虎杖种质资源的分子标记研究   总被引:2,自引:0,他引:2  
本文应用RAPD、ISSR和SRAP标记对26份虎杖种质资源遗传多样性进行检测.在22个引物中有17个引物(77.3%)扩增产物具多态性,多态性水平相对较高.22个引物共得到98条扩增DNA片段,其中90.8%具有多态性.每个多态性引物平均可扩增出5.24个多态性片段.聚类分析表明,利用BAPD、ISSR和SRAP技术相结合可将全部供试材料区分开,26份材料在Gs值0.54水平上全部聚为一类,以所有材料间的平均遗传相似遗传系数0.71为阈值,将其分为11类.虎杖种质资源在分子水平上确实存在较大遗传差异,RAPD、ISSR和SRAP标记可作为构建虎杖DNA指纹图谱的有效工具.  相似文献

利用RAPD标记分析大麦种质资源的遗传多样性   总被引:6,自引:4,他引:6  
利用RAPD标记对19份西藏近缘野生大麦材料、33份我国不同省市的地方品种以及8份国外引进大麦品种共60份大麦种质资源的遗传多样性进行检测.结果表明材料间遗传差异明显.32个RAPD引物中,有25个引物(占78.13%)可扩增出清晰且具多态性的条带,另外7个引物能扩增出1~3条清晰但无多态性的条带.每个引物可扩增出1~8条多态性带,平均为3.72条.32个引物共产生119条DNA片段,其中87条具有多态性,多态性比率(PPB)为73.11%,平均多态信息量(PIC)为0.434;每个位点平均有效等位基因数(Ne)为2.304;材料间遗传相似系数GS变化范围为0.757~0.981,平均值为0.871.19份来源于西藏的近缘野生大麦材料间GS值变幅为0.818~0.969,平均为0.892;33份我国栽培大麦地方品种间的GS值变化范围为0.783~0.981,平均为0.879;8份分别来自8个国家的栽培大麦品种间的GS值变幅为0.820~0.956,平均为0.882.根据RAPD标记分析的结果,对60份大麦种质资源进行聚类分析,在平均GS值0.871水平上60份大麦材料可聚为5类,聚类结果能在一定程度上反应材料的地理分布关系,但某些相同地理来源的材料也较分散地分布在整个聚类树中.本研究从分子水平上进一步证明了我国栽培大麦丰富的遗传多样性,是世界栽培大麦的遗传多样性中心之一.  相似文献

云南黑籽南瓜种质遗传多样性的RAPD和ISSR分析   总被引:5,自引:0,他引:5  
摘要: 采用RAPD和ISSR分子标记技术对来源于云南省6个地州13份的黑籽南瓜种质进行遗传多样性分析。结果表明:6个RAPD和6个ISSR引物分别扩增出43条和41条带,多态性比率分别为90.70%和51.21%;RAPD和ISSR标记检测供试材的遗传相似性系数(Gs)范围,分别为0.340-0.895和0.162-0.941,ISSR(平均GS值0.698)检测多态性效果高于RAPD(平均GS值0.481)。RAPD标记聚类分析将供试种质分为3个类群5组;ISSR标记聚类分析将供试种质分为4个类群6组,RAPD和ISSR标记的遗传相似性系数呈显著相关(r=0.536)。基于UPGMA聚类结果,可为黑籽南瓜的引种栽培或品种改良提供参考。  相似文献

苦瓜种质遗传多样性的RAPD和ISSR分析   总被引:10,自引:1,他引:9  
采用RAPD和ISSR分子标记技术对38份苦瓜种质进行遗传多样性分析。结果表明:10个RAPD和10个ISSR引物分别扩增出93条和81条带,多态性比率分别为50.54%和61.29%;RAPD和ISSR标记检测供试材料的遗传相似性系数(GS)范围,分别为0.287~1和0.221~1,ISSR(平均GS值0.672)检测多态性效果高于RAPD(平均GS值0.694)。RAPD标记聚类分析将供试种质分为3个类群6组,分类结果与苦瓜瓜瘤的表型分类比较相似;ISSR标记聚类分析将供试种质分为3个类群7组,ISSR标记划分类群与形态上以颜色分类比较接近。RAPD和ISSR标记的遗传相似性系数呈显著相关(r=0.550)。两个标记整合后聚类分析可检测到更大的遗传变异,结果与苦瓜的农艺性状分类和地理分布有一定的相关性。  相似文献

旨在探讨灵芝原生质体单核体间遗传多态性,为育种材料的筛选提供分子水平依据。采用原生质体单核化获得灵芝Ganoderma lucidum 36株单核体,从筛选出的12组SRAP引物,13条ISSR引物和13条RAPD引物对供试菌株的多态性综合分析结果显示,共扩增出320个条带,其中265条表现出多态性,多态位点百分率为82.81%;根据扩增结果采用软件NTsys2.10e计算菌株间的遗传相似系数(GS值)并进行聚类分析,37个菌株间GS值的变化范围为0.3741~0.9852;130-29、130-33和130-53等7株相同交配型单核体聚为一类,其余29株单核体与出发菌株C0130聚为一类,但并非严格按照交配型聚类,且单核体表现出发菌株全部或部分带型,同时也表现其所不具有的带型。研究表明,SRAP、ISSR和RAPD标记可以用于灵芝单核体多态性研究,并可为交配型鉴定提供参考,这将为育种材料的筛选提供帮助,减少育种工作量和风险。  相似文献

尖孢镰刀菌可造成不同瓜类的枯萎病.为明确不同寄主、不同地区的瓜类枯萎病菌菌株间的遗传多样性及亲缘关系,采用相关序列扩增多态性(SRAP)分子标记技术,对来源于不同地区、不同寄主的95株尖孢镰刀菌的基因组DNA进行多态性扩增.以筛选出的19对引物共扩增出238条带,多态性比率为100%,平均每对引物扩增出12.5个位点和12.5个多态性位点;尖孢镰刀菌苦瓜专化型共扩增出166条带,其中145条为多态性条带,多态性比率为87.4%,平均每对引物扩增出8.7个位点和7.7个多态性位点,说明尖孢镰刀菌的遗传变异较为广泛.瓜类枯萎病菌株间的遗传相似系数范围为0.68~0.99,样品间的平均Nei遗传多样性指数和Shannon指数分别为0.2390和0.3718.在遗传相似系数为0.74时,可将供试的95株尖孢镰刀菌划分为苦瓜、黄瓜、西瓜、甜瓜4个专化群.在SRAP聚类树中,同一寄主的尖孢镰刀菌聚在一个分支上,其中尖孢镰刀菌苦瓜专化型菌株间的遗传相似系数范围为0.78~0.99,Nei遗传多样性指数为0.1811,平均Shannon指数为0.2750,表明尖孢镰刀菌苦瓜专化型的遗传变异较大,且菌株的聚群与地理来源存在相关性.  相似文献

粗壮女贞(苦丁茶)不同种质材料遗传多样性的RAPD分析   总被引:1,自引:0,他引:1  
木犀科苦丁茶是我国西南地区一大类重要的代茶饮料植物,其中以粗壮女贞[Ligustrum robustum (Roxb.) Blume]为代表.本研究应用随机扩增多态DNA(randomly amplified polymorphic DNA,RAPD)分子标记技术,系统地研究了粗壮女贞种质材料的亲缘关系和遗传多样性.用所筛选出的19条RAPD随机引物对79份不同地域来源的粗壮女贞种质材料进行扩增,共扩增出369条谱带,多态性带百分率为86.0%;各种质材料间的GS值变化范围为0.466 4~0.973 6,平均GS值为0.662 3;有效等位基因数Ne=1.432 1,Nei’s基因多样性指数He=0.259 5,Shannon多样性信息指数Ho=0.396 0.这表明,粗壮女贞不同地域来源或不同类群种质材料之间存在着显著的遗传差异,其遗传多样性丰富; RAPD技术可以应用于粗壮女贞种内遗传多样性分析.基于RAPD遗传相似系数的UPGMA聚类结果表明,所有各供试材料均按其地理起源聚类,可把供试的79份粗壮女贞材料分为两大类群,即A类群(贵州 四川类群)和B类群 (云南类群).  相似文献

为了解来自广东和广西的瓜类疫霉的遗传多样性,利用从180条RAPD引物中所筛选出的多态扩增性强、重复性好的12条引物,对分离自两省区的96株瓜类疫霉进行了全基因组DNA遗传多样性分析和指纹图谱构建。通过对供试菌株的RAPD-PCR扩增,共获得135条DNA标记谱带,其中124条为多态性谱带,多态检测率为91.9%。利用NTSYSpc Version2.1软件对供试菌株间的遗传距离进行聚类分析并构建系统树,以遗传相似系数0.81为阈值,将96个供试菌株划分为12个RAPD群,多数分离物之间遗传相似性较低,在DNA水平上存在显著的遗传变异,具有较丰富的遗传多样性。不同地区间菌株的遗传分化程度不同,分离自黄瓜的菌株遗传分化明显高于分离自冬瓜的菌株。RAPD群与菌株地理来源、分离寄主、致病力、交配型及甲霜灵抗性均无明显的相关性。  相似文献

试验用SSR分子标记对60份宁夏粳稻种质资源进行遗传多样性分析。103对SSR引物表现多态性的有58对,共扩增出212条多态性条带,等位变异范围为2~9,平均每对引物3.7个;多态性信息含量(PIC)变幅为0.032~0.788,平均为0.403;高多态性位点主要发生在3号、6号和11号染色体上,而无多态性或低多态性位点主要发生在1号和10号染色体上;成对供试材料的遗传相似系数GS值变幅为0.642~0.958,平均为0.790,单个供试材料的平均GS值变幅为0.710~0.816,平均为0.781,亲缘关系较近;UPGMA聚类表明,在遗传相似系数约0.785处,供试材料可被分为11类,大部分材料被聚在一类中。  相似文献

采用SRAP分子标记技术, 对采自中国四川、重庆、贵州、西藏四省区的32份野生狗牙根(Cynodon dactylon)材料进行遗传多样性分析, 获得下述结果:(1)用14对引物组合共得到132条多态性条带, 平均每对引物扩增出9.4条多态带, 多态性位点百分率为79.8%, 材料间的遗传相似系数范围在0.591到0.957之间, 平均GS值为0.759, 这些结果说明, 供试野生狗牙根具有较为丰富的遗传多样性; (2)对所有材料进行聚类分析, 可聚为4类, 大部分来自相同或相似生态地理环境的材料聚为一类, 表明供试材料的聚类和其生态地理环境间有一定的相关性; (3)基于Shannon多样性指数估算了6个狗牙根生态地理类群内和类群间的遗传分化, 发现类群内遗传变异占总变异的65.56%, 而类群间遗传变异占总变异的34.44%; (4)对各生态地理类群基于Nei氏无偏估计的遗传一致度的聚类分析表明, 各生态地理类群间的遗传分化与其所处的生态地理环境具有一定的相关性。  相似文献

Genetic diversity within North American ginseng (Panax quinquefolius L.) grown in Ontario was investigated at the DNA level using the randomly amplified polymorphic DNA (RAPD) method via the polymerase chain reaction (PCR). A total of 420 random decamers were initially screened against DNA from four ginseng plants and 78.8% of them generated RAPD fragments. Thirty-six of the decamers that generated highly repeatable polymorphic RAPD markers were selected for further RAPD analysis of the ginseng population. With these primers, 352 discernible DNA fragments were produced from DNA of 48 ginseng plants, corresponding to an average of 9.8 fragments per primer, of which over 45% were polymorphic. The similarity coefficients among the DNA of ginseng plants analyzed were low, ranging from 0.149 to 0.605 with a mean of 0.412, indicating that a high degree of genetic diversity exists in the ginseng population. Lower levels of genetic diversity were detected among 3-year-old ginseng plants selected on the basis of greater plant height than among the plants randomly selected from the same subpopulation or over the whole population, suggesting that genetic factors at least partly contribute to morphological variation within the ginseng population and that visual selection can be effective in identifying the genetic differences. The significance of a high degree of genetic variation in the ginseng population on its potential for improvement by breeding is also discussed.  相似文献

The genomic DNA from ten isolates of the cattle tick, Boophilus microplus collected in and around Chennai, India, was analyzed by random amplified polymorphic DNA (RAPD) using PCR. Selected five random primers were used for the study of genetic variability among different isolates of B. microplus. A high degree of genetic polymorphism with a different pattern of RAPD profiles for each tick isolate was detected with all these random primers. This variability was also confirmed by similarity coefficient values and dendrogram which were performed using mean RAPD profiles for all the primers between various isolates of ticks. The findings suggest the existence of a complex genotypic diversity of the tick B. microplus in an endemic region such as Chennai.  相似文献

Bipolaris sorokiniana is a phytopathogenic fungus causing diseases of cereal crops such as common root rot, the leaf spot disease, seedling blight, and black point of the grain. Random-amplified polymorphic DNA (RAPD) assay was used to investigate the genetic diversity of 20 isolates collected from different cultivars in wheat-producing regions in Brazil. Seventy primers, with random nucleotide sequences, were tested. Reproducibility to amplify the genomic DNA of isolates was found for 30 of the 70 primers tested, generating between 1 and 17 fragments ranging from 0.35 to 2.0 kb (average size). The degree of similarity between samples was calculated through simple association and the dendrogram was assessed using the unweighted pair group method with arithmetical average. After the RAPD analyses 19 isolates were closely grouped, having a similarity coefficient of >or= 78%. Isolate I017 showed very low similarity coefficients, ranging between 38 and 46%. The RAPD analyses provided important information as to the degree of genetic variability and the relationship between the isolates investigated, revealing polymorphism and establishing electrophoretic profiles useful to characterize the phytopathogen.  相似文献

以我国主要稻区的稗草植株上分离的17株尖角突脐孢菌菌株为试验材料,采用改良的SDS法提取其基因组DNA,并运用优化的RAPD分析体系对其进行了分子标记遗传差异研究。从25个随机引物中筛选出20个扩增效果好的引物,对全部试验材料进行了RAPD扩增,共得到239条有效带,其中多态性带229条(占95.8%)。依据扩增结果建立了17株尖角突脐孢菌基因型的DNA指纹图谱并对其进行了有效区分。根据RAPD分析结果计算了菌株间的遗传距离,分析了它们的遗传差异并进行了聚类分析,结果表明,RAPD分子标记技术是能够用于杂草致病菌资源的鉴定的,并可以进一步应用于特定性状的基因标记研究。  相似文献

Shisham (Dalbergia sissoo) is one of the most preferred timber tree species of South Asia. Two DNA-based molecular marker techniques, intersimple sequence repeat (ISSR) and random amplified polymorphism DNA (RAPD), were compared to study the genetic diversity in this species. A total of 30 polymorphic primers (15 ISSR and 15 random) were used. Amplification of genomic DNA of 22 genotypes, using ISSR analysis, yielded 117 fragments, of which 64 were polymorphic. Number of amplified fragments with ISSR primers ranged from five to ten and varied in size from 180 to 1,900 bp. Percentage polymorphism ranged from 0 to 87.5. The 15 RAPD primers produced 144 bands across 22 genotypes, of which 84 were polymorphic. The number of amplified bands varied from five to 13, with size range from 180 to 2,400 bp. Percentage polymorphism ranged from 0 to 100, with an average of 58.3 across. RAPD markers were relatively more efficient than the ISSR assay. The mental test between two Jaccard’s similarity matrices gave r ≥ 0.90, showing very good fit correlation in between ISSR- and RAPD-based similarities. Clustering of isolates remained more or less the same in RAPD and combined data of RAPD and ISSR. The similarity coefficient ranged from 0.734 to 0.939, 0.563 to 0.946, and 0.648 to 0.920 with ISSR, RAPD, and combined dendrogram, respectively.  相似文献

应用RAPD技术对吐鲁番地区火焰山及艾丁湖区域分离的15株土壤绿藻(chlorophyta)品系的遗传多样性及其亲缘关系进行探讨。结果表明:从20个随机引物中,筛选出多态性和重复性较好且谱带清晰的引物8个,这8个引物扩增出的DNA片段大多在300~2 000 bp之间,所形成的多态性位点数差距较大,显示该区域土壤绿藻具有较丰富的遗传多样性;15株土壤绿藻扩增共得到74条谱带,71条多态性带,其多态性比率为95.95%;聚类分析显示15株土壤绿藻明显地聚为2大类,与其来源相对应,即隶属于同一亚组或相近亚组的不同种基本归为一类,其种间关系与传统的形态学分类结果相吻合。  相似文献

Optimization of primer screening for evaluation of genetic relationship in 34 cultivars of rose through random amplified polymorphic DNA (RAPD) markers was investigated. Four series of decamer primers were used for screening and optimization of RAPD analysis between which A and N series performed good amplification of fragments as compared with other series. The primers OPN-07 and OPN-15 produced maximum number of DNA fragments in Rosa hybrida cv. Anuraag. Some primer either did not produce amplification or produced very poor amplification. Further, ten selected primers were used for genetic analysis of 34 rose cultivars. The primer OPN-15 amplified 21 fragments in all cultivars tested. A total of 162 distinct DNA fragments (bands) ranging from 100 to 3400 base pairs were amplified by using 10 selected random primers. The cluster analysis indicated that these rose cultivars formed nine clusters.  相似文献

Hawthorn ( Crataegus spp.) has a long history as an ornamental and a source of medicine. We report the use of random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) markers to determine genetic relationships in the genus Crataegus . Twenty-eight accessions, including eight species ( Crataegus pinnatifida , Crataegus bretschneideri , Crataegus maximowiczii , Crataegus kansuensis , Crataegus altaica , Crataegus songarica , Crataegus dahurica and Crataegus sanguinea ) and two botanical varieties ( C. pinnatifida var. major and C. maximowiczii var. ninganensis ) were analysed. Twelve RAPD primers reproducibly and strongly amplified 128 fragments of which 116 were polymorphic; similarly, 13 ISSR primers generated 127 products of which 119 were polymorphic. Dendrograms based on unweighted pair group method with arithmetic average analysis were constructed from both the RAPD and the ISSR data. Similarity coefficient based on RAPD and ISSR markers ranged from 0.22 to 0.98 and 0.23 to 0.98, respectively. The range in similarity coefficient indicated that the genus has a high level of genetic diversity. The Mantel test on the similarity matrices produced by RAPD and ISSR markers gave r  = 0.86, showing high correlation between RAPD and ISSR markers in their ability to detect genetic relationships between Crataegus accessions. RAPD and ISSR appear to be reliable methods for the analysis of genetic relationships among hawthorns.  相似文献

Polymerase chain reaction was used, with single 10-mer primers of arbitrary sequence, to amplify random regions of genomic DNA from a diploid cultivated alfalfa backcross population. Segregation of the random amplified polymorphic DNA (RAPD) fragments was analysed to determine if RAPD markers are suitable for use as genetic markers. Of the 19 primers tested, 13 amplified a total of 37 polymorphic fragments, of which 28 (76%) segregated as dominant Mendelian traits. RAPD markers appear useful for the rapid development of genetic information in species like alfalfa where little information currently exists or is difficult to obtain.  相似文献

Abstract

Phosphate solubilizing bacteria (PSB) can convert insoluble form of phosphorous (P) to an available form which is a major concern in Indian agriculture. In this study, 21 isolates having phosphate solubilizing capability were isolated from different regions of Lucknow, India. Among all, six efficient PSB were confirmed by using in vitro P estimation and 16S rRNA universal primers. The similarity detection was done using random amplified polymorphic DNA (RAPD) finger printing for genotyping the PSB isolates and to determine genetic relatedness between them. Twenty different OPA primers were tested among which four primers produced prominent, highly reproducible, and polymorphic bands. An average of 10.5 polymorphic bands per primer with the amplified DNA fragments ranging from 200 to 2000?bp in size. A dendrogram constructed from these data indicated 25–76% homology. Highest similarity was found in between Bacillus anthracis and Bacillus cereus with 33.8% similarity while least dissimilarity was found in B. anthracis and Pseudomonas fragi with 12% of similarity. These findings provide that there is a great genetic diversity between bacterial isolates from different geographical regions and RAPD can be used as a specific, time consuming and also proves as a reliable molecular tool which helps in strain level discrimination.  相似文献
