Two experiments were conducted to evaluate postpartum resumption of ovulatory cycles in primiparous, suckled beef cows exposed to close physical contact or limited, fenceline contact with mature bulls. The hypotheses tested were that proportions of cows cycling and intervals from the start of exposure to resumption of ovulatory cycles did not differ among cows exposed to close physical contact; exposed to limited, fenceline contact; or not exposed to bulls. Angus x Hereford cows were assigned randomly to be exposed to close physical contact (experiment 1 (Exp. 1), BE; n = 14); limited, fenceline contact (Exp. 2, BEFL; n = 22); or not exposed (Exps. 1 and 2, NE; n = 37) to mature bulls beginning 59 days (Exp. 1) and 68 days (Exp. 2) after calving (day 0 in each experiment, respectively). In Exps. 1 and 2, anovular cows were exposed or not exposed to biostimulatory stimuli of bulls for 35 and 42 days, respectively. An increase in progesterone concentrations that exceeded 1.0 ng/mL in three consecutive samples was used as evidence of resumption of luteal function in Exp. 1. In Exp. 2, the presence of a corpus luteum in an ovary, assessed by ultrasonography, was used as evidence of resumption of luteal function. Percentage of cows that resumed luteal activity by the end of the Exp. 1 was greater (P < 0.05) for BE cows than for NE cows, whereas proportions of BEFL cows did not differ (P > 0.10) between BE or NE cows in Exp. 2. Interval from the start of the exposure period to resumption of luteal activity was shorter (P < 0.05) for BE and BEFL cows than for NE cows in Exps. 1 and 2, respectively. More (P < 0.05) BE than NE cows resumed luteal activity by 7 days after exposure and this difference was maintained throughout the exposure period in Exp. 1. However, in Exp. 2 proportions of BEFL cows that resumed luteal activity were greater (P < 0.05) than those of NE cows on days 14, 21, and 28 after exposure. By days 35 and 42, the proportion of BEFL cows that had resumed luteal activity did not differ (P > 0.10) from that NE cows. We conclude that the response of anovular, primiparous cows to the biostimulatory effect of bulls may depend on the intensity of exposure (frequency of exposure, duration of exposure, and quantity of stimuli) of pheromonal stimuli produced by bulls.  相似文献

Background  

The physiological mechanism by which bulls stimulate resumption of ovarian cycling activity in postpartum, anovular, suckled cows after calving may involve the concurrent activation of the hypothalamic-hypophyseal-ovarian (HPO) axis and hypothalamic-hypophyseal-adrenal (HPA) axis. Thus, the objectives of this experiment were to determine if characteristics of temporal patterns of cortisol and luteinizing hormone (LH) in postpartum, anovular, beef cows are influenced by acute exposure to bulls. The null hypotheses were that daily, temporal characteristics of cortisol and LH concentration patterns do not differ between cows exposed acutely to bulls or steers.  相似文献

The objective of this experiment was to determine if the proportion of first-calf suckled beef cows that resumed ovulatory cycles and the interval to resumption of ovarian cycling activity differ after exposure to either “unfamiliar” bulls or cows on d 35 postpartum, after exposure to either “familiar” bulls or cows for the first 30–32 d after calving. Fifty Angus × Hereford cows were stratified by calving date, calf BW, and calf sex by d 3 postpartum, and assigned to be exposed to familiar epididectomized bulls (BEF; n = 25) or familiar mature ovariectomized (OVX) cows (CEF; n = 25). On d 35 after calving, 12 BEF cows were assigned to be exposed to unfamiliar bulls (BEU); likewise, 12 CEF cows were assigned to be exposed to unfamiliar OVX cows (CEU). Cows were in their treatments for either 95 d (BEF and CEF) or 60 d (BEU and CEU) during the experiment. Blood samples were collected every third d from the beginning to the end of the experiment. A rise in progesterone concentration of >0.5 ng/mL in consecutive samples was used as the criterion for resumption of ovulatory cycles. Exposing cows to bulls on d 5 after calving and then switching a subset of these cows to be exposed to unfamiliar bulls 30–32 d later did not (P > 0.10) alter: (1) the proportion of cows that resumed cycling activity; and (2) postpartum interval to resumption of ovarian cycling activity compared to cows exposed to familiar bulls. However, 32% more (P < 0.05) cows exposed to bulls (BEF and BEU) resumed cycling activity by approximately 14.8 d before cows that were exposed to OVX cows (CEF and CEU). We conclude that the familiarity of first-calf cows to either bulls or ovariectomized cows, did not affect the postpartum occurrence of cycling activity or the interval from calving to resumption of ovulatory cycles. This experiment was designed to determine 1) if exposure of firstcalf suckled beef cows to mature bulls in the first 30 days, after 30 days, or continuously post partum reduces the postpartum anestrous period and 2) if exposure to bulls alters the first service pregnancy rate. Postpartum first-calf suckled crossbred (Angus x Hereford; Hereford x Angus) cows were randomly assigned to be 1) exposed continuously to mature, epididectomized bulls (BE; n = 18); 2) exposed to bulls for the first 30 days post partum (BE/NE; n = 17); 3) exposed to bulls after the first 30 days post partum (NE/BE; n = 16); or not exposed to bulls (NE; n = 18). Blood samples were collected weekly to be assayed for progesterone to determine resumption of ovarian cyclic activity. All the cows were bred for 21 days by AI while under their respective treatment regimens and were then exposed to fertile bulls for an additional 35 days. The postpartum interval to resumption of ovarian cyclic activity did not differ (P>0.10) among the 3 (BE, BE/NE, and NE/BE) treatment groups, but it was 15.4 d shorter (P<0.05) than for cows in the NE group. The overall pregnancy rates did not differ (P>0.10) among the treatment groups. The AI pregnancy rates for the BE/NE and NE/BE treatment groups were higher (P<0.05) than for the NE group. The AI pregnancy rates for the BE and NE treatment groups did not differ (P>0.10). The results showed that all three treatments (BE, BE/NE and NE/BE) similarly decrease the postpartum interval and that exposure to bulls may improve the reproductive performance of first-calf suckled beef cows.  相似文献

The objective of this experiment was to determine if duration of daily bull exposure influences length of postpartum anestrus in primiparous, anovular, suckled, beef cows. The null hypotheses were that intervals from calving or the start of bull exposure (D 0) to resumption of ovulatory activity (OA), and proportions of cows that resumed OA during the experiment does not differ among cows exposed to bulls for 0 h, 6 h, or 12 h daily, and that there is no relationship between the duration of bull exposure and interval to resumption of OA in cows exposed to bulls for 0 h, 6 h, or 12 h daily. At 51.5 ± 2.3 d (±SE) after calving, cows were assigned randomly to be exposed for 12 h (BE12; n = 15) or 6 h daily (BE6; n = 14) to bulls, or not exposed to bulls (NE; n = 10) for 45 d. Interval from calving or from D 0 to resumption of OA was shorter (P < 0.05) and the proportion of cows that resumed OA during the experiment was greater (P < 0.05) for BE12 than for NE cows. Interval from D 0 to resumption of OA did not differ (P > 0.10) between BE6 cows and either BE12 or NE cows. However, interval from calving to resumption of OA was shorter (P < 0.05) for BE6 than NE cows. The proportion of cows that resumed OA did not differ (P > 0.10) between BE6 cows and BE12 cows; however, the proportion of cows that resumed OA during the experiment tended (P = 0.08) to be greater for BE6 cows than for NE cows. There was a linear relationship between intervals from calving (b1 = −7.64 d/h; P < 0.05) and D 0 (b1 = −3.3 d/h; P < 0.05) to resumption of OA and duration of daily bull exposure. Thus, the duration of bull-pheromone stimuli that cows perceive each day is related to when primiparous, postpartum, anestrous, suckled cows respond to this stimulus and undergo the physiological changes necessary to resume ovulatory activity.  相似文献

This experiment was designed to determine if continuous bull exposure of heifers 2 wk post partum to a first calf (30 to 36 m old) or mature (42 to 72 m old) Zebú Brahman suckled cows reduces the interval to first estrus and the number of days open. Post partum first calf cows were randomly assigned to be exposed continuously to epidectomized bulls with surgical ablation of the dorsal ligament of the penis (Group 35 FBE, n = 25) or not exposed to bulls (Group FNE, n = 22). Post partum mature cows were likewise assigned to be continuously exposed (Group MBE, n = 24) or not exposed to bulls (Group MNE, n = 25). Milk samples were collected twice a week for progesterone assays to determine resumption of ovarian cyclic activity. All the cows were artificially inseminated for 120 d while in their respective treatment groups. The first postpartum estrus percentages were 60 < 90 d, 24.4%; and > 90 d, 33.7%. The interval to first estrus f-or Groups FBE and FNE was 75.4 and 104.2 d, respectively, and differed significantly (P < 0.01). For Groups MBE and MNE the interval to first estrus was 76.3 and 95.0 d, respectively (P < 0.05). The number of open days for Groups FBE and FNE was 77.3 and 114.9, respectively (P < 0.01); while for Groups MBE and MNE it was 73.0 and 98.6 d, respectively (P < 0.01). The results of the present study indicate that introduction of bulls to first calving cows or mature Zebú Brahman cows after parturition provides a practical and economical option for reducing the extended post partum anestrus interval and days open under tropical conditions.  相似文献

In Experiment I, 38 crossbred suckled beef cows grazing fescue pastures and 34 crossbred beef cows grazing bluestem pastures were randomly allocated at the time of calving into a group with 4 teaser bulls or no bulls. Two blood samples were collected 7 d apart from the cows to determine cyclic activity 67 and 76 d after calving in the fescue and bluestem pastures, respectively. Progesterone greater than 1.0 ng/ml in one or both samples indicated cyclic activity. There was no difference in the percentage of cows cyclic among the different groups. The number of cyclic cows in the fescue pasture with bulls was 16/19 (84%); in the fescue pasture with no bulls, 14/19 (74%); in the bluestem pasture with bulls, 17/17 (100%); and in the bluestem pasture with no bulls, 16/17 (94%). Overall cyclic activity among all cows for teaser bull-exposed and no bull was similar, 33/36 (91%) and 30/36 (83%). Overall cyclic activity was greater (P < 0.05) in cows grazing bluestem (33/34), 97% than fescue pastures (30/38), 80%. Measurements of cyclic activity were initiated too late in the postcalving period to quantify differences in estrous activity between the bull and no bull treatment groups. Another trial was planned for the following year with a modified protocol. In Experiment II, blood samples were collected for progesterone concentrations soon after calving and were repeated at intervals to characterize both the occurrence and duration of estrous cycles. In this experiment, 29 crossbred suckled beef cows grazing fescue pastures were randomly allocated 12 d after calving (Day 0) into 1 of 2 groups with teaser bulls or without bulls. Nineteen crossbred beef cows grazing bluestem pastures were allocated similarly 10 d after calving (Day 0). Bulls were added to the groups with bulls in fescue and bluestem pastures on day 6 after the initial allocations. Blood samples were collected from all cows on Day 0 and every 3 d until Day 46. Means (+/- SEM) of the cumulative progesterone concentrations (ng/ml) per cow for the 16 samples from cows grazing fescue were 12.5 +/- 3.5 for cows exposed to bulls, 2.5 +/- 0.16 for cows not exposed to bulls, 27.6 +/- 4.42 for cows grazing bluestem pastures and exposed to bulls, and 16.0 +/- 2.75 for cows without exposure to bulls. Progesterone concentrations were higher in cows exposed to bulls (P < 0.01). The percentages of both short and normal cycles increased (P < 0.01) in groups exposed to bulls (88%, 21/24 and 63%, 15/24) when compared with the no bull groups (29%, 7/24 and 21%, 5/24), respectively. The duration of anestrus in cattle was usually shortened when cows were exposed to bulls. The objective of the present experiment was to determine if weekly bull exchange accelerated the resumption of cyclicity in postpartum suckled beef cows. We tested the hypothesis that exposure of postpartum, anestrous, suckled beef cows (extensively managed) to weekly exchange of bulls, accelerates the resumption of cyclicity, compared to continuous exposure to the same bulls. Ninety-one multiparous suckled Hereford and Hereford x Angus cows, <60d postpartum, were assigned to two homogeneous groups. Beginning on December 1st (late spring), the control group (C, n=45) remained with one pair of bulls throughout the breeding period (7 weeks), whereas in the "exchanged" (E, n=46) group two pairs of bulls were exchanged weekly. Based on weekly ultrasonographic examinations of all cows, none had a CL at the start of the experiment and for 2 weeks after the start of bull exposure. However, the accumulated frequency of cows with a CL was greater in group E than in group C cows on week 4 (P=0.024), as well as on weeks 5-7 (P<0.001) after the start of bull exposure. Furthermore, in group E versus group C, there was a higher pregnancy rate 30d after the end of bull exposure (26 of 46, 56.2% vs. 16 of 45, 35.6%; P=0.045). In conclusion, weekly exchange of two pairs of bulls shortened postpartum anestrus in suckled multiparous cows, compared to continuous exposure to a single pair of bulls.  相似文献

The objective of this study was to investigate the relationships among body condition score (BCS), body weight and plasma metabolite concentrations, and resumption of ovarian cycles postpartum in high-producing dairy cows in a subtropical region in Japan. The study was conducted between May 2001 and January 2003 in a commercial dairy farm in Hiroshima. Cows were kept in free-stall barns with sawdust and wood shavings for bedding, and fed a total mixed ration consisting of alfalfa, timothy and oat hay, corn, tofu ground-wet, beet pulp, cottonseed and soybean. Milk samples were collected twice weekly from 2 to 11 weeks postpartum, and progesterone concentrations in skim milk were determined by ELISA. At 8 and 2 weeks before calving and 3, 5, 7, 9, and 11 weeks after calving, both BCS and body weights were determined and blood samples were collected to determine plasma concentrations of total cholesterol (T-cholesterol), non-esterified fatty acids (NEFA), total protein, and urea nitrogen. The average 305 days milk yield was 10,422 kg. Out of 110 cows, 44.5% had normal resumption (ovulation occurred < or =45 days after calving, followed by regular ovarian cycles), 33.6% had a prolonged luteal phase (luteal activity >20 days), 12.7% had delayed first ovulation (no ovulation until < or =45 days after calving), 4.6% had a short luteal phase (luteal activity <10 days, except in the first cycle), and the remaining 4.6% had cessation of cyclicity (no luteal activity > or =14 days between cycles). The BCS of cows with delayed first ovulation were lower than the BCS of cows with normal resumption and prolonged luteal phase at 5 weeks (2.9+/-0.1 versus 3.1+/-0.1 and 3.2+/-0.1; P<0.05), 7 weeks (2.8+/-0.1 versus 3.1+/-0.1 and 3.2+/-0.1; P<0.01), 9 weeks (2.7+/-0.1 versus 3.0+/-0.1 and 3.2+/-0.1; P<0.01) and 11 weeks (2.6+/-0.1 versus 3.0+/-0.1 and 3.2+/-0.1; P<0.01) after calving. None of the plasma parameters differed (P>0.05) among different groups of cows. Cows loosing > or =1 unit BCS after calving had a prolonged interval to commencement of luteal activity, and were at greater risk of having delayed first ovulation. Poor postpartum nutritional status was associated with delayed first ovulation postpartum. Regular monitoring of BCS before and after calving would be useful in nutritional management, and help in preventing delayed first ovulation in high-producing dairy cows.  相似文献

A total of 135 postpartum suckled zebu cows were assigned randomly at calving to two treatments: cows exposed to vasectomised bulls (Mature Bull Exposure, MBE) and cows not exposed to bulls (No Bull Exposure, NBE) for a period of 150 days postpartum. This study was conducted to determine the influence of bull biostimulation, season and parity on postpartum reproductive performance of Bos indicus cattle. The trials were conducted in two seasons: cows calving in the dry season and cows calving in the rainy season. Cows with an increase in serum progesterone (P(4)) concentration of >1 ng/ml from the weekly blood samples were used to analyse the number of days from calving to the time of resumption of ovarian activity. The introduction of vasectomised bulls shortened the postpartum anoestrus in cattle following calving. Mean interval from calving to resumption of ovarian activity for the MBE cows was 71.7 days, which was significantly earlier following calving than the NBE cows with a duration of 77.8 days. By 60 to 80 days postpartum, the proportion of cows at resumption of postpartum activity for MBE cows was greater than for the NBE cows.Mean interval from calving to resumption of ovarian activity for cows that calved in the dry season was 71.3 days, which was significantly earlier than for cows that calved in the rainy season (78.6 days). At 60 days postpartum, the proportion of cows at resumption of postpartum ovarin activity for cows that calved in the dry season was greater than the cows that calved in the rainy season. Mean interval from parturition to resumption of ovarian activity for cows with three to five calvings was 65.1 days, which was significantly earlier than the value of 71. 2 days for cows with one to two calvings. By 60 to 80 days postpartum, the proportion of cows at onset of ovarian activity for cows with three to five calvings was greater than those cows with one to two calvings. Cows that calved in the dry season completed uterine involution by 24.4 days, which was significantly shorter than the duration of 26.5 days for cows that calved in the rainy season. Cows with three to five calvings completed uterine involution earlier than those with one to two calvings. It is concluded that bull-cow biostimulation influences reproductive activity in the cow possibly through olfactory cues (pheromones).  相似文献

Concentrations of cortisol were determined in pooled fluid of small (less than 10 mm) and large (greater than or equal to 10 mm) follicles of cyclic cattle (Exp. 1), and in fluid of the largest follicle of 17 post-partum anovulatory cows (Exp. 2). In Exp. 1, concentrations of cortisol in small follicles were greater (P less than 0.05) than in large follicles (14.7 versus 13.2 ng/ml), and varied significantly with stages of the cycle; small and large follicles had the highest cortisol concentration during the early luteal phase of the cycle. Large follicles had 2-fold greater concentrations of oestradiol than did small follicles, whereas small follicles had 2-fold greater concentrations of androstenedione than did large follicles. Across pools of follicular fluid, cortisol concentrations were correlated only to androstenedione concentrations (r = 0.65, P = 0.07). In Exp. 2, concentrations of cortisol did not significantly differ between oestrogen-active (oestradiol greater than progesterone in follicular fluid) and oestrogen-inactive (progesterone greater than oestradiol) follicles, although oestrogen-active follicles had a 24-fold greater concentration of oestradiol than did oestrogen-inactive follicles. Cortisol concentrations were correlated to hCG binding capacity of thecal cells (r = -0.35, P = 0.08) and to follicular diameter (r = 0.45, P less than 0.05). These results suggest that normally fluctuating concentrations of cortisol in follicular fluid of cattle play little or no active role in follicular differentiation in vivo.  相似文献

The objective of this study was to investigate the resumption of ovarian cyclicity postpartum in high-producing dairy cows in commercial dairy farms under subtropical conditions. The cows were kept in a free-stall or tie-stall barn. Milk samples were collected from cows twice weekly, and progesterone in the skim milk was assayed by double-antibody ELISA. Cows were examined rectally and vaginoscopically at 2-week intervals after calving. Body condition score (BCS) and body weights were taken before and after calving. A cow was considered to have resumed ovarian cyclicity on the day of ovulation if followed by regular ovarian cycles. Thirty seven percent (n=20/54) of the cows had normal resumption of ovarian cyclicity (resumption within 45 days after calving), and 63% (n=34/54) had delayed resumption (resumption did not occur until >45 days after calving). Delayed resumption Type I (one or more ovarian cycles with luteal phase >20 days, i.e. prolonged luteal phase; 31.5%) and delayed resumption Type II (first ovulation did not occur until > or =45 days after calving, i.e. delayed first ovulation; 24.1%) were the most common types of delayed resumptions. Almost half (46.3%) of the cows did not resume their ovarian cyclicity until >65 days postpartum. Cows with delayed resumption Type I had a higher incidence of abnormal cervico-vaginal discharge (64.7%) and incomplete uterine involution (94.1%) compared to cows with normal resumption (P<0.01). The BCS of cows with delayed resumption Type II were lower than those of normal resumption cows at 5 weeks and later in the postpartum period (P<0.05). Approximately two-thirds of high-producing cows had delayed resumption of ovarian cyclicity postpartum. Prolonged luteal phase and delayed first ovulation were two important ovarian dysfunctions that delayed postpartum resumption of cyclicity in high-producing dairy cows.  相似文献

The objective of this study was to investigate whether bull exposure affects LH profiles in postpartum, anoestrous dairy cows. Eight cows between 10 and 17 days after parturition were used. On Day 1, blood samples were taken at 10 min intervals for 8 h. On Day 2, blood sampling continued at 10 min intervals and after 2 h a bull was introduced behind a fence, and blood sampling continued for another 8 h. Time of resumption of luteal activity was between 25 and more than 80 days after parturition for these animals and was not related (P>0.1) with frequency of LH pulses, amplitude of pulses and basal LH concentration on either Day 1 or Day 2. In 6 of the 8 cows, average and basal LH concentration were greater (P<0.001) during the 8 h of bull presence (0.56 +/- 0.33 and 0.39 +/- 0.26 ng/ml, respectively) compared to the 8 h without a bull (0.50 +/- 0.30 and 0.35 +/- 0.24 ng/ml, respectively). Pulse amplitude did not differ (P=0.85) between Day 2 (0.45 +/- 0.24 ng/ml) or Day 1 (0.45 +/- 0.14 ng/ml). LH pulse frequency was greater (P<0.1) on Day 2 (5.3 pulses/8h) compared to the Day 1 (4.6 pulses/8h). In conclusion, fenceline bull exposure early postpartum seems to have an acute effect on LH-release in anoestrous dairy cows. Whether sustained bull exposure can hasten first ovulation after calving through an effect on LH release in dairy cows is an interesting area of research.  相似文献

Concentrations of LH, cortisol, estradiol-17beta (E(2)), prolactin and 13,14-dihydro-15-keto-prostaglandin F(2alpha) (PGFM) were determined in cows with experimentally induced clinical mastitis during early lactation. Cows free of intramammary infection (IMI) and in the luteal phase of the estrous cycle were balanced by lactation number and days in milk and assigned to either control (n=5) or treatment (n=5) groups. Treated cows were infected experimentally (day 0), in two mammary quarters, with Streptococcus uberis and developed clinical mastitis within 60 h after inoculation as evidenced by increased mastitis scores, elevated rectal temperatures, mammary swelling and isolation of S. uberis pathogen. Four days following bacterial challenge, blood samples were collected every 20 min for 8 h for determination of PGFM and LH following administration of oxytocin and GnRH, respectively. Blood samples were also collected on days 0, 4 and 7 of the experiment to determine concentrations of E(2), prolactin and cortisol. Four days after bacterial challenge, concentrations of cortisol were higher (P=0.04) in experimentally infected cows than controls. Experimentally challenged cows had increased (P=0.02) concentrations of cortisol on days 4 and 7 compared with day 0. Control cows had no significant increase in blood cortisol during the experimental period. Baseline concentrations of PGFM did not differ between groups; however, peak concentrations of PGFM following oxytocin challenge were elevated (P=0.006) in cows with clinical mastitis compared with control animals. Prolactin, E(2) and LH did not differ between cows with clinical mastitis or controls. Experimentally induced mastitis during early lactation elevated concentrations of cortisol during the luteal phase of the estrous cycle. Furthermore, mastitic cows demonstrated an increased PGFM response following oxytocin administration. Altered reproductive efficiency in cows with clinical mastitis caused by Gram-positive pathogens may be the result of increased uterine sensitivity to prostaglandin F(2alpha) (PGF(2alpha)).  相似文献

Suckled postpartum Zebu (Bos indicus) cows were exposed either to estrous females alone or together with bulls to determine if this regimen would stimulate the resumption of ovarian activity. The study was carried out on 60 Zebu cows. The animals were allocated at parturition to 1 of 3 treatment groups. Group FB cows (n = 14) were exposed to progestogen treated females (approximately 3 cows per progestogen treated cow) and a bull for 42 d. Group F cows (n = 31) were exposed to progestogen treated females and after 21 d a bull was introduced and remained with the group until the end of the experiment 21 d later. Estrus was induced with Syncro Mate B treatment and the cows were in estrus when introduced into Groups FB and F. Group N cows (n = 15) were introduced to a bull 21 d after the treatments had begun in Groups FB and F, and the bull remained with these cows until the end of the study. On average, resumption of ovarian activity occurred in 57% (8/14), 19% (6/31) and 7% (1/15) of the cows in Group FB, F and N, respectively, within 21 d after the start of treatments. The difference was significant between Group FB and Groups F and N (P < 0.05). During the second part of the experiment, in which the bull was introduced to Groups F and N, an additional 32% (10/31) of the cows in Group F and 33% (5/15) in Group N resumed ovarian activity. No cows in Group FB resumed ovarian activity during this period. There were significant differences in body condition between cows that resumed ovarian activity and those that remained in anestrus (P < 0.05). We conclude that biostimulation effectively enhanced resumption of ovarian activity in anestrous suckled Zebu cows. This technique offers a potentially useful and practical way to improve reproductive efficiency in anestrous Zebu cattle in the tropics.  相似文献

Ovarian function of nutritionally induced anoestrus cows was evaluated in vivo (Expt 1) and in vitro (Expt 2). In Expt 1, 32 nutritionally induced anoestrous beef cows were divided into four treatment groups receiving: (1) saline infusions at one pulse every 4 h for 13 days (control); (2) 2 micrograms GnRH at one pulse every 4 h (2 micrograms infused in 1.8 ml saline over 5 min) for 13 days (GnRH-4); (3) 2 micrograms GnRH at one pulse every 1 h for 13 days (GnRH-1); and (4) continuous infusion of 2 micrograms GnRH (a total of 2 micrograms in 34 ml h-1) for 13 days (GnRH-C). On the last day of treatment, cows were killed, ovaries were removed and follicular fluid samples (n = 149) were collected. The percentage of cows with luteal activity on day 13 was significantly different (P < 0.01) among treatments (0, 25, 75 and 25% for control, GnRH-4, GnRH-1 and GnRH-C cows, respectively). Owing to the large percentage of ovulatory cows in the GnRH-1 group (n = 6), anovulatory cows (n = 2) were removed from this treatment group for statistical analysis, as were cows with luteal tissue from the GnRH-4 (n = 2) and GnRH-C (n = 2) groups. The numbers of small (1.0-4.9 mm) and medium plus large (> or = 5 mm) follicles were not affected (P > 0.10) by treatment. However, GnRH-4 cows (n = 6) had greater (P < 0.05) concentrations of oestradiol in follicular fluid than did control (n = 8) but not GnRH-1 (n = 6) or GnRH-C (n = 6) cows. Concentrations of insulin-like growth factor I were greater (P < 0.05) in the follicular fluid of GnRH-1 cows than in all other treatment groups. Concentrations of androstenedione and progesterone in follicular fluid were not affected (P > 0.10) by treatment or follicle size. The binding activity of insulin-like growth factor binding proteins was not affected by GnRH treatment. However, the binding activity of insulin-like growth factor binding protein 2, 29-32 kDa and 22 kDa insulin-like growth factor binding proteins were greater (P < 0.05) in small versus medium plus large follicles. In Expt 2, granulosa cells were collected from nutritionally anoestrous cows to determine whether ovarian cells from anoestrous cows have the capacity to respond to insulin-like growth factor I or insulin in vitro. Both insulin-like growth factor I (20 and 200 ng ml-1) and insulin (10, 100 and 1000 ng ml-1) increased (P < 0.05) granulosa cell proliferation and progesterone production. In conclusion, pulsatile infusion of 2 micrograms GnRH (every 1 or 4 h) for 13 days into nutritionally induced anoestrous cows results in increased intrafollicular oestradiol and insulin-like growth factor I concentrations and can stimulate ovulation without markedly affecting concentrations of androstenedione or progesterone, or the binding activity of insulin-like growth factor binding proteins, in follicular fluid. In addition, granulosa cells from nutritionally induced anoestrous cows have the capacity to respond to insulin-like growth factor I and insulin in vitro, indicating that the decrease in trophic factors observed with restricted feeding does not reduce the response of the ovary to insulin-like growth factor I and insulin.  相似文献

Background

The main objective of this study was to estimate the effect of supplementation with Saccaromyces cerevisiae (SC) (Yea-Sacc® 1026) on milk production, metabolic parameters and the resumption of ovarian activity in early lactation dairy cows.

Methods

The experiment was conducted during 2005/2006 in a commercial tied-house farm with an average of 200 milking Estonian Holstein Friesian cows. The late pregnant multiparous cows (n = 46) were randomly divided into two groups; one group received 10 g yeast culture from two weeks before to 14 weeks after calving. The groups were fed a total mixed ration with silages and concentrates. Milk recording data and blood samples for plasma metabolites were taken. Resumption of luteal activity was determined using milk progesterone (P4) measurements. Uterine bacteriology and ovarian ultrasonography (US) were performed and body condition scores (BCS) and clinical disease occurrences were recorded. For analysis, the statistical software Stata 9.2 and R were used to compute Cox proportional hazard and linear mixed models.

Results

The average milk production per cow did not differ between the groups (32.7 ± 6.4 vs 30.7 ± 5.3 kg/day in the SC and control groups respectively), but the production of milk fat (P < 0.001) and milk protein (P < 0.001) were higher in the SC group. There was no effect of treatment on BCS. The analysis of energy-related metabolites in early lactation showed no significant differences between the groups. In both groups higher levels of β-hydroxybutyrate (BHB) appeared from days 14 to 28 after parturition and the concentration of non-esterfied fatty acid (NEFA) was higher from days 1–7 post partum (PP). According to US and P4 results, all cows in both groups ovulated during the experimental period. The resumption of ovarian activity (first ovulations) and time required for elimination of bacteria from the uterus did not differ between the groups.

Conclusion

Supplementation with SC had an effect on milk protein and fat production, but did not influence the milk yield. No effects on PP metabolic status, bacterial elimination from the uterus nor the resumption of ovarian activity were found.

Peripheral concentrations of cortisol, growth hormone and testosterone were determined in two experiments which examined the endocrine and behavioral responses of sexually mature Angus bulls to an estrous female (Experiment 1) and to female exposure 5 hours following an adrenocorticotropin (ACTH) injection (Experiment 2). Sexual activity of bulls in Experiment 1 significantly increased levels of cortisol when compared with concentrations before exposure to a female. Administration of ACTH in Experiment 2 consistently elevated levels of cortisol by 30-fold (P<0.01) when compared with pre-ACTH concentrations. This heightened level of cortisol persisted throughout the period of exposure to an estrous cow, although a gradual decline in cortisol concentrations occurred over time (P<0.05). In Experiment 1, growth hormone profiles tended to increase in response to sexual activity (P<0.10), whereas in Experiment 2, growth hormone increased in response to ACTH administration (P<0.01) and to female exposure (P<0.01). Concentrations of testosterone were unaffected (P>0.10) by mating activity in Experiment 1. In Experiment 2, acute suppression (P<0.01) in testosterone concentrations 5 hours after ACTH administration coincided with the exposure period to the estrous female. Frequencies of mounting behavious (penis extension, mounting, intromission and ejaculation) exhibited by ACTH-treated bulls were significantly lower compared with the frequencies two days earlier. Exogenous ACTH administration suppressed reproductive behaviors of bulls and altered secretion of cortisol, growth hormone and testosterone. Furthermore, these data provide evidence that specific mating behaviors of the bull can be influenced by circulating steroids.  相似文献
