Genetic structure and differentiation among four Lethenteron taxa, L. japonicum, L. kessleri, and two groups of L. reissneri, collected from Japan and the Far Eastern region of Russia, were investigated by electrophoretic analysis. Several complete-allele substitutions were found between all possible pairs of taxa in regions of sympatry, strongly suggesting the existence of reproductive isolation between them. Therefore, four Lethenteron taxa should be regarded as discrete species, respectively. In each taxon, the genetic variability within each population (H=0.062 – 0.127 for L. japonicum, H=0.062– 0.128 for L. kessleri, H=0.026 – 0.148 for the northern group of L. reissneri, and H = 0.015 – 0.102 for the southern group of L. reissneri) was considerable, suggesting large effective sizes for most populations. The sample of L. japonicum collected from Kyiya, the basin of Amur, was somewhat divergent from the other intraspecific samples. This may have resulted mainly because it does not migrate to the sea. In contrast L. kessleri has a fluvial life style, but the genetic differentiation between populations (GST=0.117, D = 0.000 – 0.061) was less than that for each group of L. reissneri (GST = 0.493, D=0.000 – 0.226 for the northern group and GST=0.660, D = 0.008 – 0.422 for the southern group), probably meaning more recent dispersal of L. kessleri than the two groups of L. reissneri. The parasitic and anadromous L. japonicum appeared to be closely related to L. kessleri (D=0.042 – 0.090) and the northern group of L. reissneri (D=0.163 – 0.355), which have nonparasitic and fluvial life styles, whereas the southern group of L. reissneri was greatly divergent from the other three Lethenteron species (D=0.559 – 0.926), suggesting that the former three species might be monophyletic.  相似文献

Ten qualitative traits were observed in two isolated human populations in Bosnia for prewar and postwar periods. Due to recent war in Bosnia and Herzegovina two million citizens was forced to migrate. Dramatic migration effects occurred in the investigated populations. Population characteristics found were geographic isolation, high levels of marital distance “zero”, patrilocality and propagation isolation. Relative recessive allele frequencies of ten qualitative traits were calculated. Average heterozigosity, genetic differentiation, total heterozigosity, within populations heterozigosity, genetic distance and relative measures based on results of genetic distance analysis were observed. The total number of observed individuals was 1875. We have observed 1209 individuals in prewar period (Bijela population 731 and Memici population 478) and 666 individuals in the postwar period (Bijela population 248 and Memici population 478). Results ofFst andGst analysis showed changed degree of genetic differentiation across observed loci. Significant changes in differentiation were recorded for crooked little finger and PTC tasting. Modest changes of gene differentiation were recorded for ear lobe type and thumb proximal extensibility. Genetic distance analisis between Bijela and Memici has lower value after then before the war, but genetic distance between Bijela-prewar and Bijela-postwar has higher value, as well as Memici-prewar and Memici-postwar genetic distance. Genetic distance between Bijela-prewar and Memiciprewar, as well as Bijela-postwar and Memici-postwar showed similarity. Constructed dendrogram based on results of genetic distance analyses indicated two clusters groups (1. Bijela-prewar, Memici-prewar; 2. Bijela-postwar, Memici-postwar). Changed genetic differentiation and results of genetic distance analyses indicated possibility of significant impact of forced migration in genetic structure of observed populations.  相似文献

Blood protein polymorphism of gelada baboon (Theropithecus gelada) to the south of the Rift Valley, Arsi Region, were examined for 36 genetic loci using three electrophoresis techniques for 48 blood samples from three localities, and compared with the northern geladas. New variant alleles and genetic markers of Hb-α, PA-2, and TBPA loci were detected. The distribution patterns of the variant alleles of Hb-α, PA-2, TBPA, Pi, Gc, PGM-II, and TBPA loci were localized in the geographic regions of south and north gelada populations, respectively. Genetic variability of southern geladas was estimated as Ppoly=0.083 and , which was comparable to northern geladas. A remarkably high genetic differentiation between the two geographic populations was shown byNei's genetic distance=0.071 and GST value=0.420. Our results of genetic analysis suggest that the southern and northern gelada populations have been separated for several hundred thousand years, and gene flow between the two geographic populations is severely restricted. The southern gelada baboon may be regarded as a distinct subspecies.  相似文献

Habitat fragmentation may influence the genetic make-up and adaptability of endangered populations. To facilitate genetic monitoring of the endangered European ground squirrel (EGS), we analyzed 382 individuals from 16 populations in Central Europe, covering almost half of its natural range. We tested how fragmentation affects the genetic architecture of presumably selectively neutral (12 microsatellites) and non-neutral (the major histocompatibility class II DRB gene) loci. Spatial genetic analyses defined two groups of populations, “western” and “eastern”, with a significantly higher level of habitat fragmentation in the former group. The highly fragmented western populations had significantly lower genetic diversity in both types of markers. Only one allele of the DRB gene predominated in populations of the western group, while four alleles were evenly distributed across the eastern populations. Coefficient of inbreeding values (F IS) calculated from microsatellites were significantly higher in the western (0.27–0.79) than in eastern populations (−0.060–0.119). Inter-population differentiation was very high, but similar in both groups (western F ST = 0.23, eastern F ST = 0.25). The test of isolation by distance was significant for the whole dataset, as well as for the two groups analyzed separately. Comparison of genetic variability and structure on microsatellites and the DRB gene does not provide any evidence for contemporary selection on MHC genes. We suggest that genetic drift in small bottlenecked and fragmented populations may overact the role of balancing selection. Based on the resulting risk of inbreeding depression in the western populations, we support population management by crossbreeding between the western and eastern populations.  相似文献

The Egyptian vulture (Neophron percnopterus) is a species in decline throughout Europe, with the largest remaining breeding populations found in northern Spain. Iberian Peninsula populations of this species (about 1000 pairs) migrate to Africa in winter, while small populations in both the Canary and Balearic Islands (less than 40 pairs in each case) are apparently sedentary. We found that Egyptian vultures from both of these island groups were significantly differentiated from Iberian Peninsula populations (R ST = 0.065–0.129, p = 0.000–0.007), using nine microsatellite loci isolated in a related species, the bearded vulture. The greatest degree of genetic differentiation was observed between the two island groups (R ST = 0.279, p = 0.000). These island populations were more distinct from mainland groups than was a small sample of a well-defined separate subspecies from India (N. p. ginginianus; R ST = 0.083–0.091, p = 0.023–0.024). This implies that these two island populations have been isolated from peninsular populations for many generations, despite the long-distance migration capabilities of the species. In constrast, populations within the Peninsula were not differentiated from one another at these microsatellite loci (R ST = $-$0.004–0.007, p = 0.442–0.675). Introductions of Egyptian vultures from the larger northern breeding groups might therefore be appropriate in southern Spain, if necessary, but mainland birds should not be introduced to the islands if the genetic distinctiveness of these groups is to be preserved. Independent conservation plans are urgently required to protect these two island populations from extinction. We investigated the phylogeny and phylogeography of four closely related Ainsliaea species (Asteraceae) on the continental Ryukyu Islands of eastern Asia, which consist of two flood-adapted “rheophyte” and two non-rheophyte (inland) species, based on 12 nuclear microsatellite loci. Phylogenetic analyses using 420 individuals from 26 populations showed that rheophytic A. linearis and A. oblonga are genetically distinct. Each species was clustered with the inland species that occur on the same islands, suggesting a different ancestry for the two rheophytes that evolved independently by local adaptation to flooded habitats. The results from the neighbor-joining clustering and principal coordinate analysis (PCoA) indicate that the southern populations of A. macroclinidioides are distinct lineages and ancestral to the northern populations as well as the other diverse species complex in the Ryukyus. These results suggest a pattern of colonization initially from the Asian mainland to the southern islands, followed by the northern islands via land bridges generated during the Quaternary glaciations. After isolation from southern populations, species radiation and regional differentiation within the northern clade occurred possibly via local adaptation and/or geographic isolation of the subdivided island bridge.  相似文献

Abstract Carex misera is a rare sedge, endemic to rocky outcrops and mountain summits within the southern Appalachian Mountains from northern Georgia to northern North Carolina. We assessed allozyme diversity for 406 individuals from nine populations over most of the geographic range. Twenty-seven putative loci were assayed and eight (30%) were found to be polymorphic. Nei's gene diversity statistics ( H T =0.043, H S =0.019, G ST =0.551) indicated low levels of variation but relatively highly differentiated populations, suggesting little gene flow. Significant deviations from genotypic expectations under Hardy-Weinberg equilibrium, high positive fixation indices, and the existence of small genetic neighborhoods within populations suggest that at least some inbreeding occurs. Cluster analysis of Nei's genetic identity statistics and principal component analysis of allele frequency data showed high similarity among the six southern populations with the two northern populations more differentiated from them and from each other. These results suggest that preservation of the northern populations is necessary to conserve the already low levels of genetic diversity within the species.  相似文献

Genetic variability within and between populations of the wild progenitor of barley was studied electrophoretically. Thirty enzyme loci were assayed in 437 individuals representing 11 populations ofH. spontaneum in Turkey. The results indicated that: (a)H. spontaneum in Turkey is genetically rich in allozyme variation, but because of predominant self-pollination the variation is maintained as different homozygotes in the population; (b) genetic differentiation of populations includes clinal, regional, and local patterns, sometimes displaying sharp geographic differentiation over short distances; (c) overall indices of allozymic diversity and some allele frequencies of wild barley are significantly correlated with the environment and are predictable ecologically, chiefly by combinations of temperature and humidity variables; (d) a high percentage of alleles (66%) occur in local areas or are distributed sporadically rather than widespread; (e) Wright Fixation index was very high, F=0.995; (f) genetic distance was high (D=0.11, ranging from 0.031 to 0.288) between populations, and (g) average relative genetic differentiation was high among populations (Gst=0.47, ranging from 0.02 to 0.66). The spatial patterns and environmental correlates and predictors of genetic variation ofH. spontaneum in Turkey, indicated that genetic variation in wild barley populations is not only common, but also at least partly, adaptive. Therefore, a much fuller exploitation of these genetic resources by breeding is warranted.  相似文献

Allozyme diversity was examined in 30 populations of the maritime perennial plant Hedyotis strigulosa var. parviflora , which is distributed from subtropical islands to the central mainland of Japan. Genetic diversity within populations tended to be larger in southern island populations than in northern mainland populations. In the southern part of the distribution, the population size is generally large and populations are distributed more continuously than in the northern area, resulting in the larger effective size of southern populations as a whole. These factors play a major role in maintaining greater genetic diversity in the southern populations. By contrast, genetic diversity in the northern populations is very low, probably resulting from bottlenecks of population establishments during recolonization from refugial area to the northern areas. Geographically close populations were located near each other in the multidimensional scaling and the phenogram based on genetic distances, suggesting that gene flow among remote populations is rather limited. The pattern of genetic diversity in H. strigulosa var. parviflora is likely caused by the distribution expansion of the species; in the last glacier era, the species was restricted to the southern area; its advance to the northern area is relatively recent. The coastal deserts of northern Chile show an important latitudinal gradient of aridity with more arid areas to the north of the Atacama Desert than to the south. Several plant species have disjunct distributions that correspond with the extremes of this latitudinal gradient. In this study, using genetic (chloroplast and nuclear DNA), morphological (vegetative and floral traits of various kinds) and climatic and topographic information, we explored ecological and historical events that have putatively shaped patterns of variation among Monttea chilensis populations—a species that shows this disjunct distribution. Through phylogeographic and phylogenetic analyses, two divergent lineages were identified located at the latitudinal extremes. The lineage located northern lineage (NG) of the Atacama Desert showed more genetic diversity and better-resolved phylogeographic structure than the southern lineage (SG). Considerable morphological variation across the geographical range corresponds with these genetic groups. We observed contrasting relationships between floral and vegetative traits: populations from the most arid region NG possessed larger flowers, but smaller vegetative values, and vice versa. Niche modelling and multivariate analyses, including environmental data, revealed different environmental requirements for each lineage. NG plants occur in regions with warmer and drier climatic conditions and at higher altitudes, while SG populations inhabit colder and more humid environments and lower altitudes. The evolutionary history of M. chilensis exhibits a phylogeographical footprint consistent with past fragmentation and allopatric differentiation, where the hyper-arid zone formed by the Atacama Desert clearly acted as an important gene flow barrier. This barrier has led to considerable differentiation in morphology and ecology, resulting in two ecotypes or geographical races, suggesting incipient speciation promoted by local adaptation and geographical isolation.  相似文献

The present study investigates the genetic structure of 12 roe deerCapreolus capreolus Linnaeus, 1758 population samples from Serbia, by screening a total of 334 individuals. We examined whether genetic differentiation exists in local populations in Serbia, and addressed the question whether management policies may affect genetic structure. The populations were analysed by multilocus protein electrophoresis, with 33 protein loci examined. Screening of 20 enzymes and one group of general proteins revealed polymorphism at the following 12 loci: Sdh, Mdh-1, Me-1, Idh-2, 6-Pgd-1,αGpd, Ak, Pgm-1, Pgm-2, Ca, Mpi andGpi. Among samples, the proportion of polymorphic loci varied between 3–15.2% (mean 11.9%), while the average gene diversity was in the range of 1.1–4.2%. The overall genetic differentiation was low (θ = 0.03). The comparison of two regional population groups (northern-southern, separated by the Danube River) showed an absence of genetic differentiation between regions. Gene flow was estimated at 8.96 migrants per generation, and was higher in the lowland than in the highland group. Three loci (Ca, 6-Pgd andGpd-1) showed clinal variation along a geographical gradient. Additional five alleles of four loci (Ak, Pgm-1, Gpi, 6-Pgd) showed significant spatial autocorrelation. Genetic distances were small (D = 0–0.004). Northern and southern populations clustered separately. For at least three populations game management practices provide evidence for outlying genetic parameters. The observed heterogeneity in the inbreeding level was deemed more under the influence of non-random mating strengthened by game management, than by overall selective pressure.  相似文献

The genetic variation of brown trout from Duero, one of the main Atlantic Iberian river basins, was assessed at 34 enzymatic loci in 62 native populations. A strong intrabasin differentiation was detected (G(ST) = 0.46; range D: 0-0.066), mainly attributable to the existence of two divergent groups of populations within Duero: southern and northern groups. This divergence was mainly a consequence of the unequal distribution of *75 and *100 alleles at sMDH-B1,2* isoloci, which were correlated with substantial differences in genetic diversity among regions. The Lower Course region (nearly fixed for the *100 allele) and Pisuerga River (nearly fixed for the *75 allele) showed lower heterozygosities (H approximately 0.8%) in contrast with adjacent areas, which evidenced intermediate frequencies for both alleles and higher heterozygosities (H: 2.2-3.1%). Vicariance appeared as the more probable explanation for the significant positive correlation detected between genetic and geographical distances in Duero Basin. Genetic relationships with adjacent Iberian drainages indicate a close similarity between the southern group and Cantabric trout, whereas the northern group constitutes an ancient form from this basin. This study confirmed complex genetic relationships in brown trout from northwest Iberia, reasserting the existence of clines at several loci and for genetic diversity. The interaction between Cantabric and Duero trout, as well as the location of the limit of the anadromous form around the 42 degrees N parallel, are both required to understand the genetic characteristics of brown trout from this area.  相似文献

The Atlantic sand fiddler crab Uca pugilator is an extremely abundant crab found along the eastern coast of the United States. Fiddler crabs have a life cycle with an obligatory planktonic larval phase of 30-90 days, which might be expected to lead to widespread larval dispersal and consequent genetic homogeneity over considerable distances. However, a large amount of morphological and behavioral variation is found between northern and southern populations along the eastern coast. This study was undertaken to determine the population genetic structure of U.pugilator and to determine whether these differences may have a genetic basis. The population structure of the fiddler crab was analyzed using 472 individuals collected from 12 sites along the eastern coast. PCR-based single stand conformation polymorphism (SSCP) was used to investigate between-site variation in the mitochondrial 16S rRNA gene of these individuals. Analysis of genetic variation indicated frequent gene flow between nearby localities, but much reduced levels between populations separated by larger geographic distances. Thus, despite the potential for high dispersal by planktonic larvae, population differentiation and isolation by distance is evident between northern and southern populations of U.pugilator. A high amount of genetic differentiation (FST=0.3468) was found between northern and southern regions suggesting that the morphological and behavioral differences between these two regions have a genetic basis and may represent subspecies [Current Zoology 55(2):150-157,2009].  相似文献

In order to elucidate genetic composition of European grayling (Thymallus thymallus) populations in the Western Balkans, the partial mitochondrial DNA (mtDNA) control region was sequenced and 12 microsatellite loci genotyped in 14 populations originating from tributaries of the Adriatic and Danube drainages. Eleven mtDNA haplotypes were found, one confined to the Adriatic clade, one to the Alpine group and the rest to the ‘Balkan’ grayling phylogenetic clade. Haplotypes from the Balkan clade were confined to the Danube drainage and constituted two groups: northern group with haplotypes found in the Slovenian part of the Danube drainage, and southern group, consisting from Bosnia–Herzegovina and Montenegro. Substantial genetic distance between northern and southern groups of haplotypes (0.75–1.8%) and well supported divisions within the northern group indicate very structured grayling population within the studied Danube basin that most probably did not evolve due to vicariance but rather as a consequence of multiple colonization waves that might have occurred during the Pleistocene. Furthermore, genetic distance of ~4% between Adriatic and Danube populations’ haplotypes, suggest that their separation occurred in mid-Pliocene. These findings imply a complex colonization pattern of the Western Balkans drainages. Microsatellite data also confirm high genetic diversity in Western Balkans populations of grayling (on average 7.5 alleles per microsatellite locus and H exp 0.58). Limited stocking activities were detected based on microsatellites and mtDNA data. Regarding current knowledge of grayling phylogeography appropriate management strategies were proposed to preserve unique, autochthonous grayling populations in Western Balkan.  相似文献

The phylogenetic relationships among 23 local sheep breeds and varieties in East Asia were determined based on allele frequency data for five informative and polymorphic blood protein and nonprotein loci (transferrin, arylesterase, hemoglobin-β, X-protein, and potassium transport) using electrophoresis and ion-densitometric techniques. Comparatively striking differences in the frequency distributions, especially of hemoglobin-β and X-protein alleles, were seen between the northern population of the Khalkhas, Bhyanglung, Baruwal, Jakar, Sakten, and five Chinese breeds and the southern population of the Bengal, Kagi, Lampuchhre, Myanmar, and Sipsu breeds. Clustering analyses using UPGMA and NJ methods and principal component analysis, using the data of all populations and loci tested, clearly demonstrated the difference. The two population groups are divided by the boundary of the Himalayas, and each diverged into three subgroups: Mongolian, Tibetan, and Himalayan groups in the north and Indian groups I, II, and III in the south. It was noted that the genetic differentiation of populations was more distinct in the northern group. These findings strongly suggest the existence of at least two large and phylogenetically different gene pools of sheep in East Asia.  相似文献

We use a combination of microsatellite marker analysis and mate-choice behavior experiments to assess patterns of reproductive isolation of the túngara frog Physalaemus pustulosus along a 550-km transect of 25 populations in Costa Rica and Panama. Earlier studies using allozymes and mitochondrial DNA defined two genetic groups of túngara frogs, one ranging from Mexico to northern Costa Rica (northern group), the second ranging from Panama to northern South America (southern group). Our more fine-scale survey also shows that the northern and southern túngara frogs are genetically different and geographically separated by a gap in the distribution in central Pacific Costa Rica. Genetic differences among populations are highly correlated with geographic distances. Temporal call parameters differed among populations as well as between genetic groups. Differences in calls were explained better by geographic distance than by genetic distance. Phonotaxis experiments showed that females preferred calls of males from their own populations over calls of males from other populations in about two-thirds to three-fourths of the contrasts tested. In mating experiments, females and males from the same group and females from the north with males from the south produced nests and tadpoles. In contrast, females from the south did not produce nests or tadpoles with males from the north. Thus, northern and southern túngara frogs have diverged both genetically and bioacoustically. There is evidence for some prezygotic isolation due to differences in mate recognition and fertilization success, but such isolation is hardly complete. Our results support the general observation that significant differences in sexual signals are often not correlated with strong genetic differentiation.  相似文献

Plants with natural variation in their floral traits and reproductive ecology are ideal subjects for analyzing the effects of natural selection and other evolutionary forces on genetic structure of natural populations. Agave lechuguilla shows latitudinal changes in floral morphology, color, and nectar production along its distribution through north-central Mexico. Both the type and abundance of its pollinators also change with latitude. Using starch electrophoresis, we examined the levels and patterns of variation of 13 polymorphic allozyme loci in 11 populations of A. lechuguilla. The overall level of genetic variability was high (H(e) = 0.394), but the levels of genetic variation had no geographic pattern. However, the southern populations exhibited an excess of heterozygotes in relation to expectations for Hardy-Weinberg equilibrium, whereas the northern populations had an excess of homozygotes. Total differentiation among populations was low (θ = 0.083), although gene flow estimates (Nm) varied among groups of populations: southern populations had the lowest levels of genetic differentiation, suggesting high levels of gene flow; northern populations had greater levels of genetic differentiation (θ = 0.115), suggesting low gene flow among them. The patterns and inferences of the genetic structure of the population at the molecular level is consistent with variation in floral traits and pollinator visitation rates across the range of the species.  相似文献

A survey of genetic variation of lotic breeding salamander Hynobius naevius by starch gel electrophoresis using 26 populations from southwestern Japan revealed the presence of two genetically distinct groups (GROUP-A and B) which overlap distributional ranges in some parts of Kyushu. Degree of genetic differentiation between the two groups was larger in Kyushu, including sympatric area, than in allopatric areas. We surmise that the two groups first differentiated allopatrically and overlapped their distributions secondarily. The first genetic differentiation is considered to have occurred in the Pliocene as a result of isolation between northern (GROUP-A) and southern (GROUP-B) ancestral populations by the formation of the Seto Inland Sea, largely separating the present distributions of the two groups. Genetic diversities within the GROUP-A seem to have become complicated by isolations, disappearances of local populations, and subsequent range expansion of distributions in Chugoku and northern half of Kyushu regions. Within the GROUP-B, diversities first formed by isolation by distance have been retained and accelerated secondarily by the formations of the Kii and Bungo straits. Sympatric populations of the two groups in northern half of Kyushu seem to have overlapped their ranges by niche segregation.  相似文献

Aim  The aim of this study is to analyse the genetic population structure of Meum athamanticum Jacq. in order to explore the alternative hypotheses (1) that the central and northern highland populations are the result of post-glacial recolonization from southern refugia, and the disjunct distribution of M. athamanticum can be explained by modern ecological conditions, or (2) that extant populations north of the Alps and Pyrenees persisted in situ during glacial periods.
Location  Europe.
Methods  Variation of amplified fragment length polymorphisms (AFLPs) was analysed for 23 populations from the entire range of the species. We used band-based approaches and methods based on allele frequencies to measure genetic diversity and to identify population structure.
Results  Our analyses reveal a north–south differentiation within M. athamanticum . High levels of genetic diversity, as well as private fragments, are found in populations both north and south of the Alps. Differentiation among populations is lower in the northern than in the southern population group, and significant isolation-by-distance is found only in the latter group.
Main conclusions  Our results indicate that M. athamanticum survived the last ice age in multiple refugia throughout its contemporary range and did not expand into areas north of the Alps from southern refugia. We found evidence that regional-scale migration in northern, formerly periglacial, parts of the species range has resulted in the intermingling of populations. In contrast, southern populations are characterized by long-term isolation. The south-west Alps represent an area where immigration and mixing of populations from northern and southern refugia appears to have taken place.  相似文献
