Investigations on the meiotic behaviour of chromosomes in interspecific hybrids (2n=6x=42) between Hordeum lechleri (6x) and H. procerum (6x) and in their component haploids have been utilized to assess the nature of pairing and the extent of genome homology between the two species. In the F1 hybrids an average of 25 (60%) chromosomes associated at metaphase I, mostly as bivalents. A majority (60%) of the pollen mother cells (PMCs) in H. procerum haploids (2n=3x=21) displayed 21 univalents and even in the remainder, a maximum of two rod bivalents were formed resulting in an average of 0.52 bivalents per cell. In haploids of H. lechleri (2n=3x=21) however, 30% of chromosomes pair. The sum of the chromosomal associations in the component haploids represents only 17% of the complement, far below the observed frequency (60%) in the hybrids. Thus, the pairing displayed in hybrids between H. lechleri and H. procerum was mostly allosyndetic and suggestive of two genomes being common in these species.In haploid H. procerum 1/3 of the PMCs displayed a tripolar organisation of chromosomes leading to triad and hexad formation after divisions I and II respectively. The significance of hexad formation in the trihaploid H. procerum and a possible suppression of homoeologous pairing in H. procerum haploids are discussed.  相似文献

4′,5′-Unsaturated nucleosides are obtained by the action of 1,5-diazabicyclo-[5.4.0]undec-5-ene on N1- and N3-(methyl 2,3,4-tri-O-acetyl-β-d-glucopyranosyluronate)-5-fluorouracil. The 2H1 conformation of N1- and N3-(methyl 4-deoxy-α-l-threo-hex-4-enopyranosyluronate)-5-fluorouracil has been established by 1H-n.m.r. and c.d. methods. Interaction of the heterocyclic base and the double bond of the sugar moiety is demonstrated.  相似文献

The thermodynamic stability of self-complementary oligoribonucleotides containing N6-isopentenyladenosine (i(6)A) or N6-isopentanyladenosine (p(6)A) was determined. The base pairs i(6)A.U and p(6)A.U were placed in either an internal (separated and tandem) and a terminal position within the duplex, or unpaired i(6)A and p(6)A as a 3'-dangling ends. The thermal unfolding of the oligomers was determined by means of UV melting profiles and the thermodynamic parameters: enthalpy (DeltaH degrees ), entropy (DeltaS degrees) and free energy (DeltaG degrees (37)) as well as the melting temperature (T(m)) were calculated. Both modified nucleosides destabilized the duplexes, however, the effect depended on the position of the modified adenosine within the duplex. The similarity of the behavior of oligomers containing i(6)A and p(6)A suggests a negligible effect of the double bond on the thermal stability. The largest destabilization was observed when derivatives of adenosine were placed in an internal position. The effect of 3'-dangling ends suggests that the presence of the N6-isopentenyl- or N6-isopentanyl substitutent affects hydrogen bonding rather than stacking within duplex.  相似文献

Enzyme-linked immunosorbent assays (ELISAs) were developed for determination of N 6-benzyladenosine, N 6-(meta-hydroxybenzyl)adenosine, and structurally related cytokinins. The use of the ELISAs allowed detection over the range of 0.05–70 pmol for N 6-benzyladenine and 0.01–20 pmol for the N 6-(meta-hydroxybenzyl)adenine cytokinins. Polyclonal antibodies used in the assays were specific for N 6-benzyladenine and N 6-(meta-hydroxybenzyl)adenine and their corresponding N 9-substituted derivatives. By the use of internal standardization, dilution assays, authentic [2-3H]cytokinin recovery markers, and immunohistograms, the ELISAs have been shown to be applicable for the estimation of N 6-benzyladenine and N 6-(meta-hydroxybenzyl)adenine-type cytokinins in plant tissues. For the analysis of cytokinins in the tissues of young poplar leaves and Solarium teratoma shoot culture, the extracts were fractionated by high performance liquid chromatography (HPLC) and the fractions analyzed by ELISAs. The synthesis and biological evaluation of novel N(6)-substituted adenosine derivatives is reported. The first series of compounds was obtained using an established procedure for the nucleophilic substitution of a 1-(6-chloro-purin-9-yl)-beta-D-1-deoxy-ribofuranose with various amines. In addition, attachment of two different amino-functionalised spacer arms at the N(6)-position of adenosine enabled derivatisation by an innovative polymer-assisted protocol. Thus, we were able to prepare three series of substituted derivatives that displayed activity versus the multiresistant Plasmodium falciparum strain Dd2 in cell culture experiments.  相似文献

A series of novel 3'-amido-3'-deoxy-N(6)-(1-naphthylmethyl)adenosines was synthesized applying a polymer-assisted solution phase (PASP) protocol and was tested for anti-malarial activity versus the Dd2 strain of Plasmodium falciparum. Further, this series and 62 adenosine derivatives were analyzed regarding 1-deoxy-d-xylulose 5-phosphate (DOXP) reductoisomerase inhibition. Biological evaluations revealed that the investigated 3',N(6)-disubstituted adenosine derivatives displayed moderate but significant activity against the P. falciparum parasite in the low-micromolar range. On the molecular level, DOXP reductoisomerase utilizing an adenosyl-containing substrate was identified as a promising metabolic target for ligands of adenosine binding motifs.  相似文献

In this paper we describe the reduced recombination and distorted segregation in an interspecific hybrid between Solanum tuberosum and Solanum spegazzinii. To study these phenomena, a cross was made between a (di)haploid S. tuberosum, used as a female parent, and a diploid wild potato species, S. spegazzinii, used as a male parent. Next, a backcross (BC) population was made with F1 genotype 38 that was backcrossed to S. tuberosum. In the backcross, S. tuberosum was used as the male parent. RFLP linkage maps were made using the F1 and the BC populations, yielding linkage maps of the interspecific hybrid, S. spegazzinii, and S. tuberosum from which male and female linkage maps could be constructed. The computer program JOINMAP was used to construct and combine the separate linkage maps. Subsequently, the separate linkage maps were compared with each other, and reduced recombination was observed in the linkage maps of the male S. tuberosum and the interspecific hybrid. The reason for this reduced recombination is discussed. Another common feature in linkage maps is the observation of distorted segregation. The distorted segregation of alleles from the interspecific hybrid was studied in more detail in the BC population. Most of the distortion was probably caused by gamete selection, but for 3 loci, on chromosomes 2, 3, and 4, we found evidence for the presence of a strong selection force acting at the zygote level against homozygous genotypes.  相似文献

We report an improved synthesis of N 6-(6-aminohexyl)FAD (1) using an efficient one-pot conversion of inosine to the N-trifluoroacetyl protected N 6-(6-aminohexyl)adenosine 3. The N(6)-adenine DNA-methyltransferase was isolated from the vacuolar vesicle fraction of wheat coleoptiles. In the presence of S-adenosyl-L-methionine the enzyme de novo methylates the first adenine residue in the TGATCA sequence in the single- or double-stranded DNA substrates but it prefers single-stranded structures. Wheat adenine DNA-methyltransferase (wadmtase) is a Mg(2+)- or Ca(2+)-dependent enzyme with a maximum activity at pH 7.5-8.0. Wadmtase seems to be responsible for mitochondrial DNA modification that might be involved in the regulation of replication of mitochondria in plants.  相似文献

Much of the variation in hexaploid Atriplex tridentata appears to have come from tetraploid A. canescens by introgression. Among the recombinations, three types appear to have become established as new, adaptive, hexaploid derivatives. One of these is a robust, woody form near Knolls, Utah, another is a widespread, low-growing, shrubby form in Lander County, Nevada, the other is an upright bushy, canescens-like form near Grantsville, Utah.  相似文献

Despite the presence of intraspecific polyploidy (2x, 4x, 5x and 6x) in Agrimonia eupatoria, origin of these cytotypes has never been addressed adequately. The aim of the present study was to record the original chromosome counts and characterize chromosomal pairing during meiosis and microsporogenesis in the 5x cytotype, and discussing the hypothesis regarding the possible origin of polyploid cytotypes (4x, 5x and 6x) in the species. The geographical distribution pattern of cytotypes in the Indian Himalayas and elsewhere has also been analyzed. The present meiotic analysis revealed three chromosomes counts, the tetraploid (2n?=?4x?=?56), the pentaploid (2n?=?5x?=?70) and the hexaploid (2n?=?6x?=?84) cytotypes based on x?=?14. Meiotic course was perfectly normal in the 4x and 6x cytotypes resulting into high pollen fertility (94–100 %). Meiotic course in the imbalanced 5x cytotype has been found to be irregular characterized by the presence of high frequency of univalents at diakinesis and metaphase-I. Abnormal meiotic course contributed towards high pollen sterility (74–88 %). Even the apparently fertile/stained pollen grains were of irregular shape and of heterogeneous sizes. Meiotic behaviour of the 5x cytotype is like typical of allopolyploid. Individuals of 5x cytotype did not produce seeds and propagate vegetatively (root suckers) while 4x and 6x cytotypes exploited sexual (seeds) as well as vegetative means for propagation. Chromosomal pairing in pentaploid cytotype is like typical of an allopolyploid and we assume that it might have originated owing to natural inter-cytotype hybridization between 4x and 6x cytotypes in a mixed population. Analysis of geographical distribution pattern of cytotypes shows that Indian Himalayas represent the most cytotype-diverse region for A. eupatoria with the existence of all the four cytotypes (2x, 4x, 5x, 6x). This shows the dynamic nature of the species at chromosomal level in this part of the world.  相似文献
