Productivity gradients in the boreal forest are largely determined by regional-scale changes in soil conditions, and bacterial communities are likely to respond to these changes. Few studies, however, have examined how variation in specific edaphic properties influences the composition of soil bacterial communities along environmental gradients. We quantified bacterial compositional diversity patterns in ten boreal forest sites of contrasting fertility. Bulk soil (organic and mineral horizons) was sampled from sites representing two extremes of a natural moisture-nutrient gradient and two distinct disturbance types, one barren and the other vegetation-rich. We constructed 16S rRNA gene clone libraries to characterize the bacterial communities under phylogenetic- and species-based frameworks. Using a nucleotide analog to label DNA-synthesizing bacteria, we also assessed the composition of active taxa in disturbed sites. Most sites were dominated by sequences related to the α-Proteobacteria, followed by acidobacterial and betaproteobacterial sequences. Non-parametric multivariate regression indicated that pH, which was lowest in the natural sites, explained 34% and 16% of the variability in community structure as determined by phylogenetic-based (UniFrac distances) and species-based (Jaccard similarities) metrics, respectively. Soil pH was also a significant predictor of richness (Chao1) and diversity (Shannon) measures. Within the natural edaphic gradient, soil moisture accounted for 32% of the variance in phylogenetic (but not species) community structure. In the boreal system we studied, bacterial β-diversity patterns appear to be largely related to “master” variables (e.g., pH, moisture) rather than to observable attributes (e.g., plant cover) leading to regional-scale fertility gradients. 相似文献

From a total woody flora of ca. 1000 species, a suite of 121 species forms an oligarchy dominating the cerrado biome. This mirrors patterns of dominance described in western Amazonian rain forests. Widespread sampling shows that across the biome this suite of species contributes on average 66% of the total species composition, and 75% of the total Importance Value Index in cerrado communities. An analysis of the floristic similarity between six cerrado phytogeographic provinces (southern, central and south-eastern, central-western, far-western, north-eastern, disjunct Amazonian) reveals great heterogeneity within the biome, principally of the less common species. Of the 951 species recorded from 375 floristic surveys across the biome, 494 species (more than half of the total) are found in only one of the provinces, with very few species (37, i.e., 3.9%) found in all six provinces. Each of the provinces contains a significant number of species which are apparently confined to it, ranging from 15 species (1.6% of the total woody cerrado flora) in the far-western province (primarily in the state of Rondônia) to 162 species (17%) in the central-western province. At the local level, floristic similarity can be very high. An analysis of the floristic composition of 13 sites within the Federal District shows a woody flora of 236 species, indicating that 25% of the total woody flora of the cerrado biome is represented in 0.3% of its area, illustrating the great conservation importance of this region. Floristic similarity between sites within the Federal District is high, although much of this similarity is accounted for by oligarch species which account for between 59 and 89% of total species recorded per site. Informed conservation judgements within the cerrado need to take account of regional floristic patterns to ensure maximum protection of biodiversity, as the majority of species are not geographically widespread within the biome. 相似文献

Several studies report the decline of β diversity caused by agricultural impacts in river ecosystems. However, the susceptible scale of β diversity to agricultural impacts and the indirect effects on β diversity within hierarchically nested ecosystems are unclear. We first tested the hypothesis that β diversity between microhabitats is significantly influenced by agricultural land use. We also examined the indirect effects of agricultural land use on α and β diversities at the microhabitat scale. Twelve microhabitat samples (25 cm2) were collected at 27 reaches within Kitamihorobetsu River, Japan. All reaches were classified into three agricultural intensities based on pasture area (low, middle, and high), and their hierarchical diversities (microhabitat, reach, and catchment) were calculated using additive partitioning. Indirect effects were demonstrated by structural equation modelling using indirect and direct environmental gradients. The α and β diversities at the microhabitat scale decreased significantly with agricultural intensity. Increasing pasture cover within catchments showed a negative correlation with habitat heterogeneity and water quality (habitat homogenization and water degradation) and a positive correlation with sand cover on streambeds (sedimentation of fine particles). The α diversity decreased with sedimentation of fine particles, and the β diversity decreased with habitat homogenization and water degradation. Our findings suggest that species diversity of aquatic insects at the microhabitat scale would be susceptible to agricultural land use. Furthermore, we emphasize that the diversity index used to evaluate restoration projects should be carefully selected because influential abiotic factors were different between α and β diversities. 相似文献

Black layer is a condition of high-sand-content golf greens that results in a subsurface blackened layer in the sand produced by sulfate-reducing bacteria. Black layer can be the product of an interaction of cyanobacteria and sulfate-reducing bacteria and may or may not be toxic to the grass growing on the sand. The organic byproducts of the cyanobacteria coat and plug the sand thereby creating an anoxic environment for development of the sulfate- reducing bacteria. The present study was initiated to determine the range of gaseous hydrocarbons evolved from black layered sand produced by the interaction of two genera of cyanobacteria, Nostoc and Oscillatoria, and Desulfovibrio desulfuricans. The gaseous hydrocarbons measured included methane, ethane, ethylene, and propylene. In nonblackened sand, Nostoc evolved the highest levels of these gases, Oscillatoria evolved relatively low levels except for propylene, and D. desulfuricans evolved the smallest quantities of the gases. When the cyanobacteria and D. desulfuricans were combined to develop black layered sand some changes occurred in the evolution of the gases. Evolution of the gases from Nostoc + D. desulfuricans decreased or remained the same relative to Nostoc alone, and increased relative to D. desulfuricans alone. Except for propylene evolution, gases from Oscillatoria + D. desulfuricans increased relative to Oscillatoria or D. desulfuricans alone. Propylene evolution from Oscillatoria + D. desulfuricans remained unchanged relative to Oscillatoria alone, but increased relative to D. desulfuricans alone. The gases measured are discussed relative to the organisms observed and the conditions of the study. 相似文献

Small soda lakes represent one of the most vulnerable ecosystem types due to their high hydrological sensitivity to climate change and anthropogenic interventions. Since diatoms are excellent bioindicators, determining the β-diversity and the structuring dynamics of diatom metacommunities can provide valuable information for conservation planning for soda pans. In this study, two diatom metacommunities were surveyed monthly during a one-year period from distinct regions of the Carpathian basin: the Fert?-Hanság National Park (FH) between 2013 and 2014, and the Danube-Tisza Interfluve (DT) between 2014 and 2015. We explored whether β-diversity of diatom assemblages in the two regions is enhanced by species turnover or nestedness (related to richness differences) and investigated the role of deterministic and stochastic processes in shaping β-diversity patterns. Furthermore, we evaluated the contribution of environmental variables, geographic distance and temporal variation to community structure. High β-diversity (>90%) was revealed for both metacommunities, and was maintained primarily by species turnover. Within the metacommunity of the DT where the natural hydrological cycle of soda pans is not disturbed, diatom communities assembled mainly due to the selection force of environment at a spatiotemporal scale. In the soda pans located in the habitat reconstruction area of the FH, besides species-sorting, significant temporal variation in community structure appeared as a result of water management and periodic water supply. Our results point to the need for a conservation management strategy which maintains the natural hydrological regime of small saline lakes, and therefore their habitat heterogeneity which is of high conservation value. 相似文献

Growing evidence suggests the presence of active lysosomal enzymes in extra-lysosomal compartments, such as the plasma membrane. Although in the past little attention was paid to glycohydrolases acting on cellular compartments different from lysosomes, there is now increasing interest on plasma membrane-associated glycohydrolases because they should be involved, together with glycosyltransferases, in glycosphingolipids oligosaccharide modification processes regulating cell-to-cell and/or cell-environment interactions in both physiological and pathological conditions. Starting from the previous evidence of the presence of β-hexosaminidase and β-galactosidase at the plasma membrane of cultured fibroblasts, we here investigated the association of these glycohydrolases with lipid microdomains of Jurkat T-lymphocytes. Monosialoganglioside GM3 represents the major glycosphingolipid constituent of T-cell plasma membrane and its amount largely increases after T-cell stimulation. β-hexosaminidase and β-galactosidase cleave specific β-linked terminal residues from a wide range of glycoconjugates and in particular are involved in the stepwise degradation of GM1 to GM3 ganglioside. Here we demonstrated that fully processed plasma membrane-associated β-hexosaminidase and β-galactosidase co-distribute with the lipid microdomain markers and co-immunoprecipitate with the signalling protein lck in Jurkat T-cell. Furthermore, Jurkat cell stimulation up-regulates the expression and activity of lysosomal β-hexosaminidase and β-galactosidase and increases their targeting to lipid microdomains. The non-random distribution of plasma membrane-associated β-hexosaminidase and β-galactosidase and their localization within lipid microdomains, suggest a role of these enzymes in the local reorganization of glycosphingolipid-based signalling units. 相似文献

Riparian forests and environments close to watercourses support high biodiversity, which may be modified by human activities. In the Paraná River Delta region, cattle raising is one of the activities with the greatest impact, altering vegetation structure. These changes are reflected in bird communities that inhabit these environments. We hypothesize that the absence of large herbivores (whether cows or native deer) will produce an increase in the vegetation cover of the lower strata of forests, due to the greater height of the herbaceous stratum and greater coverage of the shrub stratum, and that these changes mainly affect the species of birds that use resources associated with these strata. Our objective was to understand the changes in richness and abundance of the bird community among different types of cattle management using a functional aggregation approach of bird species. In the areas with cattle, we found less coverage of the shrub layer, lower height of the herbaceous layer and fewer climbing vines. We found that changes in richness and abundance of bird community were strongly related to species associated with the lower vegetation strata (ground-feeding guild, shrub guild and low canopy guild), and that the responses of the different guilds were not homogeneous. Understanding the direction of changes in bird communities occurring in response to modifications of the environment, allows us to optimize conservation efforts. If these efforts are based on conservation of the environment in its natural state, we should adjust the management of the herbivory to the pristine conditions of the environment. On the other hand, if conservation efforts are based on particular species or groups of species associated with certain resources, the correct management of herbivory by cattle can be essential to obtain successful results. 相似文献

Cora pavonia, a lichen having a basidiomycetous mycobiont, is rich in protein (36%), of which 10% is tyrosine. α,α-Trehalose is present and was isolated in 4.4% yield. The lichen was found to contain polysaccharides typical of basidiomycetes and different from those of ascomycetes and ascomycetous lichens. Isolated and characterized were a β-d-glucan and a heteropolysaccharide containing l-rhamnose, l-fucose, d-xylose, d-mannose, d-glucose, and d-galactose. The β-d-glucan was highly branched with 21% of nonreducing end-groups, contained 3-O-, 6-O-, and 3,6-di-O-substituted β-d-glucopyranosyl units, and had a main chain consisting of (1→3)- and (1→6)-links. The heteropolysaccharide component contained mainly mannose and xylose, having a mannose-containing nucleus and a main chain with preponderant (1→3)-linked α-d-mannopyranosyl residues. These were unsubstituted (10%), and 4-O- (10%) and 2,4-di-O-substituted (10%) with residues of β-d-xylopyranose. On methylation analysis of the heteropolysaccharide, a capillary column of DB-210 proved to be particularly useful for gas-liquid chromatographic resolution of partially O-methylated alditol acetates. 相似文献

By using the proton microprobe technique we have investigated the elemental composition of both pancreatic -cells and exocrine pancreas from fed and 24 h or 48 h starved obese hyperglycemic mice. Among the 15 elements measured in the -cells both Ca and Fe increased while Mg and S decreased significantly after 24 h of starvation, the effects being more pronounced after 48 h. When animals were starved for 48 h there was a decrease in the contents of Cl, Rb and Cu, whereas that of Al and Mn increased with 152 and 55%, respectively. There was an initial decrease in Na after 24 h of starvation, which was followed by an increase after 48 h. This is in contrast to Cd, which first increased and then decreased to a value lower than that obtained in the fed animal. The content of K showed a small decrease and that of Pb showed an increase only in the 24 h starved group. In the -cells the contents of Zn and P did not change subsequent to starvation. In the exocrine pancreas Na, Cl and P decreased after 24 h of starvation and except for Na, the decrease was maintained when the starvation period was increased to 48 h. After 24 h there was a significant, though transient, increase in K, Mg and Rb. With regard to the contents of Zn, Cu and S there was a progressive decrease as the starvation continued. In contrast to the endocrine pancreas the content of Al in the exocrine pancreas did not change after 48 h of starvation. There was no change in islet insulin content subsequent to starvation. The extent to which the observed changes in -cell elemental composition is involved in the impaired insulin release associated with starvation, merits further investigations. 相似文献

The aim was to estimate the impacts of invasive Impatiens parviflora on forests' herbal layer communities. A replicated Before-After-Control-Impact field experiment and comparisons with adjacent uninvaded plots were used. The alien's impact on species richness was tested using hierarchical generalized mixed effect models with Poisson error structure. Impact on species composition was tested using multivariate models (DCA, CCA, RDA) and Monte-Carlo permutation tests. Removal plots did not differ in native species richness from neither invaded nor adjacent uninvaded plots, both when the treatment's main effect or its interaction with sampling time was tested (Chi(2)?=?0.4757, DF?=?2, p?=?0.7883; Chi(2)?=?7.229, DF?=?8, p?=?0.5121 respectively). On the contrary, ordination models revealed differences in the development of plots following the treatments (p?=?0.034) with the invaded plots differing from the adjacent uninvaded (p?=?0.002). Impatiens parviflora is highly unlikely to impact native species richness of invaded communities, which may be associated with its limited ability to create a dense canopy, a modest root system or the fact the I. parviflora does not represent a novel and distinctive dominant to the invaded communities. Concerning its potential impacts on species composition, the presence of native clonal species (Athyrium filix-femina, Dryopteris filix-mas, Fragaria moschata, Luzula luzuloides, Poa nemoralis) on the adjacent uninvaded plots likely makes them different from the invaded plots. However, these competitive and strong species are more likely to prevent the invasion of I. parviflora on the adjacent uninvaded plots rather than being themselves eliminated from the invaded communities. 相似文献

β-site APP-cleaving enzyme (BACE1) cleaves the wild type (WT) β-site very slowly (k(cat)/K(m): 46.6 m(-1) s(-1)). Therefore we searched for additional β-secretases and identified three cathepsins that split the WT β-site much faster. Human cathepsin S cleaves the WT β-site (k(cat)/K(m): 54 700 m(-1) s(-1)) 1170-fold faster than BACE1 and cathepsins B and L are 440- and 74-fold faster than BACE1, respectively. These cathepsins split two bonds flanking the WT β-site (K-MD-A), where the K-M bond (85%) is cleaved more efficiently than the D-A bond (15%). Cleavage at the major K-M bond yields Aβ (amyloid β-peptide) extended by N-terminal Met that should be removed to generate Aβ initiated by Asp1. The activity of cytosol and microsomal aminopeptidases on relevant peptides revealed rapid removal of N-terminal Met but not N-terminal Asp. Brain aminopeptidases showed similar specificity. Thus, aminopeptidases would convert Aβ extended by Met into regular Aβ (Asp1) found in amyloid plaques. Earlier studies indicate that Aβ is likely produced in the endosome and lysosome system where cathepsins S, B and L are localized and cysteine cathepsin inhibitors reduce the level of Aβ in cells and animals. Taken together, cathepsins S, B and L deserve further evaluation as therapeutic targets to develop disease modifying drugs to treat Alzheimer's disease. 相似文献

Several hypotheses have been proposed to explain biotic resistance of a recipient plant community based on reduced niche opportunities for invasive alien plant species. The limiting similarity hypothesis predicts that invasive species are less likely to establish in communities of species holding similar functional traits. Likewise, Darwin’s naturalization hypothesis states that invasive species closely related to the native community would be less successful. We tested both using the invasive alien Ambrosia artemisiifolia L. and Solidago gigantea Aiton, and grassland species used for ecological restoration in central Europe. We classified all plant species into groups based on functional traits obtained from trait databases and calculated the phylogenetic distance among them. In a greenhouse experiment, we submitted the two invasive species at two propagule pressures to competition with communities of ten native species from the same functional group. In another experiment, they were submitted to pairwise competition with native species selected from each functional group. At the community level, highest suppression for both invasive species was observed at low propagule pressure and not explained by similarity in functional traits. Moreover, suppression decreased asymptotically with increasing phylogenetic distance to species of the native community. When submitted to pairwise competition, suppression for both invasive species was also better explained by phylogenetic distance. Overall, our results support Darwin’s naturalization hypothesis but not the limiting similarity hypothesis based on the selected traits. Biotic resistance of native communities against invasive species at an early stage of establishment is enhanced by competitive traits and phylogenetic relatedness. 相似文献

Anthropogenic activity, such as conversion and degradation of habitats, is causing global biodiversity declines. However, our understanding of how local ecological communities are responding to these changes taxonomically and functionally is still limited. The effects of the replacement of native by introduced pastures on biodiversity are some of those examples with limited understanding. Here, we sampled dung beetles in native and introduced pastures using standardised sampling protocols during the dry and rainy seasons in the Brazilian Pantanal. We used multifaceted β-diversity partitioning of taxonomy-, abundance- and trait-based approaches to evaluate spatial (i.e. between pasture types) and temporal (i.e. between seasons) patterns of dung beetle changes in composition, abundance and species traits. Spatially, we found no effects of pasture type, season and their interaction on taxonomy-based β-diversity and its components. For abundance-based β-diversity and its components, pasture type had effects on both Bray-Curtis dissimilarity and balanced variation in abundance. Higher values of both metrics were found in native pastures. For functional-based β-diversity, Sorensen dissimilarity with higher values in the dry season, while pasture type also had an effect on functional nestedness, where higher values of functional nestedness were found in introduced pastures. Seasonally, we also found no effects of pasture type, season and their interaction on taxonomy-based β-diversity and its components. However, pasture type had effects on Bray-Curtis dissimilarity, with higher values found in introduced pastures. No effects on balanced variation in abundance and abundance gradients were found. Pasture type influenced the functional turnover, but not functional dissimilarity or nestedness. Higher values of functional turnover were found in native pastures. In summary, we demonstrate that the type of pastures and climatic seasonality have effects on abundance- and functional- but not on taxonomy-based β-diversity patterns of dung beetles in the Brazilian Pantanal. 相似文献

-globin gene haplotypes were determined for 196 normal (-A) and 419 thalassaemia (-Th) chromosomes of individuals from four different regions of the Indian subcontinent; North-west Pakistan, Gujarat, Punjab and Sindh. Analysis of -A and -Th haplotypes and haplotype-mutation associations in each regional group along with a consideration of Indian history provided information about the origin and spread of -thalassaemia mutations on the Indian subcontinent. The data are consistent with relatively recent and local origins for most -thalassaemia mutations. The frequencies of particular alleles differ markedly in various regions and these may be useful population markers. Of the high frequency alleles, intervening sequence 1 (IVS-1) nucleotide 5 (G-C) and codons 41/42 (-CTTT) appear to be older as suggested by multiple haplotype associations and a widespread geographical distribution. The microepidemiology of -thalassaemia in this region reflects considerable ethnic diversity, gene flow from population migration and natural selection by malaria infection. 相似文献

Although a number of recent studies have demonstrated biotic homogenization, these have mainly focused on larger spatial scales. Homogenizing effects are equally important at finer resolutions, e.g. through increasing similarity between habitats, which may result in a simplification of ecosystem structure and function. One major cause of homogenization is the expanding ranges of alien species, although it is not clear whether they are inherently homogenizing at smaller scales. We therefore assessed whether the alien flora is less complex across habitats than the resident native flora of Mediterranean Islands. From a regional data base, we examined floristic lists for between‐habitat taxonomic and functional similarity, and within‐habitat functional diversity, using resampled data sets to control for sample size biases. Aliens and natives showed equivalent complexity in most respects. At the taxonomic level, between‐island and between‐habitat similarities were almost identical, and when ecosystem function was measured by a functional group classification system, this was also true of between‐habitat similarities and within‐habitat diversities. When ecosystem function was measured using Grime's CSR classification, aliens were found to be more functionally homogenous between‐habitats and less functionally diverse within habitats. However, since the CSR profiles of aliens and natives differed, simplification is not inevitable due to ecological segregation of the two floras (aliens tend to be recruited to disturbed habitats rather than displacing natives). One deficiency is a lack of large scale species abundance data. A simple simulation exercise indicated that this is likely to lead to substantial overestimation of true levels of similarity, although would only influence the comparison between aliens and natives if they have different abundance distribution curves. The results indicate that alien floras are not intrinsically more simple than natives, but a higher proportion of competitive strategists among aliens may still cause small‐scale homogenization as these include many strong competitors that are likely to dominate communities. 相似文献

Antigenic similarity between the β-subunits of the ATPases of a bacterium,a yeast and a higher plant
Antiserum raised against the β-subunit of wheat (Triticum aestivum) chloroplast ATPase cross-reacts with a 51000 protein located in the membrane fraction of Escherichia coli. The differential solubility of this polypeptide after chloroform treatment of unc+ and uncD409 strains indicates that this cross-reacting polypeptide is the bacterial β-subunit of ATPase. Thus a high degree of conservation of antigenic determinant sites exists between a bacterial β-subunit and the β-subunit of a monocot. This conservation also seems to extend to the β-subunit of mitochondrial ATPase of yeast (Saccharomyces cerevisiae). 相似文献

Rust-red wild and black mutant strains of the red flour beetle, Tribolium castaneum, were used to investigate temporal patterns of catecholamine and β-alanine content during sclerotization and pigmentation of adult cuticle and to relate these patterns to corresponding changes in cuticle resistance to puncture. Rust-red elytral cuticle sclerotized more rapidly than black cuticle until 6 days after adult eclosion when both became equal in puncture resistance. The cuticular concentrations of (NBAD), β-alanine and 3,4-dihydroxyphenylacetic acid (DOPAC) increased more rapidly in the rust-red strain than in the black strain during the first 7 days following adult eclosion. Conversely, cuticular dopamine increased more rapidly in black than in the red strain. Thus the rust-red pigmentation and rapid sclerotization appear to be related to the availability of β-alanine, and DOPAC. Melanization was prevented and rust-red pigmentation induced by injections of β-alanine or NBAD into newly ecdysed black mutant beetles. Crosses of the two strains generally had intermediate levels of cuticular dopamine and β-alanine, but the NBAD levels were similar to those of the rust-red strain. Dopamine, NBAD and DOPAC levels became similar in both black and rust-red strains about 6 days after adult ecdysis as did resistance to puncture. Therefore, dopamine appears to be directed initially into the melanin pathway in black adults due to a temporary lack of with β-alanine. After the melanization phase, dopamine is metabolized to sclerotization precursors eventually resulting in normal physical properties of the exoskeleton. 相似文献
