The study assessed the effects of different roughage to concentrate ratios on enteric methane production, rumen fermentation and microbial counts. These ratios were 80:20, 50:50, and 20:80 for diets 1, 2, and 3, respectively. No significant differences were observed in total gas production among diets; however, methane emissions increased (P?<?0.05) with increased roughage in diet. The pH was greater (P?<?0.05) in diet 1 compared to diets 2 and 3 (6.38 vs 6.17 and 6.07). In vitro dry matter digestibility increased with decreased roughage ratios (47.67, 61.67, 67.33 % for diets 1, 2 and 3, respectively). Similarly, total volatile fatty acids (mM/100 mL) also increased with decreased roughage ratios [diet 1 (5.38); diet 2 (6.30); diet 3 (7.37)]. Methanogen counts, total bacterial counts and protozoal counts were lower (P?<?0.05) in diet 3 compared to diet 1 and 2. However, total fungal counts were higher in diet 1 compared to diet 2 and 3. The results indicate that methane emission, enteric fermentation patterns, and change in methanogens population appear only with higher level of roughage. These findings are important for reducing methane without any impact on rumen performance.  相似文献

Rancid and tart flavours are the most common off-flavours in Norwegian goat milk. In a feeding trial, we examined the effects of different levels of concentrate and of supplementary hay feeding on the occurrence of these off-flavours. Forty-eight experimental goats grazed cultivated pasture in the lowland during spring and autumn, and mountain pasture in summer. Twenty-four of the goats were fed 0.2 kg/day of concentrate (low concentrate, LC) and the remaining 24 goats 0.7 kg/day (high concentrate, HC). Twelve animals in LC and HC groups grazed freely, while the remaining goats in the two groups were given feeding challenges (limited periods of roughage restriction and ad libitum hay feeding). This was done to examine the effect of differing roughage availability on milk flavour. The animals fed low level of concentrate and with free grazing represented the control group. During the challenge periods, the goats were kept indoors and fed 0.2 kg hay/day for 2 days (roughage restriction) or hay according to appetite for 2 days (ad libitum feeding). At least 10 days of uninterrupted grazing were allowed between the challenges. Milk samples for sensory and chemical analysis were taken at the end of the challenge periods.The level of concentrate feeding did not influence the flavour of the milk when the goats were on lush spring pasture. However, the low level of concentrate feeding (LC) increased the frequency of off-flavour during the mountain and autumn experimental periods (P < 0.05).In the end of the mountain experimental period when the frequency of rancid/tart flavour increased in all groups, the sensory quality of milk produced by goats fed hay ad libitum (S1) had better taste ratings (P < 0.001) than milk from goats that were grazing. When the goats were given restricted amounts of hay, they adapted to the treatment by reducing their milk yield. In the spring and mountain experimental periods, restricted roughage feeding resulted in higher fat content and better taste ratings, especially in the group fed low level of concentrate (LC/S1). However, this connection was not clear in the autumn.Probably, the goats are able to handle a negative energy balance as long as mobilizable body reserves exist, but start to produce milk with lower dry matter content and higher frequencies of rancid/tart flavour when the body reserves are empty and the energy deficit is still pronounced.It is concluded that increasing the level of concentrate and feeding hay supplements will reduce the frequency of flavour defects at times when grazing is suboptimal or the pasture is of medium or low quality.  相似文献

Four lactating dairy cows were used in two experiments to study the effects of the roughage to concentrate ratio in the diet on nitrogen balance, plasma urea, urinary urea, milk urea and urinary purine derivatives. The use of the allantoin to creatinine ratio in spot samples of urine as an index of the urinary allantoin excretion was also evaluated. Four isoenergetic and isonitrogenous diets were formulated according to a 2 × 2 factorial arrangement. Factor I was concentrate content. The roughage to concentrate ratios were 65:35 and 35:65 for the high roughage and high concentrate diets, respectively. Factor II was fat content, which was 2.8% and 5.8% for the low and high fat diets, respectively. In Experiment 1 cows were fed diets with low fat content, and in Experiment 2 cows were fed diets with high fat content. In both experiments, diets were fed according to a change-over design. Nitrogen balance was not affected by the treatments. In cows fed high concentrate diets the amount and the proportion of nitrogen excreted in milk, as well as milk production was higher than in cows fed the high roughage diets. In both experiments, as an overall effect, the urea levels in plasma, urine and morning milk were higher, although the total urinary excretion of urea was lower, for the high concentrate diets. Urinary allantoin excretion was higher, although not significantly in Experiment 1, for the high concentrate diets. The allantoin to creatinine ratio in spot samples of urine showed the same pattern as the total allantoin excretion. Urinary creatinine excretion appeared to be affected by the diet.  相似文献

The objective was to reduce the incidence of liver abscesses (LAs) in young bulls by reducing the starch content of the concentrate and increasing the straw intake by adding molasses without reducing performance. Eighty-five Danish Friesian bulls (146 ± 2.6 kg live weight (LW)) were allocated to four treatment groups in a 2 × 2 factorial design and were ad libitum fed either a high (H) (43%) or a low (L) (25%) starch concentrate, and either chopped barley straw (S) or a mixture (Sm) of chopped barley straw and sugar-beet molasses (75:25). The bulls were slaughtered at 440 (±4.2) kg LW (11.3 months of age) on average. The concentrate dry-matter intake (DMI) was 7.5% higher (P < 0.001), the roughage DMI was 12% higher (P < 0.01), whereas the total net energy intake was 7% lower (P < 0.001) for the L compared with the H concentrate. By adding molasses to the straw, roughage DMI increased from 0.50 to 0.96 kg/day (P < 0.001). Average daily gain (ADG) was not affected by treatment. Feed conversion efficiency (FCE) for dry matter (FCEDM) was decreased (P < 0.001), whereas FCE for net energy (FCENE) was increased with the L concentrate (P < 0.01), suggesting a lower utilisation of the cell wall fraction with the H starch concentrate. There were no major effects of treatment on carcass characteristics. The higher roughage intake with Sm prevented rumen wall damage, but did not reduce the number of animals with LAs. The L concentrate did not affect the rumen wall but reduced the level of LA (2 v. 9 animals, P < 0.02). There was a higher level of respiratory diseases in animals fed the H concentrate (P < 0.05). The experiment showed that it was possible to maintain performance and reduce LA by using a lower starch content of the concentrate.  相似文献

A study was carried out to determine if the protozoal fauna of indigenous African wild ruminants was different from that found in their domestic counterparts and if the animal's diet influenced the number and types of protozoa. Samples of rumen contents were collected in 1997 and 2001 from various indigenous African wild ruminants in Kenya. All three ruminant feeding types were sampled: browsers or concentrate selectors (giraffe and Guenther's dik-dik); intermediate or adaptable mixed feeders (impala, Thomson's gazelle, Grant's gazelle and eland); grass or roughage eaters (hartebeest and wildebeest). Total concentration of ciliate protozoa and percentage generic distribution were determined. In general, protozoal concentrations were higher in concentrate selectors, followed by the intermediate or opportunistic mixed feeders and lowest in the grass and roughage eaters. Both Thomson's and Grant's gazelle were protozoa-free in the 2001 samples. Entodinium percentages were considerably higher in concentrate selectors and intermediate mixed feeders, compared to roughage eaters. Two genera of protozoa previously found in only a few African ruminants, Epiplastron and Opisthotrichum, were observed in several additional animal species and represent new host records. A difference was noted in the protozoal species composition of the indigenous wild ruminants from that previously observed in African domestic ruminants.  相似文献

The ruminal microbiome in herbivores plays a dominant role in the digestion of lignocellulose and has potential to improve animal productivity. Kankrej cattle, a popular native breed of the Indian subcontinent, were used to investigate the effect of different dietary treatments on the bacterial diversity in ruminal fractions using different primer pairs. Two groups of four cows were assigned to two primary diets of either dry or green forages. Each group was fed one of three dietary treatments for six weeks each. Dietary treatments were; K1 (50% dry/green roughage: 50% concentrate), K2 (75% dry/green roughage: 25% concentrate) and K3 (100% dry/green roughage). Rumen samples were collected using stomach tube at the end of each dietary period and separated into solid and liquid fractions. The DNA was extracted and amplified for V1–V3, V4–V5 and V6–V8 hypervariable regions using P1, P2 and P3 primer pairs, sequenced on a 454 Roche platform and analyzed using QIIME. Community compositions and the abundance of most bacterial lineages were driven by interactions between primer pair, dietary treatment and fraction. The most abundant bacterial phyla identified were Bacteroidetes and Firmicutes however, the abundance of these phyla varied between different primer pairs; in each primer pair the abundance was dependent on the dietary treatment and fraction. The abundance of Bacteroidetes in cattle receiving K1 treatment indicate their diverse functional capabilities in the digestion of both carbohydrate and protein while the predominance of Firmicutes in the K2 and K3 treatments signifies their metabolic role in fibre digestion. It is apparent that both liquid and solid fractions had distinct bacterial community patterns (P<0.001) congruent to changes in the dietary treatments. It can be concluded that the P1 primer pair flanking the V1–V3 hyper-variable region provided greater species richness and diversity of bacterial populations in the rumen of Kankrej cattle.  相似文献

The critical roughage part (CRP) of 2 diet types was determined in a cross-over design with 6 double-muscled and 6 normally conformated Belgian Blue bulls fitted with rumen cannulae. The roughage:concentrate ratio was lowered weekly until signs of a lack of physical structure were observed. For diet 1, consisting of maize silage and concentrates, the initial proportion of maize silage was 25% of DM but it decreased weekly with 5% units of DM. For the second diet, consisting of wheat straw and concentrate, 12% straw (DM basis) was provided during the first week and thereafter the proportion of straw decreased weekly with 3% units of DM. Several directly observable parameters (rumen pH, feed intake, bloat, faecal consistency) were evaluated weekly for each bull. Apart from these direct indicators of acidosis, also other parameters, whose results were only available after the end of the trial, were determined (volatile fatty acid profile, lactic acid concentration, chewing time). The roughage part between the part fed when signs of a lack of physical structure was first observed and the part that was fed the week before, was considered as the CRP. Most animals showed no acute signs of clinical acidosis (directly observable parameters) and finished the trial on a 100% concentrate diet. However, in sacco rumen DM-degradabilities of maize silage, grass silage and wheat grain was depressed considerably when low roughage diets were fed. Based on all observed parameters, the mean CRP was calculated to be 14.7% for diet 1 and 8.1% for diet 2. The beef type (double-muscled or not) had no influence on the CRP.  相似文献

It is well observed that feeding energy-dense diets in dairy cows during the dry period can cause metabolic imbalances after parturition. Especially dairy cows with high body condition score (BCS) and fed an energy-dense diet were prone to develop production diseases due to metabolic disturbances postpartum. An experiment was conducted to determine the effects of an energy-dense diet and nicotinic acid (NA) on production and metabolic variables of primiparous and multiparous cows in late pregnancy and early lactation which were not pre-selected for high BCS. Thirty-six multiparous and 20 primiparous German Holstein cows with equal body conditions were fed with energy-dense (60% concentrate/40% roughage mixture; HC group) or adequate (30% concentrate/70% roughage mixture; LC group) diets prepartum. After parturition, concentrate proportion was dropped to 30% for all HC and LC groups and was increased to 50% within 16 days for LC and within 24 days for HC cows. In addition, half of the cows per group received 24 g NA supplement per day and cow aimed to attenuate the lipid mobilisation postpartum. Feeding energy-dense diets to late-pregnant dairy cows elevated the dry matter (p < 0.001) and energy intake (p < 0.001) as well as the energy balance (p < 0.001) without affecting the BCS (p = 0.265) during this period. However, this did not result in any metabolic deviation postpartum as the effects of prepartum concentrate feeding were not carried over into postpartum period. Multiparous cows responded more profoundly to energy-dense feeding prepartum compared with primiparous cows, and parity-related differences in the transition from late pregnancy to lactation were obvious pre- and postpartum. The supplementation with 24 g NA did not reveal any effect on energy metabolism. This study clearly showed that energy-dense feeding prepartum did not result in metabolic imbalances postpartum in multiparous and primiparous cows not selected for high BCS. A genetic predisposition for an anabolic metabolic status as indicated by high BCS may be crucial for developing production diseases at the onset of lactation.  相似文献

The objectives of this trial were to develop multiple linear regression (MLR) models and three-layer Levenberg-Marquardt back propagation (BP3) neural network models using the Cornell Net Carbohydrate and Protein System (CNCPS) carbohydrate fractions as dietary variables for predicting in vitro rumen volatile fatty acid (VFA) production and further compare MLR and BP3 models. Two datasets were established for the trial, of which the first dataset containing 45 feed mixtures with concentrate/roughage ratios of 10∶90, 20∶80, 30∶70, 40∶60, and 50∶50 were used for establishing the models and the second dataset containing 10 feed mixtures with the same concentrate/roughage ratios with the first dataset were used for testing the models. The VFA production of feed samples was determined using an in vitro incubation technique. The CNCPS carbohydrate fractions (g), i.e. CA (sugars), CB1 (starch and pectin), CB2 (available cell wall) of feed samples were calculated based on chemical analysis. The performance of MLR models and BP3 models were compared using a paired t-test, the determination coefficient (R2) and the root mean square prediction error (RMSPE) between observed and predicted values. Statistical analysis indicated that VFA production (mmol) was significantly correlated with CNCPS carbohydrate fractions (g) CA, CB1, and CB2 in a multiple linear pattern. Compared with MLR models, BP3 models were more accurate in predicting acetate, propionate, and total VFA production while similar in predicting butyrate production. The trial indicated that both MLR and BP3 models were suitable for predicting in vitro rumen VFA production of feed mixtures using CNCPS carbohydrate fractions CA, CB1, and CB2 as input dietary variables while BP3 models showed greater accuracy for prediction.  相似文献

Based on repeatedly reported extensive pantothenic acid disappearance in the rumen, the present study is aimed at examining if pantothenic acid is used for a more efficient ruminal fermentation and microbial growth in an artificial rumen (Rusitec). Three substrates differing in roughage/concentrate ratio were incubated with and without the addition of Ca-D-pantothenate. Pantothenic acid was extensively degraded without notably influencing fermentation, microbial protein synthesis and the status of other B-vitamins such as riboflavin, vitamin B6 and niacin. Therefore, pantothenic acid supplementation cannot be expected to contribute to microbial benefit for the ruminant animal.  相似文献

Summary A review is made of the ruminant digestive system in its morphophysiological variations and adaptations relating to foraging behaviour, digestive physiology, to interactions between plants and ruminants and to geographic and climatic diversity of ruminants' ecological niches. Evidence is provided for evolutionary trends from an extreme selectivity mainly for plant cell contents and dependence upon a fractionated fore- and hindgut fermentation, to an unselective intake of bulk roughage subjected to an efficient plant cell wall fermentation, mainly in the forestomachs. The review is based on detailed comparative morphological studies of all portions of the digestive system of 65 ruminant species from four continents. Their results are related to physiological evidence and to the classification of all extant ruminants into a flexible system of three overlapping morphophysiological feeding types: concentrate selectors (40%), grass and roughage eaters (25%) and intermediate, opportunistic, mixed feeders (35%). Several examples are discussed how ruminants of different feeding types are gaining ecological advantage and it is concluded that ruminants have achieved high levels of digestive efficiency at each evolutionary stage, (including well-documented seasonal adaptations of the digestive system) and that ruminant evolution is still going on. Deductions made from the few domesticated ruminant species may have, in the past, biased scientific evaluation of the free-ranging species' ecology. The aim of this study was to investigate the effect of different dietary levels of concentrate on feed intake, digestibility, ruminal fermentation and microbial population in steers. Eight Nellore steers fitted with ruminal cannulas were used in a double 4 × 4 Latin square design experiment. The dietary treatments consist of four different proportions of concentrate to roughage: 30:70, 40:60, 60:40 and 80:20% in the dry matter, resulting in Diets 30, 40, 60 and 80, respectively. The roughage was corn silage, and the concentrate was composed of corn, soybean meal and urea. Apparent digestibility of organic matter and crude protein showed a linear association with concentrate proportion (= 0.01), but the increased concentrate levels did not affect the digestibility of fibre. The lowest ruminal pH-values were observed in animals fed with Diet 80, remaining below pH 6.0 from 6 h after feeding, while in the other diets, the ruminal pH was below 6.0 not before 12 h after feeding. After feeding Diet 80, the ammonia concentration in the rumen was significantly the highest. Higher dietary concentrate levels resulted in a linear increase of propionic acid concentrations, a linear reduction of the ratio acetic acid to propionic acid (p < 0.01) and a linear increased synthesis of microbial nitrogen (p < 0.001). The predicted production of methane was lower in diets with greater amounts of concentrate (p = 0.032). The population of methanogens, R. flavefaciens and R. albus decreased with higher concentrate levels, while the population of S. ruminantium increased (p < 0.05). The results indicate that greater amounts of concentrate do not decrease ruminal pH-values as much as expected and inhibit some cellulolytic bacteria without impairing the dry matter intake and fibre digestibility in Nellore steers.  相似文献

A study was made on the numbers and species of bifidobacteria present in the rumen of calves fed high-roughage and high-concentrate diets. With the roughage ration the bifidobacteria were not detectable in a 10(-3) dilution, whereas with the concentrate ration their number was high, usually in the order of 10(8) to 10(9)/ml of rumen fluid. The species most represented, identified by means of deoxyribonucleic acid-deoxyribonucleic acid hybridization tests, included Bifidobacterium ruminale, Bifidobacterium globosum, and an apparently new species.  相似文献

AIM: To determine the effect of different carbohydrate-based finishing diets on fermentation characteristics and the shedding of Escherichia coli and enterohaemorrhagic E. coli (EHEC) virulence genes in cattle faeces. METHODS AND RESULTS: The size of faecal E. coli populations and fermentation characteristics were ascertained in three experiments where cattle were maintained on a range of finishing diets including high grain, roughage, and roughage + molasses (50%) diets. Increased E. coli numbers, decreased pH and enhanced butyrate and lactate fermentation pathways were associated with grain diets, whereas roughage and roughage + molasses diets resulted in decreased concentrations of ehxA, eaeA and stx(1) genes, this trend remaining at lairage. In one experiment, faecal E. coli numbers were significantly lower in animals fed roughage and roughage + molasses, than animals fed grain (4.5, 5.2 and 6.3 mean log10 g(-1) digesta respectively). In a second experiment, faecal E. coli numbers were 2 log lower in the roughage and roughage + molasses diets compared with grain-fed animals prior to lairage (5.6, 5.5 and 7.9 mean log10 g(-1) digesta respectively) this difference increasing to 2.5 log at lairage. CONCLUSIONS: The type of dietary carbohydrate has a significant effect on E. coli numbers and concentration of EHEC virulence genes in faeces of cattle. SIGNIFICANCE AND IMPACT OF THE STUDY: The study provides a better understanding of the impact finishing diet and commercial lairage management practices may have on the shedding of E. coli and EHEC virulence factors, thus reducing the risk of carcass contamination by EHEC.  相似文献

In the captive Indian rhinoceros (Rhinoceros unicornis), two disease complexes with a high incidence—chronic foot problems and uterine leiomyomas—may be linked to excess body weight (BW). In this study, intake and digestion trials were conducted (by means of 7‐day weigh‐backs, and 5‐day total fecal collections, respectively) with 11 Indian rhinoceroses at four zoological institutions in Europe and the United States to quantify energy and mineral nutrition on conventional or roughage‐only diets. Diets comprising a variety of forages (grass hay only, a combination of grass hay and grass silage, straw, or a mixture of grass and legume hay) were offered as the roughage source, along with various concentrates, produce, and supplements. Water intake was quantified, and urine samples were obtained opportunistically. The animals consumed 0.5–1.1% of their BW in dry matter (DM) daily, with calculated digestible energy (DE, in megajoules MJ) values ranging from 0.27 to 0.99 MJ DE/kg BW0.75/day compared to an estimated requirement of 0.49–0.66 MJ DE/kg BW0.75/day. Seven of 11 rhinos (64%) fed restricted levels of concentrate plus forage consumed DE in excess of this estimate. Even on roughage‐only diets, some individuals consumed energy well above the presumed metabolic requirements. Hence, restriction of both concentrates and roughage may be important for weight management in this species. Water intake ranged from 30 to 49 mL/kg BW daily (3.4–5.2 L/kg ingested DM), similar to values that have been reported for domestic equids. Excretion amounts and patterns also resembled those found in horses. Endogenous fecal losses measured for Ca, P, Cu, Fe, and Zn indicate that the maintenance requirements of these minerals should be met in Indian rhinoceroses by diets that meet recommendations for domestic horses. It is particularly important to evaluate dietary adequacy in mineral nutrition in this species in concert with the need for restricted energy intake, especially with regard to the hypothetical involvement of a low Zn supply in chronic foot problems. AIM: To study the diversity of commensal Escherichia coli populations shed in faeces of cattle fed on different diets. METHODS AND RESULTS: Thirty Brahman-cross steers were initially fed a high grain (80%) diet and then randomly allocated into three dietary treatment groups, fed 80% grain, roughage, or roughage + 50% molasses. Up to eight different E. coli isolates were selected from primary isolation plates of faecal samples from each animal. Fifty-two distinct serotypes, including nine different VTEC strains, were identified from a total of 474 E. coli isolates. Cattle fed a roughage + molasses diet had greater serotype diversity (30 serotypes identified) than cattle fed roughage or grain (21 and 17 serotypes identified respectively). Cluster analysis showed that serotypes isolated from cattle fed roughage and roughage + molasses diets were more closely associated than serotypes isolated from cattle fed grain. Resistance to one or more of 11 antimicrobial agents was detected among isolates from 20 different serotypes. Whilst only 2.3% of E. coli isolates produced enterohaemolysin, 25% were found to produce alpha-haemolysin. CONCLUSIONS: Diverse non-VTEC populations of E. coli serotypes are shed in cattle faeces and diet may affect population diversity. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides new information on the serotype diversity and phenotypic traits of predominant E. coli populations in cattle faeces, which could be sources of environmental contamination.  相似文献

The purpose of this study was to improve the prediction of the quantity and type of Volatile Fatty Acids (VFA) produced from fermented substrate in the rumen of lactating cows. A model was formulated that describes the conversion of substrate (soluble carbohydrates, starch, hemi-cellulose, cellulose, and protein) into VFA (acetate, propionate, butyrate, and other VFA). Inputs to the model were observed rates of true rumen digestion of substrates, whereas outputs were observed molar proportions of VFA in rumen fluid. A literature survey generated data of 182 diets (96 roughage and 86 concentrate diets). Coefficient values that define the conversion of a specific substrate into VFA were estimated meta-analytically by regression of the model against observed VFA molar proportions using non-linear regression techniques. Coefficient estimates significantly differed for acetate and propionate production in particular, between different types of substrate and between roughage and concentrate diets. Deviations of fitted from observed VFA molar proportions could be attributed to random error for 100%. In addition to regression against observed data, simulation studies were performed to investigate the potential of the estimation method. Fitted coefficient estimates from simulated data sets appeared accurate, as well as fitted rates of VFA production, although the model accounted for only a small fraction (maximally 45%) of the variation in VFA molar proportions. The simulation results showed that the latter result was merely a consequence of the statistical analysis chosen and should not be interpreted as an indication of inaccuracy of coefficient estimates. ABSTRACT. Protozoal concentrations were determined in rumen and cecal contents of 20 blue duikers ( Cephalophus monticola ). Ten animals of each sex were fed either a high concentrate or high roughage diet. Rumen protozoa were present in 19 of the 20 animals and concentrations ranged from 4.5 to 33.7 × 106 per g of rumen contents. At the higher concentrations, protozoal cells equaled between 30–40% of the total rumen contents volume. No protozoa were found in cecal contents. Weight of rumen contents was higher in females than in males ( P < 0.01), and rumen protozoa concentrations were higher in males ( P < 0.05) and in those animals fed the high concentrate diet ( P < 0.05). All the protozoa were identified as belonging to a single species, Entodinium dubardi . However, an average of about 30% of the E. dubardi cells varied from the typical morphology of this species. These cells appeared to be on variation lines leading toward 7–10 other non-caudate species of Entodinium . The study aimed to examine effects of supplemented CLA to periparturient dairy cows receiving different concentrate proportions antepartum (a.p.) to investigate CLA effects on metabolism and immune function. Compared with adapted feeding, high-concentrate diet a.p. should induce a ketogenic metabolic situation postpartum (p.p.) to better understand how CLA works. A total of 64 pregnant German Holstein cows had ad libitum access to partial mixed rations based on concentrate and roughage 3 weeks before calving until day 60 p.p. A.p., cows received 100 g/day control fat (CON) or a CLA supplement, either in a low-concentrate (20%, CON-20, CLA-20) or high-concentrate diet (60%, CON-60, CLA-60). P.p., concentrate proportion was adjusted to 50% while fat supplementation continued. After day 32 p.p., half of the animals of CLA-groups changed to CON supplementation (CLA-20-CON, CLA-60-CON). A ketogenic metabolic state p.p. was not achieved and respective impacts of CLA could not be examined. Blood samples for isolation of peripheral blood mononuclear cells (PBMC) were collected on day −21, 7, 28 and 56 relative to calving. Blood chemistry samples were taken over the entire experimental period. Mitogen-stimulated proliferation of PBMC remained unaffected. Besides serum concentrations of triglycerides, total bilirubin, total protein, albumin and IGF-1, clinical-chemical serum characteristics remained uninfluenced by treatments. No post-supplementation effect could be observed. Measured blood metabolites and mitogen-stimulated proliferation of PBMC indicate that all groups had an increased metabolic stress around calving, whereby group CLA-20 was affected more severely. Overall, supplemented CLA did not positively affect metabolism or immune function of periparturient dairy cows. However, feeding CLA in a low-concentrate diet a.p. seems to increase liver stress around calving via reduced DMI.  相似文献

Biotin is involved in many vital metabolic pathways and must be provided for an efficient fermentation in the rumen, as well as for the intermediary metabolism of the host animal. Factors influencing ruminal biotin metabolism and output are widely unknown at present. Therefore, dairy cows fitted with permanent cannulas in the dorsal rumen and in the proximal duodenum were fed differently composed diets, and the biotin flow at the proximal duodenum was measured. The diets (on DM basis) consisted of 8.9 kg grass hay (Diet 1), 8.9 kg corn silage plus 2.0 kg concentrate (Diet 2), or 7.3 and 7.4 kg grass silage plus 10.0kg concentrate (Diets 3 and 4). The concentrate in Diets 3 and 4 contained 87% wheat and corn grain, respectively. The cows were pre-fed the rations for 21 days. Thereafter duodenal digesta was sampled every two h for 5 days. Cr2O3 served as a flow marker and the microbial proportion of total nitrogen at the duodenum was estimated by near infrared spectroscopy (NIRS). The duodenal flow of biotin was not related to biotin intake, but to the amount of fermented organic matter (FOM) and the amount of microbial protein (Biotin [mg/d] = 0.518 kg FOM - 0.300; r=0.85 and biotin [mg/d] = 0.012 x g microbial protein + 1.478; r = 0.84), irrespective of the composition of the diet fed. Mean daily biotin flow was 0.48 +/- 0.11 mg/kg FOM without any systematic effect of diet composition. The ruminal biotin balance, calculated as the difference between biotin flow at the duodenum and biotin intake, was positive (1.4 - 2.0 mg/d) in cows fed the mixed roughage/concentrate diets and negative (-0.71 mg/d) when the pure hay diet was fed.  相似文献
