Sieve tube sap exuded from the cut hypocotyl of castor bean seedlings (Ricinus communis L.) was found to contain 0.2–0.5 mmol m?3abscisic acid (ABA). The ABA concentration in the sieve tube sap always exceeded that in root pressure exudate under a wide range of water supply. Exudation of sieve tube sap from the cut hypocotyls caused water loss, and this induced ‘water shortage’ in the cotyledons which resulted in the ABA concentration in the cotyledons increasing by 3-fold and that in the sieve tube sap increasing by up to 50-fold within 7h. The wounded surface of the cut hypocotyl was not responsible for the ABA increase. Incubation of the cotyledons of endosperm-free seedlings in various ABA concentrations (up to 100 mmol m?3) increased the ABA concentration in sieve tube sap. The concomitant increase in ABA, both in cotyledons and in sieve tube sap, had no effect on the phloem loading of sucrose, K+ and Mg2+ within the experimental period, i.e. up to 10h. It can be concluded that (i) the phloem is an important transport path for ABA, (ii) water stress at the phloem loading sites elevates phloem-mobile ABA, which may then serve as a water stress signal for sinks, for example stem and roots (not only for stomata), and (iii) the ABA concentration of cells next to or in the phloem is more important than the average ABA content in the whole cotyledon for determining the ABA concentration in sieve tube sap.  相似文献

Sieve tube sap was obtained from Tanacetum by aphid stylectomy and from Ricinus after apical bud decapitation. The amino acids in sieve tube sap were analyzed and compared with those from leaves. Arginine and lysine accumulated in the sieve tube sap of Tanacetum more than 10‐fold compared to the leaf extracts and they were, together with asparagine and serine, preferably selected into the sieve tube sap, whereas glycine, methionine/tryptophan and γ‐amino butyric acid were partially or completely excluded. The two basic amino acids also showed a close covariation in sieve tube sap. The acidic amino acids also grouped together, but antagonistic to the other amino acids. The accumulation ratios between sieve tube sap and leaf extracts were smaller in Ricinus than in Tanacetum. Arginine, histidine, lysine and glutamine were enriched and preferentially loaded into the phloem, together with isoleucine and valine. In contrast, glycine and methionine/tryptophan were partially and γ‐amino butyric acid almost completely excluded from sieve tube sap. The covariation analysis grouped arginine together with several neutral amino acids. The acidic amino acids were loaded under competition with neutral amino acids. It is concluded from comparison with the substrate specificities of already characterized plant amino acid transporters, that an AtCAT1‐like transporter functions in phloem loading of basic amino acids, whereas a transporter like AtGAT1 is absent in phloem. Although Tanacetum and Ricinus have different minor vein architecture, their phloem loading specificities for amino acids are relatively similar.  相似文献

Chemical Forms of Manganese and Zinc in Phloem Exudates   总被引:3,自引:0,他引:3  
Investigations were performed to study the chemical form in which manganese and zinc are transported in sieve tubes. As a test plant Ricinus communis was used. From this plant phloem sap can be obtained in a rather pure state. The plants received labelled manganese or zinc. In the experiments on manganese translocation a radioactive phosphorus isotope was added to the nutrient solution to test if complex compounds of the cation contain also phosphorus. Components of the phloem exudate were determined by means of physicochemical separation methods. Almost all zinc but only a part of the manganese were bound to organic compounds. The major part of the manganese was in ionic form. The molecular weight of the complexing com-pound(s) of manganese was estimated to be between 1000 and 5000 and of zinc between 1000 and 1500. The complexes probably contain some phosphorus. The charge of the zinc complex is negative.  相似文献

Pure phloem sap was collected from insects feeding on rice (Oryza sativa L.) leaves by a laser technique similar to the aphid stylet technique. Rapid circulation of nitrogen in the sieve tubes was demonstrated directly using 15N as a tracer. Application to the roots of the metabolic inhibitors of amino acids, aminooxyacetate and methioninesulfoximine, changed the amino acid composition in the sieve tubes. Feeding methionine to leaf tips resulted in its bulk transfer into the sieve tubes. In vitro experiments confirmed the existence of protein kinases in the pure rice phloem sap. The phosphorylation status of the sieve tube sap proteins was affected by the light regime. The possibility that changes in chemical composition or protein modification such as phosphorylation in the sieve tubes might affect plant growth are discussed.Analysis of pure phloem sap collected from rice plants by insect laser technique has shown dynamic changes in the chemical composition and the quality of proteins in the sap.  相似文献

Reports about diurnal changes of assimilates in phloem sap are controversial. We determined the diurnal changes of sucrose and amino acid concentrations and fluxes in exudates from cut aphid stylets on tansy leaves (Tanacetum vulgare), and sucrose, amino acid and K(+) concentrations and fluxes in bleeding sap of castor bean pedicel (Ricinus communis). Approximately half of the tansy sieve tubes exhibited a diurnal cycle of sucrose concentrations and fluxes in phloem sap. Data from many tansy plants indicated an increased sucrose flux in the phloem during daytime in case of low N-nutrition, not at high N-nutrition. The sucrose concentration in phloem sap of young Ricinus plants changed marginally between day and night, whereas the sucrose flux increased 1.5-fold during daytime (but not in old Ricinus plants). The amino acid concentrations and fluxes in tansy sieve tubes exhibited a similar diurnal cycle as the sucrose concentrations and fluxes, including their dependence on N-nutrition. The amino acid fluxes, but not the concentrations, in phloem sap of Ricinus were higher at daytime. The sucrose/amino acid ratio showed no diurnal cycle neither in tansy nor in Ricinus. The K(+)-concentrations in phloem sap of Ricinus, but not the K(+) fluxes, decreased slightly during daytime and the sucrose/K(+)-ratio increased. In conclusion, a diurnal cycle was observed in sucrose, amino acid and K(+) fluxes, but not necessarily in concentrations of these assimilates. Because of the large variations between different sieve tubes and different plants, the nutrient delivery to sink tissues is not homeostatic over time.  相似文献

Phloem sieve elements have shut‐off mechanisms that prevent loss of nutrient‐rich phloem sap when the phloem is damaged. Some phloem proteins such as the proteins that form forisomes in legume sieve elements are one such mechanism and in response to damage, they instantly form occlusions that stop the flow of sap. It has long been hypothesized that one function of phloem proteins is defence against phloem sap‐feeding insects such as aphids. This study provides the first experimental evidence that aphid feeding can induce phloem protein occlusion and that the aphid‐induced occlusions inhibit phloem sap ingestion. The great majority of phloem penetrations in Vicia faba by the generalist aphids Myzus persicae and Macrosiphum euphorbiae triggered forisome occlusion and the aphids eventually withdrew their stylets without ingesting phloem sap. This contrasts starkly with a previous study on the legume‐specialist aphid, Acyrthosiphon pisum, where penetration of faba bean sieve elements did not trigger forisome occlusion and the aphids readily ingested phloem sap. Next, forisome occlusion was demonstrated to be the cause of failed phloem ingestion attempts by M. persicae: when occlusion was inhibited by the calcium channel blocker lanthanum, M. persicae readily ingested faba bean phloem sap.  相似文献

The non-proteinogenic amino acid nicotianamine (NA) is ubiquitous among plants. In meristematic tissues it reaches concentrations of about 400mol (g fresh weight)–1. NA forms complexes, among others, with the metal micronutrients (MN) copper, zinc, iron and manganese (logK MeNA 18.6-8.8). Calculations of the dissociation curves of the metal-NA complexes based on the complex formation constants and on the acid dissociation constants of NA revealed their stability at the neutral or weak alkaline pH of cytoplasm and sieve tube sap. For the Mn-NA complex, dissociation begins at about pH 6.5, for all others dissociation occurs at more acid pHs. Thus, metal-NA complexes could theoretically persist also in the apoplasm and in xylem sap. The octanol water partition coefficient of NA is about 1 and those of its metal complexes are in the range of 0.3–0.4. The reason for this shift is perhaps the negative charge of the complexes. The higher lipophilicity of the free NA indicates that the NA supply to sites of requirement is faster than the removal of the complexes as long as membranes are an integral part of the transport paths. Changing phloem transport rates of MN-NA complexes by manipulation of the cotyledon apoplasm of Ricinus commuais L. suggest a competition of MN for NA at the site(s) of phloem loading. Thus, NA could control MN transport via phloem including recirculation.  相似文献

Investigations were performed to study the chemical form in which nickel and cobalt are transported in sieve tubes. Ricinus communis plants were used since they allow easy collection of relatively pure phloem sap. In the experiments labelled nickel or cobalt were given to the nutrient solution. The tapped phloem exudate was separated by gel filtration on Sephadex or by paper electrophoresis. Nickel and cobalt were bound to complexing organic compounds with a molecular weight in the range of 1000—5000. The overall charge of the nickel complex was negative, and the bulk of cobalt also appeared to complex to a compound with negative overall charge.  相似文献

In yeast, eIF5A, in combination with eEF2, functions at the translation step, during the protein elongation cycle. This result is of significance with respect to functioning of the enucleate sieve tube system, as eIF5A was recently detected in Cucurbita maxima (pumpkin) phloem sap. In the present study, we further characterized four CmeIF5A isoforms, encoding three proteins, all of which were present in the phloem sap. Although hypusination of CmeIF5A was not necessary for entry into the sieve elements, this unique post‐translational modification was necessary for RNA binding. The two enzymes required for hypusination were detected in pumpkin phloem sap, where presumably this modification takes place. A combination of gel‐filtration chromatography and protein overlay assays demonstrated that, as in yeast, CmeIF5A interacts with phloem proteins, like eEF2, known to be involved in protein synthesis. These findings are discussed in terms of a potential role for eIF5A in regulating protein synthesis within the enucleate sieve tube system of the angiosperms.  相似文献

To test for the existence of an apoplastic unloading/reloadingstep in phloem translocation, as envisaged in the relay hypothesisof phloem transport, isotopic trapping experiments were performedon Ricinus comunis L. var. Gibsonii. A CO2 buffer system wasused to supply 14CO2 at constant partial pressure and constantspecific activity to a photosynthesizing leaf. The subtendinginternode was perfused with solutions of sugars or of mannitoland transolation of 14C past the perfused zone was monitoredby the collection of phloem exudate. Trapping of activity inthe perfusate was enhanced by the presence of sugars as wouldbe expected with an unloading/reloading process. However, therewas no evidence that introduction of the unlabelled sugars tothe apoplast also reduced activity in the phloem exudate. Moreover,the rate of loss of activity to the perfusing solutions representedonly 1% of the rate of appearance in the exudation. It is suggestedthat the trapping results may reflect an unloading of tracerfrom the phloem associated with a subsequent reloading by adjacenttissues rather than by the sieve tubes. To investigate the length of sieve tube continuity in Ricinus,a horizontal incision was made to the bark and the rate of exudationof phloem sap was monitored. Successive circumferential cutswere made above the exuding incision and progressively closerto it. In general, a girdling incision produced a transientdecrease in the rate of change of exudation rate (i.e. the firstderivative became more negative/less positive). The magnitudeof this response rose with exudation rate and fell with thedistance at which a girdling cut was made. Fitting an appropriatemodel yielded an estimate for contributory length of 69 ±6 cm. This was comparable with the distance of the initial tangentialincision from the stem apex, suggesting a continuous sieve tubesystem in Ricinus. A similar investigation on the petiole yieldedan estimate of around 7.0 cm. This lower estimate for contributorylength is believed to reflect a rapid sealing process that limitsthe distance of propagation of turgor-release rather than alimited length of sieve tube continuity. The results of this investigation do not support the relay hypothesisof phloem transport. Rather they suggest a continuous sievetube system which has a distributed capacity to load and unloadsolutes, and which may exhibit a sealing response when injured. The biosynthesis of 14C-IAA from 14C-tryptophan applied to abraded leaves of Ricinus communis and its subsequent export through the phloem were studied. Phloem sap was collected at intervals from incisions made in the stem below the IAA fed leaf. Any upward movement of label through the phloem or downward movement of phloem mobile compounds from leaves above the treated one were restricted by bark-ringing the plants.TLC and HPLC analyses of the collected sap indicate that some conversion of 14C-tryptophan to 14C-IAA had occurred. Subsequent GC-MS analysis of the HPLC purified samples of phloem sap revealed high levels of endogenous IAA transported from the fed leaf. The high ratio of unlabelled/labelled IAA in the phloem sap makes unequivocal confirmation by GC-MS of the predicted biosynthesis of 14C-IAA impossible. It is postulated that IAA is synthesised from tryptophan in mature leaves and exported to developing sink tissues with the flow of photoassimilates in the phloem.  相似文献

Abstract. Ricinus communis plants have an unusually high capacity to exude considerable quantities of phloem sap from bark incisions. We have used Ricinus as an experimental system to study different aspects of sap exudation. Dimensional changes in the bark, monitored by a displacement transducer, showed that pressure release in the sieve tubes was accompanied by elastic shrinkage. The rate of exudation was also controlled by the degree of pressurization and elastic properties of the sieve tubes. A displacement transducer was used to measure the elastic modulus (ɛ) of phloem samples by immersing them in a range of different osmotica. The cells had a low elastic modulus (ɛ= 1.62 ± 0.41 MPa at full turgor). ɛ of phloem tissue in massage pretreated bark, from which exudation was enhanced, was not significantly different from that of unmassaged bark in contrast with the suggestion of Lee (1981). However, anatomical studies showed that massage pretreatment has a stimulating influence on the cambial cell division, which increased the phloem tissue cross-section up to 160%. The newly-formed sieve tubes were 'spliced' into existing ones in the unmassaged zone to re-establish vascular continuity. Plants with a greater capacity to exude phloem sap from a given stem location had a greater cross-sectional area of sieve tubes in the vicinity.
The significance of observations with respect to other sap exudation phenomenon is discussed. The importance of the present work in understanding the technique of palm tapping, on which the palm sugar industry depends, is also considered.  相似文献

The metal micronutrients (MN) copper, iron, manganese, and zinc are transported via the phloem in the course of remobilization and circulation. The extent of these processes and transport species are still largely unknown. The Ricinus seedling was used to study the transport of these metal micronutrients as well as their interactions with the plant-endogenous chelator nicotianamine (NA) by daily measurements of the concentrations in the seedling parts and in the sieve tube sap obtained from a cut at the hypocotyl hook. The concentrations of these micronutrients in the phloem exudate decreased slightly from day 4 to day 8 of seedling development. Maximum values at day 4 were 65 μM for Zn, 63 μM for Fe, 27 μM for Cu, and 12 μM for Mn. The phloem transport rates reached maxima of 0.12 nmol cm?2h?1 for Zn and Fe at days 6 and 7, corresponding to the maximum exudation rates. The magnitude of these transport rates were in agreement with the net translocation rates estimated by analyses of the concentrations in the individual seedling parts. The NA content of the seedlings increased from day 0 (seed before sowing) until day 8, from 16 nmol to 474 nmol, which corresponds to an average net synthesis rate of about 100 nmol day?1 between the days 4 and 8. The NA:MN ratio was constant at 0.5 in the seedlings within this period. The NA concentrations and the sum of the concentrations of all four micronutrients in the sieve tube sap showed a constant ratio of 1.25 over the entire experimental period. Thus, both complex partners were subject to a cotransport in the phloem. Removal of the supplying endosperm led to a decrease in MN and NA concentrations in the sieve tube sap to about 80% while an average excess of NA of 1.1 was maintained. Since the concentrations of other amino acids, also possible chelators of metal micronutrients, fall to about 10% after removal of the endosperm, their role seems to be negligible as vehicles of MN transport in the phloem. Thus it is suggested that the divalent micronutrients considered in this study are loaded and maybe transported as NA complexes.  相似文献

Resistance of the melon line TGR‐1551 to the aphid Aphis gossypii is based on preventing aphids from ingesting phloem sap. In electrical penetration graphs (EPGs), this resistance has been characterized with A. gossypii showing unusually long phloem salivation periods (waveform E1) mostly followed by pathway activities (waveform C) or if followed by phloem ingestion (waveform E2), ingestion was not sustained for more than 10 min. Stylectomy with aphids on susceptible and resistant plants was performed during EPG recording while the stylet tips were phloem inserted. This was followed by dissection of the penetrated leaf section, plant tissue fixation, resin embedding, and ultrathin sectioning for transmission electron microscopic observation in order to study the resistance mechanism in the TGR. The most obvious aspect appeared to be the coagulation of phloem proteins inside the stylet canals and the punctured sieve elements. Stylets of 5 aphids per genotype were amputated during sieve element (SE) salivation (E1) and SE ingestion (E2). Cross‐sections of stylet bundles in susceptible melon plants showed that the contents of the stylet canals were totally clear and also, no coagulated phloem proteins occurred in their punctured sieve elements. In contrast, electron‐dense coagulations were found in both locations in the resistant plants. Due to calcium binding, aphid saliva has been hypothesized to play an essential role in preventing/suppressing such coagulations that cause occlusion of sieves plate and in the food canal of the aphid's stylets. Doubts about this role of E1 salivation are discussed on the basis of our results.  相似文献

It is commonly believed that the synthetic triazole growth regulator paclobutrazol (PAC) is exclusively xylem mobile within plants. By contrast, the triazole amitrole and many natural growth regulators are phloem mobile. This raises some doubt as to whether PAC must necessarily be exclusively xylem mobile. PAC was introduced into castor oil plants (Ricinus communis L.) through their hollow petioles. PAC was detected in xylem and phloem sap collected above the point of introduction but not in xylem sap below this point. This finding shows that PAC is not exclusively xylem mobile as believed previously. These results also raise the possibility of introducing PAC into plants in a different way so that it is carried by both the xylem and phloem and thus optimizing its effectiveness. Aphid salivation in sieve elements and phloem sap ingestion were linked to waveforms in the Electrical Penetration Graph (EPG). Non-viruliferousRhopalosiphum padi (L.) (Hemiptera, Aphididae) on barley yellow dwarf virus (BYDV) infected wheat could acquire the virus, which was used as an indication for phloem sap ingestion, whereas virus inoculation by viruliferous aphids on healthy plants was associated with salivation in sieve elements or other phloem cells. Probing was monitored and the waveforms recorded were related to ELISA results of test plants. The EPG patterns A, B, and C are indicative of the stylet pathway phase, whereas patterns E1 and E2 reflect the phloem (sieve element) phase with an unknown activity (E1) or with ingestion and concurrent salivation (E2). Aphids showing pathway and E1 rarely acquired virus, suggesting that little or no phloem sap ingestion can occur during these patterns, whereas those showing additionally pattern E2 did so substantially, indicating phloem sap ingestion. The main pattern related to virus inoculation was E1, although some aphids were able to inoculate plants during pathway. Pattern E1 clearly reflects the most important salivation into sieve elements. Pattern E2 had no clear contribution to virus inoculation, supporting the present hypothesis that during this pattern the saliva is mixed with the phloem sap in the single canal at the stylet tips and ingested immediately, without reaching the plant tissue. Sustained sap ingestion did not affect virus inoculation. So, BYDV inoculation mainly occurs during the first period of a sieve element puncture which is always formed by E1. Implications on persistent virus transmission are discussed.  相似文献

A theory is presented to explain the phloem mobility of certain systemic xenobiotics that are not weak acids. It is shown that there is a theoretically optimum permeability that permits optimum circulation through the symplasm and apoplast (including the phloem and xylem) of Solanum tuberosum plants. The optimum permeability is large enough to permit substantial passive permeation into sieve cells in the source leaf and yet is small enough to permit phloem transport with some retention. The optimum permeability is a function of the velocity of sap flow in sieve tubes, the radius of the sieve tube, the over-all length of the plant, and the length of the carbohydrate and xenobiotic sources. It is argued that the nematicide, oxamyl, is near the optimum permeability under some experimental conditions. It is shown that depending on the strength of the carbohydrate sink in roots or growth points and depending on the permeability of the xenobiotic, there can be passive accumulation of xenobiotics in the sieve tubes in the carbohydrate sink regions.  相似文献

Amino acid and sucrose contents were analyzed in the chloroplastic, cytosolic, and vacuolar compartments and in the phloem sap of illuminated spinach leaves (Spinacia oleracea L.). The determination of subcellular metabolite distribution was carried out by nonaqueous fractionation of frozen and lyophilized leaf material using a novel three-compartment calculation method. The phloem sap was collected by aphid stylets which had been severed by a laser beam. Subcellular analysis revealed that the amino acids found in leaves are located mainly in the chloroplast stroma and in the cytosol, the sum of their concentrations amounting to 151 and 121 millimolar, respectively, whereas the amino acid concentrations in the vacuole are one order of magnitude lower. The amino acid concentrations in the phloem sap are found to be not very different from the cytosolic concentrations, whereas the sieve tube concentration of sucrose is found to be one order of magnitude higher than in the cytosol. It is concluded that the phloem loading results in a preferential extraction of sucrose from the source cells.  相似文献

Histochemical localization of ATPase was carried out on phloemtissues from vegetative and reproductive sinks of Ricinus communis,using lead precipitation procedures. Reaction products werelocalized mainly at the plasma membrane of the sieve elements,companion cells and phloem parenchyma cells. Activity was alsopresent in plasmodesmata, the tonoplast of companion cells anddispersed P-protein within the sieve element lumen. The resultsare discussed in relation to the possible involvement of a plasmamembrane ATPase in apoplastic and symplastic unloading fromthe phloem conducting tissues. Aphididae feed upon the plant sieve elements (SE), where they ingest sugars, nitrogen compounds and other nutrients. For ingestion, aphid stylets penetrate SE, and because of the high hydrostatic pressure in SE, phloem sap exudes out into the stylets. Severing stylets to sample phloem exudates (i.e. stylectomy) has been used extensively for the study of phloem contents. Alternative sampling techniques are spontaneous exudation upon wounding that only works in a few plant species, and the popular EDTA-facilitated exudation technique. These approaches have allowed fundamental advances on the understanding of phloem sap composition and sieve tube physiology, which are surveyed in this review. A more complete picture of metabolites, ions, proteins and RNAs present in phloem sap is now available, which has provided large evidence for the phloem role as a signalling network in addition to its primary role in partitioning of photo-assimilates. Thus, phloem sap sampling methods can have remarkable applications to analyse plant nutrition, physiology and defence responses. Since aphid behaviour is suspected to be affected by phloem sap quality, attempts to manipulate phloem sap content were recently undertaken based on deregulation in mutant plants of genes controlling amino acid or sugar content of phloem sap. This opens up new strategies to control aphid settlement on a plant host.  相似文献
