The objective of this study was to characterize facial whorl characteristics in Holstein stud bulls (n = 485) and to investigate their association with measures of semen quantity and quality. Bulls had 1 (n = 454), 2 (n = 21) or no facial hair whorls (n = 5). Some 39% of bulls had whorls in the middle of their forehead, whereas 43% had whorls very low down on their forehead. Spiral whorls were present on 77% of bulls while 22% had elongated whorls. Abnormally shaped whorls were found more often on bulls with low whorls (27% versus 17%, P = 0.01) than on bulls with high or middle whorls. Semen production data were obtained from the AI center database to provide a representative sample of each sires most recent semen production characteristics. The number of semen collections per bull averaged 38 with a range of 1 (n = 9) to 272 (n = 1). The average number of days between collections was 5 but varied from 1 to 425 days. Average age of bulls at first recorded collection was 679 days and ranged from 307 to 4555 days. There were no significant differences between black and white bulls in semen quality or quantity. Semen quality over the summer months was reduced in comparison to the winter months. Age of the bull at time of collection had no effect on any of the semen quality traits. Older bulls had larger scrotal circumference and produced more semen (P > 0.01). Bulls with whorls located in the center of the forehead were not significantly different in semen quality or quantity for the 10 traits considered when compared to bulls with whorls located outside the center. In contrast to previous research findings with Angus cattle, semen attributes were not significantly different between bulls characterized with round (or spiral) epicenters and those with abnormally shaped whorls. The difference between the two studies may be due to the fact that Angus bulls have a higher percentage of abnormal elongated whorls compared to Holsteins.  相似文献

Detailed semen evaluations were carried out on approximately 363 Santa Gertrudis, 5/8 Brahman and Brahman bulls on 12 different properties across northern Australia, as part of systematic breeding soundness examinations. A subset of bulls (n=245) were subsequently mated in groups, to cows and heifers at bull:female ratios of 2.5-6.0%, with the paternity of resulting calves being determined by microsatellite DNA testing. Motility traits of semen and spermatozoa were moderately repeatable and correlated with each other, but were unrelated to calf output. The percentage of morphologically normal spermatozoa in ejaculates was moderately to highly repeatable (e.g. r=0.10-0.64). The most common morphological abnormalities seen were mid-piece abnormalities, in particular, distal mid-piece reflex associated with a cytoplasmic droplet. Semen quality, particularly percent normal spermatozoa, was consistently related to calf output. In general, bulls with <50% normal spermatozoa sired few calves while bulls with the highest calf outputs had >70% normal spermatozoa. The presence or absence of heparin binding proteins in semen did not influence calf output. Semen from 93% of tested bulls was positive for heparin binding proteins. These results confirm that examination of semen, in particular, evaluation of percent morphologically normal spermatozoa, should be included in the breeding soundness examination of bulls.  相似文献

The objectives were to standardize some methodological and analytical aspects of a direct technique to detect sperm-bound antisperm antibodies (ASAs) in bovine semen using flow cytometry, including the effects of prefixation of sperm membranes with formalin buffer solution and inclusion of dead cells in the analysis. Fourteen Angus bulls, including ASA-positive (experimentally induced ASAs) and 10 reproductively normal ASA-negative bulls, were used. Fixation of sperm membranes had no significant effect on the percentage of IgG- or IgA-bound spermatozoa detected by flow cytometry. However, including dead cells in the analysis increased the percentage of IgG-bound spermatozoa in fixed (live and dead 18.6 ± 9.7% and live 1.3 ± 0.5%; median ± SEM) and nonfixed samples (live and dead 18.8 ± 9.2%, live 1.5 ± 0.6%; P = 0.0029), as well as IgA-bound spermatozoa in fixed (live and dead 16.3 ± 6.4%, live 0.3 ± 0.5%) and nonfixed samples (live and dead 21.4 ± 4.6%, live 1.0 ± 0.5%; P = 0.0041) in semen from ASA-negative bulls. Intrasample, intra-assay, and interassay coefficients of variation (CV) were 0.8%, 4.6%, and 5.3%, respectively, for determination of sperm-bound IgG, and were 2.8%, 8.4%, and 40.3% for determination of sperm-bound IgA. Despite the high interassay CV for IgA determination, all ASA-positive bulls consistently had high percentages of IgA-bound spermatozoa. Flow cytometry correctly identified ASA-positive bulls. Confocal laser microscopy confirmed binding of ASAs to sperm heads and cytoplasmic droplets, and less frequently to midpieces and principal piece. In conclusion, although fixation was not necessary, dead cells should be excluded from the analysis, because ejaculates with a large proportion of dead cells can yield false-positive results. Flow cytometry was accurate and reliable for detection of sperm-bound IgG and IgA and discrimination between ASA-positive and ASA-negative bulls.  相似文献

Fertilizing potential of semen containing a high percentage of sperm with a proximal droplet was evaluated using IVF. Design criteria: (a) specified semen with >100 x 10(6) sperm/mL with >40% progressively motile spermatozoa, after collection via electro-stimulation; (b) designated a droplet group, bulls whose semen contained >30% spermatozoa with a proximal droplet and <25% with other morphological abnormalities, and a control group, with <25% abnormalities of any type; and (c) stipulated evaluations at 11 to 13 mo of age and again -4 wk later. At the initial evaluation, when a bull was assigned to the droplet group, the next bull meeting control criteria was designated his pair; 15 pairs in four herds were studied. Semen was extended in egg-yolk citrate, cooled to 5 degrees C over approximately 2.5 h, and held at 5 degrees C. After 20 to 44 h, spermatozoa were processed by swimup, incubated with heparin, and co-cultured with oocytes (35 to 56 oocytes/sample; 18 h). Ova were observed for cleavage approximately 42 h after co-culture, and further development was evaluated on day 8. At first evaluation, cleavage rates were 18 and 46% for droplet and control groups (P < 0.01); semen had 34 to 70% and 0 to 12% droplet spermatozoa. For 10 of 15 droplet bulls, <10% of ova were cleaved whereas cleavage rate was >15% for all control bulls. At second evaluation, only three droplet bulls still had >30% of spermatozoa with a proximal droplet. Cleavage rates increased accordingly; only four droplet bulls had <10% cleaved ova and 10 had >34% cleaved ova. Three control bulls had <10% cleaved ova and nine had > or = 34% cleaved ova. Considering all 60 ejaculates, correlation between percentage of spermatozoa with a proximal droplet and percentage of cleaved ova was -0.49 (P < 0.0 1). Correlations between percentages of motile or normal spermatozoa in field evaluations and outcome in IVF were 0.28 and 0.52. Laboratory evaluations of spermatozoa concomitant with preparation for IVF revealed that only incidence of proximal droplets appeared related to outcome in IVF. We concluded: (a) semen from most yearling beef bulls with a high incidence of proximal droplet spermatozoa had severely compromised IVF fertility; (b) as these bulls matured, the incidence of proximal droplets decreased, and IVF fertilizing potential increased; and (c) semen containing >30% spermatozoa with a proximal droplet is strong evidence that fertilizing potential of the bull will be low until the incidence decreases.  相似文献

Semen from 200 Holstein bulls in an artificial insemination center was examined for the frequency of craters on the surface of sperm heads, as visualized with the aid of differential interference contrast microscopy. Semen from 100 of these bulls was examined in more detail in 2 experiments by staining with eosin-aniline blue to determine the relationship of unstained spermatozoa, and spermatozoa with normal acrosomes with apical ridges to the incidence of craters and fertility. Only 3 of 100 bulls had a substantial incidence of craters (15 to 23%), whereas the average of the other 97 bulls in 2 experiments was 1 to 3%. The percentage of sperm cells with craters was correlated (P < 0.05) with the percentage of unstained spermatozoa (r = -0.29 and sperm cells with normal acrosomes (r = -0.52) but was not significantly correlated (r = -0.24) with the nonreturn rate. One bull with many sperm cells with craters was slaughtered, and the epididymal spermatozoa were examined. The high incidence of sperm cells with craters was limited to one side, with the testis on that side having 2 Sertoli cell tumors. The remaining 2 bulls as well as one other that produced 16% of sperm cells with craters did so only temporarily. Within a few months crater sperm production had decreased and semen quality increased. The condition usually appears to be transitory, presumably due to temporary stress.  相似文献

Yearling, grass-fed, beef bulls at the USDA Subtropical Agricultural Research Station, Brooksville, Florida, were assessed for physical and semen traits in January, April, July and October of 1991 (Trial 1) and 1992 (Trial 2). Bulls were given a breeding soundness evaluation (BSE) using revised semen and scrotal circumference (SC) criteria. In Trial 1, the bulls consisted of Angus (n = 15), Brahman (n = 14), Hereford (n = 15) and Senepol (n = 14). In Trial 2, the breeds were Angus (n = 15), Brahman (n = 16), Romosinuano (n = 13) and Nellore x Brahman (n = 9). Trial bulls generally showed delayed growth compared with grain-fed bulls in temperate environments. Breed influenced semen traits (percentage sperm motility, normal spermatozoa and those with primary abnormalities) in both trials. Temperate Bos taurus breeds (Angus, Hereford) were generally superior to Bos indicus breeds (Brahman, Nellore x Brahman). Tropically-adapted Bos taurus breeds (Senepol, Romosinuano) were intermediate for those traits tested. In general, tropically-adapted Bos taurus breeds were more similar in reproductive development to temperate Bos taurus than to Bos indicus breeds. Breed by test period interactions occurred and were mainly influenced by delayed sexual maturity of Bos indicus bulls. Qualitative semen traits increased with bull age, particularly from 12 to 18 mo. Scrotal circumference development was slower in the Bos indicus breeds. Bulls of satisfactory BSE status at 18.1 to 22 mo of age were 73.9% in Trial 1 and 58.5% in Trial 2. Brahman bulls had the least satisfactory BSE scores in both years (Trial 1, 44.4%; Trial 2, 22.2%). Most bulls failed to achieve satisfactory BSE status due to a small SC relative to age (Trial 1, 66%; Trial 2, 72%). The most efficacious use of the BSE was > or = 15 mo in Bos taurus bulls and > 18 mo for Bos indicus bulls. Although the BSE has proven to be useful for the assessment of young, pasture-raised bulls in semi-tropical environments, use of SC thresholds linked more with growth traits than with calendar age would improve comparisons of relative reproductive development in such bulls, particularly those of Bos indicus derivation.  相似文献

Eight young reproductively normal Brahman bulls (average age and bodyweight 20 months and 500 kg, respectively) received either cottonseed meal delivering 8.2 g free gossypol/bull/d (treatment group, n=4) or soybean meal (control group, n=4) for 12 wk. After adjustment (1 wk), weekly procedures (11 wk) included blood collection, scrotal circumference measurement and electroejaculation. Semen assessments included sperm motility, percentage of live spermatozoa, general sperm morphology (using brightfield microscopy), and midpiece morphology (using DIC microscopy). After sacrifice (Week 12), sperm production rates (daily and per gram testicular parenchyma) were determined. Treated bulls did not differ from controls in scrotal circumference or the percentage of live spermatozoa. Sperm motility differed at Weeks 9 (P<0.05), 10 and 11 (both P=0.06). Treated bulls had fewer normal spermatozoa at Weeks 5 (P<0.05), 6 (P<0.01) and 7 thru 11 (P<0.001). Beginning from Week 3, treated bulls showed an increased proportion of sperm midpiece abnormalities (P<0.05) which stabilized at 52 to 62.5% between Weeks 5 and 11 (P<0.01 or P<0.001). Treated bulls also had lower sperm production than untreated bulls, both on a daily (P<0.01) and per gram testicular parenchyma (P<0.001) basis. A cottonseed supplement providing 8.2 g of free gossypol per bull per day had adverse effects upon both sperm morphology and spermatogenesis in young Brahman bulls, with the former being first evident within 3 to 4 weeks of feeding of cottonseed meal.  相似文献

The primary objectives were to describe beef bulls considered for use and those reported as used in 205 beef herds in western Canada, and to determine whether factors typically assessed during breeding soundness evaluations were associated with reproductive success. More than 100 veterinary clinics reported 2990 breeding soundness evaluations for bulls considered for natural service in client's herds. Differences among clinics explained 5.2% of the variation in scrotal circumference (SC) and 6.9% of the variation in percentage of morphologically normal sperm of all bulls considered for use (after accounting for age, breed, body condition, significant physical abnormalities, month, and year). The percentage of morphologically normal sperm was lower in bulls with an SC ≤34 versus >34 cm (P < 0.006). This study included data from 1384 and 1370 bulls used for breeding in 2001 and 2002, respectively. Most (80%) of the bulls used were Simmental, Black Angus, Charolais, Red Angus, or Hereford, and 80% were ≤4 y of age. Before the breeding season, a veterinarian evaluated 89.5% of all bulls used in these herds. Of the bulls subjected to a breeding soundness evaluation and subsequently used, 93.1% were satisfactory. In 2001 and 2002, injuries were reported in 2.5 and 2.1% of bulls and in 16.6 and 11.4% of herds, and necrobacillosis of the foot was reported in 2.5 and 1.2% of bulls and 11.2 and 6.5% of the herds. The average number of cows exposed to each bull was 26 (both years). Cows exposed to bulls with a smaller SC were less likely to be diagnosed pregnant (P < 0.047) and had a longer median interval from first bull exposure to calving (P < 0.016) than bulls with a larger SC. In conclusion, our findings emphasized the value of breeding soundness evaluations, including measurements of SC, in fertility management of beef cattle.  相似文献

The relative content of DNA in spermatozoa presumed to be the X- and Y-chromosome-bearing gametes from bulls, boars, rams and rabbits and the amount of DNA in spermatozoa of cockerels was determined by flow cytometry. Differences in the relative content of DNA and proportions of the presumed X- and Y-sperm populations in cryopreserved semen from Holstein, Jersey, Angus, Hereford and Brahman bulls were also determined. Spermatozoa were washed by centrifugation using a series of dimethyl sulfoxide solutions made in isotonic sodium citrate, fixed in ethanol, treated with papain and dithioerythritol to loosen the chromatin structure and remove cellular organelles, and stained quantitatively for DNA with the fluorochrome 4'-6-diamidino-2-phenylindole (DAPI). Approximately 5000 stained sperm nuclei, which were nonviable due to the removal of other cellular organelles during the washing procedure, were measured for DNA in an epi-illumination flow cytometer. A single distinct peak for cockerel spermatozoa and two symmetrical, overlapping peaks for species with X- and Y-spermatozoa were seen. This and other evidence strongly supports the interpretation that the peaks represent the X- and Y-sperm populations. The content of DNA in sperm nuclei from cockerels, bulls, boars, rams and rabbits, as determined by fluorescence flow cytometry, corresponded to biochemical estimates of DNA per sperm cell. Analyses of the bimodal histograms by computer-fitting two Gaussian distributions to the data showed the means of the peaks differed by 3.9, 3.7, 4.1 and 3.9% for bulls, boars, rams and rabbits, respectively. In four replicate analyses of semen from 25 bulls representing 5 breeds, the average population of sperm nuclei in the Y-peaks ranged from 49.5 to 50.5% for all breeds. The X-Y peak differences did not vary within each breed, but were significantly different when the breeds were compared. Spermatozoa from Jersey bulls had larger X-Y peak differences (P less than 0.001) than spermatozoa from Holstein, Hereford, and Angus bulls; spermatozoa from Brahman bulls had smaller X-Y differences (P less than 0.004). It is suggested from the evidence obtained in these studies that flow cytometry can be used to assess the proportion of X- and Y-spermatozoa in semen of domestic animals and is thereby applicable to verification of the effectiveness of enrichment techniques for X- or Y-spermatozoa.  相似文献

Poor semen quality has long been associated with bull infertility. However, the molecular basis in spermatozoa cells underlying the mechanisms of bull infertility remain unknown. The purpose of this study was to determine whether there is any protein in bovine spermatozoa related to semen quality. Semen samples from 18 Brahman bulls, 3 to 10 yrs of age, were assessed for semen quality in terms of spermatozoa motility and spermatozoa morphology. Spermatozoa extracts were separated using 2D-PAGE followed by staining with Coomassie blue. At least one duplicate gel was performed for each sample. Each gel was scanned with an ImageScanner System and analyzed for spots by ImageMaster 2D platinum software. The related protein spot(s) with semen quality was cut from the gel and identified by LC MS/MS. The results showed that at least 600 protein spots were detected in the spermatozoa extracts of the Brahman bulls. Of all these spots, there were 3 of 56 kDa at pI 6.4, 6.6 and 6.8 (Z1, Z2 and Z3, respectively) that clearly showed different expression pattern among 18 Brahman bulls. Of 18 bulls (a) five showed the presence of spot Z1 and Z2 (pattern A) (b) one of spot Z3 (pattern B) (c) five of spot Z2 and Z3 (pattern C) (d) one of spot Z1 (pattern D) and (e) six of spot Z2 (pattern E). Identification of spot Z1, Z2 and Z3 by LC MS/MS had a similar result as matched to the tektin-4 protein of Bos taurus with a respective score of 171, 557 and 591. The statistical analysis of the 56 kDa protein patterns, tektin-4, indicated a significant effect on spermatozoa motility (P < 0.05) albeit non-significant on spermatozoa morphology. The bulls which showed pattern A had a higher percentage of spermatozoa motility than pattern E (P < 0.05) and not different from pattern C (P > 0.05). The statistical analysis also revealed that the presence of spot Z1 had an effect on the percentage of spermatozoa motility (P < 0.01), whereas the presence of spot Z2 and Z3 had no effect (P > 0.05). The correlation coefficient between the relative protein content of spot Z1 and the percentage of spermatozoa motility was 0.49. Our study demonstrates that the expression patterns of tektin-4 were a proxy for an effect on spermatozoa motility and consequently bull infertility. It may be that these protein patterns can be used as markers for improving bovine reproduction.  相似文献

Bull breeding soundness parameters, semen characteristics and sexual behavior were evaluated for effects on reproductive performance in single-sire beef herds. A total of 155 cow herds (Angus, 50 herds; Hereford, 40 herds; Brahman, 46 herds; and Senepol, 19 herds) bred to bulls of the same breed were observed for 8 yr. All bulls produced adequate quality semen and had scrotal circumference (SC) >/= 30 cm. Reproductive performance was evaluated by the conception rate (CON), conception rate during the first 21 d of the breeding season (21dCON), mean calving date (MCD), and mean calving date of the first half of the herd to calve (HHCD). Correlations were determined between breeding soundness parameters and reproductive performance for all bulls combined, by breed, and by age. The Cp statistic was used to select models for the effects of parameters on CON, 21dCON, MCD and HHCD. Breeding season length and breed had significant effects. The percentages of normal cells, proximal droplets, detached heads and the semen score (motility plus percentage of normal cells) had a significant effect on CON when all bulls were considered. After the effect of season was deleted, the most significant parameter affecting CON in the Brahman was the percentage of detached sperm heads. In the Angus, motility was significantly correlated with all reproductive performance indices. In the Hereford, breeding soundness examination score (BSE) was positively correlated with 21dCON.  相似文献

Spermatozoa were not found in the urine collected from 12 bulls before electroejaculation, whereas spermatozoa were found in the urine collected after electroejaculation. The concentration of spermatozoa in four consecutive samples of urine collected during the first postelectroejaculation micturition did not differ (P > 0.80) within bulls, suggesting that the spermatozoa found in the urine were those that had flowed into the urinary bladder during electroejaculation. The mean percentage of retrograde flow was 21% and ranged from 1 to 50% for the 12 bulls. These findings demonstrate that there was a significant urinary loss of spermatozoa during electroejaculation.  相似文献

An IVF and culture system was used to determine the effect of the knobbed acrosome defect in bovine spermatozoa on fertilization and early embryonic development. Three bulls affected with knobbed acrosomes were identified as K+ (flattened acrosome), K2+ (indented acrosome) or K3+ (deep indentation of the acrosome) based on the predominant type of acrosomal aberration present in sperm of the respective bulls. After swim-up, all semen traits, except for acrosome morphology, were similar between bulls with varying degrees of the knobbed acrosome defect and a control bull, C. The mean number of spermatozoa bound to the zona pellucida was lower (P< 0.05) for the bulls with the knobbed acrosome defects (40.3 +/- 2.3, 29.5 +/- 1.6, 14.6 +/- 1.3, respectively, for Bulls K+, K2+ and K3+) than for Bull C (52.3 +/- 2.3). The percentages of zonae pellucidae penetrated by spermatozoa from Bulls K+ (51.2%), K2+ (49.5%) and K3+ (37.1%) were lower than that of Bull C (84.5%). No sperm with knobbed acrosome defects were found to have penetrated the zona pellucida. Fertilization rates for bulls with the knobbed acrosome defect, K+ (63.0%), K2+ (62.7%) and K3+ (22.6%), were significantly lower than that of the control bull (82.8%). Percentages of cleaved embryos, morulae and blastocysts produced were also lower for the bulls with knobbed acrosomes than that of the control bull. Results indicate that sperm with the knobbed acrosome defect had a reduced ability to bind to the zona pellucida, depending upon the severity of the defect, and that these aberrant spermatozoa did not penetrate the zona pellucida. The apparently normal spermatozoa coexisting in the inseminate of bulls with a high percentage of knobbed spermatozoa were also functionally deficient; oocytes penetrated by these spermatozoa had a reduced potential for fertilization, and resulting zygotes had a reduced ability for cleavage and embryonic development to the blastocyst stage. The results of the present study do not support the hypotheses that the knobbed acrosome defect is compensable.  相似文献

The semen characteristics of 12 Zebu/Friesian crossbred bulls, aged 2 to 3 years, were studied during a 21-month period. At the 12th month of the study, the commencement of the rainy season, the bulls were infected naturally with Dermatophilus congolensis . Lesions were scattered over the body and limbs, but were particularly pronounced on the scrotum. Monthly treatments with injection of terramycin were begun as soon as lesions were detected and continued until the end of the study. The lesions worsened and became pronounced particularly on the scrotum of all the bulls. Scrotal scab formation caused by infection became prominent at the 14th month of the study. Until that period, the bulls had normal semen characteristics. From the 15th month until the end of the study, there was progressive deterioration of semen characteristics in all the bulls; this was manifested by some or all of the following effects: decreased volume, increased percentage of dead spermatozoa, increased percentage of total sperm morphological abnormalities, decreased percentage of progressively motile spermatozoa, oligospermia and terminal azoospermia. Therefore, severe chronic scrotal dermatophilosis may be a significant cause of infertility or sterility in bulls.  相似文献

Nine Simmental X Angus bulls (2-yr of age) were used in 2 experiments. In Experiment 1, the scrotal neck was insulated (from Day 1 to Day 8) in 5 bulls, and semen was collected from all 9 bulls by electroejaculation approximately every 3 d until Day 35. Bulls with insulated scrotal necks had lower percentages of normal spermatozoa (P < 0.08) and higher percentages of spermatozoa with head defects (P < 0.06) or droplets (P < 0.08) than the untreated bulls. There was a time-by-treatment interaction (P < 0.04) for midpiece defects; the incidence was higher (P < 0.05) in the insulated than noninsulated bulls from Day 5 to Day 32. Spermatozoa within the epididymis or at the acrosome phase during insulation appeared to be the most affected. Compared with the noninsulated bulls, the insulated bulls had twice as many (P < 0.02) spermatozoa with midpiece defects and 4 times as many (not significant) with droplets on Day 5, fewer (P < 0.04) normal spermatozoa and 3 times as many with midpiece defects (P < 0.05) and with droplets (not significant) on Day 8, fewer (P < 0.02) normal spermatozoa on Days 15 and 18, and more sperm cells (P < 0.05) with head defects on Days 18 and 21. In Experiment 2, scrotal subcutaneous temperature (SQT; degrees C, mean +/- SE) prior to and after the scrotal neck had been insulated for 48 h in all 9 bulls was 30.4 +/- 0.7 and 32.4 +/- 0.6 (P < 0.01) at the top, 30.3 +/- 0.7 and 31.8 +/- 0.6 (P < 0.03) at the middle, and 30.2 +/- 0.8 and 30.7 +/- 0.6 (P < 0.05) at the bottom of the scrotum. Concurrently, there was an increase (0.9 degrees C) in intratesticular temperature (ITT) at the top (P < 0.07), middle (P < 0.04), and bottom (P < 0.04) of the testes. Scrotal surface temperature (SST) prior to and after the scrotal neck had been insulated for 24 h was 29.2 +/- 0.7 and 28.2 +/- 0.4 (P < 0.05) at the top of the scrotum and 24.7 +/- 0.6 and 25.3 +/- 0.7 (not significant) at the bottom, resulting in SST gradients of 4.6 +/- 0.6 and 2.9 +/- 0.5, respectively (P < 0.05). However, after the scrotal neck had been insulated for 48 h, none of the SST end points were significantly different from those prior to insulation. It appears that compensatory thermoregulatory mechanisms restored SST but were not able to restore SQT and ITT. Insulation of the scrotal neck affected SST, SQT, ITT and semen quality, emphasizing the importance of the scrotal neck in scrotal/testicular thermoregulation.  相似文献

A study was designed to investigate relationships between testicle size and histological, sperm production and endocrinological traits in yearling beef bulls at the end of performance test. Twenty-five beef bulls, (Hereford, n=16; Angus, n=4; and Charolais, n=5), with scrotal circumference (SC) measurements ranging from 28.5 to 36.0 cm, were used. Just prior to slaughter at 15 mo of age, SC measurements were taken, semen was collected, and a GnRH response test was conducted. Testicles were processed for daily sperm production (DSP), epididymal sperm reserves (ESR), seminiferous epithelial area (SEA), and degree of germinal epithelial loss (DGEL). There were significant positive correlations between SC and testicular weight (P<0.05), DSP/g (P<0.02), and DSP/bull (P<0.01) and ESR (P<0.01); however, the correlation between SC and SEA was not significant (P=0.4). Scrotal circumference was negatively correlated with DGEL (P<0.05). Degree of germinal epithelial loss was also negatively correlated with DSP/g, DSP/bull and ESR (P<0.01). Morphological characteristics of spermatozoa were diversely related to sperm production traits, and the percentage of normal spermatozoa was positively related to SC (P<0.02) and negatively related to DGEL (P<0.001). Gonadotropin releasing hormone stimulation did not reveal evidence of gonadotropin deficiency in any of the bulls. However, peak testosterone levels were lower in bulls with SC below 31 cm (P<0.05) than those with SC measurements above 31 cm.  相似文献

A study was designed to determine the in vitro fertilizing characteristics of bovine semen with a high percentage of spermatozoa with multiple nuclear vacuoles. In Experiment 1, a total of 620 oocytes was divided into 2 groups and inseminated with spermatozoa from 1 of 2 different bulls at a concentration of 2 × 105/ml. After Percoll washes, 73.5 ± 3.0% of spermatozoa from Bull A contained multiple nuclear vacuoles, while no sperm cells from Bull B contained vacuoles. After 19.5 ± 0.5 h of co-incubation of oocytes with spermatozoa, loosely attached sperm cells were removed by washing, and the oocytes were fixed between 2 poly-l-lysine coated glass slides. Mean (±SD) percentage of fertilization was significantly lower (P < 0.05) in Bull A (19.7 ± 7.0%) than in Bull B (67.6 ± 4.5%). In one-third of the oocytes fertilized by spermatozoa from Bull A, sperm head decondensation was incomplete and normal male pronucleus formation did not occur. All oocytes fertilized by Bull B had normally decondensed sperm heads. Although fewer (P < 0.05) spermatozoa from Bull A were bound to the zona pellucida than from Bull B, the percentage of vacuolated sperm cells bound to the zona pellucida (73.3 ± 7.8%) did not differ from that in the inseminate. The mean number of sperm cells binding to fertilized oocytes was higher than to unfertilized oocytes for both bulls (P < 0.05). In Experiment 2, 748 salt-stored oocytes (zonae) were inseminated with semen from the same 2 bulls to determine the ability of spermatozoa to penetrate the zona pellucida. The percentage of zonae penetrated by spermatozoa from Bull A (69.9 ± 3.5%; a mean of 2.4 ± 2.3 spermatozoa) was lower (P < 0.05) than from Bull B (96.5 ± 14.7%; a mean of 11.3 ± 9.9). Although the proportion of vacuolated sperm cells from Bull A that bound to the zona pellucida did not differ from that in the inseminate, the proportion of those penetrating the zona pellucida (52.7%) was lower (P < 0.05). In summary, vacuolated sperm cells apparently gained access to the oocyte and bound to the zona pellucida, but they penetrated the zona pellucida at a lower rate and apparently did not form normal male pronuclei.  相似文献

The diadem/crater defect was studied over several months in two related 20-month-old Angus bulls. In bull 1, diadem/crater defects were present in 2-99% of ejaculated spermatozoa at various times during the evaluation period. In bull 2, affected cells varied from 20% to 94%, with other abnormalities (head and acrosome defects, coiled tails, proximal cytoplasmic droplets) also common. Single sire mating trials conducted over 26 days during an apparent recovery phase showed normal fertility (approximately 50% pregnancies per estrus exposed). Both resting and gonadotropin-releasing hormone (GnRH)-stimulated testosterone values were within nor-mal limits. Histopathological evaluation of testes showed no obvious hypoplastic, inflammatory, or degenerative condition. Electron microscopy of ejaculated spermatozoa demonstrated the characteristic diadem pattern of craters in the equatorial region of the head. Many cells from bull 2 contained large craters in other regions of the nucleus. Electron microscopy of testicular tissue demonstrated nuclear invaginations lined by a single unit membrane in round spermatids. Lesions in elongated spermatids were more pronounced, with curling of the nucleus and large membrane-filled cavities in the chromatin occurring in addition to craters in the equatorial region of the nucleus.  相似文献

The relationship between nonreturn rates of bulls in a commercial artificial insemination program and in vitro induction of acrosome reactions in frozen-thawed spermatozoa by the calcium ionophore, A23187, was investigated. Washed spermatozoa from 3 to 5 ejaculates, collected from each of 23 Holstein bulls, were incubated for 1 h with 1 microM A23187. Acrosome reactions were determined by fluorescence microscopy. The percentage of increase in acrosome reaction in the ionophore-treated compared with control samples was significantly correlated to the 90-d nonreturn rate of the bulls (r = 0.86; P < 0.001). In a second experiment, a significant correlation was obtained between the fertility of bulls predicted on the basis of induced acrosome reaction and achieved 90-d nonreturn rate (r = 0.84; P < 0.005). No other assessments of semen quality (post-freezing motility, percentage of morphologically normal spermatozoa) was significantly correlated with fertility. Finally, the regression between acrosome reaction induction obtained from young bulls was used to predict the fertility of mature bulls whose semen was in widespread use (actual versus predicted nonreturn rate, r = 0.88; P < 0.0001).  相似文献

To determine the effect of breed on growth and reproductive development, weaned bulls in each of 2 yr were managed as a single group for approximately a year. In Year 1, the study group consisted of 24 Angus, 24 Brahman, 20 Hereford and 14 Senepol bulls, while in Year 2, it contained 25 Angus, 17 Brahman. 13 Romosinuano and 9 Nellore x Brahman bulls. Body and testicular growth measurements were recorded at 6-wk intervals. At approximately 1 yr of age and quarterly thereafter (4 periods), bulls were evaluated for libido, pubertal status, and GnRH-induced LH and testosterone secretion. Significant breed-by-age interactions occurred for most growth measurements. Brahman bulls (Bos indicus ) were (P < 0.05) older and heavier at puberty than Angus, Hereford, Senepol and Romosinuano bulls (Bos taurus ). Libido scores were lowest for Brahman and Nell ore x Brahman bulls (Bos indicus ). highest for Angus and Hereford bulls (temperate Bos taurus breeds) and intermediate for Senepol and Romosinuano bulls (tropical Bos taurus breeds; P < 0.05). Differences were not consistent among breeds or between years for GnRH-induced LH secretion. In both years, basal testosterone concentrations and areas under the GnRH-induced testosterone curve were higher (P < 0.05) for Angus and Hereford bulls (temperate breeds) than for Brahman, Senepol, Romosinuano and Nellore x Brahman bulls (tropical breeds). In conclusion, reproductive development of Senepol and Romosinuano bulls (tropical Bos taurus breeds) was more similar to Angus and Hereford bulls (temperate Bos taurus breeds) than to Brahman and Nellore x Brahman bulls (Bos indicus ).  相似文献
