This paper presents the results of an investigation on the distribution of210Po in Mutharasanallur pond ecosystem. It has been demonstrated that210Po is non-uniformly distributed within the ecosystem. The results of the study show a dissolved210Po concentration in pond water of 1 4mBq 1−1. The sediment samplso recorded a210Po activity of 59 9 Bq kg−1. The aquatic organisms showed differential accumulation of the radionuclide with enhanced bioaccumulation in soft tissues and muscle. The210Po activity in the biota fell within the range of 1·2–53 3 Bq kg−1 (wet wt). The bivalve mussel,Lamellidens marginalis was identified to accumulate higher concentration of210Poin soft tissues, suggesting that these organisms could serve as a bio-monitor of210Po radionuclide in a freshwater system. The concentration factors of210Po for the biotic components ranged from ∼102–∼104. Analyses of the results indicate that prawn and fish represent an important source of supply of210Po to humans via dietary intake. Results of210Po activity in the abiotic and biotic components of the pond ecosystem were higher when compared with those of Cauvery river system, the primary water source of the pond.  相似文献

Concentrations of 210Po, a natural alpha-radiating radionuclide were determined in various Black Sea fish species sampled along the coastal zone and in the bays of Sevastopol from May to June, 1999. Its value increased in the ecological row from the bottom through the demersal to pelagic region. Among the pelagic species, the Black Sea anchovy and sprat displayed the highest concentrations of 210P. In fishes, the liver was the 210Po accumulating organ.  相似文献

A study to determine activity concentrations of 210Pb and 210Po in the urine of certain Finnish population groups was conducted, to investigate the variation in natural background level of urinary excretion. The study participants were divided into three groups mainly based on their diet. The first group comprised recreational fishermen and the second group represented people consuming more reindeer meat than an average Finn, while people using drinking water with very high activity concentrations of 210Po were selected for the third group. The fourth group was a control group. The mean urinary excretion of 210Po in groups 1 and 2 was 73 and 100 mBq d−1, respectively. These values were higher than the value of the control group (20 mBq d−1) and the mean values reported in the literature. The mean daily urinary excretion of 210Pb in groups 1 and 2, 70 and 52 mBq d−1, was also slightly higher than that in the control group (32 mBq d−1). In contrast, the excretion rates of both 210Po and 210Pb for the members of group 3 were one to two orders of magnitude higher than those reported in the literature. This was clearly due to the elevated levels of natural radionuclides in their drinking water. The present study demonstrates the importance of possessing good knowledge of the background levels, in order to allow the determination of the additional exposure due, for example, to the malevolent use of radiation.  相似文献

The effect of dose rate on the induction of lung cancer in Syrian hamsters by 5.3 MeV alpha particles was examined by varying the number of weekly intratracheal instillations of carrier-free 210Po. By this technique, most of the radiation dose to the lungs was delivered over intervals ranging from 10 to 120 days. Protraction of exposure over 120 days was slightly more carcinogenic at lower total lung doses (24 rad), but slightly less carcinogenic at higher doses (240 rad), than exposure limited to a 10-day interval. No synergism was observed between very low radiation exposures (2.4 rad) and simultaneously administered benzo[a]pyrene. The carcinogenic effect of a single intratracheal instillation of 210Po in isotonic saline was markedly enhanced by subsequent weekly instillations of 0.2 ml of saline alone, emphasizing the importance of noncarcinogenic secondary factors in the expression of radiation-induced lung cancer.  相似文献

A simple, inexpensive and convenient radioimmunoassay for rat liver metallothionein has been developed. The double-antibody assay involves the labeling of homogeneous, rat liver zinc thionein with trace amounts of 109Cd(II) to a specific activity of 1–2 × 106 cpm/μg protein; the binding of this antigen by rabbit anti-rat liver metallothionein antiserum; the displacement of this antigen by unlabeled zinc thionein or cadmium, zinc-thionein; the precipitation of the rabbit antibody-rat antigen complex by goat anti-rabbit IgG immunoglobulins; and the binding of this precipitate to cellulose acetate filters. The radioimmunoassay is useful in the range of concentration of metallothionein of 10–500 ng protein. The assay is accurate as compared with a previous technique of quantitating metallothionein in extracts of rat liver. A radial immunodiffusion assay for metallothionein is also described.  相似文献

CALCULATIONS of maximum permissible body burdens of polonium-210 for radiation workers and the general public made by the International Commission on Radiological Protection (ICRP) assume 0.06 as the fraction going from the gastrointestinal tract to blood1. This estimate is based on the work of Delia Rosa et al.2, in which freshly neutralized polonium chloride solution was given orally to rats by stomach intubation (gavage). Excretion functions and tissue retention were measured and used to calculate the transfer of polonium across the gut. Hill3 and Kauranen and Miettinen4, who studied the transfer of Po along the lichen-reindeer or caribou-man food chain, suggested that the 210Po found in the soft tissues of herbivores may be organically bound and therefore readily available to humans. Their data indicated that the 210Po ingested from reindeer meat was from five to twenty-five times more available than predicted from the transfer values adopted by the ICRP. Experiments reported here indicate that polonium secreted in the milk of a goat which had been given an acute oral dose of PoO2 is absorbed across the gut of rats in concentrations approximately twenty times greater than that absorbed when rats are fed PoO2 or PoCl4 in drinking water. These results are at least partial verification of the greater availability of 210Po through biological modification as estimated by Hill.  相似文献

The mechanisms by which DNA-incorporated radionuclides impart lethal damage to mammalian cells were investigated by examining the capacity of dimethyl sulfoxide (DMSO) to protect against lethal damage to Chinese hamster V79 cells caused by unbound tritium ((3)H(2)O), DNA-incorporated (125)I- and (131)I-iododeoxyuridine ((125)IdU, (131)IdU), and cytoplasmically localized (210)Po citrate. The radionuclides (3)H and (131)I emit low- and medium-energy beta particles, respectively, (125)I is a prolific Auger electron emitter, and (210)Po emits 5.3 MeV alpha particles. Cells were radiolabeled and maintained at 10.5 degrees C for 72 h in the presence of different concentrations of DMSO (5-12.5% v/v), and the surviving fraction compared to that of unlabeled controls was determined. DMSO afforded no protection against the lethal effects of the high-LET alpha particles emitted by (210)Po. Protection against lethal damage caused by unbound (3)H, (131)IdU and (125)IdU depended on the concentration of DMSO in the culture medium. Ten percent DMSO provided maximum protection in all cases. The dose modification factors obtained at 10% DMSO for (3)H(2)O, (131)IdU, (125)IdU and (210)Po citrate were 2.9 +/- 0.01, 2.3 +/- 0.5, 2.6 +/- 0.2 and 0.95 +/- 0.07, respectively. These results indicate that the toxicity of Auger electron and beta-particle emitters incorporated into the DNA of mammalian cells is largely radical-mediated and is therefore indirect in nature. This is also the case for the low-energy beta particles emitted by (3)H(2)O. In contrast, alpha particles impart lethal damage largely by direct effects. Finally, calculations of cellular absorbed doses indicate that beta-particle emitters are substantially more toxic when incorporated into the DNA of mammalian cells than when they are localized extracellularly.  相似文献

The seaweed Fucus vesiculosus is a member of the brown algae family. Kille and co-workers [Biochem. J. 338 (1999) 553] reported that this species contains the gene for metallothionein. Metallothionein is a metalloprotein having low molecular weight, and high cysteine content, which binds a range of metals. F. vesiculosus bioaccumulates arsenic from the aquatic environment [Mar. Chem. 18 (1986) 321]. In this paper we describe arsenic binding to F. vesiculosus metallothionein, characterized by electrospray ionization mass spectrometry. Five arsenic-MT species were detected with increasing As to protein ratios. These results provide important information about the metal-chelation behaviour of this novel algal metallothionein which is a putative model for arsenic binding to F. vesiculosus in vivo.  相似文献

The risk of an enhanced level of radionuclides of the uranium and thorium decay series in the environment for reference plant species (Pinus sylvestris and Vicia cracca) was assessed. 238U, 230Th, 226Ra, 210Po, 232Th and 228Th concentration factors for plants were found to be lower than one. The aboveground parts of Vicia cracca sampled from the area of the radium production waste storage mainly accumulated 22Ra, Pinus sylvestris branches--210Pb, 226Ra and 210Po. LOEDR calculated for the chromosome aberration frequency in both plant studies was 17-71 microGy/h. LOERD values for the reproductive capacity decrease in P. sylvestris and V. cracca were 17-71 microGy/h and 116-258 microGy/h, correspondingly. EDR10 for the chromosome aberration frequency in P. sylvestris and V. cracca were 148 and 347 microGy/h, that is, correspondingly, 255 and 708 times higher that background values. EDR10 for the plant reproductive capacity was 11-34 microGy/h, which 19-69 times increases the background values.  相似文献

Rats and hamsters, (pre)-treated with copper and cadmium, were used to investigate whether species-differences in renal metallothionein synthesis in response to gold were determined by changes in the kidney concentrations of other metals. The effects of both dietary copper limitation and excess on the renal metabolism of gold also were studied in the rat. In this species, all of the pre-treatments affected the renal concentrations of total and metallothionein-bound copper, but none of them altered either the kidney uptake or thionein-binding of gold. Incorporation of zinc into the metallothionein, which accompanied the binding of gold in this fraction of the kidney, however, was influenced slightly by the pretreatments. In hamsters, pretreatment with cadmium, which increased the concentrations of total and thionein-bound zinc in the kidneys, also did not affect the renal uptake of gold, although it increased significantly the binding of gold to the metallothionein fraction of the renal cytosol. This increased binding of gold also was accompanied by further increases in the zinc and copper contents of the metallothionein; the contents of total and thionein-bound cadmium, however, remained essentially unchanged. Concentrations of copper and zinc in the hamster kidney were not affected significantly by subcutaneous administration of copper alone (five daily doses, each of 3.2 mg Cu/kg body wt.), but were increased when gold was given during the copper-treatment. The concentrations of gold, copper and zinc in the renal metallothionein fraction also were increased under these conditions. From these results it seems that kidney metallothionein synthesis in response to gold may be related to the changes in either the concentration or distribution of zinc, rather than copper.  相似文献

A sensitive and rapid method to estimate concentrations of functional metallothionein in small biological samples, based upon the acid stability of 203Hg binding and solubility of this protein in trichloroacetic acid is described. Sephadex G-10 minicolumns supported in centrifuge tubes afforded separation and quantitation of isotope bound metallothionein from unbound metal. Elution of metallothionein bound 203Hg was achieved by short term-low speed centrifugation that segregated chelator-ligand complex into the eluate while unbound ligand remained in the gel. A well characterized standard of pure metallothionein protein was utilized to verify the specificity and sensitivity of the modified assay. Metallothionein levels were estimated by 203Hg binding in extracts of wild type and cadmium resistant Chinese hamster ovary cells treated with maximum tolerable concentrations of CdCl2. Similar separation methods demonstrated [35S]-cysteine incorporation into induced metallothionein. Additionally, induction of metallothionein was observed after treatment with particulate CdS but not crystalline NiS particles. These results demonstrate that the modified assay system is easily applied to serial measurement of metallothionein levels in multiple small biological samples.  相似文献

In Lahontan Valley, Nevada, arsenic, cobalt, tungsten, uranium, radon, and polonium-210 are carcinogens that occur naturally in sediments and groundwater. Arsenic and cobalt are principally derived from erosion of volcanic rocks in the local mountains and tungsten and uranium are derived from erosion of granitic rocks in headwater reaches of the Carson River. Radon and 210Po originate from radioactive decay of uranium in the sediments. Arsenic, aluminum, cobalt, iron, and manganese concentrations in household dust suggest it is derived from the local soils. Excess zinc and chromium in the dust are probably derived from the vacuum cleaner used to collect the dust, or household sources such as the furnace. Some samples have more than 5 times more cobalt in the dust than in the local soil, but whether the source of the excess cobalt is anthropogenic or natural cannot be determined with the available data. Cobalt concentrations are low in groundwater, but arsenic, uranium, radon, and 210Po concentrations often exceed human-health standards, and sometime greatly exceed them. Exposure to radon and its decay products in drinking water can vary significantly depending on when during the day that the water is consumed. Although the data suggests there have been no long term changes in groundwater chemistry that corresponds to the Lahontan Valley leukemia cluster, the occurrence of the very unusual leukemia cluster in an area with numerous 210Po and arsenic contaminated wells is striking, particularly in conjunction with the exceptionally high levels of urinary tungsten in Lahontan Valley residents. Additional research is needed on potential exposure pathways involving food or inhalation, and on synergistic effects of mixtures of these natural contaminants on susceptibility to development of leukemia.  相似文献

Salps, salp fecal pellets and other zooplankton species were analyzed for a suite of elements and natural radionuclides to assess their role in the biogeochemical cycling of nuclides in oceanic waters. The nuclide/Al ratios in organisms normalized to the same ratio in crustal rock indicated that Ca, Sr, Zn, Cu, U,210Po, and210Pb are enriched in the organisms. The concentrations of Fe, Al, Th isotopes and210Pb in salps and fecal pellets were about an order of magnitude higher than those in salps, whereas Ca, Cu, Zn, Mn and Po were higher by factors of about 2–5. Fluxes via salp defecation were higher than those which have been measured in crustacean zooplankton species, a result primarily due to the high defecation rates characteristic of salps. High nuclide levels in salp fecal pellets coupled with high defecation rates and presumed high salp biomass in many areas underscore the importance of these indiscriminate filter feeders in packaging and transporting to depth particulate-associated nuclides in surface waters.  相似文献

Objectives To identify public perceptions of the risk to health after the poisoning of Alexander Litvinenko with polonium-210 (210Po) in London and to assess the impact of public health communications.Design Cross sectional telephone survey and qualitative interviews.Setting London, United Kingdom.Participants 1000 people completed the cross sectional survey and 86 potentially exposed people completed the qualitative interviews.Main outcome measures Perception of risk to personal health after the 210Po incident. Qualitative interviews were analysed with an emphasis on information needs.Results 11.7% of the survey sample (n=117) perceived their health to be at risk. Aside from personal variables the main predictors of perceived risk to health were believing that the incident was related to terrorism (odds ratio 2.7, 95% confidence interval 1.5 to 4.6) rather than to espionage, that it was targeted at the wider public rather than one person (5.9, 3.2 to 10.9), and that it could affect people who had not been in the contaminated area (3.2, 2.1 to 5.1). Participants in the qualitative interviews were generally satisfied with the information they had received, although they would have preferred more information about their individual risk of exposure, the results of their urine tests, and the health implications of the incident.Conclusions Perceptions of the public that the 210Po incident in London in 2006 was related to espionage helped to reassure them that the risks to personal health were low. In the event of future incidents it is important to ensure that detailed, comprehensible information about the risks of any exposure is available.  相似文献

The gene encoding metallothionein, a low mol. wt. metal binding and stress response protein, is expressed in white adipose tissue. In the present study, metallothionein (MT-1) gene expression and factors regulating metallothionein production have been examined in adipocytes induced to differentiate from fibroblastic preadipocytes in primary cell culture. On the induction of differentiation, the metallothionein-1 gene was strongly expressed in the cells and metallothionein released into the medium. A peak in metallothionein-1 mRNA level and metallothionein secretion occurred at 2 and 10 days post-differentiation, respectively, with a decrease in protein release after this time. The metallothionein-1 gene was expressed in the adipocytes prior to the adipsin and lipoprotein lipase genes, suggesting that it is an early marker of adipocyte differentiation. The addition of the glucocorticoid, dexamethasone, led to a substantial increase in metallothionein-1 mRNA in the cells and metallothionein secretion. Insulin and leptin also stimulated metallothionein production, although the effect was small. Neither noradrenaline nor the beta3-adrenoceptor agonist, BRL 37 344, altered metallothionein release but forskolin and bromo-cAMP were stimulatory, markedly increasing both metallothionein-1 level and metallothionein secretion. It is suggested that metallothionein is a novel secretory product of the differentiated white adipocyte and that its production is regulated particularly by glucocorticoids and through a cAMP-dependent pathway.  相似文献

1. An antimetallothionein antibody, raised against Cd-carrying metallothionein, was applied in Western blotting of metallothionein. 2. Treatment of the electroblotted nitrocellulose sheets with metals belonging to the periodic system transition groups Ib and IIb, or with Pb, Ni or Cr, considerably enhanced binding of anti-metallothionein. A similar effect was found when the electroblotted sheets were treated with the strong alkylator N-ethylmaleimide. 3. It seems that the binding of metal to metallothionein modifies the configuration of the antibody binding sites by the formation of metal thiolate complexes. 4. Metal treatment of the nitrocellulose sheets after electroblotting, but before application of the primary antibody, offers a convenient method for use in Western blotting to significantly potentiate the reaction between metallothionein and the antimetallothionein antibody.  相似文献

Three simple methods were developed to estimate the proportion of particles in lake water derived from resuspended material. These techniques use the different distributions of long and short-lived radioisotopes sorbed onto particles and were tested in the three basins of Lake Erie using 7Be, 137Cs and 210Po/210Pb.While the concentration of 210Po on particles did not vary significantly in the lake, resuspended particles were characterized by high concentrations of 137Cs and low concentrations of 7Be. The distribution of these radioisotopes is consistent with a simple mixing model in which the fraction of particles in the lake water derived from resuspension ranged from 8% to about 100%. Higher concentrations of resuspended particles were found in deeper samples from the nepheloid layer and in the shallow western basin where thermal stratification was very weak.  相似文献

Catfish hepatic metallothionein was purified to homogeneity by Sephadex G-75 gel filtration, DEAE-Sephadex A-25 column chromatography and preparative polyacrylamide gel electrophoresis. Induction by cadmium and zinc, characteristic UV spectrum, cadmium binding property and its low MW established that it was a metallothionein. Antibody was raised in rabbit against catfish metallothionein. Catfish antimetallothionein cross-reacted with other fish metallothioneins but not with chicken or rodent metallothionein. Catfish metallothionein is more electronegative as compared to mouse, rat, chicken or hamster metallothionein. Catfish MT appeared to aggregate readily on storage and to be less electronegative.  相似文献
