The objective was to synchronize follicular wave emergence among cattle for synchronization of estrus and ovulation, and to determine pregnancy rate after AI at observed estrus. At random stages of the estrous cycle, a controlled internal drug release device (CIDR-B) was inserted intravaginally (Day 0) in 67 cross-bred beef heifers, and they were randomly allocated to receive either no further treatment (Control; n = 18); 5 mg of estradiol-17beta and 100 mg of progesterone im (E/P; n = 16); 100 microg im of GnRH (GnRH; n = 16); or transvaginal ultrasound-guided follicular ablation of all follicles > or = 5 mm (FA; n = 17). All heifers received a luteolytic dose of PGF (repeated 12 h later), and CIDR-B were removed on Days 9, 8, 6 or 5, in Control, E/P, GnRH or FA groups, respectively, so the dominant follicle of the induced wave was exposed to exogenous progesterone for a similar period of time in each group. Mean (+/- SEM) intervals (and range, in days) from treatment to follicular wave emergence in these groups were 3.5 +/- 0.6 (-2 to 8), 3.4 +/- 0.1 (3 to 4), 1.5 +/- 0.3 (-1 to 4), and 1.0 +/- 0.1 (0 to 2), respectively. Although the interval was longest (P<0.01) in the E/P and Control groups, it was least variable (P<0.01) in the E/P and FA groups. Intervals (and range, in days) from CIDR-B removal (and first PGF treatment) to estrus were 2.3 +/- 0.2 (1.5 to 4.5), 2.2 +/- 0.2 (1.5 to 3.0), 2.1 +/- 0.1,(1.5 to 3.5), and 2.5 +/- 0.1 (2.0 to 3.5), and to ovulation were 3.5 +/- 0.2 (2.5 to 5.5), 3.4 +/- 0.1 (3.0 to 4.5), 3.5 +/- 0.1 (2.5 to 4.5), and 3.8 +/- 0.1 (3.0 to 4.5), for Control, E/P, GnRH and FA groups, respectively (ns). The proportion of heifers displaying estrus was higher in the Control than in the FA group (94% versus 65%, P<0.05) and intermediate in EP and GnRH groups (87% and 75%). Heifers were inseminated approximately 12 h prior to ovulation (based on estrous behavior and ultrasound examinations). Pregnancy rates were 78%, 80%, 69% and 65% for Control, E/P, GnRH and FA groups, respectively (P=0.73). Results support the hypothesis that synchronous follicular wave emergence results in synchronous follicle development and, following progesterone removal, synchronous estrus and ovulation with high pregnancy rates to AI. The synchrony of estrus and ovulation in the E/P, GnRH and FA groups suggest that these treatments, in combination with CIDR-B, could be adapted to fixed-time insemination programs.  相似文献

The effects on estrus and fertility of 3 estrus synchronization protocols were studied in Brahman beef heifers. In Treatment 1 (PGF protocol; n=234), heifers received 7.5 mg, i.m. prostianol on Day 0 and were inseminated after observed estrus until Day 5. Treatment 2 (10-d NOR protocol; n = 220) consisted of norgestomet (NOR; 3 mg, s.c. implant and 3 mg, i.m.) and estradiol valerate (5 mg, i.m.) treatment on Day -10, NOR implant removal and 400 IU, i.m. PMSG on Day 0, and AI after observed estrus through to Day 5. Treatment 3 (14-d NOR+PGF protocol; n = 168) constituted a NOR implant (3 mg, sc) on Day -14, NOR implant removal on Day 0, PGF on Day 16, and AI after observed estrus through to Day 21. All heifers were examined for return to estrus at the next cycle and inseminated after observed estrus. The heifers were then exposed to bulls for at least 21 d. During the period of estrus observation (5 d) after treatment, those heifers treated with the PGF protocol had a lower (P<0.01) rate of estrual response (58%) than heifers treated with the 10-d NOR (87%) or 14-d NOR+PGF (88%) protocol. Heifers treated with the 10-d NOR protocol displayed estrus earlier and had a closer synchrony of estrus than heifers treated with either the PGF or the 14-d NOR+PGF protocol. Heifers treated with the 14-d NOR+PGF protocol had higher (P<0.05) conception and calving rates (51 and 46%) to AI at the induced estrus than heifers treated with the PGF (45 and 27%) or the 10-d NOR (38 and 33%) protocol. Calving rate to 2 rounds of AI was greater (P<0.05) for heifers treated with the 14-d NOR-PGF (50%) protocol than heifers treated with the 10-d NOR (38%) but not the PGF (43%) protocol. Breeding season calving rates were similar among the 3 protocols. The results show that the 14-d NOR+PGF estrus synchronization protocol induced a high incidence of estrus with comparatively high fertility in Brahman heifers.  相似文献

The purpose of the experiment was to test the hypothesis that a variable and/or insufficient level of progestagen at the end of a treatment to synchronize estrus in goats could explain variability in the onset of estrus. The experiment was performed during the anestrous season on 2 herds, one of Alpine (n = 49) the other of Saanen (n = 53) dairy goats. The animals were allocated to 1 of 3 treatments: Group 1 received a vaginal sponge impregnated with 45 mg of fluorogestone acetate (FGA) on Day 0; Group 2 received a sponge on Day 0 plus a second sponge on Day 7; Group 3 received a sponge on Day 0 plus a second sponge on Day 9. The sponges were withdrawn on Day 11. All goats received 400 or 500 IU eCG and 50 mug PGF(2alpha) analog 48 h prior to sponge removal. They were inseminated with frozen-thawed semen 24 h after the onset of estrus. Among treatment groups no difference (P > 0.05) was observed for the following parameters: percentage of goats in estrus, percentage of goats ovulating, mean time and variability of onset of estrus. The fertility of Alpine goats in Group 3 was significantly decreased (P < 0.05). No effect on prolificacy was noticed. These observations show that to increase progestagen level at the end of treatment did not improve estrus synchronization. They provide further evidence that treatments with too high progestagen amounts can decrease fertility.  相似文献

The main objective of this study was to investigate the effectiveness of certain progestagen-gonadotrophin treatments on synchronization of estrus in sheep. In Experiment I, 30 Chios ewes were treated at the beginning of the breeding season with medroxyprogesterone acetate (MAP) intravaginal sponges for 12 days and a single i.m. treatment of either FSH (Group 1,10 IU, n = 8; Group 2, 5 IU, n = 8; Group 3, 2.5 IU, n = 8) or eCG (Group 4, 400 IU, n = 6) at the time of sponge removal. Ten days after sponge removal laparotomy was performed to record ovarian response. Clinical estrus was observed in more (though not at a significant level) FSH treated than eCG treated sheep (62.5% versus 33.3%). Administration of 400 IU eCG resulted in the highest mean number of CL perewe ovulating (2.8 +/- 0.2), with administration of 10 IU FSH producing the next best results (2.1 +/- 0.3). Statistically significant differences in the mean number of CL per ewe ovulating were found only between ewes in Group 3 (1.7 +/- 0.4) and Group 4 (2.8 +/- 0.2) (P < 0.05). In Experiment II, 53 Chios and 30 Berrichon ewes were treated during the mid-breeding season with MAP intravaginal sponges for 12 days and a single i.m. treatment of either 10 IU FSH (27 Chios and 16 Berrichon ewes) or 400 IU eCG (26 Chios and 14 Berrichon ewes), at the time of sponge removal. Ewes that were in estrus on Days 2-4 and 19-23 after sponge removal were mated to fertile rams. No significant differences were recorded between treatment or breed groups in the proportions of ewes observed in estrus after treatment. In the Berrichon breed, FSH administration resulted in higher lambing rates (93.8% versus 57.1%, P < 0.05) and higher mean number of lambs born per ewe exposed to rams (1.4 +/- 0.2 versus 0.8 +/- 0.2, P < 0.05) than that of eCG. After treatment with eCG, the mean number of lambs born per ewe exposed to rams was higher in the Chios than the Berrichon breed (1.4 +/- 0.2 versus 0.8 +/- 0.2, P < 0.05). After treatment with FSH, the lambing rate was higher in the Berrichon than the Chios breed (93.8% versus 63.0%, P < 0.05). In conclusion, a single FSH treatment (5 or 10 IU) at the end of progestagen treatment appears to be more effective than eCG for the induction of synchronized estrus in sheep at the beginning of the breeding season, with no cases of abnormal ovarian response observed. During the mid-breeding season FSH (10 IU) appears to be equally as effective as eCG (400 IU) in respect of lambing rate and mean number of lambs born per ewe.  相似文献

In the present study, two new short estrus synchronization methods have been developed for lactating dairy cows. The study was completed in three consecutive phases. In experiment (Exp) 1, 32 cows, that were not detected in estrus since calving between the 50th and 84th post-partum days, were treated with PGF2alpha (PGF, d-cloprostenol, 0.150 mg), estradiol propionate (EP, 2mg) and GnRH (lecirelina, 50 microg) at 24h intervals, respectively, and timed artificial insemination (TAI) was performed 48 h after PGF. Different from Exp 1, EP and GnRH were given at 48 and 60 h, respectively after PGF in Exp 2 (n=20), instead of 24 and 48 h. Ovulations were investigated by ultrasound for 7 days starting from the day of PGF treatment, and ovulation rates were compared with the ones obtained in Exp 1. In Exp 3, cows were given the same treatments as Exp 2, but treatments started at certain estrus stages. Cows detected in estrus and with a confirmed ovulation (n=27) after the second PGF given 11 days apart were assigned to three treatment groups. Treatment was initiated at Day 3 (group metestrus, n=9), Day 12 (group diestrus, n=9) and Day 18 (group proestrus, n=9) after ovulation. All cows included in Exp 3 were TAI between 16 and 20 h after GnRH treatment. In Exp 2 and 3, blood samples were obtained once every 2 days, starting from Day 0 to the 10th day after GnRH injection, and once every 4 days between the 10th and the 22nd days after GnRH to examine post-treatment luteal development. During the study, animals exhibiting natural estrus were inseminated and served as controls (n=85). The rate of estrus was found to be significantly higher in cows with an active corpus luteum (CL) at the start of Exp 1 (72.7% vs. 30.0%, P<0.05) and the pregnancy rate tended to be higher than cows without an active CL (40.9% vs. 10.0%, P=0.08). Compared to those in Exp 1, cows in Exp 2 had higher rates of synchronized ovulation (94.1% vs. 59.1%, P=0.013). In Exp 3, estrus (P<0.001) and pregnancy rates (P=0.01) were found to be significantly higher in cows in the proestrus group than in those in the metestrus group. Comparable pregnancy rates were obtained from the first and second inseminations in Exp 1 and 3 with results from those inseminated at natural estrus (P>0.05). It was concluded from the study that the treatment in Exp 1 and 3 could result in comparable pregnancy rates after timed AI of lactating dairy cows at random stages of the estrus cycle relating to those inseminated at natural estrus, but the stage of the estrus cycle can have significant effects on pregnancy rates.  相似文献

Genetic engineering of miniature pigs has facilitated the development of numerous biomedical applications, such as xenotransplantation and animal models for human diseases. Manipulation of the estrus is one of the essential techniques for the generation of transgenic offspring. The purpose of the present study was to establish a useful method for induction of the estrus in miniature gilts. A total of 38 pubertal miniature gilts derived from 4 different strains were treated with exogenous gonadotropins. Estrus and ovulatory response were examined after treatment with pregnant mare serum gonadotropin (PMSG) and human chorionic gonadotropin (hCG) as 200 IU PMSG and 100 IU hCG, 300 IU PMSG and 150 IU hCG, or 1,500 IU PMSG only, followed by 100, 150 or 750 IU hCG 72 h later, respectively. The optimal protocol was determined to be the combination treatment of 200 IU PMSG and 100 IU hCG followed by 100 IU hCG. The administration of 200 IU PMSG and 100 IU hCG was effective in inducing estrus regardless of the strain, although there was a strain difference in the ovulatory response. These results indicate that treatment with a low-dose combination of PMSG and hCG provides one of the simplest methods for induction of estrus and ovulation in pubertal miniature pigs.  相似文献

Mature, cyclic Ethiopian Menze ewes (n = 72) were used in this study. They were divided into 6 equal groups in a 2x3 factorial experimental design. Estrus and ovulation were synchronized in all ewes using either 2 dosages of prostaglandin F2 alpha 12 days apart (n = 36) or intravaginal progestogen sponges for an equal length of time (n = 36). At sponge removal or at the second prostaglandin injection, equal groups of ewes were injected with either 0, 200, or 300 IU of PMSG. Prostaglandin-synchronized ewes exhibit estrus significantly earlier (P = 0.025) than the progestogen-synchrcnized group. Although PMSG treatment increased twinning rates and therefore total number of lambs born, the differences between groups did not reach significant levels (P>0.10).  相似文献

The induction of estrus in 17 previously cycling nulliparous ewes, 9 to 10 months of age, was attempted with Medroxyprogesterone acetate (MAP) pessaries during the early anestrous period (March-April). Ewes were verified to be anestrous by the lack of estrous behavior in the presence of a vasectomized ram and by a radioimmunoassay for serum progesterone in two samples taken 7 days apart showing less than 1 ng/ml serum progesterone. Superovulation was attempted with injections of either FSH or FSH + LH. MAP vaginal pessaries remained in place for a period of 12 days and FSH was administered to all ewes (IM) at 12 hr intervals over a 3 day period; 5 mg was injected twice on day 11 after pessary insertion, followed by 4 and 3 mg injections twice daily on each succeeding day, for a total of 24 mg per ewe. Nine ewes were given 25 mg LH (IV) within 8 hrs after the onset of behavioral estrus in addition to FSH. Ewes were hand-mated to several rams at 12 hr intervals throughout the estrus period. Ovulation and fertilization rates were recorded for each ewe following midline laparotomy and embryo collection. All ewes were in estrus between 36 and 48 hrs after removal of the MAP pessaries. In ewes injected with FSH only, 8 of 8 ovulated with a mean ovulation rate of 6.0 +/- 4.4 and a fertilization rate of 70%. Nine of 9 ewes receiving both FSH + LH ovulated with a mean ovulation rate of 13.9 +/- 13.1 and a fertilization rate of 72%. Statistical analysis by Students t-test resulted in differences in number of ova recovered (P<.05) between FSH only and FSH + LH treated ewes and a trend towards increased ovulation rate in FSH + LH treated ewes. These results show that early seasonally anestrous ewes can be successfully induced and synchronized for estrus with MAP pessaries and the number of ova recovered is increased with the inclusion of LH in the superovulation regime.  相似文献

The use of tailhead paste and chin-ball markers for estrus detection in beef cows following estrus synchronization was evaluated. The estrous cycles of 85 cows were synchronized with a prostaglandin analogue in a two-injection program. At the time of the second injection, each cow's tailhead was painted with an estrus detection paste and the cows were placed in pastures with four intact bulls with chin-ball markers. Blood samples for plasma progesterone assays were obtained at the time of each injection and daily for five days following the second injection. Based on progesterone values, tailhead paste was incorrect 27% of the time in indicating estrus and chin-ball markers were incorrect 24% of the time. Detecting particular cows in estrus in groups of synchronized cows is difficult even with estrus detection aids.  相似文献

The longer lactation period required in organic piglet producing herds reduces the potential number of produced litters per sow per year compared with that of conventional production. Induction and use of lactational estrus may be a way to increase the productivity in organic production. However, if lactational estrus is to be beneficial under practical husbandry conditions, it is crucial that the majority of sows are successfully mated within a few days to make batch farrowing procedures possible. The objective of this study was to investigate the occurrence and timing of lactational estrus in an organic outdoor system based on ad libitum feeding, individual housing until Day 35 in lactation, followed by grouping and introduction of a boar and weaning of piglets after 8 wk. Five groups with four sows ((Danish Yorkshire × Danish Landrace) × Danish Duroc) in each were observed, and rank was determined by a food competition test. All sows showed lactational estrus, and 84% of these sows showed estrus within 1 wk, on average 43.5 d and 7.3 d after farrowing and boar introduction, respectively. The number of days from boar introduction to estrus increased significantly with increasing feed competition rank (the lowest number being the top rank position). Eighty-four percent of all sows were diagnosed pregnant 5 wk after estrus. Behavioral observations revealed that the average total number of copulations per estrus sow was 2.3 with a range of 0 to 5 copulations. The findings of the current study indicate that it is possible to combine lactational estrus and batch farrowing procedures to increase the number of weaned piglets per year per sow in organic piglet production based on 8 wk of lactation or more.  相似文献

P-Chlorophenylalanine (p-ChPhe) was found to induce estrus in estrogen-primed ovariectomized C57BL/6J and DBA/2J mice but not in adrenalectomized females. α-Methyltyiosine had no such effects. It was concluded that serotonergic pathways are involved in reserpine- or p-ChPhe-induced estrus. Both p-ChPhe and reserpine were found to be ineffective in induction of estrus in hypophysectomized female mice. It appears that pituitary ACTH released by these drugs causes release of an adrenal hormone (probably progesterone) which is of functional significance in estrus.  相似文献

Reproductive performance in cows following synchronization of estrus with intravaginal progesterone releasing devices (IVD) has varied with the length of treatment, cyclic status and prolonged return to estrus intervals in some cows following first AI. The objective of this study was to compare two methods of synchronizing and resynchronizing estrus on the reproductive performance of lactating dairy cows. Cows were treated with an IVD (Day 0) for 7 days (n = 350) or 8 days (n = 350), cloprostenol (0.5 mg i.m.) at the time of device removal and estradiol benzoate (EB) at the time of device insertion (1.5mg i.m.), and again 9 days later (1.0 mg i.m.). Cows were also resynchronized starting on Days 23 and 46 by reinsertion of IVDs for either 7 or 8 days and treatment with EB (1mg i.m.) at the time of device insertion and again 9 days later. Cows were inseminated on detection of estrus for 4 days after removal of devices at each of the synchronized estrous cycles. No significant differences in reproductive performance were detected between each treatment throughout the study period. Synchrony of estrus was more precise at the first and second estrus after treatment with an IVD for 8 days compared to 7 days. Cows classified as anestrous had lower reproductive performance than cows classified as cycling and had longer intervals to estrus at the second (P < 0.001) and third estrus (P < 0.06), but not at the first estrus (P = 0.09). Mean time to onset of estrus after IVD removal was less in cows treated with an IVD for 8 days compared to 7 days at each synchronized estrus (P < 0.01). More Holstein-Friesian cows were classified as non-pregnant and not detected in estrus than crossbreed cows (15.7%, 54/343 versus 9.0%, 24/266; [P < 0.05). The results of the study suggested that the main effects of the treatments that were used to synchronize and resynchronize estrus were to alter the timing and synchrony of estrus without affecting fertility.  相似文献

Two sources of gonadotropin, combined with norgestomet implants and cloprostenol, were compared to induce estrus in 137 lactating dairy goats during the period of transition from anestrus to estrus. Does received either reagent-grade PMSG or commercially available hCG/PMSG product labeled for use in swine. Of the does treated with hCG/PMSG, 63 of 70 (90%) became pregnant compared with 50 of 66 (76%) does treated with reagent-grade PMSG. The pregnancy rates were significantly associated with the source of gonadotropin (P = 0.047). No difference in estrus response or litter size was detected; however, more does in the PMSG group appeared to have an extended interval from implant removal to breeding than did the hCG/PMSG-treated does. The results of this study indicate that the hCG/PMSG combination product is a satisfactory source of PMSG for extra-label use with norgestomet implants and cloprostenol to synchronize estrus in transitional period does.  相似文献

The impact of male presence or absence on the timing of the preovulatory LH surge and estrus was studied in 3 experimental groups (n = 6/group) of Eld's deer hinds pretreated with intravaginal progesterone-releasing devices (CIDR-type G) as follows: Group 1 = indirect male contact barn; Group 2 = direct male contact barn; and Group 3 = male isolation barn. For all hinds, the duration of the preovulatory LH surge averaged 2.5+/-0.5 h, whereas mean peak preovulatory and basal LH concentrations were 2.9+/-0.2 ng mL(-1) and 0.27+/-0.03 ng mL(-1), respectively. Nine of 12 male-exposed hinds exhibited a preovulatory LH surge within 24 to 32 h postCIDR device withdrawal, whereas 0 of 6 male-isolated hinds exhibited a preovulatory LH surge during the same time period. Onset of behavioral estrus (45.2+/-2.3, 52.7+/-5.7 and 66.3+/-1.8 h, respectively) was significantly advanced (P<0.05) after CIDR device withdrawal in male exposed hinds (Groups 1 and 2) compared with male isolated hinds (Group 3). These data suggest that stag exposure is important for modulating the timing of the preovulatory LH surge and behavioral estrus after synchronization of estrus with exogenous progestagens.  相似文献

The results of selection of mice for the selection index, with the use of gestagenic synchronization of oestrus for 12 generations, allows to affirm that: The synchronization effect decreases with successive generations, which is connected with the existence of a negative correlation between the synchronizability and fertility. The selection effect in the control population was higher than that in experimental one, subjected to oestrus synchronization. It can be supposed that gestagen dimishes the correlation between the genotype and the phenotype, thus decreasing the selection efficiency in the experimental population. A similar tendency was found for the number of implantations and litter size at the age of 1, 10 and 21 days after birth.  相似文献
