The effect of plum pox virus (PPV) infection on the response of some antioxidant enzymes was studied in two apricot cultivars, which behaved differently against PPV infection: cultivar Real Fino (susceptible) and cultivar Stark Early Orange (cv. SEO, resistant). In the susceptible cultivar, PPV produced a decrease in Φ PSII, F 'v/ F 'm and Q p. PPV infection produced a drop in p -hydroxy mercury benzoic acid (pHMB)-sensitive ascorbate peroxidase, dehydroascorbate reductase and peroxidase in the soluble fraction from susceptible plants, whereas in the resistant apricot cultivar, pHMB-insensitive ascorbate peroxidase, monodehydroascorbate reductase, glutathione reductase and superoxide dismutase increased. However, catalase decreased in the soluble fractions from both infected cultivars. Long-term PPV infection also produced a decrease in the chloroplastic ascorbate–glutathione cycle enzymes only in the susceptible plants. As a consequence of PPV infection, an oxidative stress, indicated by an increase in lipid peroxidation and in protein oxidation, was produced only in the leaves from the susceptible cultivar which was also monitored by the diaminobenzidine peroxidase-coupled H2O2 probe. The loss of Φ PSII, indicative of activated oxygen species production, and the decrease in the levels of antioxidant enzymes in chloroplasts from susceptible plants could be responsible for the chlorosis symptoms observed. The results suggest that the higher antioxidant capacity showed by cv. SEO could be a consequence of a systemic acquired resistance induced by PPV penetration in stem tissue at the graft site and could be related, among other factors, to their resistance to PPV.  相似文献

The peroxidase activities in leaves from resistant and susceptible cultivars of wheat infected and non-infected by Erysiphe graminis DC were studied. In non-infected wheat, soluble and ionic bound peroxidase activity level was found to be higher in the resistant cultivar than that in the one susceptible to Erysiphe graminis DC. After infecting wheat leaves with Erysiphe graminis DC a remarkable increase in the activity of soluble and ionic bound peroxidases was detected 5 days after inoculation only in the resistant cultivar. In the susceptible cultivar a high increase in the activity of the soluble and ionic bound peroxidases occurred only 15 days after inoculation. Using ion exchange chromatography four peroxidase fractions were obtained from infected susceptible and resistant cultivars as from non-infected ones. The fraction II in non-inoculated resistant cultivars was much higher than that in the susceptible one. This fraction increased after inoculation in both cases reaching a higher level in resistant cultivars. Fraction I was higher in the susceptible cultivar. Electrofocusing profiles of peroxidase from the susceptible and resistant cultivar differed from one another. New peroxidase bands after inoculation appeared only in the resistant cultivar.  相似文献

The content of phenolic compounds as well as the activity of polyphenoloxidase and peroxidase in the red raspberry callus tissue after D. applanata infection were investigated. Two red raspberry cultivars: Latham — relatively resistant and M. Promise — susceptible were used. In the M. Promise cultivar the amount of phenols, especially o-diphenols, decreased after infection and stayed more or less on the same level in Latham cultivar. The activity of polyphenoloxidase increased in both of the investigated cultivars, however in the tissue of Latham cultivar this rise was more pronounced than in the M. Promise one. The changes in peroxidase activity after infection were inconsiderable. The relation between the changes observed after infection in the phenolic metabolism and the differentiated susceptibility of callus tissue to D. applanata of the investigated cultivars was discussed. The obtained results explain partially the browning of infected callus tissue of one of the investigated cultivar i.e. M. Promise which had already been observed in the earlier investigations.  相似文献

Investigations of the effects of elevated ozone (O(3)) on the virus-plant system were conducted to inform virus pathogen management strategies better. One susceptible cultivar of tobacco (Nicotiana tabacum L. cv. Yongding) and a resistant cultivar (Nicotiana tabacum L. cv. Vam) to Potato virus Y petiole necrosis strain (PVY(N)) infection were grown in open-top chambers under ambient and elevated O(3) concentrations. Above-ground biomass, foliage chlorophyll, nitrogen and total non-structural carbohydrate (TNCs), soluble protein, total amino acid (TAA) and nicotine content, and peroxidase (POD) activity were measured to estimate the effects of elevated O(3) on the impact of PVY(N) in the two cultivars. Results showed that under ambient O(3), the resistant cultivar possessed greater biomass and a lower C/N ratio after infection than the susceptible cultivar; however, under elevated O(3), the resistant cultivar lost its biomass advantage but maintained a lower C/N ratio. Variation of foliar POD activity could be explained as a resistance cost which was significantly correlated with biomass and C/N ratio of the tobacco cultivar. Chlorophyll content remained steady in the resistant cultivar but decreased significantly in the susceptible cultivar when stressors were applied. Foliar soluble protein and free amino acid content, which were related to resistance cost changes, are also discussed. This study indicated that a virus-resistant tobacco cultivar showed increased sensitivity to elevated O(3) compared to a virus-sensitive cultivar.  相似文献

The aim of the present studies was to compare H2O2 and ascorbate contents as well as peroxidase (PO) and catalase (CAT) activities in leaves of less susceptible cultivar Perkoz and more susceptible Corindo after B. cinerea infection. Increase in H2O2 contents in both Perkoz and Corindo cytosol was observed, however, it appeared earlier in the less susceptible cultivar. The increase in PO activity in the cytosol fraction was observed 48 hours after infection in both cultivars but it was greater in the less susceptible Perkoz. No significant differences between the tested cultivars were observed in ascorbate peroxidase (APX) activity and in reduced and oxidated ascorbate contents. PO activity was thoroughly analyzed in the apoplast fraction. It was measured with syringaldazine (S), tetramethylbenzidine (TMB) and ferulic acid (FA)—substrates characteristic of isoenzymes involved in lignification and stiffening of a cell wall. Increase in PO activity with these substrates was observed earlier in cultivar Perkoz than in cultivar Corindo. Similarly, increase in PO activity with NADH appeared significantly earlier in cultivar Perkoz. Apoplastic PO was separated with DEAE Sepharose and two fractions binding and non-binding were obtained. Binding PO fraction was significantly more active especially with S, TMB and NADH after B. cinerea infection. The increase in the enzyme activity was mostly observed in cultivar Perkoz. Binding PO was separated by electrophoresis on acrylamide gel and revealed six enzymatic forms from which three were much more active after infection in cultivar Perkoz. The obtained results suggest that cell wall strengthening mediated by apoplast PO is a key factor responsible for different resistance of tomato cultivars Perkoz and Corindo to B. cinerea infection.  相似文献

Quantities of free amino acids in segments of cotton roots resistant and susceptible to Meloidogyne incognita were compared. Following infection, the root-knot susceptible cultivar, M8, had greater percentage increases of certain individual free amino acids than the resistant cultivar, Clevewilt, but the sum total of free amino acids was greatest in the resistant cultivar. More free amino acids were present in infected than in noninfected plants of both cultivars. The overall concn of glycine declined over the I 0-day period following inoculation. The concns of the aromatic amino acids, tyrosine and phenylalanine, varied as functions of infection, cultivar, and time of harvest. Proline in susceptible M8 increased nearly 2000-fold 10 days after infection, when considerable thickening of syncytial walls is occurring.  相似文献

The phospholipid content and phospholipase D activity in the leaves of two tobacco (Nicotiana tabacum L.) cultivars were investigated. These cultivars are characterized by different response to the infection with tobacco mosaic virus (TMV). In the infected leaves of a susceptible cv. Samsun, phospholipid content and phospholipase D activity did not change within seven days after TMV infection. The development of a hypersensitive response in the leaves of a resistant cv. Xanthy necrotic was not accompanied by a change in the total phospholipid content as compared to the noninfected leaves. However, the appearance of necrotic lesions and their subsequent expansion resulted in a steady decrease in the level of phosphatidylglycerol in infected leaves. At the same time, phosphatidic acid and diphosphatidylglycerol contents increased. Leaf zones remote from the regions of necrosis development were also characterized by an increased level of phosphatidic acid. There was a tendency for an increase in phospholipase D activity in both the sites of necrosis development and in the leaf regions remote from these sites. The changes in phosphatidic acid content were of similar nature, and therefore a relative increase in phosphatidic acid could result from the phospholipase D activity. This fact suggests a possible involvement of phospholipase D in the development of the hypersensitive response, and this suggestion is supported by a higher enzyme activity in the leaves of healthy plants of the resistant cultivar as compared to the susceptible one. Causes for the changes in the content of some phospholipids, as well as the physiological role of phospholipase D in the hypersensitive response are discussed.  相似文献

Isoperoxidases were detected in resistant Rossol and susceptible Roma VF tomato roots uninfected and infected by Meloidogyne incognita. Syringaldazine, guaiacol, p-phenylenediamine-pyrocatechol (PPD-PC), and indoleacetic acid (IAA) were used as substrates, and the corresponding peroxidative activities were detected either in cytoplasmic or in cell wall fractions, except for IAA oxidase, which was measured in soluble and microsomal fractions. Isoperoxidase activities and cellular locations were induced differently in resistant and susceptible cultivars by nematodes. Nematode infestation markedly enhanced syringaldazine oxidase activity in cell walls of the resistant cultivar. This isoperoxidase is involved in the last step of lignin deposition in plants. Conversely, the susceptible cultivar reacted to M. incognita infection with an increase in cytoplasmic PPD-PC oxidase activity, which presumedly is involved in ethylene production; no changes in cell wall isoperoxidases were observed. IAA oxidase was inhibited in susceptible plants after nematode inoculation, whereas in resistant plants this activity increased in the soluble fraction and decreased in the microsomal fraction.  相似文献

Hydroxyproline-rich glycoproteins (HRGPs) play a defensive role in host–pathogen interactions. However, specific roles of individual HRGPs in plant defense against pathogen are poorly understood. Changes in extracellular distribution and abundance of individual cell wall HRGPs were investigated on root sections of two wax gourd (Benincasa hispida Cogn.) cultivars (Fusarium wilt resistant and susceptible, respectively), which were analyzed by immunolabelling with 20 monoclonal antibodies recognizing different epitopes of extensins and arabinogalactan proteins (AGPs) after being inoculated with Fusarium oxysporum f. sp. Benincasae or treated with fusaric acid (FA). These analyses revealed the following: (1) The levels of JIM11 and JIM20 interacting extensins were higher in the resistant cultivar. Either treatment caused a dramatic decrease in signal in both cultivars, but some new signal appeared in the rhizodermis. (2) The AGPs or rhamnogalacturonan containing CCRCM7-epitope were enhanced in the resistant cultivar, but not in the susceptible one by either treatment. (3) Either treatment caused a slight increase in the levels of the AGPs recognized by LM2 and JIM16, but there were no differences between two cultivars. (4) The MAC204 signal nearly disappeared after FA treatment, but this was not the case with pathogen attack. (5) The LM14 signal slightly decreased after both treatments in both cultivars, but a less decrease was observed with the resistant cultivar. These results indicate that the CCRCM7 epitope likely contributed to the resistance of wax gourd to this pathogen, and JIM11 and JIM20 interacting extensins as well as LM2, LM14, MAC204 and JIM16 interacting AGPs were involved in the host–pathogen interaction.  相似文献

以不同抗病性梨品种为材料,研究了梨黑星菌粗毒素对梨离体叶片的过氧化物酶(POD)活性、苯丙氨酸解氨酶(PAL)活性、丙二醛(MDA)含量、细胞膜相对透性以及叶绿素含量变化的影响.研究表明,毒素接种后抗病和感病品种叶片的POD和PAL活性、MDA含量和相对电导率均呈上升趋势,其间会有1个或2个峰值出现,且抗病品种叶片的POD和PAL活性高于感病品种,而细胞内MDA含量和相对电导率增加比率低于感病品种;同时,毒素使梨离体叶片的叶绿素、类胡萝卜素含量下降,且感病品种的下降幅度大于抗病品种.总之,梨离体叶片POD和PAL的活性变化与梨品种抗病性呈正相关,叶片MDA含量和相对电导率变化与梨品种抗病性呈负相关,抗病品种离体叶片对毒素的毒害有更强的抵抗力.  相似文献

The relationship between age-related resistance of peper plants to Phytophthora capsici and contents of carbohydrates, amino acids, phenolics and mineral nutrients in pepper stems was studied using two pepper cultivars, Hanbyul (susceptible) and Kingkun (resistant). With increasing age of pepper plants, the two cultivars, which differ in their susceptibility to Phytophthora blight, became gradually resistant to the disease. The cultivar Kingkun distinctly showed the age-related resistance to Phytophthora blight at the second branch stage. The weight of dry matter in healthy stems of pepper plants at the second branch stage was twice that at the six leaf stage. The resistant cultivar Kingkun contained lower levels of fructose, glucose and sucrose in stems than the susceptible cultivar Hanbyul at the different developmental stages. No consistent differences between the developmental stages of the plants were recognized with regard to their glucose content. However, the contents of fructose and sucrose in the cultivar Hanbyul greatly increased at the second branch stage. The levels of inositol reduced in both pepper cultivars during plant development. In view of the fact that there were only slight changes in the amount of total amino acids, it seems unlikely that there is a relationship between the amino acid metabolism and the retardation of Phytophthora infection during plant development. The amounts of total phenolic compounds in pepper stems were relatively low at the later growth stages of the plants and also in the resistant cultivar Kingkun. The contents of macroelemental nutrients such as nitrogen, phosphorus, potassium, calcium and magnesium were drastically reduced in pepper stems at the later plant growth stage. No significant differences between the cultivars or the plant growth stages were found in the silicon and microelemental nutrients such as sodium, iron, zinc and manganese. These results suggest that the expression of age-related resistance of pepper plants may be due to the morphological and nutritional changes in tissues of pepper stems during ageing, i.e. the pronounced increase in weight of dry matter, the significant decrease in amounts of mineral nutrients such as nitrogen, phosphorus, potassium, calcium and magnesium, and the tow contents of fructose, glucose and sucrose in the stem tissues.  相似文献

The roots of date palm contain four cell wall‐bound phenolic acids identified as p‐hydroxybenzoic, p‐coumaric, ferulic and sinapic acids. The ferulic acid represents the major phenolic compound since it constitutes 48.2–55.8% of cell wall‐bound phenolic acids. All these phenolic acids were present in the resistant cultivar (BSTN) and the susceptible cultivar (JHL). However, the pre‐infection contents of p‐coumaric, ferulic and sinapic acids were greater in the resistant cultivar than in the susceptible one. For the contents of p‐hydroxybenzoic acid, there was no significant difference between the resistant cultivar and the susceptible cultivar. Similarly, the pre‐infection contents of lignin were approximately equal for both cultivars. Inoculation of the date palm roots by Fusarium oxysporum f. sp. albedinis induced important modifications to the contents of the cell wall‐bound phenolic compounds and lignin, which made it possible to distinguish between resistant and susceptible cultivars. The post‐infection contents of cell wall‐bound phenolic compounds underwent a rapid and intense increase with a maximum accumulation on the tenth day for p‐hydroxybenzoic acid (1.54 μmol/g), p‐coumaric acid (2.77 μmol/g) and ferulic acid (2.64 μmol/g) and on the fifteenth day for sinapic acid (1.85 μmol/g). The maximum contents accumulated in the resistant cultivar were greater than those in the susceptible cultivar, namely, 11 times for p‐hydroxybenzoic acid, 2.6 times for p‐coumaric acid, 1.8 times for ferulic acid and 12.3 times for sinapic acid. In the susceptible cultivar, p‐coumaric acid and ferulic acid contents also increased after inoculation although they did not reach the pre‐infection contents of the resistant cultivar. The contents of p‐hydroxybenzoic acid in the susceptible cultivar roots did not present post‐infection modification and those of sinapic acid decreased instead. The lignin contents increased in both cultivars with a maximum accumulation on the fifteenth day. However, the maximum contents accumulated in the resistant cultivar roots were 1.5 times greater than those of the susceptible cultivar. These results showed clear differences between the resistant BSTN and the susceptible JHL cultivars. The implication of cell wall‐bound phenolic compounds and lignin in the resistance of date palm to F. oxysporum f. sp. albedinis appears to be dependent on the speed and intensity of their accumulation with greater contents in the first stage of infection.  相似文献

We conducted laboratory experiments on two field-collected populations of the squash bug, [Anasa tristis (De Geer) (Hemiptera: Coreidae)], to compare their responses to five generations of selection on a resistant squash cultivar Waltham Butternut, a susceptible cultivar Early Prolific Straightneck, or alternating generations on the resistant and susceptible cultivars. Before testing, one population was maintained for 1 generation on susceptible squash plants, while the other was maintained on the same cultivar for 5 generations. In both populations, nymphal survival decreased markedly in the first generation on resistant squash, but then approximately doubled in the second generation reared on the resistant cultivar. By the end of the experiments, survival on the rearing host was the same for A. tristis maintained continuously on a single cultivar, regardless of which cultivar. Immediately following one or more generations on resistant plants, A. tristis nymphs had lower survival on the susceptible cultivar than did nymphs that had been maintained for consecutive generations on the susceptible cultivar. Results of reciprocal crosses between lines of A. tristis reared on either resistant or susceptible squash for five generations indicated a genetic basis for differences in nymphal survival on the two cultivars.In contrast with the changes in nymphal survival, the preoviposition period became progressively longer when A. tristis was reared for several generations on the resistant cultivar compared to the susceptible one. Prior rearing on resistant plants increased the preoviposition period of adults placed on susceptible plants. However, adult feeding on resistant plants following nymphal development on susceptible plants did not result in an increase in the preoviposition period on resistant plants. Our findings indicate that squash bugs can adapt to resistant plants within a relatively short period in the laboratory. However, ecological and agricultural factors should prevent squash bugs from overcoming resistance in the field.  相似文献

The inoculation of the seedling roots of the resistant (Bousthami Noir) and susceptible (Jihel) date palm (Phoenix dactylifera) cultivars by Fusarium oxysporum f. sp. albedinis induced an increase in phenylalanine ammonia-lyase (PAL) activity. The response of the PAL activity in the resistant cultivar was faster and higher than in the susceptible one. However, the elicitation of the seedlings with the hyphal wall elicitor (HWE) of the pathogen induced identical PAL activity in both cultivars. In the resistant cultivar, the the PAL activity elicited with the HWE was not influenced by the addition of the fungal culture filtrate (FCF) whereas it was suppressed in the susceptible cultivar. This FCF suppressor effect was dose-dependent, not influenced by sodium periodate, whereas it was strongly reduced by the heat (121 °C for 45 min) and pronase E. These results show that differential induction of the defence mechanisms in both cultivars was not related to differences in the induction of the PAL activity, but to the suppression of its elicitation in the susceptible cultivar.  相似文献

Abstract

Macroscopic symptoms were observed in two strawberry cultivars, with the degree of symptom intensity varying depending on the susceptibility of the cultivars, i.e. resistant or susceptible. The symptoms presented as red spots and were observed 30 d following leaf tissue inoculation with the Mycosphaerella fragariae pathogen. A comparison of the superoxide dismutase isoform profiles obtained by gel electrophoresis in all samples extracted from both resistant and susceptible cultivars indicated one constant sharp band, identified as Mn[sbnd]SOD with a molecular mass of 19 kDa. The intensity of this band was higher in all samples derived from the resistant cultivar than in those from the susceptible cultivar. Another superoxide dismutase (SOD) isoform, identified as CuZn[sbnd]SOD with a molecular mass of 16 kDa, was detected in all soluble proteins derived from the resistant cultivar. This isoform was not observed in the susceptible cultivar; however, following an incremental increase in the amount of loaded protein, it was illuminated as a faint band in a sample collected 3 d after inoculation, indicating insufficient production of the CuZn[sbnd]SOD isoform in the susceptible cultivar during an oxidative burst induced by the M. fragaria pathogen. Several bands were also characterized in both cultivars containing Fe and Mn as their co-factors (Fe, Mn[sbnd]SOD). Unlike in the resistant cultivar, where the activity of Fe, Mn[sbnd]SOD isoforms gradually and regularly increased and reached its highest level on the third day after inoculation, the activity of the isoforms changed irregularly over 20 days of study in the susceptible cultivar.  相似文献

The phospholipid (PL) and fatty acid (FA) composition of major membrane lipid constituents, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), as well as the cholesterol/phospholipid (CL/PL) ratio were assayed in the muscles, gills and liver of the black plaice Pleuronectes (Liopsetta) obscura at different ambient temperatures (18, 9 and 0°C). PL and CL were shown to be actively involved in adaptation of the fish to changes in the seawater temperature. As temperature declines, the monounsaturated FA (MUFA) level increases while the polyunsaturated FA (PUFA) fraction in gills and liver PC and PE, on the contrary, decreases, resulting in diminished functional activity of the fish. However, in muscles this correlation is lacking. The PC and PE composition was shown to be organ- and ambient temperature-dependent. Major PC forms are saturated FA (SFA)/PUFA and MUFA/PUFA composed of a relatively small number of major molecular species. A temperature drop results in an increased SFA/PUFA level and decreased MUFA/PUFA and PUFA/PUFA levels in muscles and gills, and this may promote a drop in the viscosity of the outer lipid monolayer of membranes and in their functional activity. In contrast to PC, the PE composition in all organs tested is characterized by a decrease in the SFA/ PUFA level and an increase in MUFA/PUFA and PUFA/PUFA levels. Such changes promote the retention of functional activity of the inner lipid monolayer of membranes and are not synchronized with rearrangements in their outer monolayer. Due to intermolecular transfer of acyl radicals at a constancy of their composition, functional rearrangement of the lipid matrix appears to be achieved through changes in the membrane viscosity. Our data support the idea that different adaptation strategies in fish are driven by certain sets of PL molecular species.  相似文献

在家蚕疾病中,病毒性疾病是危害茧产品的主要且最普遍的疾病。病毒性蚕软腐病是由BmIFV, BmDNV1 和 BmDNV2引起的。对印度种质库(Indian Germ plasm stock)中的BmDN V1有抗性和敏感的家蚕品种进行了鉴定。利用标准方法对在BmDNV1感染过程中抗性和敏感品系的主要有机成分的变化(包括总蛋白、碳水化合物和脂类)进行了检测。结果表明: 随着幼虫年龄的增长,对照组和处理组中有机成分(即蛋白质和碳水化合物)的含量也随之增长,但是处理组的增长水平明显地低于各自对照组的增长水平。接种BmDNV1后,与对照组比较,敏感品种体内的血淋巴和中肠组织的总蛋白的量显著地下降。在抗性品种,接种4天后的总蛋白的量有了显著地下降,之后,下降水平小于各自的对照组。在抗性和敏感品种中,血淋巴和中肠组织中的总碳水化合物的量略有下降。在变化不显著的抗性和敏感品种中,血淋巴和中肠组织中的脂质的量有显著地提高。在敏感家蚕品种,生化变化清晰地显示: BmDNV1感染消耗作为主要能量来源的总蛋白和碳水化合物。这些成分的损耗导致了被感染家蚕的生长受阻。  相似文献

Synergistic effects of Fusarium — toxins mixture (moniliformin, fumonisin B1, fusaproliferin, zearalenone, zearalenol and deoxynivalenol, each at concentration 3.5 μg mL-1) on maize plants of resistant and susceptible cultivars were studied. After 72-hour treatment the biomass production with both cultivars was approximately 6% lower than in the respective controls. In the resistant cultivar chlorophyll content was increased comparing to control, with higher increase in chlorophyll b. In susceptible cultivar chlorophyll content was slightly decreased, particularly with chlorophyll b.No significant differences between treated and non-treated plants were found in the cell ultrastructure. In susceptible plants the young root cells were more vacuolated and plasmolysis occurred in the cells of outer cortex. In leaves of this cultivar also disorganization of thylakoids in some chloroplasts was observed.  相似文献

This study investigated the infection process of Mycosphaerella graminicola and enzyme activities related to reactive oxygen species (ROS) or oxylipin biosynthesis in four French wheat cultivars with variable resistance to M.?graminicola infection. At field level, cultivars Caphorn, Maxyl and Gen11 were susceptible, whereas Capnor showed high levels of quantitative resistance. Moreover, Capnor and Gen11 were tolerant, i.e., their yield was less affected by infection compared to non-tolerant Maxyl and Caphorn. These four cultivars were inoculated under laboratory conditions with the M.?graminicola IPO323 reference strain. Cytological and biochemical responses were studied on collected first plantlet leaves and several features discriminated between cultivars. However, resistance and tolerance had no impact on the fungal infection process. Levels of lipoxygenase (LOX), peroxidase (PO) and glutathione-S-transferase (GST) activities were also compared with regard to cultivar resistance or tolerance to M.?graminicola. LOX, PO and GST activities did not discriminate resistance and tolerance profiles, although a low level of PO in inoculated and non-inoculated plants could be associated with tolerance. In addition, cell necrosis correlated positively with LOX in non-tolerant cultivars, while mycelia surrounding stomata were positively correlated with PO in the resistant cultivar. GST activity presented correlations between cytological and biochemical parameters only for susceptible cultivars. Stomatal and direct penetration were positively correlated with GST activity in the susceptible non-tolerant cultivars, while these correlations were negative in the tolerant cultivar. When combining cytological and biochemical observations with resistance and tolerance profiles, for each cultivar and at each time point, cultivars could be classified in tight accordance with their previous field characterisation. Moreover, tolerance allowed us to distinguish susceptible cultivars when both biochemical and cytological parameters were considered together.  相似文献

The effects of the ring rot causal agent Clavibacter michiganensis subsp. sepedonicus (a virulent strain 5369) on the peroxidase activity of various tissues of potato plants grown under axenic conditions were studied. Root infection enhanced peroxidase activity in all plant tissues (roots, leaves, and stems). In the resistant cv. Lugovskoi, peroxidase activity was much higher than in the susceptible cv. Luk'yanovskii. Co-culturing of the suspension cells of these potato cultivars with the bacterial pathogen also activated peroxidase in the cells of the resistant cultivar; in the cells of the susceptible cultivar, peroxidase activation was less pronounced. Treating suspension cell with exopolysaccharides secreted by the pathogen enhanced the activity of extra- and intracellular peroxidases, and the degree of this enhancement differed in the two potato cultivars.  相似文献
