The process of the denaturation of “myosin B” solution was studied by the measurement of ATPase activity, SH groups, sedimentation behaviour and flow birefringence. When “myosin B” solution was stored at lower temperature, lower pH or higher ionic strength, the interaction between myosin A and actin became less strong, and further storage brought about an irreversible dissociation.

The condition for measuring Mg-modified ATPase activity of “myosin B” at low ionic strength was explained in the relation with superprecipitation.  相似文献

This study examined social behaviour of juvenile red sea urchins, Strongylocentrotus franciscanus (Agassiz). At four coastal locations in British Columbia, Canada, distributions of juvenile sea urchins were examined in relation to adults. One-third of all juveniles were found underneath an adult “spine canopy”, another third outside the spine canopy but close to an adult, and the remainder were found apart from adults. Juveniles associated with adults were contagiously distributed with each other. In the laboratory, juveniles chose adults over other target locations. They selected adults in particular locations over others, and in one case showed selection among adults; but they did not select between fed and non-fed adults, nor between adults that had been “protective” and “non-protective” of juveniles in the field. In time-lapse film studies, starved juveniles showed little change in social behaviour when food was introduced, but showed a dramatic change in behaviour when predators were active. We suggest that juvenile red sea urchins are found under the spine canopies of adults as a result of juvenile behaviour; and that this interaction functions to protect juveniles from predation. The strong gregarious nature of juveniles among themselves may have evolved in a different selective situation as a defence against predation and to ensure reproductive success.  相似文献

Earlier studies on social behaviour in the domestic pig have only been performed on groups housed artificially. The present study was undertaken to investigate the social behaviour of free-ranging sows in a well-established flock, so that it could be compared with the behaviour of sows housed indoors that had been studied earlier. The free-ranging flock, which contained 13 animals, 5 of which were sows, lived in an enclosure, approximately 1.1 ha in area. The interaction sequences were registered during 1 h after feeding and during foraging in the enclosure. With sequence analysis, the effect of different behaviours on the behaviour of the receiver animal was measured. This allowed the behaviour patterns to be classed after function. “Aiming”, a behaviour pattern not previously seen, was interpreted as a threat behaviour, being “statistically strongly directive” towards submissive and flight behaviour. The functions of the other 7 behaviours included in the study were found to be the same as in the indoor groups studied earlier. The dominance order was not found to be better established than in the most extensively housed indoor group, but the level of aggression was very low, indicating that a stable dominance order does not automatically lead to a low level of aggression. It was found that the dominance order was mainly maintained through the behaviour of the subordinate animals, supporting the use of an “avoidance order” instead of an “aggression order” as a measurement of social dominance. On the whole, no dramatic differences were found between the behaviour of the free-ranging sows and that of the most extensively housed of the indoor groups studied earlier, although great differences were found between free-ranging sows and sows kept in confinement indoors.  相似文献

The influence of the class IV calcium antagonist flunarizine on the phase behaviour of different species of the major phospholipid classes of mammalian plasma membranes has been examined using differential scanning calorimetry. We show that it has the ability to substantially influence the phase behaviour of phospholipids. Flunarizine significantly influences the gel to liquid-crystalline transition temperature of phosphatidylserines whilst having little effect on those of the phosphatidylethanolamines tested. The liquid-crystalline to inverted hexagonal phase transition of phosphatidylethanolamines is, however, strongly induced by the presence of flunarizine. Examination of the effect of flunarizine on the phase behaviour of different phosphatidylcholine species revealed an acyl-chain dependent influence. Dissimilar results with phosphatidylcholines, phosphatidylethanolamines and phosphatidylserines reveal different locations and ionization states for the drug in the different phospholipid bilayers. These results not only indicate an essential role for the ionization state of the drug in determining drug-phospholipid interactions but also the role of the phospholipid in determining the ionization state of the drug and have important implications for drug-membrane interactions demonstrating that drug interaction with one phospholipid may bear no relation whatsoever to its interaction with another.  相似文献

Abstract

The principle purpose of this paper is to demonstrate the use of the Inverse Monte Carlo technique for calculating pair interaction energies in monoatomic liquids from a given equilibrium property. This method is based on the mathematical relation between transition probability and pair potential given by the fundamental equation of the “importance sampling” Monte Carlo method. In order to have well defined conditions for the test of the Inverse Monte Carlo method a Metropolis Monte Carlo simulation of a Lennard Jones liquid is carried out to give the equilibrium pair correlation function determined by the assumed potential. Because an equilibrium configuration is prerequisite for an Inverse Monte Carlo simulation a model system is generated reproducing the pair correlation function, which has been calculated by the Metropolis Monte Carlo simulation and therefore representing the system in thermal equilibrium. This configuration is used to simulate virtual atom displacements. The resulting changes in atom distribution for each single simulation step are inserted in a set of non-linear equations defining the transition probability for the virtual change of configuration. The solution of the set of equations for pair interaction energies yields the Lennard Jones potential by which the equilibrium configuration has been determined.  相似文献

Based on an analysis of L.S. Vygotsky's concepts of “units” and “elements” of psychological systems, this article highlights five of their attributes. It shows that these attributes are logically symmetrical, since in their wording they can be converted into one another by negation or by replacing some words with their opposites. This suggests that the concepts of the “unit” and “element” of a system are different poles of one theoretical construct of the activity of human psychology. Thus methods for the study of psychological systems by breaking them down into elements or by separating them into units can be seen as complementary. The article describes differences among the concepts of “unit,” “minimal unit,” and “cell” of a psychological system. It reviews several problems that are solvable using the “method of units,” as well as some concepts of the theory of psychological systems that are understood as holistic, conceptual, and active processes and/or results of human interaction with the world. Among the examples of such systems are “systems of psychological functions” (according to Vygotsky), as well as separate activities (according to A.N. Leontiev), human actions and operations (interactions with the world on the level of objects and mental or physical means). The “component” of a psychological system is defined as any “something” that in some sense belongs to or is included in human interaction with the world. A component that belongs to the system is called an “element” of it, but a component that is included in the functioning and development of the system is called a “part” of it. The article presents the mathematical and psychological foundation of these definitions. It identifies and discusses the substantial (independently existing) components of psychological systems and their attributes (properties and conditions). It describes the relationships between them using the bipolar theoretical constructs “part-element” and “substantial-attributive” component of a system.  相似文献

A general theory of the structural changes and fluctuations of proteins has been proposed based on statistical thermodynanic considerations at the chain level.The “structure” of protein was assumed to be characterized by the state of secondary bonds between unique pairs of specific sites on peptide chains. Every secondary bond changes between the bonded and unboned states by thermal agitation and the “structure” is continuously fluctuating. The free energy of the “structural state” that is defined by the fraction of secondary bonds in the bonded state has been expressed by the bond energy, the cooperative interaction between bonds, the mixing entropy of bonds, and the entropy of polypeptide chains. The most probable “structural state” can be simply determined by graphical analysis and the effect of temperature or solvent composition on it is discussed. The temperature dependence of the free energy, the probability distribution of structural states and the specific heat have been calculated for two examples of structural change.The theory predicts two different types of structural changes from the ordered to disordered state, a “structural transition” and a “gradual structural change” with rising temperature, In the “structural transition”, the probability distribution has two maxima in the temperature range of transition. In the “gradual structural change”, the probability distribution has only one maximum during the change.A considerable fraction of secondary bonds is in the unbonded state and is always fluctuating even in the ordered state at room temperature. Such structural fluctuations in a single protein molecule have been discussed quantitatively.The theory is extended to include small molecules which bind to the protein molecule and affect the structural state. The changes of structural state caused by specific and non-specific binding and allosteric effects are explained in a unified manner.  相似文献

Social animals interact frequently with conspecifics, and their behaviour is influenced by social context, environmental cues and the behaviours of interaction partners, allowing for adaptive, flexible adjustments to social encounters. This flexibility can be limited by part of the behavioural variation being genetically determined. Furthermore, behaviours can be genetically correlated, potentially constraining independent evolution. Understanding social behaviour thus requires carefully disentangling genetic, environmental, maternal and social sources of variations as well as the correlation structure between behaviours. Here, we assessed heritability, maternal, common environment and social effects of eight social behaviours in Neolamprologus pulcher, a cooperatively breeding cichlid. We bred wild‐caught fish in a paternal half‐sibling design and scored ability to defend a resource against conspecifics, to integrate into a group and the propensity to help defending the group territory (“helping behaviour”). We assessed genetic, social and phenotypic correlations within clusters of behaviours predicted to be functionally related, namely “competition,” “aggression,” “aggression‐sociability,” “integration” and “integration‐help.” Helping behaviour and two affiliative behaviours were heritable, whereas there was little evidence for a genetic basis in all other traits. Phenotypic social effects explained part of the variation in a sociable and a submissive behaviour, but there were no maternal or common environment effects. Genetic and phenotypic correlation within clusters was mostly positive. A group's social environment influenced covariances of social behaviours. Genetic correlations were similar in magnitude but usually exceeding the phenotypic ones, indicating that conclusions about the evolution of social behaviours in this species could be provisionally drawn from phenotypic data in cases where data for genetic analyses are unobtainable.  相似文献

Throughout the animal kingdom, individual variation in reproductive success is commonly observed, even under similar environmental conditions. However, the mechanisms behind such differences remain unclear. The notion of behavioural consistency in animals has developed rapidly since the early 21st century partly as an approach to understand among‐individual differences. In this context, a number of studies have highlighted the influence of pair assortment in personality on breeding success. In this study, we related breeding success to individual behaviour, specifically a risk‐taking behaviour, and pair assortment per behaviour in African penguins (Spheniscus demersus) over two breeding seasons of contrasting food availability. On Bird Island, Algoa Bay, South Africa, we used indices of boldness and overall mobility in penguins’ nest defence behaviour as a response to a standard pedestrian approach during chick‐rearing. These behaviours were consistent over the trials and indicated these traits may be related to personality in African penguins. Individuals were categorized as risk‐prone (“bold,” “mobile”) or risk‐averse (“shy,” “non‐mobile”). We then assessed their breeding success through chick growth and survival over 4 weeks in 2015 and 2016. There was weak positive assortment of pairs in relation to nest defence behaviour. However, pair assortment did not significantly influence birds’ breeding success. Shy penguins were generally the most successful (had the highest chick growth rates), which was especially apparent during a food shortage in 2016, possibly reflecting a higher energy investment when foraging. In contrast, chicks from bold parents grew significantly slower, especially in 2016. Bold parents may defend their nest successfully against predation or intra‐specific aggression when food is abundant, but when predation risks are limited and food availability is low, this strategy may not be beneficial. In the context of climate change, where food shortage events may become more frequent, risk‐averse individuals may be favoured and genetic diversity may be reduced in African penguins.  相似文献

An exactly solvable model of the interaction of small nonpolar molecules with biological membranes is developed. This model, which is based upon a “decorated dimer model” extension of Nagle's membrane model, is demonstrated to qualitatively reproduce many of the changes in the order-disorder phase transition seen when biological membranes are exposed to anesthetic gases. The decorated dimer model is itself interesting because it provides an example of an exactly solvable monomer-dimer model in which phase transitions can occur in the presence of monomers.  相似文献

The synthesis of a protein unique to the nervous system, the “S100-protein,” has been studied in a clonal line of rat glial cells. It has been shown that these cells do not begin to accumulate “S100-protein” until the cultures enter a phase of density-dependent inhibition of cell proliferation. Further experiments indicate that the regulation of “S100-protein” accumulation resides at least in part in an interaction involving the cell surface.  相似文献

Abstract

Simulations of the lamellar phase transitions of symmetric amphiphilic chains are carried out on a cubic lattice, with the amphiphilic chain length N varied from 6 to 48 lattice sites, corresponding to lengths ranging from surfactants to short block copolymers. We find that the effective interaction energy parameter χN (which incorporates the effect of added solvant) at which the transition from the lamellar ordered state to the disordered state occurs is roughly equal to 18-21. While this result is consistent with an extrapolation of the Fredrickson-Helfand weak-segragation theory to N values in the range of the simulations, the amplitude of the sinusoidal compositional wave in the ordered state near the transition is large for all N studied, in disagreement with the weak segregation theories. Thus, for values of N up to 48, the transition occurs in a “moderate,” rather than weak-segregation regime. Near the disordering transition, fluctuating “bridge” or “hole” defects in the lamellae spontaneously appear; with heating these proliferate and lead to the disordering transition. These fluctuating bridges might help explain anomalous diffusion and rheological behavior observed near the disordering transition. We also find that in the ordered state near the transition, the orientational order parameter, which is proportional to the intrinsic birefringence, falls rapidly with increasing N, roughly as 1.5 N-2.  相似文献

The significance of searching-image behaviour by predators in relation to visually polymorphic prey is outlined in relation to its possible role in maintaining these polymorphisms by frequency-dependent selection. A simple experimental design is described whereby innate preferences, selection for crypsis, and searching-image behaviour can be distinguished. Four experimental situations are described ranging from an artificial predator-prey relationship (humans as predators—dyed toothpicks as prey) to a semi-natural one (wild song thrushes as predators—artificial populations of live polymorphic snails as prey). The results suggest that “naive” predators may regularly form searching-images for a frequent prey type.  相似文献

The relation between the molecular interaction of colloidal particles in solution and the formation of an anisotropic phase is examined in two special cases. A graphical method is presented which allows the prediction of phase resetting curves of circadian rhythms for both type 1 and type 0 resetting, starting from one experimentally determined phase resetting curve. Calculations were based on literature data for the pupal eclosion rhythm of Drosophila pseudoobscura. The method is based on the assumption that for all practical purposes the rhythm may be approached as a “simple clock”, i.e. an oscillator with only one state variable, namely its phase or circadian time, CT. Besides predicting both “types” of phase resetting the method is capable to locate the “position” of the phase singularity and thus the transition from type 1 to type 0 resetting. This graphical method is an elaboration of the “transformation method”, developed in 1972 by A. Johnsson and H. G. Karlsson, which was effective in predicting phase resetting by “strong” stimuli, but failed in the case of “weak” stimuli. Predictions made using the extended transformation method are in good agreement with experimental results obtained with Drosophila. Also for the flesh fly, Sarcophaga argyrostoma, a prediction is made of the position of the phase singularity of the eclosion rhythm and compared with experimental results.  相似文献

Appetite control, pubarchal timing, and catch-up growth all seem to depend, to some extent, on body mass. Information regarding this variable may be supplied to the central nervous system (“ponderostat”) by a humoral radar system employing an insulin-like molecule or dependent substrate as a signal. The intensity of this signal would vary as a function of the organism's total adipose cell surface area, which, in turn, is a product of cell size and number. The geometry of a sphere (adipose cell) is such that surface area change per unit volume change increases as the radius of the sphere is reduced. Signal shifts then are maximal when cell volume is minimal. Divergence of the adipose signal from an age-appropriate norm could influence “ponderostat” activity and, in turn, brain centers mediating appetitive and various endocrine systems. Total caloric intake and the timing of this intake with respect to somatic maturity appear to affect adipose cell volume and number respectively. If shifts in adipose surface area provide a signal source regarding organismic mass, and if this surface area reflects the quantity and chronology of antecedent nutritional events, there are obvious implications for the role of nutrition during “critical periods” in the subsequent operation of the complex systems subserving appetite, somatic growth and sexual maturation.  相似文献

Abstract The wing surfaces of 97 insect species from virtually all relevant major groups were examined by high resolution scanning-electron-microscopy, in order to identify the relationships between the wing microstructures, their wettability with water and their behaviour under the influence of contamination. Isolated wings with contact angles between 31.6° and 155.5° were artificially contaminated with silicate dusts and subsequently fogged until drops of water (“dew”) formed and rolled off. The remaining particles were counted via a digital image analysis system. Remaining particle values between 0.41% and 103% were determined in comparison with unfogged controls. Some insects with very unwettable wings show a highly significant “self-cleaning” effect under the influence of rain or dew. Detailed analysis revealed that there is a correlation between the wettability and the “SM Index” (quotient of wing surface/(body mass)0.67) with values ranging from 2.42 to 57.0. Furthermore, there is a correlation between the “self-cleaning” effect and the SM Index, meaning that taxa with a high SM Index, e.g. “large-winged” Ephemeroptera, Odonata, Planipennia, and many Lepidoptera, have very unwettable wings and show high particle removal due to dripping water drops. The “small-winged” insects, such as Diptera and Hymenoptera, and insects with elytra, such as Blattariae, Saltatoria, Heteroptera and Coleoptera, show completely opposite effects. This is clearly a result of the fact that species with a high SM Index are, in principle, more restricted in flight by contamination than species with a low SM Index which can also actively clean their own wings. The wings primarily serve a protection function in insects with elytra, so that the effects of contamination are probably of minor importance in these insects. An education reform policy and inclusive education policy have been implemented in Hong Kong for over a decade. As more students with special educational needs have entered the mainstream education system under these policies, Hong Kong's primary music classrooms offer a site where three policies interact—the education reform policy entitled “Learning to Learn,” the policy of inclusive education, and the undeclared “policy” of making savings in the government budget. This article seeks to explore the results of the interaction of these three policies. A qualitative study was carried out to investigate the views of Hong Kong primary school music teachers on the policy of inclusion in relation to music teaching. Insufficient support in inclusive learning in “non-core” subjects, such as music, is evidenced.  相似文献
