Summary The structure of myoneural junctions in the tibialis anterior and intercostal muscles was studied after histochemical reaction for myoneural acetylcholinesterase (E.C. 3.1.1.7) in human embryos.Myoneural junctions of a primitive form were first seen in the intercostal muscle at the age of 8.6 weeks (crown-rump length of 3.2 cm), and in the tibialis anterior muscle at the age of 10 weeks (4.3 cm). The postsynaptic membrane was devoid of any junctional folds typical of adult synapses up to the age of about 19 weeks. At first small, the junctional folds gradually became deeper and more prominent during the following weeks, and some ramification of the previously coherent postsynaptic area took place. Myoneural morphogenesis was not completed at birth, although well developed postsynaptic Moldings were present.  相似文献

The development of muscular tissue fibers has been studied in 8--19-day-old chick embryos by methods of electron microscopy, autoradiography and cytophotometry. Differentiated myogenic elements are derived from promyoblasts which devide during myogenesis at slightly changing parameters of the mytotic cycle. Promyoblasts fuse with each other or with symplasts or else can develop into myoblasts containing myofilaments in their cytoplasm. At later stages of myogenesis, some promyoblasts pass into "rest" condition with characteristic changes in the structure of nucleus, cytoplasm and muscular cells. These cells are situated between the basal and cytoplasmic membranes and identified as cell-satellites (myosatellites). They represent an important subpopulation of the muscular system, which preserve a certain ability to perceive stimuli to proliferation and differentiation, forming the cambial system of the skeletal-muscular tissue.  相似文献

By means of the radioautography, morphometry and alkaline dissociation methods musculus tibialis anterior has been studied in 30 chick embryos and chickens. The muscle fibers are variable in their diameter and length, number of nuclei 3H-thymidine-labelled including. The greatest variability of the structures is noted in 11-14-day-old embryos. Three classes of myotubes (MT) and symplasts are described. The primary MT are evidently the source of cellular elements, that form new cellular-symplastic structures (muscular-proliferative units) that develop, for some time, in a close connection with the primary MT. Between the extreme morphological variants of structure of the muscular elements, there is a transitional form. All the forms of the myosymplasts and MT reflect various stages of differentiation of the myogenic elements; this results in formation of the cellular-symplastic system--muscle fiber.  相似文献

Using multiple immunofluorescence labelling on human breast tissues obtained and freshly frozen at the 12th, 15th, and 18th weeks of pregnancy, we have shown that markers of mammary functional differentiation, milk proteins (beta-casein and kappa-casein), are synthesised by actively cycling (Ki67 positive) as well as non-cycling (Ki67 negative) cells. These results demonstrate that functional differentiation/maturation does not coincide with loss of proliferative potential in human mammary luminal epithelial cells. In addition, we have examined expression patterns of integrin subunits (alpha1, alpha2, alpha3, alpha6, beta1, and beta4) and extracellular matrix components (laminin, fibronectin, collagen I, and collagen IV), since they have been shown to exert influences on mammary differentiation and morphogenesis in vitro. Compared to human breast tissues obtained from non-pregnant women, a decrease in alpha2 labelling on luminal epithelial cells was observed, particularly in expanding acini that showed abundant Ki67 positivity. The expression patterns of other integrin subunits, however, did not change, indicating that the expression patterns of most integrins existing prior to pregnancy are sufficient to support the morphological and functional development associated with milk protein synthesis.  相似文献

Chondrocytes freshly explanted from the sternal cartilage of 14- to 16-day-old chick embryos proliferate and differentiate in cell culture in a serum-free medium as well as in a medium containing 10% foetal calf serum. A comparable degree of multiplication and differentiation was found in chondrocytes cultivated in the serum-free medium containing native growth-promoting alpha-globulin (GPAG). The degree of proliferation was smaller in chondrocytes cultivated in a modified Eagle MEM which is fully chemically defined and contains low molecular weight substrates only. Since this medium does not contain either hormones or proteins, it is suitable for the cultivation of chondrocytes which should be employed when studying the mechanism of the effect of hormones which influence chondrocyte proliferation or chondrogenesis.  相似文献

A comparative study has been made of erythroid cell development pathways in the peripheral blood of pigeons during severe, moderate and weak forms of anaemia. Three modes of erythrocyte formation from bone marrow precursor are described: 1. A reserve erythropoiesis--the principal process during severe anaemia; the bone marrow precursors are basophylic erythroblasts which are reversibly blocked in phase G2 of the cell cycle; in results the rapid, increase of erythrocyte population above the normal level, although the cells have 25-30 per cent deficiency in haemoglobin content. 2) A mode of erythropoiesis, whose precursors are proliferating polychromatophylic erythroblasts; this is the principal mode of erythropoiesis at the moderate anaemia, leading to restoration of the normal quantity of erythrocytes with a normal haemoglobin content. 3) A mode of erythropoiesis with proliferating orthochromatic erythroblasts being precursors (which do not divide normally); this is the principal mode during the weak anaemia to result in a slow restoration of the number of erythrocytes with an excess in haemoglobin content. It is shown that regulation of the restoration processes during anaemia are characterized by a specific combination of cell proliferation and differentiation.  相似文献

Evidence is presented from C-banding studies that the W chromosome of eleven species of snakes is not homogeneous in nature but is differentiated linearly into alternating lighter and darker C positive regions. The same is true of the W chromosome of at least some birds. There is evidence from the literature indicating a similar differentiation of the Y chromosome of some mammals and here the intermediate C positive regions are deficient in highly repetitive DNA. The significance of heterochromatinization as a means of generating differentiated sex chromosomes is discussed in the light of these findings.Dedicated to Professor M.J.D. The rate of ageing is a genetically influenced feature of an individual's life history that responds to selection on lifespan. Various costs presumably constrain the evolution of prolonged life, but these have not been well characterized and their general nature is unclear. The analyses presented here demonstrate a correlation among birds and mammals between rates of embryonic growth and ageing-related mortality, which are quantified by the exponents of fitted power functions. This relationship suggests that rapid early development leads to accelerated ageing, presumably by influencing some aspect of the quality of the adult individual. Although the mechanisms linking embryo growth rate and ageing are not known, a simple model of life-history optimization shows that the benefits of longer life can be balanced by connected costs of extended development.  相似文献

Although cancers are highly heterogeneous at the genomic level, they can manifest common patterns of gene expression. Here, we use gene expression signatures to interrogate two major processes in cancer, proliferation and tissue remodeling. We demonstrate that proliferation and remodeling signatures are partially independent and result in four distinctive cancer subtypes. Cancers with the proliferation signature are characterized by signatures of p53 and PTEN inactivation and concomitant Myc activation. In contrast, remodeling correlates with RAS, HIF-1α and NFκB activation. From the metabolic point of view, proliferation is associated with upregulation of glycolysis and serine/glycine metabolism, whereas remodeling is characterized by a downregulation of oxidative phosphorylation. Notably, the proliferation signature correlates with poor outcome in lung, prostate, breast and brain cancer, whereas remodeling increases mortality rates in colorectal and ovarian cancer.  相似文献

Summary The structure of the myoneural junction in the striated muscle of rat embryos and postnatal rats was studied by electron microscopy in order to assess at ultrastructural level the roles of neuronal and muscular elements and the sequence of events resulting in the formation of a functionally mature synaptic organization.From the observations it is concluded that the axon terminals enveloped by Schwann cells contain vesicles prior to apposition of the prospective synaptic membranes. Subsequently, subsarcolemmal thickening of the postsynaptic membrane takes place after the synaptic gap has been formed by disappearance of the teloglial cell from between the synaptic membranes but before the primary synaptic cleft in the strict sense is formed. Secondary synaptic clefts are formed later, when the primary synaptic cleft is regular in width, by local finger-like invaginations of the postsynaptic membrane, which thereafter expand basally, in a plane transverse to the axis of the axon terminal, to resemble flattened flasks. The junction is formed between multinucleated muscle cells and multiple axons, which at first lie side by side and later, when formation of adult-type secondary synaptic clefts is in progress, become separated by folds of the sarcoplasm and the teloglia. In extraocular muscles of adult rats the sarcoplasmic reticulum is closely associated with the postjunctional sarcoplasm.In the light of earlier observations on the development of contractibility after nerve stimulation, cholinesterase histochemistry and muscle fibre physiology, these observations are interpreted to indicate that functional differentiation of the myoneural synapse results from induction by the motor axon and that the association of the sarcoplasmic reticulum with the postjunctional sarcoplasm in adult extraocular muscles is related to modified fibre physiology.The author wishes to thank Prof. Antti Telkkä, M.D., Head of the Electron Microscope Laboratory, University of Helsinki, for placing the electron microscopic facilities at his disposal.  相似文献

The endosperms of Carthamus tinctorius cv. HUS-305, excised at globular to heart-shaped stages of zygotic embryo development, were cultured on Murashige and Skoog’s medium (MS) supplemented with different concentrations of 6-benzylaminopurine (BAP), kinetin, thidiazuron (TDZ), 2,4-dichlorophenoxyacetic acid (2,4-D) or α-naphthalene-acetic acid (NAA). The highest incidence of callusing was on 2,4-D supplemented media. However, embryos differentiated only from the calli developed on media supplemented with BAP, kinetin or TDZ with the last eliciting maximum embryogenic response. The addition of a reduced nitrogen source, casein hydrolysate to MS medium supplemented with BAP and/or NAA, did not stimulate the response. However, adenine sulphate (100 mg dm−3) promoted the induction of somatic embryos. Upon transfer to MS basal medium or the same supplemented with 0.61 μM gibberellic acid (GA3), plumular poles of few embryos elongated resulting in the development of shoots.  相似文献
