When fasted rats were fed a chow or fat-free diet supplemented 5% with L-histidine for three days, the brain adenosine 3′:5′-monophosphate (cAMP) level increased. A 50% increase occurred in rats fed a chow diet and 20% increase in rats fed a fat-free diet. Purification of liver fatty acid synthetase and the isolation of liver apo-, holo-a and holo-b fatty acid synthetases demonstrated that L-histidine feeding caused changes in the relative amounts of these enzymes. Apo- and holo-b fatty acid synthetases increased while the holo-a form simultaneously decreased. This effect was observed in rats fed either chow or fat-free diets supplemented with L-histidine.  相似文献

Evidence is presented in support of the hypothesis that an important step in the adaptive synthesis of fatty acid synthetase is the conversion of inactive enzyme precursors to active enzyme via the incorporation of the 4′-phosphopantetheine prosthetic group. Fatty acid synthetase activity was generated in vitro when CoA or E. coli acyl carrier protein was incubated with enzymatically inactive extracts from livers of rats fed a fat-free diet for 0–5 hr following starvation, and a factor present in liver extracts from rats refed for more than 6 hr. When (14C)-CoA, labelled in the pantetheine moiety, was used in the above system, radioactivity was incorporated into a protein bound form, from which it could be released by mild alkaline hydrolysis.  相似文献

The diurnal variations in enzyme activities including tyrosine aminotransferase (TAT), ornithine decarboxylase (ODC), ornithine aminotransferase (OAT) and serine dehydratase (SDH) have been studied in rats trained to a 2 hour meal feeding schedule (″2+22″) during metabolic transition from 12.5 to 60% protein diets over a period of 21 days. Although the maximal TAT activity on the first day was slightly lower compared with other days, both TAT and ODC activities adapted rapidly to the increased dietary protein from the first day. The responses of TAT and ODC to the food were so rapid that the maximal value was observed only 4 hrs after the onset of feeding. After each feeding ODC activity decreased rapidly after 4 hours, while TAT activity declined only after 6 hours had elapsed. No clear diurnal rhythm was observed in either OAT or SDH, though OAT activity tended to decrease from the beginning of the dark period and to resume a slow adaptation after about four hours. In contrast to ODC and TAT both OAT and SDH required about 7 days to fully adapt to the high protein diet. The activities of the four enzymes were also compared after 4 groups of rats had been adapted to the ″2+22″ feeding of 12.5, 30 and 60% protein diets and to 60% diet, adlibitum, respectively. The enzyme activities were not directly proportional to the protein content of the diets although higher activity was observed on the high protein diets. The diurnal variations in both TAT and ODC were observed in all ″2+22″ groups although the timing of the peak values were slightly different from each other. The maximal activities of TAT were found at earlier times in 12.5 and 30% protein groups than in the 60% protein group. The peak time for ODC activity was found at a later time in the 12.5% protein group than in rats fed 30% and 60% protein. Adlibitum rats fed 60% protein maintained relatively high levels of TAT activity compared to the rats on the schedule. However, the maximal activity of ODC on the 60% ″2+22″ protein diet adlibitum was so low that a diurnal rhythm was not clearly evident.  相似文献

The hepatic removal of plasma chylomicrons was determined for rats fed the following diets: a) containing no triglyceride, b) regular chow diet with 4.5% of its mass as lipid and, c) a corn oil-supplemented chow with triglyceride accounting for 20% of the mass. The fractional hepatic uptake of either radiolabeled chylomicrons or a triglyceride emulsion was reciprocally related to the amount of lipid in the diet. The animals receiving only carbohydrate and protein calories had the most active hepatic uptake of particulate triglyceride and were observed to have a significant decrease in the plasma concentration of the C apolipoproteins. The addition of either C-I, C-II, or C-III apoproteins to the triglyceride emulsion prior to intravenous injection produced a significantly lower hepatic triglyceride recovery of emulsions containing apoC-III. When the plasma of animals fed a fat-free diet was supplemented with human C-III-1 apolipoprotein, the distribution into the liver of either enterally administered fatty acid or parenteral triglyceride was diminished. The triglyceride content in the liver of the rats fed fat-free or corn oil-supplemented diets was significantly greater than that of the control rats and composition was somewhat similar to that of lymph triglyceride. The studies indicate an important influence of dietary lipid on both the partition of plasma triglyceride into the liver and the steady state hepatic triglyceride content.  相似文献

The specific activity (units activity/mg cytosolic protein) of malic enzyme was found to be three-fold higher in the livers of mice fed a semipurified diet containing 50% (ww) glucose and 15% (ww) saturated and monounsaturated but no polyunsaturated fat (hydrogenated cottonseed oil) over an 11-day period than in the livers of mice fed a standard laboratory mouse chow (Purina) diet. In contrast, when other lab chow-fed mice were fed an isocaloric diet containing 15% (ww) polyunsaturated fat (corn oil), no change in the specific activity of malic enzyme occurred over a similar period of time. Rocket immunoelectrophoresis performed on cytosols from both dietary groups demonstrated that the livers of mice consuming the hydrogenated cottonseed oil diet contained approximately three times more malic enzyme protein than did the livers from the corn oil-fed animals. In mice pulse-labeled with l-[4,5-3H]leucine, the rate of hepatic malic enzyme synthesis (relative to that for total protein) was approximately twofold greater in the hydrogenated cottonseed oil-fed mice than in their corn oil-fed counterparts whereas the rate of hepatic malic enzyme degradation was similar for both groups. Immunotitration of liver malic enzyme from hydrogenated cottonseed oil-fed and corn oil-fed mice revealed identical equivalence points, demonstrating that the catalytic efficiency of mouse liver malic enzyme had not been affected by the type of dietary fat administered. When total liver RNA, isolated from the hydrogenated cottonseed oil- and the corn oil-fed animals, was translated in cell-free translation systems (wheat germ extract and reticulocyte lysate) we found that both dietary treatments had resulted in an increase in the activity of malic enzyme messenger RNA. Furthermore, there were no significant differences between the two dietary groups in this regard. These results suggest that hepatic malic enzyme specific activity in high-carbohydrate polyunsaturated fat-fed mice is regulated principally by dietary-induced changes in the rate of enzyme synthesis and not by the activity of messenger RNA coding for the enzyme.  相似文献

Trained and untrained rats were fed either a control, high-fat, or high-carbohydrate diet and then sacrificed in either a rested or exhausted state. The in vitro activity of several muscle glycolytic and liver gluconeogenic enzymes was measured. Muscle hexokinase, phosphorylase, and phosphofructokinase activities were increased after training. Hexokinase was decreased in exhausted rats. Phosphorylase and phosphofructokinase were increased in untrained-exhausted rats but were unchanged in trained-exhausted rats. Liver pyruvate carboxylase and phosphoenolpyruvate carboxykinase activities were increased in trained-rested rats fed a high-fat diet. In trained-exhausted rats phosphoenolpyruvate carboxykinase activity was increased regardless of diet fed. Blood glucose was decreased in trained-exhausted rats, but it was increased in untrained-exhausted rats. Plasma glucocorticoid level was increased in exhausted rats. This study showed that training was associated with an increased muscle glycolytic capacity. Training was also related to the ability of liver to increase phosphoenolpyruvate carboxykinase activity during exercise, thereby increasing gluconeogenic capacity.  相似文献

The contents of plasma free amino acids, the amounts of urinary excreted amino acids and urea, and the activities of liver serine dehydratase, glutamic-oxalacetic transaminase and glutamic-pyruvic transaminase were determined in weanling rats fed ad libitum a 10% casein diet (control), a 10% casein diet containing 7% glycine and 10% casein diets containing 7% glycine supplemented with 1.4% L-arginine and/or 0.9% L-methionine for 14 days.

The remarkable increase of glycine and the moderate increase of serine in the plasma of animals fed excess glycine diets were observed. The amount of excreted glycine in the urine of animals fed the excess glycine diet supplemented with L-arginine and L-methionine was much greater than that of animals given the excess glycine diet. Urinary excreted urea of rats fed the excess glycine diet was a little greater and that of rats fed the excess glycine diet supplemented with L-arginine and L-methionine was much greater than the control. Liver serine dehydratase activity of animals given the excess glycine diets with or without L-arginine was higher than the control and the highest activity was observed in the liver of animals fed the excess glycine diet containing L-arginine and L-methionine. The activity of liver glutamic-oxalacetic transaminase of rats fed the excess glycine diet containing L-arginine and L-methionine was a little higher than that of rats given the other diets. Liver glutamic-pyruvic transaminase activity was a little higher in animals given the excess glycine diets with or without L-arginine and further higher in animals fed the excess glycine diet containing L-arginine and L-methionine than the control.  相似文献

Ketogenic diets are used in the treatment of epilepsy in children refractory to drug therapy. This study identifies changes in liver morphology in rats fed four different diets: a normal rodent chow diet, a calorie-restricted high-fat (ketogenic) diet and each diet supplemented with clofibric acid. Hepatocytes of rats fed the ketogenic diet show many lipid droplets and these are reduced to control levels when clofibrate is present in the diet. Mitochondria are enlarged in the livers of rats fed the ketogenic diet and further enlarged if clofibrate is present. Alterations in the appearance or numbers of other organelles are also found.  相似文献

ATP citrate lyase, acetyl-CoA synthetase, malic enzyme and hexose monophosphate dehydrogenase activities and rates of denovo synthesis of long chain fatty acids from labeled acetate and citrate were measured in cell-free fractions of liver from rats fed various diets, with and without D- or L- thyroxine. Diets containign sucrose (vs. isocaloric glucose) or lard (vs. isocaloric corn oil) stimulated hepatic lipogenesis both in control and in thyroxine-treated rats. The lipogenic response to thyroxine was greatly modified by diet, except for an invariable rise in malic enzyme activity. With diets providing less than 6% of calories as linoleic acid, thyroxine increased fatty acid synthesis, depleted liver glycogen and retarded growth; when linoleic acid was increased to 16% of calories, thyroxine had no effect on fatty acid synthesis or growth and liver glycogen depletion was significantly attenuated. This response to dietary linoleic acid suggests that these phenomena may be largely secondary to the increased requirement for essential fatty acid in thyrotoxicosis. Further study should reveal the extent to which observed effects of excess thyroid hormone are amenable to control by dietary polyunsaturated fat.  相似文献

Changes in the time course of the urinary excretion of creatinine, creatine and urea, and the activities of kidney transamidinase and liver urea-cycle enzymes were investigated in rats fasted and fed on a 10% casein diet and 10% casein diets supplemented with 10% glycine and/or 1.4% arginine.

The urinary total-creatinine of the fasted rats increased extremely during fasting for 7 days, while that of the animals given the 10% casein diet supplemented with glycine and arginine rose exceedingly on the 3rd day and thereafter no significant change was observed. Most of the increase of total-creatinine could be accounted for by the increase of creatine. The activity of kidney transamidinase in the fasted rats decreased in the 3rd day and thereafter kept nearly constant. The transamidinase activity of rats fed on the 10% casein diet after giving a protein-free diet for 5 days increased in the 3rd day. An inverse relation was observed between the urinary creatine and the transamidinase activity. The urinary urea increased in the rats fasted or fed on the 10% casein diets with the supplement of glycine and/or arginine. In fasting, the activities of liver urea-cycle enzymes, except arginase, had a tendency of increasing with the lapse of time. The arginase activity remained more or less constant. The reason of the extreme increase of urinary creatine during starvation was discussed.  相似文献

Erythrocyte and liver tissue δ-aminolevulinate dehydratase activity was determined in rats fed a semipurified diet under controlled nutritional intake of zinc and copper. A significant decrease in enzymatic activity was observed in animals fed low zinc diet, while dietary copper had no effect. In vitro addition of zinc to the erythrocyte preparations obtained from rats on low zinc diet produced a slight increase in enzymatic activity. It appears that, even though zinc may be the metal ion activator of δ-aminolevulinate dehydratase, the requirement of this metal is at the site of synthesis of this enzyme.  相似文献

Guinea pigs fed purified diets that contained either 0.4% or 1.0% of phosphorus were injected intraperitoneally with crude Salmonella typhimurium endotoxin. Functional activity of the RES was evaluated after endotoxin administration by determining the initial clearance rates of non-viable 32P-labeled Salmonella typhimurium. Three days after administration of endotoxin the RES clearance rates were approximately 212 times greater in animals fed 1.0% than in those fed 0.4% of dietary phosphorus. After the RES response had risen to a maximum (5–6 days), there was no difference between dietary groups and RES clearance rates remained high throughout an 18-day period. The increased clearance rate induced by endotoxin involved largely the liver RES. Endotoxin administration led to a decrease in blood inorganic phosphorus in both dietary groups but the concentration in those fed the higher phosphorus level remained significantly higher. It was concluded that the protective action of high phosphorus is mediated at least in part by its beneficial effect on RES function.  相似文献

The norepinephrine (NE) concentration of 14 discrete brain nuclei of rats fed with either purina rat chow or highly palatable foods, given ad litibum or in a restricted quantity isocaloric with the Purina rat chow diet, was measured by high performance liquid chromatography with electrochemical detection. The NE level was significantly increased in the n. suprachiasmaticus, n. paraventricularis, n. arcuatus and n. amygdaloideus (medialis, corticalis, centralis, basalis pars lateralis, lateralis pars posterior) of rats fed with palatable foods given in restricted amount. The increased NE level seems to be associated with the limitation of access to highly palatable food whilst the regimen composition and the palatability of foods are not implicated in this phenomenon. Some hypothalamic and amygdaloid NE-containing neurones can be affected by food-restriction stress, suggesting a role of this neuronal system in feeding activity.  相似文献

To determine whether changes in unsaturation of fatty acids in rat liver plasma membranes might alter activities of membrane-associated enzymes, liver plasma membranes were prepared from rats fed purified diets lacking or supplemented with essential fatty acids. Two methods of membrane purification were used. A similar degree of purification was obtained with both methods for both depleted and control membranes, as indicated by marker enzyme purification. The proportion of essential fatty acids of the linoleate series was significantly lower in phospholipids from depleted rats. The specific activity of 5′-nucleotidase was lower, and the activity, V and apparent Km for total (Na++K++Mg2+)-ATPase were higher in the depleted liver plasma membranes. Arrhenius plots of total ATPase activity showed a discontinuity at the same temperature for both the depleted and control membranes. Activity with the depleted membranes was higher at all temperatures tested. Supplementation of deficient rats with a source of essential fatty acids (corn oil) restored V and apparent Km values to normal. Adenylate cyclase activity in the presence of fluoride, glucagon or glucagon plus GTP was significantly lower in the depleted plasma membranes.  相似文献

The present investigation was designed to study the uptake of67Cu when administered directly, into the portal vein, along with either functose or glucose, by the liver and extrahepatic tissues. Following weaning, male Sprague-Dawley rats were fed for 3 wk either commercial laboratory ration (chow) or semipurified diets deficient in Cu (0.6 ppm) or supplemented with Cu (6.0 ppm) and containing 62% carbohydrate as either fructuse or cornstarch. After an overnight fast, a single dose of rat plasma (0.1 mL) containing fructose or glucose extrinsically labeled with67Cu was injected directly into their portal vein. Although not always statistically significant, rats fed chow retained more radioactivity in the liver and several extrahepatic tissues when67Cu was administered with fructose than with glucose. Regardless of Cu status, rats fed diets containing fructose retained more radioactivity in extrahepatic tissues than rats fed starch. There was an increased uptake of67Cu by the liver, blood, muscle, and fat pad when fructose as compared to glucose was injected in combination with the isotope. These data strongly suggest that Cu requirements or utilization are greater when fructose is the main dietary carbohydrate. The results may also in part explain the reason for the increased severity of Cu deficiency in rats fed fructose.  相似文献

To study the mechanism of fatty infiltration in the liver due to added sulfur-containing amino acids to low casein diet, the effect of sulfur-containing amino acids and glutathione (GSH) on the incorporation of acetate-l-14C into lipid fractions were studied in liver slices from rats fed on 8% casein diet (Basal diet) with or without added methionine (Met).

The liver acetyl Co A carboxylase activities of rats on basal diet with or without added Met were similar.

Addition of Met, cystine or cysteine to the incubation medium had little effect on lipogenesis of slices. On addition of GSH to liver slices from rats fed on basal diet, lipid formation increased appreciably. On the other hand, addition of GSH to liver slices from rats fed on Met supplemented diet showed no accelerative effect on lipogenesis.

Addition of GSH to the incubation medium of liver slices from rats fed on basal diet tended to reduce the incorporation of acetate into the phospholipid fraction and to increase into the fatty acid fraction of liver slices.

The content of liver GSH was lower in rats on basal diet than in those on Met supplemented diet. The higher GSH level in rats on Met supplemented diet may be one factor causing fatty infiltration in the liver of these animals.  相似文献

Six experiments on growing rats were conducted to measure the nutritive value of diets based on cereals (wheat and barley, 1:1) and ethanol-grown yeast. Optimum protein provision in isonitrogenous diets (11.3% crude protein) determined on the basis of live weight gain, feed efficiency and protein efficiency ratio was found to be 60 and 40% for cereal and yeast protein, respectively, corresponding to a diet containing 84.5% cereals and 13.2% yeast (Experiments 1 and 2). The addition of single amino acids (l-lysine, 0.05–0.15%; dl-methionine, 0.1–0.3%) or their combinations (l-lysine + dl-methionine, 0.05 + 0.1%; 0.1 + 0.1%; 0.05 + 0.2%; and 0.1 + 0.2%) to the diet containing 13.2% yeast had no significant effect on either growth rate or feed efficiency (Experiments 3 and 4). A non-fortified diet alone proved to be comparable in nutritive value to a standard diet containing high-quality protein components.The diet with yeast providing 20% and cereals providing 80% of the total protein (5.55% yeast, 92.15% cereals) with combined supplement of amino acids (dl-methionine 0.2% + l-lysine 0.2% + l-isoleucine 0.1%; Experiment 5) promoted growth rate and feed efficiency similar to the standard diet, in spite of the fact that the experimental diet protein-content was about 3% lower. In a separate experiment (Experiment 6), similar results were achieved with the supplement of l-lysine alone (0.15% of the diet).As suggested by the results of model experiments on rats, the nutritive value of diets based on cereals and ethanol-grown yeast, possibly supplemented with amino acids, is comparable with that of diets containing animal protein components.  相似文献

Effects of dietary supplementation of orotic acid to a diet containing the casein protein were compared with diets containing egg protein, soy protein, or wheat gluten on lipid levels in the liver and serum and activities of ornithine carbamoyltransferase (OCT) and alanine aminotransferase in the serum of rats. We found that supplementation of orotic acid to each diet increased the contents of the liver total lipids, triacylglycerol, and phospholipids compared with those not supplemented. The contents of liver total lipids, triacylglycerol, cholesterol, and phospholipids in rats fed the casein diet were significantly higher than those of rats fed the other three diets when orotic acid was supplemented. The levels of triacylglycerol, cholesterol, and phospholipids in the serum of rats fed the casein diet were markedly decreased by addition of orotic acid. The supplementation of orotic acid significantly increased the activities of both serum OCT and alanine aminotransferase in rats fed the casein diet, but not in rats fed the other diets. In conclusion, liver lipid accumulation induced by dietary orotic acid depends on the type of dietary protein. The enhancement of serum OCT activity may result from liver lipid accumulation in rats fed the casein diet supplemented with orotic acid, demonstrating hepatic damage.  相似文献

Supplementation of high fat/cholesterol-enriched diets with polyoxypropylene-polyoxyethylene copolymers containing 90% hydrophobic constituents has been found to impair enteric secretion of chylomicrons, lower plasma levels of very low density (VLDL) and low density (LDL) lipoprotein cholesterol and prevent diet-induced hypercholesterolemia and atherosclerosis. These agents are known to be absorbed from the gastrointestinal tract and excreted in bile. In order to determine whether dietary supplementation with this group of hydrophobic poloxalenes influences hepatic secretion of triglyceride-rich lipoproteins, groups of rats were maintained for 21-34 days on either standard chow, semisynthetic diet containing 10.0% safflower oil/1.0% cholesterol, or each of the above diets supplemented with the hydrophobic poloxalene Pluronic L-81. At the end of the feeding period, newly secreted hepatic VLDL were isolated from 2-hr recirculating liver perfusates, quantitated, and characterized. Compared to perfusions in chow-fed rats, perfusion experiments in rats fed the high fat/cholesterol-enriched semisynthetic diet revealed a 3.1-fold increased net hepatic VLDL secretion rate; enrichment of secretory VLDL in cholesteryl esters and in C18:2 core lipid fatty acids; and a shift in the size distribution of secretory VLDL towards larger particles. When the 0.5% Pluronic L-81 was included in the high fat/cholesterol-enriched semisynthetic diet, the net hepatic VLDL secretion rate fell significantly and the physicochemical properties of secretory VLDL in these rats were found to resemble those of chow-fed animals. Supplementation of the chow diet with L-81 resulted in a significant fall in the net hepatic VLDL secretion rate from that observed in rats fed chow alone. Compared to rats fed chow alone, perfusate VLDL from rats fed each of the other experimental diets contained markedly lower amounts of both apoB molecular weight variants, as analyzed by gradient gel electrophoresis and densitometric gel scanning. Since previous studies have demonstrated that VLDL are the major cholesterol transport lipoproteins following fat/cholesterol feeding; a precursor-product relationship exists between fat/cholesterol-induced hepatic VLDL and plasma VLDL; such particles are capable of delivering cholesterol to the arterial wall; and dietary supplementation with hydrophobic poloxalenes prevents both the increase in plasma VLDL-cholesterol and diet-induced atherosclerosis, it is possible that dietary supplementation with hydrophobic poloxalenes may influence the atherogenic process through direct and/or indirect effects on hepatic VLDL transport.  相似文献
