孑遗植物桫椤种群遗传变异的RAPD分析   总被引:14,自引:2,他引:14  
应用 RAPD标记分析了桫椤分布在海南尖峰岭及霸王岭 ,广东塘朗山、黑石顶及大西山孑遗种群的遗传变异。 2 8个引物共检测到 1 1 8个位点 ,其中多态位点 33个 ,多态位点比率 2 7.97%。Shannon多样性、Nei遗传分化和分子方差分析 ( AMOVA)结果一致显示尖峰岭种群遗传多样性最大 ,霸王岭种群次之、塘郎山种群第三 ,黑石顶种群第四 ,而大西山种群最小。种群的遗传变异主要发生在种群间 ;Shannon指数测出的种群间遗传多样性所占比率为 79.1 0 % (基于表型频率 )和 77.85 % (基于基因频率 ) ,Nei基因分化系数 ( Gst)达 80 .69% ,分子方差分析也表明种群分化极为显著 ( Φst=0 .82 86,P<0 .0 0 1 )。桫椤的种内遗传多样性水平很低 ,种内 Shannon多样性仅为 0 .0 641 (基于表型频率 )和 0 .1 1 2 4 (基于基因频率 ) ,总基因多样性只有 0 .0 770。另外 ,Jaccard相似性系数的 UPGMA分析结果显示 5个种群分为两个分支 :尖峰岭种群和霸王岭种群组成一分支 ;黑石顶、塘郎山和大西山种群组成另一分支 ,并且三者中前两者的遗传关系更为密切。聚类分析结果还得到主成分分析的支持。根据桫椤种群的遗传变异特点 ,初步探讨了保护策略。  相似文献

Four new species of Cyatheaceae from Ecuador are described: Alsophila conantiana Lehnert, Cyathea brucei Lehnert, C. moranii Lehnert, and C. sylvatica Lehnert. Range extensions are documented for Alsophila esmeraldensis R.C. Moran and Cyathea macrocarpa (C. Presl) Domin.

Abstract Breeding-system data have been available for a large number and diverse array of angio-sperms for a relatively long time. In contrast, breeding systems of ferns and their allies (pteridophytes) have only recently been examined, and breeding-system data from natural populations of sporophytes are still lacking for pteridophytes representing many life-history strategies. Few studies, for example, have examined breeding systems of tropical pteridophytes, and no breeding-system data are available for tree ferns. We therefore examined the breeding systems of three species of tree ferns from Costa Rica, Alsophila firma (Cyatheaceae), Cyathea stipularis (Cyatheaceae), and Lophosoria quadripinnata (Lophosoriaceae) using enzyme electro-phoresis. Genetic data were used to estimate intragametophytic self-fertilization and F , the fixation index. Analysis of genetic data indicates that the gametophytes of these three species predominantly cross-fertilize; all three species would be characterized as outcrossers. However, some population-to-population variation in breeding system was detected in all three species. Outcrossing also typifies a diverse array of temperate ferns. Thus, despite the potential for self-fertilization, outcrossing appears to characterize the majority of pteridophytes representing a variety of evolutionary lineages, life-history strategies, and environments.  相似文献

Two species were newly identified in Japan. Lachnum lanariceps was characterized by a central and cylindrical stipe and hairs containing pale yellow pigment with red or garnet resinous matter. Lachnum oncospermatum was identified after comparison with the holotype of Dasyscyphus oncospermatis. This fungus has the characteristics of wrinkled apothecium and branched stipe.  相似文献

Several plants invade the cushions of Laretia acaulis in the high Andean communities of central Chile. Humus develops among the rosettes, and 42 different species settle on the cushions. Most of these species are also found outside the cushions. Semi-woody chamaephytes, hemicryptophytes and cryptophytes have a similar cover on both substrates, while the cover of woody chamaephytes and therophytes is smaller on the cushions. The diversity and cover of invading plants, which vary with altitude, result from interactions among the environmental conditions, cushion morphology and the surrounding vegetation.This study is part of the Chilean contribution to the Man and the Biosphere (MAB) Programme, within the framework of the UNEP/UNESCO Andean Project (1105-77-01).Nomenclature is given in the Appendix.We thank M. Investigation of chromosome numbers of allAzolla species, and for the first time of hybrids, has been undertaken. Removal of wax from the leaf surface proved invaluable in achieving clear cytological preparations and providing unambiguous chromosome numbers. In contrast to previous records, the speciesA. pinnata, A. filiculoides, A. filiculoides var.rubra, A. caroliniana, A. microphylla, andA. mexicana were found to be 2n=44, andA. nilotica to be 2n=52. Several triploids (2n=66) and one tetraploid (2n=88) were identified. No geographical pattern could be observed in the distribution of triploids which probably derive from the function of unreduced gametes. The chromosome number of hybrids occasionally deviates from the diploid chromosome number (2n=44). The small chromosome size limits karyotypic analysis and only differences in overall chromosome size can be observed. Taxonomic implications of chromosome numbers and sizes are discussed.  相似文献

The development of spatial genetic structure (SGS) in seed plants has been linked to several biological attributes of species, such as breeding system and life form. However, little is known about SGS in ferns, which together with lycopods are unique among land plants in having two free‐living life stages. We combined spatial aggregation statistics and spatial genetic autocorrelation analyses using five plastid microsatellites and one nuclear gene to investigate SGS in two populations of the outcrossing tree fern Alsophila firma (Cyatheaceae). We assessed how the observed patterns compare with those estimated for other ferns and seed plants. Populations of A. firma exhibited strong SGS, spatial clustering of individuals, substantial clonal diversity and no inbreeding. SGS in ferns appears to be higher than in most seed plants analysed to date. Contrary to our expectations, an outcrossing breeding system, wind dispersal and an arborescent life form did not translate into weak or no SGS. In ferns, SGS is probably being affected by the life cycle with two free‐living life stages. The reproductive biology of ferns appears to be more complex than previously thought. In Andean forests, Cyathea caracasana grows across a range of successional habitats. This study documents variation in several measures of plant performance (stem growth, leaf production, leaf longevity, and spore production) in C. caracasana growing in open habitat, low-canopy secondary forest, and high-canopy secondary forest, based on 33 mo of observation. In open habitat, C. caracasana displayed significantly higher growth rates, leaf production rates, and leaf turnover than in either of the forested habitats. The highest rates of spore production were also observed in open-habitat individuals, with only one plant in the forest understory producing spores during the study. Despite low growth and no reproduction, I observed no mortality among ferns in the forest understory. These data show that C. caracasana performs best under conditions of full sun but can persist under the closed canopy. This suggests a life history in which periods of rapid growth, spore production, and recruitment in forest gaps alternate with low growth rate and persistence in the understory. A phylogenetic perspective suggests that the habitat flexibility, which might conventionally mark C. caracasana as a habitat generalist, is better interpreted as specialization for the dynamic forest in which it grows.  相似文献

Multiple bud formation was induced from shoot apices of Matteuccia struthiopteris cultured on semi-solid Knudson's medium supplemented with 10-5 and 10-6 M kinetin. Summary With 11 currently recognised species, the genusEriocnemis (Reichenbach, 1849) is one of the most diversified Andean trochilid groups occupying mainly open montane habitats such as the edges of cloud forest or páramos. On the basis of distributional and morphological patterns, this study highlights the geographical variation and biogeography of the taxon. Characteristics common to all these species are the greenish dorsal plumage, the conspicuous and mostly whitish tibial tufts, and a fairly pronounced tail bifurcation. With the help of plumage synapomorphies for a cladistic analysis (PAUP*), several species groups or superspecies can be distinguished: theE. vestitus group (incl.E. vestitus, E. godini, E. nigrivestis), theE. luciani group (incl.E. luciani, E. cupreoventris, E. sapphiropygia), and theE. alinae group (incl.E. alinae, E. mirabilis).E. glaucopoides, E. mosquera, andE. derbyi differ quite widely in morphology and ecological requirements from the other species. Three new subspecies are described,E. vestitus arcosi from southern Ecuador and northern Peru, andE. luciani baptistae from central and southern Ecuador. A previously overlooked specimen ofE. luciani from the Andes of Mérida represents the first species record for Venezuela, about 1100 km northeast of the main population range, and should be recognised taxonomically asE. luciani meridae, subsp. nov., on the basis of its unique plumage morphology and geographical separation. Additionally, the unique type ofE. ventralis (Salvin, 1891) is probably of hybrid origin (E. vestitus × cupreoventris). The genus may have evolved in the northern Andes, subsequently spreading southward and invading the central Andes. Its recent range and phylogenetic patterns indicate vicariance events as the major speciation factor inEriocnemis.In memoriam Dr. Luis F. Baptista (1941–2000)  相似文献

桫椤(Alsophila spinulosa),为中生代古老孑遗物种,国家二级保护植物。贵州赤水桫椤国家级自然保护区是以桫椤及其生存环境为保护对象的自然保护区,是拥有世界上数量最多、面积最广的桫椤林区之一。该文采用植物生态学、区系学等方法,对保护区内群落及物种多样性、伴生物种区系成分、生活型谱、群落结构等进行研究。结果表明:(1)群落内维管植物共计60科96属122种,其中蕨类植物12科14属17种,裸子植物2科2属2种,被子植物46科80属103种。(2)群落内植物的科、属区系成分类型多样,热带成分占优势,温带成分次之,表现出从热带区系向温带区系过渡的特征。(3)生活型谱以高位芽植物为主(占57.38%),反映出中亚热带森林以高位芽植物为优势的特点。(4)群落垂直分层显著,乔木层树高在4~12m的比例最大为76.64%;灌木层树高在2~3m的比例最大为69.78%;草本层各区间分布较为均匀,但以0~0.2 m的比例较大,群落层次明显。(5)毛竹(Phyllostachys heterocycla cv.pubescens)和桫椤在该地乔木组成中占主导地位;灌木层中,桫椤为最优物种;草本层主要以红盖鳞毛蕨(Dryopteris erythrosora)、翠云草(Selaginella uncinata)为主;物种丰富度分析显示,乔木层灌木层草本层;多样性指数、均匀度指数分析表明,乔木层为聚集分布、组成较简单,灌木层和草本层为均匀分布,但数量较少。  相似文献

Fourteen polymorphic microsatellite loci in the endangered fern Adiantum reniforme var. sinense were developed and characterized using the Fast Isolation by AFLP of Sequences Containing repeats (FIASCO) protocol. Polymorphism of each locus was assessed in a bulked sample of 30 individuals from 8 natural populations. The number of alleles per locus varied from 2 to 11, with an average value of 6.2. The ranges of observed and expected heterozygosity were 0.000–0.895 and 0.226–0.868, respectively. These microsatellite markers provide useful tools for the ongoing conservation genetic studies of Adiantum reniforme var. sinense.  相似文献

The pre-meiotic, meiotic and tetrad stages of development in microsporangia of Alsophila setosa were studied with particular emphasis on the early establishment of patterning in the microspore wall and the subsequent development of the sporoderm. The data obtained were compared with corresponding ontogenetic stages of Psilotum nudum. Tapetal behaviour was also examined. During the tetrad period, only one layer, a thin undulating sheet, appeared alongside the plasma membrane of the tetraspores, and this was evidently formed on a pre-patterned structure – a fibrillar layer, corresponding to a kind of primexine matrix. The early free microspores had a wavy plasma membrane with a parallel, sinusoidal, thin initial sporoderm layer. The proximal apertural fold was observed to be an extended outgrowth of this initial spore envelope. Sporoderm ontogeny during the tetrad period in Alsophila and Psilotum show some common points, but also fundamental differences, mainly in the relative timing of events: in Alsophila the end of the tetrad period is the starting point for exospore development, whereas in Psilotum the exospore is already complete at this stage. Considerable differences were also observed in the tapetum of the two species.  相似文献

桫椤(Alsophila spinulosa)对研究物种形成和植物地理区系、重建侏罗纪古生态环境、研究地质变迁具有重要参考价值。依据现有189个桫椤分布样点和21个环境数据,应用最大熵模型(MaxEnt)和地理信息系统(GIS)软件预测末次间冰期、末次冰盛期、中全新世、现今以及未来(2070年)气候条件下桫椤潜在适生区影响因素、空间分布和变化规律。结果表明:1)桫椤潜在适生概率(P)高于0.5的环境条件为:昼夜温差月均值低于7.6℃、最冷月最低温为3-10℃、年温变化在21.5-27.5℃范围,年降水量1200-2100 mm、最干月降水量16-46 mm、降雨量变化方差在57-76区间,海拔160-800 m,坡度12-28°之间;末次间冰期至末次冰盛期,桫椤潜在适生区主要受最冷月最低温(高于-3℃)和最干月降水量(高于10 mm)影响,中全新世至2070年,还同时受降雨量变化方差(大于50)影响;2)桫椤潜在适生区主要位于中国约32°20''N以南的热带亚热带山地,末次间冰期至末次冰盛期,桫椤潜在适生区中心由广东中部向西北方向移动至广西北部且距离最远,末次冰盛期至2070年则由广西北部向东北方向的湖南西部移动;末次间冰期至2070年,II(0.1 < P < 0.3)、III(0.3 < P < 0.5)级潜在适生区平均海拔总体升高,在现今气候下达到最大、之后降低,IV(0.5 < P < 0.7)、V(P > 0.7)级潜在适生区平均海拔先升高、随后呈波浪状升降趋势;3)末次间冰期至2070年,潜在适生区总面积、III、IV、V级潜在适生区面积均增多,在现今气候下达到最大,之后有所减少,II级潜在适生区面积在末次间冰期至中全新世增多,中全新世至现今减少、随后增多;各级潜在适生区面积在海拔约300-400 m处达最大值;约77%-98%的II-V级潜在适生区分布于海拔1200 m以下,约81%-97%的IV、V级潜在适生区分布于海拔800 m以下。本文研究结果可为保护桫椤应对气候变暖、引种栽培和保护区选址提供参考。  相似文献

Summary The body temperature of free-ranging Andean toadsBufo spinulosus was measured either directly or radiotelemetrically during two 15-day periods at 3200 m elevation in the Mantaro Valley, Central Perú. All toads attempted to maintain their diurnal sum of body temperature within a narrow range. Consequently thermoregulatory behaviour differed according to cloud cover and precipitation. If the sky was clear, toads emerged from their hiding place and exposed themselves to solar radiation during 3–5 h in the morning. Core temperature increased up to 15° C above the air temperature in shade and reached maximum values of about 32° C. At air temperatures (in sun) exceeding 29° C, toads maintained body temperatures below 32° C by evaporative cooling. Following heliothermic heating during the moring toads retreated to the shade, thereby decreasing body temperature below air temperature. Under overcast sky toads remained exposed during the whole day displaying body temperatures at or slightly above ambient levels. Quantitative models to predict the core temperature of toads under the different weather conditions demonstrated that the substrate temperature was the main energy source accounting for 64.6–77.9% of total variance whereas air temperature was of minor importance (1.5–4.4%). The unexplained variance was probably due to evaporative cooling. The volume of urine stored into the urinary bladder of toads varied diurnally; during basking in the morning hours most bladders contained large volumes of urine, whereas during the afternoon the bladders were mostly empty. The bladder contents probably serve as water reserves during basking when evaporative water loss was high. Toads preferred sites that provided shady hiding places as well as sun-exposed bare soil within a radius of 5 m. However, they frequently changed their centers of activity and moved to other sites in 20–70 m distance after periods of 2–5 days. The helio-and thigmothermic behaviour of the Andean toad permits the maintenance of high core temperature during morning which probably increases the digestion rate and accelerate growth. Evaporative cooling and preference of shady sites were employed to regulate body temperature below the morning levels in response to the constraints of water balance. Periodic changes between thigmothermic behaviour and locomotory activity during the night maintains body temperature above air temperature and prolongs the period of food uptake.Dedicated to Prof. Dr. H. Summary Biotechnology has offered a nonconventional method of plant propagation and has been intensively applied as a conservation strategy for sustaining biodiversity for rare plants. In vitro conservation through micropropagation of Ochreinauclea missionis, a rare, endemic and medicinal tree species of Western Ghats in Karnataka region of India is reported. Multiple shoots were initiated from nodal explants on Murashige and Skoog (MS) medium supplemented with 8.8 μM 6-benzylaminopurine (BA) and 0.3% (w/v) activated charcoal. Shoots were elongated in MS medium with a combination of 2.2 μM BA and 5.3 μM α-naphthaleneacetic acid (NAA) or growth regulator-free medium. Individual shoots with a minimum of one node were excised and rooted in vitro on MS medium with 0.3% activated charcoal or ex vitro rooted by treatment with 49 μM indole-3-butyric acid (IBA) for 30 min. Regenerants acclimated in Soil-rite exhibited 65% survival in the greenhouse.  相似文献

Summary Cortical microtubule arrays in tip-growing protonemal and rhizoid cells of the fernAdiantum gametophytes were observed by immunofluorescence microscopy. A circular arrangement of cortical microtubules was demonstrated around the subapical part of protonemal cells growing under red light conditions. However, such an arrangement was not found in growing rhizoids either by immunofluorescence microscopy or by electron microscopy. The different patterns of microtubule arrays around the apices of tip-growing protonemal and rhizoid cells suggest the possible existence of different mechanisms in regulating the cell diameter in the two types of cylindrical cell.  相似文献
