Vitamin B6 induces in vivo as well as in vitro the appearance of a puff at region 2–48C in Drosophila hydei. At concentrations of 10–2 M or lower, region 2–48C is the only region responding to vitamin B6 provided that oligomycin is present in the incubation medium. Pyridoxal phosphate and pyridoxamine phosphate supplied to medium containing oligomycin induce upon incubation of salivary glands a larger puff at 2–48C. — Puff 2–48C produces large quantities of a unique RNP-product; globular 140–220 Å particles which aggregate to stable complexes of 0.1–0.2 in diameter. Upon continuous in vitro incubation with vitamin B6, puff 2–48C becomes loaded with these aggregates which have never been observed in any other puff of Drosophila hydei.  相似文献

It is reported that chromatin can be prepared from highly purified polytene nuclei from the salivary glands of third instar larvae of Drosophila hydei; such chromatin differs from that of diploid nuclei mainly by deficiencies in certain nonhistone chromosomal proteins. It is suggested that these proteins are important components of constitutive heterochromatin, which is severely underrepresented in polytene chromosomes. Chromosome morphology, including the pattern of induced puffs, is maintained throughout the mass isolation of glands and sucrose gradient purification of nuclei, as indicated by studies on temperature-shocked and control larvae. No significant alteration in the chromosomal proteins following puff induction by heat shock could be detected on analysis of the isolated protein fractions by disc gel electrophoresis. More sensitive techniques must be developed to study the apparent rearrangement or accumulation of protein at puff sites, and to elucidate the role of this protein in gene activation.  相似文献

Summary In Drosophila hydei abnormal puffing activities could be induced by temperature shocks and treatments with 1.2% and 1.4% KCl solutions. After temperature shocks in vivo and in vitro, a number of puffs showed a similar change in activity.Other puffs were found to show a change in activity only after a distinct treatment.Some of the puffs, specific for temperature shocks, showed a change in activity only at a distinct stage of development.In discussing the results, particular attention is paid to puffs observed in common after all treatments.Dedicated to Prof. H. The banding pattern of the salivary gland chromosomes of D. hydei was investigated in the electron microscope. We compared the banding pattern of squashed chromosomes with non-squashed preparations and observed that the fixation and squash procedure we used does not introduce artificial changes in the banding pattern of the chromosome. An electron microscopic map was made of the banding pattern of the distal half of the second salivary gland chromosome. On the basis of the number of bands in this part of the second chromosome we calculated a total of about 3700 bands for the whole set of polytene chromosomes of D. hydei. Our data indicate a similar number of bands in the salivary gland chromosomes of evolutionary remote Drosophila species like D. hydei and D. melanogaster.  相似文献

Surface-spread polytene (SSP) chromosomes of salivary glands from late third-instar larvae were used for the construction of an electron microscopic (EM) photo map of the entire genome of D. hydei. In comparison with the light microscopic chromosome map of Berendes (1963), based on squash preparations, the EM micrographs depict some 40%–50% more bands. — Two different types of chromosome constrictions are described. Differential staining of the core and RNP particles of RNP complexes in puff 2–48 BC in salivary gland chromosomes of Drosophila hydei was achieved with aqueous uranyl acetate (UA) at low pH, with UA in acetone, with phosphotungstic acid (PTA) in organic solvents, and with aqueous PTA at pH 5 and 6. A comparison of the results of UA and PTA staining under various conditions indicate that the proteins in the core region and in the RNP particles connected to it differ with respect to their amino-acid composition (arginine and lysine residues). — The staining mechanism of PTA and UA is discussed.  相似文献

Five hours after the injection of glucosamine into the hemolymph of Drosophila virilis larvae, puff 55E in the salivary gland chromosomes is significantly reduced in its activity. This observation in connection with the circumstances under which the activity of puff 55E decreases during normal development led to the proposition that its activity is involved in mucoprotein synthesis in the salivary glands during the third larval instar.
