Salivary (s)-cortisol, s-amylase, s-DHEA are used extensively in stress research. Vasoactive Intestinal Peptide (VIP) is also detectable in saliva using a standard RIA kit. VIP is a 28 amino acid neuropeptide that belongs to the secretin/glucagon family of peptides and acts as a neurotransmitter/neuromodulator. VIP has also been detected in the parasympathetic nerves enervating the salivary glands. Here we measured the level of s-cortisol, s-DHEA, s-amylase and s-VIP in three different stress exercises of different duration and intensity. The results indicate that a brief intense exercise lasting minutes elicited a stress response with significant increases in s-cortisol, s-DHEA, s-amylase and s-VIP. A less rigorous exercise did not elicit a stress response with no significant increases in s-cortisol, s-DHEA, s-amylase and s-VIP. A longer intense exercise lasting hours elicited a stress response with significant increases only in s-cortisol. 相似文献

We examined the relations of 84 preschoolers' (43 boys; mean age = 54 months) situational stress reactivity to their observed emotions and mothers' reports of temperament and adjustment. Salivary cortisol and salivary alpha-amylase (sAA) were collected prior to, and following, a frustrating task. Children's anger, sadness, and positive affect were measured, and mothers reported on preschoolers' dispositional emotionality, regulation, impulsivity, and problem behaviors. Forty-seven percent of children had an increase in sAA and 52% had an increase in cortisol following the challenging task. On average, sAA levels showed the predicted pattern of rise following the frustrating task, followed by return to baseline. For cortisol, there was a mean increase from pre-task to 40 min post-test. sAA reactivity was associated with relatively low levels of dispositional anger and impulsivity and relatively high regulation, particularly for girls. sAA reactivity also was related to low externalizing problems for girls, but not boys. Although cortisol reactivity was unrelated to children's emotions and maladjustment, it was positively related to mothers' reports of regulation. The findings suggest that sAA reactivity in response to a frustrating social task may reflect girls' constrained behavior. 相似文献

Salivary alpha-amylase has been implemented as renewed non-invasive biomarker of interest to measure the levels of stress in humans. In the current clinical study, two-factor repeated-measures cross-sectional design was implemented to determine the salivary alpha-amylase levels and its rhythm characteristics in newly diagnosed female breast cancer in- and out-patients. Alpha-amylase levels were determined using the Salimetrics® salivary alpha-amylase assay kit. Rhythm characteristics of alpha-amylase were computed using the Cosinor Rhythmometry technique. Results showed that the levels of alpha-amylase significantly increased from/to the increasing number of chemotherapy cycle in patients of both group; however the levels were higher in in-patients. Further, results revealed significant differences in rhythm characteristics of alpha-amylase characterized by higher MESOR, lower amplitude, and advanced acrophase in in-patients as compared to out-patients. Current findings indicate that patients of both the groups were associated with progression under stress levels across the number of chemotherapy cycle; however, the levels of stress were higher in in-patients as compared to out-patients. Higher level of stress in cancer in-patients could be attributed to the possible effect of hospitalization and/or relative social isolation. 相似文献

Psychosocial stress has been shown to induce inflammatory reactions, followed by the release of immunosuppressive glucocorticoids. This may be mediated by catecholamines or other stress reactive substances such as neuropeptides or cytokines. We here set out to explore the effects of acute psychosocial stress on plasma levels of substance P (SP), a possible mediator of stress-induced inflammatory reactions, and interleukin-1 receptor antagonist (IL-1ra). Twelve healthy male subjects (mean age 27 yrs.) were subjected to the psychosocial stress test "Trier Social Stress Test" (TSST) and a resting control condition. Blood and saliva samples were taken before, as well as 1, 20, 45, and 90 min after TSST or rest, respectively. Salivary cortisol and plasma SP and IL-1ra were measured using immunoassays, salivary alpha-amylase (sAA) was measured by an enzyme kinetic method, and plasma epinephrine (E) and norepinephrine (NE) were measured by HPLC. The TSST induced immediate increases of E, NE, and sAA, and a delayed increase of free cortisol. Plasma IL-1ra showed an even further delayed peak at 90 min after stress. Plasma levels of SP did not respond to stress. No significant associations between changes of stress hormones and IL-1ra or SP were found. We conclude that substance P, epinephrine, and norepinephrine are probably not involved in mediating peripheral inflammation following psychosocial stress, at least with respect to IL-1ra. Further studies have to reveal the mechanisms involved in the stress-induced up regulation of IL-1ra. 相似文献

The maturation of the hypothalamo-pituitary-adrenal (HPA) axis is a key-component of the changes that occur during adolescence. In guinea pigs, HPA responsiveness during late adolescence depends strongly on the quantity and quality of social interactions: Males that lived in a large mixed-sex colony over the course of adolescence exhibit a lower stress response than males that were kept in pairs (one male/one female). Since colony-housed males have higher testosterone (T) levels than pair-housed males, and inhibiting effects of T on HPA function are well known, we tested the hypothesis that the decrease in stress responsiveness found in colony-housed males is due to their high T concentrations. We manipulated T levels in two experiments: 1) gonadectomy/sham-gonadectomy of colony-housed males (which usually have high T levels), 2) application of T undecanoate/vehicle to pair-housed males (which usually have low T levels). As expected, gonadectomized males showed a significantly increased stress response in comparison with sham-gonadectomized males, and T-injected males had a significantly lower stress response than vehicle-injected males. Both experiments thus confirm an inhibiting effect of T on HPA responsiveness during adolescence, which can mediate the influence of social interactions. The reduction in stress responsiveness is hypothesized to have a biologically adaptive value: A sudden increase in glucocorticoid concentrations can enhance aggressive behavior. Thus, pair-housed males might be adapted to aggressively defend their female (‘resource defense strategy’), whereas colony-housed males display little aggressive behavior and are capable of integrating themselves into a colony (‘queuing strategy’). 相似文献

The salivary glands and salivary pumps were investigated by means of dissection and serial semithin sections in order to expose the anatomy and histology of Nymphalidae in relation to feeding ecology. The paired salivary glands are tubular, they begin in the head, and extend through the thorax into the abdomen. The epithelium is a unicellular layer consisting of a single cell type. Despite the uniform composition, each salivary gland can be divided into five anatomically and histologically distinct regions. The bulbous end region of the gland lies within the abdomen and is composed of highly prismatic glandular cells with large vacuoles in their cell bodies. The tubular secretion region extends into the thorax where it forms large loops running backward and forward. It is composed of glandular cells that lack large vacuoles. The salivary duct lies in the thorax and also shows a looped formation but is composed of flat epithelial cells. The salivary reservoir begins in the prothorax and reaches the head. Its cells are hemispherical and bulge out into the large lumen of the tube. In the head the outlet tube connects the left and right halves of the salivary gland, and its epithelial cells are flat. The salivary pump lies in the head ventral to the sucking pump and leads directly into the food canal of the proboscis. It is not part of the salivary gland but is derived from the salivarium. Both the thin cuticle of the roof of the salivary pump and the thick bottom are ventrally arched. Paired muscles extend from the hypopharyngeal ridges and obviously serve as dilators for the pump. A functional interpretation of the salivary pump suggests that when not in use, the dilators are not contracted and the pump is tightly closed due to its own elasticity. When the dilator muscles repeatedly contract, the saliva is forced forward into the food canal of the proboscis. The salivary gland anatomy was found to be similar to other Lepidoptera. Furthermore, the histology of the salivary glands is identical in all examined butterflies, even in the species which exhibit specialized pollen-feeding behavior. 相似文献

In two studies, one with an all-male German sample and the other with a mixed-sex U.S. sample, subjects competed in pairs on reaction time-based cognitive tasks. Participants were not aware that contest outcome was experimentally varied. In both studies, implicit power motivation, defined as the non-conscious need to dominate or have impact on others, predicted changes in salivary cortisol from before to after the contest. Increased cortisol post-contest was associated with high levels of power motivation among losers but with low levels of power motivation among winners, suggesting that a dominance success is stressful for low-power individuals, whereas a social defeat is stressful for high-power individuals. These results emerged only in participants tested in the afternoon, possibly because of greater variability in cortisol in the morning due to the rapid decline after the morning peak. These studies add to the evidence that individual differences greatly influence whether a social stressor like losing a contest activates the HPA axis in humans. 相似文献

This study examines the family environments and hormone profiles of 316 individuals aged 2 months-58 years residing in a rural
village on the east coast of Dominica, a former British colony in the West Indies. Fieldwork was conducted over an eight-year
period (1988–1995). Research methods and techniques include radioimmunoassay of cortisol and testosterone from saliva samples
(N=22,340), residence histories, behavioral observations of family interactions, extensive ethnographic interview and participant
observation, psychological questionnaires, and medical examinations.
Analyses of data indicate complex, sex-specific effects of family environment on endocrine function. Male endocrine profiles
exhibit greater sensitivity to presence of father than do female endocrine profiles. Father-absent males tend to have (a) low cortisol levels during infancy, (b) high or abnormal cortisol profiles during childhood and adolescence, and (c) high cortisol and low testosterone levels during adulthood compared with those of males raised with a resident father. These
results indicate that early family environment has significant effects on endocrine response throughout male life histories.

Fluctuating asymmetry (FA) is small deviations from perfect symmetry in normally bilaterally symmetrical traits. We examined
the relationship between FA of five body traits (ear height, length of three digits, and ankle circumference) and self-reported
scores of physical and verbal aggression in a sample of 90 boys aged 10 to 15 years. The relationships between FA and scores
of aggression (particularly physical aggression) were found to be negative; in other words, the most symmetrical boys showed
highest aggression. One trait (ankle circumference) showed the characteristics of “ideal” FA—parametric mean of zero and a
normal distribution. Mean asymmetries calculated from six repeated measures of ankle FA in 30 subjects taken over a period
of five months showed strong negative associations with scores of physical aggression which were independent of age, height,
and weight.
It is argued that soft tissue “cyclical” FA (as opposed to “fixed” bony FA) is dependent on the secretion of hormones: for
example, cortisol. Causal associations between behavioral traits such as aggresion and hormones will lead to similar correlations
between FA and behavior.

The development of antisocial behavior in youth has been examined with neurobiological theories that suggest that adolescents who are less responsive to their environments are less likely to develop empathy in the absence of extant physiological arousal. However, little attention is paid to these individuals' social context. Individuals with adverse early experiences can also exhibit attenuated physiological arousal. The current investigation examines whether psychopathic symptoms or life stress exposure is associated with cortisol and its diurnal rhythm within 50 incarcerated adolescent boys (14–18 years old). Ten saliva cortisol samples were collected 1–2 weeks after admission to a maximum-security juvenile facility. Hierarchical Linear Modeling distinguished waking cortisol levels, the awakening response (CAR) and the diurnal rhythm. Multiple interviews and self-report measures of CU traits and stressor exposure were collected. Boys with higher levels of CU traits or greater life stress exposure had flat diurnal rhythms and a steeper awakening response in analyses with lifetime stress exposure specifically. Nonetheless, boys who were elevated on both CU traits and prior stress exposure had steeper diurnal rhythms. These results extend neurobiological theories of cortisol and illustrate that boys with the combination of severe stress with CU traits have a unique physiological profile. 相似文献

Lack of social support and social exclusion is associated with adverse effects for mental and physical health. Additionally, women appear to be more vulnerable to social triggers of health disturbances. The hypothalamus–pituitary–adrenocortical-axis (HPA axis) might play a key role in this context as it has been shown both to relate to psychosocial conditions and health outcomes and to respond differentially depending on gender. In a previous experiment we found no effects of exclusion alone (operationalized via Cyberball) on cortisol secretion. Here we examine the effects of a social exclusion pre-experience on psychological and cortisol responses to a public speaking stressor. Subjects (33 m, 34 f) were randomly assigned to social exclusion (SE) or one of two control conditions (exclusion attributed to technical default (TD) and social inclusion (SI)). Afterwards salivary cortisol and psychological responses to a public speaking paradigm were assessed. Exclusion pre-treatment does not affect psychological responses to public speaking stress though with respect to cortisol significant. Cyberball by gender and Cyberball by gender by time interactions are found. SE-women show a blunted cortisol stress response to public speaking while cortisol responses of SE-men fall between SI-men and TD-men. Pre-experience of social exclusion leads to a blunted cortisol response to stress in women but not in men. This factor might contribute to the higher vulnerability to social triggers of health disturbances observed in women. 相似文献

Dermatoglyphic prints and salivary samples were taken on a sample of 39 adult males. A statistical relationship between dermatoglyphic asymmetry and adult testosterone levels as measured in saliva was examined for seven dermatoglyphic variables by means of correlation, regression, and analysis of covariance, controlling for age and stature when necessary. The first two types of analyses indicated a significant effect of testosterone level upon the asymmetry of three dermatoglyphic variables: a-b ridge count, palmar pattern intensity, and the combined pattern intensity of palm and digits. Analysis of covariance, which examined the effect of testosterone level as a categorical variable, while holding age or stature constant, demonstrated the asymmetry of five variables to be significantly affected by testosterone level: radial digital count, digital pattern intensity, palmar pattern intensity, total digital ridge count, and the combined palmar and digital intensity. Although there is as yet only associational evidence linking levels of prenatal and secondary testosterone, the results of the present study lend support to the hypothesis that prenatal testosterone levels may have a significant effect on the development of dermatoglyphics. © 1993 Wiley-Liss, Inc. 相似文献

The aim of this study was to evaluate differences of cAMP-PDE activity in human salivary glands, between a control group and some different benign tumours groups and, where present, with 2 malignant tumors groups. The value of the enzymatic activity in the groups analysed was 50% lower than control samples. The differences between the control group (82 ± 7.9 nmols/mg of protein) and the 3 pathologic groups (Benign A: 44 ± 6.2; Malignant A: 40 ± 16; Benign B: 40 ± 14.2; Malign A: 9.1; Benign C: 22 nmols/mg of protein) are statistically significant. 相似文献

The circadian variation in salivary Cortisol concentration was investigated in four men during a traverse on foot, of the Greenland ice cap and of salivary Cortisol and testosterone on a summer expedition to Spitzbergen. In both instances circadian rhythmicity was demonstrated both before leaving the U.K. and throughout the expedition. The acrophase of the rhythms followed the changes in activity patterns on both expeditions although there was a dissociation between the Cortisol and testosterone following an acute 8 hr phase shift in Spitzbergen. The mesor of Cortisol in both instances tended to be higher than in the U.K. but the difference was never statistically significant. This work demonstrates the feasibility of using salivary steroid concentrations to investigate their circadian rhythmicity in circumstances where frequent blood sampling would be precluded. 相似文献

Experiments were performed with the perfused rat submandibular gland in vitro to investigate the nature of the coupling between
transported salt and water by varying the osmolarity of the source bath and observing the changes in secretory volume flow.
Glands were submitted to hypertonic step changes by changing the saline perfusate to one containing different levels of sucrose.
The flow rate responded by falling to a lower value, establishing a new steady-state flow. The rate changes did not correspond
to those expected from a system in which fluid production is due to simple osmotic equilibration, but were much larger. The
changes were fitted to a model in which fluid production is largely paracellular, the rate of which is controlled by an osmosensor
system in the basal membrane. The same experiments were done with glands from rats that had been bred to have very low levels
of AQP5 (the principal aquaporin of the salivary acinar cell) in which little AQP5 is expressed at the basal membrane. In
these rats, salivary secretion rates after hypertonic challenges were small and best modelled by simple osmotic equilibration.
In rats which had intermediate AQP5 levels the changes in flow rate were similar to those of normal rats although their AQP5
levels were reduced.
Finally, perfused normal glands were subject to retrograde ductal injection of salines containing different levels of Hg2+ ions (0, 10 and 100 μM) which would act as inhibitors of AQP5 at the apical acinar membrane. The overall flow rates were progressively diminished
with rising Hg2+ concentration, but after hypertonic challenge the changes in flow rates were unchanged and similar to those of normal rats.
All these results are difficult to explain by a cellular osmotic model but can be explained by a model in which paracellular
flow is controlled by an osmosensor (presumably AQP5) present on the basal membrane. 相似文献

BACKGROUNDMethylmercury (MeHg) is still considered a global pollutant of major concern; thus, it becomes relevant to investigate and validate alternative diagnostic methods to track early-life human exposure. This study aimed to evaluate the salivary parameters and to characterize potential mechanisms of oxidative damage on the salivary glands (SG) of offspring rats after pre- and postnatal environmental-experimental MeHg exposure.METHODSPregnant Wistar rats were daily exposed to 40 μg/kg MeHg during both gestational and lactation periods. Then, the saliva of offspring rats was analyzed in terms of flow rate, amylase activity, and total protein concentration. The SG of the offspring rats were dissected to perform the oxidative biochemistry analyses of antioxidant capacity against peroxyl radicals (ACAP), lipid peroxidation (LPO), and nitrite levels.RESULTSExposure to MeHg significantly decreased the ACAP, increased LPO and nitrite levels, decreased salivary flow rate, amylase activity, and total protein concentration.CONCLUSIONSaliva analyses can predict damages induced by early-life MeHg exposure and may be used as an auxiliary diagnostic method. 相似文献

In male golden hamsters, agonistic behavior matures during puberty, changing from play fighting to adult-like aggression. In addition, this transition is accelerated by repeated social subjugation early in puberty. However, little is known about the development of agonistic behavior in females. In the present study, we compared the development of agonistic behavior in male and female golden hamsters. Furthermore, we also tested the effects of repeated social subjugation on the development of agonistic behavior during puberty. Hamsters were tested for agonistic behavior in the presence of a smaller intruder at different intervals during puberty. Several observations were made. First, the frequency of attacks remained stable in females, while varying in males. Second, the transition from play fighting to adult-like aggression occurred at earlier time periods in females than in males. Finally, a clear transitional period marked by attacks focused on the flanks was observable in males around mid-puberty. However, this transitional period was not apparent in females. In addition, juvenile females were exposed to aggressive adult males or females. In both cases, repeated exposure to stress had no statistically significant effect on the development of agonistic behavior. After 2 weeks of subjugation, exposure to aggressive adults had no effect on serum cortisol levels, indicating that juvenile females habituate to repeated social stress. These data show significant sex differences in the development of agonistic behavior and adaptation to repeated stress in juvenile golden hamsters. 相似文献

Glucocorticoids (GCs) have been related to social rank in many studies across species, a particular rank giving rise to a particular stress-related physiological profile. Our aim was to examine the hypothesis that GCs levels in toddlers would be related to social dominance in a competitive resource situation. Subjects were 376 toddlers from the Quebec Newborn Twin Study. At 19 months of age, each subject was exposed to 2 unfamiliar situations known to be moderately stressful at that age. Saliva was collected before and after the unfamiliar situations, to assess pre-test and reactive cortisol. Then the toddler reaction to a competitive situation for a toy with an unfamiliar peer was assessed and we measured the proportion of time the child controlled the resource. In girls, no association between cortisol levels and the proportion of time the child got the toy was found. On the other hand, in boys, increased cortisol levels before the unfamiliar situation were significantly related to a decreased proportion of time they got the toy in the competitive situation (r174 = − 0.17, P = 0.02). These results show that even in toddlers with limited social experience, association between GCs levels and social dominance can be found, an association that is specific to boys. 相似文献
