Aims:  The objective of this study was to deduce and analyse equations that best describe the behaviour of faecal bacterial indicators and two decomposition parameters during broiler litter composting. Mathematical models were fitted and the order of rate equations were identified.
Methods and Results:  The Levenburg-Marquardt algorithm was used to fit nonlinear mathematical models to total coliforms (TC), faecal coliforms (FC), Escherichia coli (EC), faecal enteroccocci (FE), organic-C and volatile solids reduction, VSRed, by the least squares procedure. The rate equations showed that TC, FC and EC reductions were expressed by second-order decay kinetics. FE reduction followed first-order decay. Temperature dependency of decomposition rate was effectively verified by applying empirically derived rate equations.
Conclusions:  The governing mathematical models critically compare the inactivation kinetics of faecal indicators. TC, FC and EC were rapidly destroyed while FE was more resistant. Temperature elevation, organic-C and VSRed dynamics provide an accurate understanding of composting-induced decomposition of the broiler litter.
Significance and Impact of the Study:  The conservative performance of FE with respect to the other indicators has been established. Hence, FE presents better opportunities to encompass the totality of the composting process in terms of attainment of hygiene efficacy compared with EC.  相似文献

In this report we describe the results of a studyconducted in order to better estimate airbornemicroorganisms. A method based on a bio-moleculartechnique, Polymerase Chain Reaction (PCR) wascompared with the culture methods based on the viablecounts of total and fecal bacteria. Microbial aerosolemission from the surfaces of aeration tanks of anindustrial and municipal wastewater treatment plant(Como, Italy), at different seasons, was determined.This study was accomplished by conducting test runs inwhich SAS (Surface Air Systems, PBI) viable sampler,Sartorius MD8 with membrane gelatine filter andgravity method with Petri dishes were used to collectbacterial aerosol samples in situ. Total aerobicheterothropic bacteria, Mycetes, total and fecalcoliforms were determined. The preliminary resultsshow that: no correlation was found between the twodifferent passive and active culture techniques, dueto the different mechanisms of capture of bioaerosolagents on the media; optimal values for the recoveryof E. coli viable bacteria by MD8 samplerwith gelatine membrane, time and temperature ofstorage, were recognised. For the PCR technique, acouple of primers (URL 301–URR 432) to detect E. coli, on definite air samples, was used, operativeconditions were defined, and then, applied inmonitoring on in situ bioaerosol samples. At thewastewater plant, the highest total aerobic bacteriaemission rate during the preliminary mechanicaltreatments and in correspondence of the enclosedactivated sludge phase, as a consequence of theremarkable aeration of the tanks, were registered. Thesensitivity (82 CFU/m3) and rapidity (less than 8hours) of the biomolecular methods to determine thepresence and the fecal coliforms (E. coli) ratein bioaerosols was considered satisfactory.  相似文献

Seasonal variations and aeration effects on water quality improvements and the physiological responses of Nymphaea tetragona Georgi were investigated with mesocosm experiments. Plants were hydroponically cultivated in six purifying tanks (aerated, non-aerated) and the characteristics of the plants were measured. Water quality improvements in purifying tanks were evaluated by comparing to the control tanks. The results showed that continuous aeration affected the plant morphology and physiology. The lengths of the roots, petioles and leaf limbs in aeration conditions were shorter than in non-aeration conditions. Chlorophyll and soluble protein contents of the leaf limbs in aerated tanks decreased, while peroxidase and catalase activities of roots tissues increased. In spring and summer, effects of aeration on the plants were less than in autumn. Total nitrogen (TN) and ammonia nitrogen (NH4 +-N) in aerated tanks were lower than in non-aerated tanks, while total phosphorus (TP) and dissolved phosphorus (DP) increased in spring and summer. In autumn, effects of aeration on the plants became more significant. TN, NH4 +-N, TP and DP became higher in aerated tanks than in non-aerated tanks in autumn. This work provided evidences for regulating aeration techniques based on seasonal variations of the plant physiology in restoring polluted stagnant water.  相似文献

A biofilm fluidized sand bed column reactor (14 L) has been operated in the three-phase mode on a soluble glucose-yeast hydrolysate substrate in which the biofilm-sand phase (1-2.5 L) was suspended by direct aeration of the bed. Within two weeks a tight biofilm was formed whose activity resulted in a 90% reduction, with loads of 10.7 kg TC/m(3)day. The residence time was 1 h. The biofilm remained intact during operation with high residence times (up to 23 h) over three weeks. Oxygen transfer coefficients varied with aeration rate and sand quantity between 0.02 and 0.04 s(-1) during non growth conditions; they decreased with increasing amounts of clean sand and were higher and relatively independent of the sand fraction with biofilm-covered sand. Aeration rates used in the 14 L reactor were 23-40 L/min (2.4-4.1 cm/s) and were sufficient to suspend 78-92% f the biofilm-covered sand. Clean sand was 50-75% suspended. Oxygen uptake rates varied between 15.4 and 23.1 mol/m(3) h.  相似文献

To optimize the aeration conditions for microalgal biomass production in a vertical flat-plate photobioreactor (VFPP), the effect of the aeration rate on biomass productivity was investigated under given conditions. Air enriched with 5% or 10% (v/v) CO(2) was supplied for the investigation at rates of 0.025-1 vvm. The CO(2) utilization efficiency, change of pH in the medium, and the optimum aeration rate were determined by evaluating biomass productivity. To investigate the VFPP mass transfer characteristics, the overall volumetric mass transfer coefficient, k(L)a, was evaluated for several different flat-plate sizes. Increasing the height of the VFPP could improve both the mass transfer of CO(2) and the illumination conditions, so this appeared to be a good method for scaling up. Based on a comparison of the k(L)a value at the optimum aeration rate with previously reported results, it was confirmed that the range of CO(2) concentration used in the experiments was cost-effective for mass culture.  相似文献

The effects of the sensitizer methylene blue at concentrations of 0 to 10 mg/liter, different radiation times from 0 to 2 h, and sunlight intensities of 0, 68, and 2,030 muE/m2 per s were investigated. In laboratory-scale experiments, 1.3 X 10(9) coliforms in 100 ml of oxidation pond municipal sewage water containing 0.5 mg of methylene blue were destroyed in about 30 min. The results of the study on relation between tetracycline biosynthesis and the specific power input for agitation in pilot plant apparatus was studied. No correlation was observed between the levels of tetracycline biosynthesis and changes in the specific power input within a range of 0.6 to 2.3 kW/m3 at the expense of changes in the mixer diameter and the agitation rate, when the aeration rate was constant. It was shown that the aeration conditions were most significant for tetracycline biosynthesis. The study provided determination of the optimal aeration conditions for biosynthesis of tetracycline.  相似文献

Both long term and batch experiments were carried out to identify the sources of the N2O emission in anoxic/aerobic sequencing batch reactors (A/O SBRs) under different aeration rates. The obtained results showed that aeration rate has an important effect on the N2O emission of A/O SBR and most of the N2O was emitted during the aerobic phase. During the anoxic phase, nitrate ammonification was the major source of N2O emission while denitrification performed as a sink of N2O, in all three bioreactors. The N2O emission mechanisms during the aerobic phase differed with the aeration rate. At low and high aeration rates (Run 1 and Run 3), both coupled-denitrification and nitrifier denitrification were ascribed to be the source of N2O emission. At mild aeration rate (Run 2), nitrifier denitrification by Nitrosomonas-like ammonia oxidizing-bacterial (AOB) was responsible for N2O emission while coupled-denitrification turned out to be a sink of N2O because of the presence of inner anaerobic region in sludge flocs.  相似文献

应用TGGE指纹图谱技术对两个曝气池细菌种群的动态变化及多样性进行了研究。每3d进行1次,共8个监测时期中同一曝气池活性污泥的16SrDNAV3-PCRTGGE指纹图谱基本一致,图谱间的相似性系数(Cs)为100%。同一曝气池不同位点活性污泥的TGGE指纹图谱也完全一致。功能不同曝气池活性污泥TGGE指纹图谱存在差异,Cs为83.3%。对TJ1活性污泥TGGE图谱中9条主要条带回收、扩增、克隆建库,每个条带选4个转化子进行序列分析,结果显示TGGE条带是由序列不同的片段组成。32个序列在97%的相似性下分成16个分类操作单元(OTU),14个OTU与GenBank中已登录的细菌种群的同源性≥97%,2个OTU的同源性为95%和94%。与10个OTU同源性较高的细菌类群是在活性污泥或污染环境分离或发现的,与8个OTU相似的细菌类群目前尚无法分离培养。  相似文献

Inactivation (loss of culturability) by sunlight of enterococci and fecal coliforms within sewage effluent diluted in seawater was investigated in field experiments. In most experiments, 500-ml flasks of pure silica were used to confine activated sludge effluent diluted to 2% (vol/vol) in seawater. Inactivation of bacteria in these flasks (diameter, 0.1 m) was faster than in either open chambers (depth, 0.25 m) or patches of dyed effluent (depth of order, 1 m), probably because of the longer light paths in the latter two types of experiment, which caused greater attenuation of sunlight. Inactivation of 90% of enterococci generally required 2.3 times the insolation required for 90% inactivation of fecal coliforms, because of both the presence of larger initial shoulders on survival curves and a lower final inactivation rate. Two parameters are required to model inactivation of enterococci, a shoulder constant as well as a rate coefficient. The depth dependence of inactivation rate for both fecal indicators matched the attenuation profile of UV-A radiation at about 360 nm. Inactivation by UV-B radiation (290 to 320 nm), which penetrates much less into seawater, is of minor importance compared with the UV-A and visible radiation in sunlight, contrary to expectations in consideration of published action spectra for bacterial inactivation.  相似文献

Aims: To measure enteric bacterial contamination of tubewells in three flood prone areas in Bangladesh and the relationship of bacteriological contamination with tubewell sanitary inspection scores. Methods and Results: Microbiologists selected 207 tubewells in three flood prone districts, assessed physical characteristics of the tubewells and collected a single water sample from each tubewell. Tubewell water samples were contaminated with total coliforms (41%, n = 85), thermotolerant coliforms (29%, n = 60) and Escherichia coli (13%, n = 27). Among contaminated wells, the median CFU of contamination per 100 ml was 8 (interquartile range, 2–30) total coliforms, 5 (interquartile range, 2–23) thermotolerant coliforms and 6 (interquartile range, 1–30) E. coli. There was no significant association between tubewell contamination with E. coli, thermotolerant coliforms or total coliforms and a poor sanitary inspection score, though a history of inundation was associated with contamination with both E. coli and thermotolerant coliforms. Conclusions: Tubewells in flood-prone regions of Bangladesh were commonly contaminated with low levels of faecal organisms, contamination that could not be predicted by examining the tubewell’s external characteristics. Significance and Impact of the Study: The forms currently used for sanitary inspection do not identify the most important causes of drinking water contamination in these communities.  相似文献

Sunlight inactivation rates of somatic coliphages, F-specific RNA bacteriophages (F-RNA phages), and fecal coliforms were compared in seven summer and three winter survival experiments. Experiments were conducted outdoors, using 300-liter 2% (vol/vol) sewage-seawater mixtures held in open-top chambers. Dark inactivation rates (k(D)s), measured from exponential survival curves in enclosed (control) chambers, were higher in summer (temperature range: 14 to 20 degrees C) than in winter (temperature range: 8 to 10 degrees C). Winter k(D)s were highest for fecal coliforms and lowest for F-RNA phages but were the same or similar for all three indicators in summer. Sunlight inactivation rates (k(S)), as a function of cumulative global solar radiation (insolation), were all higher than the k(D)s with a consistent k(S) ranking (from greatest to least) as follows: fecal coliforms, F-RNA phages, and somatic coliphages. Phage inactivation was exponential, but bacterial curves typically exhibited a shoulder. Phages from raw sewage exhibited k(S)s similar to those from waste stabilization pond effluent, but raw sewage fecal coliforms were inactivated faster than pond effluent fecal coliforms. In an experiment which included F-DNA phages and Bacteroides fragilis phages, the k(S) ranking (from greatest to least) was as follows: fecal coliforms, F-RNA phages, B. fragilis phages, F-DNA phages, and somatic coliphages. In a 2-day experiment which included enterococci, the initial concentration ranking (from greatest to least: fecal coliforms, enterococci, F-RNA phages, and somatic coliphages) was reversed during sunlight exposure, with only the phages remaining detectable by the end of day 2. Inactivation rates under different optical filters decreased with the increase in spectral cutoff wavelength (50% light transmission) and indicated that F-RNA phages and fecal coliforms are more susceptible than somatic coliphages to longer solar wavelengths, which predominate in seawater. The consistently superior survival of somatic coliphages in our experiments suggests that they warrant further consideration as fecal, and possibly viral, indicators in marine waters.  相似文献

Cattle and other animals infected by Salmonella can emit high numbers of these bacteria. To determine an effective means for reducing this bacterial group in animal slurry, samples were subjected to aeration in laboratory experiments and in farm-scale slurry tanks. A clear reduction in Salmonella levels was found in laboratory experiments at temperatures from 4 to 40 °C. Aeration in farm-scale slurry tanks increased the temperature above the ambient temperatures (often less than 0 °C) to maxima ranging between 19 and 40 °C. Farm-scale aeration resulted in similar reductions in Salmonella as those achieved in laboratory experiments. Thus, reductions, ranging from greater than 99% of the initial number to no detectable Salmonella , could be reached after 2–5 weeks using aeration processes with cattle slurries contaminated by Salm. infantis or pig slurry contaminated by Salm. typhimurium . These results suggest that farmers can control the spread of Salmonella from slurry to agricultural fields. The reduction mechanisms remain unknown, though the increase in pH (to 7·6–9·0) found in slurries after aeration might exert a decreasing effect on these bacteria.  相似文献

Safe water quality criteria on the load and types of microbial populations are important for human use from fishery, tourism and navigational viewpoints. To understand the variations in sewage pollution indicator and certain human pathogenic bacteria, data collected from various locations along central west coast of India during 2002–2007 were analyzed. Water and sediment samples were examined for total viable counts (TVC), pollution indicator bacteria (total coliforms – TC, fecal coliforms – FC and Escherichia coli – EC) and potential pathogens (Vibrio cholerae – VC, Shigella – SH, and Salmonella spp. – SA). In both Mandovi and Zuari estuaries, where fishing and tourist-related activities are sizable and long-term data collection was regular, we observed high counts of TC, FC, VC, SH and SA in particular during monsoon due to increased land runoff. Further, the abundance of TC and FC has increased significantly over the years in the water column to much above either USEPA or India permissible limits. The concentrations of Vibrio cholerae, and Shigella correlated with those of coliforms. Pathogenic bacteria were detected even 20 km and/or 25 km offshore mainly due to dumping of raw or improperly treated sewage effluents either from land, fishing trawlers and/or ships in the anchorage. Higher concentrations of fecal coliforms and pathogenic bacteria in neretic waters signify threats to environmental and human health.  相似文献

AIMS: To describe the extent of faecal pollution and point of use water treatment strategy during and after the 2004 flood in Dhaka. METHODS: A total of 300 water samples were collected from 20 different drinking water sources in Kamalapur, Dhaka city from August 2004 to January 2005. The level of faecal contamination was estimated using measurements of faecal indicator bacteria (total coliforms, faecal coliforms and faecal streptococci) and isolation of Vibrio cholerae was carried out following standard procedures. Total dissolved solids, dissolved oxygen, hardness, chloride and pH were also monitored. The efficacy of four disinfectants including Halotab, Zeoline-200, alum potash and bleaching powder were tested as point of use water treatment agents. The unacceptable level of contamination of total coliforms (TC), faecal coliforms (FC) and faecal streptococci (FS) ranged from 23.8% to 95.2%, 28.6% to 95.2% and 33.3% to 90.0%, respectively. The isolation rates of V. cholerae O1 and O139 were both 0.33%, and non-O1/non-O139 was 7.0%. CONCLUSION: Water collected during and after floods was contaminated with TC, FC, FS and V. cholerae. Although alum potash, bleaching powder, Halotab and Zeoline-200 were all effective general disinfectants, Halotab and Zeoline-200 were superior to bleaching powder and alum potash against FC. SIGNIFICANCE AND IMPACT OF THE STUDY: During and after floods, point of use water treatment could reduce waterborne diseases among flood-affected people.  相似文献

The major cationic peroxidase in sorghum grain (SPC4) , which is ubiquitously present in all sorghum varieties was purified to apparent homogeneity, and found to be a highly basic protein (pI approximately 11). MS analysis showed that SPC4 consists of two glycoforms with molecular masses of 34,227 and 35,629 Da and it contains a type-b heme. Chemical deglycosylation allowed to estimate sugar contents of 3.0% and 6.7% (w/w) in glycoform I and II, respectively, and a mass of the apoprotein of 33,246 Da. High performance anion exchange chromatography allowed to determine the carbohydrate constituents of the polysaccharide chains. The N-terminal sequence of SPC4 is not blocked by pyroglutamate. MS analysis showed that six peptides, including the N-terminal sequence of SPC4 matched with the predicted tryptic peptides of gene indice TC102191 of sorghum chromosome 1, indicating that TC102191 codes for the N-terminal part of the sequence of SPC4, including a signal peptide of 31 amino acids. The N-terminal fragment of SPC4 (213 amino acids) has a high sequence identity with barley BP1 (85%), rice Prx23 (90%), wheat WSP1 (82%) and maize peroxidase (58%), indicative for a common ancestor. SPC4 is activated by calcium ions. Ca2+ binding increased the protein conformational stability by raising the melting temperature (Tm) from 67 to 82 degrees C. SPC4 catalyzed the oxidation of a wide range of aromatic substrates, being catalytically more efficient with hydroxycinnamates than with tyrosine derivatives. In spite of the conserved active sites, SPC4 differs from BP1 in being active with aromatic compounds above pH 5.  相似文献

A pilot-scale active aeration reactor was studied for composting food wastes in an open-top container aerated with negative pressure by vacuum. A biological filter bed was used to remove NH(3) from the emission. In addition to monitoring system parameters, the compost stability and fertilizer content were also examined to ensure product quality. The compost temperature rose to above 65 degrees C the first day and maintained at the high temperature fermentation level for 30 days. The pH decreased from the initial 5.2 to 4.3 on the first day and then rose slowly to 7.4 on the 60th day. After 60 days, the C/N ratio dropped from 32 to below 20. The final compost contains 1.6% nitrogen, 0.6% phosphate, and 1.4% potassium with total coliforms below 3 MPN/100mL. Using the biological filter to remove NH3, the emission contains less than 1 ppm of NH3. This system proves to be effective and environmental friendly.  相似文献

通过田间试验,采用静态箱法研究相同施氮量条件下,施用尿素、控释肥及尿素与控释肥配施(尿素与控释肥以3∶7配合施用)对稻田N2O排放的影响.结果表明:与单施尿素处理相比,配施处理和控释肥处理水稻生长期N2O排放量分别减少40.4%和59.6%(P<0.05),其中烤田期分别减少65.1%和83.9%;与配施处理相比,施用控释肥处理N2O排放量略微减少(P>0.05),其中烤田期减少53.9%.施用控释肥可增加水稻产量,与尿素处理相比,施用控释肥和配施处理水稻产量分别增加7.8%和9.8%(P>0.05).施用控释肥使土壤无机氮峰值出现时间延后,烤田期N2O排放减少.水稻生长期N2O排放通量与土壤氧化还原电位(Eh)和土壤温度均无明显相关性(P>0.05).  相似文献

Both k(L)a and k(L) measurements were carried out by an unsteady state technique at impeller speeds ranging from 1.6 to 5.8 s(-1) in a mechanically agitated animal cell culture vessel of working volume 1.5 L. Checks were made that the time constant of the oxygen electrode was negligible compared to the time for aeration and that the oxygen electrode reading was not a function of agitator speed in the range employed. The k(L) values by surface aeration of (1.18-3.54) x 10(-5) m/s and k(L)a values by sparged aeration of (2.8-8.5) x 10(-4) s(-1) were found. The former are in reasonable agreement with published experimental values and the latter in accord with values estimated from published correlations based on agitator power input and aeration rate. The fluids used were water, basal medium, and basal medium supplemented with 5% (v/v) foetal calf serum; for each of these, k(L) and k(L)a values were similar. However, the addition of silicone antifoam (6 PPM) reduced the k(L)a value by ca. 50%.  相似文献
