The effects of ABA, isobutyric acid (IBA) and nicotine on K+and Cl+ efflux were studied in Arabidopsis thaliana seedlings,and the role of pHcyt, and Em in the regulation of the effluxof these ions was discussed. The data show that treatments withIBA and nicotine influenced in opposite directions the effluxof either K+ or Cl: K+ efflux was increased by nicotineand reduced in the presence of IBA, whereas Cl effluxwas stimulated by IBA and decreased by nicotine treatment. Underall the conditions tested ABA induced cytoplasmic acidificationand inhibition of K+ and Cl net efflux. Experiments aimedto estimate the individual contribution of pHcyt and Em in modulatingK efflux indicated that, within the range of acidic pHcytvalues, a regulation of K+ efflux was imposed by pHcyt on thecontrol exerted by Em, the efflux being inhibited by lower pHcytvalues. Conversely, in the alkaline side of pHcyt K+ effluxseemed linked only to the Em values. These results are consistentwith the hypothesis that the decrease in K+ efflux observedin non-stomatal tissues in the presence of ABA may be mediatedby the cytoplasmic acidification induced by the hormone. Resting or basal intracellular pH (pHi) measured in cultured human syncytiotrophoblast cells was 7.26 ± 0.04 (without HCO3) or 7.24 ± 0.03 (with HCO3). Ion substitution and inhibitor experiments were performed to determine whether common H+-transporting species were operating to maintain basal pHi. Removal of extracellular Na+ or Cl or addition of amiloride or dihydro-4,4'-diisothiocyanatostilbene-2,2'-disulfonate (H2DIDS) had no effect. Acidification with the K+/H+ exchanger nigericin reduced pHi to 6.25 ± 0.15 (without HCO3) or 6.53 ± 0.10 (with HCO3). In the presence of extracellular Na+, recovery to basal pHi was prompt and occurred at similar rates in the absence and presence of HCO3. Ion substitution and inhibition experiments were also used to identify the species mediating the return to basal pHi after acidification. Recovery was inhibited by removal of Na+ or addition of amiloride, whereas removal of Cl and addition of H2DIDS were ineffective. Addition of the Na+/H+ exchanger monensin to cells that had returned to basal pHi elicited a further increase in pHi to 7.48 ± 0.07. Analysis of recovery data showed that there was a progressive decrease in pH per minute as pHi approached the basal level, despite the continued presence of a driving force for H+ extrusion. These data show that in cultured syncytial cells, in the absence of perturbation, basal pHi is preserved despite the absence of active, mediated pH maintenance. Several studies suggest the involvement of Na+ and HCO3 transport in the formation of cerebrospinal fluid. Two Na+-dependent HCO3 transporters were recently localized to the epithelial cells of the rat choroid plexus (NBCn1 and NCBE), and the mRNA for a third protein was also detected (NBCe2) (Praetorius J, Nejsum LN, and Nielsen S. Am J Physiol Cell Physiol 286: C601–C610, 2004). Our goal was to immunolocalize the NBCe2 to the choroid plexus by immunohistochemistry and immunogold electronmicroscopy and to functionally characterize the bicarbonate transport in the isolated rat choroid plexus by measurements of intracellular pH (pHi) using a dual-excitation wavelength pH-sensitive dye (BCECF). Both antisera derived from COOH-terminal and NH2-terminal NBCe2 peptides localized NBCe2 to the brush-border membrane domain of choroid plexus epithelial cells. Steady-state pHi in choroidal cells increased from 7.03 ± 0.02 to 7.38 ± 0.02 (n = 41) after addition of CO2/HCO3 into the bath solution. This increase was Na+ dependent and inhibited by the Cl and HCO3 transport inhibitor DIDS (200 µM). This suggests the presence of Na+-dependent, partially DIDS-sensitive HCO3 uptake. The pHi recovery after acid loading revealed an initial Na+ and HCO3-dependent net base flux of 0.828 ± 0.116 mM/s (n = 8). The initial flux in the presence of CO2/HCO3 was unaffected by DIDS. Our data support the existence of both DIDS-sensitive and -insensitive Na+- and HCO3-dependent base loader uptake into the rat choroid plexus epithelial cells. Malignantgliomas exhibit alkaline intracellular pH (pHi) and acidicextracellular pH (pHe) compared with nontransformedastrocytes, despite increased metabolic H+ production. Theacidic pHe limits the availability ofHCO3, thereby reducing both passiveand dynamic HCO3-dependent buffering.This implies that gliomas are dependent upon dynamic HCO3-independent H+buffering pathways such as the type 1 Na+/H+exchanger (NHE1). In this study, four rapidly proliferating gliomas exhibited significantly more alkaline steady-state pHi(pHi = 7.31-7.48) than normal astrocytes(pHi = 6.98), and increased rates of recovery fromacidification, under nominallyCO2/HCO3-free conditions.Inhibition of NHE1 in the absence ofCO2/HCO3 resulted inpronounced acidification of gliomas, whereas normal astrocytes wereunaffected. When suspended inCO2/HCO3 medium astrocytepHi increased, yet glioma pHi unexpectedlyacidified, suggesting the presence of anHCO3-dependent acid loadingpathway. Nucleotide sequencing of NHE1 cDNA from the gliomasdemonstrated that genetic alterations were not responsible for thisaltered NHE1 function. The data suggest that NHE1 activity issignificantly elevated in gliomas and may provide a useful target forthe development of tumor-selective therapies.

The vacuolar H+-ATPase (V-ATPase) acidifies compartments of the vacuolar system of eukaryotic cells. In renal epithelial cells, it resides on the plasma membrane and is essential for bicarbonate transport and acid-base homeostasis. The factors that regulate the H+-ATPase remain largely unknown. The present study examines the effect of glucose on H+-ATPase activity in the pig kidney epithelial cell line LLC-PK1. Cellular pH was measured by performing ratiometric fluorescence microscopy using the pH-sensitive indicator BCECF-AM. Intracellular acidification was induced with NH3/NH4+ prepulse, and rates of intracellular pH (pHi) recovery (after in situ calibration) were determined by the slopes of linear regression lines during the first 3 min of recovery. The solutions contained 1 µM ethylisopropylamiloride and were K+ free to eliminate Na+/H+ exchange and H+-K+-ATPase activity. After NH3/NH4+-induced acidification, LLC-PK1 cells had a significant pHi recovery rate that was inhibited entirely by 100 nM of the V-ATPase inhibitor concanamycin A. Acute removal of glucose from medium markedly reduced V-ATPase-dependent pHi recovery activity. Readdition of glucose induced concentration-dependent reactivation of V-ATPase pHi recovery activity within 2 min. Glucose replacement produced no significant change in cell ATP or ADP content. H+-ATPase activity was completely inhibited by the glycolytic inhibitor 2-deoxy-D-glucose (20 mM) but only partially inhibited by the mitochondrial electron transport inhibitor antimycin A (20 µM). The phosphatidylinositol 3-kinase (PI3K) inhibitor wortmannin (500 nM) abolished glucose activation of V-ATPase, and activity was restored after wortmannin removal. Glucose activates V-ATPase activity in kidney epithelial cells through the glycolytic pathway by a signaling pathway that requires PI3K activity. Continuous measurements of cytoplasmic pH (pHc) in Sinapis roothairs have been carried out with double-barrelled pH-micro-electrodesin order to gain information on translocation of protons acrossthe plasmalemma and cytoplasmic pH control. (i) The cytoplasmicpH of Sinapis (7–33 ? 0–12, standard conditions)changes no more than 0.1 pHc, per pHo-unit, regardless of whethercyanide is present or not. (ii) Weak acids rapidly acidify pHcand hyperpolarize, while weak bases alkalize pHc and depolarizethe cells, (iii) 1.0 mol M,3 NaCN acidifies the cytoplasm by0.4 to 0.7 pH-units, but alkalizes the vacuole. (iv) 20 mmolm–3 CCCP has no significant effect on pHc, if added atpH 9.6 or 7.2, but acidifies pHc by 1.3 units at pH 4.3. Inthe presence of CCCP, cyanide acidifies the cytoplasm, (v) Chloridetransiently acidifies pHc, while K+, Na+, and have no significant effects, (vi) Cytoplasmic buffer capacityforms a bell-shaped curve versus pHc with an optimum of about50 mol m–3 H+pHc-unit. The modes of proton re-entry and the effects of active and passiveproton transport on cellular pH control are critically discussed.It is suggested that the proton leak, consisting of H+-cotransport(e.g. H+/Cl) rather than H+-uniport, is no threat topHc. The proton export pump, although itself reacting to changesin pHc, influences pHc only to a minor extent. It is concludedthat buffer capacity and membrane transport play moderate rolesin pHc control in Sinapis, while the interlocked H+-producingand -consuming reactions of cellular metabolism are the mainregulating factors. Using thepH-sensitive dye2',7'-bis(2-carboxyethyl)-5(6)-carboxyfluorescein (BCECF),we examined the effect of hyperosmolar solutions, which presumablycaused cell shrinkage, on intracellular pH(pHi) regulation in mesangialcells (single cells or populations) cultured from the rat kidney. Thecalibration of BCECF is identical in shrunken and unshrunken mesangialcells if the extracellular K+concentration ([K+])is adjusted to match the predicted intracellular[K+]. ForpHi values between ~6.7 and~7.4, the intrinsic buffering power in shrunken cells (600 mosmol/kgH2O) is threefold larger than in unshrunken cells (~300mosmol/kgH2O). In the nominalabsence ofCO2/HCO3,exposing cell populations to a HEPES-buffered solution supplementedwith ~300 mM mannitol (600 mosmol/kgH2O) causes steady-statepHi to increase by ~0.4. The pHi increase is due to activationofNa+/H+exchange because, in single cells, it is blocked in the absence ofexternal Na+ or in the presence of50 µM ethylisopropylamiloride (EIPA). Preincubating cells in aCl-free solution for atleast 14 min inhibits the shrinkage-induced pHi increase by 80%. Wecalculated the pHi dependence oftheNa+/H+exchange rate in cell populations under normosmolar and hyperosmolar conditions by summing 1) thepHi dependence of the totalacid-extrusion rate and 2) thepHi dependence of theEIPA-insensitive acid-loading rate. To determine the role ofmembrane transporters in intracellular pH (pHi) regulationunder conditions of low microenvironmental O2, we monitoredpHi in isolated single CA1 neurons using the fluorescentindicator carboxyseminaphthorhodafluor-1 and confocal microscopy. Aftertotal O2 deprivation or anoxia (PO2 0 Torr), a large increase in pHi was seen in CA1neurons in HEPES buffer, but a drop in pHi, albeit small,was observed in the presence of HCO. Ionicsubstitution and pharmacological experiments showed that the largeanoxia-induced pHi increase in HEPES buffer was totallyNa+ dependent and was blocked by HOE-694, stronglysuggesting the activation of the Na+/H+exchanger (NHE). Also, this pHi increase in HEPES bufferwas significantly smaller in Na+/H+ exchangerisoform 1 (NHE1) null mutant CA1 neurons than in wild-type neurons,demonstrating that NHE1 is responsible for part of the pHiincrease following anoxia. Both chelerythrine and H-89 partly blocked,and H-7 totally eliminated, this anoxia-induced pHiincrease in the absence of HCO. We conclude that1) O2 deprivation activatesNa+/H+ exchange by enhancing protein kinaseactivity and 2) membrane proteins, such as NHE, activelyparticipate in regulating pHi during low-O2states in neurons.

The stimulatory pathways controlling HCO3- secretion by the pancreatic ductal epithelium are well described. However, only a few data are available concerning inhibitory mechanisms, which may play an important role in the physiological control of the pancreas. The aim of this study was to investigate the cellular mechanism by which substance P (SP) inhibits pancreatic ductal HCO3- secretion. Small intra/interlobular ducts were isolated from the pancreas of guinea pigs. During overnight culture the ducts seal to form a closed sac. Transmembrane HCO3- fluxes were calculated from changes in intracellular pH (measured using the pH-sensitive dye BCECF) and the buffering capacity of the cells. We found that secretin can stimulate HCO3- secretion in guinea pig pancreatic ducts about fivefold and that this effect could be totally blocked by SP. The inhibitory effect of SP was relieved by spantide, an SP receptor antagonist. SP had no effect on the activity of basolateral Na+-HCO3- cotransporters and Na+/H+ exchangers. Corneal transparency and hydration control are dependent on HCO3 transport properties of the corneal endothelium. Recent work (13) suggested the presence of an apical 1Na+-3HCO3 cotransporter (NBC1) in addition to a basolateral 1Na+-2HCO3 cotransporter. We examined whether the NBC1 cotransporter contributes significantly to basolateral or apical HCO3 permeability and whether the cotransporter participates in transendothelial net HCO3 flux in cultured bovine corneal endothelium. NBC1 protein expression was reduced using small interfering RNA (siRNA). Immunoblot analysis showed that 5–15 nM siRNA decreased NBC1 expression by 80–95%, 4 days posttransfection. Apical and basolateral HCO3 permeabilities were determined by measuring the rate of pHi change when HCO3 was removed from the bath under constant pH or constant CO2 conditions. Using either protocol, we found that cultures treated with NBC1 siRNA had sixfold lower basolateral HCO3 permeability than untreated or siCONTROL siRNA-treated cells. Apical HCO3 permeability was unaffected by NBC1 siRNA treatment. Net non-steady-state HCO3 flux was 0.707 ± 0.009 mM·min–1·cm2 in the basolateral-to-apical direction and increased to 1.74 ± 0.15 when cells were stimulated with 2 µM forskolin. Treatment with 5 nM siRNA decreased basolateral-to-apical flux by 67%, whereas apical-to-basolateral flux was unaffected, significantly decreasing net HCO3 flux to 0.236 ± 0.002. NBC1 siRNA treatment or 100 µM ouabain also eliminated steady-state HCO3 flux, as measured by apical compartment alkalinization. Effects of removal of external Ca2+ on the cytoplasmic pH (pHc)of Chara corallina have been measured with the weak acid 5,5-dimethyl-oxazolidine-2,4-dione(DMO) as a function of external pH (pH0) and of the externalconcentration of K+. Removal of Ca2+ always decreased pHc whenpH0 was below about 6.0; the decrease was about 0.2–0.4units at pH0 5.0, increasing to about 0.5 units at pH0 4.3.When pH0 was 6.0 or higher the removal of Ca2+ had little orno effect on pHc. This situation was not altered by changingthe concentration of K+, though in some experiments at pH0 5.0–5.2there was a slight decrease in pH0 (about 0.2 units) when K+was increased from 0.2 to 2.0 mol m–3, an effect apparentlyreversed when K+ was higher (5.0 or 10.0 mol m–3). Theresults suggest that H+ transport continues in the absence ofexternal Ca2+, despite previous suggestions to the contrary,and that the H+ pump does not necessarily run near thermodynamicequilibrium with its chemical driving reaction. They indicate,rather, that the H+ pump is under kinetic control and providefurther evidence for the inadequacy of present models for theoperation of the H+ pump in charophyte cells, especially inrelation to its proposed role in regulating pHc. A rise in cytosolic Ca2+ concentration ([Ca2+]cyt) in pulmonary artery smooth muscle cells (PASMC) is a trigger for pulmonary vasoconstriction and a stimulus for PASMC proliferation and migration. Multiple mechanisms are involved in regulating [Ca2+]cyt in human PASMC. The resting [Ca2+]cyt and Ca2+ entry are both increased in PASMC from patients with idiopathic pulmonary arterial hypertension (IPAH), which is believed to be a critical mechanism for sustained pulmonary vasoconstriction and excessive pulmonary vascular remodeling in these patients. Here we report that protein expression of NCX1, an NCX family member of Na+/Ca2+ exchanger proteins is upregulated in PASMC from IPAH patients compared with PASMC from normal subjects and patients with other cardiopulmonary diseases. The Na+/Ca2+ exchanger operates in a forward (Ca2+ exit) and reverse (Ca2+ entry) mode. By activating the reverse mode of Na+/Ca2+ exchange, removal of extracellular Na+ caused a rapid increase in [Ca2+]cyt, which was significantly enhanced in IPAH PASMC compared with normal PASMC. Furthermore, passive depletion of intracellular Ca2+ stores using cyclopiazonic acid (10 µM) not only caused a rise in [Ca2+]cyt due to Ca2+ influx through store-operated Ca2+ channels but also mediated a rise in [Ca2+]cyt via the reverse mode of Na+/Ca2+ exchange. The upregulated NCX1 in IPAH PASMC led to an enhanced Ca2+ entry via the reverse mode of Na+/Ca2+ exchange, but did not accelerate Ca2+ extrusion via the forward mode of Na+/Ca2+ exchange. A rise in cytosolic Ca2+ concentration ([Ca2+]cyt) in pulmonary artery smooth muscle cells (PASMC) is an important stimulus for cell contraction, migration, and proliferation. Depletion of intracellular Ca2+ stores opens store-operated Ca2+ channels (SOC) and causes Ca2+ entry. Transient receptor potential (TRP) cation channels that are permeable to Na+ and Ca2+ are believed to form functional SOC. Because sarcolemmal Na+/Ca2+ exchanger has also been implicated in regulating [Ca2+]cyt, this study was designed to test the hypothesis that the Na+/Ca2+ exchanger (NCX) in cultured human PASMC is functionally involved in regulating [Ca2+]cyt by contributing to store depletion-mediated Ca2+ entry. RT-PCR and Western blot analyses revealed mRNA and protein expression for NCX1 and NCKX3 in cultured human PASMC. Removal of extracellular Na+, which switches the Na+/Ca2+ exchanger from the forward (Ca2+ exit) to reverse (Ca2+ entry) mode, significantly increased [Ca2+]cyt, whereas inhibition of the Na+/Ca2+ exchanger with KB-R7943 (10 µM) markedly attenuated the increase in [Ca2+]cyt via the reverse mode of Na+/Ca2+ exchange. Store depletion also induced a rise in [Ca2+]cyt via the reverse mode of Na+/Ca2+ exchange. Removal of extracellular Na+ or inhibition of the Na+/Ca2+ exchanger with KB-R7943 attenuated the store depletion-mediated Ca2+ entry. Furthermore, treatment of human PASMC with KB-R7943 also inhibited cell proliferation in the presence of serum and growth factors. These results suggest that NCX is functionally expressed in cultured human PASMC, that Ca2+ entry via the reverse mode of Na+/Ca2+ exchange contributes to store depletion-mediated increase in [Ca2+]cyt, and that blockade of the Na+/Ca2+ exchanger in its reverse mode may serve as a potential therapeutic approach for treatment of pulmonary hypertension. sodium-calcium exchange; calcium homeostasis; vascular smooth muscle  相似文献

Satellite cellsare glial cells wrapped around somata of sensory and autonomic ganglionneurons. Neither their functional roles nor electrical properties havebeen fully clarified so far. Using immunohistochemistry, we found thatinwardly rectifying K+ channelsubunit Kir4.1 (also called Kir1.2 orKAB-2) was expressed prominently in the satellite cells of cochlear ganglia. The Kir4.1 immunoreactivity was localized specifically at the myelin sheaths ofsatellite cells wrapping the somata of the ganglion neurons. Developmental expression of Kir4.1 in satellite cells paralleled development of the action potential in the auditory nerve. These results suggest that this channel in satellite cells may be responsible for the regulation of K+ extrudedfrom the ganglion neurons during excitation.

Earlymucosal restitution occurs by epithelial cell migration to resealsuperficial wounds after injury. Differentiated intestinal epithelialcells induced by forced expression of the Cdx2 gene migrateover the wounded edge much faster than undifferentiated parental cellsin an in vitro model. This study determined whether thesedifferentiated intestinal epithelial cells exhibit increased migrationby altering voltage-gated K+ (Kv) channel expression andcytosolic free Ca2+ concentration([Ca2+]cyt). StableCdx2-transfected IEC-6 cells (IEC-Cdx2L1) with highly differentiated phenotype expressed higher basal levels of Kv1.1 andKv1.5 mRNAs and proteins than parental IEC-6 cells. Neither IEC-Cdx2L1cells nor parental IEC-6 cells expressed voltage-dependent Ca2+ channels. The increased expression of Kv channels indifferentiated IEC-Cdx2L1 cells was associated with an increase inwhole cell K+ currents, membrane hyperpolarization, and arise in [Ca2+]cyt. The migration rates indifferentiated IEC-Cdx2L1 cells were about four times those of parentalIEC-6 cells. Inhibition of Kv channel expression by polyamine depletiondecreased [Ca2+]cyt, reduced myosin stressfibers, and inhibited cell migration. Elevation of[Ca2+]cyt by ionomycin promoted myosin IIstress fiber formation and increased cell migration. These resultssuggest that increased migration of differentiated intestinalepithelial cells is mediated, at least partially, by increasing Kvchannel activity and Ca2+ influx during restitution.

Sodium-independent Cl movement (i.e., Cl-anion exchange) has not previously been identified in the basolateral membranes of rat colonic epithelial cells. The present study demonstrates Cl-HCO3 exchange as the mechanism for 36Cl uptake in basolateral membrane vesicles (BLMV) prepared in the presence of a protease inhibitor cocktail from rat distal colon. Studies of 36Cl uptake performed with BLMV prepared with different types of protease inhibitors indicate that preventing the cleavage of the COOH-terminal end of AE2 protein by serine-type proteases was responsible for the demonstration of Cl-HCO3 exchange. In the absence of voltage clamping, both outward OH gradient (pHout/pHin: 7.5/5.5) and outward HCO3 gradient stimulated transient 36Cl uptake accumulation. However, voltage clamping with K-ionophore, valinomycin, almost completely (87%) inhibited the OH gradient-driven 36Cl uptake, whereas HCO3 gradient-driven 36Cl uptake was only partially inhibited (38%). Both electroneutral HCO3 and OH gradient-driven 36Cl uptake were 1) completely inhibited by DIDS, an anion exchange inhibitor, with a half-maximal inhibitory constant (Ki) of 26.9 and 30.6 µM, respectively, 2) not inhibited by 5-nitro-2-(3-phenylpropylamino)benzoic acid(NPPB), a Cl channel blocker, 3) saturated by increasing extravesicular Cl concentration with a Km for Cl of 12.6 and 14.2 mM, respectively, and 4) present in both surface and crypt cells. Intracellular pH (pHi) was also determined with 2',7'-bis(2-carboxyethyl)-5(6)-carboxyfluorescein-acetomethylester (BCECF-AM) in an isolated superfused crypt preparation. Removal of Cl resulted in a DIDS-inhibitable increase in pHi both in HCO3-buffered and in the nominally HCO3-free buffered solutions (0.28 ± 0.02 and 0.11 ± 0.02 pH units, respectively). Human NBC3 is an electroneutral Na+/HCO3 cotransporter expressed in heart, skeletal muscle, and kidney in which it plays an important role in HCO3 metabolism. Cytosolic enzyme carbonic anhydrase II (CAII) catalyzes the reaction CO2 + H2O HCO3 + H+ in many tissues. We investigated whether NBC3, like some Cl/HCO3 exchange proteins, could bind CAII and whether PKA could regulate NBC3 activity through modulation of CAII binding. CAII bound the COOH-terminal domain of NBC3 (NBC3Ct) with Kd = 101 nM; the interaction was stronger at acid pH. Cotransfection of HEK-293 cells with NBC3 and CAII recruited CAII to the plasma membrane. Mutagenesis of consensus CAII binding sites revealed that the D1135-D1136 region of NBC3 is essential for CAII/NBC3 interaction and for optimal function, because the NBC3 D1135N/D1136N retained only 29 ± 22% of wild-type activity. Coexpression of the functionally dominant-negative CAII mutant V143Y with NBC3 or addition of 100 µM 8-bromoadenosine to NBC3 transfected cells reduced intracellular pH (pHi) recovery rate by 31 ± 3, or 38 ± 7%, respectively, relative to untreated NBC3 transfected cells. The effects were additive, together decreasing the pHi recovery rate by 69 ± 12%, suggesting that PKA reduces transport activity by a mechanism independently of CAII. Measurements of PKA-dependent phosphorylation by mass spectroscopy and labeling with [-32P]ATP showed that NBC3Ct was not a PKA substrate. These results demonstrate that NBC3 and CAII interact to maximize the HCO3 transport rate. Pulmonary vasoconstriction and vascularmedial hypertrophy greatly contribute to the elevated pulmonaryvascular resistance in patients with pulmonary hypertension. A rise incytosolic free Ca2+ ([Ca2+]cyt)in pulmonary artery smooth muscle cells (PASMC) triggers vasoconstriction and stimulates cell growth. Membrane potential (Em) regulates[Ca2+]cyt by governing Ca2+influx through voltage-dependent Ca2+ channels. Thusintracellular Ca2+ may serve as a shared signaltransduction element that leads to pulmonary vasoconstriction andvascular remodeling. In PASMC, activity of voltage-gated K+(Kv) channels regulates resting Em. In thisstudy, we investigated whether changes of Kv currents[IK(V)], Em, and[Ca2+]cyt affect cell growth by comparingthese parameters in proliferating and growth-arrested PASMC. Serumdeprivation induced growth arrest of PASMC, whereas chelation ofextracellular Ca2+ abolished PASMC growth. Resting[Ca2+]cyt was significantly higher, andresting Em was more depolarized, inproliferating PASMC than in growth-arrested cells. Consistently, wholecell IK(V) was significantly attenuated in PASMCduring proliferation. Furthermore, Emdepolarization significantly increased resting[Ca2+]cyt and augmented agonist-mediatedrises in [Ca2+]cyt in the absence ofextracellular Ca2+. These results demonstrate that reducedIK(V), depolarized Em, and elevated [Ca2+]cyt may play a criticalrole in stimulating PASMC proliferation. Pulmonary vascular medialhypertrophy in patients with pulmonary hypertension may be partlycaused by a membrane depolarization-mediated increase in[Ca2+]cyt in PASMC.

Cytoplasmic pH (pHc) in Chara corallina was measured (from [14C]stribution)as a function of external pH (pH0)and temperature. With pH0near 7, pHc at 25?C is 7.80; pHcincreases by 0.005 pH units?C–1 temperature decrease, i.e. pHc at 5 ?C is 7.90. WithpH? near 5.5, the increase in pHc with decreasing temperatureis 0.015 units ?C–1 between 25 and 15?C, but 0.005 units?C–1 between 15 and 5?C. This implies a more precise regulationof pHc with variations in pHo at 5 or 15 ?C compared with 25?C. The observed dp Hc/dT is generally smaller than the –0.017units ?C–1 needed to maintain a constant H+/OH–1,or a constant fractional ionization of histidine in protein,with variation in temperature. It is closer to that needed tomaintain the fractional ionization of phosphorylated compoundsor of CO2–HCO3 The value of dpHc/dT has importantimplications for several regulatory aspects of cell metabolism.These include (all as a function of temperature) the rates ofenzyme reactions, the H+ at the plasmalemma(and hence the energy available for cotransport processes),and the mechanism for pHc regulation by the control of bidirectionalH+ fluxes at the plasmalemma.  相似文献

Tg737orpk mice have defects in cilia assembly and develop hydrocephalus in the perinatal period of life. Hydrocephalus is progressive and is thought to be initiated by abnormal ion and water transport across the choroid plexus epithelium. The pathology is further aggravated by the slow and disorganized beating of motile cilia on ependymal cells that contribute to decreased cerebrospinal fluid movement through the ventricles. Previously, we demonstrated that the hydrocephalus phenotype is associated with a marked increase in intracellular cAMP levels in choroid plexus epithelium, which is known to have regulatory effects on ion and fluid movement in many secretory epithelia. To evaluate whether the hydrocephalus in Tg737orpk mutants is associated with defects in ion transport, we compared the steady-state pHi and Na+-dependent transport activities of isolated choroid plexus epithelium tissue from Tg737orpk mutant and wild-type mice. The data indicate that Tg737orpk mutant choroid plexus epithelium have lower pHi and higher Na+-dependent HCO3 transport activity compared with wild-type choroid plexus epithelium. In addition, wild-type choroid plexus epithelium could be converted to a mutant phenotype with regard to the activity of Na+-dependent HCO3 transport by addition of dibutyryl-cAMP and mutant choroid plexus epithelium toward the wild-type phenotype by inhibiting PKA activity with H-89. Together, these data suggest that cilia have an important role in regulating normal physiology of choroid plexus epithelium and that ciliary dysfunction in Tg737orpk mutants disrupts a signaling pathway leading to elevated intracellular cAMP levels and aberrant regulation of pHi and ion transport activity. cAMP; ion transport  相似文献
