Two new eremophilane sesquiterpenes, compounds 1 and 2, were isolated from the endophytic fungus Microdiplodia sp. KS 75-1, together with the known compounds phomadecalins C (3) and D (4). Their structures were determined by extensive 1D– and 2D–NMR and MS spectral analyses. The previously reported stereochemistry at C-8 of 3 and 4 were revised on the basis of NOEs experiments. Compounds 1 and 2 showed antimicrobial activity against Pseudomonas aeruginosa. 相似文献

An endophytic streptomycete (NRRL 30566) is described and partially characterized from a fern-leaved grevillea (Grevillea pteridifolia) tree growing in the Northern Territory of Australia. This endophytic streptomycete produces, in culture, novel antibiotics - the kakadumycins. Methods are outlined for the production and chemical characterization of kakadumycin A and related compounds. This antibiotic is structurally related to a quinoxaline antibiotic, echinomycin. Each contains, by virtue of their amino acid compositions, alanine, serine and an unknown amino acid. Other biological, spectral and chromatographic differences between these two compounds occur and are given. Kakadumycin A has wide spectrum antibiotic activity, especially against Gram-positive bacteria, and it generally displays better bioactivity than echinomycin. For instance, against Bacillus anthracis strains, kakadumycin A has minimum inhibitory concentrations of 0.2-0.3 microg x ml(-1) in contrast to echinomycin at 1.0-1.2 microg x ml(-1). Both echinomycin and kakadumycin A have impressive activity against the malarial parasite Plasmodium falciparum with LD(50)s in the range of 7-10 ng x ml(-1). In macromolecular synthesis assays both kakadumycin A and echinomycin have similar effects on the inhibition of RNA synthesis. It appears that the endophytic Streptomyces sp. offer some promise for the discovery of novel antibiotics with pharmacological potential. 相似文献

Chemical investigation of the mangrove-derived fungus Aspergillus sp. AV-2 following fermentation on solid rice medium led to the isolation of a new phenyl pyridazine derivative (1) and a new prenylated benzaldehyde derivative, dioxoauroglaucin (2), together with fourteen known compounds (3-16). Chemical structures of the new compounds were unambiguously determined based on HRESIMS and extensive 1D and 2D NMR spectroscopic analyses. Compounds 2-7, 8 and 13 were assessed for their antiproliferative activity against Caco-2 cell lines, where flavoglaucin (6) revealed the most potent cytotoxicity with IC50 of 2.87 μM. 相似文献

The endophytic strain Mesorhizobium sp. CCNWGX022 was isolated from licorice (Glycyrrhiza uralensis Fisch.) grown in the arid and semi-arid regions of Northwest China. The new stearic acid derived gamma-lactone 1, named rhizobialide (= (5S)-4,5-dihydro-5-(8-oxotetradecyl)furan-2(3H)-one), was isolated from the petroleum-ether extract of the fermentation broth of this strain. Two new cadinane-type sesquiterpenes, hypocreaterpenes A (1) and B (2), along with five known compounds (3–7) were isolated from a marine-derived fungus Hypocreales sp. strain HLS-104 isolated from a sponge Gelliodes carnosa. Their structures were determined by a combination of spectroscopic methods. All compounds were tested for the inhibitory effects on the nitric oxide (NO) production in lipopolysaccharide (LPS)-treated RAW264.7 cells. Among them, compounds 3 and 6 showed moderate anti-inflammatory activity with average maximum inhibition (Emax) values of 10.22% and 26.46% at 1 μM, respectively. 相似文献

Studies comparing fungal endophytes between above- and belowground woody crop organs and the factors that may structure their communities are lacking. Due to its great impact on the Mediterranean Basin, the olive tree was chosen for the isolation of endophytic fungi from roots, leaves and twigs in two seasons in north-eastern Portugal. Nine hundred seventy-six isolates belonging to 38 fungal species were obtained. Phomopsis columnaris, Fusarium oxysporum and Trichoderma gamsii were the most frequently isolated, collectively representing 69% of the isolates. Fungal diversity in the roots was higher than in the aboveground organs and higher in spring than autumn. Endophyte community similarity between the above- and belowground organs and between seasons was low. Species composition also varied spatially, with the fungal composition of the roots varying more among locations than that of the aboveground organs. Our results suggest olive tree endophyte community structure is affected by plant organ, location and season. 相似文献

Two new xanthones, smeathxanthone A (1) (2-(3,7-dimethyl-2,6-octadienyl)-1,3,5,8-tetrahydroxyxanthone) and smeathxanthone B (2) (5,7,10-trihydroxy-2-methyl-2-(4-methylpent-3-enyl)[2H, 6H]pyrano[3,2-b]xanthen-6-one), have been isolated from the stem bark of Garcinia smeathmannii, and their structures elucidated on the basis of 1D and 2D NMR experiments. 1,3,5-Trihydroxyxanthone and 1,5-dihydroxyxanthone were also obtained. A Xylaria sp. was isolated as an endophyte from a surface-sterilized Concord grape leaf (Vitis labrusca). Dereplication and LC–MS analysis of the filtrate extracts identified two new structures, compound 1, 13-O-methyl-(5R) diplosporin, and 2 a new agistatine. Additionally, five known compounds 3–7 including an agistatine, coriloxin, (5R)-diplosporin, and two mellein derivatives were also isolated. This is the first report of these structurally-related metabolites isolated from the same fungus, suggesting they share a related biosynthesis. Metabolites were isolated using liquid chromatography-solid phase extraction (LC-SPE) and semi-preparative HPLC. 目的分离火欣麻的内生菌,探讨ERIC-PCR引物在植物内生细菌分型中的优化及其与AP-PCR的结合在植物内生菌分型中的应用价值。方法通过改进的碾碎法对火欣麻的内生菌进行分离,依据形态学与生理生化特性同时对比AP-PCR和ERIC-PCR图谱,对所分离菌进行分类鉴定。结果所筛选的12株内生细菌,2株为奈氏菌属,1株为葡萄球菌属,1株为链球菌属,3株为芽胞杆菌属,2株为短杆菌属,3株为肠杆菌属。优化引物后的ERIC-PCR条带稳定、丰富、清晰。结论ERIC-PCR与AP-PCR结合使用分型效果明显增强。 相似文献

From the extracts of the fermentation broth of Streptomyces sp. Lz531, two new and four known compounds were isolated. Two isoflavanones, 5,7-dihydroxy-2'-methoxy-3',4'-methylenedioxyisoflavanone (2) and 4',5-dihydroxy-2',3'-dimethoxy-7-(5- hydroxyoxychromen-7yl)-isoflavanone (4) along with six known compounds including isoflavanones, triterpenes and steroids were isolated from the roots of Uraria picta. The structures of these compounds were established unambiguously by UV, IR, MS and a series of 1D and 2D NMR analyses. The minimum inhibitory concentrations (MIC) for these compounds were found to be in the range of 12.5-200 microg/ml against bacteria (both Gram positive and Gram negative) and fungi. 相似文献

Five new germacrane sesquiterpene lactones, petrophins A–E (1–5), were isolated from the whole herbs of Salvia petrophila. The structures were established using spectroscopic analysis (1D and 2D NMR, HR-ESI-MS) and compatible with values in the literature. These sesquiterpenes are unusual with an endocyclic double bond in the γ-lactone ring. 摘要:【目的】华重楼内生菌PCE45具有较强的抗菌活性,本文将对PCE45产生的抗菌物质进行分离纯化和性质分析报道。【方法】PCE45发酵液经硫酸铵盐析、丙酮沉淀、SephadexG75柱、DE52纤维素柱和SephadexG25凝胶柱纯化分离得到抗菌肽PCP-1。【结果】稳定性测试表明该抗菌肽对蛋白酶不敏感,对高温、强酸、强碱有较好的耐受性,可造成稻瘟病菌菌丝畸形并抑制孢子萌发。抑菌谱表明该抗菌肽对玉米弯胞病菌等真菌和大肠杆菌等细菌有较强的抑菌效果。质谱测得其分子量为1058.3D。氨基酸组成分析表明该 相似文献

ABSTRACTBlack pepper endophytic Pseudomonas putida BP25 produced diverse antimicrobial volatile organic compounds having potential for plant disease management. Chemically synthesised volatiles such as 2, 5-dimethyl pyrazine; 2-methyl pyrazine; dimethyl trisulphide; 2-ethyl 5-methyl pyrazine; and 2-ethyl 3, 6-dimethyl pyrazine showed inhibitory activity against oomycete pathogens, Phytophthora capsici & Pythium myriotylum; fungal pathogens, Rhizoctonia solani, Colletotrichum gloeosporioides, Athelia rolfsii, Gibberella moniliformis & Magnaporthe oryzae; bacterial pathogen, Ralstonia pseudosolanacearum and plant parasitic nematode, Radopholus similis. Among them, dimethyl trisulphide completely inhibited oomycete and fungal pathogens as well as R. similis at a concentration of 2.65?µg?cm?3 under in vitro conditions. Pyrazines suppressed Phytophthora lesions on shoot cuttings of black pepper upon in planta volatile treatment. Dimethyl trisulphide was the only compound that exhibited soil fumigant activity against P. capsici, R. solani and A. rolfsii (6.25?µg?cm?3), C. gloeosporioides and G. moniliformis (12.5?µg?cm?3), and R. similis (50?µg?cm?3). Phytochemical investigation of the seeds of Salacia longipes var. camerunensis led to the isolation of four sesquiterpenoid derivatives, salaterpene A (1) (1α,2β,8β-triacetoxy-6β,9β-dibenzoyloxy-4β-hydroxy-dihydro-β-agarofuran), salaterpene B (2) (1α,2β,8β-triacetoxy-9β-benzoyloxy-6β-cinnamoyloxy-4β-hydroxy-dihydro-β-agarofuran), salaterpene C (3) (1α,2β-diacetoxy-6β,9β-dibenzoyloxy-4β-hydroxy-dihydro-β-agarofuran) and salaterpene D (4) (2β-acetoxy-1α,6β-dibenzoyloxy-4β-hydroxy-9β-nicotinoyloxy-dihydro-β-agarofuran) together with two known compounds (5 and 6). The structures of the compounds were established by means of NMR spectroscopy. 从水浮莲(Pistia stratiotes L.)的叶中分离出1株对西瓜枯萎病有明显拮抗作用的内生细菌XJPL-YB-26, 其发酵液上清在280 nm处有最大紫外吸收峰。利用软件Primer 6.0 设计16S rDNA引物并对其基因组DNA进行扩增并测序得到XJPL-YB-26的部分16S rDNA序列, GenBank接收号为EU251191。经Blastn调出与菌株16S rDNA同源的序列, 并用软件MEGA 3.1按Neighbor- Joining方法构建16S rDNA系统发育树。菌株XJPL-YB-26与AB271744处于同一分支, 相似性为99%, 最终鉴定为枯草芽孢杆菌Bacillus subtilis。 相似文献
