The inhibition of a chromatin-bound proteolytic activity   总被引:1,自引:0,他引:1  
A chromatin-bound proteolytic activity found in rabbit thymus nuclear lysates has been studied. Incubation of washed chromatin fibers led to degradation of histone H1 resulting in products which ran on sodium dodecyl sulfate-gels slightly ahead of the three native H1 fractions. The activity has also been demonstrated in a chromatin preparation purified by chromatography on Sephadex G-200. Of the five histones, H1 appeared to be the most readily degraded although with extended incubation times the core histones were also affected. The proteolytic activity was not inhibited by Na2SO3, PMSF, STI, pepstatin, or EDTA. Effective inhibitors included leupeptin, tosyllysine chloromethyl ketone, and tosylaminophenylethylchloromethyl ketone.  相似文献

Prostaglandin A isomerase has been purified 120-fold from rabbit serum by the use of ammonium sulfate fractionation, isoelectric focusing, and Sephadex G-200 chromatography. The molecular weight of the enzyme was estimated to be 110,000 from the elution volume on Sephadex G-200. Prostaglandin A isomerase is a heterogeneous protein with respect to charge. This has been concluded from the spread of enzymatic activity over 1 pH unit after isoelectric focusing. The enzymatic activity is inhibited by N-ethylmaleimide but not by other sulfhydryl blocking agents. The Km was determined to be 5 × 10?5m.  相似文献

Streptomyces sp. No. 280 produced several kinds of amylase inhibitors (amylase inhibitor A, B, B' and C). Two amylase inhibitors (designated as AI-A1 and AI-A2) were obtained from an amylase inhibitor A fraction by paper chromatography. AI-A1 inhibited muscle phosphorylase a much more than AI-A2 and was hydrolyzed by sweet potato β-amylase whereas AI-A2 was not. Both amylase inhibitors had a carbohydrate and were hydrolyzed by some kinds of amylases or acids. They lost their inhibitory activity against phosphorylase a after treatment with acids or hog pancreatic α-amylase, but they showed increased inhibitory activity toward porcine small intestinal sucrase.

Both AI-A1 and AI-A2 were composed of glucose and a basic moiety which gave a positive ninhydrin reaction. The molecular weights of AI-A1 and AI-A2 were estimated to be approximately 1300 ? 1500 by gel filtration on a Sephadex G-15 column. The nitrogen content of the amylase inhibitors was found to be about 1.3% by elementary analysis  相似文献

The polyphenoloxidase (PPO) from black poplar senescent leaves has been purified to almost complete homogeneity by a combination of ammonium sulphate precipitation, Sephadex G75 filtration and DEAE-cellulose chromatography. The purified enzyme has a MW of 60 000 and is probably a Cu+ enzyme. Peroxidase (PO) activity co-purifies with PPO and has the same MW as it. The two enzymes differ in pH optimum and in response to the effect of ionic strength. Natural phenols are either substrates, inhibitors or activators of black poplar PPO. This enzyme is an o-diphenoloxidase which binds substrates with Km in the millimolar range. With caffeic and chlorogenic acids inhibition by excess substrate is observed. Benzoic acid phenols and cinnamic acid phenols are either competitive or non-competitive inhibitors of PPO. Hydroquinone is a highly potent non-competitive inhibitor of the enzyme (Ki  90 μM). Ferulic acid is a potent activator of the PPO-catalysed oxidation of catechol (Ka  0.34 mM, νsato  7.7).  相似文献

Lysosomal carboxypeptidase B has been purified from rabbit lung acetone powder by acid precipitation and ammonium sulfate fractionation followed by further purification on Sephadex G-100, DEAE-Sephadex, Organomercurial-Sepharose, preparative isoelectric focusing, Sephadex G-75, and carboxymethyl-Sephadex. This procedure resulted in a homogeneous preparation as determined by polyacrylamide gel electrophoresis at pH 4.5, 8.3 and with sodium dodecyl sulfate. This enzyme has a molecular weight of 52,000, is composed of two subunits of approximately equivalent molecular weight, and is a glycoprotein with a carbohydrate content estimated to be 10% by weight. The amino acid composition is also reported. The enzyme is active on two synthetic substrates, α-N-benyoyl-l-arginineamide and hippuryl-l-arginine. With these two substrates, respectively, lysosomal carboxypeptidase B has pH optima of 5.7 and 5.0, temperature optima of 40 and 50 °C, and Km values of 10 and 16 mm. With each substrate, the enzyme requires the presence of a reducing agent for maximal activity and is inhibited to the same extent with several inhibitors. The most potent inhibitors were leupeptin and antipain at low concentrations (1 μm). Iodoacetate and Ac-(Ala)3-Ala-chloro-methyl ketone also inhibited at higher concentrations (10 μm). However, compounds such as leucyl-chloromethyl ketone, bestatin, pepstatin, phenylmethylsulfonyl fluoride, soybean trypsin inhibitor, and α-1-antitrypsin did not inhibit. When tested with peptides as substrates, this proteinase exhibited strong carboxypeptidase activity on the tetrapeptide, ThrProArgLys, and on angiotensin I, AspArgValTyrIle HisProPheHisLeu, liberating Lys, and Leu, respectively. Substance P (containing 11 amino acids plus a C-terminal amide group) was virtually inactive as a substrate for this enzyme. However, with oxidized insulin B chain as substrate, lysosomal carboxypeptidase B exhibited significant carboxypeptidase and endopeptidase activities.  相似文献

荔枝果皮采后褐变是影响这一重要热带水果经济价值的主要问题,酚类物质的酶促氧化一直被认为是造成植物组织褐变的关键因素,其中多酚氧化酶被研究得最多.过氧化物酶在植物体中分布很广,能够氧化多种底物,在荔枝果皮中的含量也很高.非结合性过氧化物酶已经被证明在果实的采后成熟与老化过程中参与多种过程.在这项研究中,用磷酸缓冲液提取荔枝果皮的非结合性过氧化物酶,并通过硫酸铵沉淀,DEAE Sephadex A-50离子交换柱层析以及Sephadex G-100凝胶过滤进行纯化.对得到的酶溶液进行了酶学性质的研究,发现荔枝果皮过氧化物酶具有较高的热稳定性和高的最适反应pH值(6.8),能够氧化许多底物尤其是单酚和各种多酚类物质,反应抑制剂专一性与其他植物来源的过氧化物酶略有不同.显示了过氧化物酶参与荔枝果皮褐变过程的可能性,并为提高荔枝采后贮藏性提供了新的思路.  相似文献

Starch phosphorylase from tapioca leaves has been purified to homogeneity, using the technique of ammonium sulfate fractionation, heat treatment, DEAE-cellulose chromatography, filtration through Sephadex G-100 and Sephadex G-200, and DEAE-Sephadex chromatography. The enzyme has a molecular weight of 450,000, as determined by gel filtration through Sephadex G-200 and contains 22 sulfhydryl groups per mole of the enzyme protein. Several types of evidence indicate the absence of pyridoxal 5′-phosphate as a prosthetic group of the enzyme. The kinetic data show a sequential type of the reaction mechanism. The enzyme activity is inhibited by tyrosine (Ki = 2.15 mm).  相似文献

A specific radioimmunoassay for human pancreatic anionic trypsin has been developed. The trypsin employed as radioiodinated tracer in the assay was inactivated with tosyl-L-lysine chloromethyl ketone in order to prevent binding of the tracer to the serum inhibitors α1-antitrypsin and α2-macroglobulin. A normal serum level of immunoreactive anionic trypsin of 5.45 ng/ml was determined. The results of experiments in which serum was fractionated by Sephadex G-200 gel filtration suggest that essentially all of the immunoreactive material in normal human serum is trypsinogen. This finding implies that a small fraction of the zymogens synthesized in the pancreas are released directly into the circulation.  相似文献

Many peroxidase inhibitors have been used in horseradish peroxidase (HRP) mediated immunostaining and in situ hybridization to quench background peroxidase activity. However, the efficacy of these inhibitors has been controversial, partially due to the lack of a quantitative study. Tyramide signal amplification (TSA) is much more sensitive than other HRP-mediated methods but its super-sensitivity also demands effective inhibition of background peroxidase activity. In searching for an effective peroxidase inhibitor, we have systematically evaluated the efficacy of several peroxidase inhibitors by quantifying the fluorescence intensity in cultured fibroblasts and tissue sections treated with the inhibitors. For cultured cells, 0.05 mM of phenylhydrazine and 1 unit/ml of glucose oxidase gave only moderate inhibition of HRP activity while 1 mM of sodium azide (NaN3), 3% of hydrogen peroxide (H2O2), NaN3/H2O2 combined and 0.02 N hydrochloric acid (HCl) provided more complete inhibition. However, the inhibitory effect of NaN3/H2O2 is reversible upon removal of the inhibitors and followed by incubation and wash to mimic antibody interactions. Similar results were obtained from rat skin wound tissues that have strong endogenous peroxidase activity. Our results recommend the use of HCl and caution the use of phenylhydrazine, glucose oxidase, NaN3 and H2O2 as potent peroxidase inhibitors.  相似文献

Candida utilis alkaline phosphatase has been detected in vacuoles. Liberation of the vacuoles was carried out by protoplast disruption under isotonic conditions. The polybase DEAE-dextran was used to induce damage to the yeast plasmalemma. The vacuoles were purified by centrifugation on sorbitol-Ficoll gradients. Alkaline phosphatase from a purified fraction of vacuoles was characterized after gel filtration on Sephadex G-200. We have found 15 mU of enzyme activity per 108 vacuoles. This enzyme activity elutes on Sephadex G-200 at a volume-to-void-volume ratio of 1.65. The approximate molecular weight is 1.35×105. TheK m value forp-nitrophenyl-phosphate is 2.5×10–3 M. The pH for maximum activity is 8.9, and the enzyme is stable at pH values between 7.0 and 9.0. Rapid inactivation occurs at temperatures above 45°C. The enzyme catalyzes the hydrolysis of phosphomonoester bonds of a wide variety of molecules, especially polyphosphates. Thus, vacuolar polyphosphates are probably the natural substrate of this enzyme. Orthophosphate, arsenate, ethylenediaminetetraacetate, molybdate, and borate act as inhibitors. Fluoride is not an inhibitor, and the activity is not affected byp-hydroxymercuribenzoate. Some metal ions also affect the activity of vacuolar alkaline phosphatase. This may indicate that this enzyme is a metalloprotein.  相似文献

Methylmalonyl coenzyme A (CoA) mutase has been purified to apparent homogeneity from human liver by a procedure involving column chromatography on DEAE-cellulose, Matrex-Gel Blue A, hydroxylapatite, and Sephadex G-150. The overall purification achieved is 500- to 600-fold, yield 3–5%. Electrophoresis of the native purified protein on nondenaturing polyacrylamide gels shows a single diffuse band coincident with the enzyme activity; dodecyl sulfate/polyacrylamide gels show a single protein band with an apparent molecular weight of 77,500. The native protein has a molecular weight of approximately 150,000 by Sephadex G-150 chromatography, suggesting that it is composed of two identical subunits. The activity of the purified enzyme is stimulated only slightly (10–20%) by the addition of its cofactor, adenosylcobalamin, indicating that the purified enzyme is largely saturated with coenzyme. The spectrum of the enzyme is consistent with the presence of about 1 mole of adenosylcobalamin per mole of subunit. The enzyme displays complex kinetics with respect to dl-methylmalonyl CoA; substrate inhibition by l-methylmalonyl CoA appears to occur. The enzyme activity is stimulated by polyvalent anions (PO43? > SO42? > Cl?); monovalent cations are without effect, but high concentrations of divalent cations are inhibitory. The enzyme activity is insensitive to N-ethylmaleimide, is rapidly destroyed at temperatures > 50 °C, and shows a broad pH optimum around pH 7.5.  相似文献

In vitro effects of ketamine and bupivacaine drugs on bovine lactoperoxidase (LPO; E.C. 1.11.1.7) enzyme activity were investigated. Lactoperoxidase was purified with Amberlite CG 50 resin, CM Sephadex C-50 ion-exchange chromatography, and Sephadex G-100 gel filtration chromatography from skimmed bovine milk. Rz(A412/A280) value for the purified LPO was found to be 0.8. Inhibition or activation effects of the drugs on LPO enzyme were determined using 2,21-azino-bis (3-ethylbenzthiazoline-6 sulfonic acid) diammonium salt (ABTS) as a chromogenic substrate at pH = 6.0. The I50 values of ketamine and bupivacaine were 0.29 mM and 0.155 mM, respectively and the Ki constants for ketamine and bupivacaine were 0.019 ± 0.031 and 0.015 ± 0.021 mM, respectively; they were non-competitive inhibitors.  相似文献

Whole rat brain dipeptidyl carboxypeptidase (E.C. 3.4.15.1) was heterogeneously distributed among 10 brain regions studied. In corpus striatum, the enzyme was enriched in the P2 pellet, a subfraction high in myelin and nerve terminals. Using [3H]benzoylphenylalanyl-alanyl-proline as a substrate, dipeptidyl carboxypeptidase manifested a different anion requirement than has been reported for other substrates. Endogenous inhibitors of the enzyme were found in corpus striatum and could be removed by dialysis or Sephadex G25 chromatography. Boiled striatal cytosol inhibited membrane-bound enzyme activity in a concentration-dependent manner and confirmed the presence of an endogenous soluble, heat-stable inhibitor in rat brain. The inhibitor apparently could be degraded by a component of the striatal P2 membranes. The inhibitor was present in all 10 brain regions studied and its levels did not appear to be related to the specific activity of dipeptidyl carboxypeptidase. Potential mechanisms for biological regulation of dipeptidyl carboxypeptidase activity are discussed in light of the above findings.  相似文献

Seven closely related protease inhibitors were isolated from commercial bromelain acetone powder in electrophoretically pure form by gel filtration on Sephadex G-75, followed by ion exchange chromatography on DEAE Sephadex at pH 7.55. The inhibitors are proteins of MW 5000-6000, which inhibit competitively the bromelaincatalyzed hydrolysis of CLN (Ki ≈ 10?7 M). This inhibition is optimal at pH 3 to 4,. and it depends upon the ionization of two acidic residues of pK = 4.5 and 5.0. In the acidic pH range the inhibitors are also effective toward papain, ficin and trypsin.  相似文献

The F420-dependent NADP reductase of Methanobacterium thermoautotrophicum has been purified employing a combination of DEAE-cellulose ion-exchange chromatography, affinity chromatography with Blue Sepharose, Sephadex G-200 column chromatography and Red Sepharose affinity chromatography. The enzyme, which requires reduced F420 as an electron donor, has been purified over 3000-fold with a recovery of 65%. A molecular weight of 112000 was determined by Sephadex G-200 chromatography. A subunit molecular weight of 28 500 was determined by Sephadex G-200 chromatography. A subunit native enzyme is a tetramer. The optimal temperature for enzymatic activity was found to be 60°C with a pH optimum of 8.0. The NADP reductase had an apparent Km of 128 μMJ for reduced F420 and 40 μM for NADP. The enzyme was stable for at least 4 h at 65°C and pH 7.5. No loss of enzyme activity was detected when purified enzyme was stored aerobically in buffer containing 2-mercaptoethanol for 10 days at 4°C. Neither FMNH2 nor FADH2 could serve as electron donors; NAD was not utilized as electron acceptor.  相似文献

A carboxylesterase (EC 3.1.1.1) was purified from germinated finger millet by ammonium sulphate fractionation, diethylaminoethyl-cellulose chromatography and Sephadex G-200 filtration. The homogeneity of the enzyme was established by Polyacrylamide gel electrophoresis, isoelectric focussing and sodium dodecyl sulphate-polyacrylamide gel electrophoresis. The enzyme has a single polypeptide chain with a molecular weight of 70,000. The amino acid analysis of the purified enzyme revealed that it contained a greater number of neutral and acidic, compared to, basic amino acid residues. The isoelectric pH of the enzyme was found to be 5·1. Studies with different organophosphate and carbamate inhibitors showed that this enzyme was more sensitive to organophosphate inhibitors than carbamates. The rate constantsk i andl 50 for different inhibitors were calculated. The product inhibition studies with this enzyme showed linear competitive inhibition with acetate and linear noncompetitive inhibition with 1-naphthol  相似文献

A trypsin inhibitor from seeds of faba bean (Vicia faba L.) was purified to near homogeneity as judged by native-PAGE with about 11 % recovery using ammonium sulphate fractionation, ion-exchange chromatography on DEAE-cellulose and gel filtration through Sephadex G-100. The inhibitor had a molecular weight of 18 kD as determined by SDS-PAGE and Sephadex G-100. The inhibitor inhibited trypsin and chymotrypsin to the extent of 48 and 12 %, respectively. The inhibtion was of non-competitive type with dissociation constant for the enzyme inhibitor complex in the region of 0.07 mg·ml−1. The inhibtor was stable between pH 4 and 5. It completely lost its activity when heated at 125 °C for 1 h or at 100 °C for 2 h. The inhibitor also lost its activity on exposure to 2-mercaptoethanol. Based on these properties, it could be concluded that Vicia faba trypsin inhibitor belongs to Bowman-Birk type of inhibitors, as it has molecular weight lower than generally observed for Kunitz type inhibitors.  相似文献

An inducible l-mandelate-4-hydroxylase has been partially purified from crude extracts of Pseudomonas convexa. This enzyme catalyzed the hydroxylation of l-mandelic acid to 4-hydroxymandelic acid. It required tetrahydropteridine, NADPH, Fe2+, and O2 for its activity. The approximate molecular weight of the enzyme was assessed as 91,000 by gel filtration on Sephadex G-150. The enzyme was optimally active at pH 5.4 and 38 °C. A classical Michaelis-Menten kinetic pattern was observed with l-mandelate, NADPH, and ferrous sulfate and Km values for these substrates were found to be 1 × 10?4, 1.9 × 10?4, and 4.7 × 10?5m, respectively. The enzyme is very specific for l-mandelate as substrate. Thiol inhibitors inhibited the enzyme reaction, indicating that the sulfhydryl groups may be essential for the enzyme action. Treatment of the partially purified enzyme with denaturing agents inactivated the enzyme.  相似文献

A series of rationally designed ROS1 tyrosine kinase inhibitors was synthesized and screened. Compound 12b has showed good potency with IC50 value of 209 nM, which is comparable with that of the reference lead compound 1. Molecular modeling studies have been performed, that is, a homology model for ROS1 was built, and the screened inhibitors were docked into its major identified binding site. The docked poses along with the activity data have revealed a group of the essential features for activity. Overall, simplification of the lead compound 1 into compound 12b has maintained the activity, while facilitated the synthetic advantages. A molecular interaction model for ROS1 kinase and inhibitors has been proposed.  相似文献
