The objective was to determine reproductive performance following AI in beef heifers given estradiol to synchronize ovarian follicular wave emergence and estradiol or GnRH to synchronize ovulation in a two-dose PGF-based protocol. In Experiment 1, 561 cycling (confirmed by ultrasonography), Angus heifers received 500 microg cloprostenol, i.m. (PGF) twice, 14 days apart (days 0 and 14) and were equally allocated to four groups in a 2 x 2 factorial design. On Day 7, heifers received either 2 mg estradiol benzoate (EB) and 50 mg progesterone (P), i.m. in oil (EBP group) or no treatment (NT group). Half the heifers in each group received 1mg EB, i.m. in oil on Day 15 (24h after the second PGF treatment) with TAI 28 h later (52 h after PGF), and the other half received 100 microg GnRH, i.m. on Day 17 (72 h after PGF) concurrent with TAI. All heifers were observed for estrus twice daily from days 13 to 17; those detected in estrus more than 16 h before scheduled TAI were inseminated 4-16 h later and considered nonpregnant to TAI. Overall pregnancy rate (approximately 35 days after AI) was higher in heifers that received EBP than those that did not (61.6% versus 48.2%, respectively; P < 0.002); but was lower in heifers that received EB after PGF than those that received GnRH (50.0% versus 59.8%; P < 0.02). Although estrus was detected prior to TAI in 77 of 279 heifers (27.6%) treated with EBP (presumably due to induced luteolysis), they were inseminated and 53.2% became pregnant. Overall pregnancy rates were 51.4, 68.3, 45.0, and 55.0% in the NT/GnRH, EBP/GnRH, NT/EB, and EBP/EB groups, respectively (P < 0.05). In Experiment 2, 401 cycling, Angus heifers were used. The design was identical to Experiment 1, except that 1.5mg estradiol-17beta (E-17beta) plus 50mg progesterone (E-17betaP) and 1mg E-17beta were used in lieu of EBP and EB, respectively. All heifers receiving E-17beta 24h after the second injection of PGF (NT/E-17beta and E-17betaP/E-17beta) were TAI 28 h later without estrus detection, i.e. 52 h after PGF. Heifers in the other two groups received 100 microg GnRH, i.m. 72 h after PGF and were concurrently TAI; heifers in these two groups that were detected in estrus prior to this time were inseminated 4-12h later and considered nonpregnant to TAI. Estrus rate during the first 72 h after the second PGF treatment was higher (P < 0.05) in the E-17betaP/GnRH group (45.0%; n = 100) than in the NT/GnRH group (16.0%; n = 100), but conception rate following estrus detection and AI was not different (mean, 57.4%; P = 0.50). Overall pregnancy rate was not significantly different among groups (mean, 46.9%; P = 0.32). In summary, the use of EB or E-17beta to synchronize follicular wave emergence and estradiol or GnRH to synchronize ovulation in a two-dose, PGF-based protocol resulted in acceptable fertility to TAI. However, when 2mg EB was used to synchronize follicular wave emergence, early estrus occurred in approximately 28% of heifers, necessitating additional estrus detection. A combination of estrus detection and timed-AI in a two-dose PGF protocol resulted in highly acceptable pregnancy rates.  相似文献

Four experiments were conducted to investigate modifications to gonadotropin releasing hormone (GnRH)-based fixed-time Al protocols in beef cattle. In Experiment 1, the effect of reducing the interval from GnRH treatment to prostaglandin (PGF) was examined. Lactating beef cows (n = 111) were given 100 mg gonadorelin (GnRH) on Day 0 (start of treatment) and either 500 microg cloprostenol (PGF) on Day 6 with Al and 100 microg GnRH 60 h later, or PGF on Day 7 with Al and GnRH 48 h later (6- or 7-day Co-Synch regimens). Pregnancy rates were 32/61 (53.3%) versus 26/50 (52.0%), respectively (P = 0.96). In Experiment 2. cattle (n = 196) were synchronized with a 7-day Co-Synch regimen and received either no further treatment or a CIDR-B device (Days 0-7). Pregnancy rates were 32/71 (45.1%) versus 33/77 (42.9%) in cows (P < 0.8), and 9/23 (39.1 %) versus 17/25 (68.0%) in heifers (P < 0.05). In Experiment 3, 49 beef heifers were randomly assigned to receive 12.5 mg pLH on Day 0, PGF on Day 7 and 12.5 mg of pLH on Day 9 with Al 12 h later (pLH Ovsynch), or similar treatment plus a CIDR-B device from Days 0 to 7 (pLH Ovsynch + CIDR-B), or 1 mg estradiol benzoate (EB) and 100 mg progesterone on Day 0, a CIDR-B device from Days 0 to 7 (EB/ P4 + CIDR-B), PGF on Day 7 (at the time of CIDR-B removal) and 1 mg i.m. EB on Day 8 with AI on Day 9 (52 h after PGF). Pregnancy rate in the EB/P4 + CIDR-B group (75.0%) was higher (P < 0.04) than in the pLH Ovsynch group (37.5%): the pLH Ovsynch + CIDR-B group was intermediate (64.7%). In Experiment 4, 266 non-lactating cows were allocated to a 7-day Co-Synch protocol (Co-Synch), a 7-day Co-Synch plus 0.6 mg per head per day melengestrol acetate (MGA) from Days 0 to 6 inclusive (Co-Synch + MGA) or MGA (Days 0-6) plus 2 mg EB and 50 mg progesterone on Day 0. 500 microg PGF on Day 7, 1 mg EB on Day 8 and fixed-time Al 28 h later (EB/ P4 + MGA). Pregnancy rates (P < 0.25) were 44.8% (39/87: Co-Synch), 47.8% (43/90; Co-Synch + MGA), and 60.7% (54/89: EB/P4 + MGA). In conclusion, a 6- or 7-day interval from GnRH to PGF in a Co-Synch regimen resulted in similar pregnancy rates in cows. The addition of a progestin to a Co-Synch or Ovsynch regimen significantly improved pregnancy rates in heifers but not in cows. Progestin-based regimens that included EB consistently resulted in high pregnancy rates to fixed-time Al.  相似文献

The objectives of this experiment were to compare estrous synchronization responses and AI pregnancy rates of beef heifers using protocols that included either CIDR or MGA as the progestin source. The hypotheses tested were that: (1) estrous synchronization responses after (a) progestin removal, and (b) PGF(2alpha); and, (2) AI pregnancy rates, do not differ between heifers synchronized with either progestin source. At the start of the experiment (Day 0) in both years, heifers were assigned randomly to receive, MGA supplement for 14 days (MGA-treated; n=79) or CIDR for 14 days (CIDR-treated; n=77). On Day 14 progestin was removed and heifers were observed for estrus up to and after PGF(2alpha) on Days 31 and 33 for CIDR-treated and MGA-treated heifers, respectively. Heifers that exhibited estrus within 60h after PGF(2alpha) were inseminated by AI 12h later; the remaining heifers were inseminated at 72h after PGF(2alpha) and given GnRH (100mug). More (P<0.05) CIDR-treated heifers exhibited estrus within 120h after progestin removal than MGA-treated heifers. Intervals to estrus after progestin removal were shorter (P<0.05) for CIDR-treated heifers than MGA-treated heifers. More (P<0.05) CIDR-treated heifers exhibited estrus and were inseminated within 60h after PGF(2alpha) than MGA-treated heifers. Pregnancy rates did not differ (P>0.10) between MGA-treated (66%) and CIDR-treated (62%) heifers. In conclusion, the use of CIDR as a progestin source in a 14-day progestin, PGF(2alpha), and timed AI and GnRH estrous synchronization protocol was as effective as the use of MGA to synchronize estrus and generate AI pregnancies in beef heifers.  相似文献

One aim of this study was to compare the reproductive performance of cows and heifers when resynchronizing returns to estrus for a second insemination by treating with an intravaginal progesterone-releasing device (IVD) for 7 or 8d when estradiol benzoate (EB) was administered at the start of treatment and again 24h after device removal. An additional aim was to document the pattern of onset and characteristics of estrus with each resynchrony treatment. Lactating cows in three herds were synchronized for a first estrus and AI by treatment with an IVD for 8d, starting on Day 0, cloprostenol (0.5 mg im) at device removal and EB at device insertion (2.0 mg im) and 24h after removal (1.0 mg im). Cows were resynchronized for a second estrus starting on Day 23 by reinsertion of IVDs for 7 (IVD-7-EB; n=449) or 8d (IVD-8-EB; n=445) with EB (1.0 mg im) administered at device insertion and 24h after removal. Cows were resynchronized for a third estrus by administration of EB (1.0 mg im) on Day 46, but subsequent treatments (no further treatment, reinsertion of CIDR or administration of EB on Day 55) varied among herds as part of separate studies. Maiden heifers (7-Day, n=68; 8-Day, n=69) were similarly treated as cows in a separate herd, but doses of EB were always 1.0 mg im at device insertion and 0.75 mg im 24h after removal. Heifers were not resynchronized for a third estrus. Cattle were inseminated on detection of estrus at each synchronized estrus. Cumulative pregnancy rates 4 week (66.0%, 276/418 versus 59.1%, 247/418) and 7 week (72.7%, 304/418 versus 67.7%, 283/418) after the start of AI were greater (P<0.05) in the IVD-7-EB cows compared to the IVD-8-EB cows, respectively; this was associated with a 9% increase in conception rates at the second estrus (P=0.051) in the IVD-7-EB cows. Treatment did not significantly affect reproductive performance in heifers. Characteristics of estrus measured with radiotelemetry did not differ significantly between the two treatment groups, but more cows were detected in estrus 36 h after removal of IVDs in the IVD-8-EB cows compared to the IVD-7-EB cows (P<0.05). We concluded that reproductive performance in resynchronized dairy cows but not heifers was greater following resynchronization of estrous cycles after AI with an IVD for 7 compared to 8d when EB was injected at the start of treatment and 24h after device removal.  相似文献

In Experiment 1, the effects of two pre-synchronization treatments on synchronized AI pregnancy rates of lactating dairy cattle were compared. Lactating Holstein cows (n=159) received 100 microg of GnRH (im) on day -7 and 25mg of PGF(2alpha) (im) on day 0 and were observed once daily for signs of estrus from day -3 to day 3. Cows detected in standing estrus and those that had lost significant amounts of tail-chalk in the previous 24h were immediately inseminated in a once-daily observation/AI program. Cows not detected in estrus by 72 h after PGF(2alpha) received fixed-time AI (TAI) and a concurrent 100 microg injection of GnRH (im). Cows were randomly assigned by parity and calving date to receive one of the following pre-synchronization treatments: (1) 25mg of PGF(2alpha) (im) on day -35 and day -21 (PGF-PGF) or (2) 100 microg of GnRH (im) on day -14 (GnRH). Fewer (P<0.05) GnRH- (49%, 41/84) than PGF-PGF-pretreated cows (65%, 49/75) were detected in estrus, however, overall pregnancy rates were not affected by pre-synchronization treatment (30 versus 32%, respectively). In Experiment 2, lactating Holstein cows received 100 microg of GnRH (im) on day -7, 25mg of PGF(2alpha) (im) on day 0 and TAI at 60-64 h after PGF(2alpha). Cows were randomized by parity and postpartum interval into pre- and post-synchronization treatments in a 2 x 2 factorial design. Pre-synchronization treatments included: (1) 25mg of PGF(2alpha) (im) on day -35 and on day -21 (PGF-PGF; n=168) or (2) 25mg of PGF(2alpha) (im) on day -21 and 100 microg of GnRH (im) on day -14 (PGF-GnRH; n=180). Within each pre-synchronization treatment, cows were further allocated by parity and postpartum interval to receive as a post-synchronization treatment 100 microg of GnRH (im) at either 48 h (Ovsynch; n=175) or 60-64 h (Cosynch; n=173) after PGF(2alpha). Pregnancy rates at TAI were not affected by pre- (PGF-PGF=26%, 44/168 versus PGF-GnRH=24%, 44/180) or post-synchronization treatments (Ovsynch=29%, 50/175 versus Cosynch=22%, 38/173). However, the numeric shift towards reduced pregnancy rates in Cosynch-treated cows suggests the 12h interval between GnRH and AI may be important to optimize conception rates in GnRH-PGF(2alpha)-based TAI protocols in dairy cattle. In conclusion, each of the pre-synchronization protocols evaluated in present study performed with comparable efficacy. Although the Cosynch protocol facilitates more efficient labor utilization, numeric trends toward reduced conception warrants further investigation.  相似文献

The present study determined whether a 5-d progesterone-based CO-Synch protocol with a single dose of prostaglandin F (PGF) at progesterone withdrawal on Day 5, would yield a timed AI pregnancy rate similar to two doses of PGF given 6 h apart on Day 5. Angus cross beef heifers (N = 562) at six locations were used. All heifers received 100 µg of gonadorelin hydrochloride (GnRH) and a controlled internal drug release (CIDR) insert on Day 0. Within farm, heifers were randomly allocated to receive one dose of 25 mg dinoprost (PGF) at CIDR removal on Day 5 (1 PGF; N = 264), or two doses of 25 mg PGF, with the first dose given on Day 5 at CIDR removal, and the second dose 6 h later (2 PGF; N = 298). Most heifers (N = 415) received a heat detector patch at CIDR removal. After CIDR removal, heifers were observed twice daily through Day 7 for estrus and heat detector aid status was recorded. On Day 8, heifers were given 100 µg of GnRH, heat detector aid status was recorded, and heifers were inseminated approximately 72 h after CIDR removal. Accounting for significant variables such as location (P < 0.01), heifers in estrus at or prior to AI (P < 0.001), and a treatment by location interaction (P < 0.01), two doses of PGF on Day 5 tended to have higher pregnancy rates to timed AI compared to those that received one dose of PGF (P = 0.06). In conclusion, heifers given two doses of PGF at CIDR removal on Day 5, in a 5-d CIDR-CO-Synch protocol, tended to have a higher pregnancy rate than those that received only one dose of PGF.  相似文献

In the present study, two new short estrus synchronization methods have been developed for lactating dairy cows. The study was completed in three consecutive phases. In experiment (Exp) 1, 32 cows, that were not detected in estrus since calving between the 50th and 84th post-partum days, were treated with PGF2alpha (PGF, d-cloprostenol, 0.150 mg), estradiol propionate (EP, 2mg) and GnRH (lecirelina, 50 microg) at 24h intervals, respectively, and timed artificial insemination (TAI) was performed 48 h after PGF. Different from Exp 1, EP and GnRH were given at 48 and 60 h, respectively after PGF in Exp 2 (n=20), instead of 24 and 48 h. Ovulations were investigated by ultrasound for 7 days starting from the day of PGF treatment, and ovulation rates were compared with the ones obtained in Exp 1. In Exp 3, cows were given the same treatments as Exp 2, but treatments started at certain estrus stages. Cows detected in estrus and with a confirmed ovulation (n=27) after the second PGF given 11 days apart were assigned to three treatment groups. Treatment was initiated at Day 3 (group metestrus, n=9), Day 12 (group diestrus, n=9) and Day 18 (group proestrus, n=9) after ovulation. All cows included in Exp 3 were TAI between 16 and 20 h after GnRH treatment. In Exp 2 and 3, blood samples were obtained once every 2 days, starting from Day 0 to the 10th day after GnRH injection, and once every 4 days between the 10th and the 22nd days after GnRH to examine post-treatment luteal development. During the study, animals exhibiting natural estrus were inseminated and served as controls (n=85). The rate of estrus was found to be significantly higher in cows with an active corpus luteum (CL) at the start of Exp 1 (72.7% vs. 30.0%, P<0.05) and the pregnancy rate tended to be higher than cows without an active CL (40.9% vs. 10.0%, P=0.08). Compared to those in Exp 1, cows in Exp 2 had higher rates of synchronized ovulation (94.1% vs. 59.1%, P=0.013). In Exp 3, estrus (P<0.001) and pregnancy rates (P=0.01) were found to be significantly higher in cows in the proestrus group than in those in the metestrus group. Comparable pregnancy rates were obtained from the first and second inseminations in Exp 1 and 3 with results from those inseminated at natural estrus (P>0.05). It was concluded from the study that the treatment in Exp 1 and 3 could result in comparable pregnancy rates after timed AI of lactating dairy cows at random stages of the estrus cycle relating to those inseminated at natural estrus, but the stage of the estrus cycle can have significant effects on pregnancy rates.  相似文献

The objective was to synchronize follicular wave emergence among cattle for synchronization of estrus and ovulation, and to determine pregnancy rate after AI at observed estrus. At random stages of the estrous cycle, a controlled internal drug release device (CIDR-B) was inserted intravaginally (Day 0) in 67 cross-bred beef heifers, and they were randomly allocated to receive either no further treatment (Control; n = 18); 5 mg of estradiol-17beta and 100 mg of progesterone im (E/P; n = 16); 100 microg im of GnRH (GnRH; n = 16); or transvaginal ultrasound-guided follicular ablation of all follicles > or = 5 mm (FA; n = 17). All heifers received a luteolytic dose of PGF (repeated 12 h later), and CIDR-B were removed on Days 9, 8, 6 or 5, in Control, E/P, GnRH or FA groups, respectively, so the dominant follicle of the induced wave was exposed to exogenous progesterone for a similar period of time in each group. Mean (+/- SEM) intervals (and range, in days) from treatment to follicular wave emergence in these groups were 3.5 +/- 0.6 (-2 to 8), 3.4 +/- 0.1 (3 to 4), 1.5 +/- 0.3 (-1 to 4), and 1.0 +/- 0.1 (0 to 2), respectively. Although the interval was longest (P<0.01) in the E/P and Control groups, it was least variable (P<0.01) in the E/P and FA groups. Intervals (and range, in days) from CIDR-B removal (and first PGF treatment) to estrus were 2.3 +/- 0.2 (1.5 to 4.5), 2.2 +/- 0.2 (1.5 to 3.0), 2.1 +/- 0.1,(1.5 to 3.5), and 2.5 +/- 0.1 (2.0 to 3.5), and to ovulation were 3.5 +/- 0.2 (2.5 to 5.5), 3.4 +/- 0.1 (3.0 to 4.5), 3.5 +/- 0.1 (2.5 to 4.5), and 3.8 +/- 0.1 (3.0 to 4.5), for Control, E/P, GnRH and FA groups, respectively (ns). The proportion of heifers displaying estrus was higher in the Control than in the FA group (94% versus 65%, P<0.05) and intermediate in EP and GnRH groups (87% and 75%). Heifers were inseminated approximately 12 h prior to ovulation (based on estrous behavior and ultrasound examinations). Pregnancy rates were 78%, 80%, 69% and 65% for Control, E/P, GnRH and FA groups, respectively (P=0.73). Results support the hypothesis that synchronous follicular wave emergence results in synchronous follicle development and, following progesterone removal, synchronous estrus and ovulation with high pregnancy rates to AI. The synchrony of estrus and ovulation in the E/P, GnRH and FA groups suggest that these treatments, in combination with CIDR-B, could be adapted to fixed-time insemination programs.  相似文献

Our objective was to determine whether extending the interval from 17 to 19 d between removal of melengestrol acetate (MGA) feed and administration of PGF2 alpha would alter conception rates, pregnancy rates and the degree of synchrony in replacement beef heifers. A commercial heifer operation in north-central Kansas purchased 591 Angus x Hereford heifers from 12 sources. Prior to the spring breeding season, 14% of the heifers were culled. The remaining heifers were assigned randomly to 2 MGA-PGF2 alpha synchronization systems. All heifers were fed MGA (0.5 mg/head/d) for 14 d, and PGF2 alpha was administered either 17 or 19 d after the completion of MGA feeding. Heifers were inseminated artificially for 30 d followed by 30 d of natural mating. Based on each source, first-service conception rates ranged from 66 to 90%, whereas overall pregnancy rates ranged from 91 to 100%. Heifers given PGF2 alpha on Day 17 after MGA had first-service conception rates of 75.9% compared with 81.4% for heifers receiving PGF2 alpha on Day 19. In response to the PGF2 alpha injection, 99% of the Day 19 heifers that were detected in estrus were inseminated artificially by 72 h after the PGF2 alpha injection, whereas 74% of the heifers in the Day 17 treatment were inseminated by that time. Average interval to artificial insemination (AI) after PGF2 alpha was greater (P < 0.01) for the Day 17 heifers (73.1 +/- 1.1 h) than for the Day 19 heifers (56.2 +/- 1.1 h). No differences in conception rates or overall pregnancy rates occurred; however, heifers receiving PGF2 alpha on Day 19 after MGA had shorter intervals to estrus, and a greater proportion was inseminated within 72 h after PGF2 alpha, thus possibly facilitating successful timed insemination of the remaining heifers not yet inseminated by that time.  相似文献

The objective of this study was to evaluate the effect of a PGF2α-analogue (PGF) on ovulation and pregnancy rates after timed artificial insemination (TAI) in cattle. In experiment 1, crossbred dual-purpose heifers, in a crossover design (3 × 3), were given an intravaginal progesterone-releasing insert (controlled internal drug release [CIDR]) plus 1 mg estradiol benzoate (EB) intramuscularly (im) and 250 μg of a PGF-analogue im on Day 0. The CIDR inserts were removed 5 days after follicular wave emergence, and the heifers were randomly divided into three treatment groups to receive the following treatments: (1) 1 mg of EB im (EB group, n = 13); (2) 500 μg of PGF im (PG group, n = 13); or (3) saline (control group, n = 13), 24 hours after CIDR removal. Ovulation occurred earlier in EB (69.81 ± 3.23 hours) and PG groups (73.09 ± 3.23 hours) compared with control (83.07 ± 4.6 hours; P = 0.01) after CIDR removal. In experiment 2, pubertal beef heifers (n = 444), 12 to 14 months of age were used. On Day 0, the heifers were given a CIDR insert plus 2 mg EB im. On Day 9, the CIDR was removed and the heifers were given 500 μg of PGF im. Heifers were randomly assigned into one of three treatment groups: (1) 1 mg of EB (EB group; n = 145); (2) 500 μg of PGF (PG group; n = 149), both 24 hours after CIDR removal; or (3) 600 μg of estradiol cypionate (ECP group; n = 150) at CIDR removal. Timed artificial insemination occurred 48 hours after CIDR removal in the ECP group and 54 hours in the PG and EB groups. The percentage of heifers ovulating was higher in the PG group compared with the other groups (P = 0.08). However, the pregnancy rates did not differ among groups (47.6%, 45%, and 46.6%, for EB, PG, and ECP, respectively; P = 0.9). In experiment 3, 224 lactating beef cows, 40 to 50 days postpartum with 2.5 to 3.5 of body condition score were treated similarly as described in experiment 2, except for the ECP group, which was excluded. The treatments were as follows: 1 mg EB (EB group; n = 117) or 500 μg PGF (PG group; n = 107), 24 hours after CIDR removal. The calves were temporarily separated from their dams from Days 9 to 11. No difference was detected on the pregnancy rate between the EB and PG groups (58.1% vs. 47.6%, respectively; P = 0.11). Taken together, the combined results suggested that PGF2α could be successfully used to induce and synchronize ovulation in cattle undergoing TAI, with similar pregnancy rates when compared with other ovulatory stimuli (ECP and EB).  相似文献

The aim was to compare the estrous response in heifers given either gonadotropin-releasing hormone (GnRH) or estradiol benzoate (EDB) at the start of a progesterone treatment initiated at emergence or dominance of the first or second follicular wave of the estrous cycle. Cross-bred beef heifers (n=134) were assigned to 1 of 3 treatments; 0.75 mg EDB given at insertion of a progesterone-releasing intravaginal device (PRID) treatment of 10 days duration (10dE2), 0.75 mg EDB at insertion of a PRID treatment of 8 days duration with 15 mg luprostiol (PGF) a luteolytic agent, given 1 day before PRID removal (8dE2) or 250 microg GnRH at insertion of a PRID treatment of 8 days duration with 15 mg PGF given 1 day before PRID removal (8dGnRH). Treatments were initiated on Days 2, 5, 10 or 13 of the estrous cycle. Estrous detection was conducted six times daily. Twice daily blood samples were taken, from 2 days before PRID insertion until detection of estrus. The proportion of heifers detected in estrus was higher (P < 0.05) for heifers in the 8dE2 treatment group (40/40) compared with those in the 8dGnRH group (38/42) and tended to be higher (P = 0.08) than heifers in the 10dE2 group (38/41). The onset of estrus was earlier (P < 0.05) for heifers in the 10dE2 treatment group (median 41 h, range 92 h) compared with either the 8dE2 (median 49 h, range 64 h) or 8dGnRH groups (median 49 h, range 92 h). Submission rate at 72 h was higher (P < 0.01) in the 8dE2 (95%) group than for those in the 10dE2 (74%) and 8dGnRH (69%) groups. In conclusion, EDB given at PRID insertion, with PGF given 1 day before PRID removal, was more effective at synchronizing estrus than was GnRH at PRID insertion. Two experiments evaluated a modified delivery of prostaglandin F2alpha (PGF2alpha) after a melengestrol acetate (MGA) treatment in Angus and Bos indicus x Bos taurus (BI) heifers. Experiment 1 was replicated three times with yearling BI heifers (n = 695). Heifers received MGA (0.5 mg head(-1) day(-1)) for 14 days. In Replications 1 and 2, heifers received either 25 mg of PGF2alpha im 19 days after MGA (single) or 12.5 mg of PGF2alpha im 19 and 20 days after MGA (split). In Replication 3, heifers received the same treatments, with PGF2alpha initiated either 18 or 19 days after MGA. Estrus was detected for 72 h after PGF2alpha, with AI commencing 8-12 h after a detected estrus. Heifers not observed in estrus by 72 h were timed-AI concomitant with GnRH (100 microg im). Heifers from Replication 2 (n = 146) had blood samples collected at the initial PGF2alpha and at timed-AI to determine corpus luteum (CL) regression by evaluating plasma progesterone concentrations. The interval from MGA withdrawal to PGF2alpha did not have a significant effect on any variable in Replication 3 and there were no treatment by replication effects for any variables, therefore data were pooled. Modifying the PGF2alpha treatment from a single treatment to two treatments on consecutive days increased (P < 0.05) 72 h estrous response (43.2% versus 50.1%), timed-AI (23.9% versus 33.5%) and total-AI pregnancy rates (34.5% versus 42.5%), and CL regression (79.1% versus 92.5%), respectively. In Experiment 2, yearling Angus (n = 66) and 2-year-old BI (n = 68) heifers were synchronized as per Experiment 1 (with the initial PGF2alpha 19 days after MGA). Neither breed nor PGF2alpha treatment effected (P > 0.05) 72 h estrous response, total-AI pregnancy rate, or CL regression rate. In conclusion, treating yearling BI heifers with split treatments of PGF2alpha (given on two consecutive days) improved estrous response and pregnancy rates by increasing PGF2alpha-induced luteolysis.  相似文献

Gonadotropin releasing hormone (GnRH) was given to 109 cows and heifers during the course of 224 superovulations. Follicle stimulating hormone (FSH) was administered twice daily (5 or 6 mg) for 3.5 to 4 days beginning on any of Days 9 to 14 of the estrous cycle; prostaglandin (45 mg PGF(2)alpha or 750 ug cloprostenol) was given in a split dose on the fourth day. Donor cows and heifers were placed into four groups according to previous superovulation treatments, which consisted of one to three treatments or of no previous treatment. Every other cow or heifer within each of the four subgroups was treated with GnRH (200 mug i.m.) at standing estrus. Only donors that exhibited estrus within 32 to 72 h after the first prostaglandin treatment were used in the study. Animals were inseminated artificially 12 and 24 h after standing estrus was first observed. No differences were noted in the number of ovulations, total ova or transferable embryos recovered from the GnRH or control groups; however, two interactions were detected. Cows given GnRH had fewer palpable corpora lutea than control cows (P < 0.05), but this difference was not seen in heifers. The second interaction was that GnRH seemed to depress ovulation rate in donors not previously superovulated, but this effect was not observed with subsequent superovulations. Cows yielded more total ova than heifers (P < 0.01). There was no difference in return to estrus between GnRH and control groups after a second prostaglandin treatment at the time of embryo recovery. Most donors within each group resumed cycling between 5 and 12 d after embryo recovery.  相似文献

In Experiment 1, cows with a history of at least two previous unsuccessful inseminations were allocated to four groups. At the repeated estrus (third or greater service), some of the cows were inseminated according to the a.m.-p.m. rule (Controls, n = 83), or received i.m. 100 mug gonadotropin-releasing hormone (GnRH, n = 32) within 30 sec after insemination. Ovaries of the remaining cows to be treated were palpated during the anticipated ensuing luteal phase to determine the presence of a corpus luteum. Cows found to have luteal tissue received i.m. 25 mg prostaglandin F(2)-alpha (PGF(2)-alpha) and were inseminated after detected estrus or at 72 and 96 h after PGF(2)-alpha in the absence of estrus. Cows given PGF(2)-alpha either received no further treatment (PGF(2)-alpha, n = 40) or were given i.m. 100 mug GnRH (PGF(2)-alpha + GnRH, n = 29) after insemination or at 72 h after PGF(2)-alpha in the absence of estrus. Conception rate of control cows (39%) was similar to that of cows given PGF(2)-alpha (40%) or PGF(2)-alpha + GnRH (43%), but it tended to be lower (P = 0.13) than that of cows given only GnRH at insemination (54%). In Experiment 2, cows with unobserved estrus and diagnosed not pregnant (palpation) were palpated to detect a corpus luteum. Cows with luteal tissue received i.m. PGF(2)-alpha (n = 52) or PGF(2)-alpha + GnRH (n = 45) and were inseminated as described above. Conception rates were similar (39% vs 33%, respectively). In Experiment 3, cows in a large commercial dairy with (n = 93) or without (n = 420) previous reproductive problems were given i.m. 100 mug GnRH after insemination (n = 169) or were left untreated (n = 344) at repeat services (third and fourth services). Treatment with GnRH improved (P < 0.05) conception in normal (47% vs 36%) and abnormal (25% vs 12%) repeat-breeding cows. Treatment with PGF(2)-alpha alone or in conjunction with GnRH failed to further improve conception rates and only delayed intervals to rebreeding when administered during the luteal phase after the repeated estrus. The use of GnRH failed to reduce intervals from treatment to insemination or improve conception in cows with unobserved estrus compared to treatment with PGF(2)-alpha alone.  相似文献

The objective was to evaluate the effects of plasma progesterone (P4) concentrations and exogenous eCG on ovulation and pregnancy rates of pubertal Nellore heifers in fixed-time artificial insemination (FTAI) protocols. In Experiment 1 (Exp. 1), on Day 0 (7 d after ovulation), heifers (n = 15) were given 2 mg of estradiol benzoate (EB) im and randomly allocated to receive: an intravaginal progesterone-releasing device containing 0.558 g of P4 (group 0.5G, n = 4); an intravaginal device containing 1 g of P4 (group 1G, n = 4); 0.558 g of P4 and PGF (PGF; 150 μg d-cloprostenol, group 0.5G/PGF, n = 4); or 1 g of P4 and PGF (group 1G/PGF, n = 3). On Day 8, PGF was given to all heifers and intravaginal devices removed; 24 h later (Day 9), all heifers were given 1 mg EB im. In Exp. 2, pubertal Nellore heifers (n = 292) were treated as in Exp. 1, with FTAI on Day 10 (30 to 36 h after EB). In Exp. 3, pubertal heifers (n = 459) received the treatments described for groups 0.5G/PGF and 1G/PGF and were also given 300 IU of eCG im (groups 0.5G/PGF/eCG and 1G/PGF/eCG) at device removal (Day 8). In Exp. 1, plasma P4 concentrations were significantly higher in heifers that received 1.0 vs 0.588 g P4, and were significantly lower in heifers that received PGF on Day 0. In Exp. 2 and 3, there were no significant differences among groups in rates of ovulation (65-77%) or pregnancy (Exp. 2: 26-33%; Exp. 3: 39-43%). In Exp. 3, diameter of the dominant ovarian follicle on Day 9 was larger in heifers given 0.558 g vs 1.0 g P4 (10.3 ± 0.2 vs 9.3 ± 0.2 mm; P < 0.01). In conclusion, lesser amounts of P4 in the intravaginal device or PGF on Day 0 decreased plasma P4 from Days 1 to 8 and increased diameter of the dominant follicle on Day 9. However, neither of these nor 300 IU of eCG on Day 8 significantly increased rates of ovulation or pregnancy.  相似文献

Three trials utilizing 231 beef heifers were conducted in 1993 to determine if a timed insemination would result in similar synchronized pregnancy rates as insemination by estrus following synchronization of estrus using the 14-d MGA-prostaglandin system. All heifers were fed 0.5 mg MGA/h/d fof 14 d and given a 25 mg injection of PGF(2)alpha im 17 d after the final day of MGA feeding. Heifers in Group 1 (timed AI treatment) were inseminated at 72 h after the prostaglandin injection independent of whether or not they were observed in estrus. Heifers in Group 2 (AI by estrus) were inseminated 12 to 18 h after the onset of estrus. Since the trial was a significant source of variation for synchronized pregnancy rate, the effect of treatment on pregnancy rate was analyzed for each trial. Synchronized pregnancy rates in Trials 2 and 3 were similar in both treatment groups; 37 vs 35% and 61 vs 58% for the timed AI vs AI by estrus (Groups 1 and 2) in Trials 2 and 3, respectively. In both of these trials the degree of estrous synchrony was high. In Trial 1, the synchronized pregnancy rate in heifers that were time-inseminated was significantly lower than that of heifers that were inseminated by estrus (29 vs 57%). The lower synchronized pregnancy rate of Group 1 (timed AI) heifers in Trial 1 appeared to be due to the low degree of estrous synchrony in this trial. Our results indicate that using timed insemination with the 14-d MGA-prostaglandin system will give similar synchronized pregnancy rates as inseminating by estrus in groups of beef heifers where the degree of synchrony is high. However, in heifers where the degree of estrous synchrony is low, a timed insemination reduces synchronized pregnancy rates.  相似文献

The objective was to determine whether timed artificial insemination (TAI) 56 h after removal of a Controlled Internal Drug Release (CIDR, 1.38 g of progesterone) insert would improve AI pregnancy rate in beef heifers compared to TAI 72 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol. Angus cross beef heifers (n = 1098) at nine locations [WA (5 locations; n = 634), ID (2 locations; n = 211), VA (one location; n = 193) and WY (one location; n = 60)] were included in this study. All heifers were given a body condition score (BCS; 1-emaciated; 9-obese), and received a CIDR insert and 100 μg of gonadorelin hydrochloride (GnRH) on Day 0. The CIDR insert was removed and two doses of 25 mg of dinoprost (PGF) were given, first dose at CIDR insert removal and second dose 6 h later, on Day 5. A subset of heifers (n = 629) received an estrus detector aid at CIDR removal. After CIDR removal, heifers were observed thrice daily for estrus and estrus detector aid status until they were inseminated. Within farm, heifers were randomly allocated to two groups and were inseminated either at 56 h (n = 554) or at 72 h (n = 544) after CIDR removal. All heifers were given 100 μg of GnRH at AI. Insemination 56 h after CIDR insert removal improved AI pregnancy rate compared to insemination 72 h (66.2 vs. 55.9%; P < 0.001; 1 - β = 0.94). Locations, BCS categories (≤ 6 vs. > 6) and location by treatment and BCS by treatment interactions did not influence AI pregnancy rate (P > 0.1). The AI pregnancy rates for heifers with BCS ≤ 6 and > 6 were 61.8 and 60.1%, respectively (P > 0.1). The AI pregnancy rates among locations varied from 54.9 to 69.2% (P > 0.1). The AI pregnancy rate for heifers observed in estrus at or before AI was not different compared to heifers not observed in estrus [(65.4% (302/462) vs. 52.7% (88/167); P > 0.05)]. In conclusion, heifers inseminated 56 h after CIDR insert removal in a 5-days CO-Synch + CIDR protocol had, on average, 10.3% higher AI pregnancy rate compared to heifers inseminated 72 h after CIDR insert removal.  相似文献

The influence of interval between insemination (AI) and estrus on subsequent fertility of PGF(2alpha)-treated (two injections of 25 mg, 11 days apart) heifers was assessed in two experiments. In Experiment I, 240 heifers were allotted to Control (AI 8 to 16 hr after estrus detection), PGF(2alpha)-E (AI 8 to 16 hr after estrus within five days of second PGF(2alpha)) or PGF(2alpha)-T (AI 80 hr after second PGF(2alpha)). In Experiment II, 130 heifers were assigned to control (AI as before) or PGF(2alpha) (AI 72 or 80 hr after second PGF(2alpha)) with half the PGF(2alpha) heifers receiving 100 mug GnRH 72 hr after first PGF(2alpha). Heifers of both experiments that were bred at a predetermined time were arrayed by interval from AI to estrus. Conception rates of heifers detected in estrus from 32 hr before AI to 24 hr after AI did not differ (x(2)=3.35, df=5, P>0.5). The percentage of GnRH-treated heifers in estrus within five days (81.8%) was not (P>0.75) greater than those not receiving GnRH (77.3%) but they had higher (P<0.05) serum progesterone (P(4)) concentration at second PGF(2alpha) (3.17 vs 2.41 ng/ml). When P(4) values were arrayed for both groups at 1 ng intervals, the percentage of heifers exhibiting estrus increased with increasing P(4) level (P<0.05).  相似文献

Following observation of estrus, 134 Holstein heifers were given injections of Prostaglandin F(2)alpha (PGF(2)alpha) between Days 5 and 10 of their cycle (estrus = Day 0). They were then randomly assigned to either a group receiving 400 mug of estradiol benzoate (E(2)B) 40 h or maintained as controls. Heifers observed in estrus within 120 h of PGF(2)alpha administration were inseminated (approximately 12 h after initial observation of estrus). Blood samples for progesterone determination were drawn from the coccygeal vein on Days 15 and 21 after insemination. Pregnancy was confirmed by palpation per rectum between Days 5.0 and 60 post insemination. When control and treated heifers were compared it was found that a higher percentage of heifers treated with E(2)B exhibited estrus after PGF(2)alpha, but there had been no effect on subsequent progesterone concentrations or pregnancy rates.  相似文献

Yearling beef heifers (n = 193) were used to evaluate reproductive performance attained with 2 MGA-PGF(2)alpha synchronization systems. These treatments were compared with an untreated control group. The 14-d MGA heifers were synchronized by feeding 0.5 mg MGA/h/d for 14 d. At 17 d after the last MGA feeding, these heifers were injected with PGF(2)alpha (25 mg, im). Heifers in the 7-d MGA treatment group were fed 0.5 mg MGA/h/d for 7 d and received a 25-mg, im injection of PGF(2)alpha on the last day of the MGA feeding period. Heifers in all 3 treatment groups were observed for estrus every 12 h for 7 d beginning 24 h after the PGF(2)alpha injection. Heifers observed in estrus during this 7-d period were artificially inseminated approximately 12 h after the onset of estrus. The percentages of heifers in estrus during the 7-d synchronized period were 75.4, 56.3 and 17.2% for the 14-d MGA, 7-d MGA and control groups, respectively. The estrous responses were significantly different in each treatment. The percentage of heifers in estrus during the peak 24-h period was higher (P < 0.05) in heifers synchronized with the 14-d MGA system than in heifers synchronized with the 7-d MGA system (75.5 vs 50.0%). The synchronized conception rate of the 14-d MGA heifers was significantly higher (65.3%) than that of both the 7-d MGA (41.7%) and control (45.4%) heifers. Synchronized conception rates were similar (P = 0.79) in the 7-d MGA and control treatments. Synchronized pregnancy rates were 55.2, 32.4 and 15.2% for the 14-d MGA, 7-d MGA and control groups, respectively. Both synchronization treatments resulted in significantly higher synchronized pregnancy rates compared with that of the controls. The synchronized pregnancy rate was higher (P < 0.05) in the 14-d MGA group than it was in the 7-d MGA group. The mean day of conception within the breeding season was 11.5 and 9.3 d shorter in the 14-d MGA heifers than in the 7-d MGA and control heifers, respectively. Our results indicate that using the 14-d MGA system to synchronize estrus in beef heifers results in better reproductive performance than that attained in heifers synchronized with the 7-d MGA system or in control heifers.  相似文献
