Many Disa species (80%) have not been germinated in vitro, fuelling conservation fears. Winter-rainfall species are typically germinable, whilst summer-rainfall species are exclusively intractable in vitro. We aimed to establish the in vitro germination requirements for previously ungerminated Disa species. Conventional asymbiotic seed culture protocols were reviewed and their efficacy in summer-rainfall Disa established. Mature seed was subjected to manipulations of media composition and viscosity, incubation temperature and illumination. Immature cultures were also established across four seed maturity classes. Four first-time germinations (D. cooperi, D. nervosa, D. pulchra and D. woodii) resulted from media with increased water availability. Germination rate (>12 weeks), percentage (<30%) and synchrony were not in accordance with values reported for winter-rainfall species (>80% germination in 8 weeks). Germination of immature seed under control conditions was similar to mature seed germination under modified conditions, but neither percentage approached the calculated germination potential (∼viability). Germination control in Disa is proposed as a trade-off between water availability and the presence of phyto-inhibitors in the environment of the embryo—two features typical of seeds exhibiting water-impermeable dormancy.  相似文献

The capacity of eight wetland seeds to be internally dispersed by common teal (Anas crecca L.) after a dry period typical of natural Mediterranean wetlands was experimentally quantified. These plant taxa (Chara spp., Echinochloacrus-galli (L.) P. Beauv., Eleocharis palustris (L.) Roem. & Schult, Polygonum lapathifolium L., Potamogeton nodosus Poir., Potamogeton pusillus L., Schoenoplectus mucronatus (L.) Palla, Scirpus maritimus L.) were frequently ingested by teal in the field. Captive teal were fed with known quantities of dry seeds and faecal samples were collected at intervals of 1-2 h for 48 h. The proportion of seeds retrieved after gut passage that germinated varied between taxa from 3% to 83%. From 2% to 63% of the seeds ingested were recovered intact, and 18-95% of these were still viable. Therefore, an overall 32% of the seeds ingested were evacuated in a viable condition, ranging from 0.2% for Chara spp. to 54% for S. mucronatus. Mean seed retention time was 3-5 h. Overall, 94% of seeds were evacuated within 12 h, but 2% were recovered after more than 24 h. Seed viability was reduced at longer retention times, although viable seeds of all taxa except Chara spp. were recovered 44-48 h after the experiment began. Germinability was increased by gut passage, although for some species this effect was only significant after stratification, indicating that they exhibit both mechanical and physical dormancy. Seed germination was accelerated by gut passage, especially at longer retention times. Gut passage increased the proportion of viable seeds, suggesting selective digestion of non-viable ones. Endozoochorous transport by teal appears to be a widespread dispersal mechanism among wetland plants.  相似文献

The impact of fire on hard-coated Cistaceae (Halimium ocymoides, Cistus ladanifer, and C. salvifolius) soil seed banks in a Mediterranean 'maquis' shrubland, and its effect on seed germinability were studied. The study also contrasts the effectiveness of two widely used techniques for quantifying seed banks, the seedling emergence and the physical separation methods, in relation to fire. The null hypothesis that a massive enhancement of physically-dormant Cistaceae seed germination by fire would make use of the time-consuming physical separation technique unnecessary was tested. Fire reduced Cistaceae seed banks in the 0–2 cm deep soil layer by both seed fire-consumption and lethal temperatures, revealed by the significant decreasing of the seed bank density and by the increase of apparently-intact but soft-unviable seeds, respectively. In contrast, no damage was recorded in the 2–5 cm soil layer. A dramatic seed bank depletion (> 90%) in both soil layers was recorded one year after fire in the burnt area, coinciding with a significant increase of seedling density confined to the first post-fire year. The ecological consequences of this massive post-fire seed bank input are discussed. A germinability test revealed that germination of surviving Cistaceae seeds was significantly enhanced in all cases except for the C. salvifolious seed bank in the deeper soil layer. However, final germination levels (60–75%) did not correspond to the magnitude of seed bank depletion, especially for C. salvifolious, which suggests that other environmental factors not exclusively associated with fire may also be important in softening Cistaceae seeds. Germination enhancement by fire soil-heating was not high enough to reject the physical separation technique, at least in the deeper soil layer. The simultaneous use of both seedling emergence and physical separation is recommended for reliable seed bank estimates when a physically-dormant hard-seeded component can be expected in the soil, as in many Mediterranean ecosystems, regardless of fire occurrence.  相似文献

Comparing AFLP, RAPD and RFLP markers for measuring genetic diversity in melon   总被引:14,自引:0,他引:14  
Three different types of molecular markers, RAPD, AFLP and RFLP were used to measure genetic diversity among six genotypes of Cucumis melo L. Each line represented a different melon genotype: Piel de Sapo, Ogen, PI161375, PI414723, Agrestis and C105. A number of polymorphic RAPD, AFLP and RFLP bands were scored on all materials and the genetic similarity measured. Clustering analysis performed with the three types of markers separated the genotypes into two main groups: (1) the sweet type, cultivated melons and (2) the exotic type, not cultivated melons. While the data obtained suggest that all three types of markers are equally informative, AFLPs showed the highest efficiency in detecting polymorphism. 目的:探讨不同含湿率及加载条件对人皮质骨纳米压痕测试的影响。方法:采用美国Hysitron纳米压痕仪,设定不同加载模式(高峰载荷分别为300、400、500 nm;加载速度分别为6、8、10 nm/s)对不同含湿率(20‰、30‰、40‰、50‰、60‰)人皮质骨进行弹性模量及硬度测量。结果:在同一加载模式下,不同含湿率标本的测试值随着含湿率增高,弹性模量及硬度值均显著降低(P0.01);三种不同加载模式对含湿率为20‰标本测试值无明显变化;对含湿率60‰标本测试值有显著性影响(P0.02)。结论:纳米压痕仪测试骨微观力学特性时,测试值不仅受标本本身含湿率的影响,当测试条件改变,对湿润标本测试结果也完全不同。当使用纳米压痕技术时,通常采用脱水包埋处理的标本测试出的机械性能,对我们在微观层面认识人体骨在湿润的生理环境下的机械性能是不够全面的。  相似文献

Moisture content (MC) is an important factor in the freshness of shiitake mushrooms (Lentinula edodes). In this study, we developed a simple prediction method for MC of shiitake mushrooms using near-infrared (NIR) imaging at 1450 nm, as the band at 1450 nm is strongly correlated with OH groups. First, a NIR image was captured by an InGaAs sensor with a bandpass filter of 1450 nm; the captured image was then calibrated using an image taken at 1100 nm to remove shade and shape effects of mushrooms. The actual MC of shiitake mushrooms was measured using an oven-drying method, and a calibration curve was obtained for the cap surface and the gill side, respectively. The correlation coefficient reached more than 0.85, and the standard error of calibration ranged from 3.1% to 8.2%. Non-destructive MC images were quantified using the constructed calibration curves, and the experimental results showed that differences in the water-holding property can be visualized over 6 h for each side using our method.  相似文献

花椒种籽油的含蜡量测定与脱蜡是长期困扰花椒种籽油处理的一项关键技术,本研究通过分析混合压榨制备的花椒种籽油,花椒籽种壳油及种仁油的含蜡量,研究了5种脱蜡方法脱除花椒籽油中蜡质的脱蜡效果,确定了脱除花椒籽油中蜡质的有效方法,研究结果表明,花椒籽油的含蜡量在15-20%,左右,而这些蜡质基本上都含在种壳油内一即种壳上,种仁油基本不含蜡质,脱除花椒籽油中蜡质的合理方法应是:(1)含蜡量相对较低的精制粗油可选用表面活性剂法脱蜡;(2)当含蜡量相对较高时,为降低脱蜡过程中油的耗损率可选用分步脱蜡法脱蜡;(3)需进行碱炼的油,可在碱炼过程中将蜡质与游离脂肪酸一半除去。  相似文献

利用 AFLP 技术对30个甜瓜材料进行多态性和聚类分析研究。从120对 MseI 和 PstI 引物中筛选出25对扩增效果好的引物,共扩增出262条多态性带。聚类分析结果新疆甜瓜中的夏甜瓜类型,新疆甜瓜中的冬甜瓜类型、美国粗皮甜瓜类型、日本甜瓜类型、梨瓜类型各聚为一组。上述材料的聚类分析结果与依据生态类型和地理起源的分类结果基本吻合,表明 AFLP 用于甜瓜种内不同材料间的遗传变异性分析是可行的。  相似文献

Differences in seed vigour of zero- and high-tannin faba beans were investigated using 25 seed lots of 12 cultivars following earlier reports of poor emergence in the zero-tannin types. Field emergence ranged from 54–96% indicating differences in seed vigour between cultivars all having high laboratory germination (>91%). Seed from zero-tannin accessions with poor emergence had a higher incidence of testa and cotyledon cracking, a smaller percentage of hard seeds, more rapid water uptake, a lower percentage of vital staining of cotyledons and a greater leaching of solutes than high-tannin types. Nevertheless, variation in these characteristics existed between cultivars and lines of both types. Seeds with more cracks in the seed coat and fewer hard seeds imbibed water more rapidly and consequently showed lower levels of vital staining and more cracks in the abaxial surface of the cotyledons. Slower imbibition in polyethylene glycol lessened the incidence of these deleterious characteristics and may provide a practical resolution to the problem of poor field emergence in zero-tannin lines of faba bean with low seed vigour.  相似文献

This study aimed to determine the effect of moisture content of three different feedstocks on overall ethanol yield. Switchgrass and sugarcane bagasse from two sources were either soaked in water (∼80% moisture) or left dry (∼12% moisture), and half each of these were impregnated with 3% w/w SO2 and all were steam pretreated. The twelve resulting substrates were compared based on overall sugar recovery after pretreatment, cellulose conversion following enzymatic hydrolysis, and ethanol yield following simultaneous saccharification and fermentation. The overall ethanol yield after simultaneous saccharification and fermentation of hexoses was 18-28% higher in samples that were soaked prior to SO2 addition than in SO2-catalyzed samples that were not soaked. In samples that were uncatalyzed, soaking made little difference, indicating that the positive effect of increased moisture content may be related to increased permeability of the biomass to SO2.  相似文献

Atriplex sagittata is an annual heterocarpic plant that produces three different fruit types (termed A, B, and C). To examine the consequence of heterocarpy on germinability patterns over long time periods, we compared seed germinability of different fruit types that had been stored for up to 8 years. While germinability of non-dormant type C fruits in distilled water was high (up to 100 %) in the first 2 years, it rapidly decreased over time. Dormant fruit types A and B showed increased germinability up to 7 years, though loss of germinability was lower for type B than for type A fruits. Eight-year-old fruits of all types had significantly lower germinability than younger fruits, probably due to loss of viability. Heterocarpy, therefore, ensures that emergence rates for seedlings of A. sagittata will be maintained over relatively long periods, even in years of strong disturbance when all adult plants may be destroyed. The experiment further showed that germinability of all fruit types in high concentrations of salt, as compared with water treatment, changed over the course of 8 years. Whilst dormant types (A and B) of A. sagittata show increased germinability with age of the seed in water treatment, they significantly lose germinability over time with salinity treatment. Type C fruit was not influenced by salt in the first year, but germinability rapidly decreased with time. It follows that the species is able to germinate under high salt concentration in the first year, but this advantage gradually disappears.  相似文献

Under suboptimal growing conditions (e.g. a lack of sunshine), floating leaves of Nymphaea alba and Nuphar lutea can become heavily infected with the fungi Colletotrichum nymphaeae and Pythium F, respectively. These fungi normally act as decomposers of senescent leaves. Mature leaves of Nymphaea alba and Nuphar lutea contain high concentrations of phenolics, secondary substances known for their fungistatic properties. The production of these compounds requires energy and primary metabolites. The hypothesis that suboptimal growing conditions reduce the ability of nymphaeids to maintain a sufficiently high level of phenolics, thereby making them more vulnerable to infection by fungi, was tested. Outdoor mesocosm experiments were used to examine the response of floating leaves of Nymphaea alba and Nuphar lutea to reduced light availability. Shading significantly reduced the phenolic content of the leaves. This was accompanied by higher disease severity. The melon, the echolocation organ of the striped dolphin (Stenella coeruleoalba), was investigated by morphological and high-resolution (13)C nuclear magnetic resonance (NMR) methods, in order to characterize structure and composition gradient at the histological and molecular level. Morphological analysis showed that the lipidic components are organized in an adipose tissue; moreover, a clear muscular component was observed. Age-related structural differences also were noted. Furthermore, NMR yielded detailed information at a qualitative-quantitative level on the lipid components.  相似文献

Summary A procedure leading to the regeneration of whole plants from protoplasts of melon is described. Protoplasts were isolated from cotyledons and leaves of plants grown in vitro. After 14 days of culture, average viability and division rates were respectively 60% and 30% for the two organs, considering total initial protoplasts plated. The manipulation of the exogenous auxin / cytokinin balance in regeneration media enabled to direct morphogenesis towards somatic embryogenesis (1 mg·l–1 2,4-dichlorophenoxyacetic acid and 0.1 mg·l–1 6-benzylaminopurine) or caulogenesis (0.5 mg·l–1 6-benzylaminopurine and 0.5 mg·l–1 kinetin). Contrary to division ability, regeneration capacity was genotype-dependent under our conditions, but the two organs expressed similar division and regeneration capacities. Maltose was superior to sucrose for the development of caulogenic nodules into buds. Blackthorn (Prunus spinosa L.) germination is often low, so new methods need to be developed with a view to improving nursery yields and to inform decision-making on natural regeneration. To this end, the effects of seed moisture content (MC) levels in combination with warm and chilling treatments on blackthorn seed dormancy release were investigated. In another experiment, the effect on seed germination of warm and chilling treatments in combination with exogenous hormones was investigated. Following treatment, the seeds were allowed to germinate at a constant 15°C with 8 h lighting per day or 20 (dark)/30°C (light). Seed lot effects were evident, but were consistent across treatments. Seeds adjusted to the lower target MC level (TMC) maintained high germination potential over a longer period of treatment than in those held in the fully imbibed (FI) state. The highest germination was achieved in the TMC seeds that were given six weeks warm treatment followed by 32 weeks chilling. Hormone treatments significantly reduced the amount of chilling needed to release dormancy in TMC seeds, but not in the FI seeds. Overall, germination response was better at 15°C test temperature than at 20/30°C.  相似文献

Fungi causes most plant disease. When fruits are stored at suboptimal conditions, fungi grows, and some produce mycotoxin which can be dangerous for human consumption. Studies have shown that the Penicillium and Monilinia species commonly cause spoilage of fruits, especially apples. Several other genera and species were reported to grow to spoil fruits. This study was conducted to isolate and identify fruit spoilage by fungi on apples collected in Riyadh, Saudi Arabia and conduct a molecular identification of the fungal isolates. Thus, we collected 30 samples of red delicious and Granny Smith apples with obvious spoilage from different supermarkets between February and March of 2012 in Riyadh, Saudi Arabia. Each apple was placed in a sterile plastic bag in room temperature (25–30 °C) for six days or until fungal growth was evident all over the sample. Growth of fungal colonies on PDA was counted and sent for molecular confirmation by PCR. Six fruit spoilage fungi were isolated, including Penicillium chrysogenum, Penicillium adametzii, Penicillium chrysogenum, Penicillium steckii, Penicillium chrysogenum, and Aspergillus oryzae. P. chrysogenum was the most frequent isolate which was seen in 14 of a total of 34 isolates (41.2%), followed by P. adametzii and A. oryzae with seven isolates each (20.6%) and the least was P. steckii with six isolates (17.6%). Penicillium species comprised 27 of the total 34 (79.4%) isolates. Sequence analysis of the ITS regions of the nuclear encoded rDNA showed significant alignments for P. chrysogenum, P. adametzii and A. oryzae. Most of these fungal isolates are useful and are rarely pathogenic; however they can still produce severe illness in immune-compromised individuals, and sometimes otherwise healthy people may also become infected. It is therefore necessary to evaluate the possible production of mycotoxins by these fungi to determine a potential danger and to establish its epidemiology in order to develop adequate methods of control.  相似文献

Amaranthus retroflexus L. is an importunate annual weed in many cropping systems of different countries. The main aim of this study was to investigate the effects of maternal nitrogen and drought stress on the seed dormancy and germinability of A. retroflexus. Field experiment was carried out in a factorial based on randomized complete block design, with four potential levels of soil water (–2, ?6, ?8 and ?10 bar) and three levels of nitrogen (0, 100 and 200 kg/ha). The germination characteristics of the seeds were measured at three different times (1 month, 6 months and 1 year after harvesting). Results showed that drought stress had positive effects on breaking of A. retroflexus seed dormancy until 6 months after seed harvesting. Seeds that were developed under severe water stress exhibited the highest germination percentage and germination rate. The results obtained from this study revealed that application of 100 kg/ha nitrogen during seed development increases germinability of A. retroflexus, whereas application of 200 kg/ha nitrogen induced seed dormancy. Furthermore, 100 kg/ha nitrogen application in the field along with 200 ppm gibberellic‐acid treatment during seed after‐ripening showed the highest germination percentage and germination rate for seeds after 6 months harvesting. Results also indicated that after‐ripening significantly increased seed germination and germination rate of A. retroflexus. These findings indicate that long‐term management of the soil seed bank in this species requires more stringent control due to the changes in germination timing, as detected in this study.  相似文献

Heavy metals are common contaminants in stormwater runoff. One of the devices that can be used to effectively and economically remove heavy metals from runoff is a yard waste compost stormwater filter. The primary goal of composting is to reduce waste volume rather than to produce stormwater filter media. Moisture content (MC) and initial pH, the two important parameters in composting, were studied for their effects on yard waste volume reduction and heavy metal adsorption performances of the compost. The main objective of this investigation was to examine whether the conditions that provided high yard waste volume reduction would also result in compost with good heavy metal removal performances. Manila grass was composted at different initial pHs (5–9) and MCs (30–70%) and the composts were used to adsorb cadmium, copper, lead and zinc from water. Results indicated that MC is more critical than initial pH for both volume reduction and production of compost with high metal adsorption performances. The most optimal conditions for the two attributes were not exactly the same but lower MCs of 30–40% and pH 7 or higher tended to satisfy both high volume reduction and effective metal adsorption.  相似文献

采用田间试验研究了421个常规籼稻、63个常规粳稻和82个籼型杂交稻品种种子含氮量的基因型差异。结果表明,栽培稻种子含氮量变幅在0.85%~2.55%,平均为1.39%。种子平均含氮量最高的是籼型杂交稻,高达1.46%,显著高于常规粳稻(1.39%)和常规籼稻(1.37%)。不同类型的栽培稻种子含氮量呈正态分布,但频数分布最高的区间存在一定的差异。以50%品种的种子含氮量(累积百分数从25%~75%)为依据,常规籼稻、常规粳稻、籼型杂交稻分别集中在1.21%~1.52%、1.21%~1.51%、1.36%~1.53%。LSR测验表明,籼型杂交稻极差显著小于常规籼稻和常规粳稻,但常规粳稻与常规籼稻的差异未达显著水平,即籼型杂交稻种子含氮量比常规籼稻和常规粳稻的分布更加集中。不同类型栽培稻内的种子含氮量极差较大,可分为若干组,且各组间达极显著差异。  相似文献
