Summary An investigation was made of the effects of temperature and of mono- and disaccharides on lipids, biomass, odd-chain and unsaturated fatty acids production of Candida lipolytica. With different sugars as carbon source at 30°C, the order for biomass production was: fructose > glucose > sucrose > lactose > galactose, while lipids production/g biomass decreased as follows: lactose, sucrose, galactose, fructose and glucose. On the other hand, the odd-chain fatty acids contents decreased in the following order: fructose, lactose, glucose, sucrose and galactose. Lowering the temperature of cultivation to 15°C, biomass, lipids and unsaturated fatty acids decreased. However, a notable decrease in the content of odd-chain fatty acids was detected.  相似文献

1. Sugar best single chorda tympani nerve fiber of rat and hamster were tested with six sugars. 2. Fibers were selected for this experiment, only if they responded to 1.0 M sucrose or 1.0 M maltose and they responded poorly to 0.1 M NaCl. 3. In rat, some single fibers gave larger responses to maltose than to sucrose, while in hamster nearly all nerve fibers responded best to sucrose. 4. The order of effectiveness of sugars was maltose greater than fructose greater than or equal to lactose greater than sucrose greater than glucose greater than galactose in rat and sucrose greater than fructose greater than or equal to glucose greater than or equal to galactose greater than maltose greater than lactose in hamster.  相似文献

Ultrasonic vocalizations were recorded from 10-day-old albinorats while they were isolated from their dam and siblings. Eachrat received a 1.0% BW intra-oral infusion of sucrose, fructose,glucose or lactose at a concentration range of 0.22–0.66M for 3 min and their vocalizations were determined during theinfusion and for an additional 7 min. Sucrose, fructose andglucose all significantly reduced vocalizations to about 50%of baseline levels, whereas lactose, the milk sugar, was ineffective.Moreover, the dose–response function was flat for thethree effective sugars. In a second experiment, the effectsof these sugars on heat escape latency were measured. Sucrose,fructose and glucose each elevated the latency with which infantrats removed a paw from a 48°C surface; lactose did not.These findings of lactose ineffectiveness and the flat dose-responsefunction for the other three sugars exactly parallel those obtainedfor human newborns. In the present study, a simple and sensitive photo‐induced chemiluminescence (CL) method for the quantitation of reducing sugars, including fructose, glucose, sucrose and lactose, was developed. This method was based on the on‐line photocatalytic reaction of the reducing sugars, using a home‐made photoreactor consisting of PTEF tube helically coiling around a high‐pressure mercury UV lamp. Reducing sugars were detected by direct CL emission resulting from the reaction between the photoproducts and luminol. To maximize the signal intensity, the effects of irradiation time, flow rate, luminol concentration, buffer pH and concentration were tested. Under optimum conditions, the linear dynamic ranges were all 0.36–18 mg/L and the relative standard deviations (RSDs) were 1.8–2.3%, with limits of detection (3σ) of 0.06 mg/L for fructose, glucose, sucrose and lactose. Finally, interference effects from ascorbic acid, amino acids (alanine, glycine, serine, leucine and methionine) and some metal ions and anions were also investigated. The effects of adding sugars to high- and low-tar cigarettes on the mutagenicity of their smoke condensates were studied using Salmonella typhimurium TA100 and TA98 with and without metabolic activation. The sugars tested were glucose, fructose, galactose, sorbitol, sucrose and lactose. The lowest mutagenicities observed with these sugars per mg of smoke condensate assayed on TA98 with metabolic activation were 37% (high-tar cigarettes) and 22% (low-tar cigaretts) of that of smoke condensate from untreated cigarettes. Addition of sugars increased the total amounts of smoke condensates, but the mutagenicities of the total condensates were also decreased by all the sugars, the lowest values being 35% (high-tar cigarettes) and 36% (low-tar cigarettes) of that of smoke condensates from cigarettes without added sugar. On assay with TA100 with metabolic activation, decreases in both specific and total mutangenicities of condensates of high-tar cigarettes were observed with all the sugars tested except galactose and sucrose. Treatment with glucose, fructose or sorbitol decreased the specific mutagenicity of condensates of low-tar cigarettes and glucose and fructose reduced also their total mutagenicity. The effects of added sugars were more marked when assayed on TA98 than on TA100 and of the sugars tested fructose and sorbitol had the greatest effects. Addition of sugars had no effect of the mutagenicity of cigarette-smoke condensate without metabolic activation.  相似文献

The suitability of various nectar and honeydew sugars as a food source for the polyphagous ant species M. rubra (L.) was studied. The sugars used included monosaccharides (fructose, glucose, galactose, mannose, rhamnose), disaccharides (sucrose, maltose, trehalose, melibiose, lactose) and trisaccharides (melizitose, raffinose, erlose). Single-sugar solutions were tested on ant workers in a long-term laboratory bioassay in which acceptance of the solutions and ant survival were recorded. The acceptance of the sugars was confirmed in a second bioassay in which feeding time was established. Enzymatic hydrolysis of sucrose, maltose and melibiose was investigated through HPLC analyses of workers fed these disaccharides. Sugar acceptance and feeding time were related to ant survival. Considering the monosaccharide units of which the sugars are composed, fructose seems especially suitable as a short-term energy source, while glucose appears to be used both directly and for storage. The presence of a galactose unit appears to reduce sugar suitability. It is suggested that the workers possess invertase and maltase and to a lesser degree also galactosidase. The gustatory perception is correlated with the profitability of sugars in further metabolic processes.  相似文献

The effect of different levels of salt, sodium nitrite, polyphosphate and various sugars on growth, pigment production, protease activity and culture pH caused by Monascus purpureus was studied in broth medium and ground meat. The addition of sodium chloride (> 50.0 g l(-1)) and polyphosphate (> 3.0g l(-1)) to broth medium decreased mycelial growth, pigment production and protease activity of M. purpureus, whereas low concentrations of sodium nitrite (< 0.2 g l(-1)) promoted mycelial growth and pigment production. When the basal medium and ground meat contained salt, 150.0 g l(-1), the mould growth was stopped. The medium with fructose as carbon source proved to be the most suitable for mycelium growth and pigment production, with maltose and glucose being the second most productive. When sucrose and lactose were used as carbon sources, mycelium growth and pigment production were inhibited but the protease activity increased significantly. The mould showed more tolerance to salt and polyphosphate in ground meat than in broth medium and used sucrose as a carbon source as well as glucose for growth and pigment production in the meat mixture.  相似文献

We were interested in determining the feeding response of the Caribbean fruit fly, Anastrepha suspensa Loew (Diptera: Tephritidae), to various sugar concentrations to develop an improved bait for adults. We compared the consumption of 0.01-1.00 M concentrations of glucose, fructose, raffinose, and sucrose in no-choice tests for 24-h- and 6-d-old male and female flies. Sucrose was the most consumed sugar or within the most consumed group of sugars at 0.02-0.20 M concentrations. There were no differences in consumption among sugars at 0.01, 0.40, and 1.00 M. Consumption generally increased with increasing sugar concentration except that sucrose consumption peaked at 0.20 M. Twenty-four-hour females consumed less fructose than other sugars; 24-h males consumed more sucrose than fructose or raffinose, with an intermediate response to glucose. Females in the 6-d group consumed more sucrose than the other three sugars, whereas 6-d males exhibited no difference in consumption among sugars. In choice tests, flies consumed more sugar solution than water, but the difference between 0.20 M fructose and water was not significant for 24-h males or 24-h females. In choice tests between 0.20 M fructose and 0.20 M sucrose, both 24-h and 6-d females showed a preference for fructose. Males of both age classes showed no preference. These results indicate that the responses of flies to different sugars can vary by sugar, gender, and age.  相似文献

Influxes of glucose, fructose and sucrose were characterised for coat cells of developing seeds of Phaseolus vulgaris L. and Vicia faba L. by monitoring uptake of [(14)C]sugars into excised seed-coat halves and two different protoplast populations derived from seed coats. Sugar influxes by the two populations of protoplasts were similar for each sugar species [sucrose > (fructose approximately glucose)] and hexoses competed with sucrose. Concentration-dependent influxes of all three sugars by excised seed coats could be described by a simple directly proportional relationship between concentration ([S]) and uptake rate (v) in the physiological range of sugar concentrations (v approximately A.[S]). Alternatively, with the exception of fructose influx by Vicia, all could be fitted to a Michaelis-Menten relationship, as could sucrose uptake by Vicia protoplasts. Apparent K(m) values were high ( approximately 100-500 mM) compared with those reported for other systems. Sucrose transport was distinct from glucose and fructose transport in both species. Sugar influx was decreased by p-chloromercuribenzenesulfonic acid, carbonylcyanide m-chlorophenylhydrazone and erythrosin B. These responses are consistent with sugar/H(+) symport acting to retrieve photoassimilates leaked to the apoplasm during post-sieve element transport within seed coats.  相似文献

Adult parasites, Isoparorchis hypselobagri were maintained in vitro in four basic salt solutions, viz. Stephenson saline, AB saline, Tyrode's solution and Locke's solution. Survival was prolonged by adding sugars to Tyrode's solution. Of all the sugars, glucose was found most effective and parasites survived for 39 days in solution containing glucose, 6 days in galactose and sucrose, 10 days in fructose and lactose and 12 days in maltose. The parasites absorb carbohydrates through the cuticle. Ligated parasites survived for a longer period than unligated parasites. The pH 9 was considered as an optimum for maximum survival of the parasites Isoparorchis hypselobagri for 39 days.  相似文献

The yeast Kluyveromyces marxianus has been pointed out as a promising microorganism for a variety of industrial bioprocesses. Although genetic tools have been developed for this yeast and different potential applications have been investigated, quantitative physiological studies have rarely been reported. Here, we report and discuss the growth, substrate consumption, metabolite formation, and respiratory parameters of K. marxianus CBS 6556 during aerobic batch bioreactor cultivations, using a defined medium with different sugars as sole carbon and energy source, at 30 and 37 °C. Cultivations were carried out both on single sugars and on binary sugar mixtures. Carbon balances closed within 95 to 101 % in all experiments. Biomass and CO2 were the main products of cell metabolism, whereas by-products were always present in very low proportion (<3 % of the carbon consumed), as long as full aerobiosis was guaranteed. On all sugars tested as sole carbon and energy source (glucose, fructose, sucrose, lactose, and galactose), the maximum specific growth rate remained between 0.39 and 0.49 h?1, except for galactose at 37 °C, which only supported growth at 0.31 h?1. Different growth behaviors were observed on the binary sugar mixtures investigated (glucose and lactose, glucose and galactose, lactose and galactose, glucose and fructose, galactose and fructose, fructose and lactose), and the observations were in agreement with previously published data on the sugar transport systems in K. marxianus. We conclude that K. marxianus CBS 6556 does not present any special nutritional requirements; grows well in the range of 30 to 37 °C on different sugars; is capable of growing on sugar mixtures in a shorter period of time than Saccharomyces cerevisiae, which is interesting from an industrial point of view; and deviates tiny amounts of carbon towards metabolite formation, as long as full aerobiosis is maintained.  相似文献

The effects of sugars (xylose, arabinose, fucose, fructose, galactose, glucose, sorbitol, maltose, sucrose, and lactose; 0-20% w/v) on the properties of the pressure-induced gel from a whey protein isolate (20%, 800 MPa, 30 degrees C, 10 min) were studied. All the sugars decreased the hardness, breaking stress and water-holding capacity of the gel at the same concentration of 55.5 mM. Increasing the sugar content changed the microstructure of the gel from a honeycomb-like structure to a stranded structure, while the strand thickness was progressively reduced. These results suggest that sugars decreased the degree of intermolecular S-S bonding of proteins and non-covalent interaction, and restrained the phase separation during gelation under high pressure.  相似文献

Glucose, maltose, sucrose, lactose, xylose, sorbose, galactose, fructose and gluconolactone were analyzed by means of immobilized pyranose oxidase as well as by the combination of immobilized glucose oxidase with immobilized glycoamylase, invertase, mutarotase, maltase (α-glucosidase) and glucose isomerase by flow injection analysis (FIA). For the simultaneous analysis of glucose and other sugars three different flow-injection configurations were applied and compared. The average error of prediction of the analyses were better than 3% in model media and better than 6% in yeast extract containing media.  相似文献

Agricultural and herbaceous feedstocks may contain appreciable levels of sucrose. The goal of this study was to evaluate the survivability of sucrose and its hydrolysis products, fructose and glucose, during dilute sulfuric acid processing at conditions typically used to pretreat lignocellulose biomass. Solutions containing 25g/l sucrose with 0.1-2.0% (w/w) sulfuric acid concentrations were treated at temperatures of 160-200 degrees C for 3-12min. Sucrose was observed to completely hydrolyze at all treatment conditions. However, appreciable concentrations of fructose and glucose were detected and glucose was found to be significantly more stable than fructose. Different mathematical approaches were used to fit the kinetic parameters for acid-catalyzed thermal degradation of these sugars. Since both sugars may survive dilute acid pretreatment, they could provide an additional carbon source for production of ethanol and other bio-based products.  相似文献

Abstract.  The longevity and nutrient levels of Pseudacteon tricuspis provided with 1  m solutions of five naturally occurring sugars, fructose, glucose, sucrose, trehalose and melezitose, are compared. All but melezitose, result in significant increases in the longevity of P. tricuspis in comparison with sugar-starved flies (flies provided with water only). Sugar-starved female and male P. tricuspis have an average longevity of 3.3 and 4.1 days, respectively. Provision of free water in addition to sugar solution is necessary for optimum longevity by female and male flies. Longevity is increased by 2.4–2.6-fold by the two monosaccharides, fructose and glucose, and by 2.6–2.8-fold by the disaccharides, sucrose and trehalose. Phorid flies provided with the trisaccharide sugar, melezitose, had a marginal increase in lifespan (approximately 1 day), but this is not significantly different from the longevity of sugar-starved flies. Significantly greater levels of total sugars are detected in P. tricuspis fed the disaccharide sugars (sucrose, trehalose) or the monosaccharide sugars (fructose, glucose), compared with flies provided with melezitose (trisaccharide), or to sugar-starved flies. Fructose is not detected in sugar-starved flies, or in flies fed glucose or trehalose. However, high levels of fructose are detected in flies fed sucrose or fructose, whereas levels of fructose in melezitose-fed flies are intermediate. In general, significantly greater glycogen levels are detected in P. tricuspis fed sucrose, glucose, trehalose or fructose, compared with melezitose-fed or sugar-starved flies. Levels of total sugars and glycogen in sugar-fed flies are positively correlated with wing length, possibly indicating a higher accumulation of storage sugars by larger flies. These results are discussed in relation to the nutritional ecology of the phorid fly.  相似文献

1. Intracellular recordings of mouse taste cell responses were made using a glass micro-electrode filled with Procion yellow dye solution. 2. Six sugars (sucrose, maltose, lactose, glucose, galactose and fructose) produced the depolarization responses. 3. Gustatory cross adaptation between sugars was determined. When the taste cell was pre-adapted with one of the six sugars, the other five sugars, cross adapted, produced depolarization, hyperpolarization or null responses. 4. From these observations, it is suggested that there are multiple sugar receptor sites on the receptor membrane of the mouse taste cell.  相似文献

The occurrence of salt‐, sugar‐sensitive neurones and a mechanoreceptor neurone in the antennal hair‐like gustatory sensilla of the click beetle Agriotes obscurus L. (Coleoptera, Elateridae) is demonstrated using the electrophysiological sensillum tip‐recording technique. The stimulating effect of 13 water soluble sugars at 100 mm is tested on the neurones of these sensilla. Sucrose and fructose are the two most stimulating sugars for the sugar‐sensitive neurone, evoking almost 30 spikes s?1 at 100 mm . The stimulating effect of arabinose, glucose, mannose, maltose and raffinose is three‐ to five‐fold lower, in the range 5.9–9.6 spikes s?1. The remaining six sugars, xylose, galactose, rhamnose, cellobiose, trehalose and lactose, have very low (<1 spikes s?1) or no ability to stimulate the sugar‐sensitive neurone. Concentration/response curves of the sugar‐sensitive neurone to sucrose, fructose and glucose at 0.01–100 mm overlap to a large extent in hibernating, cold reactivated and reproductively‐active beetles. A remarkable 9–50% decrease in the number of spikes evoked by 100 mm fructose and 10–100 mm sucrose occurs, however, in reproductively‐active beetles in June compared with beetles at the beginning of hibernation in October. These findings show that A. obscurus is capable of sensing a wide range sugars via their antennal gustatory sensilla.  相似文献

Sugar Absorption and Sucrose Inversion by Excised Tomato Roots   总被引:3,自引:0,他引:3  
The growth of excised tomato roots in sucrose is accompaniedby the appearance in the medium of glucose and fructose. Thequantitative relations between this appearance of glucose andfructose in the medium, total sugar absorption by the roots,and the decrease in the sucrose concentration of the mediumdo not suggest any causal relationship between sucrose uptakeand glucose and fructose appearance in the medium. Excised tomato roots exhibit surface invertase activity witha pH optimum at 4.0–4.4. Alterations of external pH, whichdid not affect sucrose absorption, drastically altered the levelsof glucose and fructose appearing in the medium. Glucose is preferentially absorbed from mixtures of glucoseand fructose, and by adjustment of the ratio of the two sugars,a mixture can be obtained from which equimolar absorption ofthe two sugars occurs. Root growth in this mixture is, however,very poor compared with that occurring in presence of sucrose. The results are discussed in the light of earlier studies onsucrose uptake by cultured tomato roots.  相似文献

Cultures of embryonal-suspensor mass of European silver fir (Abies alba) were screened for their response to various sucrose concentrations (29–175 mM) and to equimolar (100 or 200 mM) concentrations of glucose, fructose, galactose, sucrose, lactose and soluble starch. The higher concentrations were combined with 3.78 M abscisic acid. While fructose was ineffective in promoting any somatic embryo maturation glucose, sucrose and galactose promoted early maturation stages. Somatic embryo development up to the torpedo stage was observed only on media with soluble starch or lactose. Lactose was superior to other carbohydrate treatments and led to more synchronized maturation and more normally structured embryos. In comparison with the pronounced carbohydrate effect abscisic acid at this concentration proved to be of less importance.
