The two-competing-sites model for peptidoglycan assembly for bacterial cell shape regulation suggests that in rods, bacterial cell shape depends on the balance between two reactions (sites), one responsible for lateral wall elongation and the other responsible for septum formation. The two reactions compete with each other so that no lateral wall can be formed during septum formation and vice versa. When the site for lateral wall elongation overcomes that for septum formation, long rods or filaments are formed and cell division may be blocked. When the reaction leading to septum formation is hyperactive compared with the other, coccobacilli or cocci are formed. Other bacteria carry only one site for peptidoglycan assembly and can grow only as cocci. The two-competing-sites model predicts that two different types of cocci exist (among both morphology mutants and wild-type strains); one carries only the site for septum formation, whereas the other also carries the site for lateral wall elongation, the former site predominating over the latter. As a consequence of the inhibition (by antibiotics or by mutations) of septum formation in wild-type cocci of various species and in coccoid morphology mutants, some cocci are expected to undergo transition to rod shape and others are not. We have evaluated these predictions and show that they are in agreement. In fact, we found that among wild-type cocci belonging to 13 species, those of 6 species formed rods, whereas the remaining organisms maintained their coccal shape when septa were inhibited by antibiotics. Some coccoid morphology mutants of rod-shaped bacteria underwent coccus-to-rod transition after septum inhibition by antibiotics, whereas others maintained their coccal shape. When a mutation that causes septum inhibition was expressed in a morphology mutant of Klebsiella pneumoniae grown as a coccus, transition to rod shape was observed. A total of 914 mutants unable to form colonies at 42 degrees C were isolated from the coccoid species mentioned above. Between 75 and 95% of the mutants isolated from the species that formed rods when septum formation was inhibited by antibiotics but none of those isolated from the others underwent coccus-to-rod transition upon incubation at the nonpermissive temperature.  相似文献

Mir M7 is a spontaneous morphologically conditional mutant of Klebsiella pneumoniae which grows as round cells (cocci) at pH 7 and as normal rods at pH 5.8. We studied the rates of peptidoglycan synthesis of cocci and rods growing at pH values of 7 and 5.8, respectively. It was found that exponentially growing cocci produced a reduced amount of peptidoglycan per cell, compared with rods. Moreover, a shift of cocci to the permissive pH (5.8) caused an increase in the rate of peptidoglycan synthesis, whereas the reverse shift of rods to pH 7 determined a twofold reduction in the rate of [(3)H]diaminopimelic acid incorporation. During synchronous growth at pH 7, the rate of peptidoglycan synthesis after cell division decreased with time and rose before and during the first division. The susceptibilities of rods and cocci to beta-lactam antibiotics were also studied. It was found that cocci were more sensitive both to penicillin G and to cephalexin than were rods, but they showed a high level of resistance to mecillinam. The peculiar behavior of this mutant was interpreted as supporting the existence in bacterial rods of two different sites for peptidoglycan synthesis: one responsible for lateral wall elongation and one responsible for septum formation. In Mir M7, shape damage is described as dependent on the specific inhibition, at the nonpermissive pH, of the site for lateral wall extension.  相似文献

The study was undertaken to investigate the ability of enterococci to assimilate iron via siderophores of bacteria living in the same habitats in the human organism. The potential recipients of exogenous siderophores were six Enterococcus faecalis and six Enterococcus faecium strains, isolated from clinical materials of human origin. The donors of siderophores were Gram-negative rods (various species of the Enterobacteriaceae, Pseudomonas and Acinetobacter) and Gram-positive cocci (various species of Staphylococcus and Streptococcus). All of the investigated E. faecium and only two E. faecalis strains demonstrated the ability to utilize the siderophores of the aforementioned bacterial groups, predominantly the chelators of Gram-negative rods, those of Gram-positive cocci were utilized to a smaller extent. Four recipient strains from E. faecalis species did not demonstrate the ability to utilize siderophores synthesized by all of 40 investigated donor strains.  相似文献

Seven strains of Arthrobacter globiformis , and one each of A. crystallopoietes and A. nicotianae , isolated from the water fern Azolla, were cultured for 2–3 d at 30 °C on Pseudomonas Agar F to obtain rods and for 5 d to obtain cocci, then analysed for total cellular fatty acids. In rods, saturated iso - and anteiso -branched 13–19 carbon fatty acids averaged 85% of the total in A. globiformis , 81% in A. crystallopoietes , and 97% in A. nicotianae . Minor components included unsaturated branched chains, hydroxy and cyclopropane fatty acids. Fatty acid composition of A. globiformis was similar to that of A. crystallopoietes but different from that of A. nicotianae . Saturated straight chains in A. nicotianae averaged 1·5% of the total compared with 14–16% in A. globiformis/crystallopoietes , and anteiso -15:0 constituted 73% of the total in A. nicotianae compared with 18–19% in the other species. Composition of cocci was different from that of rods in A. globiformis and A. crystallopoietes. As cells changed from rods to cocci, percentages of saturated and unsaturated straight chains decreased and saturated iso - and anteiso -branched chains increased. In A. nicotianae there were no significant differences in the fatty acids between rods and cocci.  相似文献

Abstract Using high performance liquid chromatography and nuclear magnetic resonance techniques, the compatible solutes of some moderately halophilic bacteria were studied. The following accepted species of moderately halophilic bacteria were included: Volcaniella eurihalina and Deleya salina among Gram-negative rods, and Salinicoccus roseus and Salinicoccus hispanicus among Gram-positive cocci. Besides these strains we have also screened other new isolates, including Marinomonas species and Gram-positive cocci and rods. The tetrahydropyrimidine carboxylic acid 'ectoine' was found to be the main compatible solute in the Gram-negative strains tested when these were grown in glucose-mineral medium. In addition, betaine was accumulated from complex media containing yeast extract. Among the Gram-positive strains investigated, the solutes proline (bacillus 30, Salinicoccus ) and hydroxyectoine (coccus 28) also played an important role, while alanine, glucose, glutamate, glutamine and trehalose occurred as minor components. We also detected two recently described compatible solutes: Nδ -acetylornithine and a homologous compound, Nε -acetyllysine. Representatives of distinct phenotypic groups of Gram-positive cocci and rods were clearly distinguished by their solute pattern.  相似文献

Klebsiella pneumoniae Mir M7 is a spontaneous parentless morphology mutant which grows as cocci at pH 7 and as rods at pH 5.8. This strain has been characterized as defective in lateral wall formation (at pH7). Data suggest that the cell wall is mainly made up of poles of the rods (G. Satta, R. Fontana, P. Canepari, and G. Botta, J. Bacteriol. 137:727--734, 1979). In this work the isolation and the biochemical properties of the peptidoglycan of both Mir M7 rods and cocci and a nonconditional rod-shaped Mir M7 revertant (strain Mir A12) are described. The peptidoglycan of Mir M7 (both rods and cocci) and Mir A12 strains carried covalently bound proteins which could be easily removed by pronase treatment in Mir M7 rods and Mir A12 cells, but not in Mir M7 round cells. However, when the sodium dodecyl sulfate-insoluble residues of Mir M7 cocci were pretreated with ethylenediaminetetraacetic acid (EDTA), pronase digestion removed the covalently bound proteins, and pure peptidoglycan was obtained. EDTA treatment of the rigid layer of Mir M7 cocci removed amounts of Mg2+ and Ca2+, which were 10- and 50-fold higher, respectively, than the amount liberated from the rigid layer of Mir M7 rods and Mir A12 cells. Amino acid composition was qualitatively similar in both strains, but Mir M7 cocci contained a higher amount of alanine and glucosamine. Mir M7 cocci contained approximately 50% less peptidoglycan than rods. Under electron microscopy, the rigid layer of the Mir M7 rods and Mir A12 cells appeared to be rod-shaped and their shape remained unchanged after EDTA and pronase treatment. On the contrary, the Mir M7 cocci rigid layer appeared to be round, and after EDTA treatment it collapsed and lost any definite morphology. In spite of these alterations, the peptidoglycan of Mir M7 cocci still appeared able to determine the shape of the cell and protect it from osmotic shock and mechanical damages. The accumluation of divalent cations appeared necessary for the peptidoglycan to acquire sufficient rigidity for shape determination and cell protection. We concluded that the coccal shape in Mir M7 cells is not due to loss of cell wall rigidity but is a consequence of the formation of a round peptidoglycan molecule. The possibility that the alterations found in the Mir M7 cocci rigid layer may reflect natural differences in the biochemical composition of the septa and lateral wall of normally shaped bacteria is discussed.  相似文献

The study group were persons with risk factors of colonization by pathogenic strains and included smokers, patients suffering from paradontosis, and patients with visibly neglected oral cavity and teeth. We isolated and classified to the species or genera 488 microorganisms. Of posterior pharyngeal wall flora, 61% of were Gram-negative bacteria, represented predominantly by Haemophilus and anaerobic rods and aerobic cocci belonging to the Neisseria and Moraxella (Branhamella) genera. In the group of Gram-positive cocci (34% of the total number of microorganisms), oral streptococci, Stomatococcus mucilaginosus and Streptococcus pneumoniae were isolated most frequently. The affected by neoplastic lesions larynx, was colonized by similar bacterial groups. However, the incidence of Gram-positive cocci was higher. The main etiologic factor of purulent post-operative wound inflammations were methicillin-resistant Staphylococcus aureus strains (MRSA), which had been absent among the bacteria isolated from patients on admission.  相似文献

The pH-conditional morphology mutant of Klebsiella pneumoniae strain MirM7 grows as cocci at pH 7 and as rods at pH 5.8. The mutant has a high-level mecillinam resistance (50% lethal dose greater than 200 micrograms/ml) in both forms. When broth cultures of the rod-shaped mutant were grown with 0.7 microgram of mecillinam per ml, cells assumed a round shape and continued to divided at a higher rate than the untreated control. A MirM7 rod-shaped revertant (MirA12), when treated with the same antibiotic concentration, changed to coccal shape and stopped dividing. The penicillin-binding proteins (PBPs) of strains MirA12 and MirM7 were analyzed. K. pneumoniae had six major PBPs quite similar to those of Escherichia coli. No differences were seen in the PBPs of MirM7 cocci and rods and MirA12 cells. In particular, PBP2 was found to be present and similar in MirM7 rods and cocci and MirA12 cells. We suggest that that in gram-negative rods, a control mechanism exists which prevents further septation in the absence of lateral cell wall elongation. The unique behavior of MirM7 is due to the fact that the control mechanism is not active in this strain. This model allows us to explain the preservation of shape in bacterial rods under various conditions of growth and the mechanism of bacterial killing by mecillinam.  相似文献

As was found with the aid of capillary microscopy, the nodule bacteria of pea and lupine in soil are encountered as rods, bacteroids, and cocci. The rod-like cells form bacteroids not only in the nodules but also outside, in soil. The bacteroids are viable and later reproduce the coccoid cells (arthrospores). The rods and bacteroids prevail in soil during flowering of the legumes, and the cocci are predominant at the end of vegetation.  相似文献

Arthrobacter crystallopoietes growing exponentially as cocci were changed to rods by adding succinate to the medium. Cells were sampled before, during, and after this transition for Gram-staining and ultrastructural studies. Cells were Gram stained by the standardized method of Bartholomew, and all samples were fixed and prepared for thin sectioning in an identical manner. Cocci were gram positive, and thin sections demonstrated a gram-positive type of cell wall having an average thickness of 31 nm. Cells sampled during morphogenesis appeared as cocci with most having a single rodlike projection. The coccus portion of these transition cells was gram positive and bound by a gram-positive type of wall having an average thickness of 29 nm. The rodlike projection of the transition cells appeared to be gram negative; it was also surrounded by a gram-positive type of wall, but its average thickness was only 22 nm. Gram-negative rods of the type species, Arthrobacter globiformis, were also examined and found to produce a gram-positive type of wall with a 19-nm average thickness. Evidence for the trilaminar region, characteristic of most gram-negative bacterial cell walls, was totally lacking in both species. These results suggest that variations in cell wall thickness may be an important contributing factor to the variable Gram-staining characteristics of this genus.  相似文献

Short-term (65-h) bacterial colonization of 0.2-μm (pore size) filters submerged in water from Lake Charlotte, Nova Scotia, was characterized by a well-defined succession of cell types, in which small cocci gave way to larger, rod-shaped cells. This succession agrees with the concept of attachment as a strategy for survival, in which inactive cocci can attach to a surface and grow into larger, rod-shaped cells by using endogenous nutrients and the nutrients accumulated at the solid-liquid interface. Analyses of oxygen and CO2 microgradients above colonized surfaces indicated that a peak of respiration accompanied the succession of rods from cocci. CO2 fixation then became apparent as the rods began to bind manganese and iron to their surfaces. This means that survival by attachment may not be just the province of heterotrophs. It could also be a strategy adopted by metal-oxidizing chemoautotrophs. Long-term (34-day) colonization of similar filters indicated that, while a succession of attached cell types may indeed be a natural occurrence, other factors (such as the selective grazing of larger cells) tend to obscure the development of this succession.  相似文献

Superoxide dismutase (SOD) was present in 23 of 28 strains of the genus Bacteroides tested. Several clostridia, anaerobic cocci, and anaerobic, grampositive, nonsporing rods contained measureable SOD, but the frequency of SOD occurrence was much lower than in the bacteroides. These data indicate that there is a large variation in SOD levels between genera and among species within a genus of anaerobic bacteria. There was also no correlation between source of isolate, SOD levels, and presumed pathogenicity of the isolate.  相似文献

Calorific contents of dried cells of several representative species of bacteria (gram-negative rods and gram-positive rods and cocci), two species of yeasts, and a filamentous fungus were determined by bomb calorimetry. The grand mean was 5,383 cal per g of ash-free dry weight. This value was then used to determine quantity of energy assimilated (E(a)) during growth. Subsequently, E(a) was employed in the equation: Y(kcal) = Y/(E(a) + E(d)), where Y(kcal) is the yield of cells per kilocalorie of energy taken from a culture medium, Y is the yield per mole of substrate utilized, E(a) is Y times caloric content of the cells, and E(d) is the energy expended by oxidative dissimilation. An estimate of E(d) was obtained for a number of experiments by multiplying the moles of oxygen consumed per mole of substrate utilized during growth by the average quantity of energy utilized to reduce a mole of oxygen with electrons from organic compounds (106 kcal). From previous studies in our laboratories, a value for Y(kcal) of 0.118 g/kcal was predicted. The mean value for data from five studies of aerobic growth of prototrophic heterotrophs was found to be 0.111.  相似文献

Tonsilloliths are a potential cause of oral malodor. In this study, microbial profiles and composition of tonsilloliths were determined using culture-independent molecular methods and scanning electron microscopy. 16S ribosomal RNA bacterial genes (16S rDNAs) isolated from tonsilloliths of 6 individuals were amplified by PCR and cloned into Escherichia coli. Partial 16S rDNA sequences of approximately 600 bases of cloned inserts were used to determine species identity by comparison with sequences of known species. Characteristics of bacteria on the surface and inside the tonsillolith were analyzed using scanning electron microscopy. Anaerobic bacteria detected in tonsilloliths belonged to the genera Eubacterium, Fusobacterium, Megasphaera, Porphyromonas, Prevotella, Selenomonas and Tannerella, all of which appear to be associated with production of volatile sulfur compounds. Electron microscopy revealed cocci and rods on the surface and rods predominating inside the tonsilloliths. These results support the tonsillolith as an origin of oral malodor.  相似文献

Selective media were used to isolate a wide range of bacteria from sixty human dental plaques. Glucanase activities of the isolates were determined on dextran- and starch-containing media. All sixty samples of dental plaque yielded some colonies showing amylolytic and dextranolytic activities. The glucanase-producing organisms comprised 20% of the isolates. Of these 38% were Gram-positive rods, 27% Gram-positive cocci, 28% Gram-negative rods and 7% were Gram-negative cocci. The cultural groups most commonly represented among the glucanase-producing isolates were Actinomycetaceae, streptococci, haemophili and Gram-negative anaerobes. Species prominent among these isolates included Streptococcus sanguis, Streptococcus mitior, Actinomyces naeslundii, Actinomyces viscosus, Bacterionema matruchotii, Bifidobacterium sp. and Bacteroides sp. No isolates capable of degrading starch or dextran were identified as Streptococcus milleri, Rothia dentocariosa or Fusobacterium sp. This study has shown that a wide range of bacterial species commonly isolated from human dental plaques exhibit both amylolytic and dextranolytic activities. In order to understand glucan metabolism in human dental plaques further investigation of these catabolic activities is necessary.  相似文献

Summary The calculation based on the idea formulated as the Law of Diminishing Returns has here been applied to the development ofSc. thermophilus in milk-cultures during the phase of diminishing growth rate. The values calculated correspond fairly well with the data found in the experiment. An application of this method is given in the calculation of the relation between the acid production per cell (diplococci) per hour and the generation time. It is shown, that a quick multiplication corresponds to a high acid production and inversely a long generation time gives a small acid production. The relation between these quantities enables us to account for the rapid acidification in yoghurt. It also enables us to calculate the acid production in yoghurt by the cocci and the rods from the experimental data. By making this calculation we could prove that in the first stage the cocci are the acid producers. In the second stage the rods keep producing acid whereas the growth of cocci is checked by the lactic acid which has then for the greater part been produced by themselves. Susceptibility of intestinal bacteria to various antimicrobial agents in vitro, together with levels of those agents achieved in the gut, provides information on the likely impact of the agents on the intestinal flora. Orally administered drugs that are poorly absorbed may be useful for treatment of intestinal infections and for certain other situations in which intestinal bacteria may play a role. The antimicrobial activity of ramoplanin (MDL 62,198) against 928 strains of intestinal anaerobic bacteria was determined using the NCCLS-approved Wadsworth brucella laked-blood agar dilution method. The activity of ramoplanin was compared with that of ampicillin, bacitracin, metronidazole, trimethoprim/sulfamethoxazole (TMP/SMX), and vancomycin. The organisms tested included Bacteroides fragilis group (n=89), other Bacteroides species (n=16), other anaerobic Gram-negative rods (n=56) anaerobic cocci (n=114), Clostridium species (n=426), and non-sporeforming anaerobic Gram-positive rods (n=227). The overall MIC(90)s of ramoplanin, ampicillin, bacitracin, metronidazole, and vancomycin were 256, 32, 128, 16, and 128 mcg/ml, respectively. Ramoplanin was almost always highly active vs. Gram-positive organisms and relatively poor in activity against Gram-negative organisms, particularly Bacteroides, Bilophila, Prevotella, and Veillonella. Vancomycin was quite similar to ramoplanin in its activity. Ampicillin was relatively poor in activity vs. organisms that often produce beta-lactamase, including most of the Gram-negative rods as well as Clostridium bolteae and C. clostridioforme. Bacitracin was relatively poor in activity against most anaerobic Gram-negative rods, but better vs. most Gram-positive organisms. Metronidazole was very active against all groups other than bifidobacteria and some strains of other types of non-sporeforming Gram-positive bacilli. TMP/SMX was very poorly active, with an MIC(90) of >2048 mcg/ml.  相似文献

The distribution of culturable hyperthermophiles was studied in relation to environmental conditions in the Kubiki oil reservoir in Japan, where the temperature was between 50 and 58 degrees C. Dominant hyperthermophilic cocci and rods were isolated and shown to belong to the genera Thermococcus and Thermotoga, respectively, by 16S rDNA analyses. Using the most-probable-number method, we found that hyperthermophilic cocci were widely distributed in several unconnected fault blocks in the Kubiki oil reservoir. In 1996 to 1997, their populations in the production waters from oil wells were 9.2 x 10(3) to 4.6 x 10(4) cells/ml, or 10 to 42% of total cocci. On the other hand, hyperthermophilic rods were found in only one fault block of the reservoir with populations less than 10 cells/ml. Dominant Thermococcus and Thermotoga spp. grew at reservoir temperatures and utilized amino acids and sugars, respectively, as sole carbon sources. While organic carbon was plentiful in the environment, these hyperthermophiles were unable to grow in the formation water due to lack of essential nutrients. Concentrations of some organic and inorganic substances differed among fault blocks, indicating that the movement of formation water between fault blocks was restricted. This finding suggests that the supply of nutrients via fluid current is limited in this subterranean environment and that the organisms are starved in the oil reservoir. Under starved conditions at 50 degrees C, culturable cells of Thermococcus sp. remained around the initial cell density for about 200 days, while those of Thermotoga sp. decreased exponentially to 0. 01% of the initial cell density after incubation for the same period. The difference in survivability between these two hyperthermophiles seems to reflect their populations in the fault blocks. These results indicate that hyperthermophilic cocci and rods adapt to the subterranean environment of the Kubiki oil reservoir by developing an ability to survive under starved conditions.  相似文献
