This investigation was undertaken because biological studies to evaluate the effects of intermediate frequency magnetic fields are insufficient. White Leghorn fertile eggs (60/group) were either exposed to a 20 kHz, 1.1 mT(rms) sinusoidal magnetic field or sham‐exposed during the first 2, 7, or 11 days of embryogenesis. Lower dose exposures at 0.011 and 0.11 mT(rms) for 2 days were also conducted to elucidate possible dose–response relationships. Additional eggs given all‐trans‐retinoic acid, a teratogen, were exposed to the 1.1 mT(rms) magnetic field for the same periods to investigate the modification of embryotoxicity. After exposure, embryos were examined for mortality and developmental abnormalities. Developmental stage, number of somite pairs, and other developmental endpoints were also evaluated. Experiments were triplicated and conducted in a blind fashion. No exposure‐related changes were found in any of the endpoints in intact embryos exposed to1.1 mT(rms) or to the lower doses of 0.11 and 0.011 mT(rms) magnetic fields. Retinoic acid administration produced embryotoxic responses, which were embryonic death and developmental abnormalities, in 40–60% of embryos in the sham‐exposed groups. The magnitude of these responses was not changed significantly by the magnetic field exposures. Under the present experimental conditions, exposure to 20 kHz magnetic field up to 1.1 mT(rms) was not embryotoxic in the chick and did not potentiate the embryotoxic action of retinoic acid. For in vitro studies on the effect of extremely low frequency (ELF) magnetic field exposures in different laboratories, a programmable, high precision exposure system enabling blinded exposures has been developed and fully characterized. It is based on two shielded 4 coil systems that fit inside a commercial incubator. The volume of uniform B field exposure with 1% field tolerance is 50% larger compared to a Merrit 4 coil system with the same coil volume. The uncertainties for the applied magnetic fields have been specified to be less than 4%. The computer controlled apparatus allows signal waveforms that are composed of several harmonics, blind protocols, monitoring of exposure and environmental conditions and the application of B fields up to 3.6 mT root-mean-square amplitude. Sources of artifacts have been characterized: sham isolation >43 dB, parasitic incident E fields <1 V/m, no recognizable temperature differences in the media for exposure or sham state, and vibrations of the mechanically decoupled dish holder <0.1 m/s(2) (= 0.01 g), which is only twice the sham acceleration background level produced by the incubator and fan vibrations.  相似文献

In studies investigating adverse reproductive outcomes associated with video display terminal (VDT) usage, amounts of time spent in front of a VDT or magnetic field (MF) levels in front of the VDT are used as surrogate measures of subject's MF exposure. However, the relevance of such surrogates to actual exposures has not been demonstrated, and the validity of the use of such measures as a surrogate for the actual MF exposure is only speculative. This study examines 1) measurements of MFs at frequencies of approximately 30–1000 Hz at a fixed distance from the VDTs, 2) reported hours of VDT use, and 3) reported distance between the VDT and the subject's waist as surrogate measures for the average MF exposure level of a VDT user during one 8 h workday. The results showed a weak correlation between the average exposure level of a VDT user and the MF 46 cm from a VDT (R = 0.52, n = 67, P < 0.001). This study showed no association between self-reported hours of VDT usage, or self-reported distance between waist and VDT, and the average MF exposures. Moreover, individuals' average MF exposures did not seem to be affected by other variables, such as position of a VDT on the desk, hours of desk use, and the VDT type (color vs. monochrome). These findings indicate that VDT exposures within office settings are complex and cannot be easily predicted by surrogates. © 1996 Wiley-Liss, Inc.  相似文献

Rats were trained in ten daily sessions to perform in a 12-arm radial maze, which is a behavioral test for spatial memory functions. Exposure to a 60 Hz magnetic field (45 min, 0.75 mT) immediately before each training session retarded learning significantly. Pretreatment with the cholinergic agonist physostigmine before magnetic field exposure reversed the field's effect on spatial learning. Data from this experiment indicate that magnetic field-induced spatial learning deficit is caused by the effect of the field on cholinergic systems. © 1996 Wiley-Liss, Inc.  相似文献

Dr. Peter Valberg has written an excellent and clear paper describing our current understanding of the many facets of characterizing exposure to extremely-low-frequency (ELF) electric and magnetic fields. Dr. Valberg has directed his paper to the characterization of exposures used in laboratory experimentation. In this context, it seems to me that Valberg's program of exposure characterization can be accomplished with currently available instrumentation and with relatively modest effort. Thus, I have no fundamental problems with his recommendations and, really, have only a few minor comments to make. However, if one argues that Valberg's recommendations should be extended to the characterization of exposure in epidemio-logical research, I have serious reservations, which I will mention briefly at the end of this commentary. © 1995 Wiley-Liss, Inc.  相似文献

Bees are fundamentally important for pollination services and declines in populations could have significant economic and environmental implications. Pesticide exposure and pathogen infection are recognised as potential stressors impacting upon bee populations and recently there has been a surge in research on pesticide–disease interactions to reflect environmentally realistic scenarios better. We critically analyse the findings on pesticide–disease interactions, including effects on the survival, pathogen loads and immunity of bees, and assess the suitability of various endpoints to inform our mechanistic understanding of these interactions. We show that pesticide exposure and pathogen infection have not yet been found to interact to affect worker survival under field‐realistic scenarios. Colony‐level implications of pesticide effects on Nosema infections, viral loads and honey bee immunity remain unclear as these effects have been observed in a laboratory setting only using a small range of pesticide exposures, generally exceeding those likely to occur in the natural environment, and assessing a highly selected series of immune‐related endpoints. Future research priorities include the need for a better understanding of pesticide effects on the antimicrobial peptide (AMP) component of an individual's immune response and on social defence behaviours. Interactions between pesticide exposure and bacterial and fungal infections have yet to be addressed. The paucity of studies in non‐Apis bee species is a further major knowledge gap.  相似文献

This article addresses a classic ethnographic problem in the study of Italy: how is it that people can subscribe simultaneously to seemingly contradictory ideologies, such as Catholicism and Communism? It does so by describing examples from Italy's ‘showcase city’ of the left, ‘Red Bologna’, in which to be ‘red’ is ubiquitous but each person's ‘red’ is a different thing: being ‘red’ (differently) is the idiom in which real political distinctions are expressed over issues like religion or immigration. In parallel, I discuss the relationship between the ‘field’ as a location and the ‘field’ as a conceptual topic. My account replicates internal ethnographic differences at the analytical level by highlighting the differences between being left‐wing in Bologna and its meaning as a concept in anthropology. Hence the ‘equivocal location’: a field‐site that is productively different, from what an inexperienced ethnographer expected from it, from conceptual discussions in anthropology, and from itself.  相似文献

A recent study examining the relationship between distance to nearby power lines and childhood cancer risk re‐opened the debate about which exposure metrics are appropriate for power frequency magnetic field investigations. Using data from two large population‐based UK and German studies we demonstrate that distance to power lines is a comparatively poor predictor of measured residential magnetic fields. Even at proximities of 50 m or less, the positive predictive value of having a household measurement over 0.2 µT was only 19.4%. Clearly using distance from power lines, without taking account of other variables such as load, results in a poor proxy of residential magnetic field exposure. We conclude that such high levels of exposure misclassification render the findings from studies that rely on distance alone uninterpretable. With increasing electrical device usage, social concerns about the possible effects of 60 Hz electromagnetic fields on human health have increased. The number of people with self‐attributed electromagnetic hypersensitivity (EHS) who complain of various subjective symptoms such as headache and insomnia has also increased. However, it is unclear whether EHS results from physiological or other origins. In this double‐blinded study, we simultaneously investigated physiological changes (heart rate, respiration rate, and heart rate variability), subjective symptoms, and perception of the magnetic field to assess origins of the subjective symptoms. Two volunteer groups of 15 self‐reported EHS and 16 non‐EHS individuals were tested with exposure to sham and real (60 Hz, 12.5 µT) magnetic fields for 30 min. Magnetic field exposure did not have any effects on physiological parameters or eight subjective symptoms in either group. There was also no evidence that the EHS group perceived the magnetic field better than the non‐EHS group. In conclusion, the subjective symptoms did not result from the 60 Hz, 12.5 µT magnetic field exposures but from other non‐physiological factors. In occupational environments, an increasing number of electromagnetic sources emitting complex magnetic field waveforms in the range of intermediate frequencies is present, requiring an accurate exposure risk assessment with both in vitro and in vivo experiments. In this article, an in vitro exposure system able to generate complex magnetic flux density B‐fields, reproducing signals from actual intermediate frequency sources such as magnetic resonance imaging (MRI) scanners, for instance, is developed and validated. The system consists of a magnetic field generation system and an exposure apparatus realized with a couple of square coils. A wide homogeneity (99.9%) volume of 210 × 210 × 110 mm3 was obtained within the coils, with the possibility of simultaneous exposure of a large number of standard Petri dishes. The system is able to process any numerical input sequence through a filtering technique aimed at compensating the coils' impedance effect. The B‐field, measured in proximity to a 1.5 T MRI bore during a typical examination, was excellently reproduced (cross‐correlation index of 0.99). Thus, it confirms the ability of the proposed setup to accurately simulate complex waveforms in the intermediate frequency band. Suitable field levels were also attained. Moreover, a dosimetry index based on the weighted‐peak method was evaluated considering the induced E‐field on a Petri dish exposed to the reproduced complex B‐field. The weighted‐peak index was equal to 0.028 for the induced E‐field, indicating an exposure level compliant with the basic restrictions of the International Commission on Non‐Ionizing Radiation Protection. Iris validation is a Python package created to represent comprehensive per‐residue validation metrics for entire protein chains in a compact, readable and interactive view. These metrics can either be calculated by Iris, or by a third‐party program such as MolProbity. We show that those parts of a protein model requiring attention may generate ripples across the metrics on the diagram, immediately catching the modeler's attention. Iris can run as a standalone tool, or be plugged into existing structural biology software to display per‐chain model quality at a glance, with a particular emphasis on evaluating incremental changes resulting from the iterative nature of model building and refinement. Finally, the integration of Iris into the CCP4i2 graphical user interface is provided as a showcase of its pluggable design.  相似文献

Human exposure to magnetic fields, increased through use of new technologies like magnetic resonance imaging (MRI), has prompted investigations into possible effects of static magnetic fields (SMFs) on cellular processes. However, controversy still remains between many studies, which likely results from a lack of uniformity across experimental parameters, including the length of magnetic field exposure, the strength of the magnetic field, and the cell type or organism under investigation. The purpose of this research was to monitor effects of SMF exposure using real‐time luminescence photometry. The study investigated the potential interaction of a 100 mT SMF on a heat shock protein (hsp70)/luciferase reporter construct in stably transfected NIH3T3 cells. Changes in heat shock promoter activation following 100 mT SMF exposure were analyzed and detected as bioluminescence in real‐time. Two heat parameters were considered in combination with sham‐ and 100 mT‐exposed experiments: no heat or 1,800 s heat. As expected, there was a significant increase in bioluminescence in response to 1,800 s of heat alone. However, no significant difference in average hsp70 promoter activation between sham and 100 mT experiments was observed for no heat or 1,800 s heat experiments. Therefore, a 100 mT SMF was shown to have no effect on the activation of the heat shock protein promoter during SMF exposure or when SMF exposure was combined with a heat insult. J. Cell. Disruption of communication between transformed cells and normal cells is involved in tumor promotion. We have tested the hypothesis that exposures to radiofrequency (RF) fields using a form of digital modulation (TDMA) and a chemical tumor promoter, 12-O-tetradecanoylphorbol-13-acetate (TPA), are copromoters that enhance focus formation of transformed cells in coculture with parental C3H/10T1/2 murine fibroblasts. RF field exposures did not influence TPA's dose-dependent promotion of focus formation in coculture. Cell cultures were exposed to an 836.55 MHz TDMA-modulated field in TEM transmission line chambers, with incident energies that simulated field intensities at a user's head. Specific absorption rates (SARs) of 0.15, 1.5, and 15 μW/g were used during each digital packet, and the packet frequency was 50/s. The TEM chambers were placed in a commercial incubator at 37 °C and 95% humidity/5% CO2. The RF field exposures were in a repeating cycle, 20 min on, 20 min off, 24 h/day for 28 days. At 1.5 μW/g, TPA-induced focus formation (at 10, 30, and 50 ng/ml) was not significantly different in RF-exposed cultures compared to parallel sham-exposed cultures in ten independent experiments in terms of the number, density, and area of foci. Similarly, at 0.15 and 15.0 μW/g, in two and four experiments, respectively, RF exposure did not alter TPA-induced focus formation. The findings support a conclusion that repeated exposures to this RF field do not influence tumor promotion in vitro, based on the RF field's inability to enhance TPA-induced focus formation. A programmable system has been developed for the study of both transient and persistent effects of extremely low frequency (ELF) magnetic field exposure of cell cultures. This high‐precision exposure system enables experimental blinding and fully characterized exposure while simultaneously allowing live cell imaging. It is based on a live imaging cell around which two asymmetrical coils are wound in good thermal contact to a temperature‐controlled water jacket, and is mounted on a microscope stage insert. The applied B‐field uniformity of the active volume is better than 1.2% with an overall exposure uncertainty of less than 4.3% with very low transient field levels. The computer‐controlled apparatus allows signal waveforms that are sinusoidal or composed of several harmonics, blind protocols, and monitoring of exposure and environmental conditions. B‐fields up to 4 mT root mean square amplitude are possible with minimal temperature variation and no recognizable temperature differences between exposure and sham states. Sources of artifacts have been identified and quantified. There are no visible vibrations observable even at the highest magnifications and exposure levels. The effects of extremely low frequency (ELF) magnetic fields on membrane F0F1‐ATPase activity have been studied. When the F0F1‐ATPase was exposed to 60 Hz magnetic fields of different magnetic intensities, 0.3 and 0.5 mT magnetic fields enhanced the hydrolysis activity, whereas 0.1 mT exposure caused no significant changes. Even if the F0F1‐ATPase was inhibited by N,N‐dicyclohexylcarbodiimide, its hydrolysis activity was enhanced by a 0.5 mT 60 Hz magnetic field. Moreover, when the chromatophores which were labeled with F‐DHPE were exposed to a 0.5 mT, 60 Hz magnetic field, it was found that the pH of the outer membrane of the chromatophore was unchanged, which suggested that the magnetic fields used in this work did not affect the activity of F0. Taken together, our results show that the effects of magnetic fields on the hydrolysis activity of the membrane F0F1‐ATPases were dependent on magnetic intensity and the threshold intensity is between 0.1 and 0.3 mT, and suggested that the F1 part of F0F1‐ATPase may be an end‐point affected by magnetic fields. Microwave‐induced corneal endothelial damage was reported to have a low threshold (2.6 W/kg), and vasoactive ophthalmologic medications lowered the threshold by a factor of 10–0.26 W/kg. In an attempt to confirm these observations, four adult male Rhesus monkeys (Macaca mulatta) under propofol anesthesia were exposed to pulsed microwaves in the far field of a 2.8 GHz signal (1.43 ± 0.06 µs pulse width, 34 Hz pulse repetition frequency, 13.0 mW/cm2 spatial and temporal average, and 464 W/cm2 spatial and temporal peak (291 W/cm2 square wave equivalent) power densities). Corneal‐specific absorption rate was 5.07 W/kg (0.39 W/kg/mW/cm2). The exposure resulted in a 1.0–1.2 °C increase in eyelid temperature. In Experiment I, exposures were 4 h/day, 3 days/week for 3 weeks (nine exposures and 36 h total). In Experiment II, these subjects were pretreated with 0.5% Timolol maleate and 0.005% Xalatan® followed by 3 or 7 4‐h pulsed microwave exposures. Under ketamine–xylazine anesthesia, a non‐contact specular microscope was used to obtain corneal endothelium images, corneal endothelial cell density, and pachymetry at the center and four peripheral areas of the cornea. Ophthalmologic measurements were done before and 7, 30, 90, and 180 days after exposures. Pulsed microwave exposure did not cause alterations in corneal endothelial cell density and corneal thickness with or without ophthalmologic drugs. Therefore, previously reported changes in the cornea exposed to pulsed microwaves were not confirmed at exposure levels that are more than an order of magnitude higher. Indoor Residual Spray (IRS) with insecticides has been a procedure used for decades to protect humans from biting mosquitoes and potential vectors of pathogens. The objective of this study was to determine the responses of three wild‐caught species of malaria vectors exposed to pyrethroids of three different surface coverage percents using an excito‐repellency test box. Each species was exposed to three insecticide‐treated surfaces at varying exposure levels (full coverage, 50%, and 25% of the maximum allowable by the test system) to a single standard field dose of either lambda‐cyhalothrin or alpha‐cypermethrin. Larger numbers of mosquitoes escaped the treated chambers in the direct contact test compared to the spatial repellent chambers in all three different treated surface exposures. No significant differences in the percent of escaped mosquitoes were detected in the 50% and full coverage surface coverage exposures, whereas the 25% coverage produced significantly lower avoidance responses for both compounds. This study found that varying levels of surface exposure with synthetic pyrethroids can impact the behavioral avoidance responses of Anopheles; however, it may also be possible to reduce the amount of coverage to achieve similar avoidance actions. This information may assist policy makers in designing more cost effective strategies involving residual insecticides to control mosquito vectors.  相似文献

An expression was derived for the magnetic field between Helmholtz coils, and used to determine the intercoil region within which the field's spatial uniformity is below an arbitrary value. With different prevalence in different regions, radio frequency (RF) electromagnetic fields (EMF) are widely used for therapeutic tissue heating. Although short‐wave diathermy (27.12 MHz) is the most popular treatment modality, quantitative data on patient's exposure have been lacking. By numerical simulation with the numerical anatomical model NORMAN, intracorporal distributions of specific absorption rates (SAR) were investigated for different treatment scenarios and applicators. Quantitative data are provided for exposures of target treatment areas as well as for vulnerable regions such as the eye lenses, central nervous system, and testes. Different applicators and distances were investigated. Capacitive and inductive applicators exhibit quite a different heating efficiency. It could be shown that for the same output power therapeutic heat deposition can vary by almost one order of magnitude. By mimicking therapist's practice to use patient's heat perception as an indicator for output power setting, numerical data were elaborated demonstrating that muscle tissue exposures may be several times higher for inductive than for capacitive applicators. Presented quantitative data serve as a guide for power adjustment preventing relevant overexposures without compromising therapy; they also provide a basis for estimating target tissue heat load and developing therapeutic guidelines.
