Enzymatic syntheses of fatty acid anilides are important owing to their wide range of industrial applications in detergents, shampoo, cosmetics, and surfactant formulations. The amidation reaction of Mucor miehei lipase Lipozyme IM20 was investigated for direct amidation of triacylglycerol in organic solvents. The process parameters (reaction temperature, substrate molar ratio, enzyme amount) were optimized to achieve the highest yield of anilide. The maximum yield of palmitanilide (88.9%) was achieved after 24 h of reaction at 40 °C at an enzyme concentration of 1.4% (70 mg). Kinetics of lipase-catalyzed amidation of aniline with tripalmitin has been investigated. The reaction rate could be described in terms of the Michaelis–Menten equation with a Ping–Pong Bi–Bi mechanism and competitive inhibition by both the substrates. The kinetic constants were estimated by using non-linear regression method using enzyme kinetic modules. The enzyme operational stability study showed that Lipozyme IM20 retained 38.1% of the initial activity for the synthesis of palmitanilide (even after repeated use for 48 h). Palmitanilide, a fatty acid amide, exhibited potent antimicrobial activity toward Bacillus cereus.  相似文献

The enzyme reaction mechanism and kinetics for biosyntheses of deoxycytidine triphosphate (dCTP) and deoxythymidine triphosphate (dTTP) from the corresponding deoxycytidine diphosphate (dCDP) and deoxythymidine diphosphate (dTDP) catalyzed by pyruvate kinase were studied. The kinetic model for the two synthetic reactions was found to follow the Bi–Bi random rapid equilibrium mechanism similar to that of the biosynthesis of deoxyadenosine triphosphate (dATP) and deoxyguanosine triphosphate (dGTP) from the corresponding deoxyadenosine diphosphate (dADP) and deoxyguanosine diphosphate (dGDP). Kinetic constants involved in the reactions including the maximum reaction velocity, the Michaelis–Menten constants, and the inhibition constants for dCTP and dTTP biosyntheses were experimentally determined. This enzyme reaction requires Mg2+ ion and the optimal Mg2+ concentration was also determined. The experimental results showed a good agreement with the simulation results obtained from the kinetic model developed. The kinetics of the four biosynthetic reactions for deoxynucleoside triphosphates (dNTP) including dATP, dGTP, dCTP, and dTTP from the corresponding deoxynucleoside diphosphates (dNDP) including dADP, dGDP, dCDP, and dTDP were analyzed. The results suggest that the binding kinetics of phosphoenolpyruvate (PEP) and pyruvate are similar for all four biosynthetic reactions. The affinity of the dNDP substrates to enzyme is of the same order of magnitude as the corresponding dNTP as inhibitors. The order of reactivity and substrate specificity for dNDP is dADP > dGDP > dCDP > dTDP in the pyruvate kinase (PK) reactions. The results obtained from this study can be applied to bioreactor design and production of dCTP and dTTP for biosynthesis of DNA at a significantly lower cost compared to the currently available chemical method.  相似文献

Bio-desulphurization kinetics of dibenzothiophene (DBT) using Pseudomonas putida CECT 5279, a genetically modified micro-organism (GMO), is studied. A kinetic model describing the 4S route of DBT desulphurization is proposed. Bio-desulphurization experiments have been carried out using resting whole cells of P. putida CECT 5279 obtained at different growth times as biocatalysts. The kinetic equations proposed for each reaction have been previously checked by studying each reaction of the 4S route individually, employing different substrates in different experiments. Finally, simple Michaelis–Menten kinetic equations for the three first reactions catalyzed by two mono-oxygenases (DszC and DszA) and a kinetic equation taking into account competitive inhibition due to product for the final reaction catalyzed by a desulfinase (DszB) have been adopted. DBT has been desulphurized using cells obtained at different growth times (5, 10, 23, 30 and 45 h). The overall kinetic model proposed involving the four reactions of the 4S route was fitted to all the experimental data yielding a set of kinetic parameters able to describe the system evolution. Cell age has influence on the rates of all the reactions: reactions (1), (2) and (3) present maximum rates for cell grown during 30 h, while reaction (4) shows a maximum rate for cells with around 10 h of growth time. However, affinities of each substrate and the inhibition constant of the last reaction are not influenced by the time of growth.  相似文献

The enzymatic reaction of benzoic acid and glycerol in the absence of organic solvents to obtain the 1- or 3-monobenzoate glycerol (α-MBG) is studied. Esterification runs were batch wise performed with a concentration of enzyme of 30 g/L, changing the initial concentration of the acid in glycerol from 20 to 60 g/L, and temperature from 50 to 70 °C. In these conditions, the most active lipase among those tested for this synthesis was lipase B isozyme from Candida antarctica (CALB), obtaining conversion values higher than 80% and a significant selectivity to α-MBG. Unlike the synthesis in organic media, water did not have an inhibitory behaviour; hence control of water activity was unnecessary. Temperature and benzoic acid act synergically as deactivating factors. Considering the aforementioned, a kinetic model according to a Michaelis–Menten mechanism is proposed. This model considers a partial enzymatic deactivation mechanism with two terms, one of them accounting for the deactivating action of the acid.  相似文献

Fructosidases, in particular exo-β-fructosidases, may act on fructans such as inulins and levans of plant and bacterial origin to produce fructose. In this paper, the kinetic properties of a commercial preparation (Fructozyme L) and a recombinant exoinulinase (BfrA) from Thermotoga maritima, were studied using fructan polymer substrates from various sources. Both enzymatic preparations preferentially hydrolyzed β2-1 linkages and low molecular weight fructans. We show that chicory inulin is degraded most efficiently by both preparations, followed by bacterial inulin, in spite of its high molecular weight and branching in β2-6 positions. All bacterial levans were more slowly hydrolyzed. Michaelis–Menten kinetics describe the hydrolysis of sucrose and low molecular weight fructans (≤8.3 kDa) by both enzyme preparations, while first order kinetics were observed with respect to bacterial fructans due to the high molecular weight and, therefore, low molar concentrations. Comparison of second order rate constants indicates that bacterial inulin (Leuconostoc citreum CW28) is hydrolyzed more slowly with both enzyme preparations than chicory inulin by approximately one order of magnitude. For Leuconostoc mesenteroides NRRL B-512F levan, the second order rate constant for Fructozyme L is 200-fold lower than for chicory inulin. However, the second order rate constant for BfrA is only 22-fold lower than for chicory inulin. Taken together, our studies characterize the kinetics of fructan hydrolysis and also suggest that the kinetic parameters may be used to differentiate between fructan structures.  相似文献

Allicin, an extremely active constituent of freshly crushed garlic, is produced upon reaction of alliin with the enzyme alliinase (EC 4.4.1.4). A bacterium Cupriavidus necator with the ability of alliinase production was isolated from a soil sample and was identified by morphological, biochemical and 16S rRNA sequence. Alliinase production was optimised and it was further purified to apparent homogeneity with 103-fold purification and specific activity of 209 U/mg of protein by using DEAE Cellulose and Sephadex G-100 chromatography. The enzyme is a homodimer of molecular weight 110 kDa with two subunits of molecular weight 55 kDa each. The optimum activity of the purified enzyme was found at pH 7 and the optimum temperature was 35 °C. The enzyme exhibited maximum reaction rate (Vmax) at 74.65 U/mg and Michaelis–Menten constant (Km) was determined to be 0.83 mM when alliin was used as a substrate. The cytotoxic activity of in-situ generated allicin using purified alliinase and alliin was assessed on MIA PaCa-2 cell line using MTT assay and Acridine orange–ethidium bromide staining. This approach of in-situ allicin generation suggests a novel therapeutic strategy wherein alliin and alliinase work together synergistically to produce cytotoxic agent allicin.  相似文献

This study illustrates the benefits of Celite® supported lipase sol–gels for the transesterification of triolein to produce methyl oleate. A ping–pong bi–bi kinetic model was developed and validated taking into account the inhibition effects of methanol and glycerol as well as the effect of temperature. Although initial reaction rate models are useful for predicting the kinetics in the absence of products, a kinetic model beyond the initial conditions that considers glycerol inhibition is important. The model developed was consistent with the experimental data (R2 = 0.95) predicting an increase in methyl oleate production with increasing methanol concentration up to an optimal range of 1.3 M to 2.0 M depending on the temperature. In general, increasing the temperature increased the initial reaction rate for the immobilized lipase over the temperature range of 40–60 °C. Based on the kinetic constants, the maximum velocity of the reverse reaction is about 25% slower than that of the forward reaction and glycerol inhibition has a more significant effect on the reaction kinetics than methanol inhibition. The model developed would be useful for understanding the effects of methanol and glycerol inhibition as well as temperature on the production of methyl oleate using lipase-mediated enzymatic transesterification.  相似文献

The ring-opening polymerization of ?-caprolactone catalyzed by a novel thermophilic esterase from the archaeon Archaeoglobus fulgidus was successfully conducted in organic solvents. The effects of enzyme concentration, temperature, reaction time, reaction medium, and water activity on monomer conversion and product molecular weight were investigated. Poly(?-caprolactone) was obtained in almost 100% of the monomer conversion, with a number-average molecular weight of 1400 in toluene at 80 °C for 72 h. Furthermore, the Michaelis–Menten kinetic analysis showed that the enzyme had the highest affinity for ?-caprolactone, with a Km value of 0.093 mol/l compared with other reported lipases. The possible structural and energetic effects of the enzyme on the Km value were investigated, using molecular docking studies.  相似文献

The kinetics of catalase inactivation in buffer-free aqueous solutions within the temperature range 30–60 °C in the absence or presence of hydrogen peroxide was investigated and discussed taking into account the effect of NaCl, ethane-1,2-diol, propane-1,2,3-triol and sucrose, additives expected to increase the enzyme thermostability. Using the kinetic extended curves obtained by measuring the substrate absorbance in time and an isoconversional method, several simple kinetic models most frequently encountered in literature were fitted to the experimental data. The best model for inactivation was chosen on the basis of several statistical criteria. The half-times of inactivation and the activation energies were also reported and discussed.  相似文献

Lipase from Candida rugosa was immobilized on a polyvinylidene fluoride membrane for synthesis of rose flavor ester, 2-phenylethyl acetate. Response surface methodology (RSM) was employed for kinetic modeling of process and prediction the yield. The RSM was used in practice for determining the kinetic models by fitting the initial rate dates based on the equations of ping-pong bi–bi and order bi–bi model. The maximum reaction rate and kinetic constants were matched with the order bi–bi model. The specificity constant of the immobilized lipase was 10-folds higher than the free form indicated the enzyme–substrate affinity, and catalytic ability was enhanced after immobilization. Moreover, the effects of reaction parameters on the yield were evaluated by RSM using a Box–Behnken experimental design. Based on a ridge max analysis, the maximum conversion was 95.33 ± 2.57% at 38.78 h, 35.85 °C, and substrate mole ratio of 3.65:1. Furthermore, the order bi–bi kinetic model was simulated successfully in a batch reaction. A good prediction existed between the RSM results and integrated equation was found.  相似文献

The gene coding for the aminoglycoside adenylyltransferase (aadA6) from a clinical isolate of Pseudomonas aeruginosa was cloned and expressed in Escherichia coli strain BL21(DE3)pLysS. The overexpressed enzyme (AadA6, 281 amino-acid residues) and a carboxy-terminal truncated variant molecule ([1-264]AadA6) were purified to near homogeneity and characterized. Light scattering experiments conducted under low ionic strength supported equilibrium between monomeric and homodimeric arrangements of the enzyme subunits. Circular Dichroism spectropolarimetry indicated a close structural relation to adenylate kinases. Both forms modified covalently the aminoglycosides streptomycin and spectinomycin. The enzyme required at least 5 mM MgCl2 for normal Michaelis–Menten kinetics. Streptomycin exhibited a strong substrate inhibition effect at 1 mM MgCl2. The truncated 17 residues at the C-terminus have little influence on protein folding, whereas they have a positive effect on the enzymic activity and stabilize dimers at high protein concentrations (>100 μM). Homology modelling and docking based on known crystal structures yielded models of the central ternary complex of monomeric AadA6 with ATP and streptomycin or spectinomycin.  相似文献

Pseudomonas luteola was immobilized by entrapment in alginate–silicate sol–gel beads for decolorization of the azo dye, Reactive Red 22. The influences of biomass loading and operating conditions on specific decolorization rate and dye removal efficiency were studied in details. The immobilized cells were found to be less sensitive to changes in agitation rates (dissolved oxygen levels) and pH values. Michaelis–Menten kinetics could be used to describe the decolorization kinetics with the kinetic parameters being 36.5 mg g−1 h−1, 300.1 mg l−1 and 18.2 mg g−1 h−1, 449.8 mg l−1 for free and immobilized cells, respectively. After five repeated batch cycles, the decolorization rate of the free cells decreased by nearly 54%, while immobilized cells still retained 82% of their original activity. The immobilized cells exhibited better thermal stability during storage and reaction when compared with free cells. From SEM observation, a dense silicate gel layer was found to surround the macroporous alginate–silicate core, which resulted in much improved mechanical stability over that of alginate beads when tested under shaking conditions. Alginate–silicate matrices appeared to be the best matrix for immobilization of P. luteola in decolorization of Reactive Red 22 when compared with previous results using synthetic or natural polymer matrices.  相似文献

As a case study, we consider a coupled (or auxiliary) enzyme assay of two reactions obeying the Michaelis–Menten mechanism. The coupled reaction consists of a single-substrate, single-enzyme non-observable reaction followed by another single-substrate, single-enzyme observable reaction (indicator reaction). In this assay, the product of the non-observable reaction is the substrate of the indicator reaction. A mathematical analysis of the reaction kinetics is performed, and it is found that after an initial fast transient, the coupled reaction is described by a pair of interacting Michaelis–Menten equations. Moreover, we show that when the indicator reaction is fast, the quasi-steady-state dynamics are governed by three fast variables and one slow variable. Timescales that approximate the respective lengths of the indicator and non-observable reactions, as well as conditions for the validity of the Michaelis–Menten equations, are derived. The theory can be extended to deal with more complex sequences of enzyme-catalyzed reactions.  相似文献

The hydrolysis of substrates by cholinesterases does not follow the Michaelis–Menten reaction mechanism. The well-known inhibition by excess substrate is often accompanied by an unexpectedly high activity at low substrate concentrations. It appears that these peculiarities are the consequence of an unusual architecture of the active site, which conducts the substrate molecule over many stages before it is cleaved and released. Structural and kinetic data also suggest that two substrate molecules can attach at the same time to the free, as well as to the acetylated, enzyme. We present a procedure which provides an unbiased framework for mathematical modelling of such complex reaction mechanisms. It is based on regression analysis of a rational polynomial using classical initial rate data. The determination of polynomial degree reveals the number of independent parameters that can be evaluated from the available information. Once determined, these parameters can substantially facilitate the construction and evaluation of a kinetic model reflecting the expected molecular events in an enzymic reaction. We also present practical suggestions for testing the postulated kinetic model, using an original thermodynamic approach and an isolated effect in a specifically mutated enzyme.  相似文献

Leifsonia xyli HS0904 can stereoselectively catalyze the bioreduction of 3,5-bis(trifluoromethyl) acetophenone (BTAP) to its corresponding alcohol, which is a valuable chiral intermediate in the pharmaceuticals. In this study, a new carbonyl reductase derived from L. xyli HS0904 was purified and its biochemical properties were determined in detail. The carbonyl reductase was purified by 530-fold with a specific activity of 13.2 U mg−1 and found to be a homodimer with a molecular mass of 49 kDa, in which the subunit molecular-weight was about 24 kDa. The purified enzyme exhibited a maximum enzyme activity at 34 °C and pH 7.2, and retained over 90% of its initial activity at 4 °C and pH 7.0 for 24 h. The addition of various additives, such as Ca2+, Mg2+, Mn2+, l-cysteine, l-glutathione, urea, PEG 1000 and PEG 4000, could enhance the enzyme activity. The maximal reaction rate (Vmax) and apparent Michaelis–Menten constant (Km) of the purified carbonyl reductase for BTAP and NADH were confirmed as 33.9 U mg−1, 0.383 mM and 69.9 U mg−1, 0.412 mM, respectively. Furthermore, this enzyme was found to have a broad spectrum of substrate specificity and can asymmetrically catalyze the reduction of a variety of ketones and keto esters.  相似文献

In this study, polyurethane foam (PUF) was used for immobilization of Yarrowia lipolytica lipase Lip2 via polyethyleneimine (PEI) coating and glutaraldehyde (GA) coupling. The activity of immobilized lipases was found to depend upon the size of the PEI polymers and the way of GA treatment, with best results obtained for covalent-bind enzyme on glutaraldehyde activated PEI-PUF (MW 70,000 Da), which was 1.7 time greater activity compared to the same enzyme immobilized without PEI and GA. Kinetic analysis shows the hydrolytic activity of both free and immobilized lipases on triolein substrate can be described by Michaelis–Menten model. The Km for the immobilized and free lipases on PEI-coated PUF was 58.9 and 9.73 mM, respectively. The Vmax values of free and immobilized enzymes on PEI-coated PUF were calculated as 102 and 48.6 U/mg enzyme, respectively. Thermal stability for the immobilization preparations was enhanced compared with that for free preparations. At 50 °C, the free enzyme lost most of its initial activity after a 30 min of heat treatment, while the immobilized enzymes showed significant resistance to thermal inactivation (retaining about 70% of its initial activity). Finally, the immobilized lipase was used for the production of lauryl laurate in hexane medium. Lipase immobilization on the PEI support exhibited a significantly improved operational stability in esterification system. After re-use in 30 successive batches, a high ester yield (88%) was maintained. These results indicate that PEI, a polymeric bed, could not only bridge support and immobilized enzymes but also create a favorable micro-environment for lipase. This study provides a simple, efficient protocol for the immobilization of Y. lipolytica lipase Lip2 using PUF as a cheap and effective material.  相似文献

Short chain alkyl esters are well appreciated for fruity flavors they provide. These are mainly applied to the fruit-flavored products like jam, jelly, beverages, wine and dairy. Cutinase from Burkholderia cepacia NRRL B 2320 was found to be active in catalyzing the synthesis of alkyl esters in organic solvent. The optimal temperature range for the enzyme catalyzed synthesis was found to be from 35 °C to 40 °C. The maximum conversion (%) during synthesis of ester was obtained for butyric acid (C4) and valeric acid (C5) with butanol reflecting the specificity of the enzyme for short-chain length fatty acids. In case of alcohol specificity, butanol was found to be most preferred substrate by the enzyme and conversion (%) decreased with increasing carbon chain length of alcohol used in the esterification reaction. The kinetic analysis for the synthesis of butyl butyrate by varying concentration of one substrate at a time (butanol or butyric acid), showed that Ping–Pong Bi Bi model with acid inhibition and influence of initial water is most suitable model for the prediction of the reaction kinetics.  相似文献

The synthetic activity of lipases in biphasic o/w systems was investigated with respect to their use in the synthesis of polyester chains via transesterification reactions. Lipase-catalyzed ring-opening polymerization (ROP) of pentadecalactone (ω-PDL) dispersed in water was used as a model reaction to understand the synthetic activity of lipases in biphasic o/w system. We conducted a systematic investigation of the influence of reaction conditions on the macromolecular characteristics of oligo(ω-PDL) encompassing chemical, thermophysical and colloidal properties of the reaction medium. A model was proposed assuming Michaelis–Menten interfacial kinetics followed by chain extension via lipase-catalyzed linear polycondensation. The solidification of oligo(ω-PDL) chains with a degree of polymerization of approximately three was identified as a major factor limiting the molecular weight of obtained oligomers to ∼870 g mol−1, despite the fast reaction rate and complete conversion of ω-PDL. The addition of toluene into the dispersed phase at a volumetric ratio of 0.3–0.5 of toluene to ω-PDL allowed us to circumvent this problem and increase the molecular weight of obtained oligomers up to 1460 g mol−1. The molecular weight of polymer product according to this model was thus inversely related to the weight ratio percentage of interfacial lipase PS to ω-PDL per droplet and correspondingly correlated with the activity of lipase. Taking into account all these parameters allowed increasing the molar mass of oligo(ω-PDL) from 870 g mol−1 to 3507 g mol−1.  相似文献

The nitrogen uptake capabilities of the toxigenic diatom Pseudo-nitzschia australis (Frenguelli), freshly isolated from Monterey Bay California, were examined in unialgal laboratory cultures at saturating photosynthetic photon flux densities (100 μmol photons m−2 s−1) and 15 °C. The kinetics of nitrogen (nitrate, ammonium, urea and glutamine) uptake as a function of substrate concentration were estimated from short (20.5 min) incubations using the 15N-tracer technique. Based on the estimated maximum specific uptake rates and measures of N affinity (the initial slope of the uptake versus nutrient concentration curve), nitrate is the preferred nitrogen substrate, followed by glutamine and ammonium, which are equivalent. Rates of urea uptake by P. australis did not saturate at concentrations as high as 36 μg-at N L−1, and urea uptake as a function of concentration could not be described by Michaelis–Menten kinetics over the concentration gradient tested. Although there is a clear preference for nitrate at equivalent concentrations (compared to ammonium, urea, and glutamine), these laboratory results demonstrate the capability of this pennate diatom to utilize both inorganic and organic forms of nitrogen, supporting field observations that P. australis blooms during both upwelling and non-upwelling conditions off the west coast of North America. Substantial differences in the nitrogenous nutrition of P. australis can be expected in these environments, and anthropogenic inputs of N substrates such as ammonium and urea can support its growth, and may contribute significantly to both harmful diatom blooms and the maintenance of seed populations at non-bloom abundances, particularly during periods of reduced or absent upwelling.  相似文献
