The influence of tillage practices on native arbuscular mycorrhizal fungi (AMF) was studied in two, consecutive years in eastern Canada, in two 11 year-old long-term tillage-fertilizer experimental field soils, a sandy loam and a clay, growing corn in monoculture. The three tillage practices were: 1) conventional tillage (CT; fall plowing plus spring disking), reduced tillage (RT; spring disking) and no-till (NT). The corn crop received either inorganic (N and K) or organic (liquid dairy manure) fertilizers. Mycorrhizal hyphal density was estimated from soil samples obtained in early spring (before disking), at the 12–14 leaf stage, at silking, and at harvest. The percentage of corn root colonization by AMF at the 12–14 leaf stage, at silking and at harvest was also determined. The sandy loam was sampled over two consecutive seasons and the clay soil over one season.Densities of total and metabolically active soil hyphae, and mycorrhizal root colonization were significantly lower in CT soil than in RT and NT soil. Lowest soil hyphal densities were observed in early spring. The levels of intra- and extraradical fungal colonization always increased from spring to silking and decreased thereafter. Spring disking had only a small and transient negative effect on hyphal abundance in soil. Fertilization did not influence mycorrhizal colonization of corn or abundance of soil hyphae in the sandy loam soil, but in the clay soil metabolically active hyphae were more abundant with manure application than with mineral fertilization. In 1992, in both soils different tillage systems had same grain yield, however, in 1993, corn yield was higher in NT compared to CT system.  相似文献

Aim:  To evaluate the effects of soil physical and chemical factors (pH, conductivity, humidity, available phosphorus and organic matter) and environmental factors (temperature, relative air humidity, altitude and atmospheric pressure) on arbuscular mycorrhizal fungi (AMF)– Brachiaria decumbens grass relationship. Furthermore to establish patterns of microbiological responses that allow to differentiate the study sites in two relief types.
Methods and Results:  Mycorrhizal characteristics (spore density, external hyphae and root colonizations by hyphae, vesicles and arbuscules), physical and chemical factors in soil and environmental factors were measured.
Conclusions:  The effect of physical, chemical and environmental factors on microbiological variables was related to the type of relief 'valley and hilly terrain'; the AMF behaviour was affected only over narrower ranges of evaluated variables. Similarly, the colonization of B. decumbens roots by AMF hyphae, vesicles and the mycorrhizal spore density follow different patterns according to the relief type.
Significance and Impact of the Study:  The type of relief is one of the factors to be taken into consideration to evaluate the AMF inoculum and root colonization of these pastures, because of the influence of slope – as physical property of soil – on AMF.  相似文献

Prunella vulgaris was inoculated with different arbuscular mycorrhizal fungi (AMF) and grown at two concentrations of CO2 (ambient, 350 μl l−1, and elevated, 600 μl l−1) to test whether a plants response to elevated CO2 is dependent on the species of AMF colonizing the roots. Using compartments accessible only to AMF hyphae but not to roots, we also tested whether elevated CO2 affects the growth of external AMF hyphae. Plant biomass was significantly greater at elevated than at ambient CO2; the biomass of the root system, for example, increased by a factor of 2. The colonization of AMF inside the root remained constant, indicating that the total AMF inside the root system also increased by a factor of 2. The length of external AMF hyphae at elevated CO2 was up to 5 times that at ambient CO2, indicating that elevated CO2 promoted allocation of AMF biomass to the external hyphae. The concentration and content of phosphorus in the stolons differed significantly between ambient and elevated CO2 but this resulted in either an increase or a decrease, according to which AMF isolate occupied the roots. We hypothesized that an increase in external hyphal growth at elevated CO2 would result in increased P acquistion by the plant. To test this we supplied phosphorus, in a compartment only accessible to AMF hyphae. Plants did not acquire more phosphorus at elevated CO2 when phosphorus was added to this compartment. Large increases in AMF hyphal growth could, however, play a significant role in the movement of fixed carbon to the soil and increase soil aggregation. Arbuscular mycorrhizal fungi (AMF) enhance nutrient provision in exchange for carbon. However, a shift from nutrient to carbon limitation, induced by repeated or intense defoliation, can represent a trade-off between plant regrowth and the maintenance of mycorrhiza. The combined effects of AMF, phosphorus and defoliation on growth of Agropyron elongatum (C3 grass, low mycorrhizal responsiveness) and Brachiaria brizantha (C4 grass, high mycorrhizal responsiveness) were explored. Each species was subjected to a manipulative experiment with AMF inoculation (non-inoculated, inoculated), soluble P supply (low, high) and defoliation (non-defoliated, 60% defoliated). In A. elongatum, at low P supply, mycorrhizal plants showed increased growth rates following defoliation without substantial changes in AMF colonization. At high P supply instead, we found a clear trade-off between regrowth and the maintenance of mycorrhiza evidenced by growth depression (biomass and tillers) and lower AMF activity (reduction of arbuscules). In contrast, in B. brizantha, defoliation effects on plant regrowth were independent from AMF at any P supply (no trade-off). This indicates that cost-benefit relationship in defoliated plants is highly context-dependent and may vary with mycorrhizal responsiveness of species. This variation of responses can play a decisive role on plant recovery in pastures and natural grasslands subjected to herbivory.  相似文献

研究了在东北海伦实验站长期定位培肥实验地不同施肥处理下丛枝菌根(Arbuscular Mycorrhizal,AM)真菌生长发育状况(包括侵染率、菌丝量和孢子数),同时还分析了不同施肥处理下AM真菌群落生态分布和特征.结果表明施肥处理,尤其是磷肥(NP2K)处理显著降低AM真菌侵染玉米根系,而根外菌丝长度和孢子数并无显著变化,这和施肥处理下AM真菌的种群结构发生变化有关.随着土壤肥力的增高,土壤中AM真菌种的丰度和密度都有增加的趋势,而当肥力增高到一定程度后(磷肥和钾肥继续增加到NP2K和NPK2处理后),土壤中AM真菌种的丰度和密度都有下降的趋势;从AM真菌属在不同肥力处理下出现的频度来看,Glomus属在7个处理中出现的频度最高,在每一个肥力处理中都有分布,Acaullospora属次之,Entrohospora属则只是出现在NK处理下;而Glomus属中出现频度最高的种是Glomus mosseae,其次是Glomus caledonium;再次是Glomus diaphanium,这说明施肥处理会影响到AM真菌种属的分布,进而影响到AM真菌的群落结构和生态分布.  相似文献

A study was conducted to evaluate the effect of N and P supply levels on mycorrhizal formation and nutrient uptake in corn hybrids with different architectures and to determine arbuscular mycorrhizal fungal (AMF) development in relation to shoot N/P ratio and shoot:root ratio. Corn pot cultures with a pasteurized medium of two parts sand and one part sandy loam soil were grown in the greenhouse. Marigold plants inoculated or not with Glomus intraradices Schenck & Smith were used to establish an AMF hyphal network in the designated soil pots. Corn hybrids were seeded after removal of the marigold plant. Mycorrhizal colonization of corn hybrids and the quantity of extraradical hyphae produced in soil were greatest at the lowest P level and at the intermediate N level. Root colonization was correlated with shoot N/P ratio only at the intermediate N level. The shoot concentrations of P, Mg, Zn and Cu were significantly higher in mycorrhizal plants than in non-mycorrhizal plants. The corn phenotype with the highest shoot:root ratio had the highest root colonization. The corn hybrid with a leafy normal stature architecture had a greater mycorrhizal colonization than that of other two corn hybrids. This experiment showed that N level in soil influenced shoot N/P ratio, root colonization and extraradical hyphal production, which in turn influenced uptake of other nutrients. Mycorrhizal symbiosis can modify plant response to drying soil, but little is known about the relative contribution of soil vs. root hyphal colonization to drought resistance of mycorrhizal plants. Foliar dehydration tolerance, characterized as leaf and soil water potential at the end of a lethal drying episode, was measured in bean plants (Phaseolus vulgaris) colonized by Glomus intraradices or by a mix of arbuscular mycorrhizal fungi collected from a semi-arid grassland. Path analysis modeling was used to evaluate how colonization rates and other variables affected these lethal values. Of several plant and soil characteristics tested, variation in dehydration tolerance was best explained by soil hyphal density. Soil hyphal colonization had larger direct and total effects on both lethal leaf water potential and soil water potential than did root hyphal colonization, root density, soil aggregation, soil glomalin concentration, leaf phosphorus concentration or leaf osmotic potential. Plants colonized by the semi-arid mix of mycorrhizal fungi had lower lethal leaf water potential and soil water potential than plants colonized by G. intraradices. Our findings support the assertion that external, soil hyphae may play an important role in mycorrhizal influence on the water relations of host plants.  相似文献

Percent root length colonization may not be an appropriate measure of root colonization by arbuscular mycorrhizal fungi (AMF) in all cases. We suggest that AMF will differ in how well percent root length colonization measures the amount of AMF colonization in the root due to differences among AMF in hyphal structure and hyphal aggregation. Although soil hyphal length accounts for hyphal density, we suggest that it does not consider differences in hyphal structure in measurements of external colonization and thus might also misrepresent the true amount of AMF in the soil. To test these suggestions, we measured and compared percent root length colonization and soil hyphal length with root ergosterol and soil ergosterol, respectively, for 21 different species of AMF from three families in a greenhouse experiment. Percent root length colonization predicted intra-radical colonization best for Glomaceae and Acaulosporaceae isolates, while soil hyphal length best represented soil ergosterol for Gigasporaceae isolates. The results show that conventional methods for estimating AMF colonization are not universal for all AMF. Caution is advised when drawing inferences for different groups of AMF.  相似文献

Fungal colonization of litter has been described mostly in terms of fructification succession in the decomposition process or the process of fungal ligninolysis. No studies have been conducted on litter colonization by arbuscular mycorrhizal fungi (AMF) and their relationship with the presence of saprotrophic fungi. The aim of the present study was to evaluate the relationships that exist in simultaneous leaf litter colonization by AMF and saprotrophic fungi and the relationships between rates of litter and associated root colonization by AMF at different soil depths. We selected Eugenia sp. and Syzygium sp. in a riparian tropical forest, with an abundant production of litter (O horizon), we evaluated litter and root colonization at different depths, its C:N ratios, and the edaphic physico-chemical parameters of the A horizon immediately below the litter layer. Litter colonization by saprotrophic fungi and AMF increased with depth, but the saprotrophic fungal colonization of some litter fragments decreased in the lowermost level of the litter while AMF litter colonization continued to increase. Plant roots were present only in the middle and bottom layers, but their mycorrhizal colonization did not correlate with litter colonization. The external hyphae length of AMF is abundant (ca. 20 m g(-1) sample) and, in common with sample humidity, remained constant with increasing depth. We conclude that in zones of riparian tropical forest with abundant sufficient litter accumulation and abundant AMF external hyphae, the increase in litter colonization by AMF with depth correlates to the colonization by saprotrophic fungi, but their presence in the deepest layers is independent of both litter colonization by saprotrophic fungi and root colonization by AMF.  相似文献

A series of microcosm experiments was established to investigate the effects of simulated acid rain on the capacity of three arbuscular mycorrhizal fungi (AMF) to germinate and colonize two grasses, Calamagrostis villosa and Deschampsia flexuosa. These two grasses are normally found in degraded Norway spruce forests in the Northern Czech Republic where acid rain pollution exists and C. villosa initially outcompetes D. flexuosa for the same niche. An AM fungus isolated from acid soils (Acaulospora tuberculata BEG41) was more tolerant of acidification than two species of Glomus (isolated from agricultural soils of neutral pH) in microcosm studies. Different effects of simulated acid rain (SAR) were found at all stages of the development of three AMF studied in model systems, including spore germination, colonization of host roots, and alkaline phosphatase (ALP) and NADH diaphorase activity of the extraradical mycelium. No ALP activity was found in hyphae germinating from the spores without plants whereas it was detected in all hyphae linked to a functioning intraradical mycelium.Simulated acid rain also affected the mycorrhizal growth response and belowground competition of the two grasses. Disturbance of the ERM between the two plant species significantly reduced the growth of C. villosa but not D. flexuosa. Disturbance also decreased root colonization by AMF of both plants, the total length of ERM and the total length of extraradical hyphae with ALP and NADH diaphorase activity adjacent to both plants. D. flexuosa appeared less dependent on the mycorrhizal state, for shoot and root growth, than C. villosa under the experimental conditions. The ability, therefore, of C. villosa to thrive in forest stands suffering from acid rain pollution may be related to this dependence on its mycorrhizal hyphal links to D. flexuosa under the environmental conditions produced by the pollution including higher light levels.  相似文献

Mycorrhizal symbiosis is a key factor influencing aspects of grassland and savanna structure and functioning including plant growth, competition, population and community dynamics, and responses to fire and herbivory. This study assessed the effects of fire on mycorrhizal symbiosis and root system architecture (RSA) in South African savanna grasses. Eighteen grass species were sampled across contrasting fire frequency treatments in the Kruger National Park experimental burn plots. All eighteen species studied were highly colonized by arbuscular mycorrhizal fungi (AMF). Both mycorrhizal symbiosis and RSA were strongly affected by fire, with an increase in AMF colonization and a decrease in root branching and fine root development with decreasing fire frequency. Greater water limitation in frequently burned savanna may result in greater fine root development, thus reducing plant dependency on AMF for acquisition of soil resources. Reduced mycorrhizal colonization in frequently burned savanna may also be driven by higher phosphorus : nitrogen ratios, or indirect effects related to higher grazing intensities in frequently burned sites.  相似文献

对西双版纳热带次生林中13个科的26种植物根系的丛枝菌根真菌(Arbuscular mycorrhizal fongii,AMF)侵染情况进行了研究,并从这些植物的根围土壤中分离鉴定了隶属于球囊霉属(Clomus)、巨孢囊霉属(Gigaspora)、盾巨孢囊霉属(Scutellospora)和无梗囊霉属(Acaulospora)的11种丛枝菌根真菌。该地次生林中AMF的孢子密度为13—29个/100g土壤,平均为19个;种的丰富度在4.9之间(平均为6);平均频度为53.8%;相对多度为3.2%-26.5%;物种多样性指数和均匀度指数分别为0.94和0.93。丛枝菌根的侵染率达到44.8%.57.2%(平均为50.9%);球囊霉属(Glomus)和无梗囊霉属(Acattlospora)是热带次生林根围土壤中菌根真菌的优势类群。  相似文献

1.  Plant association with arbuscular mycorrhizal fungi (AMF) has been considered a factor increasing plant tolerance to herbivory. However, this positive effect could decrease with colonization density if the benefit : cost ratio of the AMF–plant association changes. We measured plant performance and tolerance to defoliation across a gradient of commercial AMF ( Glomus sp.) inoculum concentration.
2.  Six genetic families of Datura stramonium were grown under greenhouse conditions and subjected to five increasing levels of AMF inoculum concentration and to defoliation treatments, i.e. the presence/absence of 50% artificial damage, following a full-factorial design.
3.  AMF colonization increased linearly with inoculum concentration while foliar area, root mass, flowering phenology and seed production expressed nonlinear functions. Plant genetic variation in the benefit function of AMF colonization was also detected. We show a negative interaction between AMF concentration and plant tolerance to defoliation.
4.   Synthesis . The negative correlation between plant tolerance and AMF concentration suggests that defoliation can reduce AMF benefits and that natural variations in AMF can limit the evolution of optimum levels of tolerance. Moreover, genetic variation in the shape of the reaction norms to AMF in the presence/absence of defoliation suggests that plants may evolve in response to variation in densities of AMF and herbivores.  相似文献

The mycorrhizal status of epiphytic, rupicolous, and terrestrial bromeliad species from the Brazilian Atlantic Rain Forest has been examined. Roots of 13 species of bromeliads were analyzed for the presence of mycorrhizal structures such as arbuscules, hyphae, and vesicles as well as other fungal structures. Rhizosphere soil was sampled to identify arbuscular mycorrhizal fungal (AMF) species associated only with terrestrial bromeliad species. Most specimens collected were epiphytic bromeliads in the genera Aechmea, Bilbergia, Nidularium, Tillandsia, and Vriesea. Differentiating structures of AMF were found in only three species of bromeliads. The pattern of mycorrhizal colonization was mainly internal, and external mycelium and arbuscules were observed only in the terrestrial Nidularium procerum. Root endophytes with dark brown septate mycelium, thin external hyphae, and Rhizoctonia-like sclerotia were also detected in some root segments. A total of ten spore morphotypes were recovered from the rhizosphere of N. procerum, with Acaulospora mellea, A. foveata, and Glomus sp. being the most common species recovered. Our study demonstrated that most of the epiphytic species are not associated with AMF. We attribute this mainly to the exposed bare root conditions found in epiphytic bromeliads.  相似文献

Intra- and extraradical colonization competition and hyphal interactions among arbuscular mycorrhizal fungi (AMF) Glomus intraradices, Glomus proliferum and Gigaspora margarita were investigated in two in vitro experimental systems. AMF were polyxenically cultured with a Ri T-DNA transformed carrot root organ culture (ROC) in either big Petri plates containing three culture compartments and a common hyphal compartment (i.e. an independent host root for each AMF) or two by two in the culture compartment of regular bicompartmented Petri dishes (i.e. a common host root and a common hyphal compartment). Maps of the extraradical mycelial development of the three AMF were obtained. Two distinct substrate colonization strategies (Glomus-type and Gigaspora-type) were identified, reflecting intrinsic differences among AMF genera/families. Our data reveal a general lack of antagonism between the isolates when extraradical hyphae explore and exploit the substrate outside the root influence zone; however certain growth restrictions were imposed by Gi. margarita extraradical mycelium when developing near the host root and by G. proliferum intraradical hyphae. This work highlights once more the appropriateness of AM in vitro culture systems to perform in vivo studies on the biology of this symbiosis and opens new avenues to the formulation of in vitro AMF inoculants.  相似文献

Arbuscular mycorrhizal fungi (AMF) are key determinants of plant interactions in ecosystems. Through their effects on competition, they are regulators of the structure of communities. Conversely, the composition of plant assemblages may also influence the AMF colonization dynamics of plant species. Here, we tested under in vitro culture conditions the effects of Medicago truncatula, a highly mycorrhizal plant species, and Silene vulgaris, a weakly mycorrhizal plant species, grown single (monospecies treatments) or in combination (bispecies treatment) on the colonization dynamics of the AMF Rhizophagus irregularis MUCL 43194. The seedlings were placed in a pre-established hyphal network developing from a mature M. truncatula mycorrhizal donor plant. Extraradical mycelium (ERM) and root colonization parameters as well as root morphology were measured over a period of 12 days. An increased ERM length, total root colonization and proportion of arbuscules were noted in the bispecies treatment. Conversely, the bispecies treatment seemed to have no effect on root growth. This study also demonstrated the suitability of the in vitro culture system for studying the interactions between AMF and host plants grown as mono- and bispecies combinations.  相似文献

Material on the surface of hyphal walls of arbuscular mycorrhizal fungi (AMF) during active colonization of plant roots was detected by a monoclonal antibody. Pot-cultured isolates of Glomus, Acaulospora, Gigaspora, Scutellospora, and Entrophospora had immunofluorescent material (IM) on younger, thinner, intact hyphae, but IM was scant to absent on thicker, melanized or lysing hyphae. Colonization of corn (Zea mays L.), Sudangrass (Sorghum sudanense (Piper) Staph.) or red clover (Trifolium pratense L.) was examined during 5 months of plant growth by removing cores and performing an indirect immunoassay on roots with attached hyphae. Fresh spores of some Glomus spp. had IM on the outer layer of the spore wall. Abundant IM was seen on root hairs of plants colonized by some isolates, and some IM was detected on root surfaces of all plants examined even during early colonization. After cultures were dried, hyphae, roots and spores had little to no IM. Uninoculated control roots had very rare, small patches of IM. An immunoreactive protein was extracted from hyphae of Gigaspora and Glomus isolates by using 20mM citrate (pH 7.0) at 121°C for 90 min. Gel electrophoresis profiles indicated that all isolates tested had the same banding patterns. Lectin-binding of extracted protein is suggestive of a glycoprotein. The immunofluorescence assay can be used to examine root sections for active colonization by AMF, and the potential use of the protein to quantify AMF activity in soil is discussed.  相似文献

Rising atmospheric carbon dioxide partial pressure (pCO2) and nitrogen (N) deposition are important components of global environmental change. In the Swiss free air carbon dioxide enrichment (FACE) experiment, the effect of altered atmospheric pCO2 (35 vs. 60 Pa) and the influence of two different N‐fertilization regimes (14 vs. 56 g N m?2 a?1) on root colonization by arbuscular mycorrhizal fungi (AMF) and other fungi (non‐AMF) of Lolium perenne and Trifolium repens were studied. Plants were grown in permanent monoculture plots, and fumigated during the growth period for 7 years. At elevated pCO2 AMF and non‐AMF root colonization was generally increased in both plant species, with significant effects on colonization intensity and on hyphal and non‐AMF colonization. The CO2 effect on arbuscules was marginally significant (P=0.076). Moreover, the number of small AMF spores (≤100 μm) in the soils of monocultures (at low‐N fertilization) of both plant species was significantly increased, whereas that of large spores (>100 μm) was increased only in L. perenne plots. N fertilization resulted in a significant decrease of root colonization in L. perenne, including the AMF parameters, hyphae, arbuscules, vesicles and intensity, but not in T. repens. This phenomenon was probably caused by different C‐sink limitations of grass and legume. Lacking effects of CO2 fumigation on intraradical AMF structures (under high‐N fertilization) and no response to N fertilization of arbuscules, vesicles and colonization intensity suggest that the function of AMF in T. repens was non‐nutritional. In L. perenne, however, AM symbiosis may have amended N nutrition, because all root colonization parameters were significantly increased under low‐N fertilization, whereas under high‐N fertilization only vesicle colonization was increased. Commonly observed P‐nutritional benefits from AMF appeared to be absent under the phosphorus‐rich soil conditions of our field experiment. We hypothesize that in well‐fertilized agricultural ecosystems, grasses benefit from improved N nutrition and legumes benefit from increased protection against pathogens and/or herbivores. We investigated the functional significance of extraradical mycorrhizal networks produced by geographically different isolates of the arbuscular mycorrhizal fungal (AMF) species Glomus mosseae and Glomus intraradices. A two-dimensional experimental system was used to visualize and quantify intact extraradical mycelium (ERM) spreading from Medicago sativa roots. Growth, phosphorus (P) and nitrogen (N) nutrition were assessed in M. sativa plants grown in microcosms. The AMF isolates were characterized by differences in extent and interconnectedness of ERM. Phenotypic fungal variables, such as total hyphal length, hyphal density, hyphal length per mm of total or colonized root length, were positively correlated with M. sativa growth response variables, such as total shoot biomass and plant P content. The utilization of an experimental system in which size, growth rate, viability and interconnectedness of ERM extending from mycorrhizal roots are easily quantified under realistic conditions allows the simultaneous evaluation of different isolates and provides data with a predictive value for selection of efficient AMF.  相似文献

Effects of long-term mineral fertilization and manuring on the biomass of arbuscular mycorrhizal fungi (AMF) were studied in a field experiment. Mineral fertilization reduced the growth of AMF, as estimated using both measurements of hyphal length and the signature fatty acid 16:1ω5, whereas manuring alone increased the growth of AMF. The results of AMF root colonization followed the same pattern as AMF hyphal length in soil samples, but not AMF spore densities, which increased with increasing mineral and organic fertilization. AMF spore counts and concentration of 16:1ω5 in soil did not correlate positively, suggesting that a significant portion of spores found in soil samples was dead. AMF hyphal length was not correlated with whole cell fatty acid (WCFA) 18:2ω6,9 levels, a biomarker of saprotrophic fungi, indicating that visual measurements of the AMF mycelium were not distorted by erroneous involvement of hyphae of saprotrophs. Our observations indicate that the measurement of WCFAs in soil is a useful research tool for providing information in the characterization of soil microflora.  相似文献
