考的法庭科学检测、分析研究方法。  相似文献

Petroleum is composed of a complex mixture of hydrocarbons that readily undergo chemical and biological conversions on entering aquatic environments. These conversions lead to the formation of a host of oxygenated products, some of which are potentially toxic to marine life and to the consumer of fishery products. State-of-the-art analytical methods, as employed in our laboratories, utilize glass-capillary gas chromatography in conjunction with mass spectrometry to analyze environmental samples containing trace amounts of aliphatic and aromatic petroleum hydrocarbons. These procedures are applied on a routine basis to the analysis of seawater, sediments and tissues of marine organisms. Despite this analytical proficiency, a need exists for analyzing oxygenated and other polar petroleum products in environmental samples. For example, techniques such as high-performance liquid chromatography (HPLC), in conjunction with on-line fluorometric assay techniques and mass spectrometry, make possible the analysis of polar oxygenated compounds resulting from both chemical and biological conversions. These methodologies are first steps toward the development of routine assay procedures for environmental samples. Current techniques for hydrocarbon analyses and new methods for analyzing polar aromatic compounds are discussed.  相似文献
