PCR is a powerful tool in molecular biology, medicine and other fields of science to copy and amplify DNA of any origin. Here we demonstrate an easy way to perform this technique in school based on the „Blue Genes Kit”︁ utilizing two water baths of 94 and 63 °C. As a model experiment we show the possibilities of PCR application in the analysis of gene orientation and detecting a gene without DNA isolation by using an entire organism.  相似文献

Biosensors are obtaining an increasing significance for analytical purposes as well as for on-line monitoring and control in biotechnological processes [1]. In the special field of food industry enzyme and microbial biosensors are suited. With enzyme sensors one can analyse in mostly high specific reactions many substances – microbial sensors normally are unspecific. But they have the advantage of high stability and there is no necessity of enzyme isolation and purification. On the basis of the manifold of microbial metabolic pathways one can expect a wider range of application for microbial sensors, much more as existent for enzyme sensors. For several years now, we can notice efforts in increasing the efficiency of microbial processes by means of more intensive scientific investigation of such processes. Essential prerequisites to it are improved possibilities for process monitoring (available sensors) as well as facilities for realtime processing of process information. In this paper a microcomputer system is represented, which has been constructed on the base of computing needs for fermentation. The computing needs for fermentation experiments are outlined and the structure of the microcomputer is described. As an example it is illustrated what tasks can be solved by this microcomputer in coupled operation with a laboratory fermentor.  相似文献
