We obtained two different clones (16 kb and 13 kb) of B. mori rDNA with intron sequence within the 28S-rRNA coding region. The sequence surrounding the intron was found to be highly conserved as indicated in several eukaryotes (Tetrahymena, Drosophila and Xenopus). The 28S rRNA-coding sequence of 16 kb and 13 kb clone was interrupted at precisely the same sites as those where the D. melanogaster rDNA interrupted by the type I and type II intron, respectively. The intron sequences of B. mori were different from those of D. melanogaster. In 16 kb clone, the intron was flanked by 14 bp duplication of the junction sequence, which was also present once within the 28S rRNA-coding region of rDNA without intron. This 14 bp sequence was identical with those surrounding the introns of Dipteran rDNAs. 相似文献

Crossing over is absent in oocytes of the silkworm, Bombyx mori. Synaptonemal complexes are present during pachytene between the paired chromosomes. At leptotene, lateral components of the synaptonemal complex are attached in a bouquet to a limited region of the nuclear envelope. Before completion of lateral components, synaptonemal complex formation begins at the nuclear envelope. With synaptonemal complex formation proceeding from both ends bivalents occasionally become interlocked. After pairing is completed, the bouquet arrangement is dissolved possibly as a result of a flow of the inner membrane of the nuclear envelope thereby separating the telomeres. After the telomeres are released from the nuclear envelope, material is deposited onto the lateral components of the synaptonemal complex. The modified synaptonemal complexes are retained by the bivalents until metaphase I. It is suggested that these modified synaptonemal complexes substitute for chiasmata in order to ensure regular disjunction of homologous chromosomes in the absence of crossing over. 相似文献

The aim of this study was to understand the structure and biodegradation relationships of silk particles intended for targeted biomedical applications. Such a study is also useful in understanding structural remodelling of silk debris that may be generated from silk-based implants. Ultrafine silk particles were prepared using a combination of efficient wet-milling and spray-drying processes with no addition of chemicals other than those used in degumming. Milling reduced the intermolecular stacking forces within the β-sheet crystallites without changing the intramolecular binding energy. Because of the rough morphology and the ultrafine size of the particles, degradation of silk particles by protease XIV was increased by about 3-fold compared to silk fibers. Upon biodegradation, the thermal degradation temperature of silk increased, which was attributed to the formation of tight aggregates by the hydrolyzed residual macromolecules. A model of the biodegradation mechanism of silk particles was developed based on the experimental data. The model explains the process of disintegration of β-sheets, supported by quantitative secondary structural analysis and microscopic images. 相似文献

Bombyx mori densovirus 1 (BmDNV-1), a major pathogen of silkworms, causes significant losses to the silk industry. The structure of the recombinant BmDNV-1 virus-like particle has been determined at 3.1-? resolution using X-ray crystallography. It is the first near-atomic-resolution structure of a virus-like particle within the genus Iteravirus. The particles consist of 60 copies of the 55-kDa VP3 coat protein. The capsid protein has a β-barrel "jelly roll" fold similar to that found in many diverse icosahedral viruses, including archaeal, bacterial, plant, and animal viruses, as well as other parvoviruses. Most of the surface loops have little structural resemblance to other known parvovirus capsid proteins. In contrast to vertebrate parvoviruses, the N-terminal β-strand of BmDNV-1 VP3 is positioned relative to the neighboring 2-fold related subunit in a "domain-swapped" conformation, similar to findings for other invertebrate parvoviruses, suggesting domain swapping is an evolutionarily conserved structural feature of the Densovirinae. 相似文献

[目的]Fox家族蛋白是调控动物发育的关键转录因子.本文分析了鳞翅目昆虫家蚕Bombyxmori的BmFoxG亚家族蛋白在精巢发育中的作用,为基于生殖系统的家蚕性状优化或害虫不育技术的开发提供理论依据.[方法]利用PCR克隆家蚕BmFoxG-1 和 BmFoxG-2基因;通过生物信息学工具对BmFoxG蛋白的结构及理化性质进行分析;采用qPCR技术检测BmFoxG基因在家蚕不同发育阶段的精巢中的表达变化;在家蚕Bm12细胞株中过表达BmFoxG-1,并通过qRT-PCR分析BmFoxG-1调控的靶基因.[结果]本文克隆获得了 BmFoxG-1(933 bp)和 BmFoxG-2(702 bp)两个基因.BmFoxG-1 和 BmFoxG-2蛋白均包含保守的Forkhead结构域,但BmFoxG-1蛋白在C端多出约60个氨基酸.BmFoxG-2基因在家蚕不同发育时期的精巢中的表达量均较低;BmFoxG-1 的表达均显着高于BmFoxG-2,且随着发育时期而变化,暗示BmFoxG-1参与精巢的发育.生殖细胞发育基因BmVasa、BmCyclinA等多个细胞周期基因以及精子鞭毛发育相关基因BMSK0009828在不同发育阶段的精巢中的表达趋势与BmFoxG-1类似,且BmFoxG-1在家蚕细胞中的过表达可以显著上调BmVasa、BmCyclinA和BMSK0009828等精巢发育相关基因的表达.[结论]我们推测BmFoxG-1蛋白可能通过调节精巢细胞周期或生殖细胞的功能来调节家蚕精巢的发育. 相似文献
