Summary The pattern of diversity in forty genotypes of clusterbean (Cyamopsis tetragonoloba (L.) Taub) was analysed using the centroid method of factor analysis based on nine characters. Three factors were found to adequately account for most of the intercorrelations in the four environments. Branches, clusters, pods and seed yield/plant could be grouped as productivity factors; days to flowering, maturity and plant height as growth factors; seeds/pod and gum content as economic factor. Interestingly, the constitution of the factors were the same in all the environments. The loadings on the economic factors were relatively unstable, whereas the loadings on productivity and growth factors were stable. It is concluded that productivity and growth factors play a pivotal role towards diversity in clusterbean. The results of factor analysis provides confirmatory evidence of diversity in clusterbean, which was earlier studied by using generalized distance, and thus proves the adequacy of the centroid method in biological investigations.  相似文献

The photosynthetic potential of leaves and chloroembryos of Cyamopsis tetragonoloba (L.) Taub as measured by 14C-bicarbonate fixation, Hill activity, and in vivo fluorescence transients is compared. On a chlorophyll basis, dark fixation of NaH14CO3 in chloroembryos was 1.5 times higher than that of the leaf, whereas carbon fixation under illumination was threefold higher in the leaf than in the embryos. Rates of O2 evolution were four times more in embryo than in leaf chloroplasts. Shading of developing fruits on the day of anthesis for 10 days induced a 65% reduction in dry matter accumulation in the etiolated embryos, as compared to the normal green embryos of the same fruit half covered by a transparent Polythene sheet. The reduction in dry weight, size of the embryos, and levels of assimilates after shading the developing fruits may be ascribed to partial autotrophy of the chloroembryos.  相似文献

Pre-exposure of clusterbean plants to vitamins of the B group:thiamine, riboflavin, niacin, pantothenic acid, pyridoxine andfolic acid resulted in a significant increase in protein contentof the leaves and fruits which was also evident from the differentprotein patterns in the electrophorograms. Growth of C. tetragonoloba suffered with increase in concentration of Pb in soil. Plant biomass declined significantly at concentrations above 60 ppm of Pb. Roots showed more pronounced impact as compared to shoots. At highest applied concentration of lead (100 ppm), fresh weight of fruits decreased by 33% and dry weight by 52% as compared to control. No significant impact was noticed on the development of mycorrhiza at lower concentrations (15-45 ppm) of lead contamination. At higher concentrations of Pb (60 and 75 ppm), there was a decrease in VAM colonization. VAM hyphae had irregular size and terminated abruptly in outer cortex of root. Number of VAM fungal spores in rhizosphere also decreased with increase in the edaphic Pb concentration.  相似文献

Summary Guar (Cyamopsis tetragonoloba L. Taub) is a drought-tolerant multipurpose cash crop. A rapid regeneration system has been developed for four Indian guar genotypes. Investigations were carried out to assess the effect of different growth regulators and their combinations on a variety of explants such as the embryo, cotyledons, and cotyledonary nodes for shoot morphogenesis. It was established that Murashige and Skoog's culture medium containing 6-benzylaminopurine (13.3 μM or 3 mgl−1) in combination with indole-3-acetic acid (11.4 μM or 2mgl−1) with cotyledonary node explants gave the highest frequency of multiple shoot induction. In vitro rooting from cultured shoots was maximal on a half-salt concentration of Murashige and Skoog's culture medium fortified with indole-3-butyric acid (4.9 μM or 1 mgl−1). In vitro-regenerated plants were grown to pod setting and subsequent maturity in greenhouse conditions.  相似文献

A 2-year field trial was conducted to study the effects of seed treatment (500 ppm) or foliar application of 1000 ppm thiourea (at 25 and 40 days after sowing) or a combination of these, on growth, yield, net photosynthesis and nitrogen metabolism of clusterbean (Cyamopsis tetragonoloba (L.) Taub.) grown for two consecutive years (1999 and 2000) under rainfed conditions of the Indian arid zone. Thiourea application either as pre-sowing seed treatment or as foliar spray significantly increased plant height, leaf area, dry matter production and seed yield as compared to the untreated control plants during both the years. However, maximum favourable effects were obtained with combined application of seed treatment and foliar spray. The beneficial effects of thiourea were attributed to its role in significantly increasing the net photosynthetic rates and the concentrations of total chlorophyll and starch in the leaves. Thiourea also reflected a positive role in enhancing nitrogen metabolism as it significantly increased nitrate reductase activity and concentration of soluble protein in the treated plants. It has been concluded that seed treatment with thiourea followed by foliar spray could significantly improve growth, yield and water use efficiency of rainfed clusterbean under arid conditions due to enhanced photosynthesis and more efficient nitrogen metabolism.  相似文献

Essential oils isolated from leaves and seeds of seven umbelliferous plants were tested against the growth ofAspergillus flavus. Those from seeds ofTrachyspermum ammi, Cuminum cyminum, Carum carvi, Daucus carota and from leaves ofAnethum graveolens exhibited antifungal activity against the test fungus. Amongst these, oil from seeds ofTrachyspermum ammi was most toxic. Its minimum inhibitory concentration was 300 ppm, at which it exhibited fungistatic but not phytotoxic properties, when tested at 200, 300 and 400 ppm. The fungitoxic potency ofTrachyspermum seed oil remained unchanged after a long storage period and at high inoculum density of the test fungus. The oil was thermostable and was more efficaceous than the fungicides Agrosan G.N., Benlate, Ceresan, Dithane M-45 and Thiovit commonly used for the control of plant diseases.  相似文献

Galactomannan from seeds of Cyamopsis tetragonoloba (L.) Taub. (guar) was depolymerized using immobilized enzymatic preparation celloviridin. A set of fragments whose molecular weights varied from 12.6 to 245.6 kDa was obtained. Sulfated derivatives of components of all fractions were synthesized, in which the content of HSO 3 ? -groups was 48.05 ± 2.31%. All preparations exhibited anticoagulant activity, which was recorded in vitro in two tests—aIIa and aXa. The antithrombin activity (aIIa) was high (up to 65–87 U/mg) and did not depend on the molecular weight of a sulfated derivative; in the second test (aXa), the effect of molecular weight was observed. Biospecific electrophoresis allowed us to detect the ability of galactomannan sulfates to form complexes with protamine sulfate, a classic antidote to heparin.  相似文献

A procedure for transformation of the large-seeded endospermous legume guar (Cyamopsis tetragonoloba L.) and a study on transmission of the transgenes to offspring generations are presented. Using Agrobacterium tumefaciens with a T-DNA construct harbouring a -glucuronidase gene (uidA) and a neomycin phosphotransferase gene (nptII), maximum transformation frequencies of cotyledonary explants were obtained using 145 mg/l kanamycin sulfate as selective agent. Carbenicillin and cefotaxime, used for the elimination of Agrobacterium after co-culture, displayed considerable toxicity to guar tissues but replacing most of these -lactams by the non-phytotoxic -lactamase inhibitor sulbactam as well as addition of thidiazuron and silver thiosulfate increased transformation frequencies up to 10-fold in total. The presence of the transgenes in the primary transformants was demonstrated by genomic DNA analysis of GUS-positive shoots. Chimaeric plants (5–10%) were identified by GUS analysis at the flowering stage and were discarded. Analysis of the R1 offspring from 17 independent transformants showed that in 41% of those, the uidA gene(s) was expressed and stably inherited consistent with Mendelian genetics. This was also found for the R2 and R3 generations of single copy transformants. On the other hand, a large proportion (47%) of the primary transformants gave R1 offspring in which 100% of the plants were GUS-negative. Analysis of these plants by PCR revealed that, at least, most of the transgene sequences were absent, suggesting that they had not been transmitted from the parent transformants. This occurred at similar high frequencies (40–50%) irrespective of the estimated copy number of the transgenes. Thus, major parts of the transgenes, even when present in multiple copies, displayed aberrant transmission, at a high frequency, in the process of going from the primary transformants to the first offspring generation.  相似文献

Abstract

CIP-29, a known 29 kDa systemic antiviral resistance inducing protein isolated from Clerodendrum inerme, has been used to induce systemic resistance in Cyamopsis tetragonoloba against Sunn-hemp rosette virus (SRV). Paper reports the detection of virus inhibitory activity in induced-resistant leaf sap of C. tetragonoloba, and the purification of two virus inhibitory agents (VIAs) thereof. VIA activity was recorded as a reduction in lesion number of SRV, Tobacco mosaic virus, and Papaya ringspot virus, when they were incubated separately with resistant sap and inoculated onto susceptible C. tetragonoloba, Nicotiana tabacum cv. Xanthi-nc, and Chenopodium quinoa, respectively. The two VIAs were isolated from resistant C. tetragonoloba plant leaves using combinations of column chromatography. Both were basic proteins, and since their M r was 32 and 62 kDa, these VIAs were called CT-VIA-32 and CT-VIA-62, respectively, on the basis of their molecular mass and the host. CT-VIA-62 displayed better activity, and was thus studied further. It tested positive for a glycoprotein, and was serologically detected only in leaf tissue post-induction. Tryptic peptides generated in-gel, post SDS-PAGE of CT-VIA-62, were sequenced through LC/MS/MS. All CT-VIA-62 peptides were found to share homologies with proteins from Medicago truncatula that possess a mannose-binding lectin domain.  相似文献

The intraseminal embryo of Cyamopsis tetragonaloba is greenfrom the globular stage onwards. The chloroembryos do not photosynthesizein vivo but are capable of in vitro photosynthesis at a ratewhich is similar to that of leaf tissue and they utilize bicarbonateions in this process. Both ribulose bisphosphate (RuBP) andphospho-enol-pyruvate (PEP) carboxylases are functional in CO2fixation by the embryos. Guar seed gum, consisting primarily of a high molecular weight galactomannan, is the most cost effective natural thickener, having broad applications in the food, cosmetics, paper, pharmaceutical and petroleum industries. The properties of the polymer can potentially be enhanced by genetic modification. Development of suitable endosperm-specific promoters for use in guar is desirable for metabolic engineering of the seed gum. A ~1.6 kb guar mannan synthase (MS) promoter region has been isolated. The MS promoter sequence was fused with the GUS reporter gene and overexpressed in the heterologous species alfalfa (Medicago sativa). The potential strength and specificity of the MS promoter was compared with those of the constitutive 35S promoter and the seed specific β-phaseolin promoter. Quantitative GUS assays revealed that the MS promoter directs GUS expression specifically in endosperm in transgenic alfalfa. Thus, the guar MS promoter could prove generally useful for directing endosperm-specific expression of transgenes in legume species.  相似文献

About 377 guar (Cyamopsis tetragonoloba) rhizobacteria were isolated from cultivated soils of north-west India (Thar Desert) and their antifungal activity against Macrophomina phaseolina (strains of groundnut, mungbean and guar) and Fusarium oxysporum (strains of chickpea and cumin) was examined. Isolates were characterised for generic types and physiological/functional diversity. About 19% isolates representing 24% locations were inhibitory to fungal growth. Isolates 009071, 009073, 009078 and 102354 recorded maximum inhibition of pathogenic fungi on plates. Isolate 034206 gave highest %RI, 009073 showed maximum protease activity and 102354 gave highest salt tolerance. Net house and field screening results revealed that isolates 004052, 009071, 009073, 001001, 094340 and 102354 had potential for biocontrol of disease. Partial sequencing of 16S rRNA gene of 61 isolates showed that 85% of isolates belonged to genus Bacillus. Phylogenetically, however, there were four clusters in the Bacillus group comprising of Bacillus subtilis, B. cereus, B. pumilus and B. sphaericus. One isolate was identified as B. flexus, while six isolates were Bacillus spp. Four isolates were identified as Achromobacter xylosoxidans, two as Bacterium (unclassified bacteria), and one each as Ochrobactrum intermedium, Pseudomonas aeruginosa and Ralstonia sp.  相似文献

Protoplasts of Cyamopsis tetragonoloba were isolated from leaves of in vitro grown plants. The yield of the protoplasts, their viability and subsequent divisions were greatly influenced by the pH of the media used for isolation and culture of protoplasts. Sustained divisions of the cultured protoplasts were best supported by a modified Kao and Michayluk (1975) nutrient medium containing glucose (0.4 M), NAA (4 mgl–1), 2,4-D (1 mgl–1) and KIN (2 mgl–1 ). The protoplast derived cells developed calli on transfer to Murashige and Skoog (1962) medium supplemented with 1 mgl–1 each of 2,4-D, NAA and KIN.  相似文献

Galactomannan gums are widely used in the food and oil industries, and there is considerable interest in applying biotechnological approaches to improve their physical properties. A mannan synthase from guar ( Cyamopsis tetragonoloba ) was expressed under the control of a bean β-phaseolin promoter in transgenic Medicago truncatula . Although the expression of exogenous mannan synthase caused a slight decrease in galactomannan levels in Medicago , the molecular weight and viscosity of the polymer were significantly increased, although the mannose to galactose ratio and degree of polydispersity remained unchanged. At the same time, expression of about 2.8% of the genes was altered significantly in the seeds of transgenic Medicago lines analysed by Affymetrix genome chip, with a particularly striking induction of putative trehalose phosphate synthase genes. Mannan synthase expression also caused large alterations in the levels of a number of sugars and sugar alcohols, suggesting that over-expression of a processive glycosyltransferase perturbs the mechanisms of sugar sensing and/or homeostasis, possibly involving signalling via trehalose-6-phosphate.  相似文献

A fusion of DNA sequences encoding the SPO2 promoter, the alpha-amylase signal sequence from Bacillus amyloliquefaciens, and the mature part of the alpha-galactosidase from Cyamopsis tetragonoloba (guar) was constructed on a Bacillus subtilis multicopy vector. Bacillus cells of the protease-deficient strain DB104 harboring this vector produced and secreted the plant enzyme alpha-galactosidase up to levels of 1,700 U/liter. A growth medium suppressing the residual proteolytic activity of strain DB104 was used to reach these levels in a fermentor. Purification of the secreted product followed by NH2-terminal amino acid sequencing showed that the alpha-amylase signal sequence had been processed correctly. The molecular mass of the product estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis was slightly lower than that of the plant purified enzyme, which is most likely due to glycosylation of the latter. The alpha-galactosidase product was active both on the artificial substrate para-nitrophenyl-alpha-D-galactopyranoside and on the galactomannan substrate, guar gum. The activity of this Bacillus sp.-produced enzyme was similar to that of the glycosylated enzyme purified from guar seeds, indicating that glycosylation has no essential function for enzyme activity.  相似文献
